The high ionization potentials and low wavelength emission lines of halogens difficult the analysis of their organohalogen compounds by ICP-based methods [1]. Other techniques, such as combustion ion chromatography (CIC) or ion-selective electrodes have proven to be more reliable to determine these elements. However, these techniques are more time-consuming and/or have lower precision and accuracy.
High resolution continuum source graphite furnace molecular absorption spectrometry (HR CS GFMAS) allows the determination of solid and liquid samples by monitoring a molecule formed with the analyte and a molecule forming reagent. In the case of F, Cl and Br, several molecule forming agents have been studied for their determination. Among all of them, calcium can form a stable bond with the three elements and provides optimal analytical properties for trace analysis [2]. However, the spectral window of commercially available continuum source AAS instruments is not broad enough to cover the wavelength range necessary for a simultaneous determination. Thus, coupling it with a modular simultaneous echelle spectrograph (MOSES), which can register around 200 nm in a single measure, allows the simultaneous study of the three analytes [3].
This instrumentation is capable of register more than 200 nm of the spectra per measure keeping a high resolution. This spectral window is suitable for the monitorization of a variety of transitions of the three molecules. However, some of them are overlapped and the competitive mechanism of formation of the three molecules modify their sensitivity depending on the proportions in the sample.
In this study, a compromise in the conditions and modifiers were selected in order to maximize the sensitivity and signal to noise ratio for the three molecules. The determination was carried out by standard addition of CaF and Cl and Br were obtained from the PLS model.

The primary screening of hybridoma cells is a time-critical and laborious step during the development of monoclonal antibodies. Often critical errors occur in this phase, which supports the notion that the generation of monoclonal antibodies with hybridoma technology is difficult to control and hence a risky venture. We think that it is crucial to improve the screening process to eliminate most of the immanent deficits of the conventional approach. With this new microarray-based procedure, several advances could be achieved: Selectivity for excellent binders, high throughput, reproducible signals, avoidance of misleading avidity (multivalency) effects, and simultaneous performance of competition experiments. The latter can directly be used to select clones of desired cross-reactivity properties. In this paper, a model system with two excellent clones against carbamazepine, two weak clones and blank supernatant has been designed to examine the effectiveness of the new system. The excellent clones could be detected largely independent of the IgG concentration, which is unknown during the clone screening since the determination and subsequent adjustment of the antibody concentration is not possible in most cases. Furthermore, in this approach, the enrichment, isolation, and purification of IgG for characterization is not necessary. Raw cell culture supernatant can be used directly, even when fetal calf serum (FCS) or other complex media had been used. In addition, an improved method for the oriented antibody-immobilization on epoxy-silanized slides is presented. Based on the results of this model system, we conclude that this approach should be preferable to most other protocols leading to many of false positives, causing expensive and lengthy confirmation steps to weed out the poor clones.
During R/V Meteor cruise 141/1, pore fluids of near surface sediments were investigated to find indications for hydrothermal activity in the Terceira Rift (TR), a hyperslow spreading center in the Central North Atlantic Ocean. To date, submarine hydrothermal fluid venting in the TR has only been reported for the D. João de Castro seamount, which presently seems to be inactive. Pore fluids sampled close to a volcanic cone at 2,800‐m water depth show an anomalous composition with Mg, SO4, and total alkalinity concentrations significantly higher than seawater and a nearby reference core. The most straightforward way of interpreting these deviations is the dissolution of the hydrothermally formed mineral caminite (MgSO4 0.25 Mg (OH)2 0.2H2O). This interpretation is corroborated by a thorough investigation of fluid isotope systems (δ26Mg, δ30Si, δ34S, δ44/42Ca, and 87Sr/86Sr). Caminite is known from mineral assemblages with anhydrite and forms in hydrothermal recharge zones only under specific conditions such as high fluid temperatures and in altered oceanic crust, which are conditions generally met at the TR. We hypothesize that caminite was formed during hydrothermal activity and is now dissolving during the waning state of the hydrothermal system, so that caminite mineralization is shifted out of its stability zone. Ongoing fluid circulation through the basement is transporting the geochemical signal via slow advection toward the seafloor.
Unter den von Humboldt in Amerika erworbenen Schriftstücken der Kolonialzeit findet sich eine Tributliste aus dem Jahr 1569, die der Otomí Bevölkerung aus dem Ort Mizquahuala im Staat Hidalgo/ Mexiko zugeordnet wird. Sie ist in Form eines Kalenders ausgeführt, Wochentage und verschiedene Produkte lassen sich unterscheiden. Eduard Seler beschreibt dieses Manuskript zusammen mit einer weiteren Tributliste, die in Inhalt und Zeichnung anders gestaltet ist, jedoch ebenfalls aus dieser Region stammt.
Mit den für die Analyse von Manuskripten bewährten Untersuchungsmethoden werden Papier und Farbmittel identifiziert. Hierzu zählen Raman-, FTIR-, Röntgenfluoreszenz- und VIS- Spektroskopie. Charakteristisch für die Herstellung von schriftlichen Dokumenten in der Kolonialzeit von Amerika ist die Verwendung von indigenem Material wie Papier aus Baumrinde und Pflanzenfarben zusammen mit den aus Europa bekannten Materialien. Geklärt wird, welche Auswahl von Farbstoffen zur Herstellung der Dokumente genutzt wurde, die diese als Gruppe klassifizieren könnte.
Die den beiden Fragmenten zugeordneten Farbmaterialien sowie formale und inhaltliche Aspekte lassen vermuten, dass zwei weitere Schriftstücke der Berliner Sammlung `Manuscripta Americana´, deren Herkunft nicht bekannt ist, mit ihnen in Beziehung gebracht werden können. Stilistisch gleiche Fragmente befinden sich zudem in Mexiko, Paris und den USA.
Im Vergleich dieser Fragmente untereinander wird diskutiert, inwieweit die über die Materialanalyse identifizierten spezifischen Farbstoffe einen Hinweis auf eine gemeinsame lokale Herkunft zulassen.
