Daher bietet sich eine kamerabasierte Interaktionserkennung an. Viele existierende Verfahren benötigen dazu aber einen weitestgehend statischen Hintergrund. In der gegebenen Anwendung allerdings befindet sich im Sichtbereich der Kameras eine Projektion bewegter Inhalte, mit denen ein Benutzer interagieren kann. Im Rahmen dieser Arbeit sollte ein bestehendes, auf Infrarotlicht basierendes System verbessert werden, das sich bisher als beleuchtungsabhängig erwiesen hat. Dazu wurden zunächst mehrere Verfahren zur Trennung von Vorder- und Hintergrund auf ihre Tauglichkeit untersucht. Das favorisierte Verfahren sollte anschließend durch stereoskopische Bildaufnahme - und damit tiefenbasierte Trennung - so verbessert werden, dass der bewegte Hintergrund sicher unterdrückt wird. Dies erwies sich für die gewünschten Anwendungen zwar als nicht praktikabel, dennoch wurden mögliche andere Anwendungsbereiche gefunden. Auch die Untersuchung der einzelnen Trennungsverfahren lieferte ein Ergebnis, das - mit weiteren Tests - zur Verbesserung der bestehenden Installation beitragen kann. N2 - Modern interactive environments are tending to not impose any restrictions by additional encumbrance on the user. Therefore it seems reasonable to utilize computer vision for detecting interactions. There are many techniques available to segment an image into fore- and background, but most of them require the scene to be static. In contrast this work applies to installations where an interactive projection is in the field of vision of a camera. Within the bounds of this work an existing technique utilizing infra-red light, which has been shown to be illumination-variant, was to be improved. First several techniques for foreground-background-segmentation were checked for suitability. The most appropriate was to be refined by adding depth segmentation to suppress the animated background utilizing stereo-vision. This turned out to be impractical for the given installations, but more practicable ones were found. In addition to this even the favoured segmentationtechnique may provide an improvement on the existing one. N2 - The author presents a novel approach to add an additional degree of realism to a computer generated scene by using a sequence of images (called video texture) to light up other objects in real time. He does this by enhancing the traditional fixed shading model using the video textures as part of the light properties. An exemplary application showcases this approach by using a filmed landscape (video texture) which affects the lighting of the interior of a computer generated train (3D polygonal geometry). Combining filmed material and computer generated images has long been standard practice in special effects departments but it was not until recently that the average graphics hardware is becoming capable to produce these effects in realtime. Das künstliches neuronales Netz (KNN) soll mit dem LKR NRW vorliegenden Daten zu manuellen Entscheidungen trainiert werden. Dafür müssen die Daten vor der Verwendung umfangreich aufbereitet werden. Das Ziel der Arbeit ist die Aufarbeitung und Aktualisierung eines vorliegenden Konzepts und die Implementierung eines Prototypen des Verfahrens. Der Prototyp soll in der Programmiersprache Java umgesetzt werden. Die folgende Arbeit beginnt mit einer kurzen Einleitung über die Krankheit Krebs, das LKR NRW und KNN. Danach erläutere ich die Methodik, die sich mit und die Implementierung des Themas. In der Methodik erkläre ich was KNN sind und schneide ihre Funktionsweise an. Außerdem erkläre ich wie die Bereitstellung der Daten erfolgt und wie diese für das neuronale Netz verständlich aufbereitet werden. Zur Implementierung werde ich beschreiben was für eine Entwicklungsumgebung verwendet wurde um das Projekt umzusetzen und wie das Programm umgesetzt wurde. Die Erklärung der Umsetzung besteht aus Unified Modeling Language (UML)-Diagrammen für die Datenaufbereitung und Code-Ausschnitte für die Konfiguration und Schnittstellen des KNN. Das KNN konnte inklusive Datenaufbereitung erstellt werden und klassifiziert 94,4% der Fälle korrekt. Auf Grundlage dieser Arbeit kann das Projekt nachgebaut werden. All participants performed a ball-throwing task developed by Decharms and Davé (1965), consisting of two parts: First, participants threw the ball from fixed distances into a funnel basket. During this task, performance was measured. In the second part, the participants themselves chose distances from the basket, which allowed their risk-taking behavior to be assessed. The results indicate that listening to motivational music led to greater risk taking but did not improve ball-throwing performance. This effect was more pronounced in male participants and among those who listened to their own playlists. Furthermore, self-selected music enhanced state self-esteem in participants who were performing well but not in those who were performing poorly. Until now, little has been written about the processes underlying the selection of music in daily life. The present study aimed to disentangle some of the complex processes among the listener, situation, and functions of music listening involved in music selection. Utilizing the experience