Im Folgenden soll über aktuelle Diskussionen und Arbeitsschwerpunkte im Fachbeirat der BAM und von der Zulassungsstelle berichtet werden. Es werden Änderungen im Zusammenhang mit der Zulassungsrichtlinie für Kunststoffdichtungsbahnen vorgestellt. Es wird über die aktuelle Diskussion über die Anforderungen z. B. an metallische Sensoren für Dichtungskontrollsysteme berichtet. Änderungen der Fremdprüfung von sogenannten Sandschutzmatten, des Standards zur Qualitätsüberwachung (SQÜ) von Kunststoff Dränelementen (KDE) und der Anpassung der Formulierungen zum Überbau der in Abdichtungen verwendeten Produkte werden aufgezeigt.
Im Folgenden soll über aktuelle Diskussionen und Arbeitsschwerpunkte im Fachbeirat der BAM und von der Zulassungsstelle berichtet werden. Es werden Änderungen im Zusammenhang mit der Zulassungsrichtlinie für Kunststoffdichtungsbahnen vorgestellt. Es wird über die aktuelle Diskussion über die Anforderungen z. B. an metallische Sensoren für Dichtungskontrollsysteme berichtet. Änderungen der Fremdprüfung von sogenannten Sandschutzmatten, des Standards zur Qualitätsüberwachung (SQÜ) von Kunststoff Dränelementen (KDE) und der Anpassung der Formulierungen zum Überbau der in Abdichtungen verwendeten Produkte werden aufgezeigt.
Die BAM wurde 2009 u. a. mit der Zulassung von Bewehrungsgittern für den Deponiebau beauftragt. Der Stand von Wissenschaft und Technik bot jedoch keine ausreichende Grundlage, die tatsächlich relevanten Widerstände und die mit diesen Widerständen verbundene Wechselwirkung zwischen Gitter und Boden im Hinblick auf eine mindestens 100-jährige Funktionsdauer realistisch beurteilen zu können. Es wurde daher nur eine vorläufige Zulassungsrichtlinie für diese Produkte erarbeitet. In der Zwischenzeit hat sich ein neuer Kenntnisstand ergeben. Vor diesem Hintergrund hat die AG (Arbeitsgruppe) Geogitter des Fachbeirats der BAM am 26.09.2017 getagt. Es sollen die aktuellen Erkenntnisse und Schlussfolgerungen für die Zulassung in Kürze vorgestellt werden. Die entsprechenden Kapitel beruhen dabei auf einem Arbeitspapier von W.W. Müller und dem Autoren, das der Arbeitsgruppe zur Diskussion vorgelegt wurde.
Weiterhin soll über Entwicklungen in Zusammenhang mit Kunststoffdichtungsbahnen (KDB) berichtet werden. Die Anforderungen an die Schweißzusätze für das Extrusionsschweißen wurden angepasst. Zudem soll über die aktuelle Diskussion der Schweißbarkeit unterschiedlicher HDPE Materialien informiert werden. Die maßgebliche DVS (Deutscher Verband für Schweißen und verwandte Verfahren) Richtlinie 2225-4 wird derzeit offenbar so ausgelegt, dass Materialien mit völlig unterschiedlichen Schmelze-Massefließrate miteinander verbunden werden können. Dies steht allerdings im grundlegenden Widerspruch zum Stand der Technik.

Der Vortrag stellt einige aktuelle Herausforderungen für die Prozessanalytik und mögliche Antworten vor.
Gepulste Raman-Spektrometer akkumulieren das Raman-Signal mit Hilfe schneller optischer Schalter im Picosekunden-Bereich, bevor langlebigere Fluoreszenzanregung entsteht. Damit lassen sich stark fluoreszierende Materialien untersuchen, die bislang nicht zugänglich sind. Eine weitere interessante Entwicklung ist etwa die Shifted excitation Raman difference spectroscopy (SERDS) die besonders für biologische Anwendungen interessant ist.
Flexible, modulare Produktionsanlagen stellen einen vielversprechenden Ansatz für die kontinuierliche Produktion von Fein- und Spezialchemikalien dar. In einem EU-Projekt wurde die Feldintegration eines Online-NMR-Sensormoduls als smartes Modul für die Prozesskontrolle vorangebracht. Dieses Modul basiert auf einem kommerziell erhältlichen Niederfeld-NMR-Spektrometer, welche zurzeit für die Anwendung im Laborbereich erhältlich ist. Für die Feldintegration wurde ein ATEX-zertifiziertes, explosionsgeschütztes Gehäuse entwickelt sowie Automationsschemen für den unbeaufsichtigten Betrieb und für die kalibrierfreie spektrale Datenauswertung erstellt.
Eine sehr gut anwendbare analytische Messtechnik zur Kontrolle der elementaren Zusammensetzung von verschiedensten Materialien ist die laserinduzierte Plasmaspektroskopie (LIPS, engl. LIBS - Laser-induced Breakdown Spectroscopy). Bei der LIBS wird ein kurz gepulster Laser auf eine Probe fokussiert, um ein Leuchtplasma zu erzeugen. Das dabei erzeugte Atomemissionsspektrum ermöglicht eine qualitative und quantitative Analyse der Zusammensetzung der Probe bezüglich praktisch aller Elemente des Periodensystems. In einem aktuellen Projekt wird diese Methode neben anderen zur Online-Analyse von Ackerböden für die ortsspezifischer Bewirtschaftung (Precision Agriculture) weiterentwickelt und bewertet.
Ein Ringversuch ist eine Methode der externen Qualitätssicherung für sowohl Messverfahren als auch die Eignung von Mess- und Prüflaboratorien. Dabei werden möglichst identische Proben mit identischen Verfahren oder mit unterschiedlichen Verfahren untersucht. Die Teilnahme an Ringversuchen ist für alle akkreditierten Laboratorien ein wesentlicher Baustein der institutsinternen und -externen Qualitätssicherung. Je mehr Faktoren Einfluss auf das Endergebnis haben können, desto wichtiger ist es diese Faktoren zu kennen und deren Einflüsse durch weitestgehende Vergleichbarkeit der Messverfahren zu minimieren.
Für die Bewertung von Emissionen aus Bauprodukten haben sich Emissionsmess-kammer-Untersuchungen als Messverfahren etabliert. Um den Geruch auch für Innenraumanwendungen bewertbar zu machen sind in den letzten Jahren erste Normen entstanden (z. B. DIN ISO 16000-28 und VDI 4302-1). Um die Eignung dieser Normen und deren Anwendung zu untersuchen sind seit 2012 nunmehr 3 Ringversuche der BAM auch zum Thema Geruch durchgeführt worden.
Die bislang durchgeführten Ringversuche mit dem Ziel der Geruchsbewertung wiesen Standardabweichungen von ca. 24 bis 30 % um den jeweiligen Mittelwert auf und wiesen somit ähnliche Werte auf, die auch bei den VOC-Vergleichen ermittelt wurden. Diese Entwicklungen werden hier aufgezeigt.
In diesem Beitrag soll in Kürze darüber informiert werden, welche Themen und Fragestellun-gen den Fachbeirat der BAM, seine Arbeitsgruppen und die BAM beschäftigt haben. Bereits in den letzten Jahren wurde an dieser Stelle über die Anforderungen an Kunststoff Drän-elemente berichtet. Vor dem Hintergrund unserer Beiträge zu diesem Thema, kam eine in-tensive Fachdiskussion im Fachbeirat der BAM und in weiteren Fachgremien in Gang. So wurden die Bemessungsgrundsätze der DGGT (Deutsche Gesellschaft für Geotechnik), die GDA (Geotechnik der Deponien und Altlasten) E (Empfehlung) 2-20 (Entwässerungsschich-ten in Oberflächenabdichtungssystemen), in einigen Punkten angepasst. Eine Musterverle-geanleitung für Kunststoff Dränelemente wurde erarbeitet. Der aktuelle Bearbeitungsstand soll hier vorgestellt werden. In diesem Zusammenhang wurde ein Untersuchungsprogramm zum Einfluss von Überlappstößen auf das Wasserableitvermögen von Kunst-stoff Dränelementen angestoßen. Die Ergebnisse sollen hier in Kürze vorgestellt und die Bedeutung für die Abminderungsfaktoren und damit für die Bemessung erläutert werden. Zu welchem Zeitpunkt muss ein Kunststoff Dränelement überbaut werden und welche weiteren Regelungen müssen hier beachtet werden? Was steckt eigentlich hinter diesen strengen Regelungen? Diese Fragen sollen im Folgenden diskutiert werden.
Die Deponieverordnung (DepV) fordert eine Akkreditierung der fremdprüfenden Stellen nach verschiedenen Normen. Die fachlichen Anforderungen an den „Fremdprüfer-Kunststoff“ legt die Richtlinie Fremdprüfer der BAM fest. Um diese Anforderungen im Akkreditierungsprozess ausreichend berücksichtigen zu können, wurde nun eine sogenannte DAkkS Regel erarbeitet, die die Anforderungen der Richtlinie spiegelt. Über den aktuellen Bearbeitungsstand soll berichtet werden.
Auch über die aktuelle Arbeit in Zusammenhang mit den Kunststoffdichtungsbahnen (KDB) soll berichtet werden. Der Prüfhinweis B14 (Bestimmung der Maßhaltigkeit von geosyntheti-schen Dichtungsbahnen aus Polyethylen hoher Dichte (PEHD)) wurde überarbeitet. Voran-gegangen war eine intensive Entwicklung der Prüfeinrichtung sowie ausgiebige Ringversuche im AK GWS (Arbeitskreis Grundwasserschutz). In diesem Zusammenhang wurden neue Anforderungen an die Produkte gestellt. Die Hintergründe dazu sollen kurz vorgestellt werden. Die DVS (Deutscher Verband für Schweißen und verwandte Verfahren) Richtlinie 2225-4 regelt das Schweißen der KDB auf der Deponiebaustelle. Werden KDB im strukturierten Bereich miteinander verschweißt, werden die Strukturen in der Regel vorher abgetragen. Die DVS Richtlinie 2225-4 sieht unter bestimmten Voraussetzungen von einem Abtragen der Strukturen ab. Vor diesem Hintergrund wurde ein Untersuchungsprogramm initiiert, das die Auswirkungen aufzeigen soll. Der Bearbeitungsstand wird vorgestellt.

