The antibody A.1.1.1 was labeled and found to be very sensitive and highly selective for TNT. A novel monolithic affinity column was coated with a Trinitroaniline (TNA)-BSA affinity conjugate and a custom laser induced fluorescence detector were built to allow for continuous and sensitive detection. The affinity column combined with the high sensitivity detector resulted in a limit of detection of approx. 100 pM TNT or 20 ppt TNT for offline detection and was able to detect picogram amounts within three minutes.
Die vom BMBF unterstütze Forschungsinitiative BonaRes entwickelt Strategien um Boden als nachhaltige Ressource für die Bioökonomie zu nutzen. Das Projekt Intelligence for Soil (I4S) ist ein Teil dieser Initiative und beschäftigt sich mit der Entwicklung eines integrierten Systems zur ortsspezifischen Düngung. Ein Ziel dieses Projektes ist die Etablierung einer mobilen Sensorplattform zur Bodenkartierung. Aufgabe der BAM ist dabei das Installieren und die Optimierung eines RFA Sensors für die online Messung direkt auf dem Feld. Die RFA ist eine zerstörungsfreie Technik, die kaum Probenvorbereitung benötigt und einen schnelle Multielement Analyse ermöglicht. Das schnelle Kartieren des Bodens ermöglicht später eine ortsspezifische Düngung mit Nährstoffen wie z.B. K, Ca und P.

Der Einsatz moderner Prozessanalysentechnik führt zu einer Datenflut, deren Verarbeitung und Nutzung häufig nicht ohne den Einsatz von Chemometrie zu realisieren ist. Dafür stehen zahlreiche multivariate Verfahren, wie z.B. Hauptkomponentenanalyse (PCA) und Regressionsanalyse (PLSR) für die Klassifizierung und für die quantitative Analyse zur Verfügung. Darüber hinaus können in Spezialfällen, insbesondere in der Spektroskopie, Methoden des „Indirect Hard Modeling“ eingesetzt werden. Anhand einer Industrie-relevanten Reaktion, der Hydroformylierungsreaktion zur Bildung langkettiger Aldehyde aus der Reaktion von Alkenen und Synthesegas, die sowohl im Labormaßstab als auch in einer Miniplant in Kooperation mit der TU Berlin untersucht wurde, werden Kalibrationsstrategien (Design of Experiment, DoE) und Auswertungsansätze mittels PLSR prozessbegleitender Raman Spektren dargelegt. Auf Grundlage des chemometrischen Ansatzes ist eine on-line Überwachung der Reaktion hinsichtlich der Hauptreaktanden, 1-Dodecene und 1-Tridecanal, möglich. Weiterhin konnte mittels Raman Spektroskopie die Bildung eines unerwünschten Nebenprodukts nachgewiesen werden.
The main task of BAM in the I4S project is the development of online analytics for reliable monitoring of major and minor nutrients like K, P, Ca in soil using mobile-XRF (X-ray fluorescence spectroscopy) and mobile-LIBS (laser-induced breakdown spectroscopy). Each method provides fast and simultaneous multi-element analyses which are neccessary for mapping agricultural cropland. The first objective was to design a combined measurement setup for XRF and LIBS. The centerpiece is the rotatable sample plate which enables the measurement of moving samples. Current steps are the optimisation of the parameters for the analysis of soils.

Currently, there is almost no comprehensive mapping of agricultural cropland because of the lack of fast and affordable mapping methods for important soil properties. Arable land of some hectares in size show a broad range of different nutrient compositions. Because of this heterogeneity wrong fertilization can occur and can cause environmental pollution or lead to smaller harvests and this is simply a waste of resources. The goal of I4S (intelligence for soil) is to develop an integrated system for site-specific soil fertility management. The I4S consortium consists of 10 different institutions, which are testing various sensors for their suitability for the requested applications. Besides the preparation of reference materials, the main task of the Federal Institute for Materials Research and Testing (BAM) in this project is the method development for online-XRF (x-ray fluorescence spectroscopy) and for online-LIBS (laser-induced breakdown spectroscopy) sensor systems. Both methods have the advantage that there is no or only little sample preparation necessary. Each method provides a fast and simultaneous multi-element analysis. Both measurement setups can be utilized for a mobile application which is fundamental for reaching the goals of I4S. Having finally a set of some reference materials, further work will focus on the online-XRF and online-LIBS methods in order to obtain useable calibration models. The calibration models will then be tested using stationary and moving samples.
Der Beitrag beinhaltet die Vorstellung eines gemeinsamen Projekts zwischen der BAM und der TU Berlin (Process Dynamics and Operations Group). Mittels Raman Spektroskopie werden Hydroformylierungsreaktionen in einer Miniplant der TU prozessbegleitend untersucht. Kalibrationsstrategien, Laboruntersuchungen sowie Vorhersagen von Prozessmessungen basierend auf chemometrischen Modellen werden vorgestellt. Zum gegenwärtigen Zeitpunkt wird die Strategie als erfolgsversprechend bewertet, jedoch sind weitere Untersuchungen insbesondere zum Einfluss der Microemulsion auf die Ramanspektren notwendig.
