Sunlight is an almost unavoidable environmental cue and plays a fundamental role in the biology of pro- and eukaryotic organisms. To cope with sunlight-associated stresses e.g., high temperatures, UV radiation with associated DNA damage, accumulation of reactive oxygen species (ROS), desiccation and osmotic stresses, it is important for organisms to accurately sense and respond to changes in light. The benefits of light are obvious for green organisms such as cyanobacteria, algae and plants which use light as an energy source (photosynthesis). Less apparent are other light-dependent processes such as light-driven DNA repair by photolyases (photoreactivation) or ion pumping by microbial opsins. Fungi that can share light-flooded habitats with phototrophs may profit from their excess photosynthetic products. Rock-inhabiting Dothideomycetes and Eurotiomycetes including Knufia petricola possess many proteins for absorbing UV/blue, green, red and far-red light, produce the black 1,8 dihydroxynaphthalene (DHN) melanin and orange-red carotenoids, and may live in multispecies biofilms. Here, we are addressing the question to which extent constitutive pigment formation (melanin and carotenoids) and responses mediated by the stress-activated mitogen-activated protein (MAP) kinase contribute to the observed light (UV-B) tolerance of K. petricola.

Since 2016 ISO Technical Committee (TC) 265 is working on standardization the whole CCS-process chain. Around 30 countries are working together to provide international guidelines. As ISO 27913 was the first standard finished within these activities it is already in the stage of first revision.
The objective of ISO 27913 is “to provide specific requirements and recommendations on certain aspects of safe and reliable design, construction and operation of pipelines intended for the large-scale transportation of CO2 that are not already covered in existing pipeline standards such as ISO 13623, ASME B31.8, EN 1594, AS 2885 or other standards. Existing pipeline standards cover many of the issues related to the design and construction of CO2 pipelines; however, there are some CO2 specific issues that are not adequately covered in these standards. The purpose of this document is to cover these issues consistently. Hence, this document is not a standalone standard, but is written to be a supplement to other existing pipeline standards for natural gas or liquids for both onshore and offshore pipelines.”
This contribution shall provide information on the content, the current stage of the revision process, encourage to contribute to this standard and make listeners aware to consider the influence of ISO standard on documents currently in draft.
AbstractRed mud is the waste of bauxite refinement into alumina, the feedstock for aluminium production1. With about 180 million tonnes produced per year1, red mud has amassed to one of the largest environmentally hazardous waste products, with the staggering amount of 4 billion tonnes accumulated on a global scale1. Here we present how this red mud can be turned into valuable and sustainable feedstock for ironmaking using fossil-free hydrogen-plasma-based reduction, thus mitigating a part of the steel-related carbon dioxide emissions by making it available for the production of several hundred million tonnes of green steel. The process proceeds through rapid liquid-state reduction, chemical partitioning, as well as density-driven and viscosity-driven separation between metal and oxides. We show the underlying chemical reactions, pH-neutralization processes and phase transformations during this surprisingly simple and fast reduction method. The approach establishes a sustainable toxic-waste treatment from aluminium production through using red mud as feedstock to mitigate greenhouse gas emissions from steelmaking.

AbstractSince the 1950s, Warfarin has been used globally as both a prescription drug and a rodenticide. Research has shown that warfarin and other rodenticides are present in the environment and food chain. However, emerging contaminants are subject to degradation by biotic and abiotic processes and advanced oxidation processes. In some cases, detecting the parent compound may not be possible due to the formation of structurally changed species. This approach aims to identify hydroxylated transformation products of warfarin in a laboratory setting, even after the parent compound has undergone degradation. Therefore, the Fenton reaction is utilized to insert hydroxylation into the parent compound, warfarin, by hydroxyl and hydroperoxyl radicals generated by Fe2+/Fe3+ redox reaction with hydrogen peroxide. Using multiple reaction monitoring, a GC–MS/MS method, incorporating isotopically labeled reference compounds, is used to quantify the expected derivatized species. The analytes are derivatized using trimethyl-3-trifluoromethyl phenyl ammonium hydroxide, and the derivatization yield of warfarin is determined by using isotopically labeled reference compounds. The method has a linear working range of 30 to 1800 ng/mL, with detection limits ranging from 18.7 to 67.0 ng/mL. The analytes are enriched using a C18-SPE step, and the recovery for each compound is calculated. The Fenton reaction generates all preselected hydroxylated transformation products of warfarin. The method successfully identifies that 4′-Me-O-WAR forms preferentially under the specified experimental conditions. By further optimizing the SPE clean-up procedures, this GC–MS-based method will be suitable for detecting transformation products in more complex matrices, such as environmental water samples. Overall, this study provides a better understanding of warfarin’s degradation and offers a robust analytical tool for investigating its transformation products.

A cobalt(III) complex, [Co(L)3](DMF) (1) of Schiff base ligand HL, 2-((E)-(benzylimino)methyl)-4-bromophenol is prepared and single crystal X-ray structural analysis have also been performed. The structures of complex 1 showed hexa-coordinated mononuclear systems that adopt octahedral geometry. The complex has also exhibited the supramolecular networks through non-covalent interactions like H-bonding, C–Hπ stacking. Moreover, the complex 1 is very effective in the catalytic fixation of carbon dioxide in propergyl alcohols to produce α-alkylidene cyclic carbonates. The catalytic production of α-alkylidene cyclic carbonates have been carried out through carboxylative cyclization of propargyl alcohols using CO2 balloon of 1 atm pressure at 80 ◦C. Solvent free condition (green synthesis) made this catalytic protocol eco-friendly towards the environment. Utilizing various substrates of propargyl alcohols moderate to high percentage yield (62–95%) of respective α-alkylidene cyclic carbonates product have been isolated over this catalytic reaction. Besides, the theoretical calculations (DFT) was performed for the prediction of probable mechanism of the catalytic reaction
The European Commission has recently announced two guiding principles for EU product policy: First, product policy shall ensure that the performance of front-runner products in terms of sustainability becomes the norm, and second, the effectiveness of the current Ecodesign legislative framework is going to be significantly improved. Within this paper, already existing front-runner approaches and recent and ongoing product policy-making processes were reviewed. Based on the results, an EU front-runner approach is outlined. The presented approach (i) refers to performance levels of the best products already available on the market, (ii) aggregates information in existing databases, and (iii) works semi-automated. Together, all three attributes have a high potential to facilitate and accelerate the specification of appropriate minimum requirements for products at the EU level. This way, EU policymakers can deliver on the core objectives of the Ecodesign legislative framework much better. The basic mechanism and its legal entrenchment of the approach are illustrated for the energy efficiency of energy-related products. In addition, the Front-Runner Approach can be applied to any product group in the scope of the upcoming Ecodesign for Sustainable Products Regulation and to a wide range of product-related minimum requirements, such as durability, reparability, or recycled content. The study’s objective is to suggest a tailor-made and dynamic approach to keep the EU product legislation up to date using innovative technology based on the investigation of current regulations and identify the gap. Experiences from three international case studies suggest that a front-runner approach to setting energy-performance standards can drive innovation and reduce energy consumption via promoting energy-efficient products; transparency about available products is one of the key factors and can be established by a database. The EU front-runner approach comprises extending the existing energy label database (or making use of the digital product passport) and introducing a legislative procedure that triggers changes in the energy efficiency requirements in the specific EU regulations if the database shows that a certain threshold value is reached. Challenges such as limited EU staff capacities and opportunities such as increased dynamic are discussed.

There are two mirror entries in the European Waste Catalogue for bottom ash and slag from waste incineration: 19 01 11* and 19 01 12. Entry 19 01 11* describes a waste material containing hazardous substances. It was shown that only the hazardous property HP14 (ecotoxic) is of relevance for the classification. If substances with the hazard statement code (HSC) H400, H410, H411, H412 and H413 are present in incineration bottom ash (IBA) above certain level the waste is classified as hazardous, otherwise it is non-hazardous (in this case 19 01 12). It was further shown that only Cu, Zn and Pb compounds have an impact on the classification using a summation rule: 100xΣ c(H410) + 10xΣ c(H411) + Σ c(H412). Limit value is 25 percent. It is obvious that H410 substances have the highest impact. Therefore, the knowledge of the elemental composition of IBA is not sufficient because the content of distinct chemical compounds (with the respective molecular weight) is needed. When the exact speciation is not known a worst-case approach has to be applied. A practical guideline for the estimation of HP14 was developed based on the definition of 4 substance groups: 1. Metals and alloys, 2. Water-soluble substances, 3. Oxides and carbonates and 4. Substances not soluble in weak acids. In substance group 1 only fine Zn powder has the HSC H410. Water-soluble substances are assumed to be H410 substances. Their concentrations are, however, regularly below the cut-off value of 0.1 %. The amount of substance group 3 is estimated by a selective extraction with maleic acid. According to the worst-case approach it is assumed that CuCO3xCu(OH)2 and ZnO represent the respective Cu and Zn species in IBA because these substances have the highest stochiometric factor. Even with this worst-case approach results from operators of IBA treatment plant showed that the limit value of 25 % is not exceeded.
The requirements for utilization of IBA in Germany are defined in the Secondary
Building Materials Decree (Ersatzbaustoffverordnung EBV) which was set into force in August 2023. Leaching methods (batch test or column test) with a liquid-to-solid ratio of 2 l/kg are applied. IBA fulfills the requirements for class HMVA-2. Actually, even a classification of IBA as hazardous (i.e 19 01 11*) would not affect utilization but handling and transport.
Here, we show that calcium sulfate dihydrate (gypsum) can be directly, rapidly and reversibly converted to calcium sulfate hemihydrate (bassanite) in high salinity solutions (brines). The optimum conditions for the efficient production of bassanite in a short time (<5 min) involve the use of brines with c(NaCl) > 4 M and maintaining a temperature, T > 80 °C. When the solution containing bassanite crystals is cooled down to around room temperature, eventually gypsum is formed. When the temperature is raised again to T > 80 °C, bassanite is rapidly re-precipitated. This contrasts with the better-known behaviour of the bassanite phase in low-salt environments. In low-salinity aqueous solutions, bassanite is considered to be metastable with respect to gypsum and anhydrite, and therefore gypsum-to-bassanite conversion does not occur in pure water. Interestingly, the high-salinity transformation of gypsum-to-bassanite has been reported by many authors and used in practice for several decades, although its very occurrence actually contradicts numerical thermodynamic predictions regarding solubility of calcium sulfate phases. By following the evolution of crystalline phases with in situ and time-resolved X-ray diffraction/scattering and Raman spectroscopy, we demonstrated that the phase stability in brines at elevated temperatures was inaccurately represented in the thermodynamic databases. Most notably for c(NaCl) > 4 M, and T > 80 °C gypsum becomes readily more soluble than bassanite, which induces the direct precipitation of the latter from gypsum. The fact that these transformations are controlled by the solution provides extensive opportunities for precise manipulation of crystal formation. Our experiments confirmed that bassanite remained the sole crystalline phase for many hours before reverting into gypsum. This property is extremely advantageous for practical processing and efficient crystal extraction in industrial scenarios.

