TY - THES A1 - Erkkilä, Julia T1 - Identification of Cas9-nickase Fusions that Promote HDR-mediated Genome Editing N2 - At the heart of this project was the exploration and improvement of the CRISPR/Cas9 gene editing platform with the goal of enhancing efficiency and ensuring safety. The research carried out was primarily concerned with testing and developing a gene editing system that bypasses the need for double strand breaks (DSBs). Given that DSBs are the most damaging events for a cell and pose a major safety risk in genome editing, eliminating them was a key focus of the project. The project aimed to increase the frequency of homology directed repair (HDR) while simultaneously decreasing non homologous end joining (NHEJ), thereby improving the accuracy of genome editing. The methodology involved the use of a Cas9 protein fused to factors involved in DNA repair. Based on previous evidence that this strategy improves the HDR mediated repair of a Cas9 induced DSB, intention here was to apply the same strategy to Cas9 nickases for a safer alternative to standard genome editing procedures. A Cas9 nuclease can be transformed into a Cas9 nickase (nCas9) that cuts only one strand of DNA by mutating one of the two nuclease domains of the Cas9 either via a D10A point mutation for RuvC or an H840A for HNH. Preliminary experiments had shown that using nickases could reduce the occurrence of DSBs at both target and non target sites. However, because genome editing efficiency of nickases is limited, the project focused on amplifying nickase induced HDR frequency by fusing the D10A nCas9 with DNA repair factors that guide the repair of single strand breaks via HDR. Experiments evaluated the ability of each Cas9 nickase to direct the resolution of single strand breaks via HDR. An eGFP or mCherry cassette under its own promoter was employed as a reporter gene to measure the success of the transfection. The experiments were performed in three genomic loci, namely AAVS1, CLYBL, and H2AX. The degree of eGFP or mCherry expression after 14 days, as quantified by flow cytometry, was used to estimate the knock in success. The results obtained support the hypothesis that fusing D10A nCas9 with selected DNA repair factors leads to an increase in the knock in efficiencies as compared to the unmodified nCas9. The choice of the promoter used to express the nuclease, either CMV or CAG, did not seem to influence the knock in efficiencies. Instead, the type of repair template used did influence the knock in efficiencies with nicked plasmid templates contributing to greater HDR levels than non nicked ones. Knock in efficiencies of Cas9 nickases varied depending on which locus they were tested in. More research remains to be done to test alternative Cas9 nickase fusions. However, this approach represents a promising direction for the creation of a more efficient and safer gene editing instrument for clinical use. This rapid population growth has brought with it the challenges of poverty and food insecurity. Since agriculture and food production are the backbone of food security, there is need to make farms and enterprises operating in that sector sustainable to meet the current food demand. In Africa, most agriculture and food production is conducted by MSMEs. Therefore, taking an inventory of the main actors of agriculture and food activities within the MSMEs is important to broaden and amplify the gains from agriculture and food production. Many studies indicated that women have a high participation rate in the agriculture and food sector in Africa. However, other studies also suggested that women in the agriculture and food sector are economically and socially disempowered due to various gender based discriminations thus decreasing the productivity of agriculture and food MSMEs in Africa. In order to analyse how women empowerment can contribute to the sustainable development of the MSMEs in Africa, a survey study was conducted. The goal of the survey was to identify challenges faced by women in the agriculture and food MSMEs as well as to identify if being a woman employee or a woman entrepreneur increases or decreases women’s vulnerability to the challenges. The findings of the surveys revealed that women are subjected to gender based discrimination that include lack of access to training and financial support. N2 - The utilization of a simple test, such as Pfeiffer's Circular Chromatography (PCC), has the potential to provide deeper insights into all the parameters affecting soil health. However, despite its promise, the method has yet to attain widespread adoption among farmers and universities, primarily due to challenges in interpreting its outcomes. This thesis investigates the application of PCC as a technique for soil analysis at Hochschule Rhein Waal University. The paper analyses compost and soil samples sourced from the Rhein Ruhr region, focusing on the potential of PCC in determining chemical parameters, including nitrate content, pH, and organic matter quantity. As a result, the aforementioned chemical parameters were documented and the economic viability of the methods were compared to PCC. The development of PCC is dependent on environmental conditions. To explore this aspect, the test was conducted under different lighting conditions and assessed by various researchers to gauge its interpretability. The paper evaluates the test's reliability based on its reproducibility. The PCC results were found to be sensitive to environmental conditions; nevertheless, the method displays potential in assessing overall soil and compost health on a relative scale. The paper suggests improvements to enhance the technique's quality and provides recommendations for further research to explore the various possibilities of PCC and its interpretation. In conclusion, with additional research and data collection on the PCC method coupled with the integration of existing technologies such as machine learning and AI, the potential exists to provide farmers with a rapid technique for assessing their soil. Due to their characteristics and uses, biocatalysts are seen as promising alternatives to current methods of CRC diagnosis and therapy. This research-based bachelor thesis aims to summarize and analyze current traditional and biocatalytic methods that exist to diagnose and treat CRC, in order to seek the potential of biocatalysis in this field, as well as discuss its current state and its future in comparison to currently implemented methods. It provides an extensive overview of traditional and biocatalytic approaches to diagnosis and therapy, considering important factors such as efficacy, sensitivity, selectivity, cost, and safety. The thesis also delves into the use of biocatalysis for important points such as the early diagnosis of CRC, and the challenges faced during development, regulation, and monetary concerns. The ability to incorporate citizen science for data collection and evaluation is assessed and the quality of that data is compared using analysis of variance between group types. It was found that fertilization is strongly correlated with feeding activity (0.877). Positive correlations between temperature, soil moisture and soil organic matter content were also observed, although with varying degrees of influence. Mean feeding activities among different evaluating groups of students, staff, pupils, scout members and farming cooperative members (Solawi) did not deviate significantly from each other (p= 0.66 among students, Solawi and staff members, and p=0.52 among students, pupils and scout members, alpha= 0.05) . Data processing however for non staff and student groups was more arduous as more tallying mistakes were made than formerly mentioned groups. Results suggest that the bait lamina methodology is suited for citizen science, although data quality can easily be improved by digitizing evaluation handouts, with automation/formulas embedded for tallying. This transition necessitates an improved un-derstanding of how managerial actions influence the dynamics within these teams, to optimally align them for success. Despite the substantial research about leader effec-tiveness and team effectiveness based on organizational performance indicators, it is rather infrequent that the direct influence of leader behavior on teams is explored. This thesis aims to address this gap by answering how leader behaviors impact team pro-cesses in the context of German medium to large-scale IT-enterprises. Using a quanti-tative cross-sectional methodology, individuals working in team-based structures under direct supervision were surveyed. Analyzing data from 94 respondents via hierarchical linear regression analysis, several significant relationships were identified. In general, leader behavior has a strong positive impact on team processes. Furthermore, task-oriented leaders have the greatest positive influence on action processes, while relation-oriented leaders have the greatest positive influence on interpersonal processes. Change-oriented leaders also significantly influence action processes and interpersonal processes, albeit less strongly than their counterparts. These results provide implications for team management practices, emphasizing the importance of leader behavior for aligning team processes in the direction of success. Hierfür wurden die Daten von 210 Teilnehmenden mittels einer quantitativen Online-Befragung untersucht. Dazu wurde die Kurzskala UGK-V der SEU-3 mit 21 Items genutzt. Diese beinhaltet sieben Inhaltskalen und misst die Konstrukte Einstellung, Verhaltensintention und selbstberichtetes Verhalten. Zudem wurden drei Items zur Erfassung von Bewusstheit von Handlungskonsequenzen genutzt. Es konnten signifikante