J. A1 - Ridder, Lars T1 - MS2DeepScore: a novel deep learning similarity measure to compare tandem mass spectra JF - Journal of Cheminformatics N2 - Mass spectrometry data is one of the key sources of information in many workflows in medicine and across the life sciences. Mass fragmentation spectra are generally considered to be characteristic signatures of the chemical compound they originate from, yet the chemical structure itself usually cannot be easily deduced from the spectrum. Often, spectral similarity measures are used as a proxy for structural similarity but this approach is strongly limited by a generally poor correlation between both metrics. Here, we propose MS2DeepScore: a novel Siamese neural network to predict the structural similarity between two chemical structures solely based on their MS/MS fragmentation spectra. Using a cleaned dataset of > 100,000 mass spectra of about 15,000 unique known compounds, we trained MS2DeepScore to predict structural similarity scores for spectrum pairs with high accuracy. In addition, sampling different model varieties through Monte-Carlo Dropout is used to further improve the predictions and assess the model's prediction uncertainty. On 3600 spectra of 500 unseen compounds, MS2DeepScore is able to identify highly-reliable structural matches and to predict Tanimoto scores for pairs of molecules based on their fragment spectra with a root mean squared error of about 0.15. Furthermore, the prediction uncertainty estimate can be used to select a subset of predictions with a root mean squared error of about 0.1. Furthermore, we demonstrate that MS2DeepScore outperforms classical spectral similarity measures in retrieving chemically related compound pairs from large mass spectral datasets, thereby illustrating its potential for spectral library matching. Finally, MS2DeepScore can also be used to create chemically meaningful mass spectral embeddings that could be used to cluster large numbers of spectra. Added to the recently introduced unsupervised Spec2Vec metric, we believe that machine learning-supported mass spectral similarity measures have great potential for a range of metabolomics data processing pipelines. The present field study therefore investigated whether sound pressure level, reverberation time, and soundscape pleasantness can predict factors associated with overall restaurant quality. In total, 142 persons visiting 12 restaurants in Berlin rated relevant acoustical and non-acoustical factors associated with restaurant quality. Simultaneously, the A-weighted sound pressure level (LA,eq,15) was measured, and the reverberation time in the occupied state (T20,occ) was obtained by measurements performed in the unoccupied room and a subsequent calculation of the occupied condition according to DIN 18041. Results from linear mixed-effects models revealed that both the LA,eq,15 and T20,occ had a significant influence on soundscape pleasantness and eventfulness, whereby the effect of T20,occ was meditated by the LA,eq,15. Also, the LA,eq,15 as well as soundscape pleasantness were significant predictors of overall restaurant quality. A comprehensive structural equation model including both acoustical and non-acoustical factors, however, indicates that the effect of soundscape pleasantness on overall restaurant quality is mediated by the restaurant’s atmosphere. Our results support and extend previous findings which suggest that the acoustical design of restaurants involves a trade-off between comfort and liveliness, depending on the desired character of the place. It is our belief that context awareness has the potential to supplement the Internet of Things, when a suitable methodology including supporting tools will ease the development of context-aware applications. We believe that a meta-model based approach can be key to achieving this goal. In this paper, we present our meta-model based methodology, which allows us to define and build application-specific context models and the integration of sensor data without any programming. We describe how that methodology is applied with the implementation of a relatively simple context-aware COVID-safe navigation app. The outcome showed that programmers with no experience in context-awareness were able to understand the concepts easily and were able to effectively use it after receiving a short training. Therefore, context-awareness is able to be implemented within a short amount of time. We conclude that this can also be the case for the development of other context-aware applications, which have the same context-awareness characteristics. We have also identified further optimization potential, which we will discuss at the conclusion of this article. Therefore, we conducted a study based on the Experience Sampling Method to measure the relative contribution of a broad range of potentially relevant acoustic and non‐auditory variables in predicting indoor soundscape evaluations. Here we present the comprehensive dataset for which 105 participants reported temporally (rather) stable trait variables such as noise sensitivity, trait affect, and quality of life. They rated 6.594 situations regarding the soundscape standard dimensions, perceived loudness, and the saliency of its sound