A bead suspension array for measuring emerging pollutants and anthropogenic markers in wastewater.
(2016)
The assessment of water quality demands for high-throughput and multi-target compatible analytical methods. On the one hand, the influence of sewage on natural surface waters must be routinely monitored - according to the European Water Framework Directive. On the other hand, estimation of removal efficiencies of pollutants, such as drug residues, is in the focus of industrial and public wastewater treatment. Immunoassays, such as ELISA, are often applied in this area owing to their cost-efficiency and high-throughput capability. However, it is only possible to measure a single analyte in one measurement.
In contrast, array technology is capable for measuring multiple substances in parallel. Here we present a four-plex bead-based flow cytometric assay for measuring three drugs (carbamazepine, an anti-epileptic drug, diclofenac, an antiphlogistic and caffeine, a psychoactive substance) and the bile acid isolithocholic acid, which is proposed to be used as a fecal marker.
In this suspension array core-shell particles, consisting of a polystyrene core and a silica shell are used. They can be easily encoded with a fluorophore which is introduced to the core and functionalized with amino groups on the surface for creating binding sites for antibodies: Therefore, the marker substances are bound to the amino groups via NHS chemistry.
For the determination of the pollutants, the beads are incubated with a mixture of the analyte-specific antibodies. The antibodies undergo a competitive reaction to bind the immobilised molecules on the surface of the beads or the free analyte in the solution, respectively. Bound antibodies can then be visualized via fluorophore-labelled secondary antibodies. For read-out a flow cytometer is used to virtually separate the different encoded beads and to measure the resulting signal intensity of the immunoassay. In order to obtain highly selective binding of the antibodies, we investigated different types of surface modifications to overcome unspecific binding, finding that a PEG-based surface is suitable to support our immunoassay format. The resulting multiplexing assay is appropriate to detect the marker substances in the low µg/L range.
The screening of coatings shows that the modification by adding individual pigment was not sufficient to protect carbon steel even during a short-term exposure, indicated by the discoloration after only seven days of exposure.
Electrochemical tests indicated that the coating cathodically protects carbon steel or slows down the corrosion reaction.
A long-term exposure test confirmed that the PANI/SiO2 modified coating successfully protects the carbon steel in the Sibayak artificial geothermal water at 150 °C for 6 months.
Bisphenol A (BPA), a monomer commonly used to manufacture polycarbonate plastic products and the epoxy resins that line metal cans, has resulted in an increased human exposure because both products come in contact with food and beverages. Since BPA is an endocrine disruptor excessive exposure could affect human reproductive function. In response, various countries have adopted a range of measures, from voluntary reductions to outright bans of BPA in some products, mostly in baby bottles and containers for canned foods. Pregnancy-test like strips in combination with hand-held readers provide a simple and versatile tool to determine the presence of bisphenol A quantitatively, making it a very attractive analytical approach especially for on-site applications at critical points, such as environmental analysis, consumer protection, and materials testing.
The common characterization and identification of pollen is a time-consuming task that mainly relies on microscopic determination of the genus-specific pollen morphology. A variety of spectroscopic and spectrometric approaches have been proposed to develop a fast and reliable pollen identification using specific molecular information. Amongst them, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) shows a high potential for the successful investigation of such complex biological samples. Based on optimized MALDI sample preparation using conductive carbon tape, the application of multivariate statistics (e.g. principal components analysis, PCA) yields an enormous improvement concerning taxonomic classification of pollen species compared to common microscopic techniques.
Since multivariate evaluation of the recorded mass spectra is of vital importance for classification, it’s helpful to implement the applied sequence of standard Matlab functions into a graphical user interface (GUI). In this presentation, a stand-alone application (GUI) is shown, which provides multiple functions to perform fast multivariate analysis on multiple datasets. The use of a GUI enables a first overview on the measured dataset, conducts spectral pretreatment and can give classification information based on HCA and PCA evaluation. Moreover, it can be used to improve fast spectral classification and supports the development of a simple routine method to identify pollen based on mass spectrometry.
The repair of earthen structures is not an easy task. Earth as a construction material has comparatively weak mechanical properties. It is also susceptible towards liquid water, were it can lose cohesion very quickly if certain moisture contents are exceeded. Repair concepts of structures built with such materials are therefore challenging. Historical earthen structures often exhibit damages in form of extensive cracking, which may have been caused by static or dynamic loads. Frequently these cracks are insufficiently or inappropriately repaired because of lack of knowledge and / or technology. In particular, the behaviour of crack repair by grouting poses a challenge in earthen materials and demands specific requirements for the grouting mortar, such as low water content, good water retention, low shrinkage. Grouting materials require additional specifications such as compatible strengths and Young’s modulus as well as good adhesion to the earthen substrates. In addition, grouts have to be sufficiently flowable to fill small cracks and voids without segregation or bleeding. Therefore, the rheological behaviour has to be well understood and controlled to gain the desired effects. The repair of cracks in earthen buildings is traditionally done by stuffing manually mortar into the gap. Naturally, this method is only usable for cracks with large widths. Another disadvantage is that cracks going through thicker walls cannot be completely reached by the tools used for stuffing the mortar into the crack. Lime based grouts for earthen materials were usually used for re-attachment of plasters but less for structural reasons.Due to the nature of earthen materials grouts based on formulated or hydraulic lime (according to the definition in EN 459-1) have to meet considerable demands on a variety of properties, which are related to compatibility, durability and in particular to the ability of being injected. The goal of this study was to create a grout, which can be used to re-establish structural continuity in cracked earthen masonry or other massive earth walls (rammed earth and cob) with the focus on grouting cracks. The grout material was based on hydrated lime (calcium hydroxide) with additions of pozzolana and lime stone filler.
A new approach towards performance-related design and scientific evaluation of joint sealing systems
(2020)
While there is a permanent improvement of concrete pavement mixtures and pavement construction types over the last decades, the state-of-the-art joint sealing materials and joint constructions seem to stagnate on an antiquated empirical level. This status has been reaffirmed in the latest European standard.
The consequences in the motorway network due to unsatisfying capability and durability of joint sealing systems are unacceptable. In addition, inadequate traffic performance (noise emissions, roll-over comfort) and traffic safety losses in the joint area of concrete pavements are existing challenges. These deficits and weaknesses reflect a demand for joint sealing materials and constructions whose approval requirements take functional aspects into account. Furthermore a sufficient analysis of decisive loads and a practice-oriented method to evaluate the requirements towards performance and durability is still missed.
In this contribution decisive loads to German highways are analyzed. The design of test specimen for representative functional testing of joint sealing systems is discussed. The focus is on the geometry of the test specimens and the used concrete mixture. Finally, a new approach for a function-orientated test concept that considers representative load functions is presented. The potential of this approach to validate the durability and capability of various joint sealing systems is also presented using an example.