The presence of any aflatoxin contamination in exported figs needs to be monitored and measured through reliable and traceable methods, which require pure and matrix certified reference materials. On the other hand, certified reference materials (CRM) for determination of aflatoxins in dried fig are not yet available. Moreover, there is a lack of CRMs to be used in routine testing laboratories for method validation and quality control. The routine testing laboratories, participating in commercial proficiency testing (PT) programs, use the results available from consensus values to evaluate the performance of the participating laboratories, rather than metrologically traceable assigned values. This study initially proposed as a key comparison and presented at the EURAMET TC-MC SCOA meeting in Malta in 2015 and subsequently at the CCQM OAWG meeting in April 2015, proposes a CRM candidate for determination of levels of aflatoxins B1, B2, G1, G2 and their total in dried. Evidence of successful participation in formal, relevant international comparisons are needed to document measurement capability claims (CMCs) made by national metrology institutes (NMIs) and designated institutes (DIs).
In total nine NMI/DI participated in the Track C Key Comparison CCQM-K138 Determination of aflatoxins (AFB1, AFB2, AFG1, AFG2 and Total AFs) in Dried Fig. Participants were requested to evaluate the mass fractions expressed in ng/g units, of aflatoxins B1, B2, G1, G2 and total aflatoxin in a dried food matrix, dried fig. The CCQM-K138 results for the determination of aflatoxins (AFB1, AFB2, AFG1, AFG2 and total AFs) are ranging from 5.17 to 7.27 ng/g with an %RSD of 10.47 for AFB1, ranging from 0.60 to 0.871 ng/g with an %RSD of 11.69 for AFB2, ranging from 1.98 to 2.6 ng/g with an %RSD of 10.36 for AFG1, ranging from 0.06 to 0.32 ng/g with an %RSD of 35.6 for AFG2, and ranging from 8.29 to 10.31 ng/g with an %RSD of 7.69 for Total AFs. All participants based their analyses on LC-MS/MS, HPLC-FLD, HR-LC/MS, and IDMS. Brief descriptions of the analytical methods used by the participants, including sample preparation, analytical technique, calibrants, and quantification approach are summarized in Appendix F. Linear Pool was used to assign the Key Comparison Reference Values (KCRVs) for B1, B2, G1, G2 and total aflatoxins. Due to the traceability requirements for the calibrants not being met, results of KEBS, INTI, VNIIM and BAM were excluded from KCRV determination.
Successful participation in CCQM-K138 demonstrates the following measurement capabilities in determining mass fraction of organic compounds, with molecular mass of 100 g/mol to 500 g/mol, having high polarity (pKow > -2), in mass fraction range from 0.05 ng/g to 500 ng/g in dried food matrices.
Grasses accumulate silicon in the form of silicic acid, which is precipitated as amorphous silica in microscopic particles termed phytoliths. These particles comprise a variety of morphologies according to the cell type in which the silica was deposited. Despite the evident morphological differences, phytolith chemistry has mostly been analysed in bulk samples, neglecting differences between the varied types formed in the same species. In this work, we extracted leaf phytoliths from mature plants of Sorghum bicolor (L.) Moench. Using solid state NMR and thermogravimetric analysis, we show that the extraction methods alter greatly the silica molecular structure, its condensation degree and the trapped organic matter. Measurements of individual phytoliths by Raman and synchrotron FTIR microspectroscopies in combination with multivariate analysis separated bilobate silica cells from prickles and long cells, based on the silica molecular structures and the fraction and composition of occluded organic matter. The variations in structure and composition of sorghum phytoliths suggest that the biological pathways leading to silica deposition vary between these cell types.
Die Geräteentwicklungen im Bereich der Niederfeld-NMR-Spektroskopie im vergangenen Jahrzehnt ermöglichen den Einsatz kompakter, portabler Magnete mit geringen Streufeldern in Laborumgebungen und industriellen Produktionsanlagen. Somit werden neue Möglichkeiten für hochauflösende NMR-Experimente zur Reaktionsüberwachung eröffnet. Im Rahmen des EU-Projekts CONSENS wurden die Möglichkeiten dieser Methode umfassend anhand einer technisch bedeutenden Reaktion (elektrophile aromatische Substitutionsreaktionen) evaluiert.
Um die Flexibilität einer kontinuierlichen und modularen Pilotanlage durch Echtzeit- Qualitätskontrolle zu fördern, wurde ein vollständig automatisiertes Online- NMR-Modul entwickelt. Der Einsatz eines kommerziellen Niederfeld-NMR-Geräts im industriellen Umfeld wurde durch die entwickelten Lösungen der automatisierten Datenanlyse sowie durch ein zertifiziertes Sicherheitskonzept für den Betrieb in explosionsgefährdeten Zonen ermöglicht. Neben der Überwachung der Produktqualität wurden Online-NMR-Daten in einem neuen iterativen Optimierungsansatz zur Maximierung des Anlagenertrags eingesetzt und dienten als zuverlässige Referenz für die Kalibrierung eines Nahinfrarot-Spektrometers.
Für die Entwicklung einer robusten Datenauswertung der NMR-Spektren, die dem Anspruch der Flexibilität bei Produktwechseln genügt, wurden zunächst Versuche im Labormaßstab durchgeführt, um eine Datenbasis zu schaffen. In diesen Versuchen wurden die aromatischen Amine Anilin, p-Toluidin und p-Fluoranilin mit o-Fluornitrobenzol gekoppelt. Durch Zugabe einer Organolithium-Verbindung (Li- HMDS) findet ein Protonenaustausch zwischen dem primären Amin und Li-HMDS statt. Dies führt zu einer hohen Reaktionsenthalpie und instabilen Aryllithium-Verbindungen. Die Reaktionen wurden hinsichtlich anfallender Zwischenprodukte mittels Hochfeld-NMR-Spektroskopie analysiert.
Nachfolgend wurden die Reaktionen sowohl im Semi-Batch-Verfahren als auch im kontinuierlichen Laborbetrieb mit Online-Niederfeld-NMR-Spektroskopie und Online- Hochfeld-NMR-Spektroskopie untersucht. Da die gemessenen NMR-Spektren besonders im aromatischen Spektralbereich hohe Signalüberlappungen der Reaktanden aufweisen, wurden chemometrische Modelle entwickelt und anhand der Hochfeld-NMR-Methode validiert. Die verwendeten Durchflusszellen für die Niederfeld-NMR-Spektroskopie wurden hinsichtlich ihrer Anwendbarkeit für quantitative Messungen im kontinuierlichen Durchfluss untersucht. Es konnte gezeigt werden, dass Messungen mit einer additiv gefertigten Keramikdurchflusszelle prinzipiell möglich sind.

