Reliable analysis of chemical indicators in water, sediment and soil samples for the purpose of environmental pollution assessment poses one of the greatest analytical challenges, having in mind the complexity of sample matrix and low concentrations of pollutants. Organics (pesticides, PAHs, PFOS, etc.) and heavy metals (Hg, Cd, Ni, Pb and As) represent target parameters. Laboratories performing sampling and tests in this field regulated by respective EU directives, need strong support in order to establish a quality system. It is necessary to provide appropriate calibrators i.e. matrix CRMs relates to the unique sample matrices representing typical samples in the geomorphological and anthropological sense. In addition to that, bearing in mind the complexity and instability of environmental samples, it is very difficult to obtain appropriate referents materials with no local providers.
Our project is aiming to develop capacity to produce CRMs for environmental analysis by transferring the theoretical and practical know-how between the partners and combining their skills to focus on environmental CRM production in accordance with ISO Guide 34. Our project will have an impact on environmental monitoring in the partnering countries and on the scientific community, who will use the newly developed reference materials. Furthermore, partners will develop strategies for producing new CRMs either on their own or in cooperation. This will lead to regional CRM producers serving scientific and official laboratories.
This project aims to develop capacity to produce certified reference materials (CRMs) for environmental analysis by transferring know-how between the partners and combining their skills to focus on environmental CRM production. The production process includes good manufacturing practices for processing materials, method development, the validation and application of homogeneity, stability and characterisation tests, the calculation of individual uncertainties (between-unit inhomogeneity, long term stability, characterisation) and combination of uncertainties to determine overall uncertainty of the matrix reference materials. An inter laboratory comparison registered as a EURAMET project is set as the ultimate project outcome, confirming the partners’ capabilities in applying newly acquired skills.
Quantification of microbial load in diesel storage tanks using culture- and qPCR-based approaches
(2016)
Microbial contamination of fuels, associated with a wide variety of bacteria and fungi, leads to decreased product quality and can compromise equipment performance by biofouling and microbiologically influenced corrosion of pipelines and storage tanks. Detection and quantification of biomass are critical in monitoring fuel systems for an early detection of microbial outbreaks. The aims of this study are (i) to quantify bacterial and fungal contamination in samples from diesel storage tanks of petrol stations, using both culture dependent- and culture independent (qPCR) approaches, and (ii) to analyse the diversity of cultivable diesel-contaminating microorganisms with the purpose to create a strain collection for further use in biodeterioration experiments. Both methodological approaches revealed a high microbial contamination in all studied samples, with the bacterial load being much higher than the fungal load. The diversity of cultivable microorganisms was rather low. Based on criteria of abundance and fuel degradation potential, the most relevant microorganisms were identified as bacteria of genera Bacillus, Citrobacter, Burkholderia and Acetobacter, the filamentous fungi Paecilomyces variotii and Pseudallescheria boydii, and a Dipodascaceae yeast. Furthermore the validity and utility of qPCR-based methods are discussed.
Most microorganisms live in environments where nutrients are limited and fluctuate over time. Cells respond to nutrient fluctuations by sensing and adapting their physiological state. Recent studies suggest phenotypic heterogeneity in isogenic populations as an alternative strategy in fluctuating environments, where a subpopulation of cells express a function that allows growth under conditions that might arise in the future. It is unknown how environmental factors such as nutrient limitation shape phenotypic heterogeneity in metabolism and whether this allows cells to respond to nutrient fluctuations. Here, we show that substrate limitation increases phenotypic heterogeneity in metabolism, and this heterogeneity allows cells to cope with substrate fluctuations. We subjected the N2-fixing bacterium Klebsiella oxytoca to different levels of substrate limitation and substrate shifts, and obtained time-resolved single-cell measurements of metabolic activities using nanometre-scale secondary ion mass spectrometry (NanoSIMS). We found that the level of NH4+ limitation shapes phenotypic heterogeneity in N2 fixation. In turn, the N2 fixation rate of single cells during NH4+ limitation correlates positively with their growth rate after a shift to NH4+ depletion, experimentally demonstrating the benefit of heterogeneity. The results indicate that phenotypic heterogeneity is a general solution to two important ecological challenges - nutrient limitation and fluctuations - that many microorganisms face. Currently, we use NanoSIMS to develop a new approach that defines functionally-relevant, phenotypic biodiversity in microbial systems. In the last part of my presentation, I will highlight why the concept of phenotypic diversity is relevant for the understanding of antimicrobial resistance.

Several restoration projects of stained-glass windows have been performed in Lower Silesia (Poland) since 2010. The aim of the projects was to protect stained-glass windows against environmental impact of industial pollutants and acid rain by installing a protective glazing. The usefulness of protective glazing has been proved by climate measurements and determination of environmental impact before and after installation.
Photodegradation of the „novel“ brominated flame retardant Tris-(2,4,6-tribromophenoxy)-s-triazine
(2016)
Brominated flame retardants (BFRs) comprise a class of structurally diverse compounds that are functionally related and used in materials to reduce the flammability of polymers. Due to their global distribution, potential persistence, bioaccumulation and toxicity most of the BFRs of the first generation are banned or restricted, and replaced by new ones. However, based on similar properties these new compounds may also pose a serious risk by causing adverse effects to human health and the environment. These alternative BFRs were grouped into two classes: „novel” and „emerging” BFRs. Emerging BFRs are defined as compounds that have been identified in any environmental compartments whereas novel BFRs are only detected in materials and/or goods above 0.1 wt.-%.
The environmental fate of these alternative BFRs is partially unknown. Over the past years a multitude of studies on the photodegradation of BFRs in environmental matrices have been conducted to assess the photochemical behavior and fate in the environment. The photolytic debromination reaction is particularly induced among BFRs by UV exposure. The novel brominated triazine-based flame retardant tris-(2,4,6-tribromophenoxy)-s-triazine (TTBP-TAZ) is a BFR that is mainly used in acrylonitrile butadiene styrene and high impact polystyrene.
Ballesteros-Gómez et al. detected TTBP-TAZ in 8 of 13 plastic parts of consumer products and in 9 of 17 indoor dust samples but the photochemical behavior and fate is not known until now. The results of the present study, focused on photodegradation processes were performed for the first time for TTBP-TAZ to identify the photolysis intermediates and products to assess the environmental fate.
The scope of our study was to prepare Fe-based highly active catalysts for water treatment processes. The reactive nanomaterials were
synthesized by chemical precipitation; the post-synthesis coating of the as obtained nanostructures was achieved by capping the magnetic cores with a
polymeric surfactant, polyethylene glycol (PEG). The resulted Fe3O4/PEG nanoparticles were further used to immobilize two types of reactive materials on
their surface, either ferrous oxalate (FO) or ferric citrate (FC), yielding the final products – two green Fenton-like heterogeneous nanocatalysts. The
sensitized catalysts were characterized by several microstructure characterization techniques such as X-ray diffraction (XRD), vibrating sample
magnetometry, BET, scanning electron microscopy (SEM), and energy dispersive X-ray spectroscopy (EDX). All experiments were performed on a
laboratory scale set-up. Important variables such as effect of hydrogen peroxide concentration, UV light, the amount of catalyst and contact time for the
catalytic wet peroxide oxidation (CWHPO) of BPA were examined. The results indicate that the oxidation rate increased under UV light in the presence of
low-concentration H2O2.