Zusammenhänge zwischen den umweltrelevanten Konstrukten gefunden werden. Am stärksten korrelierte die Einstellung mit der Verhaltensbereitschaft. Die postulierten Hypothesen konnten bestätigt, müssen aber durch niedrige Reliabilität des selbstberichteten Verhaltens eingeschränkt betrachtet werden. Die Bewusstheit über Handlungskonsequenzen konnte mit allen Unterskalen und der Gesamtskala in Verbindung gebracht werden. Am höchsten korrelierte diese mit dem Umweltbewusstsein. Die angenommene Korrelation (H4) stellte den kleinsten Zusammenhang dar, was darauf hindeutet, dass Bewusstheit für Handlungskonsequenzen eher mit der Einstellung als mit Verhalten in Verbindung gebracht werden kann. Die Kausalität kann nicht festgestellt werden, so ist eine Wechselwirkung nicht auszuschließen. Uneinheitliche Definitionen der Konstrukte erschweren neben einer unzureichend repräsentativen Stichprobe eine Verallgemeinerung auf die Bevölkerung. N2 - The aim of this Bachelor Thesis is to investigate the relationships between environmental attitudes and other factors influencing pro-environmental behavior. For this purpose, the data of 210 participants was examined by means of a quantitative online survey. In this study, the short scale UGK-V of the SEU-3 with 21 items was used. It contains seven content scales and measures the constructs attitude, behavioral intention, and self-reported behavior. In addition, three items were used to measure awareness of action consequences. Significant correlations are found between the environment-related constructs. Attitude correlates strongest with behavioral intention. The postulated hypotheses can be confirmed, though this result is of limited accuracy due to the low reliability of self-reported behavior. It was possible to show a relationship between awareness of action consequences and the total scale, as well as all the subscales. This correlates the most with environmental awareness. The hypothesized correlation (H4) represented the smallest correlation, suggesting that awareness of action consequences may be associated with attitude rather than behavior. Causality cannot be established, so the different aspects may influence each other. Interaction cannot be ruled out either. Inconsistent definitions of the constructs in addition to an insufficiently representative sample make a generalization to any population difficult. The results can be compared with the values of the original sources and show valid survey instruments. Human health and welfare depend on ecosystem functions and services which are interchangeably connected to biodiversity. Amphibians are one of the most sensitive and vulnerable species to anthropogenic impacts, because of their fast reactions to changing conditions in their surroundings they are often referred to as bioindicator species. Since year 2016 an amphibian population located in the district of Wesel in North-Rhine-Westphalia in Germany experiences drastic changes in population size and composition. To find out if anthropogenic impacts may influence the development of the amphibian population, different methods for assessing the population and biodiversity are used. Data was collected from year 2017 to 2023 with the help of a drift fence with pitfall traps. The amphibian population is evaluated regarding population size, age structure, sex ratio and migration behaviour. Additionally, biodiversity indices are used for a quantitative and qualitative analysis. As last step, the amphibian population is compared to another amphibian population with similar preconditions, but experiencing less anthropogenic pressure. A high reduction of population size and changes in age and sex composition were found for the assessed amphibian population. Moreover, the migration behaviour alternates over time. The found results strongly indicate that the main cause of the negative development is a high anthropogenic pressure. Further intensive conservation and monitoring actions are needed, to restore the amphibian population and avoid the complete destruction. Die patientenseitigen Beweggründe für die vermehrte Inanspruchnahme wurden dabei bislang nicht hinreichend untersucht (Reinhold et al., 2021). Auf der Grundlage der bisherigen Literatur scheint das Nachfragewachstum jedoch häufig auf Patienten ohne medizinische Notfallindikation zurückzuführen zu sein (Durand et al., 2012). Methode: Im Zeitraum vom 01.10.-30.11.2022 wurde eine quantitative Fragebogenerhebung im BG Klinikum Duisburg durchgeführt. Der Fragebogen besteht aus insgesamt 16 Fragen. Die Datenauswertung basierte dabei auf deskriptiven sowie inferenzstatistischen Analysen und konnte mehrere Unterschiede zwischen einigen Patientengruppen aufdecken. Es wurde ein 95% Konfidenzintervall festgelegt. Ergebnisse: Im Rahmen der Fragebogenerhebung konnten 548 gültige Fragebögen erworben werden. Dabei zeigten sich die Gründe für das vermehrte Inanspruchnahme-Verhalten multifaktoriell. In erster Linie zählten hierzu antizipierte Qualitätsvorteile innerhalb der Notaufnahmen im Vergleich zu ambulanten Versorgungseinrichtungen seitens der Patienten sowie insbesondere die Unkenntnis der Bevölkerung über bestehende Gesundheitsdienstleistungen im Notfall, wie bspw. den ärztlichen Bereitschaftsdienst. Ebenso ist ein genereller Rückgang der Gesundheitskompetenz zu verzeichnen. Diskussion: Grundsätzlich ist eine fehlende medizinische Notfallindikation bei dem Großteil der Notfallpatienten zu verzeichnen. Das rund um die Uhr zur Verfügung stehende Versorgungsangebot der Notaufnahmen zeigt sich äußerst attraktiv für die Patienten und führt nicht zuletzt zu Bequemlichkeitsentscheidungen. Hierzu wurde eine Bevölkerungsgruppe mit einer besonders hohen Beanspruchung der Stimme im Beruf untersucht: die Berufssprecher. Die eingeschlossenen Studien umfassen Menschen aus westlichen Industriestaaten – überwiegend Lehrkräfte, aber auch Telefonisten von Notfallhotlines und Callcentern sowie Priester und Pastoren. Ziel der Arbeit ist es, durch eine systematische Literaturarbeit die Prävalenz von Stimmbelastungen in sprechintensiven Berufen von 2002 bis heute zu erforschen. Die Studienlage zeigt, dass es akuten Handlungsbedarf gibt. Alle Studien weisen anhand von Selbsteinschätzungs-Fragebögen eine deutlich erhöhte Prävalenz auf. Die häufigsten Symptome sind Heiserkeit, Stimmermüdung nach kurzem Stimmeinsatz und ein trockener Hals. Frauen sind aufgrund anatomischer Voraussetzungen stärker betroffen. Des Weiteren liegen häufig psychosoziale Belastungen vor. Diese verstärken das ohnehin bereits erhöhte Risiko für Stimmbelastungen bei Berufssprechern. Die hohe Prävalenz von Stimmbelastungen führt zu längeren Regenerationszeiten und einer erhöhten Anzahl von AU-Tagen. Daraus resultieren hohe Kosten für das Gesundheitssystem. Durch die Erforschung der unterschiedlichen Prävalenzarten (Punkt-/Lebenszeitprävalenz) in den in dieser Arbeit eingeschlossenen Studien, ist ein Vergleich der Ergebnisse nur eingeschränkt möglich. Zu den Risiken für die (Stimm-)Gesundheit zählen ein vorherrschendes geringes Bewusstsein über Stimmprävention sowie berufsspezifische Risikofaktoren. Folgen können Stimmverlust (Aphonie), psychische Erkrankungen sowie Arbeits- und Berufsunfähigkeit sein. Die Integration präventiver Maßnahmen in die Berufsausbildung (z.B. Stimmlehre, Stimmhygiene, Stimmtraining) sowie Eingangsuntersuchungen werden dringend empfohlen. Dazu ist eine interdisziplinäre Zusammenarbeit zwischen Fachleuten, Berufsverbänden und Politik notwendig. N2 - This paper investigates the impact of the Corona pandemic on M&A activities considering the variables M&A volume, M&A value, GDP, and Covid figures. The aim is to analyze the relationships between the development of the pandemic and M&A activities in five European countries (Germany, the UK, France, Italy, and Spain). With an introduction to the literature published so far on the topic, an excellent introduction to the situations around Corona and the M&A market is already offered. The one or other outlooks are already scientifically proven here and have thus also been very for the following analyses and results. In the first step of the research, we analyze M&A volumes and values in the countries under consideration before and during the pandemic. It can be seen that the number of M&A transactions increased overall during the pandemic, but transaction values were very fluctuating in comparison. This observation certainly reflects the general uncertainties and economic challenges of the pandemic. The next step is correlating Covid figures, M&A activity, and GDP. This shows that the pandemic outbreak led to a short-lived decline in M&A activity, as companies acted more cautiously and held back on transactions due to the uncertainty. However, M&A activity rose again as the economic situation in the countries stabilized, and companies were better able to respond to the pandemic. In addition, the impact of GDP on M&A activity is also examined. It can be seen that GDP was affected in all countries due to lockdowns and restrictions on public life. Nevertheless, there was some resilience in the economy as GDP showed an increase in all countries despite the pandemic. The analysis of the correlation between the variables shows that there is a significant relationship between Covid numbers and M&A activity. The higher the Covid numbers were in a country, the more M&A activity was affected. This suggests that the development of the pandemic had a significant impact on M&A activity. Overall, this work shows that the Corona pandemic significantly impacted M&A activity in Europe. The uncertainty and economic challenges led to a short-lived decline in activity, but the overall number of deals increased. The correlation analysis