Hierbei geht es um die Bestimmung von C, O und N in Vanadiumcarbid, Wolframcarbid und Niobcarbidmaterialien, einen hierzu durchgeführten Ringversuch mit Beteiligung der Kollegen des AK Sonderwerkstoffe und den Vergleich von Verfahrenskenngrößen.
Acoustically levitated droplets have been suggested as compartmentalized, yet wall-less microreactors for high-throughput reaction optimization purposes. The absence of walls is envisioned to simplify up-scaling of the optimized reaction conditions found in the microliter volumes. A consequent pursuance of high-throughput chemistry calls for a fast, robust and sensitive analysis suited for online interrogation. For reaction optimization, targeted Analysis with relatively low sensitivity suffices, while a fast, robust and automated sampling is paramount. To follow this approach, in this contribution, a direct coupling of levitated droplets to a homebuilt ion mobility spectrometer (IMS) is presented. The sampling, Transfer to the gas phase, as well as the ionization are all performed by a single exposure of the sampling volume to the resonant output of a mid-IR laser. Once formed, the nascent spatially and temporally evolving analyte ion cloud needs to be guided out of the acoustically confined trap into the inlet of the ion mobility spectrometer. Since the IMS is operated at ambient pressure, no fluid dynamic along a pressure Gradient can be employed. Instead, the transfer is achieved by the electrostatic potential gradient inside a dual ring electrode ion optics, guiding the analyte ion cloud into the first stage of the IMS linear drift tube accelerator. The design of the appropriate atmospheric pressure ion optics is based on the original vacuum ion optics design of Wiley and McLaren. The obtained experimental results nicely coincide with ion trajectory calculations based on a collisional model.
Perhaps due in no small part to its historical mystique, mechanochemistry has been shrowded in relative obscurity for quite some time. The successes of mechanochemistry in organic synthesis have often been curtailed by a fundamentally limited mechanistic understanding. Lately, however, the community has made great strides towards understanding the fundamentals, as well as large steps forward regarding industrially significant scale-up via twin-screw extrusion. A variety of recent work has traded in some of mechanochemistry’s mystique for simple, straight-forward chemical guidelines.
Um in einem veränderten Umfeld erfolgreich bestehen zu können, müssen Chemieunternehmen neue Pfade beschreiten. Dazu gehört insbesondere das Potential digitaler Technologien. Mit flexiblen, modularen chemischen Vielzweck-Produktionsanlagen lassen sich häufig wechselnde Produkte mit kürzeren Vorlauf- und Stillstandzeiten zwischen den Kampagnen und dennoch hoher Qualität realisieren. Intensivierte, kontinuierliche Produktionsanlagen erlauben auch den Umgang mit schwierig zu handhabenden Substanzen.
Grundvoraussetzung für solche Konzepte ist eine hochautomatisierte "chemische" Prozesskontrolle zusammen mit Echtzeit-Qualitätskotrolle, die "chemische" Informationen über den Prozess bereitstellt. In einem Anwendungsbeispiel wurde eine pharmazeutische Lithiierungsreaktion aus einer modularen Pilot-Anlage betrachtet und dabei die Vorzüge eines vollautomatischen NMR-Sensors untersucht. Dazu wurde ein kommerziell erhältliches Benchtop-NMR-Spektrometer mit Permanentmagnet auf die industriellen Anforderungen, wie Explosionsschutz, Feldkommunikation und vollautomatischer, robuster Datenauswertung angepasst. Der NMR-Sensor konnte schließlich erfolgreich im vollautomatischen Betrieb nach fortschrittlichen Regelkonzepten und für die Echtzeitoptimierung der Anlage getestet werden. Die NMR-Spektroskopie erwies sich als hervorragende Online-Methode und konnte zusammen mit einer modularen Datenauswertung sehr flexibel genutzt werden. Die Methode konnte überdies als zuverlässige Referenzmethode zur Kalibrierung konventioneller Online-Analytik eingesetzt werden.
Zukünftig werden voll integrierte und intelligent vernetzte "smarte" Sensoren und Prozesse eine kontinuierliche Produktion von Chemikalien und Pharmazeutika mit vertretbaren Qualitätskosten möglich machen.
Metabolites from Alternaria fungi exhibit a variety of biological properties such as phytotoxic, cytotoxic, or antimicrobial activity. Optimization of a literature procedure culminated in an efficient total synthesis of (−)-altenuene as well as a stable isotope-labeled derivative suitable for implementation in a LC-MS/MS method for mycotoxin analysis.
In many scientific fields, isotopic analysis can offer valuable information, e.g., for tracing the origin of food products, environmental contaminants, forensic and archaeological samples (provenance determination), for age determination of minerals (geochronological dating) or for elucidating chemical processes. Up to date, typically bulk analysis is aimed at measuring the isotopic composition of the entire elemental content of the sample. However, the analyte element is usually present under the form of different species. Thus, separating species of interest from one another and from matrix components prior to isotope ratio measurements can provide species-specific isotopic information, which could be used for tracing the origin of environmental pollutants and elucidation of (environmental) speciation. Using on-line hyphenations of separation techniques with multicollector-ICP-MS (MC-ICP-MS) can save time and effort and enables the analysis of different species during a single measurement.
In this work, we developed an on-line hyphenation of CE with multicollector-ICP-MS (CE/MC-ICP-MS) for isotopic analysis of sulfur species. With this method, the isotopic composition of sulfur in sulfate originating from river water could be analyzed without sample preparation. The results were compared with data from off-line analysis of the same samples to ensure accuracy. The precision of the results of the on-line measurements was high enough to distinguish the rivers from one another by the isotopic signature of the river water sulfate. Next to environmental applications, a future field could be species-specific isotopic analysis of biomolecules, as sulfur is the only covalently bound constituent of proteins which can be analyzed by MC-ICP-MS.
Unternehmen der chemischen Industrie müssen neuen Pfade beschreiten, um in einem veränderten Umfeld erfolgreich bestehen zu können. Dazu gehört insbesondere, das Potenzial digitaler Technologien zu nutzen. Die volle Integration und intelligente Vernetzung von Systemen und Prozessen kommt allerdings nur zögerlich voran. Dieser Beitrag ist ein Loblied auf die Feldebene. Er möchte dazu ermutigen, die Digitalisierung der Prozessindustrie auf Basis smarter Sensorik, Aktorik und Kommunikation ganzheitlicher zu denken und informiert über aktuelle technische Perspektiven, wie das Ein-Netzwerk-Paradigma, Ad-hoc-Vernetzungen, Edge-Computing, FPGAs, virtuelle Maschinen oder Blockchain. Diese geben smarter Sensorik, Aktorik und Kommunikation eine völlig neue Perspektive.
