Unter den rauen Bedingungen in der Automobilkarosseriefertigung lassen sich schweißbedingte Imperfektionen wie Risse beim Widerstandspunktschweißen von hochfesten Stählen nicht immer vermeiden. Dabei ist der Einfluss solcher Risse auf die Schwingfestigkeit insbesondere von modernen hochfesten austenitischen Stählen derzeit nicht hinreichend bekannt. Im Rahmen dieser Arbeit wurde daher der Einfluss von Schweißrissen verschiedener Lage und Ausbildung untersucht. Dabei konnte durch die Analyse der normierten Steifigkeitsverläufe von Proben sowie durch die Gegenüberstellung der Versagensschwingspielzahlen nachgewiesen werden, dass die spritzerfrei erzeugten Oberflächenrisse im Zentrum, im Übergangsbereich sowie im Randbereich der Schweißlinse keinen negativen Einfluss auf die Schwingfestigkeit des hier untersuchten hochfesten austenitischen Werkstoffs haben. Proben, die mit Schweißspritzern hergestellt wurden und Risse im Randbereich aufweisen, zeigen deutliche höhere Versagensschwingspielzahlen als rissfreie Referenzproben. ----------------------------------------------------------------------------------------------------------------------------------------------
In the rough conditions in the fabrication of automobile bodies, it is not always possible to avoid welding-induced imperfections such as cracks during the resistance spot welding of highstrength steels. In this respect, the influence of such cracks on the fatigue strength particularly of modern high-strength austenitic steels is not sufficiently well-known at present. The influence of welding cracks with various positions and formations was therefore investigated within the framework of this paper. In this case, the analysis of the standardised stiffness courses of specimens and the comparison of the numbers of failure stress cycles served to prove that the surface cracks produced without any spatter in the centre, interfacial region and peripheral region of the weld nugget do not have any negative influence on the fatigue strength of the high-strength austenitic material investigated here. Specimens which were manufactured with welding spatter and exhibit cracks in the peripheral region show considerably higher numbers of failure stress cycles than crack-free reference specimens.

Bei hochfesten austenitischen Stählen können unter den Produktionsbedingungen in der Rohkarosseriefertigung schweißbedingte Risse beim Widerstandspunktschweißen entstehen. Der Einfluss solcher Risse auf die dynamische Festigkeit ist derzeit nicht hinreichend bekannt. Im Rahmen dieser Arbeit wurden daher Schweißrisse verschiedener Lage und Ausbildung hinsichtlich ihrer Wirkung auf die Festigkeit und das Bruchverhalten von Widerstandspunktschweißverbindungen im Hochgeschwindigkeitsscherzug untersucht. Dabei konnte durch die Analyse der Kraft-Weg-Verläufe, der Scherzugfestigkeit und des Energieaufnahmevermögens nachgewiesen werden, dass Oberflächenrisse im Übergangsbereich und im Randbereich des Elektrodeneindrucks bei den hier vorliegenden Bedingungen keinen negativen Einfluss auf die dynamische Festigkeit haben. ----------------------------------------------------------------------------------------------------------------------------------------------------------------------------------
In the case of high-strength austenitic steels, welding-induced cracks may arise during resistance spot welding in the rough conditions of body shell fabrication. The influence of such cracks on the dynamic strength is not sufficiently well-known at present. Within the framework of this paper, welding cracks in various positions and with various formations were therefore investigated with regard to their effect on the strength and fracture behaviour of resistance-spot-welded joints subjected to high-speed shear tension. In this respect, the analysis of the force/displacement courses, the tensile shear strength and the energy absorption capacity served to prove that surface cracks in the interfacial region and in the peripheral region of the electrode indentation do not have any negative influence on the dynamic strength for the tested circumstances.
Genetic transformation of Knufia petricola A95 - a model organism for biofilm-material interactions

We established a protoplast-based system to transfer DNA to Knufia petricola strain A95, a melanised rock-inhabiting microcolonial fungus that is also a component of a model sub-aerial biofilm (SAB) system. To test whether the desiccation resistant, highly melanised cell walls would hinder protoplast formation, we treated a melanin-minus mutant of A95 as well as the type-strain with a variety of cell-degrading enzymes. Of the different enzymes tested, lysing enzymes from Trichoderma harzianum were most effective in producing protoplasts. This mixture was equally effective on the melanin-minus mutant and the type-strain. Protoplasts produced using lysing enzymes were mixed with polyethyleneglycol (PEG) and plasmid pCB1004 which contains the hygromycin B (HmB) phosphotransferase (hph) gene under the control of the Aspergillus nidulans trpC. Integration and expression of hph into the A95 genome conferred hygromycin resistance upon the transformants. Two weeks after plating out on selective agar containing HmB, the protoplasts developed cell-walls and formed colonies. Transformation frequencies were in the range 36 to 87 transformants per 10 µg of vector DNA and 106 protoplasts. Stability of transformation was confirmed by sub-culturing the putative transformants on selective agar containing HmB as well as by PCR-detection of the hph gene in the colonies. The hph gene was stably integrated as shown by five subsequent passages with and without selection pressure.