highlights the links between the pandemic and M&A activity and underscores the importance of thoroughly examining the variable factors influencing M&A decisions under Corona conditions. As a part of the project by Winkelmann et al. (2022), the continuos physiologically structured population model (cPSPM) of P. fluviatilis was established which appeared to have potential points for improvement. In this study, I attempted to modify mortality function of the population model. Hence based on thorough literature research 4 models for mortality estimation mortality as well as the data of life-traits of perch were found. The models include Hoenig-Then (Then et al., 2015), Pauly-Then (Then et al., 2015), Lorenzen (Lorenzen, 2022). and Weibull probability density function as function of weight dependent mortality. The methods were parametrised via arithmetic average of population specific traits and non-parametric least-square method. As a result, the mortality models were compared to each other as well as to the base model. Further on, each mortality model was integrated into cPSPM and stable population was found. At the end only Lorenzen and Weibull models were analysed. As a result of mortality model comparison, it was found that the difference between these models lies mainly in difference of magnitude. Integration of the models into cPSPM showed the potential correlation between the mortality and reproduction. The comparison of simulations results in similarity in population dynamics of our models with experimental data, with main difference in magnitude. Overall results suggest that every model has reason to be applied, and all of them have their own application. In terms of current cPSPM, it would be suggested to consider population dynamics of perch over multiple years period with different mortality method employed for each year to address the issue of variability of population dynamics on yearly basis. The tool offers several ways for playing, enabling players to perceive underlying the issue of Nitrate in groundwater from different perspectives. The tool was tested with 44 participants, in 17 single and 8 competitive game modes. Feedback was collected through survey results, performance evaluation cards, and participants’ verbal remarks and comments. The BOW test rig consists of the wheel that is actuated by the electric motor, motor encoder, and laser sensor. The desired outcome is to stabilize the ball around the equilibrium point on top of the wheel. By employing common control design techniques and establishing appropriate Simulink models the controllers for the BOW system were implemented. The dSpace software was used to capture the performance of the controllers, while MATLAB was used for analysis. However, various issues arose during the design and commissioning processes, preventing the state-space controller to achieve the desired outcome. As a result, a performance comparison was conducted based on Simulink models, as the state-space controller design failed to perform adequately on the BOW test rig. These issues were thoroughly explained within the thesis and the implications were summarized in the conclusion. The observation of the fast-growing and large-shaped leaf has drawn attention to the potential use of biomass of P. tomentosa. The study contains different chapters under the main research question with analysis. The first section starts with a brief background description of Sierra Leone's Agricultural sector in the context of livestock management and its contributions to the country's economy, poverty, and food security. Followed by the research objective, a semi-systematic literature review is conducted based on livestock management's impact and poverty reduction, lessons learned, and analysis of the significant constraint and opportunities for the sector's growth was presented using a conceptual framework. In chapter three, the writer uses a qualitative methodology to identify the different sections and data used in the thesis through a thematic approach. The results show that livestock management can contribute to the district's food security, poverty reduction, and wealth creation. However, factors such as poor road networks, weak governmental institutions/policies, insufficient funding, and lack of raw materials, among others, are critical issues affecting the development of the industry. On the other hand, traditional/cultural benefits and income generation are the district's main significant uses of livestock. The major goal of the survey was to assess the small scale and the large scale dairy farmers level of awareness on sustainability or if at all they employed sustainability practices within their farming systems. The feedback from the survey indicated that small scale farmers either knew little or nothing about sustainability practices. Dairy production is an important aspect of the Zimbabwean economy. It assures that some of the nation’s major nutritional requirements are met, farmers are economically empowered, promotes rural development and impacts positively to the gross domestic production. The thesis showed the current level of sustainability obtained at small scale dairy farms, the challenges faced by small scale dairy farmers, as well as multi-stake holder engagement and government role or involvement. However, little is known about the interactions between heavy metals and microplastics, as well as possible biological effects when the two are combined. It is possible that MPs and heavy metals can interact in soil, causing changes in their environmental behaviour, bioavailability, and potentially toxic properties, further posing ecological risks. Therefore, the objective of this study was to evaluate the individual and combined impacts of microplastics and heavy metals on soil chemical properties, microbial biomass, microbial activity, and microbial communities in soil treated with varying concentrations of these pollutants. In an incubation experiment, soil was amended with low-density polyethylene (LDPE) and a mixture of heavy metals (Zn, Cu, and Ni) in two concentrations (Low and High). Seven treatments (Control; MP-L; MP-H; HM-L; HM-H; MP+HM-L and MP+HM-H) were analysed in seven replications. The rate of microbial respiration was determined during incubation. Following incubation, soil pH, microbial biomass carbon (C), nitrogen (N), soil fluorescein diacetate hydrolytic activity, and the abundance of archaea, bacteria, and fungi were measured as well as the effects on inorganic nitrogen (NO3-, NH4+) were determined. It was found that no significant difference existed between the control treatment and LDPE treatment in terms of soil pH, FDAse activity, microbial biomass, microbial respiration, inorganic nitrogen, and abundance of archaea, bacteria, or fungi, regardless of the concentration of LDPE (0.6%, 1.2%). Heavy metal treatment did, however, result in a significant decrease in soil pH, NO3, and microbial respiration particularly at high concentrations (2.68 mg kg-1), compared with treatments with LDPE and a control, suggesting that HMs suppressed microbial activity. Additionally, HMs also reduced MBN and MBC, indicating their negative effect on microbial biomass. As a result of the combination of microplastics and heavy metals, microbial biomass, and microbial respiration decreased significantly when compared to either the control treatments or the microplastics alone treatment. Additionally, the combined treatment reduces NO3- and soil pH significantly compared to the control and sole microplastic treatment There was no significant effect observed in any treatment on the abundance of archaea or fungi, indicating that the presence of MP and/or HMs had no impact on their abundance. However, the MP+HM treatment resulted in a reduction in the number of bacteria. Using a two-factor statistical analysis, the effect of two concentrations (low and high) of microplastics and/or heavy metals on these soil parameters was further evaluated. According to these results, the concentrations of added microplastics and/or heavy metals (low and high) had no significant effect (p > 0.05) on any soil parameter except for soil pH (p < 0.05). Overall, the results indicate that the presence of heavy metals, either alone or combined with microplastics, had a detrimental effect on soil ecosystem. However, this study was limited to examining the short-term effects of these pollutants on soil microorganisms and their functioning. Additional research will be necessary to determine the long-term effects of these pollutants. In this growing trend, more and more scientists are trying to surf over the waves of bio-inspired solutions, but the fundamental differences between nature’s point of view and man-kind, on managing the resources and solving problems, has made the traditional equipment’s of scientist, obsolete; especially when it come to actuation methods. Since the traditional device and component are not suitable for cutting edge fields, such as bio-inspired robotic, artificial muscles, soft robotic and so on; this thesis has offered an especially designed syringe pump, to fulfill this need. Starting from the basic constraints and expectations for such syringe pump, a morphological box has been shaped in this project; and along with having an eye over relatively similar products in the market, borders of “design function” has been drawn. By considering the available equipment and tools for production of such device, the “design concept” of this thesis has been formed. And finally, after days of manufacturing, the prototype of this concept has been brought to life. In this syringe pump, a stepper motor (NEMA-17) is controlled by combination of computer, Arduino UNO and microstep driver