components and evaluated situational variables such as state affect, perceived control, activity, and location. To complement these subject‐centered data, we additionally crowdsourced object‐centered data by having participants make binaural measurements of each indoor soundscape at their homes using a low‐(self‐)noise recorder. These recordings were used to compute (psycho‐)acoustical indices such as the energetically averaged loudness level, the A‐weighted energetically averaged equivalent continuous sound pressure level, and the A‐weighted five‐percent exceedance level. This complex hierarchical data can be used to investigate time‐varying non‐auditory influences on sound perception and to develop soundscape indicators based on the binaural recordings to predict soundscape evaluations. These three variants of IL-6, different in the exact initiation of the mature protein (A28, P29, or V30), were expressed in Hansenula polymorpha using the Saccharomyces cerevisiae MFα prepro sequence instead of the homologous pre sequence. All three IL-6 variants were secreted but the processing by the Kex2 protease showed significant differences. V30-IL-6 showed correctly processed material but also a molecule species of higher molecular weight indicating incomplete processing of the MFα pro peptide. P29-IL-6 did not yield any correctly processed IL-6, instead only the unprocessed pro form was found in the culture supernatant. Only A28-IL-6 led to 100% correctly processed material. N-terminal sequencing of this material revealed a start at V30--obviously the first two amino acids (Ala28-Pro29) have been removed by a so far unknown protease. Thus expression of both A28-IL-6 and V30-IL-6 as MFα prepro fusion proteins resulted in the very same mature V30-IL-6, however, the ratio of correctly processed molecules was significantly higher in the case of A28-IL-6. The expression of an MFα prepro-interferon α-2a (IFNα-2a) fusion protein in H. polymorpha leads to about 50% correctly processed molecules and 50% misprocessed forms which contain part of the pro peptide at the N-termini. The insertion of A28 and P29 of IL-6 between the pro peptide and the start of the mature IFNα-2a led to correct processing and elimination of all high molecular weight isoforms observed in earlier experiments. In addition the cell's essential functions are maintained under stress conditions like high salinity, heat or aridity stress. Hansenula polymorpha was engineered to catalyze the transformation of monomeric substrates to 5-hydroxyectoine. Instead of buying music products, like CDs or even downloaded digital music files, today music is predominantly distributed via online music streaming. Therefore, it is mandatory for the music industry as well as researchers to determine which factors are affecting consumers intention to use music streaming services. This study aims to investigate drivers of Behavioral Intention (BI) to use a music streaming service. Particularly, aspects of social preferences in the context of music streaming will be explored. Therefore, a research model is created, based on the Value-based Adoption Model (VAM) (Kim et al., 2007) with an extension to take social factors into account. An online questionnaire with 102 participants was conducted to test the research model and the corresponding hypotheses. In order to evaluate reliable indications, some adjustments needed to be made resulting in a corrected research model. The final independent variables of the corrected model are Perceived Value (PV) (which is composed of Perceived Usefulness (PU) and Perceived Enjoyment (PE)), Perceived Ease of Use (PEU), Perceived Price Value (PPV), Facilitating Conditions (FC), Perceived Fairness (PF), Social Connectivity (SC), and Peer-Effects (PFX). The empirical results show that Perceived Value (PV), Perceived Price Value (PPV), and Perceived Fairness (PF) are determinants of consumers Behavioral Intention (BI) to use a music streaming service. Music streaming services have changed how music is played and perceived, but also how it is managed by individuals. Voice interfaces to such services are becoming increasingly com-mon, for example through voice assistants on mobile and smart devices, and have the poten-tial to further change personal music management by introducing new beneficial features and new challenges. Method. To explore the implications of voice assistants for personal music listening and management we surveyed 248 participants online and in a lab setting to investigate (a) in which situa-tions people use voice assistants to play music, (b) how the situations compare to established activities common during non-voice assistant music listening, and (c) what kinds of com-mands they use. Analysis. We categorised 653 situations of voice assistant use, which reflect differences to non-voice assistant music listening, and established 11 command types, which mostly reflect finding or refinding activities but also indicate keeping and organisation activities. Results. Voice assistants have some benefits for music listening and personal music management, but also a notable lack of support for traditional personal information management activities, like browsing, that are common when managing music. The transport