Antibiotic resistance is a growing global problem which poses a massive threat to human health. Although human activity contributes to the acceleration of the process, bacteria have a self-driven stabilisation mechanism to protect themselves from such and other external threats: biofilm formation. Nonetheless, it is the adhesion of a single bacterial cell to a surface that triggers the formation of such network of biomolecules and microorganisms, as well as its hazardous consequences. The main objective of this work was to quantify the adhesion force of a single E. coli cell on a Ti substrate via the AFM-related single-cell force spectroscopy, with both the cell and the substrate material being of high clinical relevance. A set of 25 x 25 force displacement curves was acquired with a maximum force of 3.2 nN without dwell time, yielding a topography map and an adhesion force map that showed to be correlated. A mean adhesion force of 0.85 ± 0.175 nN was measured and the presence of cell appendages on the bacterial cell wall was verified through individual force-displacement curves. Bacterial viability was assessed after the measurements via live/dead staining.
The application of fracture mechanics to the determination of the fatigue behavior of weldments is discussed with the focus on classic fatigue, i.e., the overall fatigue life and the fatigue strength in terms of an S-N curve and the endurance limit. The following issues are addressed: specific features of short fatigue crack propagation, an adequate initial crack size, multiple crack propagation and its statistical treatment as well as welding residual stresses. As an example, an approach of the authors is applied to the determination of FAT classes for a butt weld with varying weld toe geometry.
Water is generally considered to be an undesirable substance in fuel system, which may lead to microbial contamination. The antibacterial strategies that can turn water into things of value with high disinfection efficacy have been urgently needed for fuel system. Here, we reveal a water-fueled autocatalytic bactericidal pathway comprised by bi-metal micro-electrode system, which can spontaneously produce reactive oxygen species (mainly H2O2 and O2•–) by the electron Fenton-like reaction in water medium without external energy., The respiratory chain component of bacteria and the galvanic corrosion on the coated metals were two electron sources in the system. The specific model of Ag-Ru water-fueled autocatalytic (WFA) microelectrode particles presents extremely high disinfection efficiency (>99.9999%) in less than one hour for three aerobic bacteria (Escherichia coli, Pseudomonas aeruginosa and Bacillus subtilis) in LB media and high disinfection efficiency for the anaerobic bacteria (Desulfovibrio alaskensis) in Postgate E media without natural light irradiation. Overall, the novel WFA Ag-Ru antibacterial material explored in this study has a high potential for sterilizing applications in fuel system and this work provides the potential for the development of non-chemical and water-based antibacterial materials, such as WFA Ag-Ru antibacterial coating on stainless steel.
Aim of study: In this study, natural durability of some domestic and foreign wood species against Hylotrupes bajulus and Anobium punctatum larvae were tested on laboratory scale
Area of study: This study was conducted at Department of Forest Products Engineering in Duzce University, Turkey and Federal Institute for Materials Research and Testing (BAM), Germany.
Material and Methods: Scotch pine (Pinus sylvestris), fir (Abies nordmanniana), spruce (Picea orientalis), cedar (Cedrus libani), poplar (Populus tremula) and beech (Fagus orientalis) woods were used to test H. bajulus larvae (EN 46-1). Alder (Alnus glutinosa), oak (Quercus cerris), poplar (Populus tremula), beech (Fagus orientalis), maple (Acer carpinifolium), ash (Fraxinus angustifolia), teak (Tectona grandis), ayous (Triplochiton scleroxylon), movingui (Distemonanthus benthamianus), dahoma (Piptadeniastrum africanum), iroko (Chlorophora excelsa), bubinga (Guibourtia tessmannii) and sapele (Entandrophragma cylindiricum) woods were used for A. punctatum larvae (EN 49-1). At the end of the experiment, the mortality rates of the larvae were determined and the size and weights of the surviving larvae were measured.
Main results: F. orientalis and C. libani were found to be the most resistant wood species against H. bajulus larvae while A. nordmanniana was the least resistant. All tropical wood species and oak and maple from domestic wood species showed 100% mortality rate therefore found to be the most resistant against A. punctatum larvae. The most vulnerable wood species was found to be alder with a 35% mortality.
Research highlights: while F. orientalis, C. libani, and P. tremula were found the most resistance wood species against H. bajulus, P. sylvestris and A. nordmanniana were determined as most vulnerable. All tropical wood species and two domestic species (Q. cerris and A. carpinifolium) showed the highest mortality rate as 100%. The least durable domestic wood was determined as alder.
The characteristics of different molecules chosen as representatives for specific functionalities in conditioning layers play an important role on attachment behavior and later biofilm formation of bacteria. The chemical composition is a major component influencing the attachment but there is a conglomerate of influences.
