Line planning in public transport involves determining vehicle routes and assigning frequencies of service such that travel demands are satisfied. We evaluate how line plans, which are optimal with respect to in-motion costs (IMC), the objective function depending purely on arc-lengths for both user and operator costs, performs with respect to the value of resources consumed (VRC). The latter is an elaborate, socio-economic cost function which includes discomfort caused by delay, boarding and alighting times, and transfers. Even though discomfort is a large contributing factor to VRC and is entirely disregarded in IMC, we observe that the two cost functions are qualitatively comparable.

Background
We assessed the novel MACC1 gene to further stratify stage II colon cancer
patients with proficient mismatch repair (pMMR).
Patients and methods
Four cohorts with 596 patients were analyzed: Charité 1 discovery cohort
was assayed for MACC1 mRNA expression and MMR in cryo-preserved
tumors. Charité 2 comparison cohort was used to translate MACC1 qRT-
PCR analyses to FFPE samples. In the BIOGRID 1 training cohort MACC1
mRNA levels were related to MACC1 protein levels from
immunohistochemistry in FFPE sections; also analyzed for MMR.
Chemotherapy-naïve pMMR patients were stratified by MACC1 mRNA and
protein expression to establish risk groups based on recurrence-free
survival (RFS). Risk stratification from BIOGRID 1 was confirmed in the
BIOGRID 2 validation cohort. Pooled BIOGRID datasets produced a best
effect-size estimate.
Results
In BIOGRID 1, using qRT-PCR and immunohistochemistry for MACC1
detection, pMMR/MACC1-low patients had a lower recurrence probability
versus pMMR/MACC1-high patients (5-year RFS of 92% and 67% versus
100% and 68%, respectively). In BIOGRID 2, longer RFS was confirmed
for pMMR/MACC1-low versus pMMR/MACC1-high patients (5-year RFS of
100% versus 90%, respectively). In the pooled dataset, 6.5% of patients
were pMMR/MACC1-low with no disease recurrence, resulting in a 17%
higher 5-year RFS (95% CI (12.6-21.3%)) versus pMMR/MACC1-high
patients (P=0.037). Outcomes were similar for pMMR/MACC1-low and
deficient MMR (dMMR) patients (5-year RFS of 100% and 96%,
respectively).
Conclusions
MACC1 expression stratifies colon cancer patients with unfavorable pMMR
status. Stage II colon cancer patients with pMMR/MACC1-low tumors have
a similar favorable prognosis to those with dMMR with potential
implications for the role of adjuvant therapy.

A cubic regularization algorithm for nonconvex optimization in function space (in preparation)

We propose a cubic regularization algorithm that is constructed to deal with nonconvex minimization
problems in function space. It allows for a flexible choice of the regularization term and thus accounts for the fact
that in such problems one often has to deal with more than one norm. Global and local convergence results are established
in a general framework. Moreover, several variants of step computations are compared. In the context of nonlinear
elasticity it turns out the a cg method applied to an augmented Hessian is more robust than truncated cg.

We present a new algorithm for fairing of space curves with respect spatial constraints based on a vector valued curvature function. Smoothing with the vector valued curvature function is superior to standard Frenet techniques since the individual scalar components can be modeled similar to curvature-based curve smoothing techniques in 2d. This paper describes a curve smoothing flow that satisfies strict spatial constraints and allows simultaneous control of both curvature functions.

Subspace decompositions of finite element spaces based on $L2$-like orthogonal projections play an important role for the construction and analysis of multigrid like iterative methods. Recently several authors proved the equivalence of the associated discrete norms with the $H^1$-norm. Parameter-dependent systems of nonlinear equations with symmetry are treated by a combination of symbolic and numerical computations. In the symbolic part of the algorithm the complete analysis of the symmetry occurs, and it is here where symmetrical normal forms, symmetry reduced systems, and block diagonal Jacobians are computed. Given a particular problem, the symbolic algorithm can create and compute through the list of possible bifurcations thereby forming a so-called tree of decisions correlated to the different types of symmetry breaking bifurcation points. The remaining part of the algorithm deals with the numerical pathfollowing based on the implicit reparametrisation as suggested and worked out by Deuflhard/Fiedler/Kunkel. The symmetry preserving bifurcation points are computed using recently developed augmented systems incorporating the use of symmetry. {\bf Keywords:} pathfollowing, mixed symbolic-numeric algorithm, parameter-dependent, nonlinear systems, linear representations.

Lectures on Polytopes.

These lecture notes have several aims: \begin{itemize} \item to give an introduction to some basic facts about convex polytopes, with an emphasis on the basic methods that yield them (Fourier-Motzkin elimination, Schlegel diagrams, shellability, Gale transforms and oriented matroids), \item to discuss some important examples and elegant constructions (cyclic and neighborly polytopes, zonotopes, Minkowski sums, permutahedra and associahedra, fiber polytopes, the Lawrence construction) \item and to illustrate why polytope theory is exciting, with highlights like Kalai's new diameter bounds, the construction of non-rational polytopes, the Bohne-Dress tiling theorem, shellability and the upper bound theorem, .... \end{itemize} For several of these topics the decisive break-through is very recent, which suggests that there is much more discovered.
