To increase the competitiveness of jacket substructures compared to monopiles a changeover from an individual towards a serial jacket production based on automated manufactured tubular joints combined with standardized pipes has to be achieved. Therefore, this paper addresses fatigue tests of automatically welded tubular X-joints focusing on the location of the technical fatigue crack. The detected location of the technical crack is compared to numerical investigations predicting the most fatigue prone notch considering the structural stress approach as well as the notch stress approach. Besides, the welding process of the automated manufactured tubular X-joints is presented.
Binder remnants in historical mortars represent a record of the connection between the raw materials that enter the kiln, the process parameters, and the end product of the calcination. Raman microspectroscopy combines high structural sensitivity with micrometre to sub-micrometre spatial resolution and compatibility with conventional thin-sectional samples in an almost unique fashion, making it an interesting complementary extension of the existing methodological arsenal for mortar analysis. Raman spectra are vibrational fingerprints of crystalline and amorphous compounds, and contain marker bands that are specific for minerals and their polymorphic forms. Relative intensities of bands that are related to the same crystalline species change according to crystal orientations, and band shifts can be caused by the incorporation of foreign ions into crystal lattices, as well as stoichiometric changes within solid solution series. Finally, variations in crystallinity affect band widths. These effects are demonstrated based on the analysis of three historical mortar samples: micrometric distribution maps of phases and polymorphs, crystal orientations, and compositional variations of solid solution series of unreacted clinker grains in the Portland cement mortars of two 19th century castings, and the crystallinities of thermal anhydrite clusters in a high-fired medieval gypsum mortar as a measure for the applied burning temperature were successfully acquired.

Most knowledge about the structure, function, and evolution of early compound eyes is based on investigations in trilobites. However, these studies dealt mainly with the cuticular lenses and little was known about internal anatomy. Only recently some data on crystalline cones and retinula cells were reported for a Cambrian trilobite species. Here, we describe internal eye structures of two other trilobite genera. The Ordovician Asaphus sp. reveals preserved crystalline cones situated underneath the cuticular lenses. The same is true for the Devonian species Archegonus (Waribole) warsteinensis, which in addition shows the fine structure of the rhabdom in the retinula cells. These results suggest that an apposition eye with a crystalline cone is ancestral for Trilobita. The overall similarity of trilobite eyes to those of myriapods, crustaceans, and hexapods corroborates views of a phylogenetic position of trilobites in the stem lineage of Mandibulata.
In this paper we report on the characterization by X-ray computed tomography of calcium phosphate (CaP) and polycaprolactone (PCL) coatings on Ti-6Al-4V alloy scaffolds used as a material for medical implants. The cylindrical scaffold has greater porosity of the inner part than the external part, thus, mimicking trabecular and cortical bone, respectively. The prismatic scaffolds have uniform porosity. Surface of the scaffolds was modified with calcium phosphate (CaP) and polycaprolactone (PCL) by dip-coating to improve biocompatibility and mechanical properties. Computed tomography performed with X-ray and synchrotron radiation revealed the defects of structure and morphology of CaP and PCL coatings showing small platelet-like and spider-web-like structures, respectively.
The primary screening of hybridoma cells is a time-critical and laborious step during the development of monoclonal antibodies. Often critical errors occur in this phase, which supports the notion that the generation of monoclonal antibodies with hybridoma technology is difficult to control and hence a risky venture. We think that it is crucial to improve the screening process to eliminate most of the immanent deficits of the conventional approach. With this new microarray-based procedure, several advances could be achieved: Selectivity for excellent binders, high throughput, reproducible signals, avoidance of misleading avidity (multivalency) effects, and simultaneous performance of competition experiments. The latter can directly be used to select clones of desired cross-reactivity properties. In this paper, a model system with two excellent clones against carbamazepine, two weak clones and blank supernatant has been designed to examine the effectiveness of the new system. The excellent clones could be detected largely independent of the IgG concentration, which is unknown during the clone screening since the determination and subsequent adjustment of the antibody concentration is not possible in most cases. Furthermore, in this approach, the enrichment, isolation, and purification of IgG for characterization is not necessary. Raw cell culture supernatant can be used directly, even when fetal calf serum (FCS) or other complex media had been used. In addition, an improved method for the oriented antibody-immobilization on epoxy-silanized slides is presented. Based on the results of this model system, we conclude that this approach should be preferable to most other protocols leading to many of false positives, causing expensive and lengthy confirmation steps to weed out the poor clones.
In this paper, the impact problem and the subsequent wave Propagation are considered. For the contact discretization an intermediate non-uniform rational B-spline (NURBS) layer is added between the contacting finite element bodies, which allows a smooth contact formulation and efficient element-based integration.
The impact event is ill-posed and requires a regularization to avoid propagating stress oscillations. A nonlinear mesh-dependent penalty regularization is used, where the stiffness of the penalty regularization increases upon mesh refinement. Explicit time integration methods are well suited for wave propagation problems, but are efficient only for diagonal mass matrices. Using a spectral element discretization in combination with a NURBS contact layer the bulk part of the mass matrix is diagonal.