Fused cement clinker can be produced from molten basic oxygen furnace slag (BOFS) by way of a reductive thermochemical treatment. During the thermochemical treatment, oxidic iron is reduced to metallic iron and separated. The resulting low-iron slag has a chemical and mineralogical composition similar to ordinary Portland cement (OPC) clinker. In this study, the hydraulic reactivity of the fused clinker from BOFS with and without gypsum was investigated using isothermal calorimetry, differential scanning calorimetry, in situ X-ray diffraction and powder X-ray diffraction. Furthermore, a synthetic fused clinker without foreign ions and fused clinker produced by a mixture of both materials was studied. The hydraulic reaction of the fused clinker from BOFS was considerably slower than that of OPC. However, the reaction can be accelerated by adding gypsum as a sulfate carrier. Furthermore, the results showed an increased reaction rate with decreasing content of foreign ions such as Fe, P or Mn.
Acetogenic bacteria produce CO2-based chemicals in aqueous media by hydrogenotrophic conversion of CO2, but CO is the preferred carbon and electron source. Consequently, coupling CO2 electrolysis with bacterial fermentation within an integrated bio-electrocatalytical system (BES) is promising, if CO2 reduction catalysts are available for the generation of CO in the complex biotic electrolyte. A standard stirred-tank bioreactor was coupled to a zero-gap PEM electrolysis cell for CO2 conversion, allowing voltage control and separation of the anode in one single cell. The cathodic CO2 reduction and the competing hydrogen evolution enabled in-situ feeding of C. ragsdalei with CO and H2. Proof-of-concept was demonstrated in first batch processes with continuous CO2 gassing, as autotrophic growth and acetate formation was observed in the stirred BES in a voltage range of −2.4 to −3.0 V. The setup is suitable also for other bioelectrocatalytic reactions. Increased currents and lower overvoltages are however required. Atomically-dispersed M−N−C catalysts show promise, if degradation throughout autoclaving can be omitted. The development of selective and autoclavable catalysts resistant to contamination and electrode design for the complex electrolyte will enable efficient bioelectrocatalytic power-to-X systems based on the introduced BES.
Here, we report a comparative study of different sum parameter analysis methods for the extraction of per- and polyfluoroalkyl substances (PFAS) from manufactured consumer products, which can be measured by combustion ion chromatography (CIC). Therefore, a hydrolysis-based extraction method was further developed, which accounts for the addition of hydrolyzable covalently bound polyfluoroalkylated side-chain polymers (SFPs) to the extractable organic fluorine portion of the mass balance proposed as "hydrolyzable organically bound fluorine" (HOF). To test this hypothesis, the method was applied to 39 different consumer products containing fluoropolymers or monomeric PFAS taken from four different categories: outdoor textiles, paper packaging, carpeting, and permanent baking sheets. We also evaluated the method's efficiency by extracting four synthesized fluorotelomer polyacrylate reference compounds. The total fluorine (TF) and extractable organically bound fluorine (EOF) values were measured through CIC using established protocols. The TF values ranged from sub-ppb to %-levels, depending on the compound class. All samples showed results for hydrolyzed organofluorine (HOF) between 0.03 and 76.3 μg/g, while most EOF values were lower (<LOD to 58.1 μg/g). Moreover, the methods were contrasted with the "total hydrolyzable precursor" (THP) assay analyzed using gas-chromatography coupled mass spectrometry (GC-MS), which yielded concentrations of 6:2–10:2 fluorotelomer alcohols (<LOD-568 μg/g) for textile and paper samples. Furthermore, targeted analytical investigation of 25 compounds via liquid chromatography coupled tandem mass spectrometry (LC-MS/MS) was conducted and accounted for significantly lower sum amounts (<LOD-0.98 μg PFAS/g). To estimate the fluorine mass balance of all methods, the fluorine levels of target analytical data were calculated and compared to the sum parameter values and evaluated. Finally, the efficiency of the extraction method was approximated by the recovery analysis of four selected SFP model compounds compared to their TF values, yielding 36.7–74.0% after extraction and 3.8–36.3% after processing the extract for the HOF sum parameter.