DM542; by powering this stepper, a lead screw will be driven. Designing an aluminum structure and connecting a lead-nut to the lead-screw; linear displacement can be produced. This linear displacement is responsible for movement of a tray, which is holding onto the end of syringe’s plunger; and by clamping the cylinder to the designed station of the thesis; with this configuration, desired static pressure can be produced by displacing the plunger. Having this static pressure, is the key feature for simulating many different biomimetics projects, such as soft robotics and artificial muscles. As the results, this thesis has achieve the high precision and resolution of “100 𝜇m” for each 5 steps of stepper motor, equivalent to 9° of rotation. Furthermore, the prototype have easily and quickly delivered 300 mmHg (39.99Kpa), by using only a single 20ml syringe; and reached the highest speed of 750 rpm for the stepper motor. Such interactions and the occurrence of side effects due to incorrect dosages are also known for the antidepressant bupropion, emphasizing the importance of therapeutic drug monitoring for this medication. Therefore, this study aimed to develop and validate a method for the determination of serum levels of bupropion and its pharmacologically active major metabolite, hydroxybupropion, using LC-MS. Additionally, due to the low stability of bupropion at room temperature, the influence of this factor on the routine measurements of bupropion patient samples in the laboratory of the LVR-Klinik Viersen pharmacy was also examined. Various bupropion solutions (dissolved in methanol or human albumin) were prepared for measurement. These solutions were analyzed using an LC-MS "Exactive Plus Orbitrap" with a biphenyl column as the stationary phase and various mobile phases differing in polarity to allow for the generation of a gradient. An electron spray ionization (ESI) method and an Orbitrap detector were used for quantifying the sample concentration. The measurement of a 0.1 ng/ml bupropion solution and a 0.5 ng/ml hydroxybupropion solution resulted in a signal-to-noise ratio in the mass spectrum that was well above the required 3:1 ratio for background noise subtraction. As the concentration was already below the required detection limit of 0.2 ng/ml for bupropion and 17 ng/ml for hydroxybupropion, no further steps were taken to determine the actual limit of detection for economic reasons since sufficient sensitivity of the LC-MS method was already demonstrated. The investigations into the recovery rate (WFR) and ion suppression showed that the applied sample preparation (protein precipitation with acetonitrile) did not lead to any relevant loss of content and that no interfering effects from other components of the sample matrix occurred during the measurement. The WFR was determined to be 100 ± 5% for bupropion and 101 ± 4.8% for hydroxybupropion. The examination of ion suppression resulted in a content deviation from the target value of 4 ± 0.2% for bupropion and -9 ± 0.1% for hydroxybupropion. The linearity of the method was confirmed by measuring a 5-point calibration with a correlation coefficient for the regression line of approximately 0.99 for both analytes. The precision and reproducibility of the method were also confirmed through the evaluation of in-day and day-to-day variation, with a coefficient of variation of <10 for all measurements conducted for both analytes. Thus, a reliable method for the determination of bupropion content using LC-MS was successfully developed and validated. By participating in proficiency testing programs, the method will be further validated and potentially certified in the future. N2 - Patienten in psychiatrischer Behandlung werden in der Regel mit einer Vielzahl von Medikamenten behandelt, deren Wechselwirkungen und der daraus resultierende Einfluss auf die Serumspiegelkonzentration der Arzneistoffe nicht vorhersehbar sind. Auch für das Antidepressivum Bupropion sind solche Wechselwirkungen, sowie das Auftreten von Nebenwirkungen aufgrund falscher Dosierungen in der Therapie bekannt und unterstreichen die Bedeutung des therapeutischen Drug Monitorings für diesen Wirkstoff. In dieser Arbeit sollte daher eine Methode zur Serumspiegelbestimmung von Bupropion und seinem pharmakologisch wirksamen Hauptmetaboliten Hydroxy-Bupropion für die LCMS entwickelt und validiert werden. Aufgrund der geringen Stabilität von Bupropion bei Raumtemperatur, sollte zudem überprüft werden, welchen Einfluss dies auf die Routinemessungen von Bupropion-Patientenproben im Labor der Apotheke der LVR-Klinik Viersen hat. Es wurden verschiedene Bupropion Lösungen (in Methanol oder Humanalbumin gelöst) für die Vermessung hergestellt. Diese wurden auf einer LCMS „Exactive Plus Orbitrap“ unter Verwendung einer Biphenyl-Säule als stationäre Phase und diverser Fließmittel, die sich in ihrer Polarität unterschieden und so die Erzeugung eines Gradienten ermöglichten, analysiert. Als Ionisationsverfahren kam eine Elektronensprayionisation (ESI), sowie ein Orbitrap-Detektor zur Quantifizierung der Probenkonzentration zum Einsatz. Die Vermessung einer 0,1 ng/ml Bupropion-Lösung und einer 0,5 ng/ml Hydroxy-Bupropion-Lösung ergaben ein Signal-Rausch-Verhältnis im Massenspektrum, welches weit über dem erforderlichen 3:1-Verhältnis für die Abhebung vom Hintergrundrauschen lag. Da die Konzentration bereits unterhalb der erforderlichen Messempfindlichkeit von 0,2 ng/ml für Bupropion bzw. 17 ng/ml für Hydroxy-Bupropion lag, wurden aus wirtschaftlichen Gründen keine weiteren Schritte zur Bestimmung der tatsächlichen Nachweisgrenze unternommen, da eine ausreichende Empfindlichkeit der LCMS bereits belegt war. Die Untersuchungen zur Wiederfindungsrate (WFR) und der Ionensupression konnten zeigen, dass die verwendete Aufbereitung (Proteinfällung mit Acetonitril) zu keinem relevanten Gehaltsverlust führt und keine störenden Effekte durch andere Bestandteile der Probenmatrix während der Vermessung auftreten. Es ergab sich eine WFR für Bupropion von 100 ± 5 % und für Hydroxy-Bupropion von 101 ± 4,8 %. Bei der Untersuchung zur Ionensuppression kam es zu einer Gehaltsabweichung vom Zielwert von 4 ± 0,2 % für Bupropion und von -9 ± 0,1 % für Hydroxy-Bupropion. Die Linearität der Methode wurde durch die Vermessung einer 5-Punkt-Kalibration mit einem Korrelationskoeffizienten für die Regressionsgerade von ~0,99 für beide Analyten bestätigt. Durch die Überprüfung der In-Day- und Day-to-Day-Variation konnte auch die Präzision und Reproduzierbarkeit der Methode bestätigt werden. Dabei ergab sich ein Variationskoeffizient für alle durchgeführten Messungen von <10 für beide Analyten. Somit konnte erfolgreich eine zuverlässige Methode für die Gehaltsbestimmung von Bupropion mittels LCMS entwickelt und validiert werden. The by-product frass is a mix of larval faeces, exoskeletons and undigested substrate. Given its potential as a soil improver, frass could make insect production a “zero waste” industry. However, its positive reputation has not been thoroughly scrutinized with regard to environmental impacts. We therefore conducted experiments to investigate the effects of frass application on the soil microbial community, release of greenhouse gases and heavy metal availability. Gefördert wird das Projekt mit zweijähriger Laufzeit im Rahmen des Focus Forschung HAW Programms des Ministeriums für Kultur und Wissenschaft des Landes Nordrhein-Westfalen. Die Förderlinie FF HAW-Kooperation zielt auf eine verbesserte HS-interne interdisziplinäre Kooperation und den Auf- und Ausbau eines attraktiven Forschungsschwerpunkts. Ausgangspunkt der Arbeiten war das in der Arbeitsgruppe von F. Platte und in Kooperation mit der AG Hygiene (Prof. Bockmühl) von T. Tewes entwickelte Verfahren zur Identifizierung von Mikroorganismen mit Hilfe der Raman-Spektroskopie. Experimentelle Arbeiten wurden zu der Frage durchgeführt, ob die Methodik für die Identifizierung einzelner Bakterien in Gewässerproben anwendbar ist und welche Probenvorbehandlungs- und Datenverarbeitungsprozeduren unter Berücksichtigung der Automatisierbarkeit erforderlich sind. Die Ergebnisse bzgl. der Raman-spektroskopischen Identifizierung der Bakterienzellen waren vielversprechend. Problematisch ist allerdings, dass es gilt, wenige bakterielle Zellen zwischen einer sehr großen Zahl anderer Partikel zu identifizieren. Ideen für ein Raman-basiertes schnelles Partikel-Scan-Verfahren wurden entwickelt. Untersucht wurde auch, ob neben Mikroorganismen weitere Wasserkontaminationen mit Hilfe der Raman-Spektroskopie nachweisbar sind. Hier standen Verunreinigungen durch Mikroplastik (speziell Reifenabrieb), Ölfilme, Sulfat und Nitrat im Mittelpunkt. Im Rahmen zahlreicher „networking“-Aktivitäten wurden Kontakte zu Herstellern von Partikelmess-Systemen aufgebaut und Kooperationen mit Anbietern von Raman-Spektrometern initiiert mit dem Ziel, über F&E Projekte die Entwicklung eines kontinuierlich arbeitenden des Raman-Systems voranzubringen. Projektskizzen wurden entwickelt und Anträge auf Forschungsförderung eingereicht. Duties such as corporate social responsibility reporting as well as shifting customer demands are placing increased pressure on the association events industry to acknowledge the importance of sustainability and implement sustainable event management strategies. Association events tend to have an economic, social and environmental impact on the places in which they occur. These impacts can be both positive and negative. They can also have substantial leveraging and multiplier effects, can be used to elevate awareness of sustainability even further and, most importantly, help transform this awareness into action, for example by influencing stakeholders’ behaviour and