sampling method, data were collected from 119 participants using a smartphone application. For 10 consecutive days, participants received 14 prompts using stratified-random sampling throughout the day and reported on their music-listening behavior. Statistical learning procedures on multilevel regression models and multilevel structural equation modeling were used to determine the most important predictors and analyze mediation processes between person, situation, functions of listening, and music selection. Results revealed that the features of music selected in daily life were predominantly determined by situational characteristics, whereas consistent individual differences were of minor importance. Functions of music listening were found to act as a mediator between characteristics of the situation and music-selection behavior. We further observed several significant random effects, which indicated that individuals differed in how situational variables affected their music selection behavior. Our findings suggest a need to shift the focus of music-listening research from individual differences to situational influences, including potential person-situation interactions. This includes both the creative design process and the development of suitable methods for the evaluation of target sounds by trial participants. The main difference between e-car sound surveys and those carried out on familiar sound categories is the potential consumer’s lack of experience with electric vehicles. Thus the consumer has no, or very unspecific, expectations in this regard. Several studies have consequently shown that many subjects have to construct their personal frame of reference for evaluation within the listening experiment. However, this is possibly at odds with experience-based expectations relating to the sounds of conventional combustion engines. The result is a conflict of objectives between the traditional and the modern, familiarity and strangeness and not least between driving freedom and ecological awareness. In this context, the authenticity of the sound and the subjective interpretability of the sound information also appear as moderator variables. Moreover, associations with other vehicle categories, for example streetcars, also influence the perceived sound quality. Hinzu kommt die fehlende Erklärbarkeit der Algorithmen. Je nach Modell ist nicht eindeutig, nach welchen Prinzipien eine Entscheidung getroffen wird. Das erschwert das Aufdecken von Diskriminierung und senkt die Vertrauenswürdigkeit des Systems. In dieser Arbeit werden Ursachen für Diskriminierung und fehlende Transparenz, sowie Lösungsmethoden erarbeitet. Dabei wird erörtert, inwieweit die beiden Themen aufeinander einspielen und gemeinsam zur Qualität der Software beitragen. Advanced technologies and approaches are creating new areas of use for applications that study and imitate human behavior. At the same time, this innovative approach poses new dangers. Human behavior is not always objective and correct. Prejudice often dominates human decisions and therefore leads to discrimination. There is a risk that machines adopt this behavior. On top of this, algorithms are often not explainable. Depending on the model, the principles that are relevant for the decision are not clear. This makes it harder to detect discrimination and reduces the trustworthiness of a system. In this paper, causes of bias in Machine Learning and the lack of transparency, as well as methods to eliminate these problems, are elaborated. Hence the claim for ecological validity of such experiments is reasonable, but too vague in its bare formulation. The estimation of ecological validity requires theoretical knowledge about the meaning and the totality of factors influencing everyday listening and their interaction. Therefore the influence of “reality factors” on the sound evaluation of household appliances was investigated within the framework of nine experiments. These tests were performed in four different settings with varied reality degrees. The psychological realism and the perceived face validity of the tests were examined by means of interviews. The results reveal, on the one hand, that stimuli are perceived as more pleasant in everyday-like environments. The analysis of the relative judgments shows, on the other hand, that the influence of the experimental realism is moderated by a number of factors. These are the saliency and meaning of the sounds, differences between the assessed devices, and personality traits. The correlation and regression analysis confirms that the perceived sound character is widely independent from experimental realism. However, there is great empirical evidence, that valid assessments of sound quality require the reconstruction of everyday attention and action processes. Being a very complex system far from thermodynamic equilibrium, the cytoskeleton's ability to organize is at the same time challenging and fascinating. The extensive amounts of frequently interacting cellular building blocks and their inherent multifunctionality permits highly adaptive behavior and obstructs a purely reductionist approach. Nevertheless (and despite the field's relative novelty), the