Calcium sulfate hemihydrate (CaSO4ᐧ0.5H2O), also known as bassanite, has been used as a precursor to produce gypsum (dihydrate, CaSO4ᐧ2H2O) for various construction and decorative purposes since prehistoric times. The main route to obtain hemihydrate is a thermal treatment of gypsum at temperatures typically between 150 °C and 200 °C to remove some of the structural water.
In this contribution, we introduce (Fig. 1) a more efficient and sustainable method (T < 100 °C) that enables the direct, rapid, and reversibly conversion of gypsum to bassanite using reusable high salinity aqueous solutions (brines with c[NaCl] > 4 M). The optimum conditions for the efficientproduction of bassanite in a short time (< 5 min) involve the use of brines with c(NaCl) > 4 M and maintaining a temperature, T > 80 °C. When the solution containing bassanite crystals is cooled down to around room temperature, eventually gypsum is formed. When the temperature is raised again to T > 80 °C, bassanite is rapidly re-precipitated. This contrasts with the typical behaviour of the bassanite phase in low salt environments.
Traditionally, hemihydrate is obtained through a solid state thermal treatment because bassanite is considered to be metastable with respect to gypsum and anhydrite in aqueous solutions, and therefore gypsum-to-bassanite conversion should not occur in water. Its very occurrence actually contradicts numerical thermodynamic predictions regarding solubility of calcium sulfate phases. By following the evolution of crystalline phases with in situ and time-resolved X-ray diffraction/scattering and Raman spectroscopy, we demonstrated that the phase stability in brines at elevated temperatures is inaccurately represented in the thermodynamic databases. Our experiments confirmed that bassanite remained the sole crystalline structure for many hours before reverting into gypsum. This property is extremely advantageous for practical processing and efficient crystal extraction in industrial scenarios.
Knowledge representation in the Materials Science and Engineering (MSE) domain is a vast and multi-faceted challenge: Overlap, ambiguity, and inconsistency in terminology are common. Invariant (consistent) and variant (context-specific) knowledge are difficult to align cross-domain. Generic top-level semantic terminology often is too abstract, while MSE domain terminology often is too specific. In this paper, an approach how to maintain a comprehensive MSE-centric terminology composing a mid-level ontology–the Platform MaterialDigital Core Ontology (PMDco)–via MSE community-based curation procedures is presented. The illustrated findings show how the PMDco bridges semantic gaps between high-level, MSE-specific, and other science domain semantics. Additionally, it demonstrates how the PMDco lowers development and integration thresholds. Moreover, the research highlights how to fuel it with real-world data sources ranging from manually conducted experiments and simulations with continuously automated industrial applications.
Re-melting of scrap in an electric arc furnace (EAF) results in the accumulation of filter dust from off-gas treatment that predominantly consists of iron and zinc oxides. Filter dust is classified as hazardous waste due to its high contents of potentially toxic or ecotoxic elements such as Pb, Cr, Cd, and As. A promising processing route for this waste is selective chlorination, in which the non-ferrous metal oxides are chlorinated and selectively evaporated in form of their respective chlorides from the remaining solids via the process gas flow. Here, we investigate stepwise thermochemical treatment of EAF dust with either waste iron(II) chloride solution or hydrochloric acid at 650, 800, and 1100 ◦C. The Zn and Pb contents of the thermochemically processed EAF dust could be lowered from 29.9% and 1.63% to 0.09% and 0.004%, respectively. Stepwise heating allowed high separation between zinc chloride at the 650 ◦C step and sodium-, potassium-, and lead-containing chlorides at higher temperatures. Furthermore, the lab-scale results were transferred to the use of an experimental rotary kiln highlighting the possibilities of upscaling the presented process. Selective chlorination of EAF dust with liquid chlorine donors is, therefore, suggested as a potential recycling method for Zn-enriched steelworks dusts.
This contribution reports the development of a polymerizable BODIPY-type fluorescent probe targeting small-molecule carboxylates for incorporation into molecularly imprinted polymers (MIPs). The design of the probe crosslinker includes a urea recognition site p-conjugated to the 3-position of the BODIPY core and two methacrylate moieties. Titration experiments with a carboxylate-expressing antibiotic, levofloxacin (LEVO), showed a blue shift of the absorption band as well as a broadening and decrease in emission, attributed to hydrogen bonding between the probe’s urea group and the carboxylate group of the antibiotic. Using this probe crosslinker, core–shell particles with a silica core and a thin MIP shell were prepared for the detection of LEVO. The MIP exhibited highly selective recognition of LEVO, with an imprinting factor of 18.1 compared to the non-imprinted polymer. Transmission electron microscopy confirmed the core–shell structure and spectroscopic studies revealed that the receptor’s positioning leads to a unique perturbation of the polymethinic character of the BODIPY chromophore, entailing the favourable responses. These features are fully preserved in the MIP, whereas no such response was observed for competitors such as ampicillin. The sensory particles allowed to detect LEVO down to submicromolar concentrations in dioxane. We have developed here for the first time a BODIPY probe for organic carboxylates and incorporated it into polymers using the imprinting technique, paving the way for BODIPY-type fluorescent MIP sensors.
The emission of ultrafine particles from small desktop Fused Filament Fabrication (FFF) 3D printers has been frequently investigated in the past years. However, the vast majority of FFF emission and exposure studies have not considered the possible occurrence of particles below the typical detection limit of Condensation Particle Counters and could have systematically underestimated the total particle emission as well as the related exposure risks. Therefore, we comparatively measured particle number concentrations and size distributions of sub-4 nm particles with two commercially available diethylene glycol-based instruments – the TSI 3757 Nano Enhancer and the Airmodus A10 Particle Size Magnifier. Both instruments were evaluated for their suitability of measuring FFF-3D printing emissions in the sub-4 nm size range while operated as a particle counter or as a particle size spectrometer. For particle counting, both instruments match best when the Airmodus system was adjusted to a cut-off of 1.5 nm. For size spectroscopy, both instruments show limitations due to either the fast dynamics or rather low levels of particle emissions from FFF-3D printing in this range. The effects are discussed in detail in this article. The findings could be used to implement sub-4 nm particle measurement in future emission or exposure studies, but also for the development of standard test protocols for FFF-3D printing emissions.
There is only sparse empirical data on the settling velocity of small, non-buoyant microplastics thus far, although it is an important parameter governing their vertical transport within aquatic environments. This study reports the settling velocities of 4031 exemplary microplastic particles. Focusing on the environmentally most prevalent particle shapes, irregular microplastic fragments of four different polymer types (9–289 µm) as well as five discrete length fractions (50–600 µm) of common nylon and polyester fibers were investigated, respectively. All settling experiments were carried out in quiescent water using a specialized optical imaging setup. The method has been previously validated in order to minimize disruptive factors, e.g. thermal convection or particle interactions, and thus enable the precise measurements of the velocities of individual microplastic particles (0.003–9.094 mm/s). Based on the obtained data, ten existing models for predicting a particle’s terminal settling velocity were assessed. It is concluded that models, which were specifically deduced from empirical data on larger microplastics, fail to provide accurate predictions for small microplastics. Instead, a different approach is highlighted as a viable option for computing settling velocities across the microplastics continuum in terms of size, density and shape.
The energy demand of private households contributes globally to 36.5% of the total CO2 emissions. To analyze the emissions reduction potential, we conducted a comparative life cycle assessment of a proton exchange membrane fuel cell in a residential application and a conventional system with a stand-alone gas condensing boiler and electricity from a grid mix. The period under review was referred to as the service life of the PEMFC and is assumed to be 10 years (83,038 h of PEMFC). The applicability of this in a single-family house built between 1991 and 2000 under German climatic conditions was investigated. The functional unit is set to the thermal energy demand of 16,244 kWh/a and electricity demand of 4919 kWh/a of a single-family house. The impact assessment method “CML 2001–August 2016” was used in this investigation. The manufacturing phase of the proton exchange membrane fuel cell showed disadvantages, whereby the use phase had significant advantages in most of the environmental impact categories as compared to the conventional energy supply system. Considering the whole life cycle, the advantages from the use phase could outperform the disadvantages from the manufacturing phase in most of the impact categories, except for ADP elements and TETP.
The plastic value chain, central part of modern living, caused environmental pollution and bioaccumulation of plastic nanoparticles (PNPs). Their ubiquitous presence in different environmental and biological compartments has become a serious threat to human health and ecosystems. Frequently used plastic materials such as polypropylene (PP), polystyrene (PS) and polyethylene (PE) have been detected in the form of PNPs in the food chain, soil, water and air, as well as in human feces and blood. In this study, we aimed to provide novel insights in endocrine disrupting properties of PNPs using in vitro estrogen receptor (ER) transactivation assay. The effects of PP-NPs, PE-NPs and PS-NPs and their mixture on T47D-KBluc cell line stably transfected with luciferase as reporter enzyme was evaluated by means of cytotoxicity, cellular uptake and ER activation. Tested dose range for PNPs was 0.001 – 10 mg/L. Both cellular uptake and cytotoxicity for all PNPs was found to be dose-dependent. Only the highest dose of PP-NPs and PE-NPs induced apoptosis and cell death, while PS-NPs were not cytotoxic in tested dose range. For tested concentrations, PP-NPs and PE-NPs showed significant agonistic activity on ER, while PS-NPs cannot be considered ER active. When, applied as mixture, PNP demonstrated additive toxicity effects compared to the effect of each individual PNPs. Additivity was also observed for ER agonistic effect of PNPs mixture according to the benchmark dose-addition modelling approach. This study provides missing science-based evidence on endocrine disrupting effects of PE-NPs, PP-NPs, PS-NPs and their mixtures and highlights the importance of considering unintentional, aggregate and combined exposure to different PNPs in risk management.
Advances in ultrafast laser manufacturing: nanostructures, thin films, and scaling perspectives
(2024)
Advanced ultrafast laser technology is a rapidly growing field that currently enables many new industrial and scientific applications. During the last decades, this has been significantly driven by the availability of high-repetition-rate laser sources and novel beam delivery concepts. At the laser side, Moore’s law equally manifests for ultrafast laser technologies, since the average output power of such lasers doubles approximately every two years. This development is mainly driven by the increase of the pulse repetition rates of energetic laser pulses, currently enforcing the development of smart beam control and novel scanning strategies for preventing heat-accumulation and plasma-shielding effects during laser-based materials processing. This keynote presentation addresses the advantages, recent developments, and perspectives of laser processing with ultrashort laser pulses. A special focus is laid on the tailored structuring of thin films as well as the manufacturing and probing of sub-diffraction surface nanostructures – an ongoing race to extreme scales. Current limitations are identified and an outlook to future scaling perspectives will be provided.

Per- and polyfluoroalkyl substances (PFAS) have received global public attention because of their wide distribution in aquatic environments and potential adverse effects to humans and wildlife. Due to the lack of analytical standards and the enormous numbers of these compounds, current target-based methods (e.g., LC-MS/MS) are not suitable for the analysis of new/unknown PFAS and transformation products. Therefore, PFAS sum parameter analysis is becoming increasingly important. For PFAS determination of surface waters, solid phase extraction (SPE) is commonly implemented for clean-up and pre-concentration of samples.
Thus, within this work, single-layer SPE methods (based on Strata™-X, Strata™-X-AW, Strata™-NH2 and Oasis-HLB) were investigated for maximum PFAS coverage utilizing extractable organically bound fluorine (EOF) analysis. Thereby, the optimization procedure relied on the analysis of 3 surface water samples in Berlin, Germany, which were affected by the effluent discharge of wastewater treatment plants. To analyse the SPE elution profiles for EOF and inorganic fluorine, high resolution-continuum source-graphite furnace molecular absorption spectroscopy (HR-CS-GFMAS) and ion chromatography (IC) were used, respectively.
Highest EOF concentrations were achieved by using Strata-X/Strata-XAW as SPE sorbents and methanol as eluent. Furthermore, combinations of the most promising SPE sorbents were selected (X/XAW, XAW/X, HLB/X and WAX/GCB) to extract a wider range of PFASs. By comparing multi-layer SPE methods, lower EOF concentrations were obtained for all investigated combination phases compared to the analysed single phases. For the commercially available combination phase Strata-PFAS (WAX/GCB) for all three river water samples, lower EOF concentrations (1.5 times lower) were determined compared to Strata-X.
The results have shown that single-layer SPE systems are currently superior compared to combination phases for PFAS sum parameter analysis. The multi-layer SPE methods need further optimization regarding appropriate sorbent combinations, loading volumes and elution conditions. The developed single-layer SPE methods can help to elucidate pollutions hotspots and discharge routes.
PFAS (Per- and polyfluoroalkyl substances) are a widespread and emerging environmental problem due to their longevity and ubiquitous spread. The common structural motif of this compound class is the fluorine – carbon bond on an alkyl backbone, which are known for their chemical inertness and combination of hydrophobicity and oleophobicity. The downside of those properties is the general lack of usable molecular interactions for adsorption to extract them from the environment. The notable exception to that inertness are fluorine – fluorine interactions, which are stronger, the longer the perfluorinated chain length is. Therefore, in this project we investigated the effect of fluorination, length and presence of a charge on the alkyl chain on an adsorber. Towards that goal, in total 16 different adsorber materials were synthesized (fluorinated/non-fluorinated; C4/ C6/ C8/ C10 ; positive charge/no charge) and tested in regard to their potential to adsorb PFOA and perfluoralkylic acids of differing chain lengths. The PFAS concentration was tested via LC-MS/MS to determine the adsorption rate on the various materials. Further work includes the transition from a target analytics to sum parameter analysis for future evaluation of adsorber materials in PFAS environmental samples like river samples. Therefore a quarterly sampling and extraction of spree surface water samples was planned.

The present work demonstrates a sustainable approach of using relatively coarser iron ore particles for ironmaking. The motivation is to reduce the energy consumption in the milling of the iron ore by utilizing coarser iron ore particles (+0.05 mm) and to select a suitable binder for improving pellet properties. Iron ore fines in the range of 0.05–0.25 mm was selected and classified into three size ranges. Fluxed iron ore pellets were prepared using lime as a binder for the basicity of 0, 1, and 2. Reduction of these pellets with a packed bed of coal fines was performed in the temperature range of 900–1200 °C for a duration of 30–120 min. The direct reduction kinetics of the iron ore pellets were studied by employing diffusion and chemical reaction control models to the experimental data. The results show that pellets made with coarser iron ore particles have improved reduction behavior and kinetics. The reduction reaction is found to be a mixed control. The activation energy for the reduction reaction varies from 44.3 to 74.76 kJ mol−1 as iron ore particle size decreases from 0.25 to 0.05 mm and basicity increases from 0 to 2.