(1) Die europäischen Anwendungsnormen und das deutsche Straßenbaumerkblatt für Geokunststoffe verwenden statistische Begriffe. Deren nur „beiläufige“ Erwähnung lässt jedoch offen, was genau im Zusammenhang gemeint ist. Produkte unterschiedlicher Hersteller können daher trotz gleicher Angaben zu den Mindestwerten in den Leistungserklärungen oder Datenblättern deutlich unterschiedliche Eigenschaften haben. Um vorschriftsmäßig bemessen zu können, muss man jedoch wissen, was sich hinter den Zahlen verbirgt. Eine Zulassung oder Zertifizierung ist nur dann sinnvoll, wenn darüber hinaus einheitlich definiert wird, was die Angaben für die werkseigene Produktionskontrolle, die Inspektion durch benannte Stellen und die Kontrolle durch Dritte bedeuten. In den vergangenen Jahren seit 2009 wurde versucht, die Zulassungen stärker auf die Vorgaben aus der europäischen Normung zu stützen. Daher musste nun diskutiert werden, welche statistische Bedeutung die Angaben in den Zulassungen für Kunststoff Dränelementen und Geotextilien zum Filtern und Trennen haben. Hier musste eine einheitliche Regelung getroffen werden.
(2) Die Langzeiteigenschaften von polyolefinen Geokunststoffen sind maßgeblich von den eingesetzten Stabilisatoren abhängig. Wie kann hier eine Überwachung während der Produktion der Geokunststoffe implementiert werden, wenn die Rezepturbestandteile der Produkte sensible Firmengeheimnisse der Rohstoffhersteller sind? Hier musste eine Lösung erarbeitet werden. Über die Ergebnisse soll berichtet werden.

Darüber hinaus werden Änderungen in den Zulassungsrichtlinien für Kunststoffdichtungsbahnen, Kunststoff-Dränelemente und Geogitter kurz vorgestellt.

Vorangegangen war eine intensive Entwicklung der Prüfeinrichtung sowie ausgiebige Ringversuche im AK GWS (Arbeitskreis Grundwasserschutz). In diesem Zusammenhang wurden neue Anforderungen an die Produkte gestellt. Die Hintergründe dazu sollen kurz vorgestellt werden. Die DVS (Deutscher Verband für Schweißen und verwandte Verfahren) Richtlinie 2225-4 regelt das Schweißen der KDB auf der Deponiebaustelle. Werden KDB im strukturierten Bereich miteinander verschweißt, werden die Strukturen in der Regel vorher abgetragen. Die DVS Richtlinie 2225-4 sieht unter bestimmten Voraussetzungen von einem Abtragen der Strukturen ab. Vor diesem Hintergrund wurde vom AK GWS ein Untersuchungsprogramm initiiert, das die Auswirkungen aufzeigen soll. Die Ergebnisse und Schlussfolgerungen werden vorgestellt.

The aim of this study was to investigate the passive sampler method Diffusive Gradients in Thin-films (DGT) for ammonium and nitrate in amended soils. Therefore, we used soils from a pot experiment with maize where nitrogen (N) was supplied as ammonium sulfate nitrate (ASN), without and with a nitrification inhibitor (NI). The additional use of a NI can delay the nitrification in the soil and making the ammonium available for a longer period in the soil solution after its application. Homogenized soil samples were collected directly from each pot after one week of incubation before sowing and after harvesting the maize. Nitrate and ammonium in these soil samples were extracted using DGT devices equipped with a Putolite A520E (for nitrate) and Microlite PrCH (for ammonium) binding layer. Ammonium DGT which determined the mobile and labile ammonium forms based on diffusion and the resupplies from the solid soil phase, only showed a significantly higher amount of extractable ammonium with NI compared to that without NI for some samples. However, significantly lower values were found for nitrate of treatments with NI compared to without NI after harvest. Thus, the lower nitrate amounts for treatments with NI compared to the treatments without NI after harvest indicated the delay of the nitrification process by the NI. Furthermore, we compared also the ammonium and nitrate
DGT results to chemical extraction with KCl solutions. The results demonstrated that the trends of DGT results and chemical extraction were complimentary through all the treatments.
Microcolonial black fungi are a group of ascomycetes that exhibit high stress tolerance, yeast-like growth and constitutive melanin formation. They dominate a range of hostile natural and man-made environments, from desert rocks and salterns to dishwashers, roofs and solar panels. Due to their slow growth and a lack of genetic tools, the underlying mechanisms of black fungi’s phenotypic traits have remained largely unexplored. We chose to address this gap by genetically engineering the rock-inhabiting fungus Knufia petricola (Eurotiomycetes, Chaetothyriales), a species that exhibits all characteristics of black fungi. A cell biological approach was taken by generating K. petricola strains expressing green or red fluorescent protein variants. By applying: (1) traditional gene replacement; (2) gene editing and replacement via plasmid-based or ribonucleoprotein (RNP)-based CRISPR/Cas9, and (3) silencing by RNA interference (RNAi), we constructed mutants in the pathways leading to melanin, carotenoids, uracil and adenine. Stable single and double mutants were generated with homologous recombination (HR) rates up to 100%. Efficient, partially cloning-free strategies to mutate multiple
genes with or without resistance cassettes were developed. This state-of-the-art genetic toolkit, together with the annotated genome sequence of strain A95, firmly established K. petricola as a model for exploring microcolonial black fungi.
An alternative spectroscopic approach for the monitoring of microplastics in environmental samples

