Here, we present a new sampling method and a simplified dipstick LFA format for the detection and monitoring of cowpea chlorotic mottle virus (CCMV) as a model plant pathogen. The assay employs a monoclonal mouse antibody for capture and a poly-clonal rabbit antibody conjugated to 80 nm gold nanoparticles for detection. Conventional sample and conjugate pads are omitted, allowing the test strips to be dipped directly into wells containing plant extract and antibody-gold conjugate. No plastic casing was required, which could lead to a reduction in waste. It was shown that CCMV concentrations as low as 3.5 μg/L or 350 pg per sample could be reliably detected in 15 min. Specificity tests confirmed that other plant viruses, cowpea mosaic virus (CPMV) and tobacco mosaic virus (TMV), did not produce false-positive results. In addition, we describe a new method for on-site sampling using a manual punch and a syringe equipped with a frit. This step combines grinding the sample, extraction, filtration, and reconstitution and mixing of the antibody-gold conjugate, enabling the analysis of punched leaf disks without laboratory equipment. The specific growth rate is classified by the FDA as a critical process parameter that affects product quality and quantity, but is difficult for laboratory personnel to determine. Therefore, a simple and robust method for real-time monitoring and control is crucial. According to the current state of the art, the established Luedeking-Piret model for determining the specific growth rate requires the determination of the biomass as an absolute value to initialize the model and to determine two further model parameters. However, determining the biomass is time-consuming and error-prone. The new relative model replaces this value with the relative change in biomass, which can be easily recorded using standard laboratory methods such as optical density measurement. This eliminates the need for time-consuming and resource-intensive preliminary work. Despite this simplification, simulation tests have shown that the new model delivers identical results to the established model. It represents an independent, precise alternative and offers advantages in terms of handling. The results underline the model's potential to make bioprocesses more sustainable and efficient. The choice of process parameters is crucial for achieving desired material properties, as it directly affects the microstructure and mechanical behavior, which is important when optimizing for potential applications in several fields, such as aerospace and automotive. In this study, different scanning speeds were tested to identify optimal settings, followed by the evaluation of the effects of orientation relative to the build plate and hatching strategies to enhance performance. To assess the impact of these factors, tensile tests, microhardness measurements, and X-ray diffraction (XRD) analyses were conducted. Tensile tests revealed that higher laser scanning speed generally reduces ultimate tensile strength and elongation, likely due to an increase in porosity and a less homogeneous fusion of layers. The analysis of samples printed with different orientation relative to the build plate highlighted a strong mechanical anisotropy, with the samples printed vertically exhibiting lower tensile strength and ductility compared to horizontally printed samples. Microhardness testing further confirmed an anisotropy in material properties. XRD analysis reveals a preferential orientation of austenitic grains depending on building direction. High‑resolution CT data were converted to CAD models and additively manufactured as moulds; a gelatin‑based soft‑tissue matrix with glycerin and potassium sorbate provides elasticity and durability. Functional features include fluid‑fillable ventricular structures enabling drainage and puncture simulations, and an integrated high‑density tumour insert to model advanced scenarios. Ultrasound evaluation demonstrated clear echogenic differentiation between tumour, ventricles, and surrounding tissue, confirming the model's imaging fidelity and suitability for hands‑on training. The phantom proved cost‑efficient and reusable, facilitating standardised teaching of complex procedures without ethical approval barriers. A limitation is the material's gradual mechanical instability under repeated probe contact, motivating investigations into synthetic or hybrid matrices to enhance longevity and reuse. Electron beam focusing produces dose peaks with a tunable maximal dose depth which is interesting for healthy tissue sparing at the surface and enhancing treatment quality. Monte Carlo simulations based on the FLUKA code were performed to evaluate the depth dose curves distributions in a water phantom. Our method relies on detecting XUV fluorescence emitted from the nanodroplets as they are driven into a nanoplasma by subsequent intense infrared pulses. Nanoplasma ignition is facilitated by tunnel ionization of XUV-excited He∗ atoms attached to the droplets; it thus serves as a sensitive probe of their relaxation dynamics. Aims(s) - The primary aim is to identify the antecedents that show significant indirect generation-specific effects on turnover intention via quiet quitting. Design/methodology/approach - Utilizing a sample of 2,193 urban and suburban employees from Berlin, Germany, this study tested a