Homochirality is an obvious feature of life on Earth. On the other hand, extraterrestrial samples contain largely racemic compounds. The same is true for any common organic synthesis. Therefore, it has been a perplexing puzzle for decades how these racemates could have formed enantiomerically enriched fractions as a basis for the origin of homochiral life forms. Numerous hypotheses have been put forward as to how preferentially homochiral molecules could have formed and accumulated on Earth. In this article, it is shown that homochirality of the abiotic organic pool at the time of formation of the first self-replicating molecules is not necessary and not even probable. It is proposed to abandon the notion of a molecular ensemble and to focus on the level of individual molecules. Although the formation of the first self-replicating, most likely homochiral molecule, is a seemingly improbable event, on a closer look, it is almost inevitable that some homochiral molecules have formed simply on a statistical basis. In this case, the non-selective leap to homochirality would be one of the first steps in chemical evolution directly out of a racemic “ocean”. Moreover, most studies focus on the chirality of the primordial monomers with respect to an asymmetric carbon atom. However, any polymer with a minimal size that allows folding to a secondary structure would spontaneously lead to asymmetric higher structures (conformations). Most of the functions of these polymers would be influenced by this inherently asymmetric folding. Furthermore, a concept of physical compartmentalization based on rock nanopores in analogy to nanocavities of digital immunoassays is introduced to suggest that complex cell walls or membranes were also not required for the first steps of chemical evolution. To summarize, simple and universal mechanisms may have led to homochiral self-replicating systems in the context of chemical evolution. A homochiral monomer pool is deemed unnecessary and probably never existed on primordial Earth.

Interlaboratory studies are common tools for collecting comparable data to implement standards for new materials or testing technologies. In the case of construction materials, these studies form the basis for recommendations and design codes. Depending on the study, the amount of data collected can be enormous, making manual handling and evaluation difficult. On the other hand, the importance of the FAIR (findable, accessible, interoperable, and reusable) principles for scientific data management, published by Wilkinson et al. in 2016, is constantly growing and changing the view on data usage.
The benefits of using data management tools such as data stores/repositories or electronic laboratory notebooks are many. Data is stored in a structured and accessible way (at least within a group) and data loss due to staff turnover is reduced. Tools usually support data publishing and analysis interfaces. In this way, data can be reused years later to generate new knowledge with future insights. On the other hand, there are many challenges in setting up a data repository, such as selecting suitable software tools, defining the data structure, enabling data access, and understanding by others and
ensuring maintenance, among others.
This talk discusses the advantages and challenges of setting up and applying a data repository using the interlaboratory study on the mechanical properties of printed concrete structures carried out in RILEM TC 304-ADC as example. First, the definition of a suitable data structure including all information is discussed. The tool-dependent upload process is then described. Here, the data
management system openBIS (open source software developed by ETH Zurich) is used. Since in most cases an open compute platform allowing access from different organisations is not possible or available due to data protection and maintenance issues, tool-independent export options are discussed and compared. Finally, the different query and analysis possibilities are demonstrated.

The plant secondary metabolite families of coumarin and 4-hydroxy coumarin have a broad pharmacological spectrum ranging from antibacterial to anticancer properties. One prominent member of this substance class is the synthetic but naturally inspired anticoagulant drug and rodenticide warfarin (coumadin). A vast number of publications focus on the identification of warfarin and its major cytochrome P450-mediated phase I metabolites by liquid chromatography (LC) with mass spectrometry (MS) and tandem mass spectrometric (MS/MS) detection techniques. For the first time, electron ionization (EI) induced high-resolution quadrupole time-of-flight mass spectrometric (HR-qToF-MS) data of in-liner derivatized warfarin and selected hydroxylated species is provided in this study as an alternative to LC-MS/MS approaches. Furthermore, the characteristic fragments and fragmentation pathways of the analyzed methyl ethers are concluded. The obtained data of analytical standards, specific deuterated and 13C-labeled compounds prove inductive cleavage of the acyl or acetonyl side chain, methyl migration, and H-migration, along with consequential inductive cleavage as predominant fragmentation routes. Based on the HR-spectral data, commonalities and differences between the analyzed compounds and fragment groups were evaluated with future applicability in structure elucidation and spectra prediction of related compounds.
Organotin compounds (OTCs) have been widely used in anti-fouling paints, pesticide formulations, and as stabilizers in polyvinyl chloride over the past century. In marine ecosystems, OTCs can cause severe damage to biodiversity, leading up to the extinction of vulnerable species. Due to the extensive use of tributyltin (TBT) as a biocide on ship hulls, it has been considered one of the most hazardous substances intentionally introduced into the aquatic environment. This resulted in a global ban on TBT-containing products in the 2000s. However, recent studies indicate the emerging presence of organotin pollutants.
OTCs are known to persist and accumulate in marine sediments, posing a long-term threat to the environment. These harmful substances can be set free and dispersed even after several decades. Therefore, analyzing sediment probes is imperative for a thorough monitoring of pollution. However, species-specific analysis of OTCs at required concentration levels in complex environmental matrices remains challenging. Chromatographic systems are commonly used for their analysis, but the required sample preparation is time-consuming and prone to contamination and analyte loss. The coupling of electrothermal vaporization and inductively coupled plasma-mass spectrometry (ETV/ICP-MS) demonstrates high potential as a rapid, convenient, and chemical-saving scanning tool for environmental samples.
This method enables the direct on-line fractionation of organic compounds from an inorganic fraction and provides element-specific detection at ultra-trace levels without complex sample preparation. Since OTCs are generally more toxic than ionic or elemental tin, analyzing them as a sum parameter is advantageous. Additionally, the determination of both organic and inorganic tin, rather than just organic tin, reveals valuable information about the fate of OTCs. The main challenge in obtaining accurate quantitative data using direct solid sampling techniques like as ETV/ICP-MS is applying a suitable calibration strategy. Our isotope dilution approach overcomes matrix effects in ETV/ICP-MS analysis and is compatible with commercial systems.
The environmental fate of the frequently used broad-spectrum β-lactam antibiotic amoxicillin (AMX) is of high concern regarding the potential evolution of antimicrobial resistance (AMR). Moreover, it is known that AMX is prone to hydrolysis, yielding a variety of hydrolysis products (HPs) with yet unknown effects. Studies to identify those HPs and investigate their formation mechanisms have been reported but a long-term study on their stability in real water samples was missing. In this regard, we investigated the hydrolysis of AMX at two concentration levels in four distinct water types under three different storage conditions over two months. Concentrations of AMX and four relevant HPs were monitored by an LC-MS/MS method revealing pronounced differences in the hydrolysis rate of AMX in tap water and mineral water on the one hand (fast) and surface water on the other(slow). In this context, the occurrence, relative intensities, and stability of certain HPs are more dependent on the water type than on the storage condition. As clarified by ICP-MS, the main difference between the water types was the content of the metals copper and zinc which are supposed to catalyze AMX hydrolysis demonstrating an effective method to degrade AMX at ambient conditions.
Simulating a Gas Source Localization Algorithm with Gas Dispersion Produced by Recorded Outdoor Wind
(2023)
This paper reports the use of the first gas dispersion simulator capable of introducing large wind fluctuations into simulations. The proposed simulator enables testing of a modification made to a gas source localization algorithm in a realistic scenario in order to study how the change affects it. Gas source localization in an outdoor environment is a challenging task mainly due to the complexity of the gas spread caused by the unpredictable nature of constantly changing wind. Therefore, a novel use of outdoor wind in developing a gas source localization system by simulation is presented in this paper. To consider the characteristic of sudden but large and unpredictable changes in wind direction, we propose to use recorded outdoor wind to simulate a realistic outdoor gas dispersion which has been done for the first time to the best of our knowledge. With the use of this simulator, we have tested a modification to a mobile robot-based gas source localization algorithm. Multiple simulations of the modified and the original particle filter-based algorithm have been done to study the effect of the tested modification. The results showed that a small difference in the algorithm can greatly impact the results. From this study, we show that the use of simulation consisting of the necessary traits to evaluate outdoor gas source localization, has the potential to accelerate the development of a reliable localization system.
The ISO 16000-6 standard gives directions to adapt the analysis of volatile organic compounds (VOCs) in indoor and test chamber air to very volatile organic compounds (VVOCs). The same techniques with sorbent-based active sampling, thermal desorption and gas chromatography coupled with mass spectrometry (TD-GC/MS) should be used. However, VVOCs require gaseous standards, an adapted GC column and a reliable sampling adsorbent. This work presents experimental results to tackle those three experimental gaps. A stable standard gas mixture containing 47 VVOCs, 13 VOCs and an internal standard was successfully generated. It was employed to study the suitability of seven types of chromatography columns. The use of PLOT (Porous Layer Open Tubular) columns such as PoraBOND Q is well suitable for VVOC analysis. The recoveries of the 60 analytes on a total of 16 adsorbents and their combinations were determined: A combination of the graphitized carbon black Carbograph 5TD 40/60 and the carbon molecular sieve CarbosieveTM SII showed great recoveries for all analytes. Carbon molecular sieves adsorb water which can impair the analysis. A dry purge of the multi-sorbent in the sampling direction led to a complete water removal and promising recoveries of the analytes.
The fungus Botrytis cinerea (Botryotinia fuckeliana) infects more than 500 plant species and causes a wide range of symptoms: soft rots, accompanied by collapse and water-soaking of tissues followed by the appearance of gray masses of conidia on leaves and soft fruits (gray mold), and spots that may turn brown to full-scale soft rotting on flower petals (Botrytis blight). In general, B. cinerea is responsible for severe economic losses that are either due to the damage of growing plants in the field or the rot of harvested fruits, flowers, and vegetables during storage under cold and humid conditions. B. cinerea has adapted to the plant host and its environment by evolving strategies to use plant tissues for proliferation in terms of a necrotrophic lifestyle, and to survive biotic stresses (host responses) as well as abiotic factors of the host’s environment such as sunlight and concomitant stresses. B. cinerea maintains a complex regulatory network of light-sensitive proteins and signal transduction pathways to use light for coordinating stress responses, virulence, and reproduction. Different light-controlled reproduction cycles enable B. cinerea to live in moderate climate zones by infecting and propagating in summer and resting in winter when green host tissues are unavailable.
Remote gas sensors mounted on mobile robots enable the mapping of gas distributions in large or poorly accessible areas. A challenging task however, is the generation of three-dimensional distribution maps from these spatially sparse gas measurements. To obtain high-quality reconstructions, the choice of optimal measuring poses is of great importance. Remote gas sensors, that are commonly used in Robot Assisted Gas Tomography (RAGT), require reflecting surfaces within the sensor’s range, limiting the possible sensing geometries, regardless of whether the robots are ground-based or airborne. By combining ground and aerial robots into a heterogeneous swarm whose agents are equipped with reflectors and remote gas sensors, remote inter-robot gas measurements become available, taking RAGT to the next dimension – releasing those constraints. In this paper, we demonstrate the feasibility of drone-to-drone measurements under realistic conditions and highlight the resulting opportunities.
The chemical state of antimony and vanadium species in municipal solid waste incineration bottom ash
(2023)
Due to the large quantity as residual mineral waste, municipal solid waste incineration - bottom ash (MSWI-BA) is an interesting secondary raw material that can be utilized for road construction. However, leaching of chloride, sulfate and potentially hazardous heavy metals from MSWSI-BA into the environment may cause problems in utilization of it in civil engineering. In a previous study, we performed a long-term leaching test of MSWI-BA in a lysimeter for almost six years to investigate the efficiency of the treatment process on the release of hazardous substances. While concentrations of chloride, sulfate and the majority of the heavy metals started to decrease rapidly with progressive liquid-to-solid ratio (L/S), the ecotoxic hazardous elements antimony (Sb) and vanadium (V) behaved differently. To unravel these unusual release behaviors and the oxyanion-formation of Sb and V we will apply HERFD-XANES spectroscopy to analyze their chemical state.