The increasing pollution of terrestrial and aquatic ecosystems with plastic debris leads to the accumulation of microscopic plastic particles of still unknown fate. To monitor the degree of contamination and to understand the underlying processes of turnover, analytical methods are urgently needed, which help to identify and quantify microplastic (MP). Currently, costly collected and purified materials enriched on filters are investigated both by micro-infrared spectroscopy and micro-Raman. Although yielding precise results, these techniques are time consuming and restricted to sample aliquots in the order of micrograms precluding prompt and representative information on both, larger sample numbers and realistic material volumes. To overcome these problems, here we tested Raman and NIR process-spectroscopic methods in combination with multivariate data analysis.
For this purpose, artificial MP/soil mixture samples consisting of standard soils or sand with defined ratios of MP (0,5 – 10 mass% polymer) from polyethylene, polypropylene, and polystyrene were prepared. MP particles with diameters < 2 mm and < 125 µm were obtained from industrial polymer pellets after cryomilling. Spectra of these mixtures were collected by (i) a process FT-NIR spectrometer equipped with a reflection probe, (ii) by a cw-process Raman spectrometer and (iii) by a time-gated Raman spectrometer using fiber optic probes. For the calibration of chemometric models (partial least squares regression, PLSR) 5 – 10 spectra of defined MP/soil mixtures (consisting of 1 – 4 g material each) were collected. The obtained PLSR models served for the prediction of both, polymer type and content based on the spectra of “unknown” test samples.
Whereas MP could be detected by Raman spectroscopy in coastal sand at 0.5 mass%, in standard soils detection of MP was limited to 10 – 5 mass%. The sensitivity of Raman spectroscopy could be improved by mild treatment with hydrogen peroxide. FT-NIR was suitable for the investigation of MP in standard soils in the range of 5 – 1 mass%, however, here a non-linear effect was observed at higher polymer concentrations. When mixtures of several polymers at low concentration levels were milled together, FT-NIR spectroscopy yielded false positive polymers together with unprecise quantitative information. Recently, the investigation of “real-world” samples shall be tested and compared to the results obtained by micro-FTIR and micro-Raman.
The large number of nanomaterial-based applications emerging in the materials and life sciences and the foreseeable increasing use of these materials require methods that evaluate and characterize the toxic potential of these nanomaterials to keep safety risks to people and environment as low as possible. As nanomaterial toxicity is influenced by a variety of parameters like size, shape, chemical composition, and surface chemistry, high throughput screening (HTS) platforms are recommended for assessing cytotoxicity. Such platforms are not yet available for genotoxicity testing. Here, we present first results obtained for application-relevant nanomaterials using an automatable genotoxicity platform that relies on the quantification of the phosphorylated histone H2AX (γ-H2AX) for detecting DNA double strand breaks (DSBs) and the automated microscope system AKLIDES® for measuring integral fluorescence intensities at different excitation wavelengths. This platform is used to test the genotoxic potential of 30 nm-sized citrate-stabilized gold nanoparticles (Au-NPs) as well as micellar encapsulated iron oxide nanoparticles (FeOx-NPs) and different cadmium (Cd)-based semiconductor quantum dots (QDs), thereby also searching for positive and negative controls as reference materials. In addition, the influence of the QD shell composition on the genotoxic potential of these Cd-based QDs was studied, using CdSe cores as well as CdSe/CdS core/shell and CdSe/CdS/ZnS core/shell/shell QDs. Our results clearly revealed the genotoxicity of the Au-NPs and its absence in the FeOx-NPs. The genotoxicity of the Cd-QDs correlates with the shielding of their Cd-containing core, with the core/shell/shell architecture preventing genotoxicity risks. The fact that none of these nanomaterials showed cytotoxicity at the chosen particle concentrations in a conventional cell viability assay underlines the importance of genotoxicity studies to assess the hazardous potential of nanomaterials.

Immunochemical methods based on the binding of the analyte to specific antibodies enable fast & cost-effective on-site analyses. Magnetic bead-based immunoassays allow for implementation into an immunosensor for online testing. Particles are prepared by either coupling the analyte molecule (diclofenac) or the respective antibody (amoxicillin) to the surface. For miniaturization, detection is performed electrochemically (chronoamperometry) on a microfluidic chip. The developed immunosensor will enable detection of pharmaceuticals directly in water supply pipes.
Free-living and (ecto)-mycorrhizal fungi enhance rock weathering. In their roles as mineral weathering agents and mutualistic partners of phototrophs, fungi supply primary producers like plants and phototrophic microorganisms with mineral-derived nutrients. The exact mechanisms behind fungus-induced mineral weathering processes are however not well understood. Progress can be achieved here by reproducible experimental simulations of the natural processes, using well-characterised model organisms and minerals.
In this study, the weathering-affecting, rock-inhabiting fungus, Knufia petricola A95 and the Fe-bearing olivine (Fe0.2Mg1.8SiO4) were selected to investigate fungi-induced effects on mineral dissolution. The availability of a melanin-deficient mutant (ΔKppks) of K. petricola A95, that produced more extracellular polymeric substances (EPS) than the wild type (WT), enabled comparative studies of the role of melanin and EPS in weathering processes.
Three experimental systems, which generate long-term microbiological stability, were developed to study the impact of the WT and ΔKppks on olivine weathering: (1) batch and (2) mixed flow dissolution experiments, and (3) biofilm cultivation experiments. In addition, state-of-the-art analytical techniques were used to monitor changes in the growth medium, as well as of the mineral surface and biofilm-mineral interface.
Inductively coupled plasma optical emission spectrometry (ICP-OES) analysis of the Mg, Si and Fe concentrations in the reacted growth medium was used to quantify olivine dissolution. In abiotic controls, Mg and Si dissolved congruently, while Fe precipitated. The measured olivine dissolution rates at pH 6 were two orders of magnitude lower than previously reported, but similar at acidic pH. X-ray photoelectron spectroscopy (XPS) analyses of the olivine surface confirmed the presence of Fe (oxyhydr)oxide precipitates. Transmission electron microscopy (TEM) imaging of an abiotically reacted polished olivine section from the long-term cultivation experiment showed the presence of an amorphous layer enriched in Fe. All these observations indicate that the precipitation of Fe (oxyhydr)oxides on the olivine surface inhibits olivine dissolution.
Both tested rock-inhabiting fungal strains affect Fe precipitation as well as olivine dissolution. Evaluation of the WT and ΔKppks revealed that the WT formed less biomass but could take up higher amounts of metals (e.g. Fe) and was more efficient in its attachment to olivine. The WT and ΔKppks enhanced olivine dissolution as demonstrated by higher Mg and Si concentration in the reacted growth medium. They furthermore prevented Fe precipitation by binding Fe and retaining it in solution, thereby allowing olivine dissolution to proceed. The WT cells that were attached to the olivine surface were particularly efficient at inhibiting Fe precipitation. By binding Fe directly at the olivine surface, the WT cells removed the inhibition of olivine dissolution almost completely. TEM analysis of polished olivine sections, colonised by a fungal biofilm for seven months, supported this hypothesis. After long-term fungus-olivine interaction, the Fe-enriched, amorphous layer did not develop, and the olivine surface was stronger etched compared to the abiotic control.
To study the effect of mutualism on mineral weathering, K. petricola was grown with the cyanobacterium, Nostoc punctiforme ATCC 29133. Both partners showed an enhanced growth and formed a stratified biofilm which attached more strongly to olivine. Nevertheless, the olivine dissolution rate of the fungus-cyanobacterium consortium was moderate.
Rock weathering simulation systems developed here are promising research instruments. The experimental conditions allow for the alteration of the studied mineral surface, while the clear definition of these conditions delivers a stable growth of microorganisms. The latter makes these systems universally applicable, especially in combination with integrative multidisciplinary analytics. Processes underlying environmental and biological effects on rock weathering, metal corrosion, plastic degradation, or the deterioration of any other substrate can be studied reproducibly and over a long period of time. The chemical and biological complexity of these simulation systems mimics natural rock weathering processes. The mineral dissolution rates generated in this study are therefore relevant to natural ecosystems.
An immunochromatographic assay for the determination of bisphenol a releases from plastic materials