minimising behavioural gaps. The events and meetings industry has traditionally relied on economic performance measures, eschewing social and environmental costs and benefits. However, recent studies have underlined the necessity for a more holistic approach. This approach is fleshed out in the present research. It aims to offer stakeholders, decision-makers, event managers, associations and congress organisers a strategical instrument with which to identify the economic, social and environmental impacts of a planned event in order to utilise them in a targeted, sustainable fashion within the respective field of operation. The strategic measures for optimising event impacts were derived from the results of the empirical study. Moreover, these measures will serve to anchor the sustainable association event strategy within business operations as well as internal and external communications. Gifford’s Dragons of Inaction can be applied to determine possible barriers and, finally, to strive to minimise the gap between sustainability consciousness and sustainable behaviour. To this end, the effectiveness of cooperating attitudes, behaviour-oriented forms of intervention such as intentions or incentives for actions, and perceived consequences can be assessed and evaluated in terms of their essential dimensions. Methodology A number of research steps have been undertaken within this model, beginning with a literature review. This laid the foundation for subsequent empirical studies conducted as semi-structured expert interviews, followed by an online Delphi forecast as well as a survey among participants of an association event of the German Scouts (DPSG). Findings As shown in the primary research in both studies, social and economic measures are regarded as most important, environmental measures are most often adopted instead. Not all dimensions are considered to be part of sustainability. Social and economic aspects are considered necessary for an association’s success and are considered important strategic instruments connected mainly to positive implications on a psychological level such as image, trust and satisfaction. As it is seen as a competitive advantage and the terminology is unclear to many in the event industry, communication efforts must be targeted in order to facilitate implementation and, later, allow for the possibility of measuring sustainable event management. Holistic aspects should be made clear in order to support initiatives and ideally reduce the behavioural gap between sustainability awareness and actual behaviour. Associations like the German scouts are risk-averse by nature. They function as custodians of a sector or profession and their standards demand continuity as well as consistency. Innovation, however, is associated with risk, experimentation and (superfluous) change. Both primary and secondary research efforts have shown that barriers to innovation include a lack of time, resources (money or staff in particular), or simply innovation know-how. Therefore, implementing change or, in this case, more sustainable measures in event planning, requires an individual, tailor-made approach which is pro-active and problem-solving in order to promote broader implementation of the already developed Green Events Guidelines throughout the whole association. The findings of this mixed-method approach were used to develop a model to integrate, assess and steer event management sustainability from the perspective of event planners and associations, and propose a new concept for converting theory into applied practice for the industry. Despite the fact that groundwater is frequently abundant and widely utilized, it is difficult and costly to precisely estimate water levels when compared to surface water systems. Groundwater managers require improved digital tools to understand the system state of the aquifers they seek to interact with sustainably (e.g., accessing real-time updates on the state of the water table to check whether pumping operation is achieving or violating a desired drawdown target.) Here we propose a workflow for a pilot region that (a) temporally interpolates irregularly, manually-measured water levels in a real observation network using data from neighboring sensor-equipped observation, using a multiple linear regression approach to reduce complexity. The resulting interpolated time series are then used in conjunction with the sensor data to come up with (b) spatial interpolations of the groundwater field over time using inversed distance weighting (IDW) at any given point in time. From that, spatial estimates of the groundwater field can be transformed into maps of deviation from drawdown targets. We visualize these targets and their temporal evolution for groundwater managers in real-time on a web application. The web application displays these maps together with operational data (e.g., extraction rates) and meteorological data (recent and forecasted precipitation rates). It serves as a foundational tool for groundwater management when making control decisions to save energy in climate change mitigation and to reduce costs. Deep learning is increasingly used for modeling complex systems, which generates more interest in battery state estimation. The theoretical investigation of candidate neural networks for SOC estimation is preceded an experimental approach to evaluate the shortlisted neural networks performance. Publicly available testing datasets representing real driving cycles has been used to train the selected networks, taking into consideration different operating temperatures for the exact cycles. Testing the prediction capability of the models across a realistic range of operating conditions is the driving factor for including testing cycles with a temperature range of [0–40] °C in the training datasets. A demonstration of the full training cycle is conducted, along with the optimization problem of network parameters. This work demonstrates the applicable neural network family used for training this kind of sequential data, followed by the shortlisting of two neural networks based on their performance using literature research. After selecting the networks, finer evaluation criteria is applied, including both their prediction capability and training requirements. The results show that both networks, long short-term memory (LSTM) and gate recurrent units (GRU), offer satisfactory prediction capability with mean absolute error (MAE) of (2.9%, 2.7%) respectively, when averaged across the tested temperatures. The GRU network on the other hand had significantly higher resource requirements and a more complex architecture. Finally, a more recent architecture is briefly discussed, borrowing the strength of a parallel application field. N2 - This thesis combines the findings of several publications and studies on the influ- ence of offshore wind farms on marine life, in Northern Europe. The properties and characteristics of this offshore wind farm areas were compared to the characteristics of marine protected areas and differences as well as conformities were highlighted. It is evident, that the negative consequences accompanying the implementation of offshore wind farms are less concise, than the public opinion assumes. Although scientific knowledge gaps in multiple related fields for example, the influence of elec- tromagnetic fields on individuals or the influence of noise on benthos species, are present, offshore wind farms offer chances to contribute positive effects for marine life in terms of de facto protected areas and new possibilities for population growth. The planned extensions of offshore wind farms in geographical relation to marine protected areas could enhance the size of protected areas and connect remote ma- rine protected areas with each other. Es konnten geeignete Pflanzen für Dachbegrünungen im Allgemeinen, als auch für das zukünftige Projekt am KvGG bestimmt werden. H1 konnte innerhalb eines Vergleiches der verschiedenen Begrünungsarten bestätigt werden. Darüber hinaus konnten erfolgreich Kennzahlen bezüglich der Abflussmengen entwickelt werden. Im Rahmen dieses Versuches konnte die Hypothese H2 bestätigt werden, also dass eine Bepflanzung zu einer erheblichen Verlangsamung des Abflusses von Regen führt. Durch die Schülerumfrage konnte der Einfluss des Projekts auf die Wahrnehmung der Schüler:innen mit einem leicht negativem Ergebnis untersucht werden, H3 kann also nicht bestätigt werden. Abschließend lässt sich festhalten, dass Dachbegrünungen eine wertvolle ökologische Maßnahme darstellen. Es sind umfangreiche Einflüsse auf verschiedene umweltrelevante Bereiche möglich. Darüber hinaus eignen sich Dachbegrünungen vor allem an Schulen auch aufgrund ihres pädagogischen Mehrwerts. Zukünftig werden ähnliche Projekte hoffentlich in einem steigendem Ausmaße ihren Anteil zum Klimaschutz beitragen. Dazu wurden 93 Studierende in einer quantitativen Onlineumfrage befragt. Die Umfrage basiert auf der Employer Attractiveness Scale von Berthon, Ewing & Ha aus dem Jahr 2005 sowie auf der Authentic Living Scale von Wood et al. aus dem Jahre 2008. Ergänzungen wurden in den Bereichen ethischer Diversität, Unternehmenskultur, Glaubwürdigkeit, Authentizität und persönliche Werte vorgenommen. Diese Aspekte sind im Fragebogen von Berthon, Ewing & Ha im Vergleich mit anderen Arbeitsattraktivitätsmodellen, beispielsweise das Treiberfaktorenmodell von Nagel (2011), unterrepräsentiert, haben allerdings laut einer umfassenden Literaturrecherche für die jungen Generationen Z und Y an Wichtigkeit zugenommen. Die Ergebnisse liefern einen umfassenden Vergleich zu allgemeinen Aspekten der Präferenzen beider Generationen an den Arbeitgeber. This research aims to develop a smart gas