Quality control requirements imposed on assays used in clinical diagnostics and point-of-care-diagnostic testing (POCT), utilizing amplification reactions performed at elevated temperatures of 35 to 95 °C are very stringent. As the temperature of a reaction vessel has a large impact on the specificity and sensitivity of the amplification reaction, simple tools for local in situ temperature sensing and monitoring are required for reaction and assay control. We describe here a platform of stem-and-loop structured DNA hairpins (molecular beacons, MBs), absorbing and emitting in the visible and red spectral region, rationally designed for precise temperature measurements in microfluidic assays for POCT, and their ap-plication for temperature measurements in a common DNA-based molecular biological assay utilizing thermophilic helicase-dependent amplification (tHDA). Spectroscopic studies of these MBs, rationally designed from DNA se-quences of different thermal stabilities, chosen not to interact with the DNA probes applied in the nucleic acid amplification assay, and temperature-dependent fluorescence measurements of MB-assay mixtures revealed the suitability of these MBs for temperature measurements directly in such an assay with a temperature resolution of about 0.5 °C without interferences from assay components. Combining two spectrally distinguishable MBs provides a broader response range and an increase in temperature sensitivity up to 0.1 °C. This approach will find future application for temperature monitoring and quality control in commercialized diagnostics assays using dried reagents and microfluidic chips as well as assays read out with tube and microplate readers and PCR detection systems for temperature measurements in the range of 35 to 95 °C.
Pharmacologically active compounds are omnipresent in contemporary daily life, in our food and in our environment. The fast and easy quantification of those substances is becoming a subject of global importance. The fluorescence polarization immunoassay (FPIA) is a homogeneous mix-and-read format and a suitable tool for this purpose that offers a high sample throughput. Yet, the applicability to complex matrices can be limited by possible interaction of matrix compounds with antibodies or tracer.
Caffeine is one of the most frequently consumed pharmacologically active compounds and is present in a large variety of consumer products, including beverages and cosmetics. Adverse health effects of high caffeine concentrations especially for pregnant women are under discussion. Therefore, and due to legal regulations, caffeine should be monitored. Automated FPIA measurements enabled the precise and accurate quantification of caffeine in beverages and cosmetics within 2 min. Samples could be highly diluted before analysis due to high assay sensitivity in the low μg/L range. Therefore, no matrix effects were observed.
The antiepileptic drug carbamazepine (CBZ) is discussed as a marker for the elimination efficiency of wastewater treatment plants and the dispersion of their respective effluents in surface water. The development of a FPIA for CBZ included the synthesis and evaluation of different tracers. Using the optimum tracer CBZ-triglycine-5-(aminoacetamido) fluorescein, CBZ concentrations in surface waters could be measured on different platforms: one sample within 4 min in tubes or 24 samples within 20 min on microtiter plates (MTPs). For this study, a commercially available antibody was used, which led to overestimations with recovery rates up to 140% due to high cross-reactivities towards CBZ metabolites and other pharmaceuticals.
For more accurate CBZ determination, a new monoclonal antibody was produced. In this attempt, methods for improving the monitoring during the production process were successfully applied, including feces screening and cell culture supernatant screening with FPIA. The new monoclonal antibody is highly specific for CBZ and showed mostly negligible cross-reactivities towards environmentally relevant compounds. Measurements at non-equilibrium state improved the sensitivity and selectivity of the developed FPIA due to slow binding kinetics of the new antibody. Additionally, this measure enables for CBZ determination over a measurement range of almost three orders of magnitude. The comprehensively characterized antibody was successfully applied for the development of sensitive homogeneous and heterogeneous immunoassays.
The new antibody made the development of an on-site measurement system for the determination of CBZ in wastewater possible. After comprehensive optimization, this automated FPIA platform allows the precise quantification of CBZ in wastewater samples only pre-treated by filtration within 16 min. Recovery rates of 61 to 104% were observed. Measurements in the low μg/L range are possible without the application of tedious sample preparation techniques.
Different FPIA platforms including MTPs, cuvettes and tubes were successfully applied. For the choice of the right format, the application field should be considered, e.g. desired sample throughput, usage for optimization or characterization of antibodies or if a set-up for routine measurements is sought for. For high sample throughput and optimization, FPIA performance on MTPs is advantageous. The best results for the application to real samples were obtained using kinetic FP measurements in cuvettes.
Immunoassays are suitable tools for high-throughput screenings. The prerequisite for accurate determinations by these methods is the selection of an excellent antibody. The production and selection of monoclonal antibodies is usually a tedious process. In this study, new strategies for improving antibody production and characterization were applied. This includes the monitoring of the immunization progress in mice through antibodies extracted from feces, which allows a time-resolved and animal-friendly monitoring of the immune response. Additionally, fluorescence polarization immunoassay (FPIA) could be successfully applied for fast and easy examination of cell culture supernatants and the investigation of antibody/antigen interactions including kinetics and fluorescence properties. These methods simplify the selection of the optimal antibody. As a target analyte, carbamazepine was chosen. This is a widely used antiepileptic drug which also frequently occurs in the environment. The new antibody enables CBZ determination in the concentration range 0.66–110 µg L-1 within 10 min using a high-throughput microtiter plate-based FPIA, and between 1.4 and 79 µg L-1 within 5 min applying an automated cuvette-based FPIA instrument, and from 0.05–36 µg L-1 using ELISA. The measurements were performed at a non-equilibrium state which improved the sensitivity and selectivity of the assays. Due to low cross-reactivity especially towards the main CBZ metabolite and other pharmaceuticals (<1%), this antibody gives the opportunity for application in medical and environmental analyses.

GFS).
Only a few myxobacteria are known to date that are classified as marine, owing to their salt dependency. In this study, the salt tolerance mechanism of these bacteria was investigated. To this end, a growth medium was designed in which the mutated Escherichia coli strain BKA13 served as sole food source for the predatory, heterotrophic myxobacteria. This enabled measurement of the osmolytes without any background and revealed that the closely related strains Enhygromyxa salina SWB007 and Plesiocystis pacifica SIR-1 developed different strategies to handle salt stress. Ple. pacifica SIR-1, which was grown between 1 and 4% NaCl, relies solely on the accumulation of amino acids, while Enh. salina SWB007, which was grown between 0.5 and 3% NaCl, employs, besides betaine, hydroxyectoine as the major compatible solute. In accordance with this analysis, only in the latter strain was a locus identified that codes for genes corresponding to the biosynthesis of betaine, ectoine and hydroxyectoine.

The repair of earthen structures is not an easy task. Earth as a construction material has comparatively weak mechanical properties. It is also susceptible towards liquid water, were it can lose cohesion very quickly if certain moisture contents are exceeded. Repair concepts of structures built with such materials are therefore challenging. Historical earthen structures often exhibit damages in form of extensive cracking, which may have been caused by static or dynamic loads. Frequently these cracks are insufficiently or inappropriately repaired because of lack of knowledge and / or technology. In particular, the behaviour of crack repair by grouting poses a challenge in earthen materials and demands specific requirements for the grouting mortar, such as low water content, good water retention, low shrinkage. Grouting materials require additional specifications such as compatible strengths and Young’s modulus as well as good adhesion to the earthen substrates. In addition, grouts have to be sufficiently flowable to fill small cracks and voids without segregation or bleeding. Therefore, the rheological behaviour has to be well understood and controlled to gain the desired effects.
