Influencer marketing represents nowadays one of the most popular marketing strategies online. Given its significant expansion, it is necessary to gather a better comprehension of this widespread phenomenon. The present paper aims to explore the impact of influencer marketing on consumer behaviour in order to offer novel insights to the literature on influencer marketing and to provide the practitioners with meaningful knowledge in the domain. Specifically, the research investigates in-depth how this marketing strategy influences the consumer behaviour during the consumer decision making process and how factors of different nature such as influencers’ trustworthiness, consumers’ self-esteem and wishful identification as well as sales promotions affect their behaviour. This study applied an inductive, qualitative approach because of the exploratory nature of the research purpose. The empirical study was conducted by means of semi-structured interviews with ten young female adults from the Republic of Moldova. The interviews concentrated on respondents’ personal experiences and attitudes regarding influencer marketing and it researched particularly the beauty and fashion industries on Instagram. The findings of this study uncovered and explained the buying behaviour patterns of female consumers that appear as a result of influencer marketing influence throughout the consumer decision making process. Beyond this, the findings illustrate how different influencing factors shape the resultant consumer behaviour. Subsequently, appropriate theoretical and practical implications were derived. UX professionals are responsible for communicating user needs and UX requirements in a clear and understandable way so that other team members such as software developers can work in a user-centric way. The thesis examines known forms of requirements specification and identifies possible challenges in communicating user needs with the help of a systematic literature review. Based on design patterns, possible solutions for these problems are developed. In an interview study, these solution proposals are compared with real scenarios and adapted. As a result, the thesis presents nine guidelines that UX professionals can use to communicate user requirements. The idea is to promote a good exchange with the other team members and suitable, short forms of specification. A key point was that these should always convey a reason or the user’s motivation for a requirement to provide meaningful context information to those who have never been in touch with the user. Die Zielsetzung dieser Masterarbeit besteht darin, mit Hilfe einer Online-Umfrage die Forschungsfrage „Wie wirkt sich Online-Yoga in Zeiten von COVID-19 auf die Gesundheit aus?“ zu untersuchen. Die statistischen Ergebnisse zeigen mäßig bis starke psychische Belastungen aufgrund der COVID-19-Pandemie, welche in besonders hohem Maße die Belastungsfaktoren Antriebslosigkeit, Niedergeschlagenheit, Interessenlosigkeit sowie das Gefühl, keine Energie zu haben und Schlafstörungen umfassen. Das Praktizieren von Online-Yoga verhilft bezugnehmend auf diese Belastungsfaktoren zur Linderung und entfaltet insgesamt eine positive, gesundheitsfördernde Wirkung. Darüber hinaus stärkt die Online-Yoga-Praxis die individuellen Fähigkeiten sowie Strategien zur Anpassung und Bewältigung im Hinblick auf die viruseindämmenden Maßnahmen. Mit steigender Dauer und Häufigkeit der Online-Yoga-Praxis geht auch eine größere gesundheitsfördernde Wirkung einher. Außerdem zeichnet sich anhand der Ergebnisse ein gestärktes Gesundheitsbewusstsein ab. Diesbezüglich gilt ebenfalls, je länger Yoga praktiziert wird, desto häufiger liegt die Motivation in der Verbesserung der psychischen und körperlichen Gesundheit begründet. Die Wirkung des Online-Yoga wird als beruhigend, entspannungsfördernd und positiv im Hinblick auf die körperliche Fitness sowie Ausgeglichenheit beschrieben. N2 - This master´s thesis investigates the significance of online yoga offerings as a health-promoting measure with special regard to the COVID-19 pandemic. Yoga is one of the oldest philosophical teachings and methods, which strives for the unity and strengthening of body, mind and spirit through physical exercises in conjunction with controlled breathing exercises (Schöps 2020, p.6). Interventions to combat COVID-19 lead to severe restrictions on daily life. In particular, contact restrictions, curfews and closures of recreational, cultural and educational facilities increase psychological strain the longer they are maintained. Due to hygiene regulations and closure of sports facilities, physical exercises or sports activities are usually limited to one´s own home. Since practicing yoga in face-to-face classes is no longer possible due to virus-containment measures, online-based alternatives, such as yoga-apps, real-time video broadcasts, or videos from video portals, are necessary and becoming more frequented. The objective of this master´s thesis is to use an online survey to investigate the research question, ´What is the impact of online yoga on health in the era of COVID-19?`. The statistical results show moderate to severe psychological strain due to the COVID-19 pandemic, which particularly includes listlessness, dejection, lack of interest, lack of energy and insomnia. The study shows that practicing online yoga helps to alleviate these mental stress factors, having a positive, health promoting overall effect. Furthermore, the online yoga practice strengthens individual skills and strategies for adaption and coping with virus-control measures. With increasing duration and frequency of online yoga practice, comes greater health benefits, as several studies have stated. Moreover, the results indicate a strengthening of health awareness. Referring to this, the longer yoga is practiced, the more often the motivation is based on improving mental and physical health. The effect of online yoga is described as calming, relaxing and positive in terms of physical fitness as well as level-headedness. Concluding ultimately, the right utilization of online yoga interventions is started to be effective and health-promoting, just as practicing in face-to-face classes guided by a yoga teacher is. Yet oloids are relatively obscure and there are few works dealing with detailed analysis of their flow profile and comparison against standard stirrers. This work tries to add to this topic. In this thesis the potential of deploying oloid stirrers in bioprocesses and wastewater treatment in general is to be probed by computational fluid simulation and compared against a propeller and a Smith turbine (modification of a Rushton turbine). Parameters of interest are flow and pressure profile as well as shear stress. The results show that oloids have much lower shear at their surface compared to both propeller and Smith turbine. The flow is pulsating and in three dimensions, and while the results need verification, it seems like the oloid is well usable for agitation in ungassed bioprocesses, because it can mix a small water body at much lower stirrer speeds than rotating impellers. The application in an actively gassed vessel and whether it is usable for active gas dispersion has not been analysed. Based on data generated by others, the oloid can also be used efficiently as surface aerator in natural open water bodies. Zinc is an essential mineral for the production of alcohol as it is a co-factor in alcohol dehydrogenase, the enzyme responsible for alcohol production in yeast. The general consensus of zinc levels needed for yeast fermentation is 0.4 – 1.07 PPM (Nicola and Walker, 2011). METHODS: Data was collected using a refractometer of the brews. There were five brews in total, one with yeast nutrient, two with zinc sulfate at 0.5 PPM and two at 1PPM along with their controls. RESULTS: A total of 25 brews, in five groups of five, were performed. The results showed that supplementing the yeast with commercial yeast nutrient significantly improved the fermentation rate, while zinc sulfate at 0.5 and 1PPM showed no significant difference when compared to the controls. CONCLUSIONS: While a certain level of zinc is needed for fermentation, this study did not find supplementing homebrew cider with zinc sulfate to significantly improve the fermentation rate in comparison with control brews. When compared to yeast nutrient, zinc sulfate was not found to improve the fermentation rate. In order to consider whether or not supplementing zinc can improve fermentation in homebrewing, more studies must be conducted with different factors taken into consideration such as the yeast variant, the juice used and the initial concentration of zinc compounds in the juice. Y1 - 2021 ER - TY - THES