Iron is arguably the most essential metal in living organisms. For rock-inhabiting fungi its acquisition might be unconventional as they (1) tend to inhabit iron-deficient, oxygen-rich surfaces like marble monuments and solar panels and (2) produce the black, iron-adsorbing pigment melanin. We used a range of analytical methods, ongoing mineral dissolution experiments and gene deletion mutants of the model rock-inhabiting fungus Knufia petricola to figure out the mechanisms and substrate deteriorating effects of iron uptake by these organisms. To study both siderophore-mediated and reductive iron assimilation (RIA), genes like sidC, encoding a putative siderophore synthetase and ftr1 and fet3 encoding the subunits of an iron permease-oxidase were deleted.
At iron deficient conditions, growth of the wild type (WT) and ΔsidC mutant was similar, whereas growth of the Δftr1-fet3 mutant and the double mutant ΔsidC/Δftr1-fet3 was diminished and absent, respectively. We were not able to detect the siderophore of K. petricola and the WT and mutants were not able to grow at low concentrations of strong iron chelators. However, in a cross-feeding experiment, an overexpression strain of sidC allowed more growth of ΔsidC/Δftr1-fet3 on iron deficient medium than the WT, whereas the ΔsidC mutant could not do so at all. Compared to the WT, the sidC overexpression strain also withstood oxidative stress better and had a shorter lag time and higher growth rate. Combined, these results indicate that K. petricola relies more on RIA than siderophore-mediated uptake as it likely excretes low quantities of a primarily intracellular siderophore. Interestingly, Δftr1-fet3 had a higher iron content than the WT at iron deficient conditions. This difference disappeared upon deletion of melanin synthesis (Δpks1 vs. Δpks1/Δftr1-fet3): melanin-bound iron can likely not be used without RIA. K. petricola’s chelation incapacity implies a habitat free of competition for iron while offering us a mitigation strategy.
Small-angle X-ray scattering (SAXS) can be used for structural determination of biological macromolecules and polymers in their native states (e.g. liquid phase). This means that the structural changes of (bio-)polymers, such as proteins and DNA, can be monitored in situ to understand their sensitivity to changes in chemical environments. In an attempt to improve the reliability of such experiments, the reduction of radiation damage occurring from exposure to X-rays is required. One such method, is to use scavenger molecules to protect macromolecules against radicals produced during radiation exposure, such as reactive oxygen species (ROS). In this study we investigate the feasibility of applying the compatible solute, osmolyte and radiation protector Ectoine (THP(B)), as a scavenger molecule during SAXS measurements of the single-stranded DNA-binding protein Gene-V Protein (G5P/GVP). In this case, we monitor the radiation induced changes of G5P during bio-SAXS measurments and the resulting microscopic energy-damage relation was determined from microdosimetric calculations by Monte-Carlo based particle scattering simulations with TOPAS/Geant4 and a custom target-model. This resulted in a median-lethal energy deposit of pure G5P at 4 mg mL−1 of E1/2 = 7 ± 5 eV, whereas a threefold increase of energy-deposit was needed under the presence of Ectoine to reach the same level of damage. This indicates that Ectoine increases the possible exposure time before radiation-damage to G5P is observed. Furthermore, the dominant type of damage shifted from aggregation in pure solutions towards a fragmentation for solutions containing Ectoine as a cosolute. These results are interpreted in terms of indirect radiation damage by reactive secondary species, as well as post-irradiation effects, related to preferential-exclusion of the cosolute from the protein surface. Hence, Ectoine is shown to provide a non-disturbing way to improve structure-determination of proteins via bio-SAXS in future studies.
In construction and demolition waste (CDW) recycling, the preference to date has been to apply simple but proven techniques to sort and process large quantities of construction rubble in a short time. This contrasts with the increasingly complex composite materials and structures in the mineral building materials industry. An automated, sensor-based sorting of these building materials could complement or replace the practice of manual sorting to improve processing speed, recycling rates, sorting quality, and prevailing health conditions for the executing staff.
A joint project of partners from industry and research institutions approaches this task by investigating and testing the combination of laser-induced breakdown spectroscopy (LIBS) with near-infrared (NIR) spectroscopy and visual imaging. Joint processing of information (data fusion) is expected to significantly improve the sorting quality of CDW, and may enable the detection and separation of impurities such as SO3-cotaining building materials (gypsum, aerated concrete, etc.)
We present current advances and results about the methodological development combining LIBS with NIR spectroscopy and visual imaging. Here, applying data fusion proves itself beneficial to improve recognition rates. In the future, a laboratory prototype will serve as a fully automated measurement setup to allow real-time classification of CDW on a conveyor belt.
Bacterial biofilms can pose a serious health risk to humans and are less susceptible
to antibiotics and disinfection than planktonic bacteria. Here, a novel method
for biofilm eradication based on antimicrobial photodynamic therapy utilizing
a nanoparticle in conjunction with a BODIPY derivative as photosensitizer was
developed. Reactive oxygen species are generated upon illumination with
visible light and lead to a strong, controllable and persistent eradication of
both planktonic bacteria and biofilms. One of the biggest challenges in biofilm
eradication is the penetration of the antimicrobial agent into the biofilm and
its matrix. A biocompatible hydrophilic nanoparticle was utilized as a delivery
system for the hydrophobic BODIPY dye and enabled its accumulation within the
biofilm. This key feature of delivering the antimicrobial agent to the site of action
where it is activated resulted in effective eradication of all tested biofilms. Here,
3 bacterial species that commonly form clinically relevant pathogenic biofilms
were selected: Escherichia coli, Staphylococcus aureus and Streptococcus
mutans. The development of this antimicrobial photodynamic therapy tool for
biofilm eradication takes a promising step towards new methods for the much
needed treatment of pathogenic biofilms.
Methane emission by terrestrial invertebrates is restricted to millipedes, termites, cockroaches, and scarab beetles. The arthropod-associated archaea known to date belong to the orders Methanobacteriales, Methanomassiliicoccales, Methanomicrobiales, and Methanosarcinales, and in a few cases also to nonmethanogenic Nitrososphaerales and Bathyarchaeales. However, all major host groups are severely undersampled, and the taxonomy of existing lineages is not well developed. Full-length 16S rRNA gene sequences and genomes of arthropod-associated archaea are scarce, reference databases lack resolution, and the names of many taxa are either not validly published or under-classified and require revision. Here, we investigated the diversity of archaea in a wide range of methane-emitting arthropods, combining phylogenomic analysis of isolates and metagenome-assembled genomes (MAGs) with amplicon sequencing of full-length 16S rRNA genes. Our results allowed us to describe numerous new species in hitherto undescribed taxa among the orders Methanobacteriales (Methanacia, Methanarmilla, Methanobaculum, Methanobinarius, Methanocatella, Methanoflexus, Methanorudis, and Methanovirga, all gen. nova), Methanomicrobiales (Methanofilum and Methanorbis, both gen. nova), Methanosarcinales (Methanofrustulum and Methanolapillus, both gen. nova), Methanomassiliicoccales (Methanomethylophilaceae fam. nov., Methanarcanum, Methanogranum, Methanomethylophilus, Methanomicula, Methanoplasma, Methanoprimaticola, all gen. nova), and the new family Bathycorpusculaceae (Bathycorpusculum gen. nov.). Reclassification of amplicon libraries from this and previous studies using this new taxonomic framework revealed that arthropods harbor only CO2 and methyl-reducing hydrogenotrophic methanogens.
Numerous genus-level lineages appear to be present exclusively in arthropods, suggesting long evolutionary trajectories with their termite, cockroach, and millipede hosts, and a radiation into various microhabitats and ecological nichesprovided by their digestive tracts (e.g., hindgut compartments, gut wall, or anaerobic protists). The distribution patterns among the different host groups are often complex, indicating a mixed mode of transmission and a parallel evolution of invertebrate and vertebrate-associated lineages.
Gas source localization (GSL) helps mitigate the impact of industrial accidents and natural disasters. While GSL may be dangerous and time-consuming when performed by humans, swarms of agile and inexpensive nano aerial robots may increase the safety and efficiency of gas source localizations. Since the small payloads of nano aerial robots limit the sensing and computing resources, strategies adapted from biological swarms, such as colonies of social insects, are used to coordinate robot swarms. Most swarm GSL strategies are based on the assumption that the maxima of gas concentrations are sufficiently close to the gas sources. However, prior studies have indicated that the occurrence of “bouts”, a metric for the intermittency of gas distributions, may advantageously be used as a more accurate gas source proximity indicator. This paper presents a swarm GSL strategy employing bouts as source proximity indicators and a bio-inspired pheromone system for communication. Nano aerial robots, deployed in this study, act as agents and emit pheromone markers in an artificial environment upon detecting bouts. Leveraging the concept of artificial potential fields, the agents switch between exploiting the knowledge of the swarm by following pheromone gradients and exploring the search space by targeting a random point. The agents are repelled by each other and by walls to avoid collisions. The swarm GSL strategy is implemented into three nano aerial robots and validated in a real-world experiment in an indoor environment with a single gas source. The results indicate that the the swarm GSL strategy presented in this paper is capable of GSL in indoor environments and that the intermittency of gas distributions is a better source proximity indicator than the mean concentration.

