Our group previously synthesized 99mTc-labeled fatty acids suitable for myocardial metabolism and flow imaging. In this set of experiments, 29 new analogues were synthesized according to the 4 + 1 mixed ligand approach with some specific differences. Conventional 4 + 1 99mTc-fatty acids are built in the sequence: Tc-chelate, alkyl chain, and carboxylic group. We developed compounds following a new design with the sequence: carboxylic group, alkyl chain, Tc-chelate, and lipophilic tail. Therefore, the 99mTc-chelate was transferred to a more central position of the compound, aiming toward an improved myocardial profile and an accelerated liver clearance. In this context, several functional groups incorporated in the lipophilic tail section were tested to evaluate their influence on the compound's character. In addition to biodistribution studies in vivo, the myocardial first-pass extraction of the compounds was tested in an isolated Langendorff rat heart model. A satisfactory myocardial uptake of up to 20% of the injected dose (% ID) in the perfused heart and a fast liver clearance in vivo with only 0.29% ID/g at 60 min postinjection demonstrate that the induced molecular modifications affect the kinetics of 99mTc-radiolabeled fatty acid compounds favorably. From the data set, rules for estimating the biodistribution of fatty acids tracers are deduced.
The Perfectly Matched Layer (PML) method is an efficient approach to imposing radiation conditions at the bounded region of interest in case of wave propagation in unbounded domains. This paper presents and validates 3D FE/PML numerical schemes based on two different PML formulations for homogeneous and inhomogeneous geological media exhibiting discrete or continuous inhomogeneity. In the equation of motion for the PML domain the applied stretching behavior is expressed either as complex material properties or as complex coordinates. Both PML formulations are implemented in the FEM and verified against analytical solutions. Three different types of material inhomogeneity are considered: layered half-space, continuously inhomogeneous half-space with linear velocity profile and continuously inhomogeneous half-space with nonlinear velocity profile. Sensitivity analyses are conducted, and the performance of the developed numerical schemes is investigated taking into account a broad variation of the PML parameters. Recommendations are given for the optimal values of the PML parameters for the case of homogeneous and inhomogeneous geological media.
Unter Berücksichtigung praktischer und wissenschaftlich-technischer Erkenntnisse der letzten 15 Jahre - insbesondere auch auf der Basis verschiedener BAM-Forschungsergebnisse und Zulassungsverfahren - erfolgte eine Überarbeitung der Grundlagen, Bauprinzipien, Ausführungs- und Einbaukriterien sowie der Zulassungs- und Gütesicherungsmaßnahmen für insgesamt 4 Bauarten von Fugenfüllsystemen für hochbeanspruchte Verkehrsflächen. In diesem - für den Bundesfernstraßenbereich verbindlichen bautechnischen Regelwerk - finden insbesondere auch die Ergebnisse mehrerer wissenschaftlicher Vorhaben der BAM bezüglich labortechnischer Untersuchungen und Felduntersuchungen zum Funktionsverhalten und zur Dauerhaftigkeit erste baurechtliche Konsequenzen. Auf der Basis von Untersuchungen u.a. im Brückenbau wurden Regelungen für die Fugenausbildung in Asphalt und bei Befestigungskombinationen erarbeitet und umgesetzt. Damit konnten umfassende Regelungen für die maßgebenden Fahrbahnbeläge im Bundesfernstraßenbereich der Straßenbauverwaltung und ausführenden Industrieunternehmungen zur Verfügung gestellt werden. Die Neufassung dieser Regelungen dient darüber hinaus dem Ziel, den von der BAM vertretenen Wechsel hin zu einer performance-bezogenen Bewertungsmethodik, vorzubereiten.
Nach mehr als 15-jähriger Anwendung und mit der zwischenzeitlichen Einführung europäisch harmonisierter Normen für Fugenmaterialien war eine Anpassung Qualitäts- und Gütesicherungsanforderungen für Fugenfüllungen im Bundesfernstraßenbereich erforderlich. Hierbei ging es insbesondere darum, Maßnahmen und Verfahren zu entwickeln, um das nationale Qualitäts- und Gütesicherungsniveau weiter zu entwickeln und weiterhin sicher stellen zu können. Nach den Untersuchungserfahrungen im Labor und im Feld ging darum, neuartige Material- und Stoffkombinationen qualitätssicher dem Bauprozess zur Verfügung stellen zu können. Erforderliche Anforderungskriterien und zulässige Toleranzen der Lieferqualität wurden neu gefasst. Es wurde ein Verfahren entwickelt, europäische Bauprodukte qualitätsgesichert in den Bauprozess im Bundesfernstraßenbericht zu überführen.