Background. The worldwide increase in the use of bisphenol A (BPA), a monomer commonly used to manufacture polycarbonate plastic products and the epoxy resins that line metal cans, has resulted in an increased human exposure because both products come in contact with food and beverages. Since BPA is an endocrine disruptor excessive exposure could affect human reproductive function. In response, various countries have adopted a range of measures, from voluntary reductions to outright bans of BPA in some products, mostly in baby bottles and containers for canned foods. Nonetheless, consumers would certainly appreciate a quick test for their own assessment of contaminations by bisphenol A.
Methods. An immunochromatographic test strip was developed. This lateral-flow immunoassay (LFIA) is based on anti-BPA antibodies (from mouse) conjugated with gold nanoparticles as the marker, deposited in an elaborate pad. To form control and test zones, anti-mouse antibody and a BPA-BSA conjugate were spotted on the nitrocellulose membrane, respectively.
Results. Negative samples are revealed by red lines both in the test and control zones, respectively, whereas positive samples produce a single red line only in the control zone. The limit of detection (LOD) was found to be 0.05 µg/l, visually detected by the naked eye within 5 minutes. In addition, the intensities at the test line can be read by dedicated lateral flow readers (opTrilyzer® and Cube by opTricon, Berlin) for a more precise and documented determination.
Conclusion. Pregnancy-test like strips in combination with hand-held readers provide a simple and versatile tool to determine the presence of bisphenol A quantitatively, making it a very attractive analytical approach especially for on-site applications at critical points, such as environmental analysis, consumer protection, and materials testing.

The development of a test to evaluate the degradation of semi-volatile fuels as diesel by microorganisms is presented. This method is based on the principles described in the CEC-L-103 Standard procedure that is exclusively meant for testing the biodegradability of non-volatile lubricants. Therefore, significant modifications involve aseptic conditions for testing specific microorganisms and conducting the test in closed vessels avoiding evaporation losses, while fuel quantification using gas chromatography-flame ionization detection (GC-FID) is retained. It is suggested that the modified procedure should enable routine application for semi-volatile hydrocarbon-based fuels. GC-FID provides additionally valuable information on the alteration of fuel component patterns during biodegradation. The procedure was successfully tested using two bacteria (Pseudomonas aeruginosa and Sphingomonas sp.) and two yeasts (Moesziomyces sp. and Candida sp.) isolated from real diesel contamination cases. All tested microorganisms caused a significant degradation of diesel fuel achieving hydrocarbon degradation percentages ranging from 23% to 35%. Specific aspects on the test modification and prospects for further modification regarding targeted investigations in the field of fuel contamination by microorganisms are briefly discussed.

Pseudomonas aeruginosa MPAO1 is the parental strain of the widely utilized transposon mutant collection for this important clinical pathogen. Here, we validate a model system to identify genes involved in biofilm growth and biofilm-associated antibiotic resistance. Our model employs a genomics-driven workflow to assemble the complete MPAO1 genome, identify unique and conserved genes by comparative genomics with the PAO1 reference strain and genes missed within existing assemblies by proteogenomics. Among over 200 unique MPAO1 genes, we identified six general essential genes that were overlooked when mapping public Tn-seq data sets against PAO1, including an antitoxin. Genomic data were integrated with phenotypic data from an experimental workflow using a user-friendly, soft lithography-based microfluidic flow chamber for biofilm growth and a screen with the Tn-mutant library in microtiter plates. The screen identified hitherto unknown genes involved in biofilm growth and antibiotic resistance. Experiments conducted with the flow chamber across three laboratories delivered reproducible data on P. aeruginosa biofilms and validated the function of both known genes and genes identified in the Tn-mutant screens. Differential Protein abundance data from planktonic cells versus biofilm confirmed the upregulation of candidates known to affect biofilm formation, of structural and secreted proteins of type VI secretion systems, and provided proteogenomic evidence for some missed MPAO1 genes. This integrated, broadly applicable model promises to improve the mechanistic understanding of biofilm formation, antimicrobial tolerance, and resistance evolution in biofilms.
The European Water Framework Directive 2000/60/EC requires monitoring of organic priority pollutants in so-called whole water samples, i.e. in aqueous nonfiltered samples that contain natural colloidal and suspended particulate matter. Colloids and suspended particles in the liquid phase constitute a challenge for sample homogeneity and stability. Within the joint research project ENV08 ‘‘Traceable measurements for monitoring critical pollutants under the European Water Framework Directive 2000/60/EC’’, whole water test materials were developed by spiking defined amounts of aqueous slurries of ultrafinely milled contaminated soil or sediment and aqueous solutions of humic acid into a natural mineral water matrix. This paper presents the results of an European-wide interlaboratory comparison (ILC) using this type of test materials. Target analytes were tributyltin, polybrominated diphenyl ethers and polycyclic aromatic hydrocarbons in the ng/L concentration range. Results of the ILC indicate that the produced materials are sufficiently homogeneous and stable to serve as samples for, e.g. proficiency testing or method validation. To our knowledge, this is the first time that ready-to-use water materials with a defined amount of suspended particulate and colloidal matter have been applied as test samples in an interlaboratory exercise. These samples meet the requirements of the European Water Framework Directive. Previous proficiency testing schemes mainly employed filtered water samples fortified with a spike of the target analyte in a water-miscible organic solvent.

Die weltweite Produktion von synthetischen Kunststoffen steigt stetig und ein Teil gelangt in die Umwelt. Die Kunststofffragmente werden durch Umwelteinflüsse zerkleinert und bilden schließlich Mikroplastik, dessen Gefährdungspotential intensiv diskutiert wird. Zur Beobachtung und Bewertung der Situation ist eine leistungsstarke und schnelle Analytik erforderlich, jedoch existieren noch keine harmonisierten Verfahren. Die ThermoExtraktionDesorption-GasChromatograpie-MassenSpektrometrie (TED-GC-MS) nutzt polymer-spezifische Zersetzungsprodukte und bietet durch ihre Schnelligkeit und Robustheit Vorteile gegenüber anderen Verfahren.

