Glycolysis is one of the primordial pathways of metabolism, playing a pivotal role in energy metabolism and biosynthesis. Glycolytic enzymes are known to form transient multi-enzyme assemblies. Here we examine the wider protein-protein interactions of plant glycolytic enzymes and reveal a moonlighting role for specific glycolytic enzymes in mediating the co-localization of mitochondria and chloroplasts. Knockout mutation of phosphoglycerate mutase or enolase resulted in a significantly reduced association of the two organelles. We provide evidence that phosphoglycerate mutase and enolase form a substrate-channelling metabolon which is part of a larger complex of proteins including pyruvate kinase. These results alongside a range of genetic complementation experiments are discussed in the context of our current understanding of chloroplast-mitochondrial interactions within photosynthetic eukaryotes.
Zum verbesserten Management der umfangreichen Prozessdokumentation wurde an der Technischen Hochschule Wildau [FH] im Rahmen des fortlaufenden TQM-Prozesses (TQM – Total Quality Management) ein Dokumentationssystem für das Qualitätsmanagement auf Wiki-Basis aufgebaut. Die Organisation der Prozessdokumentation wurde im Zuge dessen von einem Dateibasierten auf einen Wiki-basierten Ansatz umgestellt. Sämtliche Inhalte wurden in das neu geschaffene TQM-Wiki migriert. Der Aufbau des TQM-Wiki erstreckte sich über ca. sechs Monate und umfasste die Phasen Vorlauf, Aufbau und Abschluss, die Gesamtarbeitszeit belief sich auf ca. 250 – 300 Stunden. Zur technischen Realisierung des Prozessdokumentationssystems wurde die freie Software DokuWiki verwendet. Das TQM-Wiki ermöglicht die kollaborative Verwaltung und effiziente Nutzung der Qualitätsmanagementdokumentation durch alle Beteiligten. Die Kommunikation über Hochschulprozesse wird transparent und bezieht die Hochschulangehörigen ein. Die Dokumente liegen im HTML-Format vor und können in weitere Standardformate exportiert werden. Der Aufwand für eine Vielzahl häufig durchzuführender Veränderungsprozesse wurde durch den Wiki-basierten Ansatz deutlich reduziert.
Femtosecond-Pulsed Laser Written and Etched Fiber Bragg Gratings for Fiber-Optical Biosensing

We present the development of a label-free, highly sensitive fiber-optical biosensor for online detection and quantification of biomolecules. Here, the advantages of etched fiber Bragg gratings (eFBG) were used, since they induce a narrowband Bragg wavelength peak in the reflection operation mode. The gratings were fabricated point-by-point via a nonlinear absorption process of a highly focused femtosecond-pulsed laser, without the need of prior coating removal or specific fiber doping. The sensitivity of the Bragg wavelength peak to the surrounding refractive index (SRI), as needed for biochemical sensing, was realized by fiber cladding removal using hydrofluoric acid etching. For evaluation of biosensing capabilities, eFBG fibers were biofunctionalized with a single-stranded DNA aptamer specific for binding the C-reactive protein (CRP). Thus, the CRP-sensitive eFBG fiber-optical biosensor showed a very low limit of detection of 0.82 pg/L, with a dynamic range of CRP detection from approximately 0.8 pg/L to 1.2 µg/L. The biosensor showed a high specificity to CRP even in the presence of interfering substances. These results suggest that the proposed biosensor is capable for quantification of CRP from trace amounts of clinical samples. In addition, the adaption of this eFBG fiber-optical biosensor for detection of other relevant analytes can be easily realized.

Begriffe wie Internet, Intranet, Multimedia, Online-Banking, World Wide Web (WWW), Hyperlinks, Surfen im Internet, E-Mail usw. findet man sowohl in einschlägiger Fachliteratur als auch in Tageszeitungen, in Zeitschriften, im Fernsehen usw. Wir befinden uns auf dem Weg von der Industrie- zur Informationsgesellschaft. Alle Bereiche, vor allem Wissenschaft, Wirtschaft, Politik, aber auch soziale und kommunikative Bereiche der Menschen werden von den neuen Informations- und Kommunikationstechnologien beeinflußt. Gewinnung, Speicherung, Verarbeitung, Vermittlung, Verbreitung und Nutzung von Informationen und Wissen sowie die interaktive Kommunikation sind in der Informationsgesellschaft entscheidende Faktoren. Der zeitliche Verlauf dieser Entwicklung wird u.a, maßgeblich vom Aufbau leistungsstarker „Datenautobahnen / Informations-Highways" bestimmt.
Die Informationsgesellschaft wird unsere bisherigen Arbeits- und Lebensgewohnheiten in einer Weise beeinflussen, die viele vielleicht für Utopie halten. Die fortschreitende Vernetzung läßt räumliche und zeitliche Grenzen unserer Welt vergessen. Wer konnte sich schon vor 5 Jahren vorstellen, daß ein Team über Kontinente verteilt, zeitgleich bei einem gemeinsamen Projekt auf die gleichen aktuellen Daten zugreifen kann?