mediation model featuring five independent variables linked to quiet quitting, with turnover intention as the outcome variable. The analysis employed Cronbach's alpha, confirmatory factor analysis, multicollinearity checks, and multiple-mediation regression techniques. Findings - The results reveal that quiet quitting partially mediated the relationship between dissatisfaction and turnover intention for GenY, as well as the relationship between negative extra-role behavior and cynicism/depersonalization for GenZ. Additionally, negative work-life balance showed partial mediation across all three generations—GenX, GenY, and GenZ. Full mediation effects were observed specifically in GenZ for both negative extra-role behavior and cynicism/depersonalization. No significant mediating effect of quiet quitting was found for disengagement. Limitations of the study - The sample is limited to Berlin and its suburbs, with no comparative data from other regions. The generational groups are represented by moderately sized subsamples. Additionally, after conducting reliability analysis and confirmatory factor analysis (CFA), most scales were reduced to just two or three items. Headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry (HS-SPME/GC-MS) enabled the identification of 312 volatile compounds, including monoterpenes (e.g., myrcene, linalool, geraniol), sesquiterpenes (e.g., humulene, caryophyllene), esters, alcohols, aldehydes, and ketones. Amarillo showed the highest myrcene content (22.61\% of the total volatile area), while Centennial was distinguished by elevated γ-muurolene (20.59\%), and El Dorado by the highest level of undecan-2-one (10.47\%), highlighting marked varietal differences in key aroma-active constituents. Multivariate, including principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA), clearly discriminated the five varieties: PC1 (41.04\% of the variance) separated samples enriched in fruity/floral monoterpenes and esters from those dominated by woody/resinous sesquiterpenes, whereas PC2 (25.93\% of the variance) reflected variation in medium-chain esters, ketones, and waxy compounds. However, a deeper look into the metrics employed to assess system performance exposes notable discrepancies in their utilization. Notably, certain indicators, such as the pooling rate, demonstrate a susceptibility to manipulation based on input parameters, potentially skewing results to give a more favorable impression of service performance. In this paper, we show that, under such circumstances, achieving a fair comparison between study outcomes and with traditional (public) transport modes becomes challenging. In light of these challenges, this study introduces a novel operational performance indicator: Operational System Efficiency (OSE). OSE is tailored to evaluate the operational efficiency of ridepooling systems in a holistic way. In particular, it combines important operational indicators such as detour factor and empty kilometers share, which are considered in numerous analyses. Hereby, OSE is fostering a more equitable assessment of service performance. We applied this methodology and calculated the indicators provided in other studies to two real-world trip datasets from ridepooling services in Berlin and M{\"u}nster, Germany, and compared the results. Distinct disparities emerge in comparison with conventional efficiency indicators. Its primary objective is to analyze how academic networking contributes to maintaining educational continuity, fostering innovation, and strengthening institutional adaptability under crisis conditions. The study focuses on two representative initiatives, the Digital Teaching Network (DTN) and the Wildau-Kharkiv IT Bridge, which exemplify the transition from traditional digital platforms to interconnected network ecosystems. Methodologically, the research combines quantitative and qualitative approaches, including an online survey, focus group discussions, and content analysis of responses from 185 participants representing 21 universities in both countries. Rib, single-bridge, and double-bridge designs with 50 μm cores and up to 150 μm propagation lengths have been evaluated. Cryogenic-etched rib waveguides achieve the highest median transmission (66\%, -1.80 dB), compared to Bosch-etched ribs (62\%, -2.08 dB). The camera, integrated into an ion imaging system based on a micro-channel plate (MCP) and a fast P47 scintillator, enables individual time-stamping of incoming ions with nanosecond timing precision and high spatial resolution. The detector successfully captured laser-induced ion events originating from free nanoparticles disintegrated by intense laser pulses. Owing to the broad size distribution of the nanoparticles (10-500 nm) and the variation in laser intensities within the interaction volume, the detected events range in occupancy from near-zero to extremely high, approaching the readout limits of the detector. By combining time-of-flight and velocity map imaging (VMI) techniques, detailed post-processing and analysis were performed. The results presented here focus on the performance of Tpx3Cam under high-occupancy conditions, which are of particular relevance to this study. These particles, dispersed in amorphous matrix thermoplastics, have shown to maintain their shape during the injection molding process. Yet, the Indonesian business context