Auf dem Betreiberforum der DWA-Plattform P-Rück wurden die Empfehlungen für den Vollzug der Phosphorrückgewinnungspflicht aus Klärschlamm vorgestellt. Diese Empfehlungen behandeln die Häufigkeit der Phosphoruntersuchungen im Klärschlamm, wo ein Jahresgang empfohlen wird und eine Empfehlung der Phosphorbestimmung über Aufschluss der Probe in der Mikrowelle und Messung mit ICP-OES. Untermauert wurden die Empfehlungen mit den Ergebnissen von Klärschlamm-Monitoring, Ringversuch und Analytikvergleich aus dem Vorhaben extraWERT.
The presence, fate and effects of microplastics (MP) in terrestrial systems are largely unknown. The few existing studies investigated either agricultural or industrial sites. Several techniques were used for analysis, primarly spectroscopic methods such as FTIR or Raman. Sample pretreatments like density separations are common to reduce matrix. A lack of harmonised and standardised sampling instructions for microplastic investigations in the terrestrial area was identified as particular critical, because different studies are barely comparable.
The aim of the project is to develop a proposal for a harmonized procedure for sampling, sample preparation and the detection of microplastics in terrestrial matrices for total content determination. By detecting specific degradation products the thermal extraction desorption gas chromatography mass spectrometry (TED-GC-MS) allows a direct determination of mass content of MP in environmental samples.
This study presents diagnostic techniques to assess the structural vulnerability of earthen buildings. Medieval structures at Ambel (near Zaragoza, Spain), once a preceptory or monastic house belonging to the Military Orders, provide a useful case study. After more than a thousand years of construction, failure and repair from the 10th century to the present day, Ambel preceptory today is characterised by marked inhomogeneities: construction materials and structural typologies are juxtaposed, with structural discontinuities at the interfaces between construction phases. This paper argues that, while static analysis is an essential prerequisite before a suitable maintenance program can be defined, no evaluation of the structural behaviour of any historic building can ignore an understanding of the building’s history.
Quantitative chemical analysis of airborne particulate matter (PM) is vital for the understanding of health effects in indoor and outdoor environments, as well as for enforcing air quality regulations. Typically, airborne particles are sampled over long time periods on filters, followed by lab-based analysis, e.g., with inductively coupled plasma mass spectrometry (ICP-MS). Within the EURAMET EMPIR AEROMET project, cascade impactor aerosol sampling was combined for the first time with on-site total reflection X-ray fluorescence (TXRF) spectroscopy to develop a tool for quantifying particle element compositions within short time intervals and even on-site. This makes variations of aerosol chemistry observable with time resolution of only a few hours and with good size resolution in the PM10 range. A proof of principles of this methodological approach and the comparison to standard methods within the scope of a field campaign will be presented. Secondly, aerosol sampling and TXRF analysis seems suitable for the quantification of elements in indoor aerosols as well and may provide an important enhancement of existing methods for the analysis of organic species in aerosols (such as sampling and TD-GC/MS). As an example, the TXRF analysis of particles emitted from laser printers under controlled conditions in an environmental test chamber will be presented.
The presence of plastic debris and especially small plastic particles in marine ecosystems has been recognized in the 1970`s. Since then various other environmental systems all over the world, like rivers, lakes and biota have been found to be polluted by plastics. Lately, agriculturally used land, waste-water treatment and biogas facilities come into focus as possibly relevant distribution vectors. To monitor the situation and investigate distribution pathways, the need for fast and reliable analytical procedures is high.
Up to now, there is no standard procedure for sampling, concentrating and analyzing plastic particles in environmental samples. Our group recently reported the development of a new analyzing method for microscale plastic particles, thermal extraction desorption gas chromatography mass spectrometry (TED-GC-MS). This method consists of thermal extraction of the sample via thermogravimetric analysis, collection of the evolved gaseous decomposition products on a solid-phase adsorber and analysis of the loaded adsorber using Thermal-Desorption-GC-MS. This technique utilizes the characteristic decomposition products of polymers for identification and even quantification of plastics in environmental samples.
In the present work we investigated samples of fermentation residues taken from a biogas plant. The results of sample preparation, concentration and analysis are presented as well as a comparison with alternative methods.
The first analysis of environmental samples shows, that the use of TED-GC-MS is suitable to analyse microplastic in environmental samples. Compared to the micro-spectroscopic methods this method is faster and gives good hints to the occurrence of high loadings of MP in environmental samples. In consequence, the method allows the monitoring of the situation and the investigation of MP distribution pathways in environment, to assess the relevance of various inputs.
While plastics have become indispensable in our daily lives over the last decades, the input into the environment has been increasing concurrently. Plastics often end up in the environment because of intensive use and poor waste management practice. They are subjected to aging and fragmentation and finally be deposited as microplastic particles or in short microplastics (MP). MP are defined as particles originating from synthetic polymers between 1 µm and 5 mm.
Although the pervasive abundance of MP in aquatic environments has been demonstrated comprehensively, less is known about the occurrence and fate of MP in terrestrial ecosystems. It is still unclear if soil functions as a MP source or a sink for aquatic environments. MP can either be transported into water bodies by soil erosion or be retained in soils. The few studies published are not comparable because of non-existent harmonized and standardized methods for sampling, sample preparation, and analysis.
For an assessment of a potential exposure situation of MP, the determination of a mass content in the soil is crucial. Consequently, spectroscopic methods like Raman or FTIR are not suitable, as they deliver information about the shape and size of individual particles. Therefore, we show the application of ThermoExtractionDesorption-GasChromatography-MassSpectrometry (TED-GC-MS) for MP analysis in the soil. In this method, the soil sample is heated up to 600 °C in a nitrogen atmosphere. The decomposition gases are sorbed on a solid phase, then transferred to a GS-MS system where they are desorbed, separated and identified. The method allows the rapid identification of individual polymers through the detection of specific decomposition products, but also the quantitative determination of the MP mass. Besides thermoplastics, elastomers originated from tire abrasion, can be detected.
In the present study, several terrestrial ecosystems in south-west Germany were systematically sampled. Subsequent sample preparation included sieving in fractions of 5-100 µm, 100-1000 µm, and 1-5 mm. MP were extracted by density separation using ZnCl2 solutions. The detection was done by TED-GC-MS measurements. Data of agricultural areas and floodplains are presented exemplarily. A quantitative assessment of highly occurring MP from littering as well as tire abrasion is conducted.
The talk is about microplastics in soil. Some results were presented with mainly state of the art according to microplastics in standardization.
Per- and polyfluoroalkyl substances (PFAS) have emerged over the course of the last twenty years as a global pollution issue. Altogether, there are currently more than 4700 known but only partly characterized fluorinated compounds identified by the Organization for Economic Cooperation and Development (OECD), as shown by a recently published classification proposal.
The ongoing production of new, yet unrestricted PFAS alternatives has become a major challenge for environmental routine analytics, since the state-of-the-art method LC-MS/MS relies on structural information and availability of isotope standards of the targeted compound. First reported by Miyake et al., fluorine sum parameters like adsorbable organic fluorine (AOF), extractable organic fluorine (EOF) and total fluorine (TF) can be applied to survey and detect the presence of large amounts of unidentified organofluorine compounds in environmental matrices. Today fluorine sum parameters have been established as a useful supplement to classic target-analytical approaches of PFAS and were implemented for the first time as a sum value “PFAS-total” in the recently revised Drinking Water Directive (2020/2184) by the European Commission.
In contrast, X-ray absorption near-edge structure (XANES) spectroscopy has been widely applied to identify low concentration of element-specific contamination without pre-treatment in environmental samples in the past. The XANES approach enables a penetration depth of approx. 1 µm (at fluorine K-edge energy; depending on the matrix) which is significantly deeper than for X-ray photoelectron spectroscopy (XPS; penetration depth approx. 10 nm). Furthermore, the method is fast, non-destructive and only simple preparation of the samples is required.
In our work, we combined both F-XANES spectroscopy and combustion ion chromatography (CIC) in order to present a new analytical perspective on the investigation of PFAS in environmental media and material samples.
Tire wear particles (TW) are generated by the abrasions of tires on the road surface through traffic. These particles can be transported by air and surface runoff and might also infiltrate the soil and consequently affect terrestrial ecosystems. The estimated tire wear (TW) emissions are immense, with 1.33 106 t a-1 in Europe. Despite this, only little is known about the environmental contents or the fate of TW.
One reason for this knowledge gap is the challenging analysis of TW in environmental samples. Detection of TW with spectroscopic methods is problematic due to high fluorescence interferences caused by contained black carbon. One analytical approach is to use zinc (Zn), a typical additive in tires, as a specific marker for the quantification of tire wear. However, any Zn originating from the sample matrix must be separated beforehand and requires elaborate sample preparation.
Car tires consist partly of synthetic rubbers, such as styrene-butadiene-rubber (SBR). This SBR could be identified and quantified via Thermal-Extraction-Desorption-Gas Chromatography-Mass Spectrometry (TED-GC-MS). This newly developed and fast screening method allows the simultaneous detection of microplastics and TW mass contents and requires minimal to no sample preparation. Firstly the sample is thermally extracted in a thermobalance under a nitrogen atmosphere. The resulting specific decomposition products are sorbed on a solid phase adsorber, which is then transferred to a GC-MS via an autosampler, where the products are desorbed, separated and identified. Cyclohexenylbenzene is used as a specific marker for SBR.
Here we investigated top layer soil samples, collected at the roadside of highly frequented German highways. Samples were analyzed without sample preparation, and SBR was detected in all investigated samples in mass contents ranging from 67.2 to 2230 mg kg-1. A correlation between SBR and Zn content in the soil was confirmed, while the correlation between SBR and Corg was hardly pronounced. We successfully demonstrated the application of TED-GC-MS as a screening method for tire wear in soil samples.
The present study will discuss these analytical results in detail as well as sampling parameters like sampling depth and distance to the roadside, and the effect of the particle size on the particle transport by water runoff and air.
Analytical Challenges for PFAS in Environmental Samples - Methods, Approaches and Applicability

