In dieser Arbeit wurden drei verschiedene Nanomaterialien auf ihre Bindungsfähigkeit zu Proteinen untersucht. Zu Beginn standen dabei die Herstellung stabiler Dispersionen der einzelnen Nanopartikel und die Stabilität der gebildeten Konjugate im Vordergrund. Der Nachweis einer erfolgreichen Konjugatbildung, sprich der Beschichtung von Nanopartikel mit Proteinen, wurde sowohl qualitativ mittels DLS-Messungen als auch über quantitative Protein-Bestimmungen erbracht. Für die Quantifizierung konnten verschiedene Methoden eingesetzt werden. Neben der klassischen Vorgehensweise, welche indirekt über die Quantifizierung von ungebundenem Protein im Überstand erfolgt, konnten ihm Rahmen dieser Arbeit verschiedene direkte Bestimmungsmethoden entwickelt werden. So wurden mittels kolorimetrischer Tests, wie dem BCA-Assay und dem Bradford-Assay, Nanodiamantdispersionen mit Hilfe einer Korrekturwellenlänge vermessen und quantifiziert. Ebenso zum Einsatz kam die Methode der Aminosäureanalytik, welche aufgrund ihrer guten Rückführbarkeit auf Aminosäurestandards Ergebnisse mit hoher Richtigkeit generieren kann und ebenso die Detektion kleiner Proteinmengen möglich macht.
Nach den erfolgten quantitativen Betrachtungen wurden die Protein-beschichteten Nanopartikel auf ihre Anwendbarkeit als Analoga von Virus-like Particles (VLP) bei einer Immunisierung zur Gewinnung von polyklonalen Antikörpern gegen humanes Ceruloplasmin in Kaninchen überprüft. Es konnte mittels ELISA gezeigt werden, dass die Konjugate erfolgreich für die Herstellung von Antikörpern eingesetzt werden können und im zeitlichen Verlauf einer Immunisierung eine Steigerung des Antikörper-Titers zu erreichen ist.

Hierzu werden chemisch-physikalische Einflüsse (Bewitterung) und mikrobielle Beanspruchungen an Modellmaterialien evaluiert. So werden die Freisetzungsraten von Schadstoffen in Abhängigkeit der Beanspruchung beschrieben. Als Modellmaterialien kommen die Polymere Polystyrol (PS) und Polypropylen (PP) zum Einsatz. Synergistische Effekte der Bewitterungsparameter und der mikrobiologischen Beanspruchung sollen dabei ebenso betrachtet werden, wie die gezielte Alterung. Auch findet eine Beschreibung des Verhaltens der ausgetragenen Schadstoffe in den Umweltkompartimenten Boden oder Wasser statt. Hier sind mit Hilfe der zu entwickelnden Screening- und non-Target-Analyseverfahren die Transformation und der Metabolismus durch Mikroorganismen zu beschreiben. Aus den Ergebnissen sollen Korrelationen zwischen den künstlichen Alterungsverfahren und realen Szenarien abgeleitet werden.
In case study one of the CONSENS project, two aromatic substances were coupled by a lithiation reaction, which is a prominent example in pharmaceutical industry. The two aromatic reactants (Aniline and o-FNB) were mixed with Lithium-base (LiHMDS) in a continuous modular plant to produce the desired product (Li-NDPA) and a salt (LiF). The salt precipitates which leads to the formation of particles. The feed streams were subject to variation to drive the plant to its optimum.
The uploaded data comprises the results from four days during continuous plant operation time. Each day is denoted from day 1-4 and represents the dates 2017-09-26, 2017-09-28, 2017-10-10, 2017-10-17.

The transition from X-ray film to digital detectors in radiography is accompanied by an increase of unsharpness due to the larger inherent digital detector unsharpness in comparison to film. The basic spatial resolution of digital detectors (see EN ISO 17636-2) is used today to describe this unsharpness. The geometrical unsharpness of the radiographic projection of object structures onto the detector plane is determined by the focal spot size of the X-ray tube and the magnification. The focal spot size is measured today (see ASTM E 1165) from pin hole camera exposures or edge unsharpness (see ASTM E 2903). The final image unsharpness is a result of a convolution of the geometrical and inherent detector unsharpness function, divided by the magnification factor of the object onto the detector plane. Different approximations of this convolution result in ASTM E 1000 and ISO 17636-2 in different optimum values for the magnification factor for a given focal spot size of a X—ray tube and the basic spatial resolution of the detector. The higher contrast sensitivity, an advantage of digital radiography, compared to film radiography is furthermore improved when using higher X-ray voltages as used with film and smaller focal spots of the X-ray tubes. This allows a higher distance between object and detector resulting in reduced object scatter in the image. The interactions between all these parameters will be discussed and simple rules for practitioners will be derived in this contribution.
What is the future of Analytical Sciences? The talk starts with a definition, comparing the current view with that from 1968. How do wie set trends? How do we get Analytics inside? Some examples of "Big Science" are given and discussed in relation to a definition of AS. How does AS face the current Grand Challenges?
As exaples for something significant, several exaples are presented, such as Climate Change of Hydrogen Storage. Another important trand are eScience and automation concepts for AS, which are highlighted.
But (Analytical) Science has to be politcal in our times to face Fake News and to breake barriers!
Nowadays, different nanoparticles have been developed for commercial applications. However, since data on toxicity are barely available, their increasing application in cosmetic products, food and their release in the environment might cause severe problems. An accurate separation, identification and characterization of nanoparticles becomes increasingly important1,2.
In this presentation, a two-dimensional separation approach based on AF4 and CE was showed and used to separate NPs with similar sizes but different coatings. Standard reference polystyrene NPs having comparable core sizes but different coatings were investigated. Different migration time and profiles were compared. Separation in either method resulted in non-baseline resolved or non-separated peaks. In contrast, two-dimensional coupling of AF4 and CE resulted in clearly separated regions in their 2 D plots in case of 20 and 50 nm particle mixtures, whereas the 100 nm NP mixture could not be separated. Various factors affecting the separation like hydrodynamic diameter or SDS concentration were discussed.
Future investigations will be focussed on inorganic NPs with differently charged coatings. There, AF4-CE coupling can be coupled with inductively coupled plasma mass spectrometry (ICP-MS) to enhance the sensitivity of this method.

The accumulation of plastics leaked into the environment achieve a high perception in the last years. It is uncontroversial that often an uncontrolled, high consumption of plastic products and a poor waste management resulted in plastic findings all over in the environment. However, less is known about the relevant pathways and fate of plastic in the environment. Especially plastic fragments smaller 5 mmm, so called microplastics, achieve a high attention, because their amount and risks arising from these fragments are still unknown. The basic mechanisms of plastic or more precise, of different polymer degradation processes are often not considered, resulting in unrealistic statements.
For generation of fundamental understanding of this problem we developed in the last years a fast thermoanalytical method for identification and mass quantification of microplastics in complex samples. The presentation will also give a short introduction about polymer degradation.
The presented development of a damping model is a research component of an experimental feasibility study about moisture in building materials measured with Bluetooth® Low Energy (BLE) signals. This study may be part of a structural health- and long-term monitoring aiming at early damage detection in the built infrastructure and is increasingly focusing on wireless sensor network technology. It is investigated, how the Received Signal Strength Indicator (RSSI) of a BLE signal, transmitted from the BLE-module embedded in building materials with changing moisture content is damped. The aim of the modelling is the derivation of a damping equation for the formal model to determine the moisture damping exponent to finding a correlation.