burning system built on reliable and secure communication channels bounded with the implemented protocol specifications. A close study of each protocol specifications as well as a bit-by-bit investigation is therefore performed, moreover, a full timing analysis of communicated data is also carried out to ensure validity and integrity of the data transmitted. It was found that logic analysis plays a very important role in constructing data frames that are accurate to the smallest time unit possible, such analysis was not possible using an old model oscilloscope, after successful construction, transmitting a predefined sequence of data successfully initiated the communication channel between all system components. With the implemented safety and security features, the system is safe to use, however, this does not make it ready for end users, this is merely a proof of concept that such system can be made smart. The results show that the microcontroller module implemented is very capable of giving the user the ability to control the system manually with attached knobs and wirelessly through the Access point server. The purpose of this thesis is to provide information on the economic ramifications of such workplace health promotion programs in the field of physical activity to allow for data-based investment decisions of firms. Within a cost-benefit evaluation framework, this thesis identified relevant costs and benefits from practically oriented literature. While most outcomes have been found inconclusive, reductions in absenteeism might be appropriately evidenced and sufficiently tangible to allow quantification. Based on population-attributable risk and insurance data, per employee 0.38-0.58 absence days, or 47.17-71.08€ were estimated to be solely attributable to physical inactivity. Oppositely, cost magnitude depends largely on the interventions’ format, cost-effectiveness, and especially the time investment of the participant. Yet, the economic evidence is largely restricted and demands further investigation. Die vorliegende Studie betrachtet daher die mediierende Wirkung der digitalen Selbstwirksamkeit auf die Beziehung zwischen Alter und Arbeitszufriedenheit. Es wurde eine online-Befragung mit 124 Arbeitnehmenden im Alter von 17-63 Jahren durchgeführt. Es wurde erwartet, dass steigendes Alter in einer negativen Beziehung zu Arbeitszufriedenheit und digitaler Selbstwirksamkeit steht. Zusätzlich wurde angenommen, dass digitale Selbstwirksamkeit positiv auf Arbeitszufriedenheit wirkt. Es wurde eine indirekte Mediation, mit einer negativen Wirkung von Alter auf Digitale Selbstwirksamkeit und einer positiven Wirkung von digitaler Selbstwirksamkeit auf Arbeitszufriedenheit identifiziert. Durch die konventionelle Fleischproduktion entstehen hohe Treibhausgasmengen sowie ein hoher Wasser- und Landverbrauch. Der Verzehr von verarbeitetem und rotem Fleisch erhöht das Krebsrisiko und begünstigt die Entstehung von Herz-Kreislauf-Erkrankungen, Diabetes und Bluthochdruck. Bei der Aufzucht der Tiere werden zur Vermeidung von bakteriellen Infektionen Antibiotika eingesetzt, die Auswirkungen auf die menschliche Gesundheit haben können. Das Wohlergehen der Tiere ist aufgrund des hohen Stressniveaus durch schlechte Haltungsbedingungen nicht gegeben. Zur Verbesserung der Risiken durch konventionell hergestelltes Fleisch werden Alternativen erforscht. Ziel ist es die sensorischen und geschmacklichen Komponenten des konventionellen Fleisches nachzuahmen und eine Verbesserung der Gesundheit, Umwelt und das Wohlergehen der Tiere zu erzeugen. Neben den auf dem Markt erhältlichen pflanzlichen Fleischprodukten, deren Sensorik noch nicht ausgereift ist, befasst sich die Forschung mit der Entwicklung von in vitro Fleisch sowie Fleisch aus dem 3D-Drucker. Die Herstellung von in vitro Fleisch nach dem Tissue Engineering Verfahren entstammt der Stammzellenforschung und Gewebezüchtung. Stammzellen eines Tieres werden entnommen und mithilfe eines Nährmediums, einem Gerüst und einem Bioreaktor zu Geweben kultiviert, die dem konventionellem Fleisch ähneln. Durch das Hinzufügen von Komponenten wie z.B. Vitamine oder gesunde Fette kann die Sensorik und der Geschmack des in vitro Fleisches verbessert werden. Der Verbrauch von Land und die Umweltbelastung durch Treibhausgase sind geringer als bei der konventionellen Fleischherstellung. Dennoch stellen sich bei der in vitro Fleischproduktion noch offene Herausforderungen in Bezug auf die Umwelt, Gesundheit und Sensorik. Damit in vitro Fleisch auf dem Markt zugelassen werden kann, muss die Sicherheit des Produktes gewährleistet sein. Die Verbraucherakzeptanz ist eingeschränkt, da wenig Informationen und Studien zu den Auswirkungen auf die Umwelt und Gesundheit vorliegen, die auf bewiesenen Ergebnissen beruhen. Since the pandemic has forced companies to reach out to their customers through digital platforms, the marketing strategies had to be adapted. A mixed method approach is used in this thesis, combining primary sources and data from a empirical study from an online survey, to answer the research question and test four hypothesis that are related to the impact of marketing automation on the customer behavior. The study found out that while there was an increase in personalized advertisements on social media, it is unclear how much customers were influenced. However, the thesis shows that marketing automation can be an effective to for improving the relationship between customers and companies. Furthermore, social media has become very importance for the customers’ purchasing decisions. The findings help to understand the role of marketing automation in the pandemic and provides insights for companies to help adapting their marketing strategies in the future. Future research could explore insights from companies and suggest improved marketing strategies to reach out to potential customers more effectively. Due to the tremendous increase in social media brand communication during the COVID pandemic, businesses need to study the views or the response of consumers to it and understand how to fully leverage it. With the objective to understand the consumer's views toward social media brand communication during the pandemic, influencing factors of social media brand communication were first examined, and then consumers' responses towards those factors were studied. Two research questions were developed to execute the study. The thesis first relies on a literature review to assess the influencing factors of social media brand communication during Covid-19. Then the thesis, in the empirical part, applies the qualitative research method by conducting semi-structured interviews with 10 participants grouped by aged 25-34 and users of both Facebook and Instagram. The interview was conducted via ZOOM, audio recorded, transcribed, and analyzed using MAXQDA. The result showed that the influencing factors of social media brand communication were informativeness, entertainment, credibility, interactivity, and electronic word of mouth. In general situation, the brand’s product information and value-related information were the central focus of the consumers but during the covid pandemic covid related information, CSR, health-related messages in entertaining form, source expertise, high level of brand interaction and word of mouth from the trusted sources were the influencing factors of social media brand communication. Consumers responded favorably to those brands that were able to incorporate all these factors in social media brand communication. Insight from a multi-dimensional characterization of informal baobab enterprises N2 - Knowledge of the structure and characteristics of informal forest enterprises is critical to determining the most appropriate course of action for their development and formalization. This study characterizes and develops a typology of informal microenterprises in the Malawian baobab processing sector; and assesses factors influencing their entry into the informal sector. Drawing on the analysis of firm-level data from 305 informal baobab pro- cessors, cost and complexity of registration, smallness (scale) of the business, and necessity-driven motivations were identified as key factors influencing informality. Coupling principal component and cluster analyses, three typologies of informal microenterprises were identified and characterized namely; high performance-survival phase enterprises (type 1), moderate performance-survival phase enterprises (type 2), and low performance-start-up phase enterprises (type 3). Annual net profit (earnings), firm age, and the extent of adoption of formal systems and controls explained the inherent segmentation. The segmentation suggests a dualism compromising of an upper- and lower-tier enterprises characterized by strong involuntary entry into the informal forest sector. For policy implications, we argue that the upper-tier enterprises (types 1 and 2) be targeted with business development programmes to promote their growth while, the lower-tier (type 3) targeted with skills development and social support. Die qualitative Studie zielt auf die Erhebung von explorativen Ergebnissen zur Erfassung der Arbeitssituation von Berufskraftfahrern ab. Resultierend aus der qualitativen Erhebung erfolgt, in Anlehnung an eine Arbeitssituationsanalyse, ein Konzept um zielgruppenspezifische Lösungsvorschläge abzuleiten. Hierbei dient eine Online-Kurzumfrage als Priorisierungsgrundlage für neun Themenbereiche (N=65). Mithilfe der Kurzumfrage wurde schließlich ein Workshop zum Thema „Belastungen und Lösungsfindung für bessere und gesündere Arbeitsbedingungen“ zu den wichtigsten fünf Themenbereichen durchgeführt (N=8). These by-products however are mostly fibrous in nature and they can hardly be valuable to be used as animal feed. Oil palm frond (OPF) is one of the oil palm’s by-product which are widely available, yet the utilization is hindered due to its high fibre content. In this study, the oil palm fronds, specifically the petiole part of it will be improved to be used as animal feed using solid state fermentation technique at the Faculty of Sustainable Agriculture (FSA) in Sandakan, Sabah by using Pleurotus ostreatus. The study was carried out to produce 30kg of treated OPF and to see if solid state fermentation technique using the mentioned fungus has the effect on the OPF. Cultivated Pleurotus ostreatus that are ready for inoculation was obtained from FSA fungus collection. It was then inoculated at the FSA general laboratory. For the OPF, they were collected from the oil palm farm in Telupid and they were ground into smaller and fine particles. 