Multi-collector inductively coupled plasma-mass spectrometry (MC-ICP-MS) has evolved significantly since its introduction in 1992. The second and third generation instruments now allow isotope ratio measurements at unprecedented precisions, 0.001 % or better. However, precision alone is not enough for producing accurate and reliable isotope ratio measurements. Metrological considerations such as the selection of suitable calibration strategies, proper assessment of instrumental biases, and the estimation of overall measurement uncertainty remain critical to the measurement process. Properly assessed, measurement uncertainty then provides the interval within which a result can be considered both accurate and precise.
All mass spectrometric measurements are affected by instrumental mass discrimination and produce isotope ratios that are biased relative to their “true” ratio. To produce accurate and traceable isotope ratio measurements, it is imperative that certified isotope reference materials (iCRMs) be used for calibration and validation purposes. iCRMs reporting absolute isotope ratios are an analyst’s first choice, particularly when its uncertainty is sufficient for the intended use. However, when smaller uncertainties are required to resolve subtle differences between samples, delta-scale measurements become important. Here, the difference between an isotope ratio measured in a sample and in an internationally accepted isotope reference material (iRM) is determined. This deviation can be positive or negative relative to the iRM, is called a delta value, and is often expressed in per mil units.
This presentation will highlight the potential for MC-ICP-MS to produce isotope ratio measurements with minimal uncertainties by examining three applications involving Mg isotopes: 1) the certification of a set of iCRMs for their absolute isotope ratio using a gravimetric isotope mixture approach; 2) the comparison of these iCRMs with currently accepted Mg delta-scale reference materials through intercalibration, and 3) the determination of isotope fractionation exponents for geochemical applications.
Gegenstand der vorzustellenden Arbeiten ist die Prüfung der Umwelt-beständigkeit und -verträglichkeit von Materialien und Produkten hinsichtlich der Emission von potenziellen Schadstoffen in die Umwelt. Hierzu werden chemisch-physikalische Einflüsse (Bewitterung) und mikrobielle Beanspruchungen an Modellmaterialien evaluiert. So werden die Freisetzungsraten von Schadstoffen in Abhängigkeit der Beanspruchung beschrieben. Als Modellmaterialien kommen die Polymere Polystyrol (PS) und Polypropylen (PP) zum Einsatz. Synergistische Effekte der Bewitterungsparameter und der mikrobiologischen Beanspruchung sollen dabei ebenso betrachtet werden, wie die gezielte Alterung. Auch findet eine Beschreibung des Verhaltens der ausgetragenen Schadstoffe (polybromierte Flammschutzmittel) in den Umweltkompartimenten Boden oder Wasser statt. Hier sind mit Hilfe der zu entwickelnden Screening- und non-Target-Analyseverfahren die Transformation und der Metabolismus durch Mikroorganismen zu beschreiben. Aus den Ergebnissen sollen Korrelationen zwischen den künstlichen Alterungsverfahren und realen Szenarien abgeleitet werden.
Warfarin als ein Vertreter der NSO-Hetereozyklen, ist seit den 1950er Jahren einer der bekanntesten Gerinnungshemmer. Beim Menschen wird es bis heute zur Vorbeugung von Thrombose und der Behandlung von Vorhofflimmern und Arrhythmie eingesetzt. Doch schon einige Jahre vor der FDA Zulassung als Medikament wurde es zur Bekämpfung von Nagetieren eingesetzt. Bis heute wurde es nur teilweise durch weitaus wirksamere antikoagulante Rodentizide (ARs) der zweiten Generation, sogenannte Superwarfarine, ersetzt. In der Fachliteratur finden sich zahlreiche Beispiele für sekundäre und tertiäre Vergiftung mit ARs bei Wildtieren und anderen Nichtzieltieren. Bisher ist relativ wenig über die Bioakkumulation, Persistenz und Toxizität von ARs und deren Transformationsprodukten (TPs) in der terrestrischen und aquatischen Umwelt, sowie der Nahrungskette bekannt.
Um Einblicke in mögliche Transformations- und Abbauprozesse von Warfarin zu erhalten und um Umwelteinflüsse zu simulieren, wurde es in einer elektrochemischen Reaktorzelle oxidativ umgesetzt und mittels Massenspektrometrie analysiert. Hinsichtlich der Wasseraufbereitung in Kläranlagen wurden technisch relevante Simulationsverfahren wie Chlorung, UV-Bestrahlung und Ozonung angewendet um potentielle Eliminierungswege aufzuklären. Gebildete Transformationsprodukte wurden daher mittels unterschiedlicher chromatografischer Verfahren getrennt, massenspektrometrisch analysiert und verglichen.
Die Wettbewerbsfähigkeit der Prozessindustrie basiert auf der Sicherung der geforderten Produktqualität bei einer optimalen Nutzung von Anlagen, Rohstoffen und Energie. Ein Weg zur wissensbasierten Produktion führt über die Betrachtung der wesentlichen Apparate-, Prozess- und Freigabedaten aus Betrieben und Labors. Das Potenzial dieser Daten wird heute vielfach noch nicht konsequent für ein umfassendes Verständnis der Produktion genutzt.
Neben Fragen zur Datenerfassung, Datenkonnektivität und Datenintegrität müssen solche Daten für eine ganzheitliche Prozessanalyse zunächst mit Kontextinformationen zusammengebracht werden. Datenquellen enthalten Zeitwertpaare, aber auch diskrete Daten aus LIMS (Laboratory Information Management Systems) oder ELN (Electronic Laboratory Notebooks) und werden zunehmend durch 2D- und 3D-Daten aus der Chromatographie-Massenspektrometrie oder bildbasierter Analytik ergänzt.
Für die automatisierte Merkmalsextraktion, etwa zur Extraktion chemischer Informationen aus den oben genannten Datenquellen werden multivariate Werkzeuge und Algorithmen genutzt. Multivariate Statistiken wie PCA (Principle Component Analysis), PLS (Partial Least Squares) und LDA (Latent Discriminant Analysis) bilden die erste Grundlage für die Datenanalyse. Für diese Verfahren sind heute Datenvorbehandlungsschritte nötig. Die Modellbildung geschieht manuell und ist sehr aufwendig.