Per- and polyfluoroalkyl substances (PFAS) are anionic, cationic and zwitterionic synthetic products, in which the hydrogen atoms on the carbon skeleton of at least one carbon atom have been completely replaced by fluorine atoms and which include up to 1.7 M compounds, depending on the definition. As a result of continuous and prolific use, mainly in aviation firefighting foams, thousands of industrial and military installations have been found to contain contaminated soil, groundwater and surface water. Furthermore, because of the continuous contamination through PFAS containing commercial products, effluents and sewage sludge from WWTPs have been shown to be an important source of PFAS discharge into the aquatic environment. In the last few years, legacy PFAS (≥C4) have been found in various environments, including soil, water and wastewater, and their environmental pathways have been partly described.
Several long-chain PFAS species, and their respective salts are considered as persistent organic pollutants by the United Nations Stockholm Convention. These pollutants have been linked to altered immune and thyroid function, liver disease, lipid and insulin dysregulation, kidney disease, adverse reproductive and developmental outcomes, and cancer. A significant shift in the chemical industry towards production of short (C4-C7) and ultrashort (C1-C3) alternatives was observed in response to recently intensified regulations and restrictions on the use of long-chain (≥C8) PFAS. PFAS analysis in environmental samples is currently mainly done by liquid chromatography tandem mass spectrometry (LC-MS/MS). This efficient method is conducted in a targeted fashion analyzing a small subset of PFAS. The US EPA method for analysis of PFAS using LC-MS/MS for example currently lists 40 PFAS (≥C4). However, to get a better overview of the amount of “total PFAS,” sum parameter methods like total oxidizable precursor (TOP) assay and methods based on combustion ion chromatography (CIC) are in development. CIC results in data regarding the sum of absorbable organic fluorine (AOF) or extractable organic fluorine (EOF), which can also quantify other organically bound fluorine compounds such as fluorinated pesticides and pharmaceutical. Moreover, non-target and suspect screening mass spectrometry can be used to identify novel emerging PFAS and partly unknown fluorinated compounds in environmental samples. Furthermore, to analyze ultrashort PFAS (C1-C3), supercritical fluid chromatography (SFC), hydrophilic interaction chromatography (HILIC) and gas chromatography-mass spectrometry (GC-MS) are available, but further research is needed to develop reliable and accurate methods to quantify several ultrashort PFAS in environmental samples.
Additionally, for research purpose several spectroscopical methods like X-ray photoelectron spectroscopy (XPS), fluorine K-edge X-ray absorption near-edge structure (XANES)spectroscopy, particular induced gamma-ray emission (PIGE) spectroscopy and 19F nuclear magnetic resonance (NMR) spectroscopy are available.
A vast number of emerging pollutants is being detected in the environment. Another lingering problem are health-threatening contaminants, such as mycotoxins, that deteriorate food and feed.
Analytical methods, suitable for trace analysis, are needed that are desirably also fast, inexpensive and, if possible, robust, and portable. Immunoanalytical, i.e., anti-body-based technologies, which are available in a broad range of formats, can be profitably used here to analyze for the distribution and the concentration trends of contaminants.
Some of these formats are single-analyte but high-throughput methods. To use them wisely, indicator substances or sum parameters must be established to be used in screening approaches. Other methods are suitable to be performed on portable in-strumentation in the field (on-site), more precisely at the point-of-need. Furthermore, there are array technologies that allow for parallel analysis of several analytes of interest (multiplexing). Seit 1968 wird durch Lunde vermutet, dass sich lipidlösliche Arsenverbindungen im Muskelgewebe fettreicher Fische, wie dem Hering (clupea harengus) anreichern. Obwohl marine Organismen ausgiebig untersucht worden sind, gibt es bisher wenige Arbeiten zur analytischen Charakterisierung dieser Spezies. In den 60iger Jahren konnte Gulbrand Lunde erstmalig den Arsengehalt in Fischölen von bis zu 20 mg/kg bestimmen. Seitdem wurden drei Klassen an Arsenolipiden identifiziert und in Ansätzen charakterisiert: Arsenokohlenwasserstoffe (As-HC), Arsenofettsäuren (As-FA) und die Arsenozuckerphospholipide (As-SugPL). In der vorzustellenden Arbeit konnten erstmalig mehr als 20 fettlösliche Arsenolipide in Heringsfilet mittels der Kopplungen HPLC-ICP-MS und ESI-Q-TOF-MS identifiziert und semiquantifiziert werden. Dabei erfolgte die strukturelle Zuordnung über die exakten Massen und durch gezielte MS-MS-Experimente, deren Fragmente eine eindeutige Charakterisierung ermöglichten. Es konnten bisher unbekannte Arsenolipide im Hering nachgewiesen werden. Eine Untersuchung weiterer Fischarten ergab, dass diese Arsenverbindungen in unterschiedlichen marinen Speisefischen weiter verbreitet sind, als bisher angenommen. In Stoffwechselstudien konnte gezeigt werden, dass es zu einer erhöhten Konzentration des kanzerogenen Dimethylarsinats (DMA) in menschlichem Urin nach dem Verzehr von Hering kommt. Weiterhin konnte aufgrund der Ergebnisse eventuell im Hering enthaltenes anorganisches Arsen, Monomethylarsonat und DMA als Ursache ausgeschlossen werden. Da Arsenobetain nachweislich nicht im menschlichen Körper metabolisiert wird, sind die in den durchgeführten Arbeiten erzielte Ergebnisse zweckdienlich, die These zu bestärken, dass die identifizierten Arsenolipide zu DMA abgebaut werden können.
Analytische Verfahren zur On-Line-Bestimmung von Elemementkonzentrationen in Pulvern und Aschen