This presentation gives a short overview of isotope ratio measurements being carried out in the past few years at BAM in different fields such as plant metabolism, food web structures and archaeology. The corresponding isotope reference materials which have been certified at BAM in parallel are presented as well. Additionally an outlook is being provided on future iRM projects.
Der Vortrag gibt eine Übersicht über aktuelle qNMR-Aktivitäten im Fachbereich 1.4. Dies umfasst beispielsweise die Online-Anwendung zur Prozessüberwachung, aber auch die Reinheitsuntersuchung von Reinstoffkomponenten für die Herstellung von Referenzgasgemischen. Zusätzlich wird ein Ausblick in die aktuelle Strategie hinsichtlich der Rückführung von qNMR-Ergebnissen mittels primärer Referenzmaterialien gegeben.
Type-I pyrethroids are frequently used for disinfection purposes against insects such as adult mosquitoes, or diseases carried by insects, like Malaria or Zika in cabins of airplanes on long-distance flights especially from tropical destinations. This treatment is mandatory at various airports but compliance with the rules is difficult to test for. Moreover, if improperly used, these compounds can entail negative health effects for crews and passengers.
The detection of the pyrethroids will be achieved thanks to an antibody-gated indicator delivery system (gAID) utilizing monoclonal antibodies and hybrid sensory nanoparticles. After the interaction of the pyrethroid with the gAID, the liberated indicator (dye) will be detected. Since only few analyte molecules are necessary for pore opening yet release a large number of dyes, the system shows intrinsic signal amplification.
The device system to be developed has to be so simple that chemically untrained personnel, such as ground or cabin crew, can use it and obtain a result in a reasonably short period of time, e.g., ≤5 min. The need for high accuracy and sufficient sensitivity, established at 0.001 g m–2, is a critical requirement and imposes another significant challenge since this value is beyond current LFTs reported in the literature for pesticide detection to date.
In order to achieve the selectivity and sensitivity required by the test itself, and to avoid cross reactivity with other type I pyrethroids, the production of a monoclonal antibody for both Permethrin and Phenontrin is necessary. The synthesis of the two hapten molecules and the subsequent immunization with different immunogens represent the first goal of the work.
For years there have been more and more reports on the presence of drugs in the aquatic environment. Due to the demographic change, the consumption of pharmaceuticals has risen sharply. After taking the drugs, they are partly metabolized in the human body. However, the metabolism is not complete so that both the metabolites and non-metabolized amounts of the parent compounds are excreted. These compounds reach the waste water and afterwards the sewage treatment plants. In sewage treatment plants transformation products can be formed by the oxidative conditions during wastewater treatment processes. The transformation products may have a higher toxicity than the actual environmental pollutants and are often only partly removed during the waste water treatment. Since a lot of these compounds are still unknown, the transformation products are not detected by target analysis used in sewage treatment plants and are often released undetected in the aquatic ecosystems. The released substances may be subject to additional transformation processes in the environment. Pharmaceuticals produced in high amounts can be already detected in the μg/L range in water bodies worldwide.
Metformin and its major transformation product guanylurea are one of the main representatives. Metformin is the drug of choice for treating type 2 diabetes. The drug therapy for diabetes mellitus has increased significantly in recent years. In the year 2015 1500 tons of metformin were prescribed in Germany (for statutory insured persons). Metformin is not metabolized in the human body and is excreted unchanged therefore concentrations between 57 μg/L and 129 μg/L are found in German waste water treatment plants influents.
In this work the transformation of the antidiabetic drug metformin is investigated. The degradation of metformin is initialize by commercial water treatment techniques like UV-radiation or noncommercial techniques like heterogenous photocatalysis based on titanium dioxide. The degradation of metformin and resulting transformation products are analyzed by LC-MS/MS and LC-HRMS.
In addition to previously reported results on the simulated aging of polystyrene samples (PS) containing 1 wt. % hexabromocyclododecane (HBCD), we present the first results of our investigations of polypropylene (PP)-samples containing 0.1 wt. % BDE-209. All studied polymer samples were exposed to a defined weathering schedule in a climate chamber in accordance to regulation EN ISO 4892-3:2006.For the determination of BDE-209 in the collected rain water samples derived from the used climate chamber, the samples were prepared in accordance with a validated protocol. Before the analyses, each sample was spiked with 2 µL of isotopically labeled BDE-209 (13C10-BDE-209) to serve as internal standard (ISTD) in the performed stable isotope dilution analysis. Subsequently the samples were extracted with isooctane, the obtained aliquots of the extracts were concentrated to 200 µL and 2 µL of the resulting solution were injected to the GC/MS for quantification.
Additionally, the total bromine contents are monitored for the aged and untreated samples using laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) as well as X-ray fluorescence analysis (XRF) as non-destructive and rapid method. Furthermore, results from surface analysis using environmental scanning electron microscopy (ESEM) for morphological characterization of the aged and untreated samples were presented and discussed. In general, the resulting data from the accelerated aging will be compared to those from the natural weathering experiments (“atmospheric exposure”, in soil).
The atmospheric exposure was performed by placing the samples on a weathering rack, which is aligned in SW direction (in a 45° angle to the horizon). The weathering data were regularly recorded by Deutscher Wetterdienst at this site. The surfaces of the test specimens (aged and stored references) were analyzed by ESEM as well as by LA-ICP-MS and by XRF. The surface of PS and PP specimens aged outdoors present the aging under real conditions and allow the comparison to the accelerated aged specimens by means of the weathering chamber. This way, we explore the efficiency of the accelerated aging procedure, which provides the advantage of well-defined and reproducible conditions compared to natural weathering, as a tool for testing different plastic materials.
Additionally “in soil” experiments were conducted in-door in a well characterized testing soil. The soil (boulder-clay, sand with 12 % loam, particle size in total 0.2-4 mm) is filled in a free-draining concrete basin inside of an air-conditioned room. In this manner, TOC, water capacity and humidity are recorded parameters. To assure a washing out process from the samples by the raining water, the target water content is calculated to 8%. The actual humidity is measured by a tensiometer, assuring the duration of the raining period. The water content is additionally monitored by weight of the basin, capturing water from raining periods. The correct humidity is a fundamental parameter for biological activity. Samples of PS resp. PP were of dimension 10x1cm and 5 specimens were placed up to the half in the soil per basin. Microbial activity of the soil, monitored by the reference polyurethane, sets HBCD resp. BDE-209 of the samples free and will be leached from the samples by raining water. Thereafter these will be captured by passive samplers placed in a distinct distance to the samples in the soil. The “in soil” experiments are complementary to the weathering experiments due to the biological activity in the soil. These experiments simulate the fate of the brominated flame retardants in the biosphere.
Environmental sustainability and –stability of Materials concerning the Migration of pollutants

MaUS is an acronym for ”Material und Umweltsimulationen“. Plastics are in the focus of environmental politics due to their long-term behaviour and therefore to their persistence. Not only that they appear as visible contaminants in the sea and on the beach, but their unknown behaviour concerning their additives as well as the related transformation products are anxious. Therefore, we wish to establish a certified reference method to provide a method for testing plastics.
Aim of this project is the development of fast motion standard reference methods for testing plastics regarding to their environmental compatibility. To establish these testing methods, we use polystyrene (PS) and polypropylene (PP) with environmental relevant brominated flame retardants, known for their persistent bioaccumulative and toxic (PBT) properties. In case of PS the material contains 1 wt% of 1,2,5,6,9,10-hexabromocyclododecan (HBCD) and in case of PP 0.1 wt% bromodiphenylether (BDE-209), which is known as a substance of very high concern (SVHC). Furthermore, we use polycarbonate (PC), which is still used as material in baby flasks and releases Bisphenol A (BPA), an estrogenic active substance.
As an additional material PTFE is used for its importance as a source for two ubiquitous environmental substances (PFOS and PFOA), whose toxicological effects are still incompletely known.
The focus in this current work is set on the transfer of potential pollutants out of applied materials mentioned above into environmental compartments like water or soil. Here an accelerated aging concept should be developed to shortened time consuming natural processes. For these resulting simulations we use a programmable weathering chamber with dry and wet periods and with high and low temperatures. These programmes run for several weeks and according to a defined sampling schedule we take water samples, run a clean-up procedure by SPE (Molecular imprinted polymers (MiPs) resp. polymer-based cartridges (Waters Oasis HLB)) and analyse them by HPLC-UV resp. LC-MS/MS. Of most interest in case of flame retardants are photocatalytic transformation products. Therefore, we conduct a non-target-screening resp. a suspected target-screening by LC-MS/MS and HRMS.

Aus den Ergebnissen sollen Korrelationen zwischen den künstlichen Alterungsverfahren und realen Szenarien abgeleitet werden, die es gestatten, Schnellprüfverfahren zu etablieren, die die Simulation der realen Beanspruchungen im Zeitraffermodell anwendbar machen. Somit sollen standardisierbare Schnellbeanspruchungs-verfahren erarbeitet werden, die als Prüfeinrichtungen etabliert werden und von externen Auftraggebern zur Prüfung der Umweltbeständigkeit und -verträglichkeit von neuen Materialien genutzt werden können. Die Umweltwirkungen (chemisch-physikalisch und mikrobiologisch) sollen so definiert eingesetzt werden, dass eine reproduzierbare Prüfung möglich wird. Aus diesen Verfahren und Methoden sollen Normen abgeleitet werden, die eine standardisierte Materialprüfung ermöglichen.

CRM (Certified Reference Material) BAM-U116/CGL306 “Cyanide in soil” was produced within a framework of cooperation between CGL (Central Geological Laboratory) of Mongolia and Federal Institute for Materials Research and Testing (BAM) of Germany in 2013-2017. The CRM BAM-U116/CGL306 represents a mixture of a sandy soil collected from a contaminated former gasworks area in the Berlin region (Germany) and an unpolluted sandy soil from Nalaikh region (Mongolia).
The bulk candidate material for this reference material was prepared at CGL CRM Laboratory exclusively destined to the preparation of reference materials and equipped with modern technical equipment. Homogeneity, stability and shelf life were studied in full compliance with ISO Guide 35. The CRM was evaluated as sufficiently homogeneous. Statistical evaluation of certification analysis
was software packages SoftCRM and ProLab Plus. Certified value of total cyanide of the CRM is 12.0 mg/kg and expanded uncertainty was assigned as 0.8 mg/kg. The intended purpose of this material is the verification of analytical results obtained for the mass fraction of total cyanide in soils and soil-like materials applying the standardized procedure ISO 11262:2011. As any reference material, it can also be used for routine performance checks (quality control charts) or validation studies.
The analysis of non-metals normally is carried out using elemental analysers which require reference material for calibration. In the lecture the CRM-program of BAM suitable for non-metal-analysis is presented. There are CRMs available with non-metal contents in the low ppm up to the high percent region.
Surface functionalization of nanomaterials is nowadays at the core of many applications of functional materials in the life and material sciences. Examples range from membranes and microarrays over bead-based assays, and next generation sequencing to nanometer-sized optical reporters, nanosensors, and magnetic and optical contrast agents. Typical function-nalization steps include silanization and grafting reactions with reactive monomers to introduce functional groups like amino or carboxylic acid groups or the attachment of ligands like polyethylene glycol (PEG) molecules and biomolecules. [1-3] This enables to tune e.g., dis-persibility, hydrophilicity and biocompatibility, minimize unspecific interactions, improve biofunctionalization efficiencies, and enhance blood circulation times and allows for the use of nanomaterials as reporters in assays or the design of targeted probes for bioimaging.
At the core of all functionalization strategies are reliable and validated methods for surface group and ligand quantification that can be preferably performed with routine laboratory instrumentation, require only small amounts of substances, and are suitable for many different types of nanomaterials. [3] There is meanwhile a considerable need to make these methods traceable. We present here versatile and simple concepts for the quantification of common functional groups, ligands, and biomolecules on different types of organic and inorganic nanomaterials, using conventional and newly developed cleavable and multimodal reporters, that can be detected with optical spectroscopy. [4-7] These reporters are chosen to enable method validation with the aid of method comparisons and mass balances. Also, strategies how to make these simple assays traceable to SI units using quantitative nuclear resonance spectroscopy (qNMR) and X-ray photoelectron spectroscopy (XPS) are derived.
Analytical Sciences has developed from Ostwald’s “unentbehrlichen Dienstmagd” to a chemical discipline at the core of many of today’s fundamental and applied scientific problems and innovations. An atomic or molecular understanding of basic processes in chemistry, soft matter physics, materials and life science is enabled only through new analytical methods and instrumentation. Similar observations can be found for pressing sociopolitical conflicts of the future: A rational discussion of global climate change or new energy sources is only possible with reliable analytical results. Progress in Analytical Sciences is only possible if the underlying interdisciplinary character is acknowledged and valued. The talk will illustrate the scope of modern Analytical Science through examples from process analysis relevant to modern process intensification and industry 4.0 to bioanalysis and the use of synchrotron radiation to elucidate fundamental reactions materials.
Two calibration-free (CF) LIBS approaches are used for the quantitative analysis of cement samples: the CF-LIBS based on the Boltzmann plot method and the Monte Carlo (MC) LIBS based on the iterative spectrum fitting. In CF-LIBS, the inverse problem is solved, i.e. the elemental concentrations are determined by the reconstruction of plasma parameters from spectra. The MC-LIBS technique solves the direct problem by finding the highest correlation between the model-generated and experimental spectrum. The accuracy of both calibration-free LIBS methods suffers from factors such as inaccurately determined instrumental function, the deviation of experimental plasma from the mathematical model used, not taking into account the collection geometry, and from the uncertainty of spectroscopic data. The both calibration-free LIBS approaches are first applied to synthetic spectra which perfectly suit the mathematical model of the method, i.e. the model of the uniform, isothermal, and stationary plasma. This test yields the accuracy of both the approaches for the ideal case. In addition, the accuracy of both the methods is investigated for non-uniform and non-isothermal plasma, because real laser-induced plasma often has high gradients in temperature and particle number densities. Finally, both calibration-free LIBS approaches are applied to experimental spectra obtained from cement samples. The figures of merits of two approaches are compared when working with both synthetic and experimental spectra.
An improved algorithm for calibration-free laser induced breakdown spectroscopy (CF LIBS) will be presented which includes several novel features in comparison with previously proposed similar algorithms. In particular, it allows using spectral lines with arbitrary optical thickness for the construction of Saha-Boltzmann plots, retrieves the absorption path length (plasma diameter) directly from a spectrum, replaces the Lorentzian line profile function by the Voigt function, and allows for self-absorption correction using pre-calculated and tabulated data rather than approximating functions. The tabulated data embody the solutions of the radiative transfer equation for numerous combinations of optical thicknesses and line widths. The algorithm is thoroughly verified using synthetic spectra.
Cancer cell lipid class homeostasis is altered under nutrient-deprivation but stable under hypoxia