physics approach has already proved to be extremely successful in revealing very fundamental concepts of cytoskeleton organization and behavior. This review aims at introducing the physics of the cytoskeleton ranging from single biopolymer filaments to multicellular organisms. Throughout this wide range of phenomena, the focus is set on the intertwined nature of the different physical scales (levels of complexity) that give rise to numerous emergent properties by means of self-organization or self-assembly. T1 - Spec2Vec: Improved mass spectral similarity scoring through learning of structural relationships JF - PLOS Computational Biology N2 - Spectral similarity is used as a proxy for structural similarity in many tandem mass spectrometry (MS/MS) based metabolomics analyses such as library matching and molecular networking. Although weaknesses in the relationship between spectral similarity scores and the true structural similarities have been described, little development of alternative scores has been undertaken. Here, we introduce Spec2Vec, a novel spectral similarity score inspired by a natural language processing algorithm-Word2Vec. Spec2Vec learns fragmental relationships within a large set of spectral data to derive abstract spectral embeddings that can be used to assess spectral similarities. Using data derived from GNPS MS/MS libraries including spectra for nearly 13,000 unique molecules, we show how Spec2Vec scores correlate better with structural similarity than cosine-based scores. We demonstrate the advantages of Spec2Vec in library matching and molecular networking. Spec2Vec is computationally more scalable allowing structural analogue searches in large databases within seconds. The actin bundle network is conductive to travelling excitations, i.e. impulses. The machine is interfaced with an arbitrary selected set of k electrodes through which stimuli, binary strings of length k represented by impulses generated on the electrodes, are applied and responses are recorded. The responses are recorded in a form of impulses and then converted to binary strings. The machine's state is a binary string of length k: if there is an impulse recorded on the ith electrode, there is a '1' in the ith position of the string, and '0' otherwise. We present a design of the machine and analyse its state transition graphs. We envisage that actin droplet machines could form an elementary processor of future massive parallel computers made from biopolymers. Recently, it became evident that this extension consists of two distinct substructures, designated lamellipodium and lamellum, which differ significantly in their kinetic and kinematic properties as well as their biochemical composition. We developed a stochastic two-dimensional computer simulation that includes chemical reaction kinetics, G-actin diffusion, and filament transport to investigate the formation of growing actin networks in migrating cells. Model parameters were chosen based on experimental data or theoretical considerations. In this work, we demonstrate the system's ability to form two distinct networks by self-organization. We found a characteristic transition in mean filament length as well as a distinct maximum in depolymerization flux, both within the first 1-2 microm. The separation into two distinct substructures was found to be extremely robust with respect to initial conditions and variation of model parameters. We quantitatively investigated the complex interplay between ADF/cofilin and tropomyosin and propose a plausible mechanism that leads to spatial separation of, respectively, ADF/cofilin- or tropomyosin-dominated compartments. Tropomyosin was found to play an important role in stabilizing the lamellar actin network. Furthermore, the influence of filament severing and annealing on the network properties is explored, and simulation data are compared to existing experimental data. In contrast to classical man-made machines, much of the cellular organization originates from inherent self-assembly and self-organization allowing a high degree of autonomy for various functional units. Recent experimental and theoretical studies revealed numerous examples of cytoskeleton components that arrange and organize in a self-regulative way. In the present review we want to shortly summarize some of the principle mechanisms that are able to inherently trigger and regulate the cytoskeleton organization. Although taken individually most of these regulative principles are rather simple with intuitively predictable consequences, combinations of two or more of these mechanisms can quickly give rise to very complex, unexpected behavior and might even be able to explain the formation of different functional units out of a common pool of available building blocks. While the formation of the lamellipodium is increasingly well understood, organizational principles underlying the emergence of the lamellum are just beginning to be unraveled. We report here on a 1D mathematical model which describes the reaction-diffusion processes of a polarized actin network in steady state, and reproduces essential characteristics of the lamellipodium-lamellum system. We observe a steep gradient in filament