Im Januar 2018 wurde die DIN EN 16516 „Bauprodukte: Bewertung der Freisetzung gefährlicher Stoffe – Bestimmung von Emissionen in die Innenraumluft“ als harmonisierte europäische Prüfnorm veröffentlicht. Für die Etablierung der DIN EN 16516 als neue Referenznorm wurden verschiedene Emissionsprüfungen mit dem Ziel durchgeführt, ein neues Prüfverfahren für Formaldehydemissionen aus Holzwerkstoffen zu erarbeiten. Das übergeordnete Ziel ist die Minimierung des Risikos von Überschreitungen des Innenraumrichtwertes für Formaldehyd. Die Untersuchungen ergaben teilweise hohe Formaldehydemissionen, insbesondere bei höheren Beladungsfaktoren und Temperaturen, sowie bei niedrigen Luftwechselraten. Eine getestete Spanplatte hätte aufgrund ihrer hohen Formaldehydemission nicht auf den deutschen Markt gebracht werden dürfen. Durch vergleichende Untersuchungen konnte ein Umrechnungsfaktor in Höhe von 2,0 für die Umrechnung von Prüfwerten nach DIN EN 717-1 zu DIN EN 16516 abgeleitet werden.
UBA und BAM schlagen die Durchführung der Prüfkammermessungen bei 23 °C und 50% relativer Luftfeuchte, einer Beladung von 1,8 m²/m³ und einem Luftwechsel von 0,5 pro Stunde vor. Diese Parameter orientieren sich an der EN 16516, die als zukünftige „Referenznorm“ zu betrachten ist. Die Schmalflächenversiegelung ist auch zukünftig wie in der EN 717-1 beschrieben vorzunehmen. Nach 28 Tagen wird der Mittelwert einer Doppelbestimmung als Endkonzentration („entsprechend der Ausgleichskonzentration“) berechnet. Alternativ dazu kann die Prüfung vorzeitig abgebrochen werden, wenn an 5 Messtagen der Prüfung eine Formaldehydkonzentration von 0,1 ppm nicht überschritten wird.
Prüfungen nach der EN 717-1 sollen weiterhin gleichberechtigt möglich sein. Ergebnisse von Messungen, die nach der EN 717-1 ermittelt wurden, sind mit dem Faktor 2,0 zu multiplizieren.
Abgeleitete Verfahren wie z.B. das Gasanalyseverfahren sollen weiterhin möglich sein. Das Perforatorverfahren entfällt hier, da eine allgemeine für alle Holzwerkstoffe gültige Korrelationen nicht existiert.
Die entsprechenden Änderungen sind in der vom BMUB veröffentlichten „Bekanntmachung analytischer Verfahren für Probenahmen und Untersuchungen für die im Anhang der Chemikalien-Verbotsverordnung genannten Stoffe und Stoffgruppen“ entsprechend zu ändern.
Comparison of Formaldehyde Concentrations in Emission Test Chambers Using EN 717-1 and EN 16516

For more than 25 years EN 717-1 (Wood-based panels - Determination of formaldehyde re-lease - Part 1: Formaldehyde emission by the chamber method) is the standard for formalde-hyde emission testing of wooden boards. In 2017 EN 16516 (Construction products - Assess-ment of release of dangerous substances - Determination of emissions into indoor air) was published as a new harmonised standard for the emission testing of construction products. Because test chamber conditions are different, both standards give different concentrations for formaldehyde. To determine a conversion factor four test series were set up with different wooden boards. For a loading of 1 m²/m³ the conversion factor is 1.6. This means that the formaldehyde concentration measured under the conditions of EN 16516 is a factor of 1.6 higher compared to EN 717-1.
Emission testing of volatile organic compounds (VOC) from materials and products is commonly based on emission test chamber measurements. To ensure the comparability of results from different testing laboratories, their measurement performance must be verified. For this purpose, Bundesanstalt für Materialforschung und -prüfung (BAM) organizes an international proficiency test (round robin test, RRT) every two years using well-characterized test materials (one sealant, one furniture board, and four times a lacquer) with defined VOC emissions. The materials fulfilled the requirements of homogeneity, reproducibility, and stability. Altogether, 36 VOCs were included of which 33 gave test chamber air concentrations between 13 and 83 µg/m3. This is the typical concentration range to be expected and to be quantified when performing chamber tests. Three compounds had higher concentrations between 326 and 1105 µg/m3. In this paper, the relative standard deviations (RSD) of BAM round robin tests since 2008 are compared and the improvement of the comparability of the emission chamber testing is shown by the decrease of the mean RSD down to 28 % in 2018. In contrast, the first large European interlaboratory comparison in 1999 showed a mean RSD of 51 %.