A1 - Schultner, Sarah T1 - Vertrauen in Künstliche Intelligenz am Beispiel des automatisierten Fahrens N2 - Als ein Bereich der Künstlichen Intelligenz erfährt automatisiertes Fahren immer mehr praktische Anwendung. Eine der wichtigsten Herausforderungen des automatisierten Fahrens ist die Entwicklung des Vertrauens der Fahrenden. Die vorliegende Arbeit soll ein Verständnis darüber schaffen, wie sich dieses Vertrauen erhöhen kann. Der Fokus liegt dabei auf der Wirkung anthropomorpher Eigenschaften des Fahrzeugs sowie auf der individuellen Neigung, Maschinen zu vertrauen, im Zusammenhang mit dem Vertrauen in automatisiertes Fahren. Innerhalb eines quantitativen Forschungsdesigns wird mit Hilfe einer onlinebasierten Umfrage das Vertrauen von insgesamt 134 Personen befragt. Eingeteilt werden diese in eine Kontrollgruppe mit 69 Personen und eine Experimentalgruppe mit 65 Personen. Zuerst wird die Neigung, Maschinen zu vertrauen, getestet. Im Anschluss werden vier ausgewählte Einsatzszenarien des vollautomatisierten Fahrens beschrieben und das jeweilige Vertrauen in diese befragt. In der vorliegenden Studie zeigt die Auswertung der Umfrage, dass anthropomorphe Eigenschaften des Fahrzeugs zu keiner Erhöhung des Vertrauens führen. Der Zusammenhang einer allgemein höheren Neigung, Maschinen zu vertrauen, kann hingegen mit dem Vertrauen in automatisierte Fahrzeuge festgestellt werden. Deutlich wird, dass nicht nur das automatisierte Fahrzeug, sondern auch der*die Fahrende selbst bei der Erhöhung des Vertrauens in automatisiertes Fahren eine wesentliche Rolle spielt. For better understanding, immense research is carried, and state of the art literature review is presented in this thesis. For geometric modeling, a meso scale model of 2x2 Twill weave pattern is imported from TexGen software. Hypo elastic constitutive model is developed for material modeling of the yarn. Inter-yarn frictional behavior is thoroughly analyzed to explore the effects of different frictional models and contact types, to produce a cost-effective process on the outcome of final composite product. For an efficient investigation, a meso-finite elemental analysis is performed by replicating picture frame test virtually in Abaqus/Explicit solver. Suitable contact formulation and multiple friction models are used to input the frictional parameters on meso scale model. Finally, the simulation results are then validated by comparing between corresponding contact algorithms and frictional models. Objective of this thesis: The main objective is to determine the mechanical properties, concentrating on effects of friction of a Twill pattern weave by performing virtual simulations by replicating picture frame test (PFT). Solution process: Adequate literature research is accomplished for better understanding of concepts of simulation. In material modeling, hypo elastic material is defined for material properties is imported through VUMAT. Multiple frictional models, including VFRIC and VFRICTION are added to the model to define frictional behavior. In-plane shear deformations are calculated by numerical simulations virtually in Abaqus/Explicit solver and then results are examined. Key Results: Satisfied simulation results are obtained as expected. The frictional behavior between yarns is sensitive to contact algorithms and friction models, and quality of material modeling. The shear locking phenomenon is captured beyond 55-degree shear angle and observed that regardless of value of coefficient of friction yarns, the type of frictional models influences simulation results and force values. Dieser Schutzmechanismus kann sich allerdings erheblich auf die Chemotherapie auswirken, indem Wirkstoffe aus der Zielzelle heraustransportiert werden. Da der Effekt von MDR1 in einigen Fällen unerwünscht ist, kommt es zum Einsatz von Inhibitoren. Diese lassen sich je nach Toxizität und Herkunft in verschiedene Kategorien einteilen. Diese Arbeit gibt zunächst einen wissenschaftlichen Hintergrund. Zuerst werden Multiwirkstoffresistenzen im allgemeinen beschrieben, bevor MDR1 genauer beleuchtet wird. Danach werden verschiedene Bereiche der Krebserkrankung erläutert, um im Anschluss die unterschiedlichen Klassen der Inhibitoren vorzustellen. Even though the vaccine BCG exists for tuberculosis since the beginning of the 20thcen-tury, it lacks efficacy and does not provide sustainable protection. An important step to develop a long-term immunity is the expansion of antigen-specific T cells in the lymph node. For T-cell priming to occur, the pathogen and/or its antigen has to be transported from the site of infection to the T-cell zone of the draining lymph node. This step is done by migratory Dendritic cells (DCs). DCs reside as sentinels in tis-sue, where after pathogen encounter, internalize the pathogen, process its antigens and migrate to the draining lymph node. Using a modified protocol, developed by Bollampali et al. (2015), during a previous study done Dr. Rothfuchs team, we investigated the mechanisms involved in the migration of DCs after injection of BCG. For this we injected mice with different inhibitors and recep-tor antagonists. It is worth mentioning that in a yet unpublished previous work we in-hibited the enzymes COX-1 and COX-2 systemically, using indomethacin, and showed a decreased number of DCs migrating to the draining popliteal lymph node after BCG in-jection compared to mice not injected with the inhibitor. Following our experiment, we decided to investigate the receptors of one of the down-stream products of COX, PGE2 and the corresponding receptors EP2 and EP4. Based on other studies two of the four receptors of PGE2, EP2 and EP4, are important for the mi-gration of DCs. We then injected mice with antagonists for these two either separate or simultaneously. Even though their affinities and the sustainability of the reaction caus-ing PGE2signalling are different, both receptors were able to compensate for each other, but a decreased number of DCs were found in the lymph node when both antagonists were injected. This and the results of the COX inhibition suggest a role for PGE2 on DC migration. Based on these results we decided to look at MMP-9, a molecule regulated by PGE2 sig-nalling. However, inhibition of MMP-9 in our model did not impact on DC migration. It is unclear if treatment with the MMP-9 inhibitor used failed or if MMP-9 is indeed not important in BCG-triggered DC migration. In conclusion in this thesis we show a role for EP2 and EP4 on DC migration after BCG injection but failed to show the same for MMP-9. Among the different classes of enzymes, PLP-dependent enzymes are of special interest for industrial biocatalysis due to their wide range of applications. As it is known, that the cofactor PLP is instable under light exposure, the aim of this thesis was to investigate, whether this affects the activity of PLP-dependent enzymes. Therefore, six PLP-dependent enzymes consisting of four amine transaminases from Chromobacterium violaceum (Cv2025), Vibrio fluvialis (VfTA), Bacillus megaterium (BmTA), and an eleven rounds mutated Arthrobacter sp. (AsTAmut11) and two lysine decarboxylases from Escherichia coli (EcLDC) and Selenomonas ruminantium (SrLDC) were analyzed in different natural and artificial light setups. Cv2025, BmTA, and SrLDC have approximately 40%, 30%, and 50% reduced activity in the light compared to dark condition respectively. EcLDC shows highest inactivation of about 90%. VfTA is stable in HEPES buffer but has 50% inactivation in TRIS and KPi buffer. AsTAmut11 is a special case, where there is no inactivation seen in light condition, but the enzyme gains about 30% and more activity over the course of 6 h. This was seen in the dark and light condition. Conclusively, not only light itself effects the enzyme activity, but also the reaction conditions in combination with the light. The whole study proves, that it can be of immense importance to protect some PLP-dependent enzymes from light. Not only during the reaction, but also during the purification procedure. To mediate proper analysis of LDCs, a new photometric assay was developed using the indicator p� nitrophenol, in contrast to the available work intensive HPLC analysis. This indicator covers pH range from 6 to higher than 8 and enables sensitive analysis of cadaverine concentration with high sensitivity below 1 mM. With optimized photometric cadaverine calibration, the photometric assay is a reasonable alternative to HPLC based analysis. Um einen hohen Sicherheits- und Qualitätsstandard des Kosmetikums zu gewährleisten, ist die Sauberkeit der Produktionsanlage fundamental [IKW, 2019; DIN EN ISO 22716: 2008]. Die chemischen und mikrobiologischen Parameter, welche das Reinigungsergebnis