Under oxygen/nutrient deprivation cancer cells modify the Balance between fatty acid (FA) synthesis and uptake, which alters the levels of individual triglyceride or phospholipid sub-species. These modifications may affect survival and drug-uptake in cancer cells. Here, we aimed to attain a more holistic overview of the lipidomic profiles of cancer cells under stress and assess the changes in Major lipid-classes. First, expressions of markers of FA synthesis/uptake in cancer cells were assessed and found to be differentially regulated under metabolic stress. Next, we performed a broad lipidomics assay, comprising 244 lipids from six major classes, which allowed us to investigate robust stress induced changes in median levels of different lipid classes -additionally stratified by fatty acid side chain saturation status. The lipidomic profiles of cancer cells were predominantly affected by nutrient-deprivation. Neutral lipid compositions were markedly modified under serum-deprivation and, strikingly, the cellular level of triglyceride subspecies decreased with increasing number of double bonds in their fatty acyl chains. In contrast, cancer cells maintained lipid class homeostasis under hypoxic stress. We conclude that although the levels of individual lipid moieties alter under hypoxia, the robust averages of broader lipid class remain unchanged.
MYCN is a transcription factor that is aberrantly expressed in many tumor types and is often correlated with poor patient prognosis. Recently, several lines of evidence pointed to the fact that oncogenic activation of MYC family proteins is concomitant with reprogramming of tumor cells to cope with an enhanced need for metabolites during cell growth. These adaptions are driven by the ability of MYC proteins to act as transcriptional amplifiers in a tissue-of-origin specific manner. Here, we describe the effects of MYCN overexpression on metabolic reprogramming in neuroblastoma cells. Ectopic expression of MYCN induced a glycolytic switch that was concomitant with enhanced sensitivity towards 2-deoxyglucose, an inhibitor of glycolysis. Moreover, global metabolic profiling revealed extensive alterations in the cellular metabolome resulting from overexpression of MYCN. Limited supply with either of the two main carbon sources, glucose or glutamine, resulted in distinct shifts in steady-state metabolite levels and significant changes in glutathione metabolism. Interestingly, interference with glutamine-glutamate conversion preferentially blocked proliferation of MYCN overexpressing cells, when glutamine levels were reduced. Thus, our study uncovered MYCN induction and nutrient levels as important metabolic master switches in neuroblastoma cells and identified critical nodes that restrict tumor cell proliferation.
Mechanochemistry is a fast and efficient method applicable for the synthesis of new organic, metal-organic, and inorganic compounds. The direct monitoring of milling reactions is still challenging. The underlying reaction mechanisms often remain unclear. In the last years, we have established a tandem in situ approach for investigating mechanochemical reactions using time-resolved in situ XRD coupled with Raman spectroscopy. A further development is the in situ coupling of synchrotron XRD, Raman spectroscopy, and thermography allowing the observation of mechanochemical reactions in real time. [5] Information on the crystalline, molecular, and temperature state of the materials during grinding could be collected. The chemical composition of the reaction mixture was found to be directly correlated with changes in the temperature profile of the reaction. Furthermore, the presented setup allows the detection of crystalline, amorphous, eutectic as well as liquid intermediates. Based on these data, metastable polymorphs of cocrystals and coordination polymorphs could be isolated and struc-turally characterized. A multi-step diffusion mechanism was identified for most systems. Crystalline phases were obtained intermediately, suggesting that the synthesis is following Ostwald’s rules of stages. The resulting deeper kinetic and thermodynamic understanding of milling processes is the key for future optimization of mechanochemical syntheses.