A new tool in material science - Targeted disruption of melanin synthesis in rock-inhabiting fungi
(2016)
The ability to survive almost absolute dehydration through air-drying is a remarkable feature of rock-inhabiting microcolonial fungi (MCF), which colonise rock surfaces in hot and cold deserts. Understanding of the underlying mechanisms which allow this group of fungi to conquer natural and man-made environments requires a set of modern biological techniques and approaches that are under development in our laboratory. We will present an overview of the targeted disruption of melanin biosynthesis genes in the rock-inhabiting Knufia petricola and give inside into the lines of research and the network of supporting laboratories that made this progress possible.
A process spectroscopic approach for the monitoring of microplastics in environmental samples
(2018)
The potential of Raman and NIR process-spectroscopic as a rapid approach for the estimation of microplastics (MP) in soil matrix were tested. For this purpose, artificial MP/soil mixture samples consisting of standard soils or sand with defined ratios of MP (0,5 – 10 mass% polymer) from polyethylene (PE), polypropylene (PP), polystyrene (PS) and polyethylene terephthalate (PET) were prepared. MP particles with diameters < 2 mm and < 125 µm were obtained from industrial polymer pellets after cryo-milling. Spectra of these mixtures were collected by (i) a process FT-NIR spectrometer equipped with a reflection probe, (ii) by a cw-process Raman spectrometer and (iii) by a time-gated Raman spectrometer using fiber-optic probes. The evaluation of process-spectra was performed by chemometric methods. Whereas MP could be detected by Raman spectroscopy in coastal sand at 0.5 mass%, in standard soil detection of MP was limited to 10 – 5 mass% with the large fraction, but samples containing particles of the 125µm mass-fraction yielded no positive result at all. One reason for the lacking sensitivity could be fluorescence by soil organic matter and thus, in a next test time-gated Raman spectroscopy was applied. However, although being indeed more sensitive to the small particles this method failed at MP < 5 mass% indicating that fluorescence was not the major problem. Finally, FT-NIR was tested. Depending on the polymer, MP contents of 0,5 or 1 mass%, respectively, could be detected in standard soils and polymers identified. Furthermore, this approach could be used for the investigation of mixtures of up to four polymers and in real-world samples from bio-waste fermenter residues.
Sewage sludge ashes (SSA) contain up to 13% P and are thus promising raw materials for fertilizer production. However, SSAs also contain heavy metals and the main P-bearing mineral phases whitlockite and aluminium phosphate are poorly bioavailable. We developed and patented a process that produces fertilizers from SSA addressing the above mentioned challenges. SSA is thermo-chemically treated at ~950°C under reducing conditions (reductive: dry sewage sludge) in a rotary kiln together with an alkali compound such as sodium or potassium sulfate, hydroxide or carbonate. Some undesired trace elements such as As, Hg, Pb and Cd are evaporated and separated via off gas treatment system. The poorly soluble phosphates are transformed into calcium alkali phosphates (CaNaPO4 or CaKPO4) that are not water soluble but completely soluble in neutral ammonium citrate solution (NAC). Pot experiments showed that the fertilizer performance of the treated SSA containing calcium alkali phosphates as the single P-bearing mineral phase was comparable to the performance of triple superphosphate. The PNAC-solubility of fertilizer products correlated very well with the resulting contents of calcium alkali phosphates and the P-uptake of plants in pot experiments. The effect of the ratio alkali/P on the PNAC-solubility was investigated by crucible experiments and trial series with a medium scale rotary kiln using different SSAs and alkali phosphates. The effects of operational parameters such as the temperature and the retention time were investigated as well as concurring side reactions of the alkali compounds e.g. with SiO2 present in SSA. The alkali/P ratio must be roughly adjusted at 2 to achieve 100% PNAC-solubility for a common type of SSA. A demonstration trial with an output of 2 t recycling fertilizer was carried out in an industrial rotary kiln (product output 30 kg/h). The PNAC-solubility of the product varied between 60% and 80% during the 4 days campaign showing that the transformation of the mineral P-phases to calcium alkali phosphates was not complete. This was observed although the amount of Na2SO4 additive was dosed according to the results of the pre-investigations. Structure analysis by XRD showed that besides the target compound CaNaSO4 also some Ca3(PO4)2 remained in the SSA as well as some unreacted Na2SO4 additive. Obviously, the conditions in the industrial rotary kiln were not optimal for the process showing that some process aspects have to be reconsidered for the scale-up.
It is of high priority to capture the extent of microplastics (MP) contamination for understanding its impact and consequences to our environment. However, due to a lack of standardized methods, MP analysis as well as its quality assurance, is still a challenge. The thermal extraction desorption-gas chromatography/-mass spectrometry (TED-GC/MS) as a novel innovative thermoanalytic method can detect MP particles in environmental samples based on their decomposition products. According to MP analysis, a distinctively larger polyethylene (PE) content in comparison to other synthetic polymers was detected in environmental samples. Various substances within the matrix which have a similar molecular structure to PE, like fats and oils, might interfere with its marker and lead to an overestimation. In this investigation an automated extraction method, the accelerated solvent extraction (ASE), is introduced as quality control tool before TED-GC/MS measurement. Thereby, not the extract like usually used, but the solid filtrate is analyzed. The aim is to clean the environmental samples from solvent removable compounds which could lead to false positive signals and thus receive a more accurate PE mass. Consequently, a sample mimicking environmental samples with a defined matrix and a known polymer mass was generated. The common polymers polyethylene, polypropylene, polystyrene and styrene-butadiene rubber, used as an indication for tire wear, were added to the matrix. Different parameters for ASE extraction were varied such as different solvents, filters and purge time. Finally, a PE recovery rate of 91 % after extraction could be reached.
Adipokinetic hormone (AKH) is a neuropeptide produced in the insect corpora cardiaca that plays an essential role in mobilizing carbohydrates and lipids from the fat body to the hemolymph. AKH acts by binding to a rhodopsin-like G protein-coupled receptor (GPCR), the adipokinetic hormone receptor (AKHR). In this study, we tackle AKH ligand and receptor gene evolution as well as the evolutionary origins of AKH gene paralogues from the order Blattodea (termites and cockroaches). Phylogenetic analyses of AKH precursor sequences point to an ancient AKH gene duplication event in the common ancestor of Blaberoidea, yielding a new group of putative decapeptides. In total, 16 different AKH peptides from 90 species were obtained. Two octapeptides and seven putatively novel decapeptides are predicted for the first time. AKH receptor (AKHRs) sequences from 18 species, spanning solitary cockroaches and subsocial woodroaches as well as lower and higher termites, were subsequently acquired using classical molecular methods and in silico approaches employing transcriptomic data. Aligned AKHR open reading frames revealed 7 highly conserved transmembrane regions, a typical arrangement for GPCRs. Phylogenetic analyses based on AKHR sequences support accepted relationships among termite and cockroach lineages to a large extent, while putative post-translational modification sites do not greatly differ between solitary and subsocial roaches and social termites. This study provides important information not only for AKH and AKHR functional research but also for further analyses interested in their development as potential candidates for biorational pest control agents against invasive termites and cockroaches.
