A vast number of emerging pollutants has been detected in the environment over the last decades. Analytical methods suitable for trace analysis are needed that are desirably also fast, inexpensive and, if possible, robust and portable. Immunoanalytical methods which are available in a broad range of formats, can be profitably used here to analyze for the distribution and the trends of concentration levels of contaminants in the environment. Some of these formats are single-analyte but high-throughput methods. In order to use them wisely, indicator substances, sometimes called anthropogenic markers, should be selected and used in screening approaches. Other methods are suitable to be performed on portable instrumentation in the field (on-site) or in facilities such as wastewater treatment plants for on-line monitoring. Furthermore, there are the socalled array technologies that allow for parallel analysis of several analytes of interest (multiplexing). The microtiter-plate based ELISA (Enzyme-linked Immunosorbent Assay) is the method of choice for the analysis of a large number of samples [1]. ELISA screening data for anthropogenic markers such as the antiepileptic carbamazepine, the analgesic diclofenac, the anti-histaminic cetirizine, the steroid hormone estrone, the antimicrobial sulfamethoxazole, the stimulants caffeine and cocaine, the priority pollutant bisphenol A, and the bile acid isolithocholic acid [2] are presented. For on-site screening and monitoring, simpler formats, like mix-and-read assays, e.g. the Fluorescence Polarization Immunoassay (FPIA) or Lateral-flow Immunoassays (LFIA) are more suitable tools. Electrochemical formats run on portable devices provide additional advantages as no light source is required. Some examples are presented and discussed. The suitability of multi-analyte formats such as immunomicroarrays depends on the choice of a signal-producing system that provides small uncertainties and good reproducibility of the measurements. Biochip (“flat”) arrays read out on slide scanners and bead-based (“suspension”) arrays read out in flow cytometers are two options and show their distinct pros and cons. Altogether these approaches show the great potential immunoanalytical methods provide for the screening for environmental contaminants in the aquatic environment.
A frequently studied environmental contaminant is the active substance diclofenac, which is removed insufficiently in sewage treatment plants. Since its inclusion in the watch list of the EU Water Framework Directive, the concentrations in surface waters will be determined throughout Europe. For this, still, more precise analytical methods are needed. As a reference, HPLC-MS is frequently employed. One of the major metabolites is 4’-hydroxydiclofenac (4’-OH-DCF). Also, diclofenac lactam is important for assessing degradation and transformation. Aceclofenac (ACF), the glycolic acid ester of diclofenac is used as a drug, too, and could potentially be cleaved to yield diclofenac again. In various sewage treatment plant influent samples, diclofenac, 4’-OH-DCF, DCF lactam and ACF could be determined with detection limits of 3 µg/L, 0.2 µg/L, 0.17 µg/L and 10 ng/L, respectively.
Diclofenac is a widely used drug against fever, inflammation, pain, and rheumatic diseases. An average of 70 % of the ingested diclofenac is excreted in the urine. Thus, 63 tons are introduced into the water cycle in Germany. Due to insufficient removal of diclofenac in wastewater treatment plants, residues of diclofenac can be found in surface water and sometimes in drinking water It exist a polyclonal against diclofenac and monoclonal antibodies against diclofenac and it`s metabolites. There all based on direct couling of diclofenac on proteins via carboxylic functions. However it was imagined advantageous to introduce a heterobifunctional spacer between the analyte and the carrier protein.
Aceclofeac is commercial available. All other spacers can only be introduced with solid phase synthesis.

