Estrone (E1), a metabolite of the estrogenic hormones 17β-estradiol (β-E2) and 17α-estradiol (α-E2), is itself a potent estrogen which can have a significant impact on the hormonal balance. Due to ist high potential for adverse effects on human health and aquatic life even at pg/L to ng/L levels, its appearance in water should be monitored. E1 has also been considered a marker substance for the presence of other estrogens. This study presents a newly developed direct competitive enzymelinked immunosorbent assay (ELISA) for quantification of E1 in environmental water samples using new monoclonal antibodies.
The quantification range of the ELISA is 0.15 μg/L to 8.7 μg/L E1, and the limit of detection is around 60 ng/L for not pre-concentrated water samples. A pre-concentration step after careful selection of suitable phases for SPE was developed, too. The influence of organic solvents and natural organic matter on the ELISA was assessed. The high selectivity of the monoclonal antibody was demonstrated by determining the cross-reactivity against 20 structurally related compounds.
For the assessment of matrix effects, a concept (“LC–ELISA”) is thoroughly exploited, i.e., separating complex samples by HPLC into 0.3 min fractions and determination of the apparent E1 concentration. Furthermore, fractions with interferences for nontarget/suspected-target analysis can be assigned. A dilution approach was applied to distinguish between specific interferences (cross-reactants) and non-specific interferences (matrix effects). In the determination of 18 environmental samples, a good agreement of the E1 concentration in the respective fractions was obtained with mean recoveries of 103 % to 132 % comparing ELISA to LC–MS/MS.
Tunnel inspection is a challenging problem because of high-volume traffic and routine operations in naturally aggressive environments. The need to keep tunnels open during inspection and minimize tunnel closures and user delays must be carefully balanced with the need to conduct in-depth lining inspections to ensure the safety of drivers. This paper describes the laboratory validation and field performance of a recently developed in-depth nondestructive testing technology for the detection of impairments in tunnel lining: linear array shear wave tomography, typically referred to as ultrasonic tomography (UST). Before this equipment is used in the field, the system is first evaluated through the use of laboratory specimens with artificial defects that mock common structural problems, such as air- and water-filled voids, delaminations, and other potential abnormalities. The device is also used to determine concrete thickness and reinforcement depth and spacing. The test results are discussed to determine the device's capabilities and limitations in locating defects in concrete structures. After the system is evaluated on the basis of its ability to detect these simulated defects, the system is taken to the field to inspect a public tunnel for natural structural defects. Potential regions of interest are first identified through high-speed air-coupled ground-penetrating radar (GPR) and visual inspection and subsequently inspected with UST. This paper shows that the combination of preliminary inspection procedures (GPR surveys and visual inspection) and an in-depth technique like UST is powerful for the assessment of the condition of tunnel linings and can detect potential anomalies, such as delamination, reinforcement depth and layout, and lining thickness.
Die Validierung dient bereits in vielen Bereichen der Technik zum Nachweis, ob mit einem Verfahren Kundenanforderungen erfüllt werden können. Da gerade die im Bauwesen verwendeten zerstörungsfreien Prüfverfahren bis auf den Rückprallhammer nicht in einer deutschen Norm geregelt sind, kann die Validierung zur verbesserten Anwendungssicherheit nicht genormter Verfahren beitragen. Die Leistungsfähigkeit des Verfahrens wird für den Kunden transparent, so dass er die Güte der Messergebnisse einschätzen kann. Das ist besonders dann von Bedeutung, wenn aus dem Messergebnis eine belastbare Aussage abgeleitet werden soll und hohe Anforderungen bezüglich der Messgenauigkeit bestehen. So entsteht z. B. die Frage, welche Bauteildicke nach zerstörungsfreien Untersuchungen für eine statische Nachrechnung auf einem bestimmten Vertrauensniveau angesetzt werden kann oder mit welcher Sicherheit Pfahlgründungen, deren Länge ebenfalls zerstörungsfrei bestimmt wurde, in eine tragende Schicht einbinden. In dem Beitrag werden die Grundzüge der Durchführung einer Validierung beschrieben und am Beispiel der Dickenmessung an Fundamenten erläutert. Die beschriebene Vorgehensweise kann prinzipiell auf eine Vielzahl im Bauwesen auftretender Prüfaufgaben angewandt werden. Sind die Anforderungen an die Prüfaufgabe bezüglich der Genauigkeit nicht so hoch, können vereinfachte Abschätzungen der Messunsicherheit vorgenommen werden.