The rational synthesis and use of nanomaterials require the characterization of many different properties, ranging from particle size and size distribution over surface chemistry to more applicationrelevant features like optical, electrochemical, and magnetic properties. In the following, several methods for the characterization of functional groups on nanomaterials, like polymer and silica nanoparticles, semiconductor quantum dots, and lanthanide-based upconversion nanocrystals are presented. Additionally, procedures for the measurement of the key spectroscopic performance parameters of nanomaterials with linear and nonlinear photoluminescence, such as the photoluminescence quantum yield, are presented for the UV/vis/NIR/SWIR.
Immunoanalytical Techniques, i.e., antibody-based analytical methods, have been used for decades in clinical diagnostics. What makes them attractive for other fields of application is their short time-to-result and high sensitivity. Microplate-based assays such as ELISA have been adopted early in environmental and food analysis. Yet, to make immunoassays even faster, more sensitive, robust, and, most desirable, portable, advanced materials, sometimes developed for other purposes, can be profitably used to achieve these goals. Materials can be novel labels, e.g., chemical or particle labels, such as fluorophores or nanoparticles. Carrier particles, such as magnetic or polymer beads, make it possible to adopt the assays to meso- or microfluidic set-ups and encoding them opens the path to multiplex analysis. Specialty electrodes can enable for higher sensitivity in electrochemical detection. All this broadens the scope of application and lowers effort and cost for analysis at the point-of-need.
Antibody-based analytical techniques have gained increasing importance in environmental analysis. Rapid assays, portable devices and formats that require a limited number of steps have a high potential for on-site analysis in outdoor environments or in environmentally relevant facilities such as wastewater treatment plants. Many of these methods have become more sensitive and more versatile due to new, advanced materials, such as novel particles, specifically prepared surfaces, new labels, specifically coated electrodes etc. The talk shows some of our works in this context.
Advances in ultrafast laser manufacturing: nanostructures, thin films, and scaling perspectives
(2024)
Advanced ultrafast laser technology is a rapidly growing field that currently enables many new industrial and scientific applications. During the last decades, this has been significantly driven by the availability of high-repetition-rate laser sources and novel beam delivery concepts. At the laser side, Moore’s law equally manifests for ultrafast laser technologies, since the average output power of such lasers doubles approximately every two years. This development is mainly driven by the increase of the pulse repetition rates of energetic laser pulses, currently enforcing the development of smart beam control and novel scanning strategies for preventing heat-accumulation and plasma-shielding effects during laser-based materials processing. This keynote presentation addresses the advantages, recent developments, and perspectives of laser processing with ultrashort laser pulses. A special focus is laid on the tailored structuring of thin films as well as the manufacturing and probing of sub-diffraction surface nanostructures – an ongoing race to extreme scales. Current limitations are identified and an outlook to future scaling perspectives will be provided.
Interlaboratory studies are common tools for collecting comparable data to implement standards for new materials or testing technologies. In the case of construction materials, these studies form the basis for recommendations and design codes. Depending on the study, the amount of data collected can be enormous, making manual handling and evaluation difficult. On the other hand, the importance of the FAIR (findable, accessible, interoperable, and reusable) principles for scientific data management, published by Wilkinson et al. in 2016, is constantly growing and changing the view on data usage.
The benefits of using data management tools such as data stores/repositories or electronic laboratory notebooks are many. Data is stored in a structured and accessible way (at least within a group) and data loss due to staff turnover is reduced. Tools usually support data publishing and analysis interfaces. In this way, data can be reused years later to generate new knowledge with future insights. On the other hand, there are many challenges in setting up a data repository, such as selecting suitable software tools, defining the data structure, enabling data access, and understanding by others and
ensuring maintenance, among others.
This talk discusses the advantages and challenges of setting up and applying a data repository using the interlaboratory study on the mechanical properties of printed concrete structures carried out in RILEM TC 304-ADC as example. First, the definition of a suitable data structure including all information is discussed. The tool-dependent upload process is then described. Here, the data
management system openBIS (open source software developed by ETH Zurich) is used. Since in most cases an open compute platform allowing access from different organisations is not possible or available due to data protection and maintenance issues, tool-independent export options are discussed and compared. Finally, the different query and analysis possibilities are demonstrated.
During the last decades the material composition of buildings has become increasingly diverse. However, largely sorted material flows are needed for generating high quality secondary building materials. The use of secondary building materials can meet the requirements of sustainability in several ways: the extended time availability of primary raw materials and, thereby, the preservation of natural resources as well as the conservation of landfill sites.
Recycling of gypsum (calcium sulfate) can be a good example for the environmental benefits of closed-loop recycling. The content of sulfates in other secondary building materials, in particular in recycled concrete aggregates, should be minimized for quality reasons. In contrast, separated gypsum can also be used in gypsum production if the high quality requirements for the recycled gypsum are met. Since almost all processing steps in the recycling process are associated with environmental impacts, an environmental evaluation of the use of recycled gypsum as a substitute in gypsum production has to be carefully conducted.
This paper focusses on the techniques for generating recycled gypsum from gypsum plasterboards, the related quality requirements and a comprehensive environmental evaluation of the complete process.
Measurements of aerosol particles are vital for enforcing EU air quality regulations to protect human health, and for research on climate change effects. Although metrics such as PM10 and PM2.5 are currently in use, the level of uncertainty of aerosol metrics is too high and the traceability is insufficient. The project AEROMET, which has been started in June 2017 aims at implementing improvements in a) the uncertainty of particle mass, size and number concentration measurements and b) in the characterization of regulated components in airborne particles. Both are demanded by existing networks within the EU as well as by global atmospheric research.
On-site measurement campaigns
One of the objects is the application of mobile x-ray spectroscopy techniques combined with aerosol sampling techniques for quantifying particle compositions in the field for real time analysis. During two in-field measurement campaigns in Budapest, Hungary in May 2018 and Cassino, Italy in September 2018 the size dependent mass concentrations of specific elements in ambient aerosols were monitored under dynamic conditions.