The repair of cracks in earthen buildings is traditionally done by stuffing manually mortar into the gap. Naturally, this method is only usable for cracks with large widths. Another disadvantage is that cracks going through thicker walls cannot be completely reached by the tools used for stuffing the mortar into the crack. Lime based grouts for earthen materials were usually used for re-attachment of plasters but less for structural reasons. Due to the nature of earthen materials grouts based on formulated or hydraulic lime (according to the definition in EN 459-1) have to meet considerable demands on a variety of properties, which are related to compatibility, durability and in particular to the ability of being injected. The goal of this study was to create a grout, which can be used to re-establish structural continuity in cracked earthen masonry or other massive earth walls (rammed earth and cob) with the focus on grouting cracks. The grout material was based on hydrated lime (calcium hydroxide) with additions of pozzolana and lime stone filler.
V. (DVL).
In-plane shear behaviour of earthen materials panels strengthened with polyester fabric strips
(2016)
An experimental investigation was carried out to study the in-plane shear behaviour of earthen material Panels strengthened with polyester fabric strips. Strengthened panels were developed to exploit the strength potential of earthen materials and to solve its lack of tensile strength, significantly improving not only strength but also ductility. Three earthen materials were considered: cob, earth block masonry (EBM) and rammed earth (RE). As first approach the strengthening configuration, based on different adhesive materials, was tested only for cob panels. As part of the study the results of a big testing campaign of unstrengthened Panels were considered. Seven strengthened panels were tested in diagonal compression/shear.A unique reinforcement orientation was used. The results of these tests are presented in this paper, and include the load-displacement behaviours, crack patterns, failure modes. The results showed that the reinforcement was the most effective in EBM panels, with increase in strength and ductility observed. In RE and cob panels the reinforcement did not likely contribute significantly to the shear resistance, due to a lack of embedment length of the strips. Instead, in EBM it was likely that the vertical reinforcement acted in tension to restrain shear induced dilation and to restrain sliding.

The paper presents a comparison between two different numerical modelling approaches aimed to simulate the in-plain behaviour of rammed earth walls, namely under axial, diagonal and cyclic shearcompression loading. In the first part of the study the mechanical characterisation of wallets tested under uniaxial compression and diagonal compression and walls tested under in-plane cyclic shear-compression loading is presented. The results were used to implement and validate the finite element simulations. The numerical modelling of the rammed earth samples tested is then discussed in the second part. A non-linear constitutive law based on the total strain rotating crack model (TSRCM) was employed as implemented in the DIANA® software [1]. The aim of the numerical analyses presented here is to simulate the behaviour of rammed earth under different inplane loading conditions. For the wallets, tests under static loading both macro- and micro-modelling approaches were considered for the simulation of the experimental tests. For the walls subjected to cyclic loading only the micro-modelling approach was applied for the simulation of the experimental tests. The respective FEM model was calibrated with the experimental results. The rammed earth layers were represented by continuum elements, the contact surfaces between layers by interface elements. This approach allowed assessing the influence of the apparent weakness of the interfaces between layers on the shear behaviour of rammed earth. The goal of the numerical simulation of the cyclic tests was to establish the adequacy of common analytical methods (e. g. used for masonry) applied to the analysis of rammed earth. Rammed earth exhibits brittle characteristics similar to masonry materials and is used in geometrical typologies, such as walls, common in masonry construction.
Lime based grouts for earthen materials were usually used for re-attachment of plasters but less for structural reasons.Due to the nature of earthen materials grouts based on formulated or hydraulic lime (according to the definition in EN 459-1) have to meet considerable demands on a variety of properties, which are related to compatibility, durability and in particular to the ability of being injected.
Biocide leaching was investigated for acrylate and vinyl acetate based coatings containing biocides either as liquid formulations or microcapsules. Laboratory and field tests indicate similar relations of results on carbendazim, diuron, terbutryn and OIT from different coatings in laboratory and field experiments. Emissions were higher in EN 16105 laboratory tests than from vertically installed test specimens exposed to weathering, and slower for microencapsulated biocides. Competing processes that cause losses of active substances can occur in both tests, but to a higher degree in field experiments. Emission curves related to runoff water were similar in repeated field experiments. Relative humidity, temperature and global radiation were identified as meteorological factors that affect leaching by complex interaction besides the amount of driving rain.
Many wooden artworks are contaminated by DDT (dichlorodiphenyltrichloroethane) as a result of a surface treatment by means of Hylotox-59©. The liquid preservative was used until the end of the 80s. DDT crystal structures are formed on the wood surfaces by the "blooming" of chlorine compounds. Contaminated waste wood with natural biocide ageing, gilded and wood carved elements of an old picture frame and wooden samples with paint layers were provided by the Schlossmuseum Sondershausen.
The removal of DDT was evaluated employing femtosecond and nanosecond laser radiation and cold atmospheric plasma technique using different working gases (air, nitrogen, and argon).
Before laser application, a chlorine measurement is done by X-ray fluorescence (XRF) analysis as reference. After laser processing, the XRF analysis is used again at the same surface position to determine depletion rates. Additionally, a documentation and characterization of the sample surface is done before and after laser and plasma treatment using optical microscopy. For plasma processing with various systems a chlorine measurement is done by gas chromatographic-mass spectrometry (GCMS) analysis.
For laser treatment a depletion of the chlorine concentration of 55-70% and 75% was achieved for 1064-nm nanosecond pulses and 800-nm femtosecond pulses, respectively. For the application of 30-fs laser pulses, no crystalline DDT residues remain on the sample surfaces. This holds also for the plasma processing with nitrogen as working gas.
The vibrations of soil and foundations are demonstrated for different types of loading. Train-induced ground vibrations are studied in a measurement campaign where a test train has run with regularly varied speeds. The measured train-induced soil vibration at 2 to 100 m distance from the track is compared with the wave propagation due to hammer excitation and with the theoretical wave field. The strong influence of the soil and the train speed on the amplitudes and frequencies of the vibration has been analysed for passages of the locomotive and the carriages. - The generation of ground vibration by strong explosions has been studied on a large testing area with sandy soil. The propagating waves were measured in a regular grid of measuring points in 10 to 1000 m. Therefore, the dominance of certain waves at certain distances and the changes of compressional waves and Rayleigh waves could clearly be observed. The results are compared with impulse hammer measurements in the range of 5 to 50 m. - A drop test facility has been built on the testing area of the Federal Institute of Materials Research and Testing (BAM). Heavy masses (containers) of up to 200 t can be dropped from 10 m height on a big reinforced concrete foundation. The foundation was instrumented by accelerometers, strain gauges and pressure cells to give information about the loading condition and by geophones to measure the vibration of the surrounding soil and building. Both excitation processes, the release of the mass and the impact, produce high vibration amplitudes. On a smaller drop foundation, the influence of the drop height and the target stiffness has been studied more systematically.