lengths at the protruding edge, a local depolymerization maximum a few microns behind the edge, as well as a differential dominance of the network destabilizer ADF/cofilin and the stabilizer tropomyosin. We identify simple and robust organizational principles giving rise to the derived network characteristics, uncoupled from the specifics of any molecular implementation, and thus plausibly valid across cell types. An analysis of network length dependence on physico-chemical system parameters implies that to limit array treadmilling to cellular dimensions, network growth has to be truncated by mechanisms other than aging-induced depolymerization, e.g., by myosin-associated network dissociation at the transition to the cell body. Our work contributes to the analytical understanding of the cytoskeletal extension's bisection into lamellipodium and lamellum and sheds light on how cells organize their molecular machinery to achieve motility. Although it is known that cytoskeletal coordination is vital for cell function, the mechanisms by which cross-linking proteins coordinate actin and microtubule activities remain poorly understood. In particular, it is unknown how the distinct mechanical properties of different actin architectures modulate the outcome of actin-microtubule interactions. To address this question, we engineered the protein TipAct, which links growing microtubule ends via end-binding proteins to actin filaments. We show that growing microtubules can be captured and guided by stiff actin bundles, leading to global actin-microtubule alignment. Conversely, growing microtubule ends can transport, stretch and bundle individual actin filaments, thereby globally defining actin filament organization. Our results provide a physical basis to understand actin-microtubule cross-talk, and reveal that a simple cross-linker can enable a mechanical feedback between actin and microtubule organization that is relevant to diverse biological contexts. Dazu wurde ein Vergleich auf Basis ausgewählter Vergleichskategorien durchgeführt. Zur Eingrenzung des Umfangs beschränkte sich der Vergleich auf native Android Apps. Die beiden App-Typen wurden im Rahmen einer Literaturrecherche und auf Basis ausgewählter Vergleichskategorien untersucht und verglichen. Zudem wurde ein Prototyp einer Progressive Web App implementiert, um die Plattformunterstützung und Funktionalität zu überprüfen. Außerdem wurde auch ein Prototyp einer nativen Android App implementiert. Dabei wurde nur die Benutzeroberfläche umgesetzt, um das Look and Feel vergleichbar zu machen. Die Untersuchung und der Vergleich haben gezeigt, dass Progressive Web Apps, anders als native Apps, mit Webtechnologien entwickelt und keine plattformspezifischen Programmiersprachen benötigt werden. Sie werden im Browser ausgeführt und sind dadurch Plattformunabhängig. Zudem wurde gezeigt, dass Progressive Web Apps über Funktionen und Eigenschaften verfügen, die bisher nur nativen Apps vorbehalten waren. Dennoch können sie noch nicht auf alle Funktionen und Komponenten eines mobilen Gerätes zugreifen. Darüber hinaus verfügen sie aber auch über Funktionen und Eigenschaften, die native Apps nicht bieten. Des Weiteren wurde mithilfe eines Prototyps die Plattformunterstützung von Progressive Web Apps überprüft. Die Überprüfung zeigte, dass die Unterstützung durch das Betriebssystem Android weit fortgeschritten und bei iOS hingegen nur sehr eingeschränkt ist. Dadurch variiert der Funktionsumfang je nach Browser und Betriebssystem. Die Untersuchung und der Vergleich der Prototypen zeigten, dass Progressive Web Apps aufgrund ihrer Eigenschaften, Funktionen und die app-ähnliche Benutzeroberfläche, ein ähnliches Look and Feel bieten können, wie native Apps. Außerdem wurde gezeigt, dass Progressive Web Apps, anders als native Apps, i. d. R. nicht über App Stores angeboten werden. Sie durchlaufen dadurch keinen Überprüfungs- und Freigabeprozess und sind nicht an die Richtlinien der App Stores gebunden. Stattdessen können sie über eine URL und über Suchmaschinen aufgerufen und aufgefunden werden. Die durchgeführte Untersuchung und der Vergleich haben gezeigt, dass Progressive Web Apps aufgrund des eingeschränkten Zugriffs auf Geräte- und Software-Funktionen sowie fehlender Browser- und Betriebssystemunterstützung, zum jetzigen Zeitpunkt native Apps noch nicht vollumfänglich ersetzen können. Continuous Delivery bietet in der agilen Softwareentwicklung mit kurzen Entwicklungszyklen die Antwort auf diesen Wandel. Die Verwendung einer Continuous Delivery-Umgebung (CD-Umgebung) erfordert einerseits zwar einen initialen Einrichtungsaufwand. Andererseits kann durch den Einsatz einer CD-Umgebung das gesamte Projekt an Schnelligkeit gewinnen. In Kombination mit Microservice-Architekturen lassen sich kleine Deployment-Einheiten umsetzen. Microservice-Architekturen bringen einen Mehrwert durch Flexibilität, Skalierung, Automatisierung, standardisierte Schnittstellen und Ausfallsicherheit. Microservices fungieren autark in verteilten Systemen und weisen nur sehr geringe Abhängigkeiten zu anderen Services auf. Die Kommunikation findet über einheitliche Schnittstellen statt, weshalb Implementierungsdetails der Microservices