Typically, airborne particles are sampled on filter substrates. During this project new sampling methods with specially designed substrate holders for an in-situ TXRF analysis were developed and applied for the first time. This approach allows a direct time and size resolved analysis without laborious digestion steps and a reduced risk of contamination.
Aerosol particles were sampled in a 13-stage DLPI impactor - size range from 0,03 µm to 10 µm - which was equipped with special adapters for acrylic discs of 30 mm diameter, serving as substrates. TXRF analysis was performed on site with the transportable spectrometer S2 PICOFOX (Bruker Nano GmbH) equipped with a Mo X-ray tube and a 30 mm² Silicon Drift Detector (SDD). Excitation conditions were 50 kV, 600 µA, measurement time 1000 s. Quantification was based on internal standardization using 50 ng of Y in solution, which was pipetted into the centre of the discs prior to sampling.
At moderate air pollution levels, i.e. PM10 ~ 20 µg/m³, sampling times of less than 2 hours were enough for the detection of elements in different particle size bins. The in-situ approach and the high sensitivity of TXRF enables the observation of rather quick changes in the quantity and distribution of elements in an ambient aerosol on the day of sampling, as the below example from the Cassino field campaign on 11 Sept. 2018 shows: The analysis of the morning and afternoon sampling shifts reveals the occurrence of the elements Fe, Ca and Si in different size bins as well as their significant temporal change in respective mass concentrations over the day while the distributions of several other elements in the aerosol remain unchanged.
The validation of these results by backup measurements is planned.
Traceable and reliable chemical element analysis of aerosols by X-ray spectrometry was investigated using aerosol samples from field campaigns which have been measured in the GIXRF-beamline at BESSY.
The reference-free XRF approach allows for a traceable analysis of the mass deposition.
Traceable quantification by means of XRF can be transfered to benchtop instrumentation used in the laboratory Chemical and dimensional analysis of deposited aerosol allows for a comprehensive analysis of aerosols, e.g. for toxicity assessment and determination of the source The folowing elements could be identified and quantified in the field samples: Al, Si, P, S, Cl, K, Ca, Ti, Cr, Mn, Fe, Ni, Cu, Zn, As, Se, Br, Rb, Sr, Y, W, Pb.
Was steckt eigentlich hinter diesen strengen Regelungen?

The aim of this study was to investigate the passive sampler method Diffusive Gradients in Thin-films (DGT) for ammonium and nitrate in amended soils. Therefore, we used soils from a pot experiment with maize where nitrogen (N) was supplied as ammonium sulfate nitrate (ASN), without and with a nitrification inhibitor (NI). The additional use of a NI can delay the nitrification in the soil and making the ammonium available for a longer period in the soil solution after its application. Homogenized soil samples were collected directly from each pot after one week of incubation before sowing and after harvesting the maize. Nitrate and ammonium in these soil samples were extracted using DGT devices equipped with a Putolite A520E (for nitrate) and Microlite PrCH (for ammonium) binding layer. Ammonium DGT which determined the mobile and labile ammonium forms based on diffusion and the resupplies from the solid soil phase, only showed a significantly higher amount of extractable ammonium with NI compared to that without NI for some samples. However, significantly lower values were found for nitrate of treatments with NI compared to without NI after harvest. Thus, the lower nitrate amounts for treatments with NI compared to the treatments without NI after harvest indicated the delay of the nitrification process by the NI. Furthermore, we compared also the ammonium and nitrate
DGT results to chemical extraction with KCl solutions. The results demonstrated that the trends of DGT results and chemical extraction were complimentary through all the treatments.
An alternative spectroscopic approach for the monitoring of microplastics in environmental samples
(2017)
The increasing pollution of terrestrial and aquatic ecosystems with plastic debris leads to the accumulation of microscopic plastic particles of still unknown fate. To monitor the degree of contamination and to understand the underlying processes of turnover, analytical methods are urgently needed, which help to identify and quantify microplastic (MP). Currently, costly collected and purified materials enriched on filters are investigated both by micro-infrared spectroscopy and micro-Raman. Although yielding precise results, these techniques are time consuming and restricted to sample aliquots in the order of micrograms precluding prompt and representative information on both, larger sample numbers and realistic material volumes. To overcome these problems, here we tested Raman and NIR process-spectroscopic methods in combination with multivariate data analysis.
For this purpose, artificial MP/soil mixture samples consisting of standard soils or sand with defined ratios of MP (0,5 – 10 mass% polymer) from polyethylene, polypropylene, and polystyrene were prepared. MP particles with diameters < 2 mm and < 125 µm were obtained from industrial polymer pellets after cryomilling. Spectra of these mixtures were collected by (i) a process FT-NIR spectrometer equipped with a reflection probe, (ii) by a cw-process Raman spectrometer and (iii) by a time-gated Raman spectrometer using fiber optic probes. For the calibration of chemometric models (partial least squares regression, PLSR) 5 – 10 spectra of defined MP/soil mixtures (consisting of 1 – 4 g material each) were collected. The obtained PLSR models served for the prediction of both, polymer type and content based on the spectra of “unknown” test samples.
Whereas MP could be detected by Raman spectroscopy in coastal sand at 0.5 mass%, in standard soils detection of MP was limited to 10 – 5 mass%. The sensitivity of Raman spectroscopy could be improved by mild treatment with hydrogen peroxide. FT-NIR was suitable for the investigation of MP in standard soils in the range of 5 – 1 mass%, however, here a non-linear effect was observed at higher polymer concentrations. When mixtures of several polymers at low concentration levels were milled together, FT-NIR spectroscopy yielded false positive polymers together with unprecise quantitative information. Recently, the investigation of “real-world” samples shall be tested and compared to the results obtained by micro-FTIR and micro-Raman.
Quantitative chemical analysis of airborne particulate matter (PM) is vital for the understanding of health effects in indoor and outdoor environments, as well as for enforcing air quality regulations. Typically, airborne particles are sampled over long time periods on filters, followed by lab-based analysis, e.g., with inductively coupled plasma mass spectrometry (ICP-MS). Within the EURAMET EMPIR AEROMET project, cascade impactor aerosol sampling was combined for the first time with on-site total reflection X-ray fluorescence (TXRF) spectroscopy to develop a tool for quantifying particle element compositions within short time intervals and even on-site. This makes variations of aerosol chemistry observable with time resolution of only a few hours and with good size resolution in the PM10 range. A proof of principles of this methodological approach and the comparison to standard methods within the scope of a field campaign will be presented. Secondly, aerosol sampling and TXRF analysis seems suitable for the quantification of elements in indoor aerosols as well and may provide an important enhancement of existing methods for the analysis of organic species in aerosols (such as sampling and TD-GC/MS). As an example, the TXRF analysis of particles emitted from laser printers under controlled conditions in an environmental test chamber will be presented.