Können diese Daten im Zeitalter von ML (Machine Learning) und KI (Artificial Intelligence) anderweitig sinnvoll genutzt werden und ohne klassische Modellbildung? Die Bezeichnung „Big Data“ als Voraussetzung für datengetriebene Auswerteverfahren ist für die Prozessindustrie allerdings unpassend, denn auch bei mengenmäßig großen Datensätzen liegen für Kampagnen typischerweise nur Informationen über einige Batches mit einer Serie von Messdaten vor, die genügend Varianz für eine datengetriebene Auswertung aufweisen – nicht vergleichbar mit den Datenmengen im WWW oder von großen Internet-Konzernen.
Ein Weg zur wissensbasierten Produktion führt über die Betrachtung der wesentlichen Apparate-, Prozess- und Freigabedaten aus Betrieben und Labors. Das Potenzial dieser Daten wird heute vielfach noch nicht konsequent für ein umfassendes Verständnis der Produktion genutzt.

Advances and Applications of Molecular Absorption Spectrometry: from Non-Metals to Isotope Analysis

The present work covers two main aspects of high-resolution continuum source graphite furnace molecular absorption spectrometry (HR-CS-GFMAS), an analytical technique for elemental trace analysis.
First, a comprehensive mechanistic study of molecule formation in graphite furnaces is presented, which is a key step into the recovery of analytical signals. For this, the molecule formation of CaF was studied, which is used for the indirect analytical determination of fluorine in HR-CS-GFMAS. A zirconium coating catalyzes the CaF formation, and its structure was investigated. The kinetics of this reaction was established by monitoring its molecular spectrum at different atomisation temperatures. An Arrhenius plot showed a pseudo-first order reaction with respect to fluorine (n = 1). An intermediate state was isolated, and its structure was elucidated by spectroscopic methods: scanning electron microscopy with energy dispersive X-ray spectroscopy (SEM-EDX), X-ray photoelectron spectroscopy (XPS), X-ray absorption spectroscopy (XANES and EXAFS), and Raman microspectroscopy. Here a mechanism is proposed, where ZrO2 works as a heterogeneous catalyst: after a pyrolytic step, an intermediate state of ZrO(OCaF) is activated, and at higher temperatures, CaF(g) is released from the zirconium-coated graphite surface.
Second, analytical methods were developed by using HR-CS-MAS as detector for non-metals and isotope analysis. Therefore, the determination of organic absorbable chlorine in water, the quantification of fluorine in consume care products with declared perfluorinated ingredients, and the determination of sulfur content in crude oils were investigated. Finally, the high resolution of the instrumentation allows to measure isotopic shifts with high precision in some observed molecular spectra. Consequently, the molecular spectra of enriched isotopes of boron and magnesium were investigated, establishing so the potential of HR-CS-MAS for the accurate and precise determination of isotopic amount ratios.

Die vorliegende Arbeit befasst sich mit zwei zentralen Aspekten der High-Resolution-Continuum-Source-Molekülabsorptionsspektrometrie mit Graphitrohrtechnik (HR-CS-GFMAS), einer Analysetechnik für elementare Spurenanalyse.
Der erste Teil der Arbeit umfasst eine mechanistische Studie zur Molekülbildung auf Graphitoberflächen. Dies ist ein wichtiger Schritt, um analytische Signale zu entdecken. Dazu wurde die Molekülbildung von CaF analysiert, welches für die indirekte, analytische Bestimmung von Fluor in HR-CS-GFMAS genutzt wird. Die CaF Bildung wurde mittels einer Beschichtung aus Zirconium katalysiert und deren Struktur analysiert. Die Kinetik dieser Reaktion wurde durch Beobachtung des jeweiligen Molekülspektrums bei verschiedenen Atomisierungstemperaturen beobachtet. Ein Arrheniusplot zeigte für Fluor (n = 1) eine Reaktion Pseudo-erster Ordnung. Ein Übergangszustand wurde über die mit Zirconium überzogene Grafitoberfläche isoliert und seine Struktur mittels spektroskopischer Methoden Energiedispersive Rasterelektronenmikroskopie / Röntgenspektroskopie (REM-EDX), Röntgenphoto¬elektronenspektroskopie (XPS), Röntgenabsorptionsspektroskopie (XAS) und Raman Spektroskopie untersucht. Auf Grundlage dieser Ergebnisse wird ein Mechanismus vorgeschlagen, bei dem ZrO2 als heterogener Katalysator fungiert; in Folge einer Pyrolysestufe wird ein Übergangszustand des ZrO(OCaF) aktiviert, welcher bei höheren Temperaturen CaF(g) an der Zirconium-Graphitoberfläche freisetzt.
Im zweiten Teil der Arbeit werden Analysemethoden entwickelt, in dem HR-CS-MAS als Detektor für Nichtmetalle und Isotopanalyse angewandt wird. Hierfür wurde organisch gebundenes Chlor im Wasser bestimmt, der Fluorgehalt in Pflegeprodukten mit perfluorierten Inhaltsstoffen quantifiziert und der Schwefelgehalt in Erdöl untersucht. Weiterhin ermöglicht die hohe Auflösung der Messgeräte eine präzise Bestimmung der Isotopenverschiebung einiger untersuchter Molekülspektren. Daher wurden die Molekülspektren angereicherter Bor- und Magnesiumisotope untersucht. Auf diese Weise wurde das Potential von HR-CS-MAS für die akkurate und präzise Bestimmung von Isotop-Mengenverhältnissen nachgewiesen.

Their drawbacks include the high costs for instruments and their operation, experienced operators and elaborate time-consuming chromatographic sample preparation. Recently, an optical spectrometric method has been proposed as faster and low-cost alternative for the analysis of isotope ratios: high-resolution continuum source graphite furnace molecular absorption spectrometry (HR-CS-GFMAS).