650 g of ground OPF was then be filled in the mushroom bag. For each mushroom bag, it was inoculated with one plate of Pleurotus ostreatus, and then all the samples were kept in the Mushroom House for fermentation. Samples were harvested after eight weeks. Dry matter, moisture content, organic matter and ash content of OPF, before and after fermentation were then evaluated in the lab. Dry matter and organic matter of treated OPF were found out to be decreasing while its moisture content and ash content were increasing. Solid state fermentation did take place on the OPF and 30 kg of treated OPF with Pleurotus ostreatus was managed to be produced. Therefore, this paper will focus on the impact of monetary policy on exchange rates and foreign trade in Türkiye since 2012. For this purpose, the Mundell-Fleming model is introduced, which is a short-run macroeconomic model that assesses the effects of fiscal and monetary policies under different exchange rate regimes. Aggregated macro data on different indicators, which are representing key variables, is analyzed by using descriptive analysis. The empirical results were interpreted and discussed in line with the underlying model. The main findings of the paper suggest that Turkish monetary policy had a negative impact on exchange rates and thus on trade, especially since 2017 after the change of governmental system. Methods such as cover cropping, the use of mulches and non- or minimal tillage are recommended in large-scale agriculture, yet are not typically found in standard versions of market gardening, as developed by J.M. Fortier. In order to redesign the market gardening system towards more sustainability, the advantages and disadvantages of mulching, possible allelopathic effects from cover crops and mulches, and a possible on-site production of transfer and insitu mulch, were investigated. For this purpose, a literature review was carried out and afterwards a market garden rotation including cover crops was designed. The review results suggested that positive (crop-promoting) as well as negative (crop- and weed-reducing) allelopathy from cover crops exist and are includable into the system as a form of integrated weed management system. It was found that the advantages of mulching outweigh the disadvantages and that it is possible to redesign the market garden system towards more sustainability. Dabei wurden die Hypothesen H1: Personen, welche im privaten Sektor arbeiten, haben eine höhere Ausprägung des Narzissmus als Personen, welche im öffentlichen Sektor tätig sind, H2: Führungskräfte haben eine höhere narzisstische Ausprägung als Mitarbeitende und H3: Je höher die Ausprägung des Narzissmus von Führungskräften, desto höher die Emotionsregulation, überprüft. Die Ergebnisse, welche durch eine Stichprobe von 178 Teilnehmenden, davon 138 Mitarbeitende und 40 Führungskräfte erreicht wurden, zeigten, dass Personen im privaten Sektor eine höhere narzisstische Ausprägung als Personen im öffentlichen Sektor haben und Führungskräfte eine durchschnittlich höhere narzisstische Ausprägung als Mitarbeitende aufweisen. Sowohl Hypothese eins als auch Hypothese zwei wurden somit bestätigt. Several advances have occurred in the field of SLE drug discovery, especially since the publishing of the first Genome Wide-Associated Studies in 2005. More than one hundred loci were found to be involved in the disease. Subsequently, the treatments were aimed towards tackling the pathways that were found after the uses of modern genetic techniques. Historically, the FDA approved only two drugs for treatment until the approval of Belimumab in 2011. Therefore, the need for novel therapeutic strategies is essential. Mesenchymal stem cells (MSCs) have been studied for a long time with encouraging results in the treatment of metastatic cancers, including breast cancer. The discovery of MSCs' therapeutic abilities to inhibit the growth of cancer, cause cell death, and trigger immune responses served as a catalyst for the development of bioengineered MSCs. The use of MSCs in cell therapy is the subject of numerous investigations now. This thesis aimed to summarize the current knowledge on traditional breast cancer treatments and compare them to novel approaches based on MSCs usage. In der vorliegenden Arbeit wird die Forschungsfrage „Sollte Selbstvermessung anhand von Wearables zur Gesundheitsförderung eingesetzt werden?“ aus ethischer Perspektive untersucht. Die Durchführung einer Online-Befragung von Wearable-Nutzenden stellt den praktischen Bezug zur Diskussion von drei verschiedenen philosophischen Ansätzen her. In der Umfrage wurden Wearable-Anwender*innen bezüglich ihres Nutzungsverhaltens und ihrer Einstellung zum Thema Datenschutz und Sicherheitsrisiken befragt. Inwieweit es sich bei der Nutzung des Wearables um Gesundheitsoptimierung handelt, wurde durch indirekte Fragestellungen evaluiert. Aus den Ergebnissen der Online-Befragung geht hervor, dass Wearables für die meisten Anwender*innen ein fester Alltagsbegleiter sind und hauptsächlich für die Motivation zur Bewegung getragen werden. Allerdings messen die Teilnehmenden der Befragung dem Tragen des Wearables im Allgemeinen eher eine mittlere Priorität bei. Currently, no study comparing plant and fish yields cultivated in coupled aquaponics (cAP) and decoupled aquaponics (dAP) could be identified. Meta-analyses were conducted to synthesize data extracted from studies reporting fish and plant performance in cAP and dAP. Three databases were used to identify relevant research papers published in English between 2016 and 2021. The effect size of each study was calculated using the standardized mean difference. The summary effect size and heterogeneity (I2) were computed using a mixed-effects model. However, a meta-regression or a sub-group analysis was not conducted to analyze and describe heterogeneity in greater detail. The results of the meta-analyses indicate that dAP is superior to cAP mainly in fish yields, whereas no significant difference in crop yields between cAP and dAP was observed. However, due to the presence of moderate heterogeneity in some meta-analyses, conclusions derived from the summary effect sizes of such meta-analyses should be carefully interpreted. Therefore, the conclusions should only provide an overall picture of how yields in cAP compare to yields in dAP. Es lassen sich zahlreiche theoretische Studien finden, bei denen die Auswirkungen und die Effektivität narzisstischer Führung in Organisationen untersucht werden. Auf Grundlage einer systematischen Literaturrecherche werden zunächst positive und negative Auswirkungen von Narzissmus auf Führungskräfte zusammengetragen und anschließend relevante Faktoren von kontraproduktivem Verhalten von narzisstischer Führung identifiziert. Die Ergebnisse zeigen, dass narzisstische Führungskräfte zu positiven Eigenschaften wie einer charmanten inspirierenden Art, positiver Selbsteinschätzung, Erfolg in der Karriere und Überzeugungskraft neigen (Back et al., 2010; Brunell et al., 2008; Campbell et al., 2004; Maccoby, 2004; Spurk et al., 2016). Zudem stellen sich negativen Eigenschaften wie einem übermäßigen Selbstvertrauen, einer kritischen Entscheidungsfindung, einem ausgeprägten Kontroll- und Machtbedürfnis, Empfindlichkeit gegenüber Kritik, mangelndem Zuhören sowie einem Mangel an Empathie heraus (Atlas & Them, 2008; Blair et al., 2008; Campbell et al., 2004; Maccoby, 2004; Rosenthal & Pittinsky, 2006). Die Eigenschaften stellen ein hohes Potenzial für kontraproduktives Verhalten im Unternehmen dar. Die Ergebnisse zeigen, dass konkrete Faktoren von CWB durch narzisstische Führung ausgelöst werden. Zu diesen Faktoren zählen unangemessenes verbales und physisches Verhalten in Form von Aggressivität und Manipulation von Mitarbeitenden sowie einzelne Faktoren der Produktionsabweichung, welche die fälschliche Nutzung von Zeit und Ressourcen und mangelhafte Arbeitsqualität beinhalten. Es wird festgestellt, dass narzisstische Führungskräfte durch ihr Verhalten insbesondere CWB in Mitarbeitenden auslösen. Aus den Ergebnissen der vorliegenden Arbeit lassen sich Implikationen für weitere Forschung sowie wichtige Anregungen für die organisationale Praxis ableiten. Numerous theoretical studies can be found that examine the effects and effectiveness of narcissistic leadership in organizations. Based on a systematic literature review, positive and negative effects of narcissism on leaders are first compiled and then relevant factors of counterproductive behavior of narcissistic leadership are identified. The results show that narcissistic leaders tend to have positive traits such as a charming inspiring manner, positive self-evaluation, career success and persuasiveness (Back et al., 2010; Brunell et al., 2008; Campbell et al., 2004; Maccoby, 2004; Spurk et al., 2016). In addition, negative traits