Bisphenol A (BPA) is a chemical that has been used in the production of plastics for more than 40 years. BPA released from bottles made of polycarbonates has been identified as a potential endocrine disrupting substance. Comprehensive research has been undertaken to test and verify its effect on animals and human beings.
Many methods have been proposed for the determination of BPA, in particular, gas and liquid chromatography. The main disadvantages of such approaches are the high cost of the equipment, the significant duration of an analysis, and difficulties in screening a large number of samples. Lateral Flow ImmunoAssay (LFIA) attributes are high throughput, high specificity and sensitivity, as well as low cost and simplicity.
An LFIA test strip was developed. This LFIA is based on anti-BPA antibodies (from mouse) conjugated with gold nanoparticles as the marker by passive absorption and covalent coupling, deposited in an elaborate pad. To form control and test zones, anti-mouse antibody and a BVA-BSA conjugate were spotted on the nitrocellulose membrane, respectively.
Negative samples are revealed by red lines both in the test and control zones, respectively, whereas positive samples produce a single red line only in the control zone. The measurement range is 0.05 – 23 μg/L, visually detected by the naked eye within 5 minutes. In addition, the intensities at the test line can be read by dedicated lateral flow readers (opTrilyzer® and Cube by opTricon, Berlin) for a more precise and documented determination.
Virtual CT acquisition and reconstruction of complex and noisy scanning trajectories in aRTist
(2019)
In modern CT imaging, simulation has become an important tool to minimize cost- and time-intensive measurements. It is increasingly used to optimize techniques for complex applications, to support the preparation of written procedures, and for educational purposes. We extended the CT simulation software ‘aRTist’ with a module to set-up arbitrary trajectories including disturbing influences during the scan. Moreover, such geometric deviations can be compensated by the internal reconstruction tool.
Presentation of latest develeopements regarding conformity assessments of pyrotechnic articles within the forum of notified bodies according to article 37 of the EU Directive 2013/29/EU OF THE EUROPEAN PARLIAMENT AND OF THE COUNCIL of 12 June 2013 on the harmonisation of the laws of the Member States relating to the making available on the market of pyrotechnic articles (recast).
Polymeric core-shell particles were synthesized in a semi-batch emulsion polymerization process. The shell of the particles consist of PVDF with a high amount of beta-phase. Small-angle X-ray scattering (SAXS) was used to quantify the size of the cores of the particles and the thickness of the shell.
The departure from the current automation landscape to next generation automation concepts for the process industry has already begun. Smart functions of sensors simplify their use and enable plug-and-play integration, even though they may appear to be more complex at first sight. Monitoring specific information (i.e., “chemical” such as physico-chemical properties, chemical reactions, etc.) is the key to “chemical” process control.
The talk introduces a smart online NMR sensor module provided in an explosion proof housing as example. This sensor was developed for an intensified industrial process (pharmaceutical lithiation reaction step) funded by the EU’s Horizon 2020 research and innovation programme (www.consens-spire.eu). Due to NMR spectroscopy as an “absolute analytical comparison method”, independent of the matrix, it runs with extremely short set-up times in combination with “modular” spectral models. Such models can simply be built upon pure component NMR spectra within a few hours (i.e., assignment of the NMR signals to the components) instead of tedious calibrations runs.
The talk also generally covers current aspects of high-field and low-field online NMR spectroscopy for reaction monitoring and process control.
Nanomaterials are a relatively new class of materials for which a regulatory framework still has to be established and regulators require comprehensive datasets of well characterized nanomaterials for the implementation. Especially information about the surface chemistry of nanoparticles is crucial, since it largely determines their biological and environmental fate. How the surface chemistry changes under relevant ageing conditions is of particular interest, because exposure will normally occur not to the pristine material but to a nanoform that underwent some kind of transformation.