For the determination of Mg isotope ratios in selected rock reference materials, the high-resolution molecular absorption spectrum of in-situ generated MgF molecule was studied applying multivariate analysis and the results compared with MC-ICP-MS. Samples were dissolved by acid digestion and Mg isotopes analyzed with and without matrix extraction. The absorption spectra were recorded for MgF for the electronic transition X 2Σ → B 2Σ+. The MgF spectrum is described as the linear combination of their isotopic components or isotopologues: 24MgF, 25MgF, and 26MgF. The isotope analysis was done by deconvolution of the MgF spectrum by partial least square regression (PLS). A PLS model was built and calibrated with enriched isotope spikes and certified reference materials. Spectra data was preprocessed by a derivate of second order and venetian blinds cross-validation was employed for finding the optimum latent variables. Finally, the model was refined by a genetic algorithm which identified the best subset of variables for a precise and accurate regression. Results are compatible with those obtained by MC-ICP-MS with an accuracy of ± 0.3‰ with uncertainties ranging between 0.02 to 0.6‰. This accuracy and precision discriminate the isotope fractionation in geological samples, and it is suitable for earth’s climate studies.

Simultaneous multielemental analysis of crude oils by high-resolutions absorption spectrometry

When crude oil arrives at a refinery it needs a lot of processing before it is suitable for cracking into lighter fractions. Sulfur has to be extracted to meet ultra-low sulfur legislation for most of the fuel grades, and desalination is a crucial process as chlorine within salts is corrosive to refinery equipment. Measuring the amounts of sulfur and chlorine within crude oil is the first step in a complex clean-up process.
Heavy metals, such as vanadium, nickel and iron need to be removed too. These metals can poison the catalyst used to crack the oil into lighter fractions. This is costly as it reduces the useful life of the catalyst. Like chlorine, heavy metals also have a corrosive effect on the plant. There is some evidence that the presence of some metals in the final product can reduce performance.
A fast determination of elements and size of suspended particles is vital for diagnosis and safeguard of refinery equipment. However, sample preparation for current analytical methods consumes precious time and lost particle size information.
High-resolution continuum source graphite furnace absorption spectrometry (HR-CS-GFAS) is proposed as a fast analytical method for elemental determination in crude oils and potentially for simultaneous multielement and particle size analysis. This HR-CS-GFAS instrumentation is coupled to a modular simultaneous echelle spectrometer (MOSES) and provides a full optical window with high resolution (from 180 to 900 nm with bandwidth λ/170,000). By using this set-up, it is possible to generate a multiparameter 3D spectral image (atomic and molecular lines, isotopic shift, atomization delay, and intensities). The generated spectral images can be analyzed by multivariate regressions for the elemental and particle size analysis. Additionally, if several atomic and molecular lines are simultaneously measured, they may be used in multi-energy calibration (MEC), a method based on the use of many wavelengths of the same absorbing/emitting entity to improve the accuracy in analytical atomic spectrometry. This MEC approach combined with multivariate image analysis allows the quantification of selected elements (Si, Al, Ni, Fe, V, S, and Cl) and overcomes matrix effects.

Isotope amount ratios in rock samples with and without matrix separation were analyzed. Calculated δ-values were accurate and obtained with precisions ranging between 0.2 ‰ and 0.5 ‰ (1 SD, n = 10). On the other hand, LAMIS allows the direct analysis of solid samples with the extended possibility of in-situ analysis. Main advantages, limitations, and scopes of both optical techniques are going to be discussed and compared to MC-ICP-MS.

Boron and Magnesium present two and three stable isotopes respectevely. It is due to their relatively large mass difference (~ 10%) that isotope fractionation leads to considerable isotope amount ratio variations in the nature. These have been used as a proof of provenance of mineral and biological samples, to estimate a contamination source and to the determination of geological processes by erosion or subduction. Additionally, boron is employed in the nuclear industry due to the capability of its isotope 10B to thermal-neutron capture and therefore 10B enriched boric acid solutions are used in the cooling system of thermonuclear facilities and in the alloying of steel and carbides for protective shielding. Traditionally, isotope ratio variations have been determined by mass spectroscopic methods.
Here an alternative faster and low cost method for isotope ratio determination is proposed: high-resolution continuum source molecular absorption spectrometry (HR-CS-MAS). Isotope amount ratios have been determined by monitoring the absorption spectrum of boron monohydride (BH) for boron and Magnesium monofluoride (MgF) for magnesium in a graphite furnace HR-CS-MAS. Bands (0→0) and (1→1) were evaluated. Partial least square regression (PLS) for analysis of samples and reference materials were applied. For this, a spectral library with different isotopes ratios for PLS regression was built. Results obtained are metrologically compatible with those reported by mass spectrometric methods. Moreover, a precision and accuracy of the method of ± 0.5 ‰. This accuracy and precision is comparable with those obtained by thermal ionization mass spectrometry (TIMS) and multiple collector inductively coupled plasma mass spectrometry (MC-ICP-MS) for boron isotope ratio measurements

Optical spectrometric methods are proposed as faster and low-cost alternative for the analysis of isotope ratios of selected elements by means of high-resolution continuum source molecular absorption spectrometry (HR-CS-MAS) and laser ablation molecular isotopic spectrometry (LAMIS). First, stable isotope amount compositions of boron (B) and magnesium (Mg) were determined based on the absorption spectra of in-situ generated heteronuclear diatomic molecules (MH or MX) in graphite furnace HR-CS-MAS. The use of a modular simultaneous echelle spectrograph (MOSES) helps to find the maximal isotope shift in the diatomic molecular spectra produced in a graphite furnace by using isotopic spike solutions. Isotopes of boron (10B and 11B) were studied via their hydrides for the electronic transition X 1Σ+ → A 1Π. The spectrum of a given sample is a linear combination of the 10BH molecule and its isotopologue 11BH. Therefore, the isotopic composition of samples can be calculated by a partial least square regression (PLS). For this, a spectral library was built by using samples and spikes with known isotope composition. Boron isotope ratios measured by HR-CS-MAS are identical with those measured by mass spectrometric methods at the 0.15 ‰ level. Similar results were obtained for a multiple isotope system like Mg (24Mg, 25Mg, and 26Mg), where isotope shifts of their isotopologues can be resolved in the MgF molecule for the electronic transition X 2Σ → A 2 Πi. Finally, the application of molecular spectrometry via emission by LAMIS is compared and discussed.
Chemical companies must find new paths to successfully survive in a changing environment. The potential of digital technologies belongs to these. Flexible and modular chemical plants can produce various high-quality products using multi-purpose equipment with short down-times between campaigns and reduce time to market for new products. Intensified continuous production plants allow for difficult to produce compounds.