In den letzten zwei Jahren wurden 11 neue Standards/Standardrevisionen zur RT veröffentlicht (ohne Strahlenschutz) und 5 werden dafür gestrichen. Es wird über die neuen Anforderungen ausgewählter Normen bzw. Normrevisionen informiert und insbesondere auch über die veränderten Parameter, die bei Prüfpraxis und bei der Klassifizierung zu berücksichtigen sind. Das wichtigste Projekt ist die Revision der DIN EN ISO 17636-1, -2, RT von Schweißverbindungen, in ISO TC 44 SC 5 WG 1. Die Standards zur Schweißnahtprüfung in der Kerntechnik, DIN 25435, wurden ins Englische übertragen und bei ISO TC 85 SC 6 bearbeitet. Hauptaktivität bei ASTM ist gegenwärtig die Überarbeitung der CT-Standards und die Berücksichtigung der Anforderungen an das dimensionelle Messen. Der Guide ASTM E 1441 zur Bestimmung von MTF, Kontrast-Detail-Funktion (CDF) und Kontrast-Diskrimination-Diagramm (CDD) wird revidiert. E 2445 zur CR Long Term Stability wird ebenfalls revidiert. Die Revision soll auch in die ISO 16371-1 eingehen. Dazu ist ein Round Robin-Test geplant. Auch die Standards zur Durchstrahlungsprüfung auf Korrosion und Ablagerungen in Rohren mit Röntgen- und Gammastrahlen (EN 16407-1, -2, 2014) wurden revidiert. Diese Revision wurde als DIN EN ISO 20769-1, -2 2018 veröffentlicht und EN 16407 wird zurückgezogen. Die Revision der Standards EN 12543 und EN 12679 zur Messung der Brennfleckgröße und der Strahlergröße ist mit Verzögerung in Überarbeitung, um die digitalen Detektoren und Messmöglichkeiten mit Bildverarbeitungsprogrammen zu berücksichtigen. Ein Entwurf zur Messung der Brennfleckgröße von nano-Fokusröhren wird derzeit vom Europäischen Metrologie-Programm EMPIR gefördert. Zur radiographischen Gussteilprüfung wurde der Standard EN 12681 revidiert und 2018 veröffentlicht. Er wurde um Zulässigkeitsgrenzen erweitert und die digitale Radiographie wurde in Teil 2 berücksichtigt.
While the absorption of X-rays and gamma radiation is determined by the Z-number of the elements a specimen is composed of, it is the hydrogen making an effective contrast with neutron imaging. As a consequence, interrogating with neutrons presents a suitable tool to study the distribution of organic consolidants in materials such as wood as encountered in impregnated wooden artworks.
Four different examples of objects are presented here to demonstrate the potential of neutron CT: 1) small wooden pieces of ship wrecks (< 2 cm thickness) interrogated with cold neutrons (0.5 meV at the ANTARES facility of the FRM II in Garching) to demonstrate the potential and the limitation of using low energy neutrons, 2) a wooden statue soaked with carbolineum (fission neutrons 1.8 MeV at the NECTAR facility of the FRM II), 3) a smaller wooden figure of a skull heavily soaked with carbolineum so it was too tight for the fission neutrons used before with accelerator neutrons (broad range about 5.5 MeV at the PTB in Braunschweig) and 4) pieces of charred wood to study the impregnation with a consolidant (NECTAR, FRM II). With the exception of the last example, all results have been combined with X-ray tomography (BAM 8.3 in Berlin). In the case of the charred wood specimens (example 4) the density histograms of the neutron tomography results were compared with those obtained from untreated references. The observed gain in specific density of the soaked specimens corresponded with an increase of specific weight. All results obtained so far showed distinct distribution patterns attributable to structural peculiarities or organic consolidants providing valuable support for subsequent restoration works.
The hydration process of Portland cement in a cementitious system is crucial for development of the high-quality cementbased construction material. Complementary experiments of Xray diffraction analysis (XRD), scanning electron microscopy (SEM) and time-resolved laser fluorescence spectroscopy (TRLFS) using europium (Eu(III)) as an optical probe are used to analyse the hydration process of two cement systems in the absence and presence of different organic admixtures.
We Show that different analysed admixtures and the used sulphate carriers in each cement system have a significant influence on the hydration process, namely on the time-dependence in the formation of different hydrate phases of cement. Moreover, the effect of a particular admixture is related to the type of sulphate carrier used.
The quantitative information on the amounts of the crystalline cement paste components is accessible via XRD analysis. Distinctly different morphologies of ettringite and calcium-silicate-hydrates (C-S-H) determined by SEM allow visual conclusions about formation of these phases at particular ageing times. The TRLFS data provides information about the admixture influence on the course of the silicate reaction. The dip in the dependence of the luminescence decay times on the hydration time indicates the change in the structure of C-S-H in the early hydration period. Complementary information from XRD, SEM and TRLFS provides detailed information on distinct periods of the cement hydration process.