Essential processes in the establishment and maintenance of rock biofilms include photosynthesis, production of extracellular polymeric substances, substrate penetration and atmospheric nutrient enrichment. Functional diversity is supported by a complex subaerial biofilm (SAB) community of heterotrophic and phototrophic microorganisms. Stress tolerant and melanised Ascomycetes dominate heterotrophic SABs while diverse algae and cyanobacteria comprise the phototrophic consortia. Laboratory simulation of SABs permits use of molecular-genetic methods coupled with geochemical and microscopic analyses to study weathering. Our in vitro model includes two free-living and symbiosis-competent, genetically tractable microorganisms: the cyanobacterium Nostoc punctiforme and the microcolonial rock-inhabiting fungus Knufia petricola. This genetically amenable cyanobacterium/fungus model biofilm allows development of quantitative methods tailored to the natural diversity of SABs. Precise, reproducible studies using this model biofilm have shown that both the melanised fungus, as well as the combined bacterial/fungal system, enhances the weathering of minerals. Geochemical signatures of these in vitro rock biofilms can now be obtained and compared with bacterial/fungal mutants of varied EPS composition and substrate penetration patterns. And finally, precise study of the model cyanobacterium/fungus biofilm will Permit prediction of the effects of conservational treatments.
The characterization of the optical properties of photoluminescent systems, that scatter, like dispersions of nanoparticles with sizes exceeding about 25 nm or solid nanophosphors is of increasing importance for many applications in the life and material sciences. Examples present nanoscale optical reporters and dye-doped microparticles for bioimaging, fluorescence assays or DNA sequencing as well as nanocrystalline emitters like semiconductor quantum dots and rods or lanthanide-based nanophosphors embedded into solid matrices for solid state lighting, display technologies, or barcoding/security applications. The assessment and comparison of material performance as well as the development of rational design strategies for improved systems require spectroscopic tools, which enable the determination of the signal-relevant optical properties like photoluminescence quantum yields and brightness values.
This encouraged us to built up an integrating sphere setup enabling absolute measurements of photoluminescence spectra and quantum yields of transparent and scattering photoluminescent dispersions and solid samples in different measurement geometries, i.e., direct and indirect illumination and the combination of both geometries and perform first measurements with selected emitters. Here, the design of this setup is presented and first recommendations concerning suitable measurement geometries are given.
A reversed phase high performance liquid chromatography coupled to an inductively coupled plasma mass spectrometer (HPLC-ICP-MS) approach in combination with isotope dilution analysis (IDA) for the separation and parallel quantification of nanostructured and ionic silver (Ag) is presented. The main focus of this work was the determination of the ionic Ag concentration. For a sufficient stabilization of the ions without dissolving the nanoparticles (NPs), the eluent had to be initially optimized. The determined Ag ion concentration was in a good agreement with results obtained using ultrafiltration. Further, the mechanism of the NP separation in the HPLC column was investigated. Typical size exclusion effects were found by comparing results from columns with different pore sizes. Since the recovery rates decreased with increasing Ag NP size and large Ag NPs did not elute from the column, additional interactions of the particles with the stationary phase were assumed. Our results reveal that the presented method is not only applicable to Ag NPs, but also to gold and polystyrene NPs. Finally, IDA-HPLC-ICP-MS experiments in single particle mode were performed to determine the particle cut-off size. The comparison with conventional spICP-MS experiments resulted in a similar diameter and particle size distribution.
The interface between materials and the environment is populated by microorganisms which organize on surfaces to form specially adapted and resistant microbial associations, so-called biofilms. Surface-bound biofilm growth is associated with substantial secretion of metabolic products that can significantly influence material corrosion. Here, a group of material-inhabiting fungi are presented, which colonize and weather air-exposed materials such as building façades, roofs and solar systems. A selected model fungus is used to carry out genetic and molecular biology experiments, which are microbiologically cutting-edge and allow for the development of novel time-saving techniques for material testing. The degree of material damage under the influence of this model fungus shows a measure of the aggressiveness of the biofilm under defined environmental conditions - and thus explores fundamental components of the interaction between material and biofilm.
The European Water Framework Directive (WFD) 2000/60/EC specifies a list of 33 priority water pollutants. For these substances, Environmental Quality Standards (EQS), equivalent to the maximum allowable concentrations, had been defined at very low levels.
EQS values refer to the whole water body, i.e. include contaminants which are associated to suspended solids or colloids present in the natural water. For several priority pollutants primary analytical methods capable of measuring the target analytes at EQS level in natural water are still missing. The project has developed and validated such methods for three selected compounds, namely Tributyltin (TBT), Polybrominated Diphenylether (PBDE) and Polycyclic Aromatic Hydrocarbons (PAH).
The occurrence of persistent organic pollutants like polychlorinated biphenyls (PCB) and organochlorine pesticides (OCP) in the environment is still a matter of concern, even years after their use was abolished. Hence, threshold values are regulated by law which reference a number of national and international standards for determination of said contaminants. In this context, soil is an especially difficult matrix, because several defining parameters can vary in a wide range. Organic matter content in particular has a crucial influence on extractability of contaminants because it largely governs formation of non-extractable or bound residues.
For this study, four soils of different total organic carbon (TOC) content were generated from uncontaminated reference soil (RefeSol 01-A) and compost which were spiked to contaminant levels representative of the Bundes-Bodenschutz-Verordnung (BBodSchV). These test materials were extracted using accelerated solvent extraction (ASE), Soxhlet extraction, and liquid-liquid extraction. For the latter method, samples were initially extracted for 15 minutes employing acetone. The follow-up extraction step was carried out using cyclohexane and the extraction time varied between 15 minutes and 16 hours. After clean-up, extracts were analysed via gas chromatography-mass spectrometry (GC MS) and with a gas chromatograph equipped with an electron capture detector (GC ECD).
Statistical analysis of the experimental values showed that recovery was not dependent on matrix TOC content. Also, no clear preference for a single method could be detected. Concerning liquid-liquid-extraction, it is merely observed that prolonged extraction time results in slightly higher recoveries, while moderate extraction times appear to yield the most robust results. Although Soxhlet and ASE show high recoveries, they also lead to larger standard deviations and are not suitable for determination of thermolabile pesticides.
In addition to recovery by different extraction methods, the agreement of values obtained by ECD and MS was evaluated. For ECD, standard deviations are generally higher but overall the values obtained by MS and ECD are comparable.