Therefore, fully automated “chemical” process control along with real-time quality control are prerequisites to such concepts and thus should be based on “chemical” information. The advances of a fully automated NMR sensor were exploited, using a given pharmaceutical lithiation reaction as an example process within a modular pilot plant. A commercially available benchtop NMR spectrometer was integrated to the full requirements of an automated chemical production environment such as , e.g., explosion safety, field communication, and robust evaluation of sensor data. It was thereof used for direct loop advanced process control and real-time optimization of the process. NMR appeared as preeminent online analytical tool and allowed using a modular data analysis tool, which even served as reliable reference method for further PAT applications.
In future, such fully integrated and intelligently interconnecting “smart” systems and processes can speed up the high-quality production of specialty chemicals and pharmaceuticals.
Multiple sclerosis (MS) is the most common acquired neurological disease affecting Young adults. It leads to myelin destruction and formation of lesions in the brain and permeabilization of the blood-brain barrier (BBB).
Gadolinium based contrast agents (GBCA), used for diagnosis and monitoring by MRI, might accumulate in tissue, including brain. Using an animal model, application of linear and macrocyclic GBCA was investigated. Analysis was performed by laser ablation inductively coupled plasma (LA-ICP) MS. This method works as a bioimaging tool for sample thin sections and allows to identify regions of accumulated Gd in the samples.
In healthy and diseased mice, inflammationmediated changes in the brain were investigated by application of GBCA. The mean objective is to understand the changes in inflamed tissue and correlate alterations of the BBB by LA-ICP-MS (Imaging Mass Cytometry).
Real-time monitoring of newly acidified organelles during autophagy in living cells is highly desirable for a better understanding of intracellular degradative processes. Herein, we describe a reaction-based boron dipyrromethene (BODIPY) dye containing strongly electron-withdrawing diethyl 2-cyanoacrylate groups at the α-positions. The probe exhibits intense red fluorescence in acidic organelles or the acidified cytosol while negligible fluorescence in other regions of the cell. The underlying mechanism is a nucleophilic reaction at the central meso-carbon of the indacene core, resulting in the loss of π-conjugation entailed by dramatic spectroscopic changes of more than 200 nm between its colorless, non-fluorescent leuco-BODIPY form and its red and brightly emitting form. The reversible transformation between red fluorescent BODIPY and leuco-BODIPY along with negligible cytotoxicity qualifies such dyes for rapid and direct intracellular lysosome imaging and cytosolic acidosis detection simultaneously without any washing step, enabling the real-time monitoring of newly acidified organelles during autophagy.
Trialkoxysilane haben sich in den vergangenen Jahren als vielseitig einsetzbare Organosilane erwiesen. Die Einsatzgebiete erstrecken sich vom Witterungsschutz von Bauwerken über haftvermittelnde Eigenschaften in der Glasfaserindustrie, bei Dicht- und Klebstoffen, in Farben und Lacken bis hin zur Modifizierung von polymeren Werkstoffen.
Kommerzielle Benchtop-NMR-Spektrometer haben das Potential auch im Bereich der Silanchemie als Online-Methode zur Reaktionsüberwachung und für die Qualitätskontrolle eingesetzt zu werden. Interessante NMR-Kerne für die oben genanntem Produkte sind 1H und 29Si. In einer gemeinsamen Forschungskooperation zwischen EVONIK und BAM wurde anhand verschiedener Fallstudien die Anwendbarkeit der Niederfeld-NMR-Spektroskopie zur chemischen Analyse von Silanen evaluiert. Im Zuge der Fallstudien wurde gezeigt, wie Niederfeld-NMR-Spektroskopie die Möglichkeiten der Konzentrationsmessung auf neue Anwendungsgebiete erweitert, in denen bestehende Technologien wie z. B. NIR, Raman, UV/VIS, etc. mangels Referenzdaten nicht quantitativ eingesetzt werden können.
Fallstudie 1: Oligomerisierung
Eine Fallstudie setzte dazu an, den Hydrolyse- und Kondensationsverlauf mit einer geeigneten Online-NMR-Analytik zu beobachten, den Reaktionsfortschritt der Hydrolyse und Kondensation auf dieser Basis besser zu verstehen und zu optimieren. Zu diesem Zweck werden durch Zugabe von Wasser zunächst die Alkoxysubstituenten eines Trialkoxysilans hydrolysiert und entsprechende Silanole gebildet. Diese können dann über eine SiOH-Funktion an den zu modifizierenden Werkstoff anbinden und über weitere Silanolgruppen unter Ausbildung von Siloxaneinheiten vernetzen.
Fallstudie 2: Spaltung von cyclischen Silanverbindungen
In einer weiteren Fallstudie wurde die Kinetik der Aufspaltungen einer cyclischen Silanverbindung untersucht. Die Online-NMR-Analytik kam hierbei sowohl im Labor als auch in der industriellen Produktionsanlage zum Einsatz. Hierfür wurde eine vollständig automatisierte Einhausung verwendet, welche den Einsatz eines kommerziellen NMR-Spektrometers in explosionsgeschützten Bereichen ermöglicht.
Fallstudie 3: Qualitätskontrolle für Produktmischungen von Trialkoxysilanen
Für Produktmischungen eines Trialkoxysilans und weiteren Bestandteilen wie u. a. org. Stabilisatoren, Organozinnverbindungen, eines aromatischen Amins und org. Peroxide wurden quantitative 1H-Spektren akquiriert und eine automatische Auswertungsmethode basierend auf Indirect Hard Modeling (IHM) entwickelt. Für die Nebenkomponenten, deren Stoffmengenanteile bis zu 3 Mol-% betragen, wurden durch die zugrunde gelegte Methode typischerweise korrekte experimentelle Stoffmengenanteile gefunden, die weniger als 0,2 Mol-% vom Referenzwert abweichen.

In most chemical reactions, stable isotopes are fractionated in a mass-dependent manner, yielding correlated isotope ratios in elements with three or more stable isotopes. The proportionality between isotope ratios is set by the triple isotope fractionation exponent θ that can be determined precisely for, e.g., sulfur and oxygen by IRMS, but not for metal(loid) elements due to the lower precision of MC-ICP-MS analysis and smaller isotopic variations. Here, using Mg as a test case, we compute a complete metrologically robust uncertainty budget for apparent θ values and, with reference to this, present a new measurement Approach that reduces uncertainty on θ values by 30%. This approach, namely, direct educt-product bracketing (sample−sample bracketing), allows apparent θ values of metal(loid) isotopes to be determined precisely enough to distinguish slopes in three-isotope space. For the example of Mg, we assess appropriate quality Control standards for interference-to-signal ratios and Report apparent θ values of carbonate−seawater pairs. We determined apparent θ values for marine biogenic carbonates, where the foraminifera Globorotalia menardii yields 0.514 ± 0.005 (2 SD), the coral Porites, 0.515 ± 0.006 (2 SD), and two specimens of the giant clam Tridacna gigas, 0.508 ± 0.007 (2 SD) and 0.509 ± 0.006 (2 SD), documenting differences in the uptake pathway of Mg among marine calcifiers. The capability to measure apparent θ values more precisely adds a new dimension to metal(loid) δ values, with the potential to allow us to resolve different modes of fractionation in industrial and natural processes.