beeinflussen, wurden am Beispiel der Tubenabfüllanlage der bb med. product GmbH untersucht und bewertet. Die chemische Rückstandsanalyse, welche mittels pH-Wert Messung der letzten Spülflotte durchgeführt wurde, diente dem Nachweis von Peressigsäurerückständen. Die Messergebnisse lieferten keinen Verdacht auf gesundheitsgefährdende Rückstände in der Produktionsanlage. Der mikrobiologische Zustand der Anlagenoberflächen wurde mittels Tupferverfahren überprüft. Die Auswertung zeigte, dass die Oberflächenkeimzahl gegenüber dem Richtwert um ein Vielfaches erhöht war. Die Wirksamkeit der eingesetzten Prozesschemikalien wurde in praxisnahen Versuchen ermittelt, bei denen Polyvinylchlorid-Oberflächen und Edelstahloberflächen artifiziell mit verschiedenen Matrices aus Sheabutter und Rinderblutserum kontaminiert wurden, in welche die Prüfkeime Staphylococcus aureus und Pseudomonas aeruginosa eingebettet wurden. Die Simulation verschiedener Anwendungsbedingungen zeigte signifikante Korrelationen zwischen dem Reduktionsfaktor, der Beschaffenheit des Oberflächenmaterials und der Art der Anschmutzung. Auf Edelstahl ließen sich konstant höhere Reduktionen erzielen als auf den PVC-Oberflächen. Des Weiteren konnte die fetthaltige Sheabutter Kontamination mit einer verminderten Wirksamkeit der Reinigungsmittel und insbesondere der Desinfektionsmittel assoziiert werden. Der zusätzliche Einfluss mechanischer Energie trug nicht eindeutig zur Steigerung der Reduktionsleistung bei. Zwischen den geprüften Desinfektionsmitteln auf der Basis von Glutaraldehyd, Isopropanol und Peressigsäure konnten keine signifikanten Wirkungsunterschiede festgestellt werden. The gathering of information was done in three ways: 1) online research to gather basic information about the species, 2) communication with the Umweltbetriebe Kleve and the Niederrhein forestry office to obtain information about the situation in Cleve, Germany 3) visits to the different black locust stands to get an understanding of the situation. Black locust is a pioneer species with a short life span and is considered as an invasive neophyte in many European countries due to its strong suckering and ability to advance into habitats which then undergo an irreversible change in soil and species composition leading to biodiversity loss in especially threatened habitats such as dry grasslands. This is opposed by black locusts’ strong biomass production, making it well suited for plantation production, resulting in a strong, durable and fast growing multiuse wood as well as nutritious foliage which can be used as animal fodder or for the production of bioenergy. The 9 visited black locust stands situated in and around Cleve, Germany are well established middle-aged stands not used for the production of high value wood. These stands were considered as not threatening to their adjacent environments due to their being surrounded by streets, waterbodies and well maintained fields and parks making a spread unlikely. As the stands are quite old already letting them fade out by natural succession would be the recommended management method. Should the need for removal present itself, girdling would be the suggested technique as, even though it is quite time intensive, it is the least forceful method and thus minimises the possibility of a strong suckering reaction. In conclusion it can be said, that its usage and management system strongly depend on the location and closeness to threatened habitats, considering that, the environmental aspect should always be put before the economic benefits. Based on the information from several kinds of literature, the composition of nut and fruit shells were studied, and the shells demonstrated the possibility to produce a wide range of materials such as bio-adsorbent like activated carbon; construction materials like concrete block, earth block, bricks, cement, etc.; and bio-composites like WPC, bio-plastic, etc. Besides these antimicrobials, drugs, insecticides, nanoparticles, and impact-resistant materials could be prepared from nut and fruit shells. Moreover, the production process for these materials was explained in the later part of the paper. Also, the yield, cost, and performance of materials prepared from nut and fruit shells were discussed and found that some of the products were as good as commercially available materials. The environmental impact being one of the important parts, materials from nut and fruit shells had a beneficial impact on nature. However, the entire life cycle assessment suggested that better techniques are required to attain a lower negative impact on the ecosystem. Since nut and fruit shells were the easy and low-cost raw material, they had a greater chance of replacing other expensive, non-environment-friendly raw materials and their products. However, the impacts on soil microbial community structure and activity have been not yet well characterized. The aim of this study was to assess the effects that frass from black soldier fly reared on 3 different diets may cause when applied fresh and dried (70°C) on a sandy loam soil, in 2 application rates (0.2 and 2% w/w). For that, frass was chemically characterized before a 28-days incubation, at the end of which samples were analysed from the chemical and biological point of view. It was found that thermal treatments and diets didn’t vary significantly in microbiota stimulation, even though a chemical analysis would suggest the potential for enriched-frass from pig effluent and wheat bran to encourage microorganisms differently from beer & coffee diet. Furthermore, fresh frass contrarily to what expected didn’t contribute with living microorganisms to enhance microbial activity and growth. However, a difference was reported for the application rates, with the 2% leading to more mineralisation than control. A trend for 2% dried frass from B&C was found, indicating the establishment of an efficient community of fungi slowly mineralizing recalcitrant and exhausted frass. Results indicate that more studies testing application rates ranging between 2 and 2.5% have to be conducted. In fact, there is evidence that BSFF between the mentioned application rates could enhance a fast microbial growth leading to a substantial mineralisation of frass, thus availability of nutrients in the short and long term. People are aware of it vaguely, it is difficult for them to see what actually the companies have or its value. According to the General Data Protection Regulation, finally, individuals can request their collected data from the companies. Nevertheless, the data has been offered as machine-readable files mostly that individuals cannot understand. Thus, a data visualisation application for individuals was designed to improve these problems in this project. The application was built as low fidelity and high fidelity prototypes and tested with 10 participants through remote usability tests to explore its possibility. In the tests, the target users showed their positive interest in the concept and visual design. Consequently, it clearly showed its potential to improve the situation. However, some participants seemed to worry about sharing their data for donating, or unsatisfied with the basic level of data visualisation. To cope with these issues and to have a better effect, some improvement of interface design, additional introduction screens and advanced data visualisation was suggested as a further improvement for the future. Since the distance is the main driver for CO2 emissions, methods were introduced to have shortest route distances after clustering of ALTANA’s shipments in space and time. This thesis aims to demonstrate methods to set benchmarks for distances or indirectly CO2 emissions. Besides complex distance calculations like Travelling Salesperson and Vehicle Routing Problems, a recommender system was also created for sea shipments. Last but not least, a simulation of social learning of consignees in a graph was also demonstrated. Geospatial and graph theory analyses were accomplished mostly with open-source tools. The main tools were PostGIS, Pysal, Geopandas, QGIS, pgRouting, Vrpy, and NetworkX. Terrestrial ecosystem plays an essential part in the uptake of “greenhouse gases (GHGs) such as Nitrous oxide (N2O), Methane (CH4) and Carbon dioxide (CO2)” through photosynthetic sequestration of atmospheric CO2 released from fossil fuel emission, cement manufacturing and deforestation. Rhizodeposits is the released carbon from the root rhizosphere containing a large amount of carbon, essential nutrients and organic compounds. This process is known as rhizodeposition and is often quantified as carbon derived from rhizodeposition (CdfR). This thesis aims to review selected existing literature on above