Trans-Zearalenone(ZEN)is a non-steroidal estrogenic mycotoxin which frequently contaminates cereal grains worldwide. Ingestion of food and feed containing ZEN causes numerous mycotoxicoses in animals and possibly humans with pronounced estrogenic effects. Due to the trans-configurated double bond, ZEN isomerizes to the cis-configuration upon the influence of light. This work investigates the instrumental analysis, metabolism and toxicity of ZEN and cis- ZEN. The first part focused on the determination of ZEN in edible oils. Due to a maximum level of 400 µg/kg ZEN in the European Union (EU), reliable analytical methods are needed. A comprehensive method comparison proved dynamic covalent hydrazine chemistry (DCHC) to be the most suitable approach. Thus, an automated solid phase extraction (SPE) coupled online to high performance liquid chromatography (HPLC) was developed with the novelty of a covalent SPE step comprising the DCHC principle. The automated online system allows an accurate, selective and reliable quantification of ZEN in edible oils in compliance with EU performance criteria while significantly reducing workload and thereby personnel costs. In contrast to ZEN, reference standards and analytical methods are missing for cis- ZEN which causes a lack of data on the occurrence, fate and risks of cis-ZEN. Therefore, a native and an isotopically labeled cis-ZEN standard were synthesized and implemented in an existing stable isotope dilution analysis HPLC tandem mass spectrometry (HPLC-MS/MS) method. Using this method, a large extent of cis-ZEN formation was observed for ZEN contaminated maize germ oils when exposed to daylight which confirms that cis-ZEN can be a relevant food contaminant and should be considered in the analysis of food and feed. Furthermore, this work investigated the in vitro phase I metabolism of ZEN and cis-ZEN in rat and human liver microsomes by using HPLC-MS and -MS/MS analyses. The metabolic pathways of cis-ZEN were found to be essentially similar to ZEN including reduction and oxidation reactions generating α- and β-cis- zearalenol as well as 13- and 15-OH-cis-ZEN. A previously unidentified oxidative metabolic pathway for both isomers of ZEN results in the formation of cis-ZEN-11,12-oxide and ZEN-11,12-oxide in human liver microsomes. The estrogenicity of cis-ZEN and its reductive metabolites was assessed using the E-Screen assay. cis-ZEN proved to be slightly more estrogenic than ZEN. Biotransformation of cis-ZEN to β-cis-ZEL corresponds to a detoxification, whereas metabolism to α-cis-ZEL resembles a metabolic activation as its estrogenicity considerably exceeds that of cis-ZEN. The catecholic metabolites can be expected to show a decreased estrogenicity as demonstrated for 15-OH- ZEN. Independent of the estrogenic effects, the catecholic and epoxidic metabolites identified in this work can be expected to act genotoxic and carcinogenic. The epoxides in particular could fundamentally change the widely accepted view of ZEN causing adverse effects exclusively through endocrine disrupting actions.
While organic pollutants have been extensively studied in urban and agricultural areas, data from forested regions are relatively rare.
Within the framework of a forest soil inventory in Germany polycyclic aromatic hydrocarbons (PAH), polychlorinated biphenyls (PCB) and organochlorine pesticides (OCP) were analyzed in 1500 soil samples taken from 3 soil horizons at 447 sites. An analytical protocol was developed and validated for the simultaneous extraction and measurement of all analytes in humic rich soil at trace level range. It included pressurized liquid extraction (PLE) and a two step cleanup procedure consisting of gel permeation chromatography (GPC) followed by solid phase extraction (SPE). Quantification was carried out using gas chromatography combined with mass spectrometry (GC-MS). In line with the monitoring campaign a comprehensive quality assurance concept was implemented to guarantee constant analytical measurement conditions for two years. The protocol is applicable for field laboratories with a high level of automation and for analyzing high sample amounts.
At most sampling sites pollutant concentrations gradually decrease with depth. Only few spots showed highest concentrations in the upper mineral soil. In the O-horizon the sum concentrations of 16 PAHs range from 104 to 14.000 µg/kg, of 6 PCBs from <1 to 106 µg/kg and the sum of DDT and metabolites range from <1 to ca. 4.000 µg/kg.
The observed concentration of PAH, PCB and OCP reflect an ubiquitous background contamination at most sites. Relatively high concentrations of PAH and PCB are related to industrial areas and peak concentrations only of PAH are associated with open cast mining of brown coal areas. The spatial distribution of the contents of DDT and metabolites in German forest soils can clearly be attributed to historic application. A contribution of ubiquitous background pollution derived from long-range atmospheric transport is likely for some compounds in the studied area, e.g., DDT in the western part of Germany. The results generally reflect the distribution of POPs in densely populated and industrialized countries located in temperate regions].
The determination of organochlorine pesticides (OCP) and polychlorinated biphenyls is challenging because they are strongly physical and chemical bound to soil organic matter. There are various valid standards for the determination of these analytes in soil that differ especially in the extraction method and extraction time. In order to make profound assessments concerning the extraction efficiency seven extraction procedures namely pressurized liquid extraction (PLE), soxhlet and shaking with different extraction times 0.5 h, 1 h, 3 h, 6 h and 16 h were compared. For the study four soils of different total organic carbon (TOC) content were generated from uncontaminated reference soil and compost which were spiked to contaminant levels representative of the Bundes-Bodenschutz-Verordnung. Clean-up and measurement conditions using gas chromatography with mass spectrometry (GC-MS) were kept constant. Statistically analysis of the experimental values showed that recovery rates were not dependent on matrix TOC content. In consideration of measurement uncertainties of 10% - 20% for the determination of OCP and PCB in soil no differences in the efficiency between the tested extraction procedures could be observed.
The aim of this work is to develop measurement procedures to provide full traceability and robust uncertainty statements for dissolved elemental mercury (Hg(0)), inorganic mercury (Hg(II)) and methyl-mercury (MeHg) in freshwater and sea water, and inorganic mercury (Hg(II)) and methyl-mercury (MeHg) in biota. Accurate measurements with low uncertainties e.g. <50 % at the EQS level are required to assess concentration levels and trends in these important environmental reservoirs for mercury.
In particular this work aims to:
Develop and validate primary measurement procedures for Hg speciation (Hg(II), MeHg and Hg(0)) in fresh and sea waters at sub-ng levels in the presence of suspended particular matter and/or colloidal fractions,
Apply the developed IDMS procedures in water in order to evaluate possible artefacts associated with sample collection and preparation, preserving speciation and/or correcting for possible post-sampling interconversions,
Develop and validate primary measurement procedures for the accurate quantification of Hg(II) and MeHg in biota, using samples from the Federal Environmental Specimen Bank,
Apply the developed IDMS procedures in biota in order to evaluate the effect of different treatments for fish sample preparation.
Sulfamethoxazol (SMX),a sulfonamide, is a widely used bacteriostatic antibiotic and therefore a promising marker for the entry of anthropogenic Pollution in the environment. SMX is frequently found in wastewater and surface water. This study presents the production of high affinity and selective polyclonal antibodies for SMX and the development and Evaluation of a direct competitive enzyme-linked immunosorbent assay(ELISA)for the quantification of SMX in environmental watersamples. The crystal structures of the cross-reacting compounds sulfamethizole, N4-acetyl-SMX andsuccinimidyl-SMX were determined by x-ray diffraction aiming to explain their high cross-reactivity. These crystal structures are described for the first time. The quantification range of the ELISA is 0.82–63 µg/L. To verify our results, the SMX concentration in 20 environmental samples, including wastewater and surfacewater, was determined by ELISA and tandem mass spectrometry(MS/MS).A good Agreement of the measured SMX concentrations was found with average recoveries of 97–113%for the results of ELISA compared to LC-MS/MS.
The polycyclic musks 6-Acetyl-1,1,2,4,4,7-hexamethyltetraline (AHTN) and 1,3,4,6,7,8-Hexahydro-4,6,6,7,8,8-hexamethylcyclopenta-ɣ-2-benzopyran (HHCB) are widely used fragrances in cosmetics, products of daily care and cleaning products for households and industry. They are in the focus of interest due to their estrogenic potential. Both compounds can be found at low μg/L concentrations in surface water where they are introduced mainly by sewage treatment plants supplied by municipal wastewater. In order to disinfect wastewater in sewage treatment plants or water in swimming pools, chlorine can be added to water as gas or hypochlorite solution. This disinfectant has a high oxidation potential resulting in the formation of disinfection by-products, the estrogenicity and toxicity of which might differ from their precursor. In this work the behaviour of AHTN and HHCB upon treatment with sodium hypochlorite solution was investigated and chlorinated possible disinfection by-products could be identified as well as a carboxylic acids and lactones derived from AHTN and HHCB. The identified species were characterized by GC-MS, LC-MSn and 1H-NMR. The elucidated structures of potential disinfection by-products are verified by X-ray crystallography. Finally the estrogenicity between the parent analytes and generated transformation products were compared. For that task, MCF-7 assay has been chosen for being most relevant to humans. Moreover metabolism studies were carried out using AHTN, HHCB and their disinfection by-products.