Neurodegenerative diseases are one of the major problems for our ageing society. Alzheimer’s disease (AD) as the most common neurodegenerative disorder affects over 46.8 million people worldwide and the number will increase as the population ages. The diagnosis of AD is challenging and only half of the patients are identified yet and often only in late stages. One reason is that existing assays for identification and quantification of AD biomarkers lack accuracy and are poorly comparable.
This study is part of the EU project “ReMiND” aiming to develop accurate, reliable and traceable methods for the detection and quantification of known and suspected AD biomarkers. Our target is the tau protein, as brain load and distribution of tau is highly correlated with the clinical progression of AD. We intend to develop a measurement method for the accurate quantification of tau by means of inductively coupled plasma mass spectrometry (ICP-MS).
ICP-MS is a powerful method for the matrix independent quantitative analysis of target elements. Developed for the use in inorganic trace analysis, ICP-MS is nowadays emerging as a valuable tool for bioanalytical questions. Especially the use of ICP-MS for quantitative proteomics by measuring heteroatoms is highly promising, considering that established quantification methods like organic mass spectrometry depend on the existence of matched protein and peptide standards or labelling of the target protein. In this work, we applied isotope dilution analysis (IDA) using ICP-MS to quantify proteins of known stoichiometry via their sulphur content. Sulphur is present in two amino acids, cysteine and methionine, and hence is omnipresent in nearly all proteins. A NIST standard bovine serum albumin (BSA) was quantified using sulfur IDA to optimize sample preparation and method parameters. Our goal is to employ the developed method in a proof of concept study for the quantification of the AD biomarker tau extracted from brains of a mouse model for AD.
Alternariol (AOH) and Alternariol monomethylether (AME) are two secondary metabolites of Alternaria fungi which can be found in various foodstuffs like tomatoes, nuts and grains. Due to their toxicity and potential mutagenic activity, the need for the development of high-throughput methods for the supervision of AOH- and AME-levels is of increasing interest. As the availability of both native and labelled AOH and AME analytical standards is very limited we herein wish to present a novel concise approach towards their synthesis employing a ruthenium-catalyzed ortho-arylation4 as the key step.
LA-ICP-MS is increasingly used for single cell analysis in two different detection modes using either the imaging mode with subcellular resolution or alternatively single spot analysis of cells with a larger laser spot size. This study compares the analytical figures of merit of both detection modes (signal to noise, precision, accuracy, throughput), as well as ease of operation and data evaluation. Adherent 3T3 fibroblast cells were stained with two metal dyes (mDOTA-Ho, Ir-DNA-intercalator) and several dozen cells were measured using both modes. We found a ten times higher throughput for single spot analysis, which has as well a straightforward data analysis, shortening the total analysis time further. The signal to noise ratio for single spot analysis was found to be slightly better compared to the signal to noise of pixels in imaging. The mean metal intensity per single cell differed by only 10% between both modes and obtained distributions were found to show no statistically significant differences. Using matrix matched calibration based on standards spotted onto nitrocellulose membrane, we achieved detection limits (10s) of 12 fg for Ir and 30 fg for Ho and quantified 57 +/-35 fg Ir and 1192 +/- 707 fg Ho per single cell.
Compared to a conventional ICP-MS measurement of a digest of about 60000 cells, 54% of Ir content and 358% Ho content was found using quantitative LA-ICP-MS. The difference might be a consequence of the two metal dyes binding to different structures of the cell and therefore might behave differently in sample preparation for conventional and LA-ICP-MS.
Gold ist eines der sieben schon im Altertum bekannten Metalle und wurde wg. seines Glanzes und seiner Seltenheit von alters her als Tauschmittel und zur Herstellung von Schmuck benutzt. Außerdem ist es einfach bearbeitbar und weitestgehend gegen chemische Einflüsse resistent. Die Untersuchungen von Gold mit synchrotronstrahlungsangeregter Röntgenfluoreszenzanalyse sind zerstörungsfrei und geben Auskunft über die in der untersuchten Probe vorhandenen chemischen Elemente. Bei den hier vorgestellten Untersuchungen an der BAMline stehen Fragestellungen wie Herkunft, Herstellungsverfahren und Zusammengehörigkeit von Goldfunden im Vordergrund. Die verschiedenen Fragestellungen werden an einer Reihe von Beispielen erläutert die vom Wikingerschatz aus Hiddensee über die Himmelsscheibe von Nebra bis hin zu Funden aus Ägypten langen. Zusätzlich werden die modernen Messmethoden vorgestellt, die am Synchrotron heutzutage zur Verfügung stehen.
Many current nanomaterials can serve as contrast agents in cellular or tissue imaging, drug delivery vehicles or therapeutics, whereas others can cause toxic effects. In order to evaluate nano-bio interactions, the number of nanoparticles (NPs) inside cells as well as their localisation within cellular substructures is of particular interest. The cellular uptake depends on the primary characteristics of the NPs (e.g. size, shape, surface coating) and on the cell type.
Laser ablation inductively coupled plasma mass spectrometry (LA‑ICP‑MS) is more and more used to study the NP pathway from uptake, via intracellular processing up to cell division. High-spatial resolution laser ablation at single cell level is achieved using novel low-dispersion LA chambers and by careful optimisation of laser energy, ablation frequency and scan speed at small laser spot sizes down to 1 µm. Different examples from BAM, Division 1.1 and cooperation partners using LA-ICP-MS to localize and quantify metal-containing nanoparticles are shown. The results demonstrate the potential of LA-ICP-MS providing insight into NP uptake, intracellular distribution and cell-to-cell variation.
qNMR provides the most universally applicable form of direct concentration or purity determination without need for reference materials of impurities or the calculation of response factors but only exhibiting suitable NMR properties. The workshop presents basic terms of statistics and uncertainty analysis, which are the basis for qNMR spectroscopy and data analysis such as, e.g., standard deviations, linear regression, significance tests, etc. and gives typical examples of applications in qNMR spectroscopy.
The Color X-ray Camera CXC or SLcam® is an energy-resolving X-ray camera capable of energy- and space-resolved measurements. It consists of a high-speed CCD detector coupled to a polycapil-lary optic that conducts the X-ray photons from the probe to distinct pixels onto the detector. The camera is capable of fast acquisition of spatially and energy resolved fluorescence images. A dedicated software enables the acquisition and the online processing of the spectral data for all 69696 pixels, leading to a real-time visualization of the elements distribution in a sample. It was developed in a joint project with BAM, IFG Berlin and PN Sensors. In this contribution we will mainly discuss the use of the CXC at our beamline, the BAMline at BESSY II and imaging applications of the CXC from different areas, like biology and archaeometry. Additionally new developments for the use of the detector without optics, like wavelength dispersive detection or 1shot-XANES, will be presented.
X-ray absorption fine structure spectroscopy (XAFS) is a frequently employed technique in order to investigate structural composition and change of chemical compounds such as catalytic species. These structural properties are essential (i) to understand underlying reaction mechanism and (ii) to further improve the design of efficient catalysts.
This investigation is based on a newly developed XAFS setup comprising both time- and lateral-resolved XAFS information simultaneously in a single-shot (S2XAFS). The primary broadband beam is generated by a filter/X-ray-mirror combination (bandpass). The transmitted beam through the sample is diffracted by a convexly bent Si (111) crystal, producing a divergent beam. This, in turn, is collected by an area sensitive detector with a theta to 2 theta geometry. This facile, stable and scanningless setup was tested at the BAMline @ BESSY-II (Berlin, Germany).
This contribution focuses on further experimental optimizations allowing the characterization of supported vanadium oxide (VOx) based catalysts at the lower hard X-ray regime (5 to 6 keV). First S2XAFS measurements of these catalysts are presented herein. Supported VOx catalysts show promising results in the oxidation of methane to formaldehyde.
S2XAFS allows determining the structural composition of the metal (i.e. vanadium) based on a fast and smart setup. It is therefore an ideal tool to identify crucial roles of chemical compounds in catalytic reactions.

X-ray Absorption fine structure spectroscopy (XAFS) is a frequently employed technique in order to investigate structural composition and Change of chemical compounds such as catalytic species or corrosion processes. These structural properties are essential (i) to understand underlying reaction mechanism and (ii) to further improve the design of materials.
While XAFS measurements are usually performed with ionization Chambers or simple fluorescence detectors, we at BAMline specialize in measurements with innovative set-ups that meet Specialrequirements such as time resolution, (3D-) spatial Resolution or demanding sample environments.
This contribution presents various available XAFS configurations with their corresponding applications. In particular, these comprise single -shot XAFS for time- resolved measurements, grazing-exit XAFS with energy and a spatially resolved detector for the characterization of thin films and an in situ grazing incidence Setup for the characterization of corrosion layers.
Additionally,the possibility of analyzing Minute samples in total-reflection geometry is demonstrated.
Certification of the mass fractions of trace elements in a medium purity graphite reference material