and below-ground biomass carbon (AGB-C and BGB-C) inputs which were used to quantify the carbon sequestration of rhizodeposits in the below-ground biomass (BGB) of different terrestrial ecosystem biomes in the world and to know where the carbon sequestration is lacking the most. Measurement of rhizodeposition in the soil is mostly carried out in the laboratory involving the use of tracing root-derived C by isotopic and non-isotopic techniques, but in this thesis, about 60 papers from different sources were reviewed, and data on above- and below-ground biomass carbon were collected to quantify the C rhizodeposition in the terrestrial ecosystem biomes. The carbon content of the dry weight biomass was estimated by multiplying the above-ground biomass (AGB) and below-ground biomass (BGB) by 0.45. The C-rhizodeposition from the BGB-C was calculated by multiplying the BGB-C by 0.5. Quantification of global C stored in the biomes, including arable crops, was quantified by multiplying the world land area of different biomes with average AGB-C and BGB-C. The estimated values of the mean, median and range of AGB-C, BGB-C and C-rhizodeposition of different biomes (tropical forest, boreal forest, temperate forest, temperate grassland, tropical grassland) and arable crops were reported in this paper. The global C-storage of above- and below-ground of different biomes and arable crops were also quantified. The values were 210, 116, 93, 7.4, 47.2 in Gt C for the tropical forest, boreal forest, temperate forest, temperate grassland, tropical grassland, and 2.4 Gt C for below-ground arable crops only. Even though the comparison between these ecosystems might be an issue because different methods were used in evaluating the above- and below-ground vegetations plant, which in turn can influence the final quantity of C allocated in above- and below-ground biomass, the findings from this thesis for the biomes show that tropical forest has the highest C stored below ground while arable crops have the least. This problem is prevalent in the Central Asian countries as well. A viable solution for this and further environmental issues would be the cultivation of a fast-growing tree species, such as poplar. Several poplar cultivars were tested on plots in Almaty, Bishkek and Khorog. The performance of the trees was assessed by investigating growth in terms of height and diameter, death rate, being bent down, pest infestation, and phenology. The influence of environmental conditions and the influence of genotype were discussed. Both factors significantly influenced height, diameter at breast height and Death Rate. For basal diameter and diameter in 2m height, the location was the single influencing variable. For characteristic of being bent down, cultivar was the single influencing variable. For the percentage of trees that were infested by pest as well as for the extend of pest infestation none of the factors were seen as having an influence. Even though there are some bio-physical and socio-economic constraints, there are many opportunities for the cultivation of poplars in Central Asia. Y1 - 2021 ER - TY - THES A1 - Ade, Carolin T1 - Bedeutung des CT-Wertes für die Epidemiologie und den klinischen Stellenwert beim pandemischen neuen Coronavirus SARS-CoV-2 - Semiquantifizierung mittels Polymerase-Kettenreaktion N2 - Einleitung: Derzeitig ist die PCR die zuverlässigste und sensitivste Nachweismethode einer Infektion mit dem neuen Coronavirus SARS-CoV-2. Die real-time PCR bietet die Möglichkeit einer Semiquantifizierung der Viruslast über die Interpretation des cycle-threshold (CT)-Wertes. Dieser gibt die Anzahl der PCR Zyklen an die nötig sind, um ein Fluoreszenzsignal der Patientenprobe zu detektieren. In dieser Arbeit wurden die CT-Werte der SARS-CoV-2-PCR untersucht und die Ergebnisse mit demographischen Daten der Patienten korreliert. Methoden: Es wurden die CT-Werte der positiv getesteten Proben retrospektiv vom 15. März bis 15. September 2020 ausgewertet. Abstriche der Patienten wurden in 500 μl 0,9-prozentiger Salzlösung ausgewaschen. Virale RNA wurde im MagNA Pure 96 mittels des „DNA and viral NA small volume“ Kit extrahiert. Die RT-PCR erfolgte im Rotor Gene Q mit 45 PCR Zyklen. Der CT-Wert des S-Gen jeder positiven Probe sowie der CT-Wert der positiven Kontrollen wurde dokumentiert. Statistische Analysen erfolgten unter anderem mit dem Mann-Whitney U-Test, dem t-Test, dem F-Test, der ANOVA und dem Bonferroni post-hoc Test. Ergebnisse: Im Untersuchungszeitraum wurden insgesamt 65.878 Patienten über Nasopharynx-Abstriche auf das neue SARS-CoV-2 untersucht. Davon wurden 1.103 (1,7%) Proben mittels RT-PCR positiv getestet. Auf Grund der Berechnung der Fehlergrenzen wurden nur Proben mit einem CT-Wert der positiven Kontrolle zwischen 25,10 und 29,70 ausgewertet. Daher wurden 26 Proben exkludiert und 1.077 positive Proben zur Auswertung herangezogen. Von den untersuchten Patienten waren 566 (52,5%) weiblich und 511 (47,4%) männlich, 944 (87,6%) Proben wurden ambulant und 133 (12,3%) stationär entnommen. Positiv getestete Frauen waren signifikant älter als Männer (50,93 versus 45,04 Jahre; P=0,006) und hatten einen geringfügig höheren CT-Wert (25,44 versus 24,77; P=0,04). Die CT-Werte der stationär und ambulant entnommenen Proben wiesen keine signifikanten Unterschiede auf (25,80 versus 25,03; P=0,1), wobei stationäre Patienten signifikant älter waren (55,11 versus 47,15 Jahre; P<0,0001). Im Vergleich der Altersgruppen wiesen lediglich die Gruppe der über 80-jährigen signifikant höhere CT-Werte auf (P<0,001). 39 von 79 (49,4%) CT-Werten von Kindern und Jugendlichen waren kleiner oder gleich 24 und waren damit infektiös. Nur 3 von 79 Kindern (3,8%) und Jugendlichen im Vergleich zu 130 von 998 Erwachsenen (13,0%) wurden stationär behandelt. Der longitudinale Vergleich der CT-Werte zeigte keine signifikanten Schwankungen im Analyse- Zeitraum. Fazit: CT-Werte eignen sich zu einer detaillierteren Überwachung der SARS-CoV-2 Pandemie. Das Alter wie auch das Geschlecht sind wichtige Co-Faktoren bei SARS-CoV-2 positiven Patienten und kann Einfluss auf die CT-Werte in der SARS-CoV-2-PCR haben. To be specific in North-Rhine-Westphalia where several cooperatives have been founded and become major player in the agribusiness sector. Those coops have been developed in the Münsterland, a region north-west located in NRW which is famous for their intensive livestock-business and small amount of arable land, and in general is the home of small, structured family-farms. In the past, cooperatives have been the only one to provide information and products to the farmers in this region. Due to those circumstances the farmers in this region have a strong attachment to “their coops”. As a result of this specialty several joint ventures of cooperatives have been observed. For this purpose, a research was conducted analyzing three Virtual Reality social apps (VRChat, Rec Room, and AltSpaceVR) to identify —under the perspective of Jenkins’ Convergence Culture— the existing activities and behaviors shaping participatory cultures inside of the platforms. Neben Vorteilen wie dem Wegfall der Anfahrt zum Arbeitsplatz und erhöhter Job-Autonomie stellt sich die Frage, ob Teleheimarbeit auch negative Konsequenzen mit sich bringt. Ziel der vorliegenden Arbeit ist es, zu untersuchen, wie Teleheimarbeit das Wohlbefinden von arbeitstätigen Personen beeinflusst. Methodik – Auf Grundlage von Fragebögen wurde eine Online-Befragung entwickelt. Teilnehmende waren Personen, welche seit Ausbruch des Corona-Virus in Deutschland arbeitstätig waren (N = 200). Es wurden die Variablen Teleheimarbeit-Intensität, subjektive soziale Isolation, arbeitsbezogene Stressreaktion sowie die dem Work-Family Conflict zuzuordnenden Variablen Family Interference with Work und Work Interference with Family erhoben. Ergebnisse – Es wurde ein positiver Zusammenhang zwischen der Teleheimarbeit-Intensität und subjektiver sozialer Isolation, arbeitsbezogener Stressreaktion sowie Family Interference with Work gefunden. Es zeigte sich eine vollständige Mediation durch subjektive soziale Isolation beim Einfluss der Teleheimarbeit-Intensität auf die arbeitsbezogene Stressreaktion. Die Anzahl von Kindern unter 18 Jahren, welche während der Teleheimarbeit betreut wurden, moderierte den verstärkenden Einfluss der Teleheimarbeit-Intensität auf Family Interference with Work, indem dieser durch die Anwesenheit von Kindern verstärkt wurde. Relevanz der Untersuchung – Die Arbeit liefert wichtige Erkenntnisse über das Verständnis von Belastungen während der Corona-Pandemie und kann als Grundlage für weitere Untersuchungen dienen. In gas distribution, optimizing is a crucial task involving control decision on the network elements