In this talk, first a short overview about the contributions of division 6.1 to multiple European-funded projects will be presented, in which surface analytical techniques are used to improve the physical-chemical characterization of nanomaterials. Secondly, a study investigating the surface-chemical transformations of a representative set of titanium dioxide nanoparticles is discussed in more detail. The ageing has partly been performed at the BAM division 7.5, and the surface chemistry was analyzed using time-of-flight secondary ion mass spectrometry (ToF-SIMS) and X-ray photoelectron spectroscopy (XPS). By analyzing the complex mass spectra with principal component analysis (PCA), it was possible to identify even subtle changes that occur upon ageing.
There is an increasing interest in bridging the gap between the photoluminescence (PL) properties of nanomaterials like semiconductor nanocrystals (QDs) commonly assessed in ensemble studies and the PL features of single QDs for life sciences applications such as bioimaging studies or use in microfluidic assays. The fluorescence quantum yield (ΦF) is a key performance parameter for all molecular and nanoscale emitters, increasingly employed in nanoscience, nanotechnology, and medical diagnostics.
ΦF determines not only the signal size together with the reporter´s molar extinction coefficient, yet it is particularly relevant for nanocrystals like QDs with coordinatively bound surface ligands and size- and surface chemistry-dependent PL characteristics.
The importance of ΦF measurements at ultralow concentration encouraged us to explore the potential of fluorescence correlation spectroscopy (FCS) for the relative determination of ΦF of ligand-stabilized CdTe nanocrystals in comparison to molecular dyes with closely matching spectral properties and known ΦF.
We describe a FCS-based method for the relative determination of ΦF of dispersed QDs at ultralow concentrations, and procedures to overcome QD-inherent challenges like complex and power-dependent blinking behavior as well as ligand- and QD-specific aggregation. We could demonstrate the potential of this approach by comparison with steady state ensemble measurements.
What exactly does a postdoc do at a research institute? What scientific questions does he or she want to address? What is their daily work routine? During our regular Lab Tours, female postdocs provide insight into their jobs. They also tell us about their career path: How did they get there? And what do they envision for their professional future?
This time, Dr. Ana Guilherme Buzanich, a scientist from the Structure Analysis division at the Bundesanstalt für Materialforschung und -prüfung (BAM), will give an overview of her Research areas. She works at the electron storage ring BESSY-II where she conducts experiments at two hard X-ray beamlines (BAMline and µSpot). The tour includes a guided visit of the research facility.
Elastomers are usually reinforced by large amount of fillers like carbon black (CB) or silica in order to improve various mechanical properties, such as Young’s modulus, hardness, tear resistance, abrasion resistance, and gas barrier properties. In recent years, such improvements were also obtained by using nanoparticles at significantly lower filler loadings. Graphene is a twodimensional (2D) sheet of a thickness in the atomic scale, composed of a honeycomb structure of sp2 carbon atoms. Besides significant mechanical reinforcement, graphene harbors the potential to be used as a multifunctional filler, as it can also increase the conductivity and weathering stability of elastomer matrices. Ultraviolet (UV) irradiation and oxidative agents can lead to the degradation of elastomers due to a multistep photooxidative process, including the formation of radicals. Carbon-based fillers have an influence on these reactions, as they can absorb UV radiation and act as radical scavengers.
This chapter summarizes the results of our larger project on multilayer graphene (MLG)/elastomer nanocomposites, previously published, which present a comprehensive case study of MLG as a multifunctional nanofiller in elastomer/graphene nanocomposites. Different elastomeric matrices are compared in order to demonstrate the outstanding impact of MLG as a general benefit. The dependency of this effect on concentration is discussed in detail. Taking into account the key role of dispersion, different mixing procedures are compared, evaluating a facile implementation of graphene nanocomposites into conventional rubber processing. Finally, the most probable commercial uses of MLG nanofillers in combination with conventional CB are studied. The nanocomposites were prepared in the kg scale in order to obtain enough specimens to investigate various properties of the uncured and vulcanized rubbers at the highest quality level, including rheology, curing, morphology, several mechanical properties, abrasion, conductivity, gas permeation, burning behavior, and weathering stability. The structure property relationships are asserted and questioned, for example, by investigating the radical scavenging ability or aspect ratio of the MLG. This chapter illustrates the state of the art of graphene/rubber nanocomposites targeted for commercial mass applications.