The increasing application of graphite materials in various fields of technology and science enhances the demand for strictly controlled properties which are often directly correlated to the contents of trace element impurities. Therefore, the availability of powerful, rapid and reliable analytical methods for the determination of trace impurities is essential for process and quality control. Certified reference materials, indispensable for the development and validation of appropriate trace analytical methods for the characterization of special purity graphite materials are still lacking. Therefore, BAM in co-operation with the working group “Special Materials” of the committee of chemists of GDMB (Gesellschaft der Metallurgen und Bergleute e.V.) certified an industrially sourced graphite material with genuine elemental impurities.
The candidate material was a commercial product (NBG 18) taken from the customary production line of the producer SGL CARBON, Chedde (France). The homogeneity of the powdered material was assessed by means of ETV-ICP OES and DC Arc-OES. Certification of the candidate reference material was based on an inter-laboratory comparison involving 17 expert laboratories from Germany, France, Slovenia, USA, South Africa. A variety of different mineralization, digestion, leaching, fusion and combustion techniques prior to ICP OES and photometry, methods without sample preparation (INAA, k0-INAA) as well as typical solid sampling methods (ETV-ICP OES, ETV-ICP-MS, SS-ET AAS, DC Arc-OES, MF-DC Arc-OES) were used to characterise the material. Certified mass fractions and their expanded uncertainties (k = 2) were assigned for 27 elements (Al, B, Ba, Be, Ca, Co, Cr, Cu, Fe, K, Li, Mg, Mn, Mo, Na, Ni, P, Pb, S, Si, Sr, Ti, V, W, Y, Zn, Zr), ranging from (0.00050 ± 0.00027) mg/kg for Be to (41 ± 6) mg/kg for Si. Informative values are given for further parameters: trace element contents of Ag, As, Au, Bi, Cd, Cs, Dy, Eu, Ga, Gd, Hg, In, La, Nb, Rb, Re, Rh, Sb, Sc, Se, Sm, Sn, Ta, Tb, Te, Th, U; method specific values obtained by ETV-ICP OES (BETV and SiETV) and ash content.
The new certified material is available as BAM Reference Material BAM-S009 Medium Purity Graphite Powder.
To establish the SI traceability of the element content w(Hg) of a gravimetrically prepared 1 g/kg mercury solution, the latter was compared to a primary reference solution by means of high accuracy inductively coupled plasma optical emission spectrometry (ICP OES) measurements. For this purpose, the ICP OES bracketing method previously described [1] was improved to yield a relative expanded uncertainty of the mercury element content of Urel(w(Hg)) = 0.5 % (with k = 2) associated with the comparison of two monoelemental solutions. In case of mercury, such a small uncertainty was achieved for the first time. Before, in general, such small uncertainties were only reported applying isotope dilution methods performed with MC-ICP-MS. However, for mercury solutions at this concentration range, several dilution steps are necessary to prepare the samples for ID-MC-ICP-MS. The sum of the uncertainty contributions stemming from the dilution steps, as well as the intrinsic difficulties of measuring mercury at low concentrations, result in an overall uncertainty of the ICP-MS measurements, which is comparable to or even larger than those, achieved with the ICP OES method applied. We will present details of the sample preparation as well as of the dedicated ICP OES measurement approach, which were crucial to achieve such a small measurement uncertainty.
The newly developed method was successfully applied in the context of the development and production of elemental solution chemical reference standards (CRS), which are distributed by the EDQM. The CRS are intended to support measurements required by the European Pharmacopoeia, which has recently incorporated a new international guideline for the control of elemental impurities in medicinal products.
Analytical glow discharges with optical (GD-OES) and mass spectrometric (GD-MS) detection are able to obtain depth resolved information about the light elements hydrogen, carbon, nitrogen and oxygen in solid samples, where most of the other analytical techniques fail. However, the interpretation or even quantification of the measured signals is still very challenging. Problems arise due to physical effects (plasma processes such as the ‘Hydrogen effect’, the Doppler effect, self absorption or diffusion of hydrogen in the sample during sputtering) chemical effects(e.g. formation of compounds with argon or the matrix, poisoning of the sample or gettering) as well as instrumental difficulties (e.g. of the sensitivity, calibration and vacuum quality)..
The GD techniques are direct solid sampling methods and require reference materials for calibration. Unfortunately the list of available certified reference materials (CRM) suited for calibration of light elements in different matrix is relatively short Therefore, sintered materials doped with the analytes H2, O2 and N2 were produced at IFW Dresden and applied as calibration standards for hydrogen, oxygen and nitrogen. Due to the high analyte concentration added, it is very likely that the real concentration agree well with the added amount of light elements in the corresponding phases. The validation of the determination of the light element concentration in the sintered samples was possible in some cases only due to the lack of suitable techniques. This fact proves the need for the development of a reliable quantification of light elements by GDS.
A systematic dependence of the sputtering rate on the composition was found and can be explained by basic principles.
Using mixtures of TiH2 and ZrH2 with Cu the ratio of hydrogen and Cu lines has a good correlation with the corresponding concentration ratio. The hydrogen emission yield however decreases over 0.3 m% hydrogen and finally the hydrogen intensity may even decrease. This behaviour can be explained by a very similar quenching of the hydrogen and copper intensity caused by the hydrogen effect. First experiments with GD-MS show no saturation of the hydrogen ion current and thus confirm the quenching of the emission yield in GD-OES.
Sintered material for the oxygen calibration (Al2O3, CuO, Cu2O and MgO mixed with Cu, Al and Mg) confirmed the blue line shift effect at O I 130.22 nm, first time reported by Köster 2009 [1]. The effect is more pronounced at Mg than in Al and Cu, which due to line interference leads to a matrix dependent EY. This effect is negligible at O I 777.19 nm and the EY is matrix independent. Using GD-MS first promising results for the calibration of oxygen with these sintered samples could be obtained, when the sputtering rate was included in the evaluation.
More recently also sintered material for nitrogen calibration (AlN mixed with Al and Si3N4 mixed with Cu) was produced and points to a matrix independent emission yield of nitrogen
Reference materials are essential, when the accuracy and reliability of measurement results need to be guaranteed in order to generate confidence in the analysis.
These materials are frequently used for determining measurement uncertainty, for validation of methods, suitability testing and quality assurance. Especially direct solid sampling methods require reference materials for calibration.
They guarantee that measurement results can be compared to recognized reference values.
This presentation gives an overview about the use of GDMS in various certification procedures. Because it represents a fast, sensitive, multielement analyses technique without extensive sample preparation it plays a special role for the purity determination of high purity standards. Various calibration strategies and the preparation of traceable matrix matched calibration standards will be discussed.
For the certification of analyte content in matrix materials mainly techniques with solvent sample preparation are used. Here GD-MS is used to identify possible loss or contamination with analytes during the sample preparation step. Typically used acids to dissolve matrices lead to interferences in the ICP- mass spectrometric detection of various analytes and their quantification. Here GD-MS as direct method can also add an important contribution in the certification process.
A candidate material for the use as primary standard for nickel determination was characterized with respect to total purity. For element determination a pure material can serve as primary standard. However, real world materials are never absolutely pure, hence the total purity of such materials need to be determined experimentally. A reasonable target uncertainty for the purity statement is 10-4 relative. Usually, the purer the material, the easier it is to achieve this target uncertainty. There are two basic ways in order to be able to establish a total purity statement. One is to determine the main component of a high purity material by a direct method such as coulometry, gravimetry or titrimetry. However, these methods are not selective enough for one element and therefore require certain efforts to analyse the material with respect to impurities with interfering analytes. Moreover, to reach the defined target uncertainty is not easy or often impossible to achieve. The second approach is to determine the sum of all possible impurities (as mass fraction) and to subtract it from the ideal purity of 100 % (1 kg/kg). In principle all impurities (all elements not being the matrix element), metals and non-metals must be considered. In this work both approaches to determine the total purity of the nickel material were followed and compared.
The primary (solid) standards are usually used to prepare primary calibration solutions to which secondary and lower order calibration solutions are linked.
Laser induced plasma (LIP) is a highly dynamic, short living event which presents significant difficulty for both diagnostics and modeling. The former requires precise spatially- and time-resolved measurements on a micron-nanosecond scale while the latter needs numerous descriptive parameters; many of them can only be obtained from experiment. Diagnostics and modeling should always complement each other for obtaining a truthful picture of LIP.
In this presentation, a newly developed collisional-dominated model will be presented. The model is based on the coupled Navier-Stokes, state, radiative transfer, material transport, and chemical equations. The model incorporates plasma chemistry through the equilibrium approach that relies on atomic and molecular partition functions. Several chemical systems are modeled including Si-C-Cl-N and B-H-Cl systems.
The model is used to study the equilibrium states of the systems as functions of the concentrations of plasma species and plasma temperature. The model also predicts the evolution of number densities of atomic and molecular species in the expanding plasma plume.
This course will provide an introduction to plasma diagnostic techniques. The major focus of the course will be on the discussions of the practical procedures as well as the underlying physical principles for the measurements of plasma fundamental characteristics (e.g., temperatures, thermodynamic properties, and electron number density). Particular emphasis will be placed on inductively coupled plasma–atomic emission spectrometry, but other analytical plasmas will also be used as examples when appropriate. Selected examples on how one can manipulate the operating conditions of the plasma source, based on the results of plasma diagnostic measurements, to improve its performance used for spectrochemical analysis will also be covered. Topics to be covered include thermal equilibrium, line profiles, temperatures, electron densities, excitation processes, microreactions, pump and probe diagnostics, tomography, temporal and spatial resolution. Basics of plasma computer modeling will be presented.
Generell ist es Ziel der Prozessführung, Reaktions- und Trennprozesse wirtschaftlich optimal zu betreiben. Hierzu werden in großen Anlagen modellbasiere Echtzeitoptimierungsverfahren (Real-time Optimization, RTO) eingesetzt. Die Qualität der Ergebnisse der Optimierung hängt dabei entscheidend von der Güte des verwendeten Modells ab. Die Entwicklung hochgenauer Modelle ist allerdings aufwändig, was für kleine Produktionsvolumina ein wesentliches Hindernis zur Nutzung solcher Methoden darstellt. Mit Hilfe der sogenannten Modifier-Adaptation ist eine iterative Betriebspunktoptimierung auch mit ungenauen Modellen möglich, wenn Messungen der für die Bewertung relevanten Größen (beispielsweise Produktkonzentrationen im Ausgangsstrom) verfügbar sein. Für eine Lithiierungsreaktion in einem bei INVITE kontinuierlich betriebenen Reaktor wurde im Rahmen des EU-Projekts CONSENS eine solche iterative Optimierung auf der Grundlage einer Messung der Zusammensetzung des Ausgangsstroms des Reaktors mit einem Online-NMR-Spektrometer realisiert. Das Spektrometer wurde an den Betrieb in einer explosionsgefährdeten Umgebung angepasst. Es konnte gezeigt werden, dass die Messdaten genau genug sind, um darauf eine Optimierung der Produktausbeute aufzubauen. Die iterative Optimierung führt den Prozess ausgehend vom mit dem Modell berechneten Betriebspunkt schrittweise zum wirtschaftlichen Optimum für die reale Anlage. Dabei reagiert die Optimierung auch auf unbekannte Abweichungen im Prozess (wie die Zusammensetzung des Feeds) und fährt iterativ den jeweils optimalen Betriebspunkt an.
Die Arbeiten wurden im Rahmen des EU-SPIRE-Projekts CONSENS, Förderkennzeichen 636942, durchgeführt.
Aufgrund des weltweit steigenden Getreidekonsums ist die Überwachung des Getreidewachstums, der Lagerung und der Verarbeitung von immer größerer Bedeutung. Besonders während der Lagerung bilden Schimmelpilze sogenannte „Hot Spots“, in denen die Gehalte der Schimmelpilzgifte (Mykotoxine) weit über den vorgegebenen Grenzwerten liegen können. Wegen der ungleichen Verteilung der Toxine stellen Probenahme und die invasive, zeitaufwändige Analyse der Mykotoxine eine Herausforderung dar.
Daher werden schnellere und einfachere Methoden benötigt, die sowohl die Probenahme als auch die Analyse beinhalten. Ein möglicher Ansatz ist die nicht-invasive Untersuchung der homogenen Gasphase über der Getreideprobe. Allerdings setzt diese Vorgehensweise die Anwesenheit von MVOC´s (microbial volatile organic compounds) voraus. Vorangegangene Studien zeigten, dass Trichodien ein Vorläufermolekül der Trichothecene – einer Mykotoxingruppe mit über 180 Substanzen – ist. Aufgrund seiner simplen sesquiterpenoiden Struktur war es bereits möglich, Trichodien mit Hilfe einer Headspace GC-MS Methode zu quantifizieren. Daher könnte es als Trichothecen-Markermolekül Anwendung finden.
Allerdings bedarf dieser Sachverhalt weiterführender Untersuchungen. Der Zusammenhang zwischen der Trichodienkonzentration in der Gasphase und dem Mykotoxingehalt in der Probe muss beleuchtet werden, um Aussagen über den Trichothecengehalt in der Probe treffen zu können. Durch die Umsetzung dieser Überlegungen könnte der Nutzen von Trichodien erweitert und die Quantifizierung der Trichothecene stark vereinfacht werden. Daher beinhaltet der erste Schritt dieser laufenden Studie die Etablierung einer Referenzmethode unter Nutzung von Gaschromatographie gekoppelt mit Massenspektrometrie zur Messung von Trichodien als volatilen Mykotoxin-Marker in Getreide. Statische Headspace und SPME (solid phase micorextraction) wurden hierfür genutzt. In einem nächsten Schritt soll diese Referenzmethode zur Validierung einer schnellen vor-Ort Methode zur Bestimmung von Trichodien in Getreide dienen.
Ionophore Antibiotika werden zum Schutz gegen Kokzidiose eingesetzt, hauptsächlich in der Geflügelmast. Rückstände dieser Substanzen und deren Transformationsprodukte (TP) gelangen über den Metabolismus zum einen in das Geflügelfleisch, aber auch durch Ausscheidung in die Umwelt, da Tiermist als Dünger verwendet wird. Ziel dieser Forschungsarbeit ist es, die TP-Bildung von vier verschiedenen Ionophoren Antibiotika (Monensin (MON), Salinomycin (SAL), Maduramicin (MAD) und Lasalocid (LAS)) zu untersuchen. TPs werden durch den Biotransformationsprozess (Metabolismus) gebildet, dieser Prozess kann in zwei Phasen eingeteilt werden. Während in Phase I Oxidations-, Reduktions- oder Hydrolysereaktionen auftreten, ist Phase II von Konjugationsreaktionen geprägt. Durch diesen Prozess werden die Substanzen besser ausscheidbar.
Natürliche Redox-Vorgänge, wie sie bei der Biotransformation (Phase I) auftreten, können mit elektrochemischen (EC) Systemen simuliert werden. In einer EC-Durchflusszelle findet die Reaktion abhängig vom angelegten Potential statt. Im positiven Potentialbereich (0.0 bis 3.0 V; vs. Pd/H2) werden die Analyten oxidiert und somit oxidative TPs generiert. Durch die online-Kopplung mit Flüssigkeitschromatografie (LC) und Massenspektrometrie (MS) wird zunächst eine säulenchromatografische Trennung der generierten TPs erhalten, gefolgt von der massenspektrometischen Detektion. Durch die online-Kopplung von EC-(LC)-MS ergibt sich eine schnelle Analysemöglichkeit von der TP-Erzeugung ausgewählter Substanzen bis hin zur Detektion, wodurch gleichzeitig eine Identifizierung möglich ist.
Die ausgewählten Ionophoren Antibiotika wurden mittels EC-(LC)-MS auf auftretende TPs untersucht. Die ersten Ergebnisse zeigen ein breites Spektrum unterschiedlicher TPs abhängig von gewählten EC-Parametern wie Lösemittel, Modifier und insbesondere vom Arbeitselektrodenmaterial der EC-Durchflusszelle. Unter den erhaltenen TPs sind sowohl bekannte als auch unbekannte TPs vertreten, so dass weitere Untersuchungen zur Strukturaufklärung und vergleichende Tests zu Phase I Metaboliten (z.B. durch Metabolismus-Studien mit Mikrosomen) geplant sind.
Carbamazepine (CBZ) and cetirizine (CTZ) are pharmaceutical drugs detected in the aquatic environment. However, the effects to marine invertebrates derived from the combination of these drugs with decreased pH conditions due to ocean acidification are poorly explored. This study investigated the biochemical parameters related to metabolic capacity and oxidative stress response and the transcription of genes related to the mode of action (MoA) of CBZ (1.0 μg/L) and CTZ (0.6 μg/L) in the clam Ruditapes philippinarum chronically exposed (28 days) to control (7.8) and low (7.5) pH conditions. The biochemical parameters applied were: the activity of electron transport system (indicator of organisms’ metabolic capacity); lipid peroxidation content (indicator of cell damage); total glutathione peroxidase activity (indicator of antioxidant defense). The transcription of genes related with drug neurotransmission and metabolism, cell defense and immunity and biomineralization was studied. No metabolic changes and oxidative stress was induced by the exposure to CBZ or CTZ at each pH level, but the transcription of several genes related with the MoA (neurotransmission, immunity and biomineralization) was altered by low pH, drug exposure and the combination of both stressors. In conclusion, it was shown that the interaction of drug exposure and low pH conditions can change bivalves’ sensitivity to drugs or alter drugs toxicity.