with respect to trader’s demand at the boundary nodes. The application of artificial intelligence is becoming popular in diverse domains, making its role in gas transport networks more and more interesting. The opposite interest of the players in the network is the reason behind the thought of establishing a game analogy here. The agents as players act on the basis of the opponent’s action. Since both agents perform actions from different space, a neural network model is designed for the dispatcher agent to learn to make decisions for active elements based on its experience from the sample gas transport network. Behavior of the network and its elements is depicted using an optimized gas network simulator. The representation of the gas transport network and its state is digitally transformed so that it is able to extract the features by an artificial neural network representing the environment and its rules. With the help of the trained neural network, the dispatcher agent decides the control decisions with less accumulated penalties. The results and findings of the proposed method are subjected to a comparison with the interval halving method. The results of the study will open the door for further research possibilities. In this study, three main analytical techniques were used to comprehensively analyse the urban surface water quality. Luminescent bacteria test is a rapid and sensitive bioassay for acute toxicity determination in water. Through the inhibition rate of bacteria, the toxicity of water quality is reflected in a short-term. In this study, Vibrio fischeri bioassay was used to determine the toxicity in urban surface water. However, when the luminescence results of samples have only a slight difference, AAS and TOC can be used as sensitive chemical analytical techniques. atomic absorption spectroscopy (AAS) can analyse the heavy metal concentration and total organic carbon (TOC) can determine the organic contamination in water. AAS and TOC can quantitatively and qualitatively analyse chemical parameters. In the study, although each technique has a certain limitation, the combination of these three experiments can be detailed explain the cause of toxicity of urban surface water. Therefore, the advantage of these three methods which are fast, highly sensitive, and easy to operate, makes the analysis of water quality more efficiently. This paper joins this conversation on how we can try to measure and understand integration in the context of which possibilities are offered by sex work. Analysing through the lens of Bourdieu’s forms of capital, and illustrating how refugee men use sex work to initiate upward social movement. For this paper seven MRSW (Male Refugee Sex Workers) were interviewed. The collected data was evaluated by the method of qualitative content analysis and by taking various important analytical categories (motivations, identity, practice) supported by the existing literature, into consideration. The findings show that MRSW use sex work in diverse ways, in order to generate different kinds of profits: While raising economic capital served as the main motivation, cultural and social capitals were generated as well. Language skills, peer bonding, and being able to understand German culture served as the greatest outcomes of sex work. As the motive, we can see that financial instability drives MRSW to sex work, but that social media plays a huge role in making sex work appear as a efficient and feasible option. For the interviewed MRSW in this research, sex work did show to have a positive impact on their ability and capability to integrate into German society. Da in Deutschland ein Psychotherapieplatzmangel besteht werden in dieser Thesis die Fragen erörtert, (1) wie die ICBT als Innovation zu charakterisieren ist, (2) ob sie sich als kosteneffektive Behandlungsform für Menschen mit einer Depression eignet und (3) welche Zielgruppen davon abgesprochen werden könnten. Theoretischer Hintergrund: Bei der Depression handelt es sich um eine häufige und oftmals unzureichend behandelte psychische Erkrankung, was zu hohen Gesundheitskosten führt. Die Digitalisierung des deutschen Gesundheitssystems ist noch nicht stark ausgeprägt, wird jedoch durch Gesetzgebungen und der Evaluation digitaler Behandlungsformen ausgebaut. Die ICBT basiert auf der kognitiven Verhaltenstherapie, welche bei Depressionen eine hohe Wirksamkeit zeigt. Durch die Erprobung der ICBT in anderen Ländern könnte diese eine wirksame Innovation für die deutsche psychiatrische Gesundheitsversorgung darstellen. Methodik: Für die Beantwortung der Fragestellung wurde eine Literaturanalyse von ICBT- Behandlungen bei unipolaren Depressionen im Zeitraum von 2015-2020 in den Datenbanken Pubmed und Cochrane Library durchgeführt. 24 RCT-Studien und Metaanalysen entsprachen den Einschlusskriterien. Zur Analyse der Innovation wurden die in den Studien verwendeten ICBT-Programme als Innovation charakterisiert, mit Erfolgsfaktoren verglichen und die Wirksamkeit erhoben. Zudem wurde eine Kosten-Effektivitätsanalyse (KEA) durchgeführt. Ergebnisse: Bei der ICBT handelt es sich um eine Prozess- sowie marktmäßige Innovation für das deutsche Gesundheitswesen, welche sich als Basistechnologie etabliert hat. In der Wirksamkeitsanalyse konnten in 20 Studien eine Reduktion der depressiven Symptome erhoben werden. Die durchschnittliche Effektstärke der ICBT gegenüber der Kontrollgruppe lag bei 0,34. Ein Problem stellte die geringere Adhärenz bei der ICBT-Behandlung dar. Bei der KEA stellte sich die ICBT-Behandlung im Vergleich zur Kontrollbedingung als kostenintensiver heraus. Diskussion: Die ICBT zeigt sich in den meisten Studien als gleichwertig zu einer Face-to- Face-Behandlung. Zur Steigerung der Adhärenz sollten geleitete ICBT-Programme bevorzugt werden. N2 - Introduction: Internet-based cognitive behavioral therapy (ICBT) for the treatment of depression has already found its way into health care in some countries. Since there is a shortage of psychotherapy places in Germany, the questions will be discussed in this thesis, (1) how Internet-based cognitive behavioral therapy can be characterized as an innovation, (2) whether it is suitable as a cost-effective form of treatment for people with depression in Germany and (3) which target groups could be discussed. Theory: Depression is a common and often inadequately treated mental illness, which leads to high healthcare costs. The digitization of the German health system develops slowly but is being expanded through legislation and the evaluation of digital forms of treatment. ICBT is based on cognitive behavioral therapy, which is highly effective in treating depression. By testing the ICBT in other countries, it could represent an effective innovation for German psychiatric health care. Methodology: To answer the question, a literature analysis of ICBT treatments for unipolar depression in the period 2015-2020 was carried out in the Pubmed and Cochrane Library databases. 24 RCT studies and meta-analyzes met the inclusion criteria. To analyze the innovation, the ICBT programs used in the studies were characterized as innovations, compared with success factors, and their effectiveness recorded. In addition, a cost- effectiveness analysis (CEA) was carried out. Result: The ICBT is a process and market-related innovation for the German healthcare system, which has established itself as a basic technology. In the analysis of effectiveness, 20 studies dealt with a reduction in depressive symptoms. The mean effect of the ICBT was 0.34. A problem of the last adherence in the ICBT treatment represents. With the KEA the ICBT treatment turns out to be more cost-intensive compared to the control condition. Discussion: Most studies show that ICBT is equivalent to face-to-face treatment. To increase adherence, guided ICBT programs should be preferred. The success factors are partially fulfilled, but this depends on the type of implementation in the healthcare system. The ICBT still has the potential to save treatment costs in the long term. Finally, possibilities were shown which could facilitate the implementation of ICBT in the German health care system. Nepal is among the countries most affected by both indoor and outdoor PM2.5 pollution in the world. This thesis is based on the recent lockdown that happened in Nepal during the pandemic of COVID-19 in 2020. This study investigates the impact of lockdown on PM2.5 emission based on the assessment of emission inventories. The annual data for PM2.5 emission was extracted from EDGAR (Emissions Database for Global Atmospheric Research) and then the extrapolation was done to calculate the assumed PM2.5 emission for 2019 and 2020 using linear regression. The assumed nationwide monthly