has also been notoriously impacted by corruption, which presents a serious challenge for international business negotiations. Interestingly, in scholarly research little is known about the indigenous management practices within the Indonesian context. We conduct an inductive analysis of rich and context-specific data generated through 55 in-depth interviews of Indonesian and non-Indonesian managers. Two-photon absorption (2PA) enables such applications yet leads to slow processing speed due to the underlying non-linear absorption process. Here, Split-Aperture Xolography (SAX), is introduced which uses stepwise excitation of dual-color responsive molecules to initiate a linear volumetric photo-reaction process that is up to several orders of magnitude more efficient than 2PA. The capabilities of SAX are investigated in a scenario study for focusing systems with high numerical aperture (NA) using a Python implementation of vectorial diffraction theory. The intersecting half-cones generated by the split illuminated entrance aperture of the objective reduce the axial focal spot size of the activation distribution by up to a factor of two compared to 2PA targeting the same electronic transition. A steep average decline of the activation probability with the fifth power away from focus is found for a wide range of directions. This is significantly better in comparison to 2PA and prevents that undesired out-of-focus excitation events sum-up with subsequent irradiations. Different rheumatic diseases present characteristic patterns (image features) in fluorescence optical imaging (FOI). We developed an atlas of FOI image features and tested its ability to differentiate various rheumatic diseases. Methods FOI images from patients with rheumatoid arthritis (RA), psoriatic arthritis (PsA), connective tissue diseases (CTD) and osteoarthritis (OA) were analysed by two readers blinded for diagnosis and calibrated against each other, using the prima vista mode (PVM) and an automated 5-phase model. Twenty-six different reoccurring typical signal enhancement patterns (features) indicating inflamed joints, nail or skin were defined and all FOI images were scored accordingly. The feature frequency in each patient cohort and phase (PVM, 5-phase) was counted. Contingency tables were created with categorical variable counts and diagnosis using common formulae. Findings Four hundred thirty-eight patients with RA (n=117), PsA (n=110), CTD (n=121) and OA (n=90) were included. Once the data had been categorised, a two-step diagnostic pathway was developed: in the first step, OA was best distinguished from the other diseases with high specificity by five patterns (specificity >0.9, diagnostic OR between 2.34 and 8.24). In a second step, the remaining autoimmune diseases were differentiated from each other by a certain number of features (five for RA, 12 for PsA and four for CTD). Consequently, we applied an AI taxonomy identified in the literature with the aim to identify the stages of the RSP in the focus of research and the algorithms mostly used. We conducted a systematic literature review underpinned by a concept matrix, complemented by a computational literature review (CLR), that employed natural language processing (NLP). The initial 4,579 studies were sourced from three databases and narrowed down to a total of 502. Our major findings indicate that the majority of studies were categorised under the stages "assessment \& selection" and "processing incoming applications" in the RSP. The predominant algorithms in use pertain to the field of NLP and machine learning. The CLR emphasised the significance of ethics in AI research. While our study has expanded the general AI taxonomy by incorporating an ethical perspective and is one of the studies with the most articles used to reflect this topic, it is solely focussing on describing the past. Universities play a pivotal role in this process through the creation and dissemination of knowledge, engaging broad sectors of society to improve understanding and to inspire action. However, universities face barriers in fulfilling their role in addressing complex societal challenges such as climate change. This contribution explores the potential of the arts to foster public engagement and stimulate responsible pathways of research at universities within this context. Using an integrative literature review, the paper provides an overview of conceptual contributions aimed at understanding different modes of the interplay between the arts, science, and society, or, more specifically, of integrating the arts into the university's third mission. The overview is organized around three pillars: (i) arts in knowledge creation, (ii) arts in the communication of scientific research, and (iii) arts in the alignment of research with societal needs. Within each pillar, specific modes of interplay between the arts, science, and society are identified and exemplified with selected climate-related artworks: i) artistic research, arts-based research, arts-science collaboration, ii) arts-based science communication and iii) arts-based responsible research and innovation. The examples hihlight resources, processes and activities enabling different modes of interplay between arts, science and society. Currently, scanning electron microscopy is the established method for investigating such structures. In