Der Übergang von der aktuellen Automatisierungslandschaft zur nächsten Generation von Automatisierungskonzepten für die Prozessindustrie hat bereits begonnen. Intelligente Funktionen der Sensoren vereinfachen ihre Anwendung und ermöglichen eine Plug-and-Play-Integration, auch wenn sie auf den ersten Blick komplexer erscheinen mögen. Dies ist die Basis für die Digitalisierung der Prozessindustrie und hilft uns, komplexere Prozesse schneller umzusetzen.
Der Vortrag fasst die derzeit diskutierten allgemeinen Anforderungen an „Smarte Feldgeräte“ zusammen und diskutiert dieses am Beispiel eines smarten Online-NMR-Sensors. NMR-Spektroskopie bietet sich durch den Vorteil der direkten Vergleichsmethode (ohne Kalibrierung) für die Prozess-Steuerung an und verringert somit die Rüstzeiten. Zudem basiert der Sensor auf physikalisch motivierten Modellen (Indirect Hard Modeling, IHM), die sich modular kombinieren lassen. Die Methoden wurden anhand eines vorgegebenen pharmazeutischen Reaktionsschrittes im Rahmen des „Horizon 2020“-Projekts CONSENS der Europäischen Union demonstriert und validiert.
Zuletzt werden Anforderungen an die Weiterentwicklung der Datenauswertemethoden diskutiert, um letztlich die semantische Information aus den Messdaten herauszulesen oder das in der Industrie 4.0 geforderte „durchgehende Engineering“ für die Automatisierungskomponenten zu ermöglichen.
Atherosklerotische Läsionen können durch Magnetresonanztomographie (MRT) unter Verwendung von sehr kleinen Eisenoxidnanopartikeln (VSOP) sichtbar gemacht werden. VSOP akkumulieren in atherosklerotischen Plaques und dienen somit als Atherosklerosesonde.
Das Ziel dieses Projektes war die Abbildung der Verteilung von Europium-dotierten VSOP (Eu-VSOP) durch Laserablation ICP-MS in histologischen Dünnschnitten der Aortenwurzelregion des ApoE-Knockout-Mausmodells, das reich an atherosklerotischen Plaques ist. Darüber hinaus wurde untersucht, ob die VSOP-Akkumulation in den Plaques mit anderen Biomarkern der Entzündung wie Makrophagen und verändertem Endothel korreliert. Somit wäre eine Beurteilung, ob es sich hierbei um instabile bzw. vulnerable Plaque-Regionen handelt möglich. Zu diesem Zweck wurden Antikörper mit verschiedenen Lanthaniden markiert und mit der Eu-VSOP-Verteilung unter Verwendung von LA-ICP-MS in einem Multiplex-Messmodus korreliert.
Eine mögliche Korrelation von reaktiven Stickstoffspezies (RNS) mit endogenem Eisen oder Eu-VSOP kann ebenfalls durch LA-ICP-MS nachgewiesen werden. Zu diesem Zweck wurden RNS-spezifische Antikörper auch mit Lanthaniden markiert.
Imaging of Eu doped very small iron oxide nanoparticle in atherosclerotic plaques via LA-ICP-MS

Atherosclerotic lesions can be visualized by magnetic resonance imaging (MRI) using very small iron oxide nanoparticles (VSOP). VSOP accumulate in atherosclerotic plaques and thus serve as an atherosclerosis probe.
The aim of this project was to image the distribution of europium doped VSOP (Eu-VSOP) by laser ablation ICP-MS in histological thin cuts of the aortic root region of ApoE knockout mouse model that is rich in atherosclerotic plaques. In addition, it was investigated whether VSOP accumulation in the plaques correlates with other biomarkers of inflammation for example macrophages and altered endothelium to assess whether it correlates with instability or vulnerability of the plaque regions. For this purpose, antibodies were labeled with various lanthanide elements and correlated with the Eu-VSOP distribution using LA-ICP-MS in a multiplex measurement mode.
A possible correlation of reactive nitrogen species (RNS) with endogenous iron or Eu-VSOP can also be detected by LA-ICP-MS. For this purpose, RNS-specific antibodies were also labeled with lanthanides.
Analysis of single cells via LA-ICP-MS is a technique with great potential, however manual targeting of single cells is laborious and therefore microarraying of cells looks promising. In this work, we investigate the potential of a commercial non-contact piezo dispenser arraying system (S3, Scienion AG, Berlin), equipped with a novel technology for single-cell isolation called CellenONE™ (Cellenion, Lyon). Usually if one aims to create a microarray of single cells via spotting a suitably diluted cell suspension, one will observe a Poisson-distributed cell number per spot. CellenONE™ overcomes this problem by controlling the number of cells optically in the piezo dispense capillary (PDC) via image recognition to obtain true single cell arrays. The figures of merit of the customized and optimized setup will be presented. In a proof of concept experiment we investigated the trace elemental fingerprint of THP-1 cells by LA-ICP-TOF-MS (Analyte G2, Teledyne Cetac; icpTOF, TOFWERK) and quantified two metal cell dyes, mDOTA-Ho (CheMatech, Dijon), and Ir-DNA intercalator (Fluidigm, San Francisco). For that, matrix matched calibration standards after Wang et al. were successfully prepared using the same arraying system. We believe that this novel approach opens new ways for automated quantitative single cell LA-ICP-MS.