emission ercentage was used to calculate the PM2.5 emission during lockdown months. The PM2.5 sources affected during the lockdown were identified and their assumed percentage contribution (6.18%) was used to calculate the emission during lockdown month. The ambient air PM2.5 concentration data was extracted from the official websites of the government of Nepal. The lockdown months were compared with the preceding year's same month and the percentage change was calculated. The study suggested that in 2020 the PM2.5 emission rate for April was reduced by 4.68% and for May it was reduced by 4.85% whereas during an assumed 'no lockdown‘ year, all the months would have had an increase in PM2.5 emission by ~1.6%. The concentration of PM2.5 for March was educed by 14.65%, similarly, for April it was reduced by 13.96% and for May it was reduced by 50.92%. The comparison of the percentage change of both the PM2.5 emission and concentration shows that the decrease in concentration was much higher compared to the reduction in emission due to the lockdown. The monthly assessment of PM2.5 during the lockdown suggested that the percentage change in emission of PM2.5 was minor compared to the change in the concentration of PM2.5 which might be due to different reasons including meteorology, underestimation of the contribution of some emission sectors by the emission inventories and the fact that the emission inventories are set up for the whole country whereas the PM2.5 concentration data are collected with monitoring stations located in cities close to major PM2.5 sources. However, Platinum is an extremely expensive material. Therefore, increasing loading could contribute to the price significantly. The aim of this work is to measure the proton exchange membrane fuel cell performance under three different catalyst loading on cathode and determine the optimum loading for performance in these three tested loading levels. The tested cathodic loadings are 0.3 mg/cm2, 0.6 mg/cm2 and 0.9 mg/cm2. By applying two methods: current-voltage measurement and electrochemical impedance spectroscopy, the open circuit voltages, polarization, power output and impedance were recorded. From the experiment, it is found that the internal resistance decreased with increasing Platinum loadings on the cathode. The simulation results indicate this change is mostly contributed by decreasing cathodic resistance. With increasing cathodic Platinum loading, the cathodic resistance decreases. It is also found during experiments, the polarization, open circuit voltage and power performance first increased and then stayed at same level. These indicators showed best performance at 0.6 mg/cm2 cathodic loading level. Combined with simulation results, the large impedance value of constant phase element in equivalent model for 0.9 mg/cm2 loading is considered as the reason why the power performance stop improving. The thesis is done in collaboration with the medical device company, FEG Textiltechnik mbH, which produces medical meshes used in hernia repair. To deal with the problem, the market of mplantable medical devices in the healthcare sector as well as market analysis tools were analysed on the basis of a literature analysis. On the basis of these information a good approach was developed and tested in a case study including two test phases. One is a comparison with a market analysis from the collaborating company, FEG Textiltechnik mbH, while the other one is a theoretical example of a market analysis on the Australian market for the previously named company. The developed good approach aims at a market entry in a foreign market. The case studies showed that the good approach has a valuable potential for companies in the industry of implantable medical devices. However, some points were falsely included, which would have been better fit at later stages of the market entry. However, the good approach can be used by companies in the industry, due to its broad fit and can be further optimised with real world applications or tests and can be used outside of the industry after some modifications. Jedoch hat nicht zuletzt der Ausbruch der COVID-19 Pandemie gezeigt, dass der Austausch Face-To-Face nicht immer gewährleistet werden kann und technologische Alternativen benötigt werden, die explizit kollaboratives Lernen unterstützen. Auf Grund des mangelnden Angebots an Anwendungen für kollaboratives Lernen, besteht die Überlegung Kollaborationstools zu nutzen, dessen theoretisches Konzept der Kollaboration eine hohe inhaltliche Übereinstimmung zum kollaborative Lernen aufweist. Um diese Forschungslücke zu füllen, können demnach zwei Fragen gestellt werden. (1) Was sind zentrale Elemente des kollaborativen Lernens und (2) können diese Elemente in dem Kollaborationstool Microsoft Teams realisiert werden? Dazu werden zunächst vier unterschiedliche Theorien zum kollaborativen Lernen dargestellt in denen sich 35 zentrale Elemente des kollaborativen Lernens identifizieren lassen. Diese werden zu 10 Faktoren zusammengefasst und mit weiteren Ergebnissen aus der Empirie betrachtet. Abschließend erfolgt der Vergleich zwischen den Faktoren und den Funktionen von Microsoft Teams. Der Vergleich zeigt, dass 6 der 10 Faktoren bereits durch die Anwendung realisiert werden. Bei weiteren 4 Faktoren bedarf es der Optimierung der Anwendung. Infolge der Limitationen, wie der begrenzten Auswahl der Theorien oder dem unstrukturierten methodischen Vorgehen bedarf es weiterer Forschungen. In zukünftigen Studien sind vor allem empirische Untersuchungen und die Interaktion der vorgestellten Faktoren interessant. The loss i.e., the decrease of biodiversity, does not only have negative effects on vegetation and animals, but also on humans. If nature is doing well, people are doing well. In order to counteract the loss, this bachelor thesis attempts to increase plant and insect biodiversity at a specific location with insect-friendly seed mixtures and by measuring the change using comparison areas. Accordingly, the two research questions were addressed: (1) "How does plant-biodiversity change in the backyard and front yard on the seed mixture plot compared to plant-biodiversity in the backyard and front yard next to the seed mixture plot?" and (2) "How does insect-biodiversity change in the backyard and front yard on the seed mixture plot compared to insect-biodiversity in the backyard and front yard next to the seed mixture plot?" To answer these research questions and to obtain more suitable data, a wildflower strip was established in the backyard and one in the front yard, each with an adjacent reference field, to ensure equal environmental conditions and to detect changes in flora and fauna. Plants and insects were observed and documented during the flowering period in order to determine differences in biodiversity sizes afterwards. The comparison of the different biodiversity sizes shows that an increase in insect and plant biodiversity can be achieved by planting insect-friendly wildflower strips. As the increase of plant biodiversity also leads to the increase of insect biodiversity. Further research in the field of biodiversity enhancement could be done by collecting more measurement points of the coefficients at additional locations to extend the data set and to improve the applied method of this bachelor thesis. The investigation of acylcarnitines in cell and tissue samples is a particularly complicated task due to their low abundance and high heterogeneity. This requires applications of sophisticated lipid separation methods. The purpose of this work was the development of an ESI-MS/MS method for the quantification of acylcarnitines with high specificity and reliability for the identification of low abundant acylcarnitine species in complex samples. General aspects of acylcarnitine analysis, including several types of solid-phase extraction, and derivatization with liquid extraction were investigated using commercially available acylcarnitine standards. The established experimental workflows, including an optimized cation-exchange extraction and derivatization with liquid extraction, were successfully applied to quantify acylcarnitine species in samples of biological origin. Consequently, murine brain tissue samples were quantitatively analyzed using two extraction procedures. The method developed in the study can be used for the qualitative and quantitative analysis of acylcarnitines in different biological samples. Die Trennung von fluiden Stoffen wird erläutert und sichtbar dargestellt. Während des Destillationsprozesses entstehen unterschiedliche Heizleistungen, die im Verlauf dieser Arbeit zur Ölausbeute zum Zeitfaktor angepasst werden. Die erstellten Diagramme geben Aufschluss über die Dauer