An immunohistochemical method is described to visualize the distribution of metallothioneins 1/2 (MT 1/2) and metallothionein 3 (MT 3) in human ocular tissue. It is making use of (a) antibodies conjugated to gold nanoclusters (AuNCs) acting as labels, and (b) laser ablation (LA) coupled to inductively coupled plasma – mass spectrometry (ICP-MS).Water-soluble fluorescent AuNCs (with an average size of 2.7 nm) were synthesized and then conjugated to antibody by carbodiimide coupling. The surface of the modified AuNCs was then blocked with hydroxylamine to avoid nonspecific interactions with biological tissue. Immunoassays for MT 1/2 and MT 3 in ocular tissue sections (5 μm thick) from two post mortem human donors were performed. Imaging studies were then performed by fluorescence using confocal microscopy, and LA-ICP-MS was performed in the retina to measure the signal for gold. Signal amplification by the >500 gold atoms in each nanocluster allowed the antigens (MT 1/2 and MT 3) to be imaged by LA-ICP-MS using a laser spot size as small as 4 μm. The image patterns found in retina are in good agreement with those obtained by conventional fluorescence immunohistochemistry which was used as an established reference method.
Die Vermeidung von Heißrissen beim Schweißen ist für die Sicherheit geschweißter Komponenten unerlässlich. Die Entstehung von Heißrissen wird durch die komplexe Interaktion von Schweißprozess, Metallurgie und der konstruktiven Umgebung beschrieben. Bislang wurden mehr als 140 Heißrisstests entwickelt, um die Neigung gegenüber einer Rissbildung festzulegen. Grundsätzlich werden diese in selbstbeanspruchende und fremdbeanspruchte Heißrisstests eingeteilt. Der Vortrag gibt einen Überblick über Forschungsthemen der BAM, welche sich in den letzten Jahren hauptsächlich mit den metallurgischen als auch mit den konstruktiven Einflüssen auf die Rissbildung beschäftigt haben. Von austenitischen und ferritischen Werkstoffen, über neuentwickelte LTT-Legierungen bis hin zur in-situ Radiographie zur Überwachung des Risswachstums während des Schweißens wird ein breites Forschungsspektrum aufgezeigt. Zum Abschluss ist ein Ausblick für zukünftige Forschungsfelder gegeben. Diese Arbeiten konzentrieren sich auf die zwei- und dreidimensionale bildliche Charakterisierung von (Mikro-)Heißrissen in innovativen Werkstoffen zur Erweiterung bestehender Modellvorstellungen zum Rissphänomen.
The knowledge of the temperature-induced changes of the superconductor volume and of the thermomechanical behavior of the different coil and tooling materials is required for predicting the coil geometry and the stress distribution in the coil after the Nb3Sn reaction heat treatment. In this paper, we have measured the Young’s and shear moduli of the HL-LHC 11 T Nb3Sn dipole magnet coil and reaction tool constituents during in situ heat cycles with the dynamic resonance method. The thermal expansion behaviors of the coil components and of a free standing Nb3Sn wire were compared based on dilation experiments.
The aim of this study was to identify specific chemical bonds and characteristic structures in melanoidins formed from D-glucose and L-alanine between 130 and 200 °C. The results might be used to control the type and amount of melanoidin produced during food processing. For this purpose, complementary techniques, such as FTIR, NMR, EPR, and MALDI-ToF, were employed. At 160 °C color, solubility and UV/Vis absorption change characteristically and consequently, structural transformations could be observed in FTIR and NMR spectra. For example, sharp signals of N-H, C-N, and C-H oscillations in the L-alanine spectrum are prone to inhomogeneous broadening in melanoidins prepared above 150 °C. These changes are caused due to formation of heterogeneous macromolecular structures and occur during condensation reactions that lead to an increasing loss of water from the melanoidins with increasing temperatures. Additionally, MALDI-ToF-MS indicates the polymerization of glyoxal/glyoxylic acid and EPR shows the formation of radical structures.
Powder based Additive Manufacturing (AM) processes are widely used for metallic and polymeric materials, but rarely commercially used for ceramic materials, especially for technical ceramics. This seemingly contradicting observation is explained by the fact that in powder based AM, a dry flowable powder needs to be used. Technical ceramics powders are in fact typically very fine and poorly flowable, which makes them not suitable for AM. The layerwise slurry deposition (LSD) is an innovative process for the deposition of powder layers with a high packing density for powder based AM. In the LSD process, a ceramic slurry is deposited to form thin powder layers, rather than using a dry powder This allows the use of fine powders and achieves high packing density (55-60%) in the layers after drying. When coupled with a printing head or with a laser source, the LSD enables novel AM technologies which are similar to *Denotes Presenter 42nd International Conference & Exposition on Advanced Ceramics & Composites 127 Abstracts the 3D printing or selective laser sintering, but taking advantage of having a highly dense powder bed. The LSD -3D printing, in particular, offers the potential of producing large (> 100 mm) and high quality ceramic parts, with microstructure and properties similar to traditional processing. This presentation will give an overview of the milestones in the development of this technology, with focus on the latest results applied both to silicate and to technical ceramics.