processes in a system involving triple convective phenomena, irregular geometry, and boundary conditions constitute a complex phenomenon. This work aims to explore the mixed thermo-bioconvection of magnetically susceptible fluid containing copper nanoparticles and oxytactic bacteria in a novel W-shaped porous cavity. The buoyant convention is generated due to the isothermal heating at the wavy bottom wall, whereas the mixed convection is induced due to the shearing motion of the top-cooled sliding wall. Furthermore, the bioconvection is induced due to the manifestation of oxytactic bacteria or organisms. The inclined sidewalls are insulated. The geometry is packed with water based Cu nanoparticle mixed porous structure, which is subjected to a magnetizing field acted horizontally. The complex transport equations are transformed into nondimensional forms, which are then computed using the finite volume-based developed code. The coupled triple-convective flow physics are explored for a wide range of involved controlling parameters, which could provide helpful insight to the system designer for its proper operation. The shape of geometry can be considered one of the important parameters to control the heat and mass transport phenomena. In general, the influence of amplitude (δ) is more compared to the waviness number (m) of the undulations. The magnitude of heat (Nu) and mass (Sh) transfer rate for the W-shaped cavity is high compared to conventional square and trapezoidal-shaped cavities. The output of the analysis could be very helpful for the designer for modeling devices operating on nanotechnology-based bioconvection, microbial fuel cells, and others. Although research programmes in psychology and economics have contributed to an improved understanding of music-related behaviour, historically, these disciplines have been unconnected. Recently, however, researchers have begun to bridge this gap by employing tools from behavioural economics. This article contributes to the literature by providing a discussion about the benefits of using behavioural economics in music-decision research. We achieve this in two ways. First, through a systematic review, we identify the current state of the literature within four key areas of behavioural economics-heuristics and biases, social decision-making, behavioural time preferences, and dual-process theory. This results in an increasing demand for mobility solutions while it is also worldwide aimed to reduce greenhouse gas emissions. This paper summarizes the results of a comparative study concerning the greenhouse gas emissions (based on carbon footprint) caused by alternative urban passenger transportation systems. The emissions for the vehicles and their infrastructure are analyszed over the entire life cycle from manufacturing up to their end of life. An existing cable car sytem in La Paz, Bolivia was analysed and evaluated in comparison to other modes of transportation such as small busses, large busses and a tram. According to the system definitions and the considered balance framework the study shows that beside the use of the systems the materials and the manufacturing as well as the infrastructure have a significant impact on the total emissions over the life cycle. To put focus on the „true and real“ impacts to the society it is preferable to consider the measurement results in total absolute emissions. The expansion of decentralized energy resources and the integration of new consumers due to sector coupling (e.g. heat pumps or electric vehicles) into low voltage grids increases the need for grid expansion and usage of flexibilities in the grid. A high observability of the current grid status is needed to perform these tasks efficiently and effectively. Therefore, there is a need to increase the observability of low voltage grids by installing measurement technologies (e.g. smart meters). Multiple different measurement technologies are available for low voltage grids which can vary in their benefit to observation quality and their installation costs. Therefore, Bayernwerk Netz GmbH and E.DIS AG in cooperation with E-Bridge Consulting GmbH and the Institute for High Voltage Equipment and Grids, Digitalization and Energy Economics (IAEW) investigated the effectiveness of different strategies for the smartification of low voltage grids. T1 - Resolution and accuracy of non-linear regression of PSF with artificial neural networks T2 - SPIE Optical Systems Design: Optical Instrument Science, Technology, and Applications, 2018, Frankfurt, Germany N2 - In a previous work we have demonstrated a novel numerical model for the point spread function (PSF) of an optical system that can efficiently model both experimental measurements and lens design simulations of the PSF. The novelty lies in the portability and the parameterization of this model, which allows for completely new ways to validate optical systems, which is especially interesting for mass production optics like in the automotive industry, but also for ophtalmology. The numerical basis for this model is a non-linear regression of the PSF with an artificial neural network (ANN). In this work we examine two important aspects of this model: the spatial resolution and the accuracy of the