Fungal pigments such as melanin and carotenoids are distinctive markers of animal and plant pathogenic fungi as well as their environmental relatives. These complex pigments play important roles in pathogenicity and stress tolerance while also being useful as biomarkers. Accordingly, it is important to be able to identify in situ the pigments in black fungi, a group of clinical and environmental importance. In this study, wild-type and genetically modified strains of Knufia petricola A95 and wild fungal cells attached to ancient rock were investigated for their spectroscopic and microscopic Raman features and morphological appearance. Knockout mutants of melanin synthesis genes pks1 (polyketide synthase), sdh1 (scytalone dehydratase), and both pks1 and the carotenoid synthesis gene phd1 (phytoene desaturase) were studied We applied two different Raman microscopes using two lasers, with 633 nm and 488 nm wavelengths. We analyzed and compared Raman spectra between the measured reference substances and the mutant and wild-type strains. In the wild strain WT:A95, the peaks close to melanin peals were found at 1353 cm−1 and 1611 cm−1. There are no characteristic melanin peaks at 1580–1600 cm−1 and around 1350 cm−1 at the spectrum of the Δpks1/Δphd1 mutant and the Δsdh1 mutant. The Δpks1 mutant spectrum has the peaks at the beta-carotene v2 C-C in-plane stretch at 1155 cm−1 and v3 C-CH3 deformation at 1005 cm−1. The peaks of carotenoids and melanin were found in all mutants and the wild strain, except the Δpks1/Δphd1 mutant. Raman spectra allow for discrimination between the various pigments. Hence, interactions between natural fungal melanin, as well as other protective pigments, and complex environmental matrices can be characterized on a range of spatial and temporal scales.
In particular, the rapid development of lateral flow assays as indispensable tools for everyone to contain the SARS-CoV-2 pandemic has fuelled the global demand for analytical tests that can be used outside dedicated laboratories. In addition to their use in medical diagnostics, rapid tests and assays have become increasingly important in various fields such as food safety, security, forensics, and environmental management. The advantage is obvious: taking the assay directly to the sample minimizes the time between suspicion and decision-making, allowing faster action. Especially today, when mobile communication devices with powerful computing capabilities and built-in cameras are ubiquitous, more people than ever before around the world have the basic skills to operate a powerful detector at their fingertips. This sets the stage for a much wider use of analytical measurements in terms of prognosis and prevention, enabling professional laypersons in particular.
However, current strip-based systems are primarily focused on single parameter analysis, whether it is SARS-CoV-2 biomarkers, blood glucose levels, or lead concentrations in water samples. Industrial applications of such methods also often still rely on single-parameter assays, requiring multiple runs even for a limited number of key parameters. Overcoming these limitations depends on developing low-number multiplexing strategies that ensure robustness, reliability, speed, ease of use, and sensitivity.
This lecture will give an overview of several generic approaches developed in recent years to address these challenges. It will highlight how the synergy of supramolecular (bio)chemistry, luminescence detection, hybrid (nano)materials and device miniaturization can result in powerful (bio)analytical assays that can be used at a point-of-need.1-5 Selected examples will introduce key aspects of such systems that include tailored signaling mechanisms and recognition elements, materials functionalization and device integration, including hybrid nanomaterials, gated indicator release systems, strip modification, and smartphone-based analysis.
Throughout their life cycles—from production, usage, through to disposal—materials and products interact with the environment (water, soil, and air). At the same time, they are exposed to environmental influences and, through their emissions, have an impact on the environment, people, and health. Accelerated experimental testing processes can be used to predict the long-term environmental consequences of innovative products before these actually enter the environment.
We are living in a material world. Building materials, geosynthetics, wooden toys, soil, nanomaterials, composites, wastes and more are research subjects examined by the authors of this book. The interactions of materials with the environment are manifold. Therefore, it is important to assess the environmental impact of these interactions. Some answers to how this task can be achieved are given in this Special Issue.
Climate change may increase the overall susceptibility of peatlands to fire. Smoldering fires in peatlands can cause substantial emissions of greenhouse gases. It is, however, less clear how smoldering affects the soil pore water quality. In this study, soil samples were collected from agricultural fen and disturbed bog study sites in Germany and Lithuania to quantify the effect of peat burning on pore water composition. The samples were air dried and smoldered under ignition temperature (approximately 200 °C) with different durations (0, 2, 5, and 10 h). Pore water samples were extracted from the soil to determine dissolved organic carbon (DOC) concentrations, dissolved organic matter (DOM) fractions, fluoride, extractable organically bound fluorine (EOF), and sulfate concentrations. The results showed that soil smoldering changes the peat pore water chemistry and that changes differ between fens and bogs. The smoldering duration is likewise influential. For fen grasslands, 2 and 5 h of smoldering of peat caused a >10-fold increase in DOC (up to 1600 mg L−1) and EOF concentrations. The fluoride (up to 60 mg L−1) and sulfate concentrations substantially exceededWHOdrinking water guidelines. In contrast, the temperature treatment decreased theDOC concentrations of samples from raised bogs by 90 %. The fluoride concentrations decreased, but sulfate concentrations increased after smoldering of the bog samples. DOC, fluoride, and sulfate concentrations of bogs varied significantly between the smoldering duration treatments. For all peat samples, the extracted DOMwas dominated by humic-like substances before smoldering, but the fraction of low molecular weight substances increased after smoldering combustion. In conclusion, smoldering alters the biogeochemical processes in both peatland types and possibly impair the water quality of adjacent water resources especially in fen peat landscapes.

Bio-SAXS of single-stranded DNA-binding proteins: Radiation protection by the compatible solute ectoine.
We aimed to increase the possible undisturbed exposure time during bio-SAXS measurements of single-stranded DNA-binding proteins. Therefore small angle X-ray scattering was performed on Gene-V Protein (G5P/GVP), which is involved in DNA repair processes. To achieve this, irradiations were performed in presence and absence of the hydroxyl-radical scavenger and osmolyte Ectoine, which showed efficient radiation protection and prevented protein aggregation, thus allows for a non-disturbing way to improve structure-determination of biomolecules.
Energy efficient products often have a higher purchase price than less efficient alternatives. However, the life cycle costs are often lower due to the lower operating costs. From a purely economic point of view, efficient appliances are not only more ecological but often also more economical and, accordingly, should be purchased more frequently than less efficient appliances. However, this is not always the case. Information deficits are one of the major reasons of this discrepancy: the purchase price is visible during the purchasing process, but the follow-up costs are not, at least not in their exact amount. The EU Energy Label can be considered as a solution to close this gap. During the past decades, there has been a discussion on whether it would be better to communicate the monetary costs associated with energy consumption, since their calculation by the consumer is difficult or needs a high level of cognitive effort.
In this study, experiments and field studies on a total of 19 product groups were evaluated with regard to the question of whether the indication of costs instead of physical units affects the decision-making process of the consumer. It has been assessed under which circumstances it might positively influence the purchase decision of the customer, i.e. helping him or her to arrive at a more “energy efficient” decision. Overall, the results indicate that monetary claims possibly exceed the effect of claims on a physical basis when the costs or the differences between the alternatives available seem “relevant” to the consumer.
The soil-feeding habit is an evolutionary novelty found in some advanced groups of termites. The study of such groups is important to revealing interesting adaptations to this way-of-life. The genus Verrucositermes is one such example, characterized by peculiar outgrowths on the head capsule, antennae and maxillary palps, which are not found in any other termite. These structures have been hypothesized to be linked to the presence of a new exocrine organ, the rostral gland, whose structure has remained unexplored.
We have thus studied the ultrastructure of the epidermal layer of the head capsule of Verrucositermes tuberosus soldiers. We describe the ultrastructure of the rostral gland, which consists of class 3 secretory cells only. The dominant secretory organelles comprise rough endoplasmic reticulum and Golgi apparatus, which provide secretions delivered to the surface of the head, likely made of peptide-derived components of unclear function. We discuss a possible role of the rostral gland of soldiers as an adaptation to the frequent encounter with soil pathogens during search for new food resources.

SCU umfasst CO2-reduzierende Maßnahmen in bestehenden Eisen- und Stahlproduktionsprozessen, Carbon Capture and Storage- (CCS) sowie Carbon Capture and Usage-Technologien (CCU).