Die Aufbereitung kommunaler Abwässer leistet einen wichtigen Beitrag zum Schutz der Umwelt und der Reinhaltung der Gewässer. Im Zuge der Abwasserreinigung entstehen deutschlandweit jährlich ca. 2,4 Millionen Tonnen Klärschlamm. Diese Massen stellen für die Anlagenbetreiber oft eine Herausforderung bei der Entsorgung dar. Es sind jedoch neben zahllosen Schadstoffen auch anorganische Wert- und Nährstoffe enthalten deren Nutzung zunehmend attraktiv wird. Ein essenzieller und in Lagerstätten begrenzt vorhandener Nährstoff ist Phosphor. Dieser ist in Klärschlämmen enthalten und erreicht in Klärschlammaschen Konzentrationen von bis zu 10 %. Eine Verwertung als Düngemittel in der Landwirtschaft ist jedoch nur für schadstoffarme Materiale mit guter Pflanzenverfügbarkeit der Nährstoffe zulässig. Für die Aufbereitung von Klärschlämmen und Schadstoffentfrachtung von Klärschlammaschen existieren verschiedene technologische Ansätze. Für jeden Prozess ist eine analytische Überwachung der Nährstoffwerte sowie der Schadstoffgrenzwerte erforderlich. Im Vortrag werden die Ergebnisse eines Projektes zur on-line Analytik thermisch aufbereiteter Klärschlammaschen vorgestellt. Es werden die Methoden der Röntgenfluoreszenz (RFA) und der Laserplasmaspektroskopie (LIBS) parallel angewendet und verglichen.
With the advent of widely accessible and cost-effective next-generation sequencing technologies, it has become increasingly feasible to study insect immunity on a deep genomic or transcriptomic level. Here we introduce a protocol that is aimed at exploiting transcriptomic data to study immunity in non-model insect organisms. We provide instructions for an entire workflow, starting with successfiil extraction of insect RNA through to bioinformatic guidelines for the effective analysis of mRNA sequencing data. The RNA extraction procedure is based on TRIzol Reagent and a spin-column clean-up Step. The bioinformatic pipeline is intended to help users identify immune genes from de novo transcriptome data and includes guidelines for conducting differential gene expression analyses on transcriptomic data. The immune gene prediction method is based on inferring protein homologs with HMMERand Blastp and talces Advantage ofthe ImmunoDB database, which is a valuable resource for research on insect immune-related genes and gene families. The differential gene expression analysis procedure utilizes the DESeq2 package as implemented in R. We hope this protocol will serve as a usefi.il resource for researchers aiming to study immunity in non-model insect species.
Working towards a comprehensive understanding of introduction pathways, number, and fate of micro¬plastics in the environment, suitable analytical methods are a precondition. Micro-spectroscopic methods are probably the most widely used techniques. Besides their ability to measure single spectra of a particle or fiber, most modern FTIR- and Raman microscopes are also capable of two-dimensional imaging. This is very appealing to microplastics research because it allows to simultaneously characterize the analytes chemically as well as their size (distribution) and shape.
Two-dimensional imaging on extensive sample areas with FTIR-micros¬copes is facilitated by focal plane array (FPA) detectors resulting in large data sets comprised of up to several million spectra. With numbers too large for manual inspection of each individual spectrum, automated data evaluation is inevitable. Identifying different polymers based on the comparison with known reference spectra (library search) has proven to be a suitable approach. For that purpose, FTIR-spectra of common plastics can be collected to create an individual reference library.
To Supplement this ‘targeted analysis’, looking for known substances via library search, an exploratory approach was tested. Principal component analysis (PCA) proved to be a helpful tool to drastically reduce the size of the data set while maintaining the significant information. Subsequently, cluster analysis was used to find groups of similar spectra. Spectra found in different clusters could be assigned to different polymer types. The variation observed within clusters gives a hint on chemical variability of microplastics of the same polymer found in the sample. Spectra labeled according to the respective cluster/polymer type were used to build a classification model which allowed to quickly predict the polymer type based on the FTIR spectrum. Classification was tested on a second, independent data set and results were compared to the spectral library search procedure.
Functional nanomaterials (NM) of different size, shape, chemical composition, and surface chemistry are of increasing relevance for many key technologies of the twenty-first century. This includes polymer and silica or silica-coated nanoparticles (NP) with covalently bound surface groups, semiconductor quantum dots (QD), metal and metal oxide NP, and lanthanide-based NP with coordinatively or electrostatically bound ligands, as well as surface-coated nanostructures like micellar encapsulated NP. The surface chemistry can significantly affect the physicochemical properties of NM, their charge, their processability and performance, as well as their impact on human health and the environment. Thus, analytical methods for the characterization of NM surface chemistry regarding chemical identification, quantification, and accessibility of functional groups (FG) and surface ligands bearing such FG are of increasing importance for quality control of NM synthesis up to nanosafety. Here, we provide an overview of analytical methods for FG analysis and quantification with special emphasis on bioanalytically relevant FG broadly utilized for the covalent attachment of biomolecules like proteins, peptides, and oligonucleotides and address methodand material-related challenges and limitations. Analytical techniques reviewed include electrochemical titration methods, optical assays, nuclear magnetic resonance and vibrational spectroscopy, as well as X-ray based and thermal analysis methods, covering the last 5–10 years. Criteria for method classification and evaluation include the need for a signal-generating label, provision of either the total or derivatizable number of FG, need for expensive instrumentation, and suitability for process and production control during NM synthesis and functionalization.
Salt tolerance in the γ-proteobacterium Halomonas elongata is linked to its ability to produce the compatible solute ectoine. The metabolism of ectoine production is of great interest since it can shed light on the biochemical basis of halotolerance as well as pave the way for the improvement of the biotechnological production of such compatible solute. Ectoine belongs to the biosynthetic family of aspartate-derived amino-acids. Aspartate is formed from oxaloacetate, thereby connecting ectoine production to the anaplerotic reactions that refill carbon into the tricarboxylic acid cycle (TCA cycle). This places a high demand on these reactions and creates the need to regulate them not only in response to growth but also in response to extracellular salt concentration. In this work, we combine modeling and experiments to analyze how these different needs shape the anaplerotic reactions in H. elongata. First, the stoichiometric and thermodynamic factors that condition the flux distributions are analyzed, then the optimal patterns of operation for oxaloacetate production are calculated. Finally, the phenotype of two deletion mutants lacking potentially relevant anaplerotic enzymes: phosphoenolpyruvate carboxylase (Ppc) and oxaloacetate decarboxylase (Oad) are experimentally characterized. The results show that the anaplerotic reactions in H. elongata are indeed subject to evolutionary pressures that differ from those faced by other gram-negative bacteria. Ectoine producing halophiles must meet a higher metabolic demand for oxaloacetate and the reliance of many marine bacteria on the Entner-Doudoroff pathway compromises the anaplerotic efficiency of Ppc, which is usually one of the main enzymes fulfilling this role. The anaplerotic flux in H. elongata is contributed not only by Ppc but also by Oad, an enzyme that has not yet been shown to play this role in vivo. Ppc is necessary for H. elongata to grow normally at low salt concentrations but it is not required to achieve near maximal growth rates as long as there is a steep sodium gradient. On the other hand, the lack of Oad presents serious difficulties to grow at high salt concentrations. This points to a shared role of these two enzymes in guaranteeing the supply of oxaloacetate for biosynthetic reactions.
Gypsum (CaSO4∙2H2O), bassanite (CaSO4∙0.5H2O), and anhydrite (CaSO4) are essential evaporite minerals for the evolution of hyper-arid surface environments on Earth and Mars (Voigt et al. 2019; Vaniman et al. 2018). The formation mechanism of especially anhydrite has been a matter of scientific debate for more than a century (van’t Hoff et al. 1903). To date, there exists no model that can reliably predict anhydrite formation at earth’s surface conditions. While thermodynamics favor its formation, it is hardly achieved on laboratory time scales at conditions fitting either the Atacama Desert on Earth, or the surface of Mars (Wehmann et al. 2023). In light of most recent developments (e.g. Stawski et al. 2016), that advocate for a complex, non-classical nucleation mechanism for all calcium sulphates, we present an analysis of natural samples from the Atacama Desert to identify key features that promote the nucleation and growth of anhydrite under planetary surface conditions. Our analyses reveal at least three distinct anhydrite facies, with differing mineralogy and micro- to nano-structures. The facies are (1) aeolian deposits with sub-μm grain sizes, (2) (sub-)surface nodules that formed from aeolian deposits and (3) selenites with secondary anhydrite rims. Possible mechanisms of their formation will be discussed.
Im Beitrag wurden auf der Basis der Ergebnisse des laufenden UFOPLAN-Forschungsvorhabens „Monitoring zur Verwertung von Bodenmaterial“ die nach BBodSchV anzuwendenden Untersuchungsverfahren bewertet und Probleme bei der Untersuchung von Bodenmaterial insbesondere im Zusammenhang mit der Analyse von PAK im Feststoff und im Eluat aufgezeigt. Dabei wurde der Einfluss der Probenaufbereitung und Probenvorbereitung vor der Analytik berücksichtigt.
In urban waters, a multitude of organic micropollutants, often termed emerging pollutants, has been found over the last decades. Analytical methods suitable for trace analysis are needed that are desirably also fast, inexpensive and, if possible, robust and portable. Immunoanalytical, i.e., antibody-based, methods which are available in a broad range of formats, can be profitably used here to screen for the distribution and to monitor the trends of concentration levels of contaminants of emerging concern in the environment. Some of these formats are single-analyte but high-throughput methods. To use them wisely, indicator substances, sometimes called anthropogenic markers, should be selected and used in screening approaches, i.e., as indicators for contamination and the pre-selection of samples at which to have a closer look by multiplex methods like LC-MS/MS. Other methods are suitable to be performed on portable instrumentation in the field (on-site) or in facilities such as wastewater treatment plants for on-line monitoring of the treatment and elimination process. Furthermore, array technologies have been established that allow for parallel (multiplex) analysis of several analytes of interest.
The microtiter-plate based ELISA (Enzyme-linked Immunosorbent Assay) is the method of choice for the analysis of a large number of samples [1]. ELISAs are available to monitor for anthropogenic markers such as the antiepileptic carbamazepine, the analgesic diclofenac, the antihistaminic cetirizine, the steroid hormone estrone, the antimicrobial sulfamethoxazole, psychoactive caffeine and cocaine, the priority pollutant bisphenol A, and the bile acid isolithocholic acid. For on-site screening and monitoring, simpler formats, like mix-and-read assays, e.g., the Fluorescence Polarization Immunoassay (FPIA) [2] or Lateral-flow Immunoassays (LFIA) [3] are more suitable tools, the latter based on dipsticks or little cassettes, with which users have become very familiar during the COVID-19 pandemic via rapid antigen tests. The suitability of multi-analyte formats such as immunomicroarrays depends on the choice of a signal-producing system that provides small uncertainties and good reproducibility of the measurements. Bead-based (“suspension”) arrays, read out in flow cytometers, are a powerful platform for multiplex assays [4]. Electrochemical formats, run on portable devices, provide additional advantages as no light source is required. They are most promising for stand-alone analysers and biosensors [5].
The speed, low cost and on-site capabilities of these methods allow to gather a lot more data on anthropogenic compounds which enables to quantify inputs, differences in degradation power of elimination processes, dilution phenomena and a more precise image of individual water cycles which is demonstrated by several examples.
The antibacterial features of natural pine/spruce rosin are well established, yet the functionality in various thermoplastics has not been surveyed. This work focuses on the processing of industrial grade purified rosin mixed with polyethylene (PE), polypropylene (PP), polylactic acid (PLA), polyamide (PA) and corn starch based biopolymer (CS). Homopolymer masterbatches were extrusion-compounded and melt-spun to form fibres for a wide range of products, such as filters, reinforcements, clothing and medical textiles. Due to the versatile chemical structure of rosin, it was observed compatible with all the selected polymers. In general, the rosin-blended systems were shear-thinning in a molten condition. The doped fibres spun of PE and PP indicated adequate melt-spinning capability and proper mechanical properties in terms of ultimate strength and Young's modulus. The antibacterial response was found dependent on the selected polymer. Especially PE with a 10 wt% rosin content showed significant antibacterial effects against Escherichia coli DH5α and Staphylococcus aureus ATCC 12598 when analysed in the Ringer's solution for 24 h.
The overarching goal of this project is to develop a predictive model for efflux-mediated antimicrobial tolerance in bacterial multicellular assemblies. Our central hypostasis is that efflux pump activity causes emergent antibiotic tolerance of multicellular bacterial populations, through the interplay of efflux mediated spatial interactions and efflux-linked persistence. To test this hypothesis, we will use a combination of microscopy, microbial killing assays, computational modelling, and data analysis, integrating information from 3 types of multicellular assembly: colonies, cell-to-cell interactions in a monolayer microfluidic device, and 3D flow chamber biofilms. Building on our preliminary observations, we will experimentally characterize the link between colony structure and spatial patterns of efflux gene expression in strains that differ in their levels of efflux. We will develop a mathematical model to test whether local growth inhibition of neighbors due to effluxing cells, coupled with local environment-dependent regulation of efflux, can account qualitatively for these results. By including persister cell formation in our model we will predict, and measure, the emergent function of antimicrobial tolerance in our colonies. To fully understand how tolerance emerges from the interplay between efflux-mediated spatial interactions and efflux-linked persister cell formation, we need quantitative measurements at the single cell level. To this end, we will use a microfluidic setup with cells growing in a monolayer to qualify in detail the dependence of efflux expression and persister cell formation on nutrient conditions, the correlation between efflux and persister formation, and the spatial range of efflux-mediated neighbour growth inhibition. To predict and quantitatively understand the emergent multicellular function of tolerance, we will perform individual-based modelling of biofilm growth, using as input the parameters measured on the single-cell level with our microfluidics experiments. Our simulations will predict biofilm spatial structure development, patterns of efflux and persister formation and, ultimately, tolerance to antimicrobial challenge. These predictions will be directly tested in flow-cell biofilm experiments. We are currently generating acrAB-tolC knockout-strain, without efflux activity, and a strain with an inducible acrAB-tolC efflux pump. To distinguish the different strains under the microscope, they were labeled with genes encoding for different fluorescent proteins. All strains are currently characterized in terms of growth, minimum inhibitory concentration of different antimicrobial substances, colony morphology, and biofilm formation ability. On the theoretical side, we are currently working on modeling the system at various scales and degree of detail, ranging from coarse-grained continuum models to stochastic, individual-based models. Some exploratory work was doe to test existing software for individual-based modelling that may be adapted for our purpose. Furthermore, we are in the process of developing more coarse-grained models. This work involves some physiological modelling and literature search, focusing on working mechanisms of efflux pumps and kinetic models for import and export of antibiotics.