Fluorophore labeled proteins and antibodies, referred to also as targeted optical probes, present a promising strategy for a variety of applications from fundamental cell-based biological studies to in vivo diagnostics and image guided surgeries in humans. In this respect, design strategies for the preparation of such conjugates from different dyes including analyte-responsive fluorophores are presented as well as their analytical and spectroscopic characterization employing Absorption spectroscopy and steady state and time-resolved fluorometry. Special emphasis is dedicated to the influence of dye hydrophilicity and labeling density on the optical properties and binding behavior of these dye-bioconjugates including their performance in in vitro and in vivo bioimaging studies.
Several environmental stressors have been identified as key and/or emerging drivers of habitat change that could significantly influence marine near-shore ecosystems. These include increasing discharges of pharmaceutical contaminants into the aquatic coastal systems. Pharmaceutical drugs are often detected in aquatic environments but still information on their toxicity impacts on inhabiting species is scarce, especially when acting in combination. Furthermore, almost no information is available on the impacts of pharmaceuticals in polychaetes, often the most abundant taxon in benthic communities and commonly used as indicator species of environmental conditions. Therefore, the present study aimed to evaluate the biochemical alterations induced in the polychaete Hediste diversicolor, from a low contaminated area at the Ria de Aveiro lagoon (Portugal), by the antiepileptic drug carbamazepine (0.0 - control, 0.3, 3.0, 6.0 and 9.0 μg/L) and the stimulant caffeine (0.0 - control, 0.5, 3.0, and 18.0 μg/L), acting alone and in combination (0.3 CBZ + 0.5 CAF and 6.0 CBZ + 3.0 CAF). Glutathione Stransferases (GSTs), superoxide dismutase (SOD) and catalase (CAT) activities was determined in Hediste diversicolor from each condition. Lipid peroxidation (LPO), glutathione reduced and oxidized (GSH and GSSG), glycogen and electron transport system (ETS) were also measured. The results obtained clearly revealed that both drugs induced oxidative stress in H. diversicolor, shown by the increase on LPO levels and decrease on total glutathione and GSH/GSSG ratio with the increase of exposure concentrations. Furthermore, the present findings demonstrated that polychaetes biotransformation capacity as well as antioxidant defense mechanisms were not sufficiently efficient to fight against the excess of reactive oxygen species (ROS) leading to LPO when organisms were exposed to both drugs. Our results also demonstrated that polychaetes tended to decrease the activity of ETSwhen exposed to drugs, avoiding energy expenditurewhich may prevent them fromgreater damages. The present study further revealed that the impacts induced by the combination of both drugswere similar to those obtained at the highest drugs concentrations acting alone.
Caffeine is known to be one of the most consumed psychoactive drugs. For this reason, caffeine is continuously released into the environment with potential impacts on inhabiting organisms. The current study evaluated the biochemical alterations induced in the clam species Ruditapes philippinarum after exposure for 28 days to caffeine (0.5, 3.0 and 18.0 mg/L). The results obtained showed that, with the increasing caffeine concentrations, an increase in clams defense mechanisms (such as antioxidant and biotransformation enzymes activity) was induced which was accompanied by an increase in protein content. Nevertheless, although an increase on defense mechanisms was observed, clams were not able to prevent cells from lipid peroxidation that increased with the increase of caffeine concentration. Furthermore, with the increase of exposure concentrations, clams increased their metabolic activity (measured by electron transport activity), reducing their energy reserves (glycogen content), to fight against oxidative stress. Overall, the present study demonstrated that caffeine may impact bivalves, even at environmentally relevant concentrations, inducing oxidative stress in organisms. The present study is an important contribution to address knowledge gaps regarding the impacts of long-term exposures to pharmaceuticals since most of the studies assessed the effects after acute exposures, most of them up to 96 h.
In the last decade studies have reported the presence of several pharmaceutical drugs in aquatic environments worldwide and an increasing effort has been done to understand the impacts induced on wildlife. Among the most abundant drugs in the environment is caffeine, which has been reported as an effective chemical anthropogenic marker. However, as for the majority of pharmaceuticals, scarce information is available on the adverse effects of caffeine on marine benthic organisms, namely polychaetes which are the most abundant group of organisms in several aquatic ecossystems. Thus, the present study aimed to evaluate the biochemical alterations induced by environmentally relevant concentrations of caffeine on the polychaete species Diopatra neapolitana and Arenicola marina. The results obtained demonstrated that after 28 days exposure oxidative stress was induced in both species, especially noticed in A. marina, resulting from the incapacity of antioxidant and biotransformation enzymes to prevent cells from lipid peroxidation. The present study further revealed that D. neapolitana used glycogen and proteins as energy to develop defense mechanisms while in A. marina these reserves were maintained independently on the exposure concentration, reinforcing the low capacity of this species to fight against oxidative stress.
Ocean acidification and increasing discharges of pharmaceutical contaminants into aquatic systems are among key and/or emerging drivers of environmental change affecting marine ecosystems. A growing body of evidence demonstrates that ocean acidification can have direct and indirect impacts on marine organisms although combined effects with other stressors, namely with pharmaceuticals, have received very little attention to date. The present study aimed to evaluate the impacts of the pharmaceutical drug Carbamazepine and pH 7.1, acting alone and in combination, on the clamScrobicularia plana. For this, a long-termexposure (28 days)was conducted and a set of oxidative stress markers was investigated. The results obtained showed that S. plana was able to develop mechanisms to prevent oxidative damage when under low pH for a long period, presenting higher survival when exposed to this stressor compared to CBZ or the combination of CBZ with pH 7.1. Furthermore, the toxicity of CBZ on S. plana was synergistically increased under ocean acidification conditions (CBZ + pH 7.1): specimens survival was reduced and oxidative stress was enhanced when compared to single exposures. These findings add to the growing body of evidence that ocean acidification will act to increase the toxicity of CBZ to marine organisms, which has clear implications for coastal benthic ecosystems suffering chronic pollution from pharmaceutical drugs.
Photodegradation of organic pollutants in water by immobilized porphyrins and phthalocyanines
(2016)
New methods for water treatment are required as a result from an increasing awareness in the reduction of the pollution impact in the environment. In the perspective of the photo-oxidation of organic pollutants present in water, the principal incentive for the preparation of heterogeneous photocatalysts is their easy recovery from the reaction mixture, which allows their reuse in successive runs, minimizing the loss of their original photocatalytic properties. Different types of supports can be used in the immobilization of photoactive species, such as porphyrins (Pors) and phthalocyanines (Pcs). This mini-review will consider the different methodologies for the immobilization of Pors and Pcs and their photocatalytic performance in the photodegradation of organic pollutants in water, addressing also their recycling ability in successive water treatments.