In a proof of concept study, metal-coded affinity tags based on click chemistry (MeCAT-Click) were used to analyze the proteome of Escherichia coli (E. coli) in response to heat stress. This allows high labeling efficiency, high detection sensitivity, and multiplex capabilities, which are pivotal for its application to protein quantification. Two approaches are presented for relative quantification of differentially lanthanide-labeled proteins. The first approach uses isotope-labeling, where ESI-MS was utilized to quantify the differentially labeled proteins from different states of E. coli. With this approach, 14 proteins were found with changed abundance, among them five proteins upregulated.
In the second approach, differentially labeled samples were separated by two dimensional gel electrophoresis (2 DE) and scanned by laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS). Comparison of the signal intensities of the different lanthanides was used to quantify different sample states. Based on this information, ESI-MS was used to identify the proteins with different abundance. The sensitivity of LA-ICP-MS allowed us to find one upregulated protein that was nearly invisible by silver staining ("Probable replication endonuclease from retron EC67"). The advantage of this approach is to locate low abundant proteins with differential expression using LA-ICP-MS, which may be overlooked otherwise.
Biological significance: This paper demonstrates the successful application of a novel metal labeling strategy to quantify the proteins from complex biological samples. In comparison with former metal labeling strategies, it reduces the steric hindrance and improves the labeling efficiency during the labeling process, which ensure its successful application. This methodology is compatible with both molecular and elemental mass spectrometry. ESI-MS/MS in combination with software-based search allows the identification and relative quantification of labeled proteins. In addition, LA-ICP-MS helps to locate the labeled proteins in 2-DE gels with superior detection capability, thus, target proteins with low abundance can be precisely followed. Its excellent sensitivity allows one to track the proteins of interest that are barely visible by silver staining.
Iridium oxide is the catalytic material with the highest stability in the oxygen evolution reaction (OER) performed under acidic conditions. However, its high cost and limited availability demand that IrO2 is utilized as efficiently as possible. We report the synthesis and OER performance of highly active mesoporous IrO2 catalysts with optimized surface area, intrinsic activity, and pore accessibility. Catalytic layers with controlled pore size were obtained by soft-templating with micelles formed from amphiphilic block copolymers poly(ethylene oxide)-b-poly(butadiene)-b-poly(ethylene oxide). A systematic study on the influence of the calcination temperature and film thickness on the morphology, phase composition, accessible surface area, and OER activity reveals that the catalytic performance is controlled by at least two independent factors, that is, accessible surface area and intrinsic activity per accessible site. Catalysts with lower crystallinity show higher intrinsic activity. The catalyst surface area increases linearly with film thickness. As a result of the templated mesopores, the pore surface remains fully active and accessible even for thick IrO2 films. Even the most active multilayer catalyst does not show signs of transport limitations at current densities as high as 75 mA cm-2.
Microplastics can affect biophysical properties of the soil. However, little is known about the cascade of events in fundamental levels of terrestrial ecosystems, i.e., starting with the changes in soil abiotic properties and propagating across the various components of soil−plant interactions, including soil microbial communities and plant traits. We investigated here the effects of six different microplastics (polyester fibers, polyamide beads, and four fragment types: polyethylene, polyester terephthalate, polypropylene, and polystyrene) on a broad suite of proxies for soil health and performance of spring onion (Allium fistulosum). Significant changes were observed in plant biomass, tissue elemental composition, root traits, and soil microbial activities. These plant and soil responses to microplastic exposure were used to propose a causal model for the mechanism of the effects. Impacts were dependent on particle type, i.e., microplastics with a shape similar to other natural soil particles elicited smaller differences from control. Changes in soil structure and water dynamics may explain the observed results in which polyester fibers and polyamide beads triggered the most pronounced impacts on plant traits and function. The findings reported here imply that the pervasive microplastic contamination in soil may have consequences for plant performance and thus for agroecosystems and terrestrial biodiversity.