While plastics have become indispensable in our daily lives over the last decades, the input into the environment has been increasing concurrently. Plastics often end up in the environment because of intensive use and poor waste management practice. They are subjected to aging and fragmentation and finally be deposited as microplastic particles or in short microplastics (MP). MP are defined as particles originating from synthetic polymers between 1 µm and 5 mm.
Although the pervasive abundance of MP in aquatic environments has been demonstrated comprehensively, less is known about the occurrence and fate of MP in terrestrial ecosystems. It is still unclear if soil functions as a MP source or a sink for aquatic environments. MP can either be transported into water bodies by soil erosion or be retained in soils. The few studies published are not comparable because of non-existent harmonized and standardized methods for sampling, sample preparation, and analysis.
For an assessment of a potential exposure situation of MP, the determination of a mass content in the soil is crucial. Consequently, spectroscopic methods like Raman or FTIR are not suitable, as they deliver information about the shape and size of individual particles. Therefore, we show the application of ThermoExtractionDesorption-GasChromatography-MassSpectrometry (TED-GC-MS) for MP analysis in the soil. In this method, the soil sample is heated up to 600 °C in a nitrogen atmosphere. The decomposition gases are sorbed on a solid phase, then transferred to a GS-MS system where they are desorbed, separated and identified. The method allows the rapid identification of individual polymers through the detection of specific decomposition products, but also the quantitative determination of the MP mass. Besides thermoplastics, elastomers originated from tire abrasion, can be detected.
In the present study, several terrestrial ecosystems in south-west Germany were systematically sampled. Subsequent sample preparation included sieving in fractions of 5-100 µm, 100-1000 µm, and 1-5 mm. MP were extracted by density separation using ZnCl2 solutions. The detection was done by TED-GC-MS measurements. Data of agricultural areas and floodplains are presented exemplarily. A quantitative assessment of highly occurring MP from littering as well as tire abrasion is conducted.
The talk is about microplastics in soil. Some results were presented with mainly state of the art according to microplastics in standardization.
Per- and polyfluoroalkyl substances (PFAS) have emerged over the course of the last twenty years as a global pollution issue. Altogether, there are currently more than 4700 known but only partly characterized fluorinated compounds identified by the Organization for Economic Cooperation and Development (OECD), as shown by a recently published classification proposal.
The ongoing production of new, yet unrestricted PFAS alternatives has become a major challenge for environmental routine analytics, since the state-of-the-art method LC-MS/MS relies on structural information and availability of isotope standards of the targeted compound. First reported by Miyake et al., fluorine sum parameters like adsorbable organic fluorine (AOF), extractable organic fluorine (EOF) and total fluorine (TF) can be applied to survey and detect the presence of large amounts of unidentified organofluorine compounds in environmental matrices. Today fluorine sum parameters have been established as a useful supplement to classic target-analytical approaches of PFAS and were implemented for the first time as a sum value “PFAS-total” in the recently revised Drinking Water Directive (2020/2184) by the European Commission.
In contrast, X-ray absorption near-edge structure (XANES) spectroscopy has been widely applied to identify low concentration of element-specific contamination without pre-treatment in environmental samples in the past. The XANES approach enables a penetration depth of approx. 1 µm (at fluorine K-edge energy; depending on the matrix) which is significantly deeper than for X-ray photoelectron spectroscopy (XPS; penetration depth approx. 10 nm). Furthermore, the method is fast, non-destructive and only simple preparation of the samples is required.
In our work, we combined both F-XANES spectroscopy and combustion ion chromatography (CIC) in order to present a new analytical perspective on the investigation of PFAS in environmental media and material samples.
Tire wear particles (TW) are generated by the abrasions of tires on the road surface through traffic. These particles can be transported by air and surface runoff and might also infiltrate the soil and consequently affect terrestrial ecosystems. The estimated tire wear (TW) emissions are immense, with 1.33 106 t a-1 in Europe. Despite this, only little is known about the environmental contents or the fate of TW.
One reason for this knowledge gap is the challenging analysis of TW in environmental samples. Detection of TW with spectroscopic methods is problematic due to high fluorescence interferences caused by contained black carbon. One analytical approach is to use zinc (Zn), a typical additive in tires, as a specific marker for the quantification of tire wear. However, any Zn originating from the sample matrix must be separated beforehand and requires elaborate sample preparation.
Car tires consist partly of synthetic rubbers, such as styrene-butadiene-rubber (SBR). This SBR could be identified and quantified via Thermal-Extraction-Desorption-Gas Chromatography-Mass Spectrometry (TED-GC-MS). This newly developed and fast screening method allows the simultaneous detection of microplastics and TW mass contents and requires minimal to no sample preparation. Firstly the sample is thermally extracted in a thermobalance under a nitrogen atmosphere. The resulting specific decomposition products are sorbed on a solid phase adsorber, which is then transferred to a GC-MS via an autosampler, where the products are desorbed, separated and identified. Cyclohexenylbenzene is used as a specific marker for SBR.
Here we investigated top layer soil samples, collected at the roadside of highly frequented German highways. Samples were analyzed without sample preparation, and SBR was detected in all investigated samples in mass contents ranging from 67.2 to 2230 mg kg-1. A correlation between SBR and Zn content in the soil was confirmed, while the correlation between SBR and Corg was hardly pronounced. We successfully demonstrated the application of TED-GC-MS as a screening method for tire wear in soil samples.
The present study will discuss these analytical results in detail as well as sampling parameters like sampling depth and distance to the roadside, and the effect of the particle size on the particle transport by water runoff and air.
Analytical Challenges for PFAS in Environmental Samples - Methods, Approaches and Applicability
(2022)
Per- and polyfluoroalkyl substances (PFAS) are anionic, cationic and zwitterionic synthetic products, in which the hydrogen atoms on the carbon skeleton of at least one carbon atom have been completely replaced by fluorine atoms and which include up to 1.7 M compounds, depending on the definition. As a result of continuous and prolific use, mainly in aviation firefighting foams, thousands of industrial and military installations have been found to contain contaminated soil, groundwater and surface water. Furthermore, because of the continuous contamination through PFAS containing commercial products, effluents and sewage sludge from WWTPs have been shown to be an important source of PFAS discharge into the aquatic environment. In the last few years, legacy PFAS (≥C4) have been found in various environments, including soil, water and wastewater, and their environmental pathways have been partly described.