Pharmacologically active compounds are often detected in wastewater and surface waters. The nonsteroidal anti-inflammatory drug diclofenac was included in the watch list of substances in the EU that requires their environmental monitoring in the member states. DCF may harmfully influence the ecosystem and the natural organization already at concentrations of ≤1 μg/l. The fast and easy quantification of DCF is becoming a subject of global importance. Fluorescence polarization immunoassay (FPIA) is a homogeneous mix-and-read method, which does not require the immobilization of reagents, they can be performed in one phase, making it possible to analyze wastewater without any complicated pre-treatment, performed within 20–30 min.
Die von der Europäischen Kommission im Jahr 2021 empfohlene, systematische Abwasserüberwachung mittels PCR-Techniken wird als ein wirksames Früh-warnsystem für künftige Pandemien durch Viren und andere Krankheitserreger betrachtet. Ein generelles Problem bei jeder quantitativen Messung von Parametern im Abwasser ist die starke Schwankung des Abwasservolumens und seiner Zusammensetzung im Tages- und Jahreszeitenverlauf. Um die Konzentrationsbestimmungen von Krankheitserregern im Abwasser in Korrelation mit menschlichen Einträgen setzen zu können, könnten sog. "anthropogene Marker" eine herausragende Rolle spielen, deren Eintrag ins Abwasser menschlichen Ursprungs sind. Im Projekt werden mittels ELISA die Konzentrationen ausgewählter anthropogener Marker in deutschlandweit gesammelten Abwasserproben erhoben und analysiert.
Increasing contamination of environmental waters with pharmaceuticals represents an emerging threat for the drinking water quality and safety. In this regard, fast and reliable analytical methods are required to allow quick countermeasures in case of contamination. Here, we report the development of a magnetic bead-based immunoassay (MBBA) for the fast and cost-effective determination of the analgesic diclofenac (DCF) in water samples, based on diclofenac-coupled magnetic beads and a robust monoclonal anti-DCF antibody. A novel synthetic strategy for preparation of the beads resulted in an assay that enabled for the determination of diclofenac with a significantly lower limit of detection (400 ng/L) than the respective enzyme-linked immunosorbent assay (ELISA). With shorter incubation times and only one manual washing step required, the assay demands for remarkably shorter time to result (< 45 min) and less equipment than ELISA. Evaluation of assay precision and accuracy with a series of spiked water samples yielded results with low to moderate intra- and inter-assay variations and in good agreement with LC–MS/MS reference analysis. The assay principle can be transferred to other, e.g., microfluidic, formats, as well as applied to other analytes and may replace ELISA as the standard immunochemical method.

Magnetic bead-based immunoassays (MBBAs) allow for implementation into an immunosensor for online testing. Particles are prepared by either coupling the analyte molecule (diclofenac) or the respective antibody (amoxicillin) to the surface. For miniaturization, detection is performed electrochemically (chronoamperometry) on a microfluidic chip. The developed immunosensor will enable detection of pharmaceuticals directly in water supply pipes.

Die zunehmende Kontamination von (Trink)-Wasser mit pharmazeutischen Rückständen stellt ein potenzielles Gesundheitsrisiko dar und verlangt nach schnellen und zuverlässigen Analysemethoden. Hierbei können antikörper-basierte Methoden helfen, da sie schnelle und kostengünstige Vor-Ort-Messungen sowie Onlineanalysen ermöglichen.
Der Übergang von konventionellen plattenbasierten Formaten, wie ELISA (Enzyme-linked Immunosorbent Assay), zu einem Immunosensor für Online-Messungen wird durch den Einsatz magnetischer Mikropartikel als mobile Messplattform ermöglicht.
Die Partikel werden dazu entweder mit dem Antikörper oder dem Analyten gekoppelt. Diese beiden Strategien werden am Beispiel zweier relevanter Kontaminanten des Wassers gezeigt: Diclofenac (DCF) & Amoxicillin (AMX), und sind leicht auf andere Analyten übertragbar. Die magnetpartikelbasierten Assays (MBBAs) weisen niedrigere Nachweisgrenzen, breitere Messbereiche sowie kürzere Analysezeiten als die entsprechenden ELISAs auf und können in einen Immunosensor mit elektrochemischer Detektion auf einem mikrofluidischen Chip integriert werden.