The toxicity induced in non-target organisms by pharmaceutical drugs has been the focus of several studies. In the aquatic environment, most of the studies have been devoted to fish and bivalves, while little is known on the impacts induced in polychaetes. The present study evaluated the impacts of carbamazepine and caffeine on the regenerative capacity of Diopatra neapolitana, a polychaete species with high ecological and economic relevance. Under laboratory controlled conditions polychaetes were exposed, during 28 days, to carbamazepine (Ctl-0.0; 0.3; 3.0; 6.0; 9.0 mg/L) and caffeine (Ctl-0.0; 0.5; 3.0; 18.0 mg/L). During the experiment, at days 11, 18, 25, 32, 39 and 46 after amputation, for each specimen, the percentage of the body width regenerated was determined and the number of new segments was counted. The regenerative capacity was assessed considering the number of days needed to achieve full regeneration and the total number of new segments. The obtained results revealed that with the increase of drugs concentrations organisms regenerated less new segments and took longer to completely regenerate.
An Annex XV restriction dossier for cadmium in artists’ paints was submitted by an EU member state to the European Chemicals Agency ECHA. By cleaning used brushes under the tap cadmium can enter the food chain via waste water treatment and subsequent agricultural application of the sewage sludge. It was estimated that 110 kg Cd per year is spread on agricultural land via this exposure route. Other sources of Cd amount to almost 120 tons per year.
The mobility of Cd from pigments was studied in a field-like scenario by leaching experiments using soil samples amended with sewage sludge and spiked with Cd pigments in percolation columns. The redox conditions were confirmed to be the decisive factor for the release of Cd. The release of Cd from artists’ paints was in most cases 1% or lower in the experiments performed.
Cd from artists’ paints in sewage sludge applied in agriculture will be mobilized for intake by crops or to leaching either by oxidation of sulfide to sulfate or by dissolution at lower pH values. The quantity of Cd from artists’ paints calculated in the restriction dossier is however negligible compared to other sources of Cd to agricultural soil. Therefore ECHA did not consider the proposed restriction to be the most appropriate EU wide measure to address the negligible level of risk. In the presentation sources of Cd on agricultural land will be discussed together with possible policy instruments to mitigate such emissions.
Large animal models are highly recommended for meaningful preclinical studies, including the optimization of cement augmentation for vertebral body defects by vertebroplasty/kyphoplasty.
The aim of this study was to perform a systematic characterization of a strictly minimally invasive in vivo large animal model for lumbar ventrolateral vertebroplasty.
This is a prospective experimental animal study.
Lumbar defects (diameter 5 mm; depth approximately 14 mm) were created by a ventrolateral percutaneous approach in aged, osteopenic, female sheep (40 Merino sheep; 6–9 years; 68–110 kg). L1 remained untouched, L2 was left with an empty defect, and L3 carried a defect injected with a brushite-forming calcium phosphate cement (CPC). Trauma/functional impairment, surgical techniques (including drill sleeve and working canula with stop), reproducibility, bone defects, cement filling, and functional cement augmentation were documented by intraoperative incision-to-suture time and X-ray, postoperative trauma/impairment scores, and ex vivo osteodensitometry, microcomputed tomography (CT), histology, static/fluorescence histomorphometry, and biomechanical testing.
Minimally invasive vertebroplasty resulted in short operation times (28±2 minutes; mean±standard error of the mean) and X-ray exposure (1.59±0.12 minutes), very limited local trauma (score 0.00±0.00 at 24 hours), short postoperative recovery (2.95±0.29 hours), and rapid decrease of the postoperative impairment score to 0 (3.28±0.36 hours). Reproducible defect creation and cement filling were documented by intraoperative X-ray and ex vivo conventional/micro-CT. Vertebral cement augmentation and osteoconductivity of the CPC was verified by osteodensitometry (CPC>control), micro-CT (CPC>control and empty defect), histology/static histomorphometry (CPC>control and empty defect), fluorescence histomorphometry (CPC>control; all p<.05 for 3 and 9 months), and compressive strength measurements (CPC numerically higher than control; 102% for 3 months and 110% for 9 months).
This first-time systematic clinical assessment of a minimally invasive, ventrolateral, lumbar vertebroplasty model in aged, osteopenic sheep resulted in short operation times, rapid postoperative recovery, and high experimental reproducibility. This model represents an optimal basis for standardized evaluation of future studies on vertebral augmentation with resorbable and osteoconductive CPC.
It is thought that the primary function of soldier castes in insect societies is to protect colonies against macroscale predators and parasites. By comparison, little attention has been paid to the role of soldiers against pathogenic microorganisms. This is a significant gap in knowledge because in non-hymenopteran societies, such as the eusocial termites, the first-evolved, and arguably most important sterile caste, is the soldier.
Here, we investigate whether soldiers of the wood-eating termite, Mastotermes darwiniensis, play a significant role in social immunity. Firstly, we show that uninfected soldiers improve the survival of worker nestmates exposed to a lethal dose of a fungal pathogen, despite the fact that soldiers (unlike workers) cannot allogroom. Hazard ratios of pathogen-exposed workers decreased from 6.616 in infected groups to 4.376 and 2.786 in infected groups containing uninfected workers and soldiers, respectively. To explore the basis of soldier protection, we applied soldier-derived oral secretions to infected worker cuticle, resulting in significantly improved worker survival. Soldiers that contacted infected workers were also more active and produced secretions with significantly greater mass variance than soldiers exposed to uninfected workers.
Significantly, we show that secretions produced by soldiers inhibit the growth of a broad range of microorganisms, including gram-negative and gram-positive bacteria, in addition to fungi. Our findings demonstrate that soldiers can contribute powerful antimicrobial factors to external immune defenses, effectively complementing behaviour-based protective activities provided by workers.
In addition to individual immunity, social insects have evolved collective defence strategies against pathogens, which consist of behavioural, physiological and organisational adaptations. The importance of social immunity in preventing disease transmission in insect societies is well known. However, we still lack a clear understanding of how collective defence mechanisms are regulated and how they interact with the other regulatory systems of the organism such us the individual immune system. Here we assess how individual immune factors shape the collective immune response to pathogens by inhibiting a key Termite immune effector: gram-negative binding protein (tGNBP-2). tGNBP-2 triggers downstream innate immunity, in addition to cleaving fungal and bacterial pathogens directly in the external environment. We established mini-colonies of Reticulitermes flavipes inside planar arenas where we were able to observe and record hygienic behaviours (i.e. allogrooming and cannibalism) in near-natural nest conditions. Colonies were either treated with the tGNBP-2 inhibitor or a control solution, after which 5 focal workers were introduced into each colony. Focal workers were pre-exposed to either: 1) the fungal pathogen Metharizium anisopliae 2) the t-GNBP-2 inhibitor + M. anisopliae 3) a control solution. Colonies were recorded and the hygienic behaviours performed by nestmates towards focal termites were analysed. We examined the impact of tGNBP-2 inhibition on individual innate immune regulation, in addition to exploring the wider impact of immune suppression on collective social behavioural responses.