Several long-chain PFAS species, and their respective salts are considered as persistent organic pollutants by the United Nations Stockholm Convention. These pollutants have been linked to altered immune and thyroid function, liver disease, lipid and insulin dysregulation, kidney disease, adverse reproductive and developmental outcomes, and cancer. A significant shift in the chemical industry towards production of short (C4-C7) and ultrashort (C1-C3) alternatives was observed in response to recently intensified regulations and restrictions on the use of long-chain (≥C8) PFAS. PFAS analysis in environmental samples is currently mainly done by liquid chromatography tandem mass spectrometry (LC-MS/MS). This efficient method is conducted in a targeted fashion analyzing a small subset of PFAS. The US EPA method for analysis of PFAS using LC-MS/MS for example currently lists 40 PFAS (≥C4). However, to get a better overview of the amount of “total PFAS,” sum parameter methods like total oxidizable precursor (TOP) assay and methods based on combustion ion chromatography (CIC) are in development. CIC results in data regarding the sum of absorbable organic fluorine (AOF) or extractable organic fluorine (EOF), which can also quantify other organically bound fluorine compounds such as fluorinated pesticides and pharmaceutical. Moreover, non-target and suspect screening mass spectrometry can be used to identify novel emerging PFAS and partly unknown fluorinated compounds in environmental samples. Furthermore, to analyze ultrashort PFAS (C1-C3), supercritical fluid chromatography (SFC), hydrophilic interaction chromatography (HILIC) and gas chromatography-mass spectrometry (GC-MS) are available, but further research is needed to develop reliable and accurate methods to quantify several ultrashort PFAS in environmental samples.
Additionally, for research purpose several spectroscopical methods like X-ray photoelectron spectroscopy (XPS), fluorine K-edge X-ray absorption near-edge structure (XANES)spectroscopy, particular induced gamma-ray emission (PIGE) spectroscopy and 19F nuclear magnetic resonance (NMR) spectroscopy are available.
A vast number of emerging pollutants is being detected in the environment. Another lingering problem are health-threatening contaminants, such as mycotoxins, that deteriorate food and feed.
Analytical methods, suitable for trace analysis, are needed that are desirably also fast, inexpensive and, if possible, robust, and portable. Immunoanalytical, i.e., anti-body-based technologies, which are available in a broad range of formats, can be profitably used here to analyze for the distribution and the concentration trends of contaminants.
Some of these formats are single-analyte but high-throughput methods. To use them wisely, indicator substances or sum parameters must be established to be used in screening approaches. Other methods are suitable to be performed on portable in-strumentation in the field (on-site), more precisely at the point-of-need. Furthermore, there are array technologies that allow for parallel analysis of several analytes of interest (multiplexing). The talk reports on methods and data for compounds con-sidered as anthropogenic contamination markers (the antiepileptic carbamazepine, the analgesic diclofenac, the anti-histaminic cetirizine, the steroid hormone estrone, the antimicrobial amoxicillin, the stimulants caffeine and cocaine, the endocrine disruptor bisphenol A, and a bile acid). Some work on mycotoxins (ochratoxin A and ergot alkaloids) is also presented. It is demonstrated that antibody-based technolo-gies have a huge potential to gain analytical insights at the point-of-need.

Gypsum (CaSO4∙2H2O), bassanite (CaSO4∙0.5H2O), and anhydrite (CaSO4) are essential evaporite minerals for the evolution of hyper-arid surface environments on Earth and Mars (Voigt et al. 2019; Vaniman et al. 2018). The formation mechanism of especially anhydrite has been a matter of scientific debate for more than a century (van’t Hoff et al. 1903). To date, there exists no model that can reliably predict anhydrite formation at earth’s surface conditions. While thermodynamics favor its formation, it is hardly achieved on laboratory time scales at conditions fitting either the Atacama Desert on Earth, or the surface of Mars (Wehmann et al. 2023). In light of most recent developments (e.g. Stawski et al. 2016), that advocate for a complex, non-classical nucleation mechanism for all calcium sulphates, we present an analysis of natural samples from the Atacama Desert to identify key features that promote the nucleation and growth of anhydrite under planetary surface conditions. Our analyses reveal at least three distinct anhydrite facies, with differing mineralogy and micro- to nano-structures. The facies are (1) aeolian deposits with sub-μm grain sizes, (2) (sub-)surface nodules that formed from aeolian deposits and (3) selenites with secondary anhydrite rims. Possible mechanisms of their formation will be discussed.
In urban waters, a multitude of organic micropollutants, often termed emerging pollutants, has been found over the last decades. Analytical methods suitable for trace analysis are needed that are desirably also fast, inexpensive and, if possible, robust and portable. Immunoanalytical, i.e., antibody-based, methods which are available in a broad range of formats, can be profitably used here to screen for the distribution and to monitor the trends of concentration levels of contaminants of emerging concern in the environment. Some of these formats are single-analyte but high-throughput methods. To use them wisely, indicator substances, sometimes called anthropogenic markers, should be selected and used in screening approaches, i.e., as indicators for contamination and the pre-selection of samples at which to have a closer look by multiplex methods like LC-MS/MS. Other methods are suitable to be performed on portable instrumentation in the field (on-site) or in facilities such as wastewater treatment plants for on-line monitoring of the treatment and elimination process. Furthermore, array technologies have been established that allow for parallel (multiplex) analysis of several analytes of interest.
The microtiter-plate based ELISA (Enzyme-linked Immunosorbent Assay) is the method of choice for the analysis of a large number of samples [1]. ELISAs are available to monitor for anthropogenic markers such as the antiepileptic carbamazepine, the analgesic diclofenac, the antihistaminic cetirizine, the steroid hormone estrone, the antimicrobial sulfamethoxazole, psychoactive caffeine and cocaine, the priority pollutant bisphenol A, and the bile acid isolithocholic acid. For on-site screening and monitoring, simpler formats, like mix-and-read assays, e.g., the Fluorescence Polarization Immunoassay (FPIA) [2] or Lateral-flow Immunoassays (LFIA) [3] are more suitable tools, the latter based on dipsticks or little cassettes, with which users have become very familiar during the COVID-19 pandemic via rapid antigen tests. The suitability of multi-analyte formats such as immunomicroarrays depends on the choice of a signal-producing system that provides small uncertainties and good reproducibility of the measurements. Bead-based (“suspension”) arrays, read out in flow cytometers, are a powerful platform for multiplex assays [4]. Electrochemical formats, run on portable devices, provide additional advantages as no light source is required. They are most promising for stand-alone analysers and biosensors [5].
The speed, low cost and on-site capabilities of these methods allow to gather a lot more data on anthropogenic compounds which enables to quantify inputs, differences in degradation power of elimination processes, dilution phenomena and a more precise image of individual water cycles which is demonstrated by several examples.