und die unterschiedlichen Heizleistungseinstellungen. Eine Trennung der Phasen während des Destillierprozesses wird detailliert dargestellt. Some of the properties include affinity, solubility, organic solvent resistance, enantioselectivity and thermostability. The probability of succeeding in directed evolution experiment depends on the size of the library generated, because the more variants are present, the higher the chances of detecting an enzyme with improved properties. The throughput represents the main bottleneck in enzyme screening, as most of the methods allow the analysis of 104 variants per round, which is not enough for a library size of 109 variants. Ultra high-throughput screening (uHTS) enables the analysis of 107 events per hour. It can reduce the time, costs and consumables needed for an applied round of directed evolution. One of its limitations is the use of a flow cytometer, which is expensive and not affordable for everyone. Also uHTS is restricted to single catalytic reactions and their coupling with synthetic fluorophores. The objective of this project is to develop an uHTS method based on fluorescence activated cell sorting (FACS). The principle of this method is to sort cells expressing the active enzyme variant, which have a fluorescent hydrogel formed on the cell membrane. Successful completion of this protocol will allow the screening of different enzyme classes, for example, laccases, alcohol dehydrogenases, organophosphorus hydrolases. In order to prove that the concept is working, laccase will be used for generating the library, which will then be displayed on the surface of E. coli. The variants which are enzymatically active, will produce the radicals that initiate the polymerization reaction of fluorescent hydrogel, allowing the flow cytometer to sort cells based on the fluorescence emitted. To reduce the effect of wind on the irrigation pattern, the sprinkler velocity and radius of throw need to be adapted to the wind’s velocity and direction. Our control valve box contains seven major components. Wind and rain sensors that will collect the weather conditions, an LCD where irrigation time will be displayed, additionally to a microcontroller communicating with a second MC at the sprinkler head box via Bluetooth and controlling an electrically actuated ball valve and solenoids connected to inline pressure reducers. These three latter components play an important role in supplying proper water conditions for the sprinkler by adjusting the pressure and water flow. The position of our sprinkler’s nozzle will be detected with a magnetometer and compared to wind direction. The servo motor will rotate the sprinkler and this latter will start irrigating in a 180° pattern for couple of minutes before rechecking again all the conditions. When the sprinkler nozzle is facing the wind the velocity of throw will be increased, and after it is rotated and is in the same direction as wind, the velocity of throw will be reduced, because wind is already assisting the water droplet reach its radius. If the wind velocity is larger than the threshold value, or if it is raining, the system will not start although the switch is turned on. Using data from non-moderated group discussions among schoolchildren aged between 14 and 18, who were involved in LGBTTIQ support projects in different schools within the German Federal State of North Rhine-Westphalia, the article reconstructs how concepts of norms and normality, heteronormativity, difference and otherness are being articulated by the students. A sequential analysis of the group discussions is used to detect the meaning of norms and normality with respect to LGBTTIQ identities, to highlight the way in which LGBTTIQ-supporters are constantly being subjected to this discourse. Simul-taneously it detects a concealed conflict between the goal setting of LGBTTIQ support and heteronormative discourse. At present, elderly homes use manual processes for data collection, which leads to poor data quality, considerable inefficiency and burden on the caregiving staff. This thesis describes the current market situation and proposes a technological solution on how to improve the quality, effectivity, and efficiency of the elderly care sector by using data collection and utilisation techniques. Moreover, it justifies the advantages of technological usage, which assists the caregiving staff in relieving the burden of daily tasks and, importantly, helps to collect high-quality data that could be used to do predictive analysis and automate the monotonous processes involved in the elderly homes. A well-documented fully functional digital application prototype was developed and tested by the elderly health industry related individuals of different age groups. The digitisation of the data collection process played a significant role in the collection of high-quality data. Furthermore, the results indicate that usage of that data has a direct correlation with the emergency response time, which could be reduced by almost half. However, further research is required to improve the predictive analysis system and the user experience of the application. The package comes with two wind sensors - i.e. wind vane and anemometer, which are mounted on a pole. The pole can be placed anywhere in the garden as long as the portable sprinkler stays within a range of 100m. When switched on, the controller on the sprinkler gets continuously updated with the current wind speed and direction using wireless transceivers. The sprinkler can also be controlled through an app on a smartphone. Simple commands such as ON/OFF switch, as well as timers for autonomous watering are possible to set up. For example, setting up the system to water the garden at sunrise, which has been suggested by experts as the most effective time to irrigate plants. Morning irrigation, on the one hand, supplies the grass and plants with enough water to sustain high temperature during the daytime and on the other hand, prevents fungal disease growth and different insects from harming the plants during the night by reducing moisture. Therefore, our sustainable irrigation system not only takes into consideration water conservation, but also provides best methods for having healthy plants in a garden. Our competitive advantage is that our sprinkler is made of durable materials and will save household costs as a long-term investment with efficient water usage methods. There are no other sprinklers in Germany that match our design and usability, which makes our product unique in the market. Because migration is always a multicausal phenomenon, this study aims to disentangle the impact of environmental factors from other migration-inducing factors to shed some light on the complex relationship between the environment and migration. Thus, we conducted quantitative microlevel studies in low-lying communities in two high-mobility countries - Ghana and Indonesia - that are particularly exposed to coastal hazards like erosion, land subsidence, storm surges and an increasing sea level, and are prone to flooding on a regular basis. Different measures of environmental threats were collected, ranging from individual perceptions over the household’s distance to the coast to expert opinions. We analyzed the relationships using logistic regressions and controlled for contextual factors on multiple levels. No statistically significant direct impacts of slow-onset environmental events on migration decisions could be detected. Perceptions of storms, a clearly sudden-onset event, however, were found to be significantly linked to out-migration decisions in Ghana. These findings support the hypothesis that environmental factors are generally not a primary cause of migration and their effects are rather context specific - especially for slow-onset changes. The results of the work demonstrate an active use of both traditional and digital marketing strategies such as direct mail, printed media in a form of catalogues and magazines, telemarketing, trade shows, sales promotions, face-to-face communication, email marketing, website, SEM (Search Engine Marketing), SMM (Social Media Marketing), blogging and video marketing in all the interviewed enterprises. Brand creation is important in the shipping industry and is the main reason for an implementation of marketing strategies by maritime firms due to a great amount of advantages a strong brand brings. Lockdown has been implemented in many countries to prevent the virus from spreading. Lockdown has prohibited people to go outside of their houses, closed industries and factories, and restricted air transportation and the free movement of vehicles, except emergency vehicles and flights. On the optimistic side, improvements