nach außen betrachtet irrelevant sind. Für die Umsetzung der CD-Umgebung wird der web-basierte Git-Repository-Manager GitLab in Kombination mit der Container-Orchestrierungsplattform Kubernetes verwendet, da sich beide Programme einfach integrieren lassen. Der qualitative Teil enthält in der Regel eine offene Frage mit der positiv formulierten Bitte um Verbesserungsvorschläge. Ground truth tracking systems require markers to create high quality motion data. But in many applications it is desired to work without markers. In recent years affordable hardware for markerless tracking systems was made available at a consumer level. Efficient depth camera systems based on Time-of-Flight sensors and structured light systems have made it possible to record motion data in real time. However, the gap between the quality of marker-based and markerless systems is high. The error sources of a markerless motion tracking pipeline are discussed and a model-based filter is proposed, which adapts depending on spatial location. The proposed method is then proven to be more robust and accurate than the unfiltered data stream and can be used to visually enhance the presence of an actor within a virtual environment in live broadcast productions. Mass fragmentation spectra are generally considered to be characteristic signatures of the chemical compound they originate from, yet the chemical structure itself usually cannot be easily deduced from the spectrum. Often, spectral similarity measures are used as a proxy for structural similarity but this approach is strongly limited by a generally poor correlation between both metrics. Here, we propose MS2DeepScore: a novel Siamese neural network to predict the structural similarity between two chemical structures solely based on their MS/MS fragmentation spectra. Using a cleaned dataset of > 100,000 mass spectra of about 15,000 unique known compounds, we trained MS2DeepScore to predict structural similarity scores for spectrum pairs with high accuracy. In addition, sampling different model varieties through Monte-Carlo Dropout is used to further improve the predictions and assess the model's prediction uncertainty. On 3600 spectra of 500 unseen compounds, MS2DeepScore is able to identify highly-reliable structural matches and to predict Tanimoto scores for pairs of molecules based on their fragment spectra with a root mean squared error of about 0.15. Furthermore, the prediction uncertainty estimate can be used to select a subset of predictions with a root mean squared error of about 0.1. Furthermore, we demonstrate that MS2DeepScore outperforms classical spectral similarity measures in retrieving chemically related compound pairs from large mass spectral datasets, thereby illustrating its potential for spectral library matching. Finally, MS2DeepScore can also be used to create chemically meaningful mass spectral embeddings that could be used to cluster large numbers of spectra. Added to the recently introduced unsupervised Spec2Vec metric, we believe that machine learning-supported mass spectral similarity measures have great potential for a range of metabolomics data processing pipelines. Although research programmes in psychology and economics have contributed to an improved understanding of music-related behaviour, historically, these disciplines have been unconnected. Recently, however, researchers have begun to bridge this gap by employing tools from behavioural economics. This article contributes to the literature by providing a discussion about the benefits of using behavioural economics in music-decision research. We achieve this in two ways. First, through a systematic review, we identify the current state of the literature within four key areas of behavioural economics-heuristics and biases, social decision-making, behavioural time preferences, and dual-process theory. Second, taking findings of the literature as a starting point, we demonstrate how behavioural economics can inform future research. Based on this, we propose the Behavioural Economics of Music (BEM), an integrated research programme that aims to break new ground by stimulating interdisciplinary research in the intersection between music, psychology, and economics. Usage of realtime-tracked avatars in co-located business-oriented applications with a ”guide-user-scenario” is examined for user-related factors of Spatial Presence, Social Presence, User Experience and Task Load. A user study was conducted in order to compare both techniques of a realtime-tracked avatar and a non-visualised guide. Results reveal that the avatar-guide enhanced and stimulated communicative processes while facilitating interaction possibilities and creating a higher sense of mental immersion for users and engagement. N2 - In the EILD.nrw project, Open Educational Resources (OER) are being developed for teaching databases. Instructors can use the tools and courses in a variety of learning scenarios. Students of computer science and application subjects learn the complete life cycle of databases. For this purpose, quizzes, interactive tools, instructional videos, and courses for learning management systems were developed and published under a Creative Commons license.