Der Computer und die weltweite Vernetzung durch das Internet sind aus unserem täglichen Leben nicht mehr wegzudenken. Auch in vielen Privathaushalten hat der PC Einzug gehalten. Zahlreiche Provider bieten auch für Privatkunden den Internetzugang an. Dabei nimmt Deutschland durchaus keine Spitzenposition ein. In Finnland haben bereits rund 60 Prozent der Bevölkerung einen Internet-Zugang. Das ist Weltspitze. In den USA sind es 16 Prozent, in Japan 18 Prozent der Bevölkerung. Über die weltweite Anzahl der Nutzer im Internet gibt es keine verläßlichen Zahlen. Geschätzt werden zwischen 23,5 und 35 Millionen Internetzugänge. Prognostiziert werden für das Jahr 2000 über 250 Millionen Internet-Nutzer. Studien haben ergeben, der Anteil der Studenten an der Anzahl der Nutzer liegt bei rund 30 Prozent! Daß das Interesse der Studierenden am Internet-Zugang sehr groß ist, kann aus der Sicht des Hochschulrechenzentrums nur bestätigt werden.
Anthracyclines like daunorubicin (DRN) and doxorubicin (DOX) play an undisputed key role in cancer treatment, but their chronic administration can cause severe side effects. For precise anthracycline analytical systems, aptamers are preferable recognition elements. Here, we describe the detailed characterisation of a single-stranded DNA aptamer DRN-10 and its truncated versions for DOX and DRN detection. Binding affinities were determined from surface plasmon resonance (SPR) and microscale thermophoresis (MST) and combined with conformational data from circular dichroism (CD). Both aptamers displayed similar nanomolar binding affinities to DRN and DOX, even though their rate constants differed as shown by SPR recordings. SPR kinetic data unravelled a two-state reaction model including a 1 : 1 binding and a subsequent conformational change of the binding complex. This model was supported by CD spectra. In addition, the dissociation constants determined with MST were always lower than that from SPR, and especially for the truncated aptamer they differed by two orders of magnitude. This most probably reflects the methodological difference, namely labelling for MST vs. immobilisation for SPR. From CD recordings, we suggested a specific G-quadruplex as structural basis for anthracycline binding. We concluded that the aptamer DRN-10 is a promising recognition element for anthracycline detection systems and further selected aptamers can be also characterised with the combined methodological approach presented here.
Für beruflich Qualifizierte ist der Weg in die Hochschulen und Universitäten in den letzten Jahren einfacher geworden. Die heterogenen Wege der beruflich Qualifizierten ins Studium werden wir in diesem Artikel beleuchten und aufzeigen, wie wir in unseren Mathematikvorkursen an die Vorkenntnisse unserer Studieninteressierten anknüpfen. Eine unter beruflich qualifizierten Studieninteressierten durchgeführte Befragung soll mehr Aufschluss darüber geben, über welche unterschiedlichen Wege und aus welchen persönlichen Gründen beruflich Qualifizierte ein Studium aufnehmen. Da Mathematik im Studium gerade für beruflich Qualifizierte meist eine große Hürde darstellt, hat die Technische Hochschule Wildau ihr Vorkursangebot auf die heterogener werdende Studierendenschaft angepasst.
The article investigates the correlation between the factors of enhancing traffic safety on highways and parameters of the economic growth in Russia and in countries with transition economy; such correlation does not always lead to traffic safety enhancement. As a rule, the population motorization (car density) level growth in such countries is not accompanied by efficient actions on accident rate decrease. Among the factors enhancing traffic safety on highways are the efficiency of traffic infrastructure planning (changing and removing traffic flows from cities’ centers, creation of a large amount of pedestrian and bicycle lanes separated from traffic flows) as well as harmonization of physical infrastructure and safety goals, ensuring conjugation of the developed transport infrastructure with the decrease of the share of private vehicle fleet and increase of the share of public transport.
The coffee sector’s active engagement with sustainability issues appears to be a relatively new phenomenon. Even newer is the necessity to deal with recycling and waste. Next to the waste produced by the “coffee to go” mobile drinking culture and coffee bars, the popularity of coffee capsules – i.e. single-use containers made of metal or plastics – is creating mountains of waste unknown to the traditional method of brewing coffee. The pioneer in this premium coffee sector has been Nespresso, a subsidiary and brand of the Swiss company Nestlé. Many other companies – from discount retailers to big-brand coffee shop chains like Starbucks – have copied Nespresso’s machine-and-capsule concept, adding to both the popularity and the waste problem. Next to the problem of waste production, coffee companies are addressing the problem of ethical sourcing. A key but by no means the only element is sourcing via “fair trade” schemes. Assuming that finding answers to these sustainability challenges can have a crucial impact on future sales, this article sets out to explore coffee companies’ sustainability programmes, using the examples of Nespresso and Starbucks. It finds that both firms engage in rather similar activities, but some critical differences can be identified. Both firms have a successful record in ethical sourcing. But neither has yet developed convincing waste solutions. Nespresso’s insistence on using aluminium for its capsules remains a critical issue.
Protein interaction and protein imaging strongly benefit from the advancements in time-resolved and superresolution fluorescence microscopic techniques. However, the techniques were typically applied separately and ex vivo because of technical challenges and the absence of suitable fluorescent protein pairs. Here, we show correlative in vivo fluorescence lifetime imaging microscopy Förster resonance energy transfer (FLIM-FRET) and stimulated emission depletion (STED) microscopy to unravel protein mechanics and structure in living cells. We use magnetotactic bacteria as a model system where two proteins, MamJ and MamK, are used to assemble magnetic particles called magnetosomes. The filament polymerizes out of MamK and the magnetosomes are connected via the linker MamJ. Our system reveals that bacterial filamentous structures are more fragile than the connection of biomineralized particles to this filament. More importantly, we anticipate the technique to find wide applicability for the study and quantification of biological processes in living cells and at high resolution.