this paper, we propose a rapid, non-destructive optical analysis technique for the simultaneous measurement of through-silicon vias (TSV) depths, silicon wafer thickness, and residual oxide thickness. The proposed method utilizes Fourier peak shift analysis (FPSA) of reflectance measurements in the near-infrared (1200 nm—2200 nm) spectral regions. The application of FPSA to representative samples taken from a commercial TSV integration process for MEMS and CMOS fabrication demonstrated good agreement with reference scanning electron microscopy measurements, confirming the feasibility of the method for in-line and in-situ metrology. PIC-based pressure sensors detect the change in optical properties, i.e. the intensity or phase of the optical carrier wave inside miniaturized waveguide structures. The detection of ultrasound is achieved by engineering the waveguide architecture such that a pressure causes a high change in the effective refractive index of the waveguide. A range of PIC-based pressure sensors have been reported, but a comparison of the sensitivity of the different approaches is not straightforward, since different pressure sensitive waveguide architectures as well as photonic layouts and measurement setups impact the performance. Additionally, the used sensitivity unit is not uniform throughout the different studies, further complicating a comparison. In this work, a detailed simulation study is carried out by finite element modeling of different pressure sensitive waveguide architectures for a consistent comparison. We analyze three different sensor architectures: (A) a free standing membrane located within a tiny air gap above the waveguide, (B) a waveguide located on top of a deflectable membrane as well as (C) a waveguide embedded inside a pressure-sensitive polymer cladding. The mechanical response of the structures and the resulting changes in mode propagation, i.e. the change of the effective refractive index, are analyzed. The waveguide sensitivities in RIU/MPa for different waveguide types (strip, slot) and polarization states (TE, TM) are compared. Successful value creation can only be achieved if all subject disciplines work together. Universities, too, must adapt to these needs and anchor the teaching of these competencies in their courses of study. In this context, interdisciplinary cooperation cannot be limited to the curricula, but must also bring together the teachers and the learners in a common context. Traditionally, universities have found it difficult to develop interdisciplinary scenarios. The teaching scenarios themselves must already provide interdisciplinary content and learning objectives. The challenge here is multi-level. First, it is a great challenge to connect the modules of a course of study in a thematically meaningful way. Second, it is the linking with other study programs that brings about the most sustainable interdisciplinary effects. One possible solution, which has been tested for several years, is to set up a fictitious company as a digital learning factory, which serves as a basis and reference for the interlinking of individual modules of the various courses of study. The scenarios that can be depicted here are extremely application-oriented, which means that not only can internal university modules be very easily aligned with them, but external partners, can also get a simple participation. Furthermore, the participating courses of study can continue to maintain their original core competencies while participating in the interdisciplinary scenarios via the modules that are aligned accordingly with the learning factory. A company from the vehicle construction sector was selected as the business model in order to be able to map another focus at the same time, autonomous driving. The individual departments of the company are linked professionally with the contents of the individual courses of study. Study programs, such as industrial engineering, form the design department, whereas technology-oriented study programs, such as business computing, represent technical development, in particular the area of driver assistance system development. In addition, the relevant departments also include economics and so on. The special feature, however, is the fact that real products in the form of vehicles on a scale of 1:14 are also created and continuously developed, which is an important success factor of the learning factory. The following article transfers the experiences made into a scheme of different levels of interdisciplinary forms of teaching within the framework of the learning factory and classifies and explains the existing scenarios accordingly. Furthermore, the teaching formats used, which have proven to be effective in imparting competences, are described.