Zu den letzten beiden Jahrestagungen der GUS wurden Verfahren und Methoden vorgestellt, die es gestatten, den Transfer von Schadstoffen aus Materialien und Produkten in die Umwelt unter umweltrelevanten Bedingungen beschreiben zu können. Dabei lag der Fokus auf den Materialien und der eingesetzten Umweltsimulationen. In einem sich anschließenden Schritt soll nun das Verhalten der freigesetzten organischen Schadstoffe unter den Bedingungen von realen und simulierten Umwelteinflüssen charakterisiert und beschrieben werden. Hier werden im Detail drei potentielle Schadstoffe betrachtet, die Inhaltsstoffe bzw. Additive in den jeweils eingesetzten Materialien sind. Im Falle von Polypropylen (PP) und Polystyrol (PS) handelt es sich um polybromierte Flammschutzmittel BDE 209 (Decabrom-diphenylether, als Einzelsubstanz) und HBCD (Hexabromcyclododecan, als technische Formulierung). Für das eingesetzte Polycarbonat ist BPA (Bisphenol A) als Schadstoff zu definieren, welcher unter umweltrelevanten Bedingungen freigesetzt wird. Im Rahmen der durchzuführenden Arbeiten sollen die aufgeführten prioritären Schadstoffe hinsichtlich möglicher Transformationsprozesse in Umwelt und Klärwerk charakterisiert werden. Die unter den Bedingungen der Ozonung, Chlorierung und UV-Bestrahlung erhaltenen Transformationsprodukte (TPs) sollen im Sinne einer Non-Target-Analytik identifiziert und eindeutig charakterisiert werden. Die Ergebnisse sollen mittels vorhandener Analysenverfahren, wie GC-EI-MS, HPLC-ESI-IT-MSn bzw. LC-MS/MS erzeugt bzw. abgesichert werden. Anhand der in der Struktur eindeutig zu beschreibenden Transformationsprodukte und der Charakterisierung von potentiellen Intermediaten sollen belastbare Reaktionswege und –mechanismen abgeleitet werden.
In addition to previously reported results on the accelerated weathering of polystyrene samples (PS) containing 1 wt.% hexabromocyclododecane (HBCD), we present the first results of our investigations of polypropylene samples (PP) containing 0.1 wt.% BDE-209. All studied polymer samples were exposed to a defined weathering schedule in a climate chamber in accordance to regulation EN ISO 4892-3:2006. For the determination of BDE-209 in the collected raining water the samples were prepared in accordance to a validated protocol. Before the analyses each sample was spiked with isotopically labeled BDE-209. Subsequently the samples were extracted with isooctane. The obtained extracts were concentrated, and the resulting solutions were analyzed by GC/MS. Additionally, the total bromine content was monitored for the weathered and untreated samples using laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) and X-ray fluorescence analysis (XRF) as a non-destructive and rapid method. In general, the resulting data from the accelerated weathering will be compared to those from the natural weathering experiments. Here, the surfaces of the test pieces were analyzed by LA-ICP-MS and XRF as well. Moreover, soil bed tests were conducted in a well characterized model soil. This soil (boulder-clay, sand with 12 % loam, particle size in total 0.2-4 mm) is filled in a free-draining basin inside of an air-conditioned cellar. In this manner, TOC, water capacity and humidity are recorded parameters. To induce a leaching process from the samples by the raining water, the target water content is calculated to 8%. The actual humidity is measured by a tensiometer, assuring the duration of the raining period. A defined humidity is a fundamental parameter for a biological activity in the soil. The test polymers were placed up to the half in the soil. Microbial activity of the soil is monitored by a reference polymer (polyurethane) and should induce the release of HBCD and BDE-209 out of the test materials. These released analytes will be captured by passive samplers (silicone tubes) placed in a distinct distance to the polymer samples in the soil. The soil bed experiments are complementary to the weathering experiments due to the biological activity in the soil.

Warfarin, also known as Coumadin is one of the most popular anticoagulant drugs used as a therapeutic in humans to prevent thrombosis, atrial fibrosis, and fibrillation since the 1950s. Because of its ability to hinder blood from clotting by blocking vitamin K-depending carboxylation of blood clotting precursors, it is also used as a rodenticide worldwide. Until today it has been partially substituted by far more potent anticoagulant rodenticides (ARs), so-called superwarfarins. There are numerous references confirming secondary and tertiary poisoning in non-target-animals and wildlife like mammals, birds, and fish associated with ARs. Up to now relatively little is known about persistence and toxicity of transformation products of those AR in the environment and food chain. Herein, warfarin was chosen as a model compound to determine andelucidate TPs which are generated technically and under environment-like conditions. Such as the simulation of the oxidative phase I metabolism by implementing electrochemical flow cells coupled to mass spectrometry (MS). Obtained results are being compared to TPs generated by UVB-irradiation and ozonation.
Analysis and separation of degradation products and TPs were achieved by a variety of gas- and liquid chromatographic techniques coupled to (high resolution) MS.

Elementanalytische Verfahren werden heute zu mehr eingesetzt als bloß zur Bestimmung von Metallgesamtgehalten in diversen Probenmatrizes. Sie stellen heute ein wichtiges Werkzeug zur Beantwortung lebenswissenschaftlicher Fragen aus Umwelt, Medizin und Biologie dar. In der vorliegenden Arbeit kommt die single-cell-ICP-Flugzeitmassenspektrometrie (single-cell-ICP-TOF-MS) zur Multielementanalytik in einzelnen Kieselalgen (Diatomeen) zum Einsatz und wird zukünftig ein wichtiges Werkzeug z.B. bei der Beantwortung
ökotoxikologischer Fragestellung sein.