Bemessungsregeln werden im Bauwesen derzeit auf der Grundlage der so genannten Zuverlässigkeitsanalyse entwickelt. Auf ihr ist auch das Sicherheitskonzept der EUROCODES gegründet. Seit ungefähr einem Jahrzehnt werden auch Dauerhaftigkeitsanalysen nach dieser Vorgehensweise durchgeführt. In diesem Beitrag wird gezeigt, wie Methoden der Zuverlässigkeitsanalyse nun auch auf Messaufgaben angewandt werden können. Dies wird am Beispiel der zerstörungsfreien Prüfung von Tunnelinnenschalen mit dem Ultraschallecho-Verfahren erläutert, dessen Anwendung in der Richtlinie RI-ZFP-TU des Bundesministeriums für Verkehr, Bau und Stadtentwicklung geregelt ist. Es wird gezeigt, wie Messdaten weitergehend ausgewertet werden können und welche Rückschlüsse daraus gezogen werden können. Damit kann die Eignung zerstörungsfreier Prüfverfahren im Bauwesen bezogen auf die vorhandenen Randbedingungen der Prüfaufgabe auch quantitativ angegeben werden.
Validation of a European standard for the determination of hexavalent chromium in solid material

A European standard for the determination of Cr(VI) in solid material has been elaborated in the framework of an international co-operation and finally validated in the course of an interlaboratory comparison. The procedure is based on the alkaline digestion prescribed by EPA method 3060A followed by ion chromatography and determines an operationally defined content of Cr(VI), including water-soluble and insoluble chromates. A preliminary robustness study was carried out in order to compare different extraction methodologies and to study the equivalency of different analytical methods for the determination of Cr(VI) in alkaline extracts of soil and waste materials. During an interlaboratory validation trial with 19 European laboratories a set of 4 samples (2 soil and 2 waste samples) was analysed to determine performance characteristics for different combinations of digestion and detection methods. With the procedures prescribed by the new European standard (EN 15192) acceptable results were obtained for both soil samples and one of the waste samples (sludge). However, for the second waste sample (fly ash) a large deviation in analytical results was observed. This indicates that particularly for waste materials a possible occurrence of strong matrix effects has to be considered and supplementary quality control data are needed in order to assess the validity of analytical results. The accuracy of the determination of Cr(VI) in solid matrices remains a challenging field in terms of maximum extraction efficiency and minimum species interconversion.
Zerstörungsfreie Prüfverfahren im Bauwesen werden zunehmend für großflächige und detaillierte Zustandsuntersuchungen von Bauwerken angewendet. Für gezielte Untersuchungen von spanngliedführenden Bereichen mit einem Schadensverdacht in Spannbetonbauwerken eignen sich die Prüfverfahren Ultraschallecho, Impakt-Echo und Radar. Eine nennenswerte Verbesserung der Ergebnisse und deren Interpretation wurde in den letzten Jahren durch die weiterführende Datenauswertung mit Hilfe von 3D-Rekonstruktionsrechnungen und der Kombination von Meßdaten durch Datenfusion erreicht. Die Anwendung der Verfahren mit automatisierter Meßwerterfassung wird für Fahrbahnplatten und Hohlkastenstege dargestellt und diskutiert. Ergebnisse einer Meßunsicherheitsbetrachtung und deren Bedeutung werden an einer Fundamentplatte aufgezeigt.

Automated Non-Destructive Assessment of Bridges
Non-destructive test methods in civil engineering are increasingly used for large-area investigations of structures. The test methods ultrasonic-echo, impact-echo and radar are well suited for detailed investigations of pre-stressed areas at pt-concrete structures. A significant improvement of the results and their interpretations has been achieved in recent years using advanced data analysis procedures such as 3D-reconstructions and the fusion of data. The automated application of the methods has been carried out successfully at decks and webs of pre-stressed box girder bridges. The achieved results are discussed in detail. The measurement uncertainty and its importance for the results are presented considering as example a foundation slab.
A neural network-based analysis method for the identification of a viscoplasticity model from spherical indentation data, developed in the first part of this work [J. Mater. Res. 21, (2006)], was applied for different metallic materials. Besides the comparison of typical parameters like Young's modulus and yield stress with values from tensile experiments, the uncertainties in the identified material parameters representing modulus, hardening behavior, and viscosity were investigated in relation to different sources. Variations in the indentation position, tip radius, force application rate, and surface preparation were considered. The extensive experimental validation showed that the applied neural networks are very robust and show small variation coefficients, especially regarding the important parameters of Young's modulus and yield stress. On the other hand, important requirements were quantified, which included a very good spherical indenter geometry and good surface preparation to obtain reliable results.