}, language = {en} } @inproceedings{BreitheckerSzymanskiStolpmann2025, author = {Breithecker, Janine and Szymanski, Mateusz and Stolpmann, Alexander}, title = {Entwicklung eines robotergest{\"u}tzten Systems zur standardisierten Applikation von Hautpflegeprodukten}, series = {Tagungsband AALE 2025 : menschenzentrierte Automation im digitalen Zeitalter}, booktitle = {Tagungsband AALE 2025 : menschenzentrierte Automation im digitalen Zeitalter}, editor = {Reiff-Stephan, J{\"o}rg and J{\"a}kel, Jens and Reichelt, Dirk}, publisher = {Hochschule f{\"u}r Technik, Wirtschaft und Kultur}, address = {Leipzig}, url = {http://nbn-resolving.de/urn:nbn:de:kobv:526-opus4-20150}, pages = {387 -- 394}, year = {2025}, abstract = {In diesem Beitrag wird ein System zur automatisierten Applikation von Hautpflegeprodukten vorgestellt, das speziell f{\"u}r dermatologische Studien entwickelt wird. In particular in the case of periodic (or cyclic) services, the Periodic Event Scheduling Problem (PESP) is well-established to compute high-quality periodic timetables. We are considering algorithms for computing good solutions and dual bounds for the very basic PESP with no additional extra features as add-ons. The first of these algorithms generalizes several primal heuristics that have been proposed, such as single-node cuts and the modulo network simplex algorithm. We consider partitions of the graph, and identify so-called delay cuts as a structure that allows to generalize several previous heuristics. In particular, when no more improving delay cut can be found, we already know that the other heuristics could not improve either. This heuristic already had been proven to be useful in computational experiments [Ralf Bornd{\"o}rfer et al., 2019], and we locate it in the more general concept of what we denote T-partitions. With the second of these algorithms we propose to turn a strategy, that has been discussed in the past, upside-down: Instead of gluing together the network line-by-line in a bottom-up way, we develop a divide-and-conquer-like top-down approach to separate the initial problem into two easier subproblems such that the information loss along their cutset edges is as small as possible. We are aware that there may be PESP instances that do not fit well the separator setting. Yet, on the RxLy-instances of PESPlib in our experimental computations, we come up with good primal solutions and dual bounds. In particular, on the largest instance (R4L4), this new separator approach, which applies a state-of-the-art solver as subroutine, is able to come up with better dual bounds than purely applying this state-of-the-art solver in the very same time.}, language = {en} } @inproceedings{PadillaMichelLucciCasalbonietal.2015, author = {Padilla-Michel, Yazmin and Lucci, Massimiliano and Casalboni, Mauro and Steglich, Patrick and Schrader, Sigurd}, title = {Mechanical Characterisation of the Four Most Used Coating Materials for Optical Fibres}, series = {Proceedings of the 3rd International Conference on Photonics, Optics and Laser Technology : vol. 1}, booktitle = {Proceedings of the 3rd International Conference on Photonics, Optics and Laser Technology : vol. 1}, editor = {Ribeiro, Paulo A.}, publisher = {SciTePress}, address = {Set{\´u}bal}, url = {http://nbn-resolving.de/urn:nbn:de:kobv:526-opus4-20063}, pages = {96 -- 102}, year = {2015}, abstract = {Optical multimode fibres have a wide variety of applications ranging from industrial to medical use. The photo injector test facility at DESY in Zeuthen (PITZ) is building up an R\&D platform, known as FLASHlab@PITZ, for systematically studying the FLASH effect in cancer treatment with its high-brightness electron beams, which can provide a uniquely large dose parameter range for radiation experiments. In this paper, we demonstrate the capabilities by experiments with a reduced parameter range on a startup beamline and study the potential performance of the full beamline by simulations. Approach. To measure the dose, Gafchromic films are installed both in front of and after the samples; Monte Carlo simulations are conducted to predict the dose distribution during beam preparation and help understand the dose distribution inside the sample. Plasmid DNA is irradiated under various doses at conventional and ultra-high dose rate (UHDR) to study the DNA damage by radiations. Start-to-end simulations are performed to verify the performance of the full beamline. Main results. On the startup beamline, reproducible irradiation has been established with optimized electron beams and the delivered dose distributions have been measured with Gafchromic films and compared to FLUKA simulations. The functionality of this setup has been further demonstrated in biochemical experiments at conventional dose rate of 0.05 Gy s-1 and UHDR of several 105 Gy s-1 and a varying dose up to 60 Gy, with the UHDR experiments finished within a single RF pulse (less than 1 millisecond); the observed conformation yields of the irradiated plasmid DNA revealed its dose-dependent radiation damage. In contrast, aberrantly high levels of triglycerides in the blood ("hypertriglyceridemia") represent a hallmark of the metabolic syndrome and type 2 diabetes. As hypertriglyceridemia has been identified as an important risk factor for cardiovascular complications, in this study we aimed to identify molecular mechanisms in aberrant triglyceride elevation under these conditions. To determine the importance of hepatic lipid handling for systemic dyslipidemia, we profiled the expression patterns of various hepatic lipid transporters and receptors under healthy and type 2 diabetic conditions. A differentially expressed lipoprotein receptor was functionally characterized by generating acute, liver-specific loss- and gain-of-function animal models. We show that the hepatic expression of lipid transporter lipolysis-stimulated lipoprotein receptor (LSR) is specifically impaired in mouse models of obesity and type 2 diabetes and can be restored by leptin replacement. Experimental imitation of this pathophysiological situation by liver-specific knockdown of LSR promotes hypertriglyceridemia and elevated apolipoprotein (Apo)B and E serum levels in lean wild-type and ApoE knockout mice. In contrast, genetic restoration of LSR expression in obese animals to wild-type levels improves serum triglyceride levels and serum profiles in these mice. The dysregulation of hepatic LSR under obese and diabetic conditions may provide a molecular rationale for systemic dyslipidemia in type 2 diabetes and the metabolic syndrome and represent a novel target for alternative treatment strategies in these patients.