Silver nanoparticles (AgNPs) nowadays appear in close to 24% of consumer products that contain engineered nanomaterials. Thus, they are expected to be released into the environment, where their fate and effect are still undetermined. Considering the evidenced efficacy of the single particle Inductively Coupled Plasma – Mass Spectrometry (sp ICP-MS) technique in the study of nanomaterials, this work reports on the use of sp ICP-MS along with an online dilution sample introduction system for the direct analysis of untreated and spiked seawater samples, as part of a larger scale experiment studying the fate of Ag (ionic and nanoparticles) in seawater mesocosm systems. Silver nanoparticles coated with branched polyethyleneimine (BPEI@AgNPs) or ionic silver (Ag+) were introduced gradually into the seawater mesocosm tanks at very low, environmentally relevant concentrations (50 ng Ag L− 1 per day, for 10 consecutive days, up to a total of 500 ng Ag L− 1 ), and samples were collected and analyzed daily, within a consistent time window. Using very low detector dwell time (75 μs) and specialized data treatment, information was obtained on the nanoparticles’ size distribution and particle number concentration, as well as the ionic silver content, of both the AgNPs and the Ag+ treated seawater mesocosm tanks. The results for the AgNP treated samples indicated the rapid degradation of the added silver particles, and the subsequent increase of ionic silver, with recoveries close to 100% for the first days of the experiment. On the other hand, particle formation was observed in the Ag+ treated seawater tanks, and even though the number concentration of silver-containing nanoparticles increased throughout the experiment, the amount of silver per particle remained relatively constant from the early days of the experiment. In addition, the online dilution sample introduction system for the ICP-MS proved capable of handling the untreated seawater matrix without significant contamination issues and downtime, while the low dwell time and data treatment procedure developed were shown to be suitable for the analysis of nanomaterials at the low nm-scale, despite the complex and heavy matrix introduced into the ICP-MS.
Bisphenol A (BPA) is used in Polycarbonate (PC), polyacrylic resins (PAR), polysulfones (PSU), epoxy resins (ER), and polyetherimides (PEI). It is also applied in recycled polyvinyl chloride (PVC). Many outdoor applications are made from these BPA-based materials which are exposed to a variety of environmental impacts, like temperature, global radiation, rain, and others. This could lead to damaging and aging of the material which might cause leaching or migration of pollutants into the environment.
Understanding the fate and behavior of the released pollutants is very important. Therefore, different transformation products (TP) will be generated and analyzed:
1. To investigate the behavior of various bisphenols in wastewater treatment plants, possible technical TP are generated by UV irradiation. Simulation of Industrial wastewater treatment is realized by using the Fenton reaction, an advanced oxidation process.
2. To study the transformation of bisphenols in surface waters, global radiation will be simulated by using UVA lamps.
3. To simulate phase 1 metabolism, an electrochemical cell coupled to a mass selective detector is applied.
The analysis of metabolites and TP are achieved by a variety of gas and liquid chromatographic techniques coupled with mass spectrometry. The elucidation of the exact structure of the obtained TP and metabolites are done by 1H- and/or 13C-NMR experiments.
Per- and polyfluoroalkyl substances (PFAS) are a large and ever-growing class of chemicals that are widely used. Due to the strength of the C-F bond, they do not decompose but accumulate in the environment posing a risk for nature and humans alike. While the use of some PFAS, like perfluorooctanoic acid (PFOA) and perfluorooctanoic sulfonic acid (PFOS) is already regulated in the EU, new ‘precursor’ substances are used to replace them. However, these precursors may be equally harmful to the environment.
In recent years, many ‘hot spot’ sites with high PFAS contaminations in the soils have been distinguished, but no German-wide background values have been determined yet. Knowing these background values is crucial to understand the degree of PFAS contamination, underpinning future regulatory decisions.
600 soil samples are freshly taken at different sites all over Germany. PFAS are extracted from all samples. The extracts are analysed using LC-MS/MS with 30 PFAS as target compounds. Eluates are produced and analysed using targeted LC-MS/MS. To determine the amount of precursor substances not included in the targeted list, total oxidisable precursor (TOP) assays are performed with different approaches. Additionally, 50 samples taken at the same locations in 2012 are analysed to determine temporal trends of PFAS contamination. The determination of total extractable organically bound fluorine (EOF) is planned for comparison.
The aim of the project is to obtain a broad overview of the status and development of the PFAS contamination in soil in Germany. By the preparation and analysis of eluates, the leaching behaviour of the analytes and thereby their mobility in the environment is investigated, helping to understand the transport of PFAS from contaminated soil to water. The use of different TOP assay methods allows to compare their results and usability in routine analysis, thereby giving insights into the amount of PFAS contamination hidden to targeted analysis and helping inspire future analysis strategies. The future comparison of these results to EOF analysis may further underline this point. The large number of samples and the geographic distribution of the sampling is unprecedented in Germany and allows to determine high-risk sites and ‘hot spots’. In the future, the insights from the project may be used as a guide for the political regulation of uses of PFAS.
Detailed knowledge about soil composition is an important prerequisite for many applications, for example precision agriculture. Current standard laboratory methods are complex and time-consuming but could be complemented by non-invasive optical techniques. Its capability to provide a molecular fingerprint of individual soil components makes Raman spectroscopy a very promising candidate. A major challenge is strong fluorescence interference inherent to soil, but this issue can be overcome effectively using shifted excitation Raman difference spectroscopy (SERDS). A customized dual-wavelength diode laser emitting at 785.2 and 784.6 nm was used to investigate 117 soil samples collected from an agricultural field along a distance of 624 m and down to depths of 1 m. To address soil spatial heterogeneity, a raster scan approach comprising 100 measurement spots per sample was applied. Based on the Raman spectroscopic fingerprint extracted from intense fluorescence interference by SERDS, 13 mineral soil constituents were identified, and even closely related molecular species could be discriminated, for example polymorphs of titanium dioxide and calcium carbonate. For the first time, the capability of SERDS is demonstrated to predict the calcium carbonate content as an important soil parameter using partial least squares regression (R2 = 0.94, root mean square error of cross-validation RMSECV = 2.1%). Our findings demonstrate that SERDS can extract a wealth of spectroscopic information from disturbing backgrounds enabling qualitative and quantitative soil analysis. This highlights the large potential of SERDS for precision agriculture but also in further application areas, for example geology, cultural heritage and planetary exploration.
Impact-based classification of extreme rainfall events using a simplified overland flow model
(2023)
In this study, we present a new rainfall classification method based on the potential of individual rainstorm events to generate and sustain overland flow on a simplified urban model surface. Conventional rainfall classification approaches using peak intensities or rainfall sums can lead to mis-judgements of the impact of rainfall events, as they do not explicitly consider the potential of rain to generate and sustain substantial amounts of overland flow on typical sealed urban surfaces. The new method of impact-based rainfall classification proposed here comprises three steps: Identification of rainfall events from high-resolution time series, computation of overland flow depths for each event, and impact-based classification of rainfall events as a function of the magnitude of overland flow. The accuracy of this approach was evaluated by its ability to describe seasonal and annual occurrences of and variations in rainstorm impacts for high-resolution series at three different urban stations in Germany.
The most massive design on the Baltic shore used geosynthetic materials, the landslide protection construction in Svetlogorsk (1300 m long, 90,000 m2 area, South-Eastern Baltic, Kaliningrad Oblast, Russian Federation) comprises the geotextile and the erosion control geomat coating the open-air cliff slopes. Due to changes in elastic properties during long-term use in the open air, as well as due to its huge size, this structure can become a non-negligible source of microplastic pollution in the Baltic Sea. Weather conditions affected the functioning of the structure, so it was assessed that geosynthetic materials used in this outdoor (open-air) operation in coastal protection structures degraded over time. Samples taken at points with different ambient conditions (groundwater outlet; arid places; exposure to the direct sun; grass cover; under landslide) were tested on crystallinity and strain at break. Tests showed a 39–85% loss of elasticity of the polymer filaments after 3 years of use under natural conditions. Specimens exposed to sunlight are less elastic and more prone to fail, but not as much as samples taken from shaded areas in the grass and under the landslide, which were the most brittle.
A collaborative trial involving 16 participants from nine European countries was conducted within the NORMAN network in efforts to harmonise suspect and non-target screening of environmental contaminants in whole fish samples of bream (Abramis brama). Participants were provided with freeze-dried, homogenised fish samples from a contaminated and a reference site, extracts (spiked and non-spiked) and reference sample preparation protocols for liquid chromatography (LC) and gas chromatography (GC) coupled to high resolution mass spectrometry (HRMS). Participants extracted fish samples using their in-house sample preparation method and/or the protocol provided. Participants correctly identified 9-69% of spiked compounds using LC-HRMS and 20-60% of spiked compounds using GC-HRMS. From the contaminated site, suspect screening with participants’ own suspect lists led to putative identification of on average ~145 and ~20 unique features per participant using LC-HRMS and GC-HRMS, respectively, while non-target screening identified on average ~42 and ~56 unique features per participant using LC-HRMS and GC-HRMS, respectively. Within the same sub-group of sample preparation method, only a few features were identified by at least two participants in suspect screening (16 features using LC-HRMS, 0 features using GC-HRMS) and non-target screening (0 features using LC-HRMS, 2 features using GC-HRMS). The compounds identified had log octanol/water partition coefficient (KOW) values ranging from -9.9 to 16 and mass-to-charge ratio (m/z) of 68 to 761 (LC-HRMS and GC-HRMS). A significant linear trend was found between log KOW and m/z for the GC-HRMS data. Overall, these findings indicate that differences in screening results are mainly due to the data analysis workflows used by different participants. Further work is needed to harmonise the results obtained when applying suspect and non-target screening approaches to environmental biota samples.
Research on per- and polyfluoroalkyl substances (PFAS) frequently incorporates organofluorine measurements, particularly because they could support a class-based approach to regulation. However, standardized methods for organofluorine analysis in a broad suite of matrices are currently unavailable, including a method for extractable organofluorine (EOF) measured using combustion ion chromatography (CIC). Here, we report the results of an international interlaboratory comparison. Seven laboratories representing academia, government, and the private sector measured paired EOF and PFAS concentrations in groundwater and eel (Anguilla rostrata) from a site contaminated by aqueous film-forming foam. Among all laboratories, targeted PFAS could not explain all EOF in groundwater but accounted for most EOF in eel. EOF results from all laboratories for at least one replicate extract fell within one standard deviation of the interlaboratory mean for groundwater and five out of seven laboratories for eel. PFAS spike mixture recoveries for EOF measurements in groundwater and eel were close to the criterion (±30%) for standardized targeted PFAS methods. Instrumental operation of the CIC such as replicate sample injections was a major source of measurement uncertainty. Blank contamination and incomplete inorganic fluorine removal may introduce additional uncertainties. To elucidate the presence of unknown organofluorine using paired EOF and PFAS measurements, we recommend that analysts carefully consider confounding methodological uncertainties such as differences in precision between measurements, data processing steps such as blank subtraction and replicate analyses, and the relative recoveries of PFAS and other fluorine compounds.
Woodworms, carpet beetles, clothes moths and termites are well known pests on artifacts of cultural importance. Lately also silverfish have gained importance. Protection of our cultural heritage for future generations is not a new task but the methods have changed. Conventional use of pesticides in the past has preserved precious objects up to the presence, which otherwise probably would have been lost over time due to the destruction by insects and microbes. However, several of the objects were contaminated with poisons in such a manner that they became unsafe to be handled and thus are useless for exhibitions or scientific studies.
Today, the concept of Integrated Museum Pest Management (IMPM) provides guidance for non-residue treatments of infested objects, followed by save storage or display under pest free (or maximum pest reduced) environments. Inert fumigants, physical and biological control measure, precise monitoring, threshold evaluations and detailed knowledge of the pest`s biology are key elements within IMPM.
Woodworms, clothes moths and termites, however, are not always considered as only pests. Some professionals have also used them to produce new pieces of art or for the installation of ephemeral displays. Under more or less controlled conditions, the insects´ destructive nature on wood or textiles has been turned around to a process of creation with aesthetic or philosophical messages.
Destructive or creative - it just depends on the point of view!

