Functionalisation of UCNPs with analyte-responsive dyes as resonance energy transfer (RET)acceptors, which is in the focus of our work, will yield chemoresponsive nanoprobes with unique features: they can be excited in the NIR range (which minimizes the photo-damage of biological samples, as well as the autofluoresence background) and one of their several sharp UC emission bands can be used as donor for RET sensing applications in combination with analyte-sensitive dyes which serve as indicators, whereas the other UC band(s) can act as internal reference. The first step upon preparation of such nanosensors is the functionalisation of the UCNPs with different surface chemistries for indicator conjugation. Desirable are systems with short distance between the UCNP(donor) and the indicator (acceptor) as a prerequisite for efficient RET. Additionally, dyes with a large spectral overlap between their absorption and one of the UC emission bands are required. Several classes of pH-responsive fluorescent dyes like xanthenes are eligible. Here, we present first studies to optimize the surface functionalization to achieve high RET efficiencies, which would improve the performance of such nanosensors for ratiometric fluorescence measurements of pH in biological matrices.