The quality of components made by laser beam melting (LBM) additive manufacturing is naturally influenced by the quality of the powder bed. A packing density <1 and porosity inside the powder particles lead to intrinsic voids in the powder bed. Since the packing density is determined by the particle size and shape distribution, the determination of these properties is of significant interest to assess the printing process. In this work, the size and shape distribution, the amount of the particle’s intrinsic porosity, as well as the packing density of micrometric powder used for LBM, have been investigated by means of synchrotron X-ray computed tomography (CT). Two different powder batches were investigated: Ti–6Al–4V produced by plasma atomization and stainless steel 316L produced by gas atomization. Plasma atomization particles were observed to be more spherical in terms of the mean anisotropy compared to particles produced by gas atomization. The two kinds of particles were comparable in size according to the equivalent diameter. The packing density was lower (i.e., the powder bed contained more voids in between particles) for the Ti–6Al–4V particles. The comparison of the tomographic results with laser diffraction, as another particle size measurement technique, proved to be in agreement.
Absorption edge tomography is a method which exploits the sudden change of the attenuation coefficient, when the photon energy crosses the absorption edge of an element. The beamline BAM line at BESSY II, which is operated by the Federal Institute for Materials Research and Testing, can provide a monochromatized beam in a photon energy range from 5 keV up to 80 keV with a bandwidth of 2%. Together with the microtomography setup, this enables differential tomography sensitive to any element with N >= 24 (Cr) by using an appropriate K- or L-edge in this range. Here, a polymer filament embedding metal organic framework (MOF) was prepared and used for 3D printing. Absorption edge tomography at the copper K edge was employed to perform a non-destructive 3D characterization of the microstructure of the embedded MOF. Data fusion was then used to determine the size distribution of the embedded MOF.

Prozessindustrie Gemeinsam Digital – Forschungsbedarf für die Digitalisierung der Prozessindustrie
(2019)

We have applied laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) with subcellular resolution as an elemental mass microscope to investigate the distributions of Ag nanoparticles (NP) in a 3-dimentional multicellular spheroid (MCS) model. The production of MCS has been optimized by changing the seeding cell number (500 to 40,000 cells) and the growth period (1 to 10 days). Incubations of MCS with Ag nanoparticle suspensions were performed with a concentration of 5 µg mL-1 for 24 hours. Thin-sections of the Eosin stained MCS were analysed by elemental mass microscopy using LA-ICP-MS to image distributions of 109Ag, 31P, 63Cu, 66Zn and 79Br. A calibration using NP suspensions was applied to convert the measured Ag intensity into the number of particles being present in each measurement pixel. The numbers of NP determined ranged from 30 up to 4,000 particles in an enrichment zone. The particle distribution was clearly correlated to 31P, 66Zn and 79Br and was localized in an outer rim of proliferating cells (confirmed by DAPI) with a width of about two-single cell diameters. For the highest seeding cell number NPs were only detected in this outer rim, whereas small molecules as for instance 79Br and 109Ag ions were detected in the core of the MCS as well. Aniline blue staining demonstrated that this outer rim was rich in collagen structures in which fibroblast cells were embedded and a thin-membrane was visible which separated the core from the biological active cell layer functioning as biological barriers for NP transport. In this presentation, we will show the possibility using this 3-dimensional model for toxicological and medical applications.