such as excessive self-confidence, critical decision-making, a strong need for control and power, sensitivity to criticism, a lack of listening skills, and a lack of empathy emerge (Atlas & Them, 2008; Blair et al., 2008; Campbell et al., 2004; Maccoby, 2004; Rosenthal & Pittinsky, 2006). The characteristics represent a high potential for counterproductive behavior in the company. The results show that specific factors of CWB are triggered by narcissistic leadership. These factors include inappropriate verbal and physical behavior in the form of aggression and manipulation of employees, as well as individual factors of production deviance, which include misuse of time and resources and poor quality work. It is found that narcissistic leaders trigger CWB in employees through their behavior. Implications for further research and important suggestions for organizational practice can be derived from the results of this paper. Einige Jahre später folgten Währungen wie Litecoin, Ripple und Ethereum. Inzwischen sind Kryptowährungen im Großteil der Welt legal und gelten in einigen Ländern bereits als offizielles Zahlungsmittel. Betrachtet man die weitere Entwicklung, stellt sich die Frage, ob Kryptowährungen eine funktionierende Alternative zu herkömmlichen Währungen bieten können, und ob diese unter aktuellen ökologischen Bedingungen tragbar wäre. Ziel ist es, die Frage zu beantworten, welche Kriterien Kryptowährungen erfüllen müssten, um weiterhin einen hohen Stellenwert einzunehmen. Es wird eine Literaturrecherche durchgeführt, welche die Funktionsweise und Problematik verschiedener Mining-Konzepte erklärt, und ökologische Daten analysiert. Dabei zeigt sich, dass es Ansätze zu ökologischen Kryptowährungen gibt, die noch in der Entwicklung sind. Although current vaccines are safe and effective, RNA technology proposes improved vaccines that are potentially able to facilitate eradication of rabies until 2030. In this review, RNA vaccine candidates are evaluated for their potential to be effective against rabies and to be implemented in immunization programs. To understand the relevance of disease prevention, rabies virus and its function in the body is clarified. RNA technology is explained to comprehend its mode of action in the human immune system and its potential to be a vaccine platform. Comparison to existing vaccines showed advantages in several characteristics that are relevant for vaccine approval. RNA vaccines scored better in efficacy, stability, production, and dosing, but there is still room for improvements in terms of safety and tolerability. Theoretical potential is proven, but practical translation of RNA vaccines into clinical candidates requires more attention, as safety and tolerability are the leading limiting factor for approval of any new drug or vaccine. Producing a safe, stable and effective RNA vaccine does not necessarily ensure less rabies deaths, because rabies control strategies heavily rely on proper funding and availability in these rabies endemic region. A framework developed based on the analysis of previous experiences N2 - As climate change continues to have negative effects on agriculture, the European Commission calls for sustainable farming approaches as solutions to challenges associated with climate change and therefore propose living labs as a transformative, transdisciplinary and participatory research mode in order to enforce the resilience of farming systems. Permaculture is considered as an approach for the living lab at the Lower Rhine as it aims at integrating sustainable management practices and ethical principles to provide the framework for a holistic approach that could provide an alternative to current management practices and their negative effects on the region. The thesis at hand aims at establishing a living lab framework that allows involvement of regional stakeholders from a range of sectors in a regional network that is part of a trinity of projects that additionally include an on-site test area and a digital knowledge platform. By performing literature research and interviews with regional primary producers and experts of the field, the thesis proceeds to place the permaculture living lab in the currently available context of living lab categorizations and provides a detailed framework with step-by-step instructions to follow as well as a selection of aspects to keep in mind in order to initiate transformative research on permaculture in the form of a living lab in the Lower Rhine region. Apart from the needs of regional primary producers, existing networks and organizations relevant for the living lab are identified, as well as successfully implemented living labs in Zurich, Lisbon, Bern and New York whose objectives align at least partially with those of the future permaculture living lab that can provide experience and suggestions for the planning process. Y1 - 2023 ER - TY - THES A1 - Walser, Heidy T1 - Personality Traits and Work Engagement among Human Services Workers: The Role of Coworker Support N2 - It has been increasingly important to study the well-being of workers in the human services industry as the society benefits from their care and effort abundantly. Most research on human services workers has been focused on the topic related to mental “un-well being” such as exhaustion and burnout, instead mental “well-being”. However, lately a more positive approach of work engagement was introduced as a form of positive psychology. Since previous studies of work engagement have focused mainly on its environmental factors, this study attempted to explore the construct from another perspective which is the personality differences (extraversion and neuroticism). The main objective is to answer the question of why some individuals are more engaged than others while working in the same conditions. Moreover, the moderating effect of perceived coworker support as a form of social support is also examined in the model. A total of 246 nurses and caregivers working in Germany participated in an online survey. Findings show that extraversion and neuroticism are significant predictors of work engagement which indicates that extraverted workers are more engaged compared to the ones with high level of neuroticism. Coworker emotional support was found to buffer the negative effect of neuroticism on work engagement. However, there is no evidence of a moderating effect from coworker instrumental support on the relationship between personality traits and work engagement. The result of this study can be useful particularly for practitioners who may benefit from designing training programs that foster helping behavior among workers hence improving their work engagement. The purpose of this research is to understand the core factors that significantly influence the adoption of blockchain transparency and traceability in supply chain management. A qualitative research design using in-depth semi-structured interviews as the research instrument was conducted with four selected experts working in organizations (Telos, Minespider, Circular Tree, StaTwig) that provide blockchain technology solutions located in USA, Germany, and India. The findings of this study emphasized on the importance of blockchain technology, its adoption and implementation and the factors regulating its adoption. Moreover, many challenging factors that are marring the adoption of blockchain technology by the organizations were deliberated, even though these are highly advantageous. It was found that blockchain technology filled gaps over the current ERP systems and can be considered as the technology of the future. This study will be helpful for the organizations that are blockchain providers, for the organizations who are being reluctant to adopt this technology, as well as for policy makers across the world as this study provides a sneak peek into the emerging issues that these providers face. Y1 - 2023 ER - TY - THES A1 - Adib, Lisa Marziyeh T1 - The Impact of Urban Green Infrastructure on the Heat Island Effect in the Urban Centre of Kleve N2 - This thesis discusses the impact of urban green infrastructure on the heat island effect in the urban centre of Kleve. Green spaces are pivotal points when it comes to reducing inner-city temperatures in times of the climate crisis. Temperature measurements were taken in three different locations (campus, woods, field) and statistical analysis revealed a significant temperature difference between all three groups. On the campus of Rhine-Waal University an urban heat island could be detected, as during the night the temperatures there were significantly higher than in the two other locations. A tree inventory was conducted in the small forest grove and on the campus. It revealed slightly higher species diversity as well as larger height and diameter at breast height diversity in the small woods compared to the campus. The vegetation on the campus is not sufficient in alleviating the urban heat island effect. However, the vegetation in the woods shows a significant cooling effect on the air temperature. The main limitation of this thesis is that - since an observational method was used in the practical part - other variables than green infrastructure, such as air flow and blue infrastructure, were not measured. Nonetheless, they also affect the air temperature. This means that more research is necessary to define the influencing factors and their exact impacts. Regardless, the results show that denser and larger vegetation can mitigate the UHI compared to faculty trees on pavement, or a grass field. Urban green infrastructure should play a central role in city planning, and, when implemented correctly, can be a huge asset in combatting the climate crisis.