model. Measurement and simulation of a PSF can have a much higher resolution then the typical pixel size used in current camera sensors, especially those for the automotive industry. We discuss the influence this has on on the topology of the ANN and the final application where the modeled PSF is actually used. Another important influence on the accuracy of the trained ANN is the error metric which is used during training. The PSF is a distinctly non-linear function, which varies strongly over field and defocus, but nonetheless exhibits strong symmetries and spatial relations. Therefore we examine different distance and similarity measures and discuss its influence on the modeling performance of the ANN. N2 - Through constant technical progress, multi-user virtual reality is transforming towards a social activity that is no longer only used by remote users, but also in large-scale location-based experiences. We evaluate the usage of realtime-tracked avatars in co-located business-oriented applications in a "guide-user-scenario" in comparison to audio only instructions. The present study examined the effect of an avatar-guide on the user-related factors of Spatial Presence, Social Presence, User Experience and Task Load in order to propose design guidelines for co-located collaborative immersive virtual environments. Therefore, an application was developed and a user study with 40 participants was conducted in order to compare both guiding techniques of a realtime-tracked avatar guide and a non-visualised guide with otherwise constant conditions. Results reveal that the avatar-guide enhanced and stimulated communicative processes while facilitating interaction possibilities and creating a higher sense of mental immersion for users. Furthermore, the avatar-guide appeared to make the storyline more engaging and exciting while helping users adapt to the medium of virtual reality. Even though no assertion could be made concerning the Task Load factor, the avatar-guide achieved a higher subjective value on User Experience. Due to the results, avatars can be considered valuable social elements in the design of future co-located collaborative virtual environments. Collecting and labeling these drive scenes is a very time-consuming and expensive process. Currently, it is not possible to reuse these drive scenes with different optical properties, because there exists no numerically efficient model for the transfer function of the optical system. We present a numerical model for the point spread function (PSF) of an optical system that can efficiently model both experimental measurements and lens design simulations of the PSF. The numerical basis for this model is a nonlinear regression of the PSF with an artificial neural network. The novelty lies in the portability and the parameterization of this model. We present a lens measurement series, yielding a numerical function for the PSF that depends only on the parameters defocus, field, and azimuth. By convolving existing images and videos with this PSF, we generate images as if seen through the measured lens. The methodology applies to any optical scenario, but we focus on the context of autonomous driving, where the quality of the detection algorithms depends directly on the optical quality of the used camera system. With this model, it is possible to reuse existing recordings, with the potential to avoid millions of test drive miles. The parameterization of the optical model allows for a method to validate the functional and safety limits of camera-based advanced driver assistance systems based on the real, measured lens actually used in the product. The numerical basis for this model is a nonlinear regression of the PSF with an artificial neural network (ANN). After briefly describing both the principle and the applications of the model, we then discuss two optically important aspects: the spatial resolution and the accuracy of the model. Using mean squared error (MSE) as a metric, we vary the topology of the neural network, both in the number of neurons and in the number of hidden layers. Measurement and simulation of a PSF can have a much higher spatial resolution than the typical pixel size used in current camera sensors. We discuss the influence this has on the topology of the ANN. The relative accuracy of the averaged pixel MSE is below 10  −  4, thus giving confidence that the regression does indeed model the measurement data with good accuracy. Traditional archers aim without a target figure. Good shooting results require an excellent body-eye coordination that allows the user to perform identical movements when drawing the bow. Our simulator provides a virtual archery experience and supports the user to learn and practice the motion sequence of traditional archery in a virtual environment. We use an infrared tracking system to capture the user’s movements in order to correct his movement. To provide a realistic haptic feedback a real bow is used as interaction device. Our system provides a believable user experience and supports the user to learn how to shoot in the traditional way. Following a user-centered iterative design approach we developed a number of prototypes and evaluated them for refinement in sequent iteration cycles.