CO2 streams captured from power stations or industrial plants may contain impurities that impact the consecutive steps of the CO2 capture and storage (CCS) chain. As the basis for an optimization of CO2 purity over the whole CCS chain, impacts of different impurities were investigated at key steps including studies on (i) corrosion of metallic materials in CO2 streams and brine, (ii) fluid and interfacial properties as a function of pressure, temperature and CO2 stream composition and their implications for CO2 transport, injection and geological storage, (iii) costs of different pipeline design options, (iv) geochemical alterations at typical reservoir conditions and their implications for geomechanical rock properties. Major findings are synthesized for two exemplary single source-single sink CCS chain scenarios involving CO2 stream compositions typical for pre-combustion capture and oxyfuel combustion. Recommendations for material selection for compression, transport and injection were derived for various CO2 stream compositions. To reliably control corrosion, a limitation of water contents to 50 ppmv is recommended for pipeline transportation of all CO2 streams. At geological storage conditions, the presence of either O2, NOx or SO2 only weakly affected fluid-mineral/rock interactions that still impacted geomechanical rock properties.
Due to the increasing presence of microplastic particles (MP) in the Environment and the unknown risks arising from them, there is an urgent need for analytical methods that allow for an efficient identification and quantification of microplastics (MP), i.e. particles < 5 mm, in environmental samples. So far, mostly timeconsuming (micro) infrared or micro-Raman spectroscopic methods are applied. Here an faster alternative Approach is presented based on a Raman processspectrometer with fiber-optical probes in combination with multivariate data analysis.
Phosphorus (P) is an essential element for all forms of life and is applied as fertilizer in agriculture. The P availability for plants may be highly dependent on the chemical state of P in fertilizers and soils; however, the nature of this dependence remains obscure due to the limitations of generally applied wet chemical and instrumental analytical approaches. This paper focuses on recently developed infrared, Raman, ultraviolet and X-ray microspectroscopic techniques for the characterization of P in soil. Microspectroscopic techniques have the advantage that discrete P phases can be distinguished and characterized even if their mass fractions are very low. However, only small volumes of soil can be analyzed by microspectroscopic methods hence a combination of macro- and microspectroscopic techniques is a promising concept.
A 400 m² soil test field with gas injection system was built up, which enables an experimental validation of linear gas sensors for specific applications and gases in an application-relevant scale. Several injection and soil watering experiments with carbon dioxide (CO2) at different days with varying boundary conditions were performed indicating the potential of the method for, e.g., rapid leakage detection with respect to Carbon Capture and Storage (CCS) issues.
This paper is a case study of complete substitution of sodium-metasilicate in zeolite Na-A synthesis by an electrostatic filterash (FA) arising in high amounts during silane waste incineration process. The silicate abundant FA is a suitable material for reinsertion in zeolite chemistry. This is shown in the presented investigation by the development of a model reaction process at low temperatures (50–60 °C) and short times (1.5–4 h). The experiments were performed under addition of NaAlO2 and variation of the alkalinity and the reaction period. Characterization and fine tuning of the process was mainly done by studying the kinetics of FA digestion and zeolite crystallization by XRD and chemical analyses (ICP-OES) of solutions and solids. It could be shown that pure FA was mostly dissolved (98%) in 8 m NaOH already after 1 h. Addition of NaAlO2 and crystallization for further 60 min under optimized conditions at 50 °C yielded to a suitable product. It consists of zeolite Na-A (92% by mass) beside some hydrosodalite (8% by mass). According to this low temperature short time process this study is a contribution for the development of energy efficient recycling solutions.

There is significant contamination of our water resources and an accelerating accumulation of hazardous species in the aquatic biosphere. Whether for instance mercury as a contaminant constantly released from a multitude of industrial processes or chemical warfare agents (CWAs) such as Tabun (GA), Sarin (GB) or Soman (GD) as highly toxic remnants of World War II, released from dumped containers upon their progressive corrosion, are concerned such hazards can cause serious problems to human health even at low concentration and monitoring them becomes increasingly essential. Conclusively, it is necessary to develop sensors for such hazards that combine high sensitivity, good selectivity, fast responses and can dispense with complex instrumentation.
Here, we present SBA-15-type mesoporous silica materials containing boron–dipyrromethene (BODIPY) probes in their pore systems that allow for the sensitive and selective detection of the two target analytes CWAs and Hg2+ in natural waters via a change in the optical properties. In case of Hg2+, the BODIPY dye is directly adsorbed on the (inner and outer) surface of the highly porous material and coordinates the metal ion, leading to an enhancement of fluorescence with a limit of detection (LOD) of 15 ppt. Using the same silica material but incorporating a covalently attached BODIPY dye, the CWAs GA, GD and GB can be detected via fluorescence quenching induced by an intraannular cyclization. The obtained LOD in natural waters is in the pM range and hence below the Maximum Permissible Concentrations for CWAs in drinking water. Aspects of system design as well as application in water and food analysis (fish extracts) will be presented.

Concrete is one of the most widely used construction materials and, accordingly, the concrete industry is an important stakeholder in the field of sustainable construction. Therefore various approaches have been implemented to increase the sustainability of concrete. Besides reducing CO2-emissions during cement production, increasing the energy efficiency of buildings and extending their life span, the end-of-life performance of concrete is also an essential aspect of sustainability. Reusing concrete as a secondary building material meets the requirements of sustainability in several ways: the extended time availability of primary raw materials and, thereby, the protection of natural resources as well as conserving landfill site. Furthermore, the production of recycled concrete aggregates (RCA) is a good example for closed-loop recycling.
However, regarding the use of RCA as a substitute for natural aggregates in concrete, attention must be paid to all issues of sustainability: this means that environmental, economic and social aspects have to be considered. Since RCA generally have inferior building material properties, such as higher porosity and lower density, the implementation of closed-loop recycling of concrete only makes sense if the technical quality assurance is secured. This paper focusses on the implementation of techniques for the deconstruction/demolition of buildings and the subsequent treatment of concrete rubble in order to improve the building material properties of RCA with regard to the requirements of sustainability and also technical rules and standards.

2,4-Dichlorophenoxyacetic acid (2,4-D) is one important and well-known herbicide that is widely used in agriculture because of its advantages to regulate plant growth. However, the use of large quantities of the treated plants as animal feed leads to residues in meat, milk and eggs. Furthermore, the herbicide can drain away and contaminate ground and drinking water. The ingestion of 2,4-D-contaminated food and water causes damage to the inner organs of humans and animals, e.g., the kidneys and the liver.
Analytical assays based on molecularly imprinted polymers (MIPs) have emerged as a valuable tool in the field of environmental analysis due to the low production costs, stability, format adaptability and the possibility to imprint and thus their ability to recognize a wide variety of target analytes.5 With regard to optical sensing technologies, however, MIPs have only been used in considerably few applications, especially in fluorescence sensors. This limitation is basically due to the fact that the incorporation of a fluorescently responding moiety into a polymer matrix is challenging. One way to overcome this limitation is to use tailor-made fluorescent indicator monomers for direct transfer of the binding event into an optical signal and coat the MIP via reversible addition-fragmentation chain transfer (RAFT) polymerization as a thin layer onto the surface of silica nanoparticles as primary sensing element.
Here, we present the response behavior of the fluorescent MIP sensor particles in terms of sensitivity for 2,4-D detection (the so-called imprinting factor), discrimination ability against structurally similar compounds and performance in a phase-transfer assay (PTA) on chip, i.e., the implementation of the assay into a microfluidic chip environment, offering a novel simple and rapid way for the detection of herbicides.
The pollution of the environment with microplastics MP (particles < 5mm) is a problem of increasing concern.1 A consistent methodology that facilitates detection of MP is still missing. Here we present the data recorded by applying different MP identification methods to a soil fortified with known amounts of MP.
