Determination of Thermal DNA-Stability With Respect to PCR or How to Debunk a Pseudoscientific Claim

Debunking pseudoscience is difficult, especially for early-career students and the public. Recently, in particular the Corona pandemic has spawned a whole range of pseudoscientific claims and conspiracy theories, many of which are publicly available in the style of scientific articles on preprint servers, in predatory journals, or in some cases even as regular scientific papers making it difficult to distinguish scientific papers from pseudoscience. One example is a recently published paper claiming that PCR is unreliable because DNA is thermolabile. While experts have the skills to recognize such pseudoscience, inexperienced early-career students usually have few chances to critically evaluate such claims. Here, we present some simple experiments that combine several aspects: (I) the analysis of a seemingly reputable publication, (II) planning experiments on the physicochemistry of DNA, (III) conducting and analyzing these experiments, and finally (IV) the refutation of the claim that PCR is unreliable. These experiments can be carried out in any standard laboratory with basic molecular biology equipment. They are therefore suitable for undergraduate programs and for high school courses. The model case described here enables students to critically evaluate these claims through practical investigations and to form their own informed opinion. The participants gained new insights into the planning of experiments and a completely new perspective on the subject of science and pseudoscience.
The question of whether PCR is reliable sounds strange at first. However, looking at the scientific literature from the 1950s and 60s, one will find many publications on the physicochemistry of DNA that have been forgotten meanwhile. Quite a few of these studies have shown that DNA is thermolabile, which consequently raises the question of whether this thermolability is relevant in the context of PCR, namely in the denaturation phase. However, it can be shown that this is not the case: losses due to thermal hydrolysis are irrelevant for the performance of contemporary PCR protocols and their specificity as well as for the significance of their results. There is now a huge amount of scientifically verified and published data on technical and molecular aspects of PCR, a small selection of which we quote here. In addition, we present some primary data that also clearly demonstrate the reliability of PCR.
Diese Arbeit beschreibt zum einen drei Instrumentierungs-Methoden für die Ablaufverfolgungsinfrastruktur Event Tracing for Windows (ETW). Diese dienen dem Abfangen der Ereignissignalisierung und Extrahieren der Event-Daten aus einer überwachten Windows-VM von einer außerhalb arbeitender Software aus. Zum anderen wird in dieser Arbeit ein Verfahren zur Interpretation und Aufbereitung der ausgeleiteten binären Event-Daten außerhalb des überwachten Systems beschrieben, das keine Windows-Abhängigkeiten besitzt.
Von den drei Extraktionsmethoden wird eine praktisch implementiert. Technisch ist die Instrumentierung mit der, dem Virtual Machine Introspection (VMI) ähnlichen, Windows-Kernel-Debugging-Schnittstelle (KD) realisiert. Für die Interpretation und Aufbereitung der ETW-Daten wurden zwei Verfahren entworfen und umgesetzt, mit denen ETL-Dateien (DFEP) und extrahierte Event-Daten (DFXP) extern verarbeitet werden können. Für Evaluationszwecke wurde eine weitere Komponente implementiert, welche die Windows-TDH-API benutzt (WBEC). Die Resultate aller drei Verfahren können automatisiert miteinander verglichen werden. Die Dekodierung der binären Event-Daten unterstützt dabei sowohl MOF-basiertes als auch Manifest basiertes ETW. Um die erwähnte Interpretation der Binärdaten außerhalb und unabhängig von Windows zu realisieren, dokumentiert diese Arbeit zahlreiche benötigte, interne Datenstrukturen des ETW-Systems. Darüber hinaus sind Teile des ETW-Ereignissignalisierungspfads und die darin befindlichen Schnittstellen wie die ETW-Provider-APIs und die Systemcall-Schnittstelle technisch beschrieben. In der Evaluation wurden die implementierten Verfahren in 20 verschiedenen Test-Szenarien geprüft. Dabei wurden in einer virtuelle Maschine mit Windows 10 x64 21H1 die ETW-Ereignissignalisierungen von bis zu 12’921 Ereignissen in 5 Minuten abgefangen, die Daten ausgeleitet und außerhalb im Host-System dekodiert und aufbereitet. Hierbei konnte eine Abweichung zwischen den extrahierten und den regulär durch das ETW-System protokollierten (ETL) Event-Daten festgestellt werden. Im schlechtesten Fall wurden dabei bis zu ca. 12 % der Ereignisse von der Extraktionsmethode nicht erfasst.
Obwohl die Verfahren Schwächen aufweisen und sporadisch Event-Objekte nicht erfasst werden, konnte dennoch generell gezeigt werden, dass die Gewinnung und Verarbeitung von ETW-Ereignis-Daten außerhalb des zu überwachenden Systems möglich ist. Die in dieser Arbeit gezeigten Ansätze erscheinen daher vielversprechend hinsichtlich zukünftiger Forschungsaktivitäten. Insgesamt, liefert die Arbeit damit wichtige Erkenntnisse für den Einsatz von Event Tracing for Windows (ETW) in Hypervisor-basierten IT-Sicherheitslösungen wie VMI-basierter Sensorik in Endpoint-Detection-and-Response-Lösungen (EDR) oder Malware-Sandboxen.