The industrial use of ultrashort laser pulses has made considerable progress in recent years. The reasons for this lie in the availability of high average powers at pulse repetition rates in the several 100 kHz range. The advantages of using ultrashort laser pulses in terms of processing precision can thus be fully exploited. However, high laser intensities on the workpiece can also lead to the generation of unwanted X-rays. Even if the emitted X-ray dose per pulse is low, the accumulated X-ray dose can become significant for high-repetition-rate laser systems so that X-ray exposure safety limits must be considered. The X-ray emission during ultrashort pulse laser processing was investigated for a pulse duration of 925 fs at 1030 nm wavelength and 400 kHz repetition rate. Industrially relevant materials such as steel, aluminum and glass were treated. Tungsten served as reference. X-ray spectra were recorded, and X-ray dose measurements were performed for laser treatment in air. For laser intensities > 2 × 10^13 W/cm2, X-ray doses exceeding the regulatory exposure limits for members of the public were found. Suitable X-ray protection strategies are proposed.
Correlating the photoluminescence (PL) properties of nanomaterials like semiconductor nanocrystals (QDs) and upconversion nanocrystals (UCNPs) assessed in ensemble studies and at the single particle level is increasingly relevant for applications of these nanomaterials in the life sciences like bioimaging studies or their use as reporters in microfluidic assays. Here we present a comparison of the spectroscopic properties of ensembles and single emitters for QDs like II/VI QDs and cadmium-free AIS/ZnS QDs as well as different UCNPs. The overall goal of this study was to derive particle architectures well suited for spectroscopic and microscopic applications.
The talk gives a historical introduction and introduces some of the biggest and most important research institutes in Germany dealing with tribology at Universities, scientific societies and companies. Some examples for research projects and focus areas are given. As an example for a research institute with a long history in tribological research the activities of BAM in Berlin are presented.
Fiber reinforced polymers (FRPs) are a well established material in lightweight applications, e.g. in automotive, aerospace or wind energy. The FRP components are subjected to multiaxial mechanical as well as hygrothermal loads. Common operation temperatures are in the range of 213 K and 373 K (-60 °C and 100 °C) at a relative humidity of 10% to 90%. In spacecraft applications, the environmental conditions are even more extreme. However, the correlation between multiaxial mechanical loading and harsh environment conditions have to-date not been investigated in detail. The project aims to investigate the fatigue behavior of FRPs dependent on multiaxial mechanical loading, temperature, and humidity. Extensive experimental testing is performed on flat plate and cylindrical tube specimens, accompanied by numerical and analytical calculations.
Intensified continuous processes are in focus of current research. Compared to traditional batch processes, intensified continuous production allows the synthesis of new and difficult producible compounds with better product uniformity and reduced consumption of raw materials and energy. Flexible (modular) chemical plants can produce various products using the same equipment with short down-times between campaigns, and quick introduction of new products to the market.
Full automation is a prerequisite to realize such benefits of intensified continuous plants. In continuous flow processes, continuous, automated measurements and closed-loop control of the product quality are required. Consequently, the demand for smart sensors, which can monitor key variables like component concentrations in real-time, is increasing. Low-Field NMR spectroscopy presents itself as such an upcoming smart sensor (as addressed, e.g., in the CONSENS project1,2).
Systems utilizing such an online NMR analyzer benefits through short development and set-up times when applied to modular production plants starting from a desired chemical reaction. As an example for such a modular process unit, we present the design and validation of an integrated NMR micro mixer based on computational modelling suited for a desired chemical reaction. This method includes a proper design of a continuous reactor, which is optimized through computational fluid dynamics (CFD) for the demands of the NMR sensor as well as for the given reaction conditions. The system was validated with a chemical reaction process.
Avoid living dangerously: non-destructive characterization of AM parts from powder to end-of-life
(2019)
The freeform and the revolutionary design possibilities offered by additive manufacturing have skyrocketed the amount of optimization studies in the realm of engineering, and metallic additive manufactured parts are becoming a reality in industry.
Not surprisingly, this has not been paralleled by a similar enthusiastic wave in the realm of materials science, and still very little is known about AM materials properties. This has the consequence that, typically, conventional materials properties are still used in design and even in simulations.
lt is necessary to dig a lot deeper than at present, in order to understand these new materials classes, and in particular their microstructure and their intemal stresses, largely different from their cast or wrought companions.