The overarching goal of this project is to develop a predictive model for efflux-mediated antimicrobial tolerance in bacterial multicellular assemblies. Our central hypostasis is that efflux pump activity causes emergent antibiotic tolerance of multicellular bacterial populations, through the interplay of efflux mediated spatial interactions and efflux-linked persistence. To test this hypothesis, we will use a combination of microscopy, microbial killing assays, computational modelling, and data analysis, integrating information from 3 types of multicellular assembly: colonies, cell-to-cell interactions in a monolayer microfluidic device, and 3D flow chamber biofilms. Building on our preliminary observations, we will experimentally characterize the link between colony structure and spatial patterns of efflux gene expression in strains that differ in their levels of efflux. We will develop a mathematical model to test whether local growth inhibition of neighbors due to effluxing cells, coupled with local environment-dependent regulation of efflux, can account qualitatively for these results. By including persister cell formation in our model we will predict, and measure, the emergent function of antimicrobial tolerance in our colonies. To fully understand how tolerance emerges from the interplay between efflux-mediated spatial interactions and efflux-linked persister cell formation, we need quantitative measurements at the single cell level. To this end, we will use a microfluidic setup with cells growing in a monolayer to qualify in detail the dependence of efflux expression and persister cell formation on nutrient conditions, the correlation between efflux and persister formation, and the spatial range of efflux-mediated neighbour growth inhibition. To predict and quantitatively understand the emergent multicellular function of tolerance, we will perform individual-based modelling of biofilm growth, using as input the parameters measured on the single-cell level with our microfluidics experiments. Our simulations will predict biofilm spatial structure development, patterns of efflux and persister formation and, ultimately, tolerance to antimicrobial challenge. These predictions will be directly tested in flow-cell biofilm experiments. We are currently generating acrAB-tolC knockout-strain, without efflux activity, and a strain with an inducible acrAB-tolC efflux pump. To distinguish the different strains under the microscope, they were labeled with genes encoding for different fluorescent proteins. All strains are currently characterized in terms of growth, minimum inhibitory concentration of different antimicrobial substances, colony morphology, and biofilm formation ability. On the theoretical side, we are currently working on modeling the system at various scales and degree of detail, ranging from coarse-grained continuum models to stochastic, individual-based models. Some exploratory work was doe to test existing software for individual-based modelling that may be adapted for our purpose. Furthermore, we are in the process of developing more coarse-grained models. This work involves some physiological modelling and literature search, focusing on working mechanisms of efflux pumps and kinetic models for import and export of antibiotics.
Biocides are in contact with soils through direct application and passive leaching from protected materials. Consequently, soil microorganisms are exposed to biocides even though they are not their primary targets. Soil is a large reservoir of microbial diversity and has been hypothesized to be a crucial factor for the evolution and spread of antimicrobial resistance.
Currently, there is little knowledge on how biocides used to protect materials affect the evolution and spread of resistance. Thus, our aim is to investigate the risk for the evolution of biocide resistance and cross-resistance to antibiotics. In addition, we aim to elucidate the affect of biocides on the spread of resistance via horizontal gene transfer (HGT).
In adaptive laboratory evolution experiments we cultured selected model soil microorganism with representative biocides under selection regimes with increasing and stable biocide concentrations followed by antibiotic and biocide cross-resistance determination. Moreover, we investigate if the selected biocides affect the rates of de novo mutations and HGT of plasmids that carry resistance genes among soil microorganism.
Our results show only small increases of biocide resistance during serial transfers under increasing biocide concentrations. One reason for this might be the narrow selective window for biocide resistance due to steep dose-response relationships. Furthermore, our results indicate that a stable low-level biocide regime did not select for high level cross-resistance to antibiotics and other biocides.
Moreover, material preservatives affected the rates of HGT via conjugation and the mutation rates at sub-inhibitory concentrations. The results will enable future risk assessment regarding resistance evolution for biocides used as material preservatives.

A wide range of analytical methods are used to estimate the plant-availability of soil phosphorus (P). Previous investigations showed that analytical methods based on the Diffusive Gradients in Thin films (DGT) technique provide a very good correlations to the amount of bioavailable nutrients and pollutants in environmental samples (Davison 2016, Vogel et al. 2017). However, the DGT results do not identify which P compound of the soil has the high bioavailability. But there are various spectroscopic techniques (infrared, Raman, P K-edge and L-edge XANES and P NMR spectroscopy) available to characterize P species in soils. Therefore, spectroscopic investigation of DGT binding layers after deployment allow us to determine the specific compounds. Nutrients such as phosphorus and nitrogen are often, together with other elements, present as molecules in the environment. These ions are detectable and distinguishable by infrared, P K- and L-edge X-ray absorption near-edge structure (XANES) and NMR spectroscopy, respectively. Additionally, microspectroscopic techniques make it also possible to analyze P compounds on the DGT binding layer with a lateral resolution down to 1 μm2. Therefore, species of elements and compounds of e.g. a spatial soil segment (e.g. rhizosphere) can be mapped and analyzed, providing valuable insight to understand the dynamics of nutrients in the environment.
Luminescence-based detection methods, ranging from fluorescence spectroscopy for photophysical and mechanistic studies over sensing applications, chromatographic separation techniques and the microarray technology with fluorescence detection to fluorescence microscopy, flow cytometry, single molecule spectroscopy, and molecular imaging to integrating sphere spectroscopy, are among the most widely used methods in the life and material sciences. This is due to e.g., their unique sensitivity enabling the detection of single molecules, potential for multiplexing, ease of combination with spatial resolution, and suitability for remote sensing. Many of these advantages are closely linked to the choice of suitable molecular and nanoscale fluorescent reporters, typically required for signal generation. This includes organic dyes without and with sensor function, fluorophore-encoded polymeric and silica nanoparticles as well as nanocrystalline systems like semiconductor quantum dots and upconversion phosphors, emitting in the visible (vis), near-infrared (NIR), and IR (infrared). Current challenges present the environment sensitivity of most fluorophores, rendering fluorescence spectra, measured intensities/fluorescence quantum yields, and fluorescence decay kinetics matrix-dependent, and instrument-specific distortions of measured fluorescence signals that need to be considered for quantification and comparability of data, particularly fluorescence spectra.
Here, current applications of luminescence-based methods and different types of reporters will be presented. In this context, suitable spectroscopic tools for the characteri-zation of the optical properties of fluorescent reporters and fluorophore-encoded microparticles, analytical tools for the determination of the surface chemistry of different types of particles, and different multiplexing strategies will be discussed.