Der Vortrag zeigt allgemeine Anforderungen an "smarte Feldgeräte" und deren Entwicklung in den vergangenen Jahren. Am Beispiel eines smarten Online-NMR-Sensors, der in einem EU-Projekt von der BAM entwickelt wurde, wird die Umsetzung der Anforderung aufgezeigt. Schließlich werden weitere Technologieanforderungen und Lösungsansätze vorgestellt.
Im Zuge der Digitalisierung der Prozessindustrie werden zunehmend modellbasiere Echtzeitoptimierungsverfahren eingesetzt, sog. „Advanced Process Control“. Mithilfe der sogenannten Modifier-Adaptation ist eine iterative Betriebspunktoptimierung auch mit ungenauen Modellen möglich, sofern zuverlässige Prozessdaten zur Verfügung stehen. Am Beispiel eines smarten Online-NMR-Sensors, der in einem EU-Projekt von der BAM entwickelt wurde, konnte das Konzept in einer modularen Produktionsanlage zur Herstellung eines pharmazeutischen Wirkstoffs erfolgreich getestet werden.
In chemical elements with three or more stable isotopes, mass-dependent stable isotope fractionation yields correlated isotope ratios, m2/m1 and m3/m1. In three-isotope space, i.e. in a δ’m2/δ’m1 vs. δ’m3/δ’m1 plot, data align along a slope θ, the so called ‘triple isotope fractionation exponent’ that scales the two isotope ratios. Theoretical calculations predict small differences in θ for kinetic- and equilibrium isotope fractionation (Young et al. 2002) and thus the precise measurement of θ allows constraining the reaction mechanism. However, due to an apparent lack of precision of stable isotope analysis by MC-ICP-MS, θ is merely used as analytical quality control, i.e. for demonstration that samples and standards plot within their analytical precision in the range of theoretical θ-values originating in δ-zero.
We show how θ can be determined precisely enough by MC-ICP-MS to distinguish kinetic- and equilibrium isotope fractionation, even when isotopic differences between samples are low. For low magnitudes of isotope fractionation, we exploit new, isotopically fractionated isotope standard materials (Vogl et al. 2016). We determine quality norms regarding interferences and measurement conditions to warrant trueness and to maximize precision. We exemplary explore the measurement of the three-isotope composition of Mg stable isotopes, budget the uncertainty of θ-values, and report the first θ-values of carbonate-water pairs and bioapatite. Our measurement approach adds a new dimension to isotope data beyond the δ-scale that has a high potential to reveal different modes of (bio)mineral precipitation in the sedimentary and biological record and thus to contribute solving conundrums in the Earth and Life Sciences.
Mass spectrometric methods (MC-ICP-MS and TIMS) are without doubt the working horse of stable isotope analysis. However, drawbacks of these methods include the high costs for instruments and their operation, experienced operators and elaborate chromatographic sample preparation which are time consuming.
We propose a lower-cost and faster optical alternative for the analysis of isotope ratios of selected elements: high-resolution continuum source molecular absorption spectrometry (HR-CS-MAS). Stable isotope amount compositions of boron (B) and magnesium (Mg) were determined based on the absorption spectra of in-situ generated mono-hydrides and halide (MH and MX) using graphite furnace HR-CS-MAS. The use of a modular simultaneous echelle spectrograph (MOSES) helps to find the maximal isotope shift in the diatomic molecular spectra produced in a graphite furnace by using isotopic spike solutions. Isotopes of boron (10B and 11B) were studied via their hydrides for the electronic transition X 1Σ+ → A 1Π. The spectrum of a given sample is a linear combination of the 10BH molecule and its isotopologue 11BH. Therefore, the isotopic composition of samples can be calculated by a partial least square regression (PLS). For this, a spectral library was built by using samples and spikes with known isotope composition. Boron isotope ratios measured by HR-CS-MAS are identical with those measured by mass spectrometric methods at the 0.15 ‰ level. Similar results were obtained for a multiple isotope system like Mg (24Mg, 25Mg, and 26Mg), where isotope shifts of their isotopologues can be resolved in the MgF molecule for the electronic transition X 2Σ → A 2 Πi. The extension of this methodology to other elements like Li, Ca, Cu, and Sr is discussed.
Imaging of elemental distributions in single cell assays as well as tissue sections can be performed by laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS). This powerful technique offers precise spatially resolved measurements at trace and ultratrace levels and has been established as an excellent tool to answer analytical, biological and biomedical questions. To date, mass cytometry is already able to simultaneously detect up to 40 cellular targets due to conjugation of isotopically pure lanthanides to affinity binders, e.g. antibodies.
To further enhance the ability of multiparametric analysis to more than 100 analytes at once, we investigated lanthanide nanocrystals as new, highly sensitive metal tags for identification of targets in clinical cell assays and tissue samples. Multiparametric analysis will be possible by encoding the lanthanide composition of nanocrystals associated to the affinity binders. Nanocrystals showed remarkable potential for sensitive detection in MS due to high stability and signal amplification compared to e.g. polymer tags, carrying fewer metal atoms. Furthermore, the nanocrystals allow multimodal imaging due fluorescence of Eu3+ as well as contrast enhancing properties of Gd3+ in magnetic resonance imaging.
Synthesis of functionalized lanthanide nanocrystals for further bioconjugation was performed with high reproducibility and monodisperse size distribution. For proof of principle, the uptake and distribution of these nanocrystals within the monolayered cell line A549 were investigated by mapping the intensities at subcellular resolution using LA-ICP-MS. It could be shown, that the cells were efficiently labeled with the nanocrystals. Additionally, the bioconjugation of the nanocrystals to antibodies and particularly the preservation of the antibody specificity was investigated using Dot Blot experiments. All in all, the results imply high sensitivity and the possibility of multiparametric analysis by doting various lanthanides into the nanocrystals.
In der klinischen Diagnostik werden für zytometrische Messverfahren bereits eine Reihe von Reagenzien eingesetzt zur Markierung von Antikörper eingesetzt, um die Detektion von Biomarkern mittels Fluoreszenz- oder Flugzeitmassenspektrometrie zu ermöglichen.
Seit kurzem ist auch eine Imaging Mass Cytometry Kombination direkt erhältlich, wodurch der Nachweis von Biomarkern in Gewebeschnitten erreicht werden kann. Dazu wird eine Kopplung von Laser Ablation und induktiv gekoppeltem Plasma Massenspektrometrie eingesetzt, wobei ähnlich der Massenzytometrie, zuvor Antikörper mit Metallen markiert, und im Anschluss mit dem Gewebeschnitt inkubiert werden. Durch die hohe Ortsauflösung können die Biomarker lokalisiert, und zukünftig vielleicht auch quantifiziert werden.
Insbesondere Lanthanide eignen sich als Markierungsmetalle, da sie einen niedrigen Untergrund und chemisch ähnliches Verhalten zueinander aufweisen. Allein durch diese Elemente können bereits etwa 15 Parameter unterschieden werden, was durch isotopenreine Standards weiter gesteigert werden kann.
Vom Markierungsgrad abhängig werden unterschiedlich viele Metalle am Antikörper gebunden, und beeinflussen so die Sichtbarkeit im ICP-MS. Nanopartikel könnten daher eine deutliche Steigerung der Sensitivität bewirken. GdVO4 Nanokristalle scheinen bisher sehr vielversprechend und bieten neben multiparametrischen Anwendungen auch Multimodalität.
Die Synthese der Nanokristalle zeigte hohe Homogenität und Reproduzierbarkeit in Partikelgröße in der Zusammensetzung. Ein erstes Experiment mit einer Zellkultur konnte bereits die effiziente Markierung der Zellen unter Beweis stellen, wobei durch hohe Signalstärke auch subzelluläre Auflösung in der LA-ICP-MS erreicht werden konnte.

Nanocrystals showed remarkable potential for sensitive detection in MS due to high stability and signal amplification compared to e.g. polymer tags, carrying fewer metal atoms. Furthermore, the nanocrystals allow multimodal imaging due fluorescence of Eu3+ as well as contrast enhancing properties of Gd3+ in magnetic resonance imaging.
Synthesis of functionalized lanthanide nanocrystals for further bioconjugation was performed with high reproducibility and monodisperse size distribution. For proof of principle, the uptake and distribution of these nanocrystals within the monolayered cell line A549 were investigated by mapping the intensities at subcellular resolution using LA-ICP-MS. It could be shown, that the cells were efficiently labeled with the nanocrystals. Additionally, the bioconjugation of the nanocrystals to antibodies and particularly the preservation of the antibody specificity was investigated using Dot Blot experiments. All in all, the results imply high sensitivity and the possibility of multiparametric analysis by doting various lanthanides into the nanocrystals.
Nanoparticles (NPs) have found a wide range of applications in research and industry. Thereby the interaction of NPs with biological systems like cells has become a major field of interest, ranging from medical applications to nanotoxicology. Size, shape and surface modification of the nanomaterials determine the uptake rate and pathway into the cells, and therefore impact specific cell components and processes.
Laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) is an established quantitative multi-elemental analysis and mapping technique. In recent years, it was shown that LA-ICP-MS can provide quantitative as well as distribution information of metal containing nanoparticles (NPs) in cell samples.
Here LA-ICP-MS was applied for the imaging of individual fibroblast cells to study the uptake and intracellular processing of NPs. Our results show that LA-ICP-MS can be used to localize nanoparticle aggregates within cellular compartments. The studied NPs accumulate in the perinuclear region in the course of intracellular processing, but do not enter the cell nucleus. The uptake efficiency depends strongly on the physico-chemical properties of the nanostructures as well as on the incubation conditions like concentration and incubation time.
ICP-MS was used to determine the composition of the nanomaterials as well as the number of NPs in cells after acid digestion of the samples.

Tutzing-Symposion vom 15.–18.04.2018 wurde mit Vorträgen und Kreativworkshops erkundet, welche speziellen Anforderungen die Prozessindustrie hat, welche digitalen Innovationen bereits umgesetzt wurden und wo noch Handlungsbedarf besteht. Ein Workshop befasste sich mit den Themenfeldern Datenkonzepte, Datenanalyse, Big Data und künstliche Intelligenz. Es geht nicht um die Digitalisierung von heute. Im Angesicht der wachsenden Digitalisierung unserer Prozesse stellt sich die Frage, ob wir den Prozess wirklich gut kennen. Ob alle Verfahrensschritte detailliert hinterlegt wurden. Nur mit einem heuristischen Ansatz kann das vorhandene Wissen nicht digitalisiert werden.
Sehr schnell werden die Mechanismen eines Massenmarktes mit denen einer Nische verwechselt. Nicht jeder Mechanismus, den wir von großen Suchmaschinen oder Einkaufsportalen kennen, gibt uns einen Hinweis auf Nutzen und Verfügbarkeit für die Prozess- oder pharmazeutische Industrie. Eine gute Analyse der Anforderungen in der Zukunft mit einem Abgleich der derzeitigen technischen Möglichkeiten ist Voraussetzung für eine Verbesserung der derzeitigen digitalen Umsetzung. Dabei ist es sinnvoll unkonventionelle Methoden einzusetzen.
Small variations in the isotopic composition of some elements have been used as proof of provenance of mineral and biological samples, to describe geological processes, and to estimate a contamination source. Routinely, isotope compositions are measured by mass spectrometry; the working horse of the isotope analysis. However, mass spectrometric methods are expensive, time-consuming and they require a high qualified analyst.
Here, an alternative faster and low-cost optical method for isotope ratio determination is investigated: high-resolution continuum source molecular absorption spectrometry (HR-CS-MAS). Stable isotope amount composition of X = Li, B, Mg, Ca and Sr have been determined by monitoring the absorption spectra of their in situ generated hydrides (XH) in graphite furnace HR-CS-MAS. For example, the system of two stable isotopes of boron (10B and 11B) was studied via its hydride for the electronic transition X1Σ+ → A1Π (Fig. 1a). The spectrum of a given sample is a linear combination of the 10BH molecule and its isotopologue 11BH. Therefore, isotopic composition of samples and reference materials are calculated by a partial least square regression (PLS). For this, a spectral library is built by using samples with known isotope composition. Results are metrologically compatible with those reported by mass spectrometric methods. [1] Similar results are obtained for n isotope systems like Mg (24Mg, 25Mg, and 26Mg), where isotope shift of their isotopologues can be resolved as shown in Fig.1 b. The extension of this methodology to other elements like Li, Ca and Sr is discussed [2].