Der Vortrag beschreibt den Stand der Arbeiten an der BAM, Fachbereich Umweltanalytik zur Entwicklung von Sensoren für die Wasseranalytik. Er stellt die bisherigen Arbeiten da, die ausgehend von ELISAs, zunehmend portable Plattformen wie FPIA und LFIA umfassen. Beas-based Formate werden von mikrotiterplatten-basierten Techniken auf mikrofluidische Lab-on-Chip-Systeme übertragen, die bei geeigneter Miniaturisierung und Automatisierung als Sensorsysteme angesehen werden können. Der Vortrag endet mit einem Blick auf ein laufendes Monitoring-Projekt von anthropogenen Markern in Abwasser und charakterisiert die Treiber, die die Entwicklung von Sensoren für die Wasseranalytik in den letzten Jahren vorantreiben.
The antimicrobial photodynamic approach has been demonstrated as an efficient and sustainable process for the eradication of microbial pathogens. In this work, silica-coated Magnetite nanoparticles (NPs) were used as carriers of glycosylated porphyrins and phthalocyanines. Their subsequent cationization
resulted in the production of stable antimicrobial photosensitizing materials, effective against E. coli. Suspensions of the photocatalysts in water present bimodal size distributions formed by big clusters and small NPs with hydrodynamic diameters between 8 and 38 nm. The presence of small NPs in the suspensions is related to an effective photodynamic inactivation (PDI) of E. coli cells. Glycosylation of the PS showed a positive effect on the PDI performance, which could be related to a higher accumulation of the photocatalyst over the bacterial cell membrane. In addition, these biocidal agents proved to be photostable and their photoactive performance decreased only between 23% and 28% upon 5 PDI cycles, mostly because of the loss of material between cycles, which makes them promising materials for water disinfection purposes.
Biocides are in contact with soils through direct application and passive leaching from protected materials. Consequently, soil microorganisms are exposed to biocides even though they are not their primary targets. Soil is a large reservoir of microbial diversity and has been hypothesized to be a crucial factor for the evolution and spread of antimicrobial resistance.
Currently, there is little knowledge on how biocides used to protect materials affect the evolution and spread of resistance. Thus, our aim is to investigate the risk for the evolution of biocide resistance and cross-resistance to antibiotics. In addition, we aim to elucidate the affect of biocides on the spread of resistance via horizontal gene transfer (HGT).
In adaptive laboratory evolution experiments we cultured selected model soil microorganism with representative biocides under selection regimes with increasing and stable biocide concentrations followed by antibiotic and biocide cross-resistance determination. Moreover, we investigate if the selected biocides affect the rates of de novo mutations and HGT of plasmids that carry resistance genes among soil microorganism.
Our results show only small increases of biocide resistance during serial transfers under increasing biocide concentrations. One reason for this might be the narrow selective window for biocide resistance due to steep dose-response relationships. Furthermore, our results indicate that a stable low-level biocide regime did not select for high level cross-resistance to antibiotics and other biocides.
Moreover, material preservatives affected the rates of HGT via conjugation and the mutation rates at sub-inhibitory concentrations. The results will enable future risk assessment regarding resistance evolution for biocides used as material preservatives.