}, language = {en} } @article{ReunerHengherrMalietal.2010, author = {Reuner, Andy and Hengherr, Steffen and Mali, Brahim and F{\"o}rster, Frank and Arndt, Detlev and Reinhardt, Richard and Dandekar, Thomas and Frohme, Marcus and Br{\"u}mmer, Franz and Schill, Ralph O.}, title = {Stress response in tardigrades: differential gene expression of molecular chaperones}, series = {Cell Stress and Chaperones}, volume = {15}, journal = {Cell Stress and Chaperones}, number = {4}, publisher = {Springer}, url = {http://nbn-resolving.de/urn:nbn:de:kobv:526-opus4-19994}, pages = {423 -- 430}, year = {2010}, abstract = {Semi-terrestrial tardigrades exhibit a remarkable tolerance to desiccation by entering a state called anhydrobiosis. In this state, they show a strong resistance against several kinds of physical extremes. Because of the probable importance of stress proteins during the phases of dehydration and rehydration, the relative abundance of transcripts coding for two α-crystallin heat-shock proteins (Mt-sHsp17.2 and Mt-sHsp19.5), as well for the heat-shock proteins Mt-sHsp10, Mt-Hsp60, Mt-Hsp70 and Mt-Hsp90, were analysed in active and anhydrobiotic tardigrades of the species Milnesium tardigradum. They were also analysed in the transitional stage (I) of dehydration, the transitional stage (II) of rehydration and in heat-shocked specimens. A variable pattern of expression was detected, with most candidates being downregulated. Gene transcripts of one Mt-hsp70 isoform in the transitional stage I and Mt-hsp90 in the anhydrobiotic stage were significantly upregulated. A high gene expression (778.6-fold) was found for the small α-crystallin heat-shock protein gene Mt-sHsp17.2 after heat shock. Based upon a merge of the dictionaries, a corporate value analysis of Germany is conducted. Design/methodology/approach The study builds on a dataset (n > 151 k) of online job advertisements which were scraped from a German job portal. It was pre-processed according to natural language processing standards. For analysing the values of an organization a dictionary based word count was applied. Therefore, the current state-of-the-art dictionaries were tested, and an enhanced dictionary was developed and translated from English to German. Finally, a cluster analysis was conducted. Findings This study supports the possibility of measuring cultural values in texts where the enhanced dictionary based on Ponitzovskiy shows the best results. It thereby supports the use of the Universal Value Structure model (Schwartz, 1992) as well as the Signalling Theory (Guest et al., 2021), that values spread across 10 core or 4 aggregated dimensions are communicated via online job advertisements. Finally, the study offers a profile of the German corporate culture average as well as 4 cultural clusters and separate organizations, all with different profiles. One common taxonomy of competency domains for health professions is from Englander et al., where this paper aims to conduct a large-scale analysis based on topic modelling to investigate the extent to which the competency framework for the healthcare sector is applied in the German job market of health professions. Design/methodology/approach The quantitative NLP analysis of a dataset consisting of 3,362 online job advertisements of nurses and doctors was scraped from a German job portal. The data was pre-processed according to Miner et al. For the analysis, the authors applied unsupervised (e.g. HDP, LDA) and supervised (BERTopic) methods and content analysis. Based on the extracted topics a word list was created and these words were coded to existing dimensions of the competency framework of Englander et al. or new dimensions were created. Findings Comparing methodologies, HDP (unsupervised) and BERTopic (supervised) were the best performing while the BERTopic algorithm outperforms HDP. For the doctor dataset 46\% of one main dimension was identified but with an overall coverage of 69\%, for the care dataset is weaker with 30.8\% but an overall coverage of 100\%. Additionally, the taxonomy was enhanced with supplementary competencies of "personality/characteristics" and "leadership" as well as two facets of job description which are "place of work" and "job conditions". Originality/value On the one hand selected dimensions of the taxonomy could be clearly identified but on the other hand, there is a documented gap between the taxonomy and the competencies advertised. One cause may lie in the NLP algorithms but applicants may also have the same difficulties when reading the OJAs. Thus, practitioners should carefully review OJAs regarding better separating explicit competencies they are searching for.