in ambient air quality due to the lockdown have been reported across the world. This study evaluated ambient air quality by analyzing the PM2.5 concentrations monitored in 7 major cities of Nepal during March, April, and May of the years 2019 and 2020. Furthermore, meteorological data were also evaluated to investigate how meteorological conditions have affected Nepal’s ambient air quality during the investigation period. The monthly mean PM2.5 concentrations during March, April, and May 2020 were found to be lower in nearly all of the 7 cities of Nepal as compared to March, April, and May 2019. To some extent, a slightly higher amount of precipitation in March-May 2020 than during March-May 2019 supressed the ambient PM2.5 concentrations in 2020. But, since the highest decrease in PM2.5 concentration was found in Nepalgunj (72.78%) followed by Kathmandu (46.2%) during May 2020 as compared to May 2019 and thus, the highest differences between 2019 and 2020 were found for the cities with the highest contribution of transport emissions to PM2.5 in ambient air, the COVID-19 lockdown might have contributed to the lower PM2.5 concentrations in 2020. On the other hand, it can be assumed that one of the major primary sources of PM2.5, biomass burning (both the residential use and natural forest fires), was not affected by the lockdown, but biomass burning might be the reason for the increment of PM2.5 concentrations found at some sites, in particular in the Kathmandu Valley, and for the only subtle decrease at other sites during the lockdown. In essense, though it is an unfortunate situation, the lockdown provided a glimpse of the situation how effective improvements in the environmental conditions could be if we were to introduce large scale mitigation measures swiftly. This quote sums up most of the current thoughts on the phenomenon of a ground-breaking invention from 2008 by Satoshi Nakamoto since a major part of the world’s population is still missing out on sophisticated knowledge about it. Especially in times of digitalization, it is of enormous value to be informed about current developments in technology. This thesis aims to evaluate the future development of money, especially if Bitcoin has the potential to totally replace conventional currencies. Furthermore, the strengths and shortcomings of Bitcoin will be discussed. It also reaches to determine characteristics for a currency’s long-term success and which of those characteristics are fulfilled by Bitcoin. To answer the research question if Bitcoin has the potential to be an alternative currency and to test the developed hypotheses, pertinent literature is reviewed and an empirical study consisting of expert interviews which are examined using a qualitative structured content analysis method, are conducted. Afterwards, the results are critically discussed. The results of this thesis reveal that Bitcoin has the potential to become an alternative currency because it fulfills most of the criteria a currency needs to meet to be successful in the long run. Furthermore, it is evident that Bitcoin still faces vast barriers and needs to overcome enormous obstacles, which shifts the time horizon for becoming a success somewhere in the farther future. Additionally, apart from the development of cryptocurrencies, it is shown that money in general, will flow much more digitally in the short run. Besides that, this thesis offers insights about the promising future of Bitcoin’s underlying core technology, the Blockchain, in various fields of application. Their applications range from pharmaceutical practice up to industrial (non-medical) purposes and, therefore, show great importance in today´s life. Most recombinant proteins are being produced by genetic modification of microorganisms to either generate those intracellularly or to secrete the desired protein into their surroundings. Dependent of the expression mechanism, the production can be influenced and improved to achieve high yields and protein purity. One aim of this study was to determine and optimise the most favorable cloning strategy for the expression and secretion of the plasma form of the actin binding protein gelsolin from Pichia pastoris. Gelsolin has two isoforms of which one of them gained particular attention due to its application in research projects concerning the current COVID-19 pandemic. For this project, a variety of oligonucleotide primers were designed to amplify the whole sequences and various parts of both gelsolin isoforms during PCR approaches. The accuracy and efficacy of primers is determined by several factors such as their length and exact structure. Also, many factors depending on the sequence of interest are influencing the outcome of the PCR. Therefore, the second aim of this study was to design and validate primer pairs with most preferable conditions to amplify the sequence for the expression of gelsolin and optimise the PCR conditions for this purpose. Durch die repräsentative Untersuchung „Gesundheitsförderung an deutschen Hochschulen“ von Michel et. al (2018) wurde deutlich, dass deutsche Hochschulen trotz der langjährigen Thematisierung von Gesundheitsförderung durch Expert*innen auf politischer wie wissenschaftlicher Ebene, nach wie vor Defizite aufweisen. Mit dem Rahmenvertrag „Gute Beschäftigungsbedingungen für das Hochschulpersonal“ setzte NRW bundesweit einen hohen Maßstab. Seine Auswirkungen auf die Beschäftigungsbedingungen waren zu diesem Zeitpunkt noch nicht absehbar und gaben mit den Erkenntnissen von Michel et. al (2018) den Anlass, das Thema innerhalb NRWs neu zu untersuchen. Das Ziel der Masterarbeit besteht darin herauszufinden, wie sich aktuell die Arbeit von Gesundheitsbeauftragten, Gesundheitsmanager*innen und Gesundheitskoordinatoren an Hochschulen in Nordrhein-Westfalen gestaltet, woran sie sich bei der Weiterentwicklung ihrer Berufspraxis orientieren und wie sie den zunehmenden psychischen Belastungsfaktoren ihrer Hochschulgemeinschaft begegnen. Um die Thematik hinreichend zu bearbeiten, wurden sechs Expert*inneninterviews mit Verantwortlichen in NRW geführt. Der Leitfaden zu den Interviews wurde auf Grundlage der zehn Gütekriterien des AGH sowie der Ergebnisse der repräsentativen Erhebung von Michel et. al (2018) konzipiert. Auf Basis des generierten Datenmaterials wurde unter Zuhilfenahme eines erstellten Kategoriensystems anschließend eine qualitative Inhaltsanalyse nach Mayring (2015) und den Modifikationen nach Steigleder (2008) angefertigt, um „einen Querschnitt durch das Material zu legen“ (Mayring, 2015: 67) und den Umsetzungsstand an den Hochschulen darstellen zu können. Gemessen an den zehn Gütekriterien des AGH und den Ausführungen der Expert*innen haben die jeweiligen untersuchten Hochschulen noch einen langen Weg vor sich, um sich als „Gesundheitsfördernde Hochschule“ bezeichnen zu können. Ein weiterer Grund ist, dass das Angebot im Internet oft unübersichtlich ist und die Suche nach geeigneten Experten viel Zeit kostet. Erste Anlaufstelle vieler Unternehmen auf der Suche nach Unterstützung ist das Internet. If we consider only European countries, Heating and cooling industry accounts for more than half of the total EU’s energy consumption. And about 75% of the heat is generated by using fossil fuels. Therefore, the major non-renewable energy sources are depleting at the rapid rate and at the same time also producing the climate-changing greenhouse gases. To achieve the climate change objectives, optimised and efficient use of the energy and the decarbonisation of the heating systems are some of the critical steps. The investigation process in this thesis involves the modelling and modifying the case study heating system by using Energy Plus and Design-Builder whole building energy simulation software. It also includes the investigation of the three different possible renewable and non-renewable solutions to optimize the current heating system and analyse their effects in terms of total energy consumption (Gas and Electricity consumption) Economic benefits, environmental and other benefits. With the detail and adequate data inputs modelling and desired modifications has been done to the heating system and the building properties. The annual simulation has been performed to all the three cases and the results have been analysed. The outcome of the analysis of the results shows a significant improvement in overall energy and electricity consumption. Also provides a detailed insight into the total economical investment needed in all three cases and simple payback periods for the investments. And Most importantly the results replicate the environmental benefits such as a considerable reduction in the overall emissions of greenhouse and other toxic gases.