Microplastics are widely distributed in aquatic and terrestrial environments, up to now less is known on toxicological impacts under realistic conditions. Research so far has focused mainly on the impacts on organisms by fresh plastic fragments or beats. However, plastic materials found in the environment are mainly a mixture of different polymers and additives and particularly degraded during aging processes. Although especially oxidized degradation products might escape from plastic materials during aging, there is a lack of information on potentially adverse effects on aquatic biota. The latter is of particular interest as oxidized chemicals become more water soluble due to higher polarity and are more bio-available, therefore.
The present study focused on plastic leachates of polystyrene (PS) and polylactic acid (PLA), which were derived from alternating stress by hydrolysis and UV radiation. Test specimen of PS, PLA or PS/PLA blends (each 50 %) were alternatingly maintained in water at 45 °C for five days and UV radiated at 45 °C for two days, for in total six weeks. Ecotoxicological effects of potentially generated degradation products of plastic materials in the storage water (mineral water, pH: 7.2) were detected by algae growth inhibition tests with Desmodesmus suspicatus and photosystem II inhibition tests with Raphidocelis subcapitata.
Results clearly indicate inhibitory effects on algae growth by contaminants in the storage water of stressed plastic materials with increasing growth inhibition of proceeding hydrolysis and UV stress times. Here, different plastics cause variable responses of algal growth. First chemical analyses indicate dissolved monomers and their oxidation products of plastic materials as possible driver of detected ecotoxicological effects, since detected microplastic particles do not seem to harm algae.
The existing data highlight the relevance of plastic aging as a framework for microplastic ecotoxicity evaluation and allow a proof of concept.
Anemophilous plants produce pollen grains, which have to be monitored to provide an information network for persons suffering from an allergy. The current conventional characterization and identification of pollen is performed by time-consuming microscopic examinations on the basis of the genus-specific pollen shape and size. These examinations are in need of proficient researchers, are not statistically validated, and additionally rely on relatively inaccurate observations of the pollination process. A variety of new analytical approaches have been proposed in order to develop a fast and reliable pollen identification using specific molecular information. Recently, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) was initially applied for the rapid investigation of such complex biological samples. The combination of the obtained patterns of pollen mass spectra and multivariate statistics provide a powerful tool for identifying taxonomic relationships. Here promising results obtained by the reduction and optimization of sample preparation prior to MALDI-TOF MS mapping are presented. Based on a novel application of conductive carbon tape as target, the signal information of the species-specific mass peak patterns of pollen was enhanced. This leads to a clear separation in a subsequent pattern analysis, which is important when analyzing pollen grains in natural species mixtures. As a proof-of-concept, artificial pollen mixtures were investigated by MALDI imaging and evaluated by innovative multivariate data analysis strategies, to assign individual pollen species in the mixtures. Our results can be used to improve the taxonomic differentiation and identification of pollen species and might be useful for the development of a routine method to identify pollen based on mass spectrometry.

Tonnen Hausmüllverbrennungs-aschen (HMVA) an.
The challenge of the project ASHES is focused on the recycling of nutrients from residues of thermochemical processing of by-products of sugar cane industry (bagasse/straw) in Brazil to increase the energy efficiency of thermal conversion and to enable the recycling of process ashes as fertilisers. Dry lignocellulosic biomass (straw, bagasse) are combusted, gasified and subsequently combined with post-thermochemical treatment in the AshDec process to increase the plant availability of phosphorus. Different fertilizer formulations are granulated/pelletized and tested regarding their storage/handling characteristics.
Nondestructive testing of gas turbine blades is essential for their maintenance and service process which is critical to ensure both safety and efficiency of these highly stressed parts. In this presentation, a novel ultrasonic testing method is explored in order to acquire part thickness information in the turbine blade’s airfoil. In established industry processes, the measurements are mainly carried out manually and only at a few specific positions of the inspected parts. The proposed method scans the part using a robot arm guiding an ultrasonic array sensor. For ultrasonic coupling to the complex-shaped surface geometry, the inspected part and sensor are immersed into water. A two-step TFM[1, 2] (Total Focusing Method) approach is used to reconstruct the outer and inner surfaces subsequently from the ultrasonic raw data, which are acquired using the FMC[3] (Full Matrix Capture) measurement principle. For each sensor position, the location and geometry of the outer surface is first identified and then used to create an image of an area inside the material. From that image, the inner surface is reconstructed. Finally, part thickness information is deducted from merging location data of inner and outer surface. The result is a high resolution, high precision mapping of the inspected part’s wall thickness.
Offshore wind energy plays an important role for the desired transition towards a carbon dioxide free industry within the next decades. However, the grounding of the offshore wind plants governs the overall installation process besides bureaucratic bottlenecks.
The application of lightweight principles in steel construction, e.g. the usage dissolved load-bearing structures, so called Jackets foundations, offer great potential in reducing the resource consumption, especially with respect to the needed amount of steel.
In this context this paper focuses on a fully digitalization of the welding manufacturing and as well as testing chain to enable a fully automated manufacturing as well as quality assessment of tubular nodes as key element of Jackets foundation structures. Furthermore, the relationship between the seam shape geometry and resulting fatigue strength is evaluated by numerical methods incorporating bionic principles. It is shown that tubular nodes can be welded fully automatically taking geometry tolerances into account. Moreover, the seam shape could be manufactured as requested by the numerical models which offers great potential for an extended lifetime. Subsequently, the improvements in resource efficiency and reduction of carbon dioxide emissions are evaluated by a life-cycle-assessment.
Essential processes in the establishment and maintenance of rock biofilms include photosynthesis, production of extracellular polymeric substances, substrate penetration and atmospheric nutrient enrichment. Functional diversity is supported by a complex subaerial biofilm (SAB) community of heterotrophic and phototrophic microorganisms. Stress tolerant and melanised Ascomycetes dominate heterotrophic SABs while diverse algae and cyanobacteria comprise the phototrophic consortia. Laboratory simulation of SABs permits use of molecular-genetic methods coupled with geochemical and microscopic analyses to study weathering. Our in vitro model includes two free-living and symbiosis-competent, genetically tractable microorganisms: the cyanobacterium Nostoc punctiforme and the microcolonial rock-inhabiting fungus Knufia petricola. This genetically amenable cyanobacterium/fungus model biofilm allows development of quantitative methods tailored to the natural diversity of SABs. Precise, reproducible studies using this model biofilm have shown that both the melanised fungus, as well as the combined bacterial/fungal system, enhances the weathering of minerals. Geochemical signatures of these in vitro rock biofilms can now be obtained and compared with bacterial/fungal mutants of varied EPS composition and substrate penetration patterns. And finally, precise study of the model cyanobacterium/fungus biofilm will Permit prediction of the effects of conservational treatments.