Studies on the development of antibodies for the highly hydrophobic plasticizers DINCH and DEHT

Diisononylcyclohexane-1,2-dicarboxylate (DINCH) and di-2-ethylhexyl terephthalate (DEHT), two of the most important substitutes for phthalate plasticizers, are used for a wide range of applications. Consequently, an increasing occurrence in urine and environmental samples is reported. Reliable and fast analytical methods for the quantification of these plasticizers are needed. So far, mainly GC-MS or LC-MS methods are used. We aimed to develop the first antibodies and immunoassays allowing for high-throughput analysis of samples. We designed two DINCH hapten structures and one DEHT hapten structure and employed hapten-protein conjugates for the immunization of rabbits. Sensitive competitive enzyme-linked immunosorbent assays (ELISAs) against each hapten using the produced polyclonal antibodies were established. Yet, binding of DINCH to the respective antibodies was not observed in neither direct nor indirect assay formats, even when using protein conjugates with the heterologous haptens and different carrier proteins in the indirect format. The use of surfactants and solvents in the sample buffer did not result in recognition of the plasticizers. Also, no binding of DEHT in ELISA employing the respective antibodies was detected. We speculate that the production of antibodies against these highly hydrophobic molecules is not possible via our route, however a different hapten design could overcome this obstacle.
A key limitation of the most constitutive models that reproduce a Degradation of quasi-brittle materials is that they generally do not address issues related to fatigue. One reason is the huge computational costs to resolve each load cycle on the structural level. The goal of this paper is the development of a temporal Integration scheme, which significantly increases the computational efficiency of the finite element method in comparison to conventional temporal integrations.
The essential constituent of the fatigue model is an implicit gradient-enhanced formulation of the damage rate. The evolution of the field variables is computed as amultiscale Fourier series in time.On a microchronological scale attributed to single cycles, the initial boundary value problem is approximated by linear BVPs with respect to the Fourier coefficients. Using the adaptive cycle jump concept, the obtained damage rates are transferred to a coarsermacrochronological scale associated with the duration of material deterioration. The performance of the developedmethod is hence improved due to an efficient numerical treatment of the microchronological problem in combination with the cycle jump technique on the macrochronological scale. Validation examples demonstrate the convergence of the obtained solutions to the reference simulations while significantly reducing the computational costs.
Pulse thermography (PT) has proven to be a valuable non-destructive testing method to identify and quantify defects in fiber-reinforced polymers. To perform a quantitative defect characterization, the heat diffusion within the material as well as the material parameters must be known. The heterogeneous material structure of glass fiber-reinforced polymers (GFRP) as well as the semitransparency of the material for optical excitation sources of PT is still challenging. For homogeneous semitransparent materials, 1D analytical models describing the temperature distribution are available.
Here, we present an analytical approach to model PT for laterally inhomogeneous semitransparent materials.We show the validity of the model by considering different configurations of the optical heating source, the IR camera, and the differently coated GFRP sample. The model considers the lateral inhomogeneity of the semitransparency by an additional absorption coefficient. It includes additional effects such as thermal losses at the samples surfaces, multilayer systems with thermal contact resistance, and a finite duration of the heating pulse. By using a sufficient complexity of the analytical model, similar values of the material parameters were found for all six investigated configurations by numerical fitting.
Combining experiments and all-atom molecular dynamics simulations, we study the conformational behavior of polyacrylamide (PAM) in aqueous alcohol mixtures over a wide range of temperatures. This study Shows that even when the microscopic interaction is dictated by hydrogen bonding, unlike its counterparts that present a lower critical solution temperature (LCST), PAM shows a counterintuitive tunable upper critical solution temperature (UCST)-type phase transition in water/alcohol mixtures that was not reported before. The Phase transition temperature was found to be tunable between 4 and 60 1C by the type and concentration of alcohol in the mixture as well as by the solution concentration and molecular weight of the polymer. In addition, molecular dynamics simulations confirmed a UCST-like behaviour of the PAM in aqueous alcoholic solutions.
Additionally, it was observed that the PAM is more swollen in pure alcohol solutions than in 80% alcoholic solutions due to y-like behaviour. Additionally, in the globular state, the size of the aggregates was found to increase with increasing solvent hydrophobicity and polymer concentration of the solutions. Above ist Phase transition temperature, PAM might be present as individual polymer chains in the coil state (r10 nm). As PAM is a widespread polymer in many biomedical applications (gel electrophoresis, etc.), this finding could be of high relevance for many more practical applications in high performance pharmaceuticals and/or sensors.
UCST-type thermoresponsive polymers (i.e. that phase separate from solution upon cooling) present a tremendous potential not only in aqueous media where they can be used in drug delivery, diagnostic and microfluidic applications, but also in water/alcohol mixtures, where they can be used for instance in sensing systems for alcohol-soluble drugs. However, only a few thermoresponsive polymers have been reported that present an UCST in a relevant temperature range and “green” solvents such as water or ethanol.
In this context, acrylamide-based monomers can be very useful building blocks for designing novel non-ionic UCST-type polymers because of their hydrophilic nature (with the appropriate side chain) and propensity to form hydrogen bonds. We will present our latest results on the UCST-type thermoresponsive behaviour of acrylamide- and 2,6-diaminopyridine-based homopolymers and copolymers in water or water/alcohol mixtures, and give some insights about the rational design of UCST polymers relying on H-bonding.