Following fluorinated drugs by molecular absorption spectrometry – from cancer cells to body fluids

Fluorine has been widely introduced into pharmaceutical drugs. Due to the high C-F bond strength, a single atom substitution with fluorine produces drastic desirable and tunable changes in the molecular properties. However, the clinical monitoring of these drugs is not straightforward.
Organofluorine compounds are elusive for a non-targeted analysis; a significant problem in fluorine determination is the lack of suitable detection techniques. Standard atomic absorption and emission spectrometers cannot access fluorine, because their resonance lines lie in the VUV spectral range below 100 nm. In the case of conventional inductively coupled argon plasmas (ICP), the plasma energy is too low to generate a significant population of excited fluorine atoms. Recently, our group introduces high-resolution continuum source absorption spectrometry (HR-CS MAS) as a new way for the indirect monitoring of fluorinated compounds. Main benefits of HR-CS-MAS includes low limits of detection, complete analyte recovery, simple to no sample preparation, and short time analysis.
Common approaches to improve the optical properties of semiconductor quantum dots and lanthanide doped nanophosphors present core/shell structures as radiationless deactivation at the particle surface is usually the main energy loss mechanism. This has led to increasingly sophisticated particle architectures using multishell systems with shells of different chemical composition and thickness and initiated an increasing number of quantitative spectroscopic studies focusing on the key performance parameter photoluminescence quantum yield to identify optimum particle structures. This is particularly challenging in the long wavelength region > 1000 nm and for nonlinear emitters like upconversion nanocrystals. Here, we present suitable absolute methods to quantify the photoluminescence of these different emitters in the vis/NIR/IR and as function of excitation power density and underline the impact of such measurements on a profound mechanistic understanding of the nonradiative deactivation pathways in semiconductor and upconversion nanocrystals of different size and particle architecture in different environments.
The antimicrobial photodynamic approach has been demonstrated as an efficient and sustainable process for the eradication of microbial pathogens. In this work, silica-coated Magnetite nanoparticles (NPs) were used as carriers of glycosylated porphyrins and phthalocyanines. Their subsequent cationization
resulted in the production of stable antimicrobial photosensitizing materials, effective against E. coli. Suspensions of the photocatalysts in water present bimodal size distributions formed by big clusters and small NPs with hydrodynamic diameters between 8 and 38 nm. The presence of small NPs in the suspensions is related to an effective photodynamic inactivation (PDI) of E. coli cells. Glycosylation of the PS showed a positive effect on the PDI performance, which could be related to a higher accumulation of the photocatalyst over the bacterial cell membrane. In addition, these biocidal agents proved to be photostable and their photoactive performance decreased only between 23% and 28% upon 5 PDI cycles, mostly because of the loss of material between cycles, which makes them promising materials for water disinfection purposes.
Molecularly Imprinted Polymers with Integrated Fluorescence as Versatile Biomimetic Sensing Matrices

Molecularly imprinted polymers (MIPs) are an established, versatile and high-performance matrix for the selective separation or enrichment of (bio)chemical species, especially small molecules of biochemical or environmental relevance. MIPs are prepared through the polymerization of a mixture of functional monomers and cross-linkers in the presence of the template with subsequent extraction of the latter. Conceptionally, this process can be seen as mimicking in a strongly accelerated, though single-step manner a biological process such as antibody formation. Because the resulting MIPs contain cavities in their matrix that are complementary in size, shape and electronic/ electrostatic or hydrogen bonding demand to the imprinted target molecule or template, these polymers are frequently termed “artificial antibodies”. Compared to natural antibodies, they are chemically and physically much more robust. Regarding sensitivity and selectivity, however, there is still a gap to bridge before MIPs can fully compete with antibodies.
Another favorable aspect that distinguishes MIPs from antibodies is that they can be endowed with an explicit function, allowing the use of MIPs in applications that require more than only an efficient binder. For instance, if specifically designed and polymerizable fluorescent indicators are integrated as functional monomers into a MIP, direct fluorescence sensing can be accomplished. Because MIPs can be prepared in a variety of different formats, their combination with miniaturized or other specific analytical techniques or sensory devices is possible, especially when the transduction mode is light. This presentation will introduce basic design considerations, challenges, limitations and the potential that lies with such sensor materials with some recent examples of our group, targeting various organic oxoanions as analytes.
Contamination of natural bodies of water with oil and lubricants (or generally, hydrocarbon derivatives such as petrol, fuel and others) is a commonly found phenomenon around the world due to the extensive production, transfer and use of fossil fuels. The timely identification of these contaminants is of utmost importance, since they directly affect water quality and represent a risk for wildlife and human health even in trace amounts.
In this work, we develop a simple system for the on-field detection of total petroleum hydrocarbons (TPH) in water and soil, the "Spectrocube". The test is based on the measurement of the fluorescence signal emitted by the molecular rotor 4-DNS-OH dye. This dye is embedded in a hydrophobic polymeric matrix (PVDF), avoiding interactions of water with the dye and providing a robust support for use in test-strip fashion. The test-strip’s fluorescence intensity increases linearly at low concentrations of TPH, reaching a saturation value at higher concentrations.
For excitation and evaluation of the test-strip fluorescence, a simple miniature optical system was designed. The system works semi-quantitatively as solvent-free TPH detection kit, as well as quantitatively when using a simple cyclopentane extraction step. To simplify the fluorescence read-out, the device is coupled to a tablet computer via Bluetooth, running a self-programmed software ("app").
Ionophore antibiotics are used to cure and prevent coccidiosis by chicken especially in broiler farming. The residues are found not only in food products (chicken and eggs) but also in the environment (manure, soil or water). In this work the ionophores monensin (MON), salinomycin (SAL), maduramicin (MAD) and lasalocid (LAS) are investigated aiming to study their transformation products (TPs) through biotransformation processes. Biotransformation can be divided into two phases, phase I: oxidation, reduction or hydrolysis and Phase II: conjugation reactions. It is necessary to further examine the biotransformation pathways to determine TPs to be able to detect residues more specifically in different matrices.
The technique of electrochemistry (EC) offers the opportunity to simulate biotransformation processes and to generate TPs for further analysis. The combination of EC with liquid chromatography and mass spectrometry (EC-LC-MS) provide a fast and simple tool to separate and determine the EC-generated TPs. The electrochemical flow through cell is coupled to the (LC)-MS system, allowing the reaction mixture to be separated by a RP-18 column and then analyzed in the MS. The oxidation products are generated at different potentials between 0.0 – 2.5 V vs. Pd/H2 using glassy carbon or boron doped diamond as working electrode materials .
The results show a broad spectrum of different TPs depending on used solvents and working electrode materials. Among the generated TPs already known as well as unknown TPs of the drugs can be found. Further investigations on structure elucidation of unkown TPs are planned.

In recent years, elemental imaging of biological samples using laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) is gaining more and more importance. Improvements concerning both spatial resolution (down to 1 µm) and signal-to-background ratio due to low-dispersion LA chambers make LA-ICP-MS particularly interesting for single cell analysis.
Here LA-ICP-MS was applied for the imaging of individual cells to study the uptake and intracellular processing of metal-containing nanostructures. The cells were incubated with different NPs under varying experimental conditions and afterwards fixed with para-formaldehyde and dried for LA analysis. High-spatial resolution LA-ICP-MS was achieved by careful optimisation of the laser ablation parameters.
Our findings show, that LA-ICP-MS is applicable to localize NP aggregates within cellular compartments. The uptake efficiency depends strongly on the physicochemical properties of the nanostructures as well as on the incubation conditions like concentration and incubation time.
The results demonstrate the potential of LA-ICP-MS providing insight into nanoparticle-cell interaction dependent on experimental parameters.
Quantification of sulphur in copper metals by isotope dilution LA-ICP-MS using polyethylene frits

Sulphur is one of the relevant impurities in copper and its alloys affecting their material properties. To ensure the quality of copper products, fast direct solid sampling techniques are very attractive. However, for the calibration suitable matrix reference materials are required. For the certification of such reference materials appropriate, SI-traceable analytical methods are essential. Therefore, a procedure was developed to quantify total sulphur in copper by combining the classical isotope dilution (ID) technique and laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS).
Here, for the first time, polyethylene (PE) frits were used to prepare appropriate solid samples for the sulphur quantification in copper metals (alloyed/unalloyed) by isotope dilution LA-ICP-MS. The properties of the PE frit meet the requirements as porous material with high absorption efficiency, thermal and chemical resistance as well as low sulphur blank. Different copper reference materials were used to develop and validate the procedure. The copper samples were spiked with 34S, digested with nitric acid and then the digests were absorbed on PE frits. After drying, the frits were analysed by LA-ICP-IDMS using a Nd:YAG laser at 213 nm coupled to an ICP sector field mass spectrometer. It could be demonstrated, that the sample solution dispersed on the frits did not influence the 32S/34S ratio significantly even though the sulphur intensities were fluctuating along the scanned lines. Relative standard deviations of the isotope ratios were below 5 % in average between three line scans (except for the pure spike solution and procedure blank). The measurement results were validated by comparing them with the