There are around 2,500 car wash lines in Germany, consuming around 30 million m3 of drinking water per year. In view of the increasing scarcity of water, this level of consumption could soon be restricted by law, as it is in some countries. There are only a few systematic studies on process water systems in car wash lines in Middle Europe; we therefore carried out a survey to determine process water quality parameters (COD, turbidity, water hardness, pH, conductivity, dissolved O2, and temperature). Data were collected and analysed from 63 different locations in Germany and the Netherlands. In addition, longitudinal parameters were measured for 1 year at a single state-of-the-art site in Germany. The scatter of the observed values was quite considerable. The average COD in water of car wash sites with biological treatment was found to be significantly lower than in car wash sites working with filter treatment. Overall, the deviation of the pollution parameters was higher than expected. Surprisingly, there is hardly any reliable data on freshwater consumption in the car wash facilities. As most of the long-established facilities do not have a modern water treatment system, freshwater consumption is up to 300 L per car washed.
This systematic literature review investigates the use of physics-informed neural networks (PINNs) in electromagnetics by examining peer-reviewed articles and conference papers. By integrating governing physical laws into the loss function of a neural network, PINNs offer a mesh-free method in scientific computing. Records published between 2020 and 2025 were retrieved from the databases Scopus, Web of Science, and IEEE Xplore. The initial dataset comprised 500 records, from which 292 unique publications were identified. These were screened, yielding a final set of 139 publications that met predefined eligibility criteria. The analysis reveals growth in research activity, with a pronounced increase from 2022 onward. The literature predominantly addresses electrodynamic problems, employs feedforward neural network architectures, and adopts physics-only training. Two-dimensional problem formulations dominate, with three-dimensional formulations concentrated almost exclusively in electrodynamics, and no publications addressing electroquasistatics were identified. Contingency tables show that methodological choices are not independent of problem characteristics: medium selection correlates with physics regime, and architectural diversity increases with spatial dimensionality. Based on these findings, priorities for future work include: addressing the gap in electroquasistatics, extending three-dimensional formulations to static and quasistatic regimes, broader architectural experimentation in lower-dimensional settings, and increased integration of labeled data in static electromagnetics. To support methodological consistency and reproducibility, a reporting checklist for future PINN-based electromagnetics publications is proposed.
This study examines the impact of cold acetone vapor treatment on the roughness of the surface, dimensional deviations, and tensile strength of ABS specimens produced using FDM technology. Although FDM is an accessible and widely used method, it presents limitations such as high surface roughness and weak interlayer cohesion. In this study, 35 specimens were printed and then treated with cold acetone vapors for periods ranging from 10 to 60 min. A maximum improvement in surface roughness of 77.63% was observed after 30 min of treatment, with the Ra roughness value decreasing from 4.9 μm to 1.02 μm. Regarding dimensional deviations, for the b dimension (width), the deviations increased from 0.006% (10 min) to a maximum of 0.75% (60 min), and for the h dimension (height), from 0.52% to 5.72%. Tensile strength gradually decreased with treatment duration, from 28.02 MPa (untreated) to 14.70 MPa (60 min), while elongation at break increased from 2.61% to 4.33%. Cold acetone vapor treatment induces a transition in the material’s behavior from rigidity to ductility, significantly affecting surface finish, dimensional stability, and the mechanical properties of FDM parts.
The sealing system of high-performance spherical valves plays a critical role in ensuring operational safety and efficiency. This paper presents a static finite element analysis (FEA) to evaluate the stress distribution, contact behavior,
and deformation of sealing system components under internal pressure. Emphasis is placed on identifying potential failure zones and understanding how material properties and design parameters influence sealing performance. The sealing system consists of a movable ring and a seat made of stainless steel. The results offer valuable insights for improving the mechanical reliability of spherical valves and contribute to the development of optimized designs suited for demanding industrial applications.
Der Beitrag befasst sich mit der kontroversen Entscheidung für oder gegen einen Stadionneubau in Oldenburg, der die Anforderungen für den kommerziellen Profisport erfüllen soll. Die Konflikte resultieren aus divergierenden Ansprüchen und zeigen die Verwobenheit politischer mit kommerziellen Interessen im Fußball. Diskutiert wird die Notwendigkeit und Legitimität eines ausschließlich durch Steuergelder finanzierten Stadionneubaus. Insbesondere die erheblichen Auswirkungen auf den kommunalen Sporthaushalt, drohende Verdrängungseffekte anderer Sportarten und die damit verbundene Beschädigung sportkultureller Vielfalt werden hinterfragt. Angesichts eines bestehenden Stadions, das Regionalligaansprüchen gerecht wird, äußert sich der Beitrag kritisch, inwieweit ein derartiges Vorhaben für einen an der Finanzierung nicht beteiligten Fußballverein in Übereinstimmung mit den Aufgaben der freiwilligen kommunalen Daseinsvorsorge und Gemeinwohlorientierung zu bringen ist.