BPA is a chemical commonly used in the production of polymer-based materials that can have detrimental effects on the thyroid gland and impact human reproductive health. Various expensive methods, such as liquid and gas chromatography, have been suggested for detecting BPA.
The fluorescence polarization immunoassay (FPIA) is an inexpensive and efficient homogeneous mix-and-read method that allows for high-throughput screening. FPIA offers high specificity and sensitivity and can be carried out in a single phase within a timeframe of 20–30 min. In this study, new tracer molecules were designed that linked the fluorescein fluorophore with and without a spacer to the bisphenol A moiety. To assess the influence of the C6 spacer on the sensitivity of an assay based on the respective antibody, hapten–protein conjugates were synthesized and assessed for performance in an ELISA setup, and this resulted in a highly sensitive assay with a detection limit of 0.05 µg/L. The lowest limit of detection was reached by employing the spacer derivate in the FPIA and was 1.0 µg/L, working range from 2 to 155 µg/L. The validation of the methods was conducted using actual samples compared to LC–MS/MS, which served as the reference method. The FPIA and ELISA both demonstrated satisfactory concordance.
Adipokinetic hormone (AKH) is a neuropeptide produced in the insect corpora cardiaca that plays an essential role in mobilising carbohydrates and lipids from the fat body to the haemolymph. AKH acts by binding to a rhodopsin-like G protein-coupled receptor (GPCR), the adipokinetic hormone receptor (AKHR). In this study, we tackle AKH ligand and receptor gene evolution as well as the evolutionary origins of AKH gene paralogues from the order Blattodea (termites and cockroaches). Phylogenetic analyses of AKH precursor sequences point to an ancient AKH gene duplication event in the common ancestor of Blaberoidea, yielding a new group of putative decapeptides. In total, 16 different AKH peptides from 90 species were obtained. Two octapeptides and seven putatively novel decapeptides are predicted for the first time. AKH receptor sequences from 18 species, spanning solitary cockroaches and subsocial wood roaches as well as lower and higher termites, were subsequently acquired using classical molecular methods and in silico approaches employing transcriptomic data. Aligned AKHR open reading frames revealed 7 highly conserved transmembrane regions, a typical arrangement for GPCRs.
Phylogenetic analyses based on AKHR sequences support accepted relationships among termite, subsocial (Cryptocercus spp.) and solitary cockroach lineages to a large extent, while putative post-translational modification sites do not greatly differ between solitary and subsocial roaches and social termites. Our study provides important information not only for AKH and AKHR functional research but also for further analyses interested in their development as potential candidates for biorational pest control agents against invasive termites and cockroaches.
Excessive discharge of quaternary ammoniumdisinfectants such as benzalkonium chloride (BAC) into aquatic systems can trigger several physiological responses in environmental microorganisms. In this study, we isolated a less-susceptible strain of Aeromonas hydrophila to BAC, designated as INISA09, froma wastewater treatment plant in Costa Rica. We characterized its phenotypic response upon exposure to three dierent concentrations of BAC and characterizedmechanisms related to its resistance using genomic and proteomic approaches. The genome of the strain, mapped against 52 dierent sequenced A. hydrophila strains, consists of approximately 4.6Mb with 4,273 genes. We found a massive genome rearrangement and thousands of missense mutations compared to the reference strain A. hydrophila ATCC 7966.
We identified 15,762 missense mutations mainly associated with transport, antimicrobial resistance, and outer membrane proteins.
In addition, a quantitative proteomic analysis revealed a significant upregulation of several efflux pumps and the downregulation of porins when the strain was exposed to three BAC concentrations.Other genes related tomembrane fatty acid metabolism and redox metabolic reactions also showed an altered expression.
Our findings indicate that the response of A. hydrophila INISA09 to BAC primarily occurs at the envelop level, which is the primary target of BAC. Our study elucidates the mechanisms of antimicrobial susceptibility in aquatic environments against a widely used disinfectant and will help better understand howbacteria can adapt to biocide pollution. To our knowledge, this is the first study addressing the resistance to BAC in an environmental A. hydrophila isolate. We propose that this bacterial species could also serve as a new model to study antimicrobial pollution in aquatic environments.
Understanding the fundamentals of bioreceptivity enables the developement of functionalized materials. Concrete as the most used building material worldwide is of special interest as microbially greened panels may represent an alternative to classic façade greening with plants. This project includes extensive outdoor experiments, in which eight differently structured concrete panels are weathered under different conditions, aiming to grow a stable biofilm of photosynthetic eukaryotic organisms. Documentation and data analysis of this experiment will be demonstrated using an example of one of the surfaces.
By tracing sustainability impacts along the supply chain, certification schemes and labels are crucial to unlock the potential of bio-based systems in achieving several SDGs. However, the rapid proliferation of certification schemes and labels led to questioning their effectiveness and robustness and to what extent they deliver important outcomes, in line with sustainability policy priorities. The STAR4BBS project develops indicators and a monitoring system for assessing the effectiveness and robustness of existing certification schemes and labels, applicable to biological feedstock and bio-based materials and products.
Per- and polyfluoroalkyl substances (PFAS) are widely used for durable water-repellent finishing of different fabrics and textiles like outdoor clothing, carpets, medical textiles and more. Existing PFAS extraction techniques followed by target analysis are often insufficient in detecting widely used side-chain fluorinated polymers (SFPs) that are barely or non-extractable. SFPs are typically copolymers consisting of a non-fluorinated backbone with perfluoroalkyl side-chains to obtain desired properties. We compared the accessible analytical information and performance of complementary techniques based on oxidation (dTOP assay, PhotoTOP), hydrolysis (THP assay), standard extraction, extractable organic fluorine (EOF), and total fluorine (TF) with five functional textiles and characterized 7 further textiles only by PhotoTOP oxidation. The results show that when applied directly to textile samples, oxidation by dTOP and PhotoTOP and also hydrolysis by the THP are able to capture large fractions of the TF in form of perfluoroalkyl side-chains present in the textiles while methods relying on extracts (EOF, target and non-target analysis) were much lower (e.g., factor ~25-50 lower). The conversion of large fractions of the measured TF into PFCAs or FTOHs from fluorinated side chains is in contrast to previous studies. Concentrations ranged from <LOQ to over ~1000 mg F/kg after oxidation/hydrolysis and <LOQ to over 2000 mg F/kg for TF, while EOF and target PFAS in extracts were detected at much lower concentrations (up to ~ 60 mg F/kg) (amount of fluorine in the order: extraction << EOF << oxidation/hydrolysis ≤ TF). Perfluoroalkyl carboxylic acids (PFCAs) and fluorotelomer alcohols (FTOHs) from THP and PhotoTOP both represented the chain-length distribution in the textiles showing that long-chain SFPs are still used in current textiles. Further advantages and disadvantages of the applied methods are discussed.
Disinfectants are important to prevent the transmission of pathogens, especially in the face of the current antibiotic resistance crisis. The crisis is further exacerbated by phenotypically tolerant persister subpopulations that can survive transient antibiotic treatment and facilitate resistance evolution. Despite the transient nature of disinfectant application, persistence to disinfectants and its role for the evolution of tolerance and cross-resistance to antibiotics has not been studied. Our work shows that E. coli displays persistence against several widely used disinfectants, including benzalkonium chloride (BAC), didecyldimethylammoniumchlorid (DDAC) and isopropanol. The molecular mechanism of BAC persistence is triggered in stationary phase and affected by several antibiotic persister genes (hipA, tisB, tolC, relA, spoT). Experimental evolution and population dynamic modeling show that repeated failure of disinfection due to persisters rapidly selects for BAC tolerance underpinned by reduced cell surface charge due to mutations in genes related to lipid A acylation (lpxML). Furthermore, evolved BAC tolerance affects the susceptibility to antibiotics, leading to positive selection of disinfectant tolerant strains at environmentally relevant antibiotic concentrations and variations in evolvability of antibiotic resistance due to epistatic effects. These results highlight the need for faithful application of disinfectants to steward their efficacy and the efficacy of antibiotics. A better understanding of the bacterial response to disinfectants is crucial to understand and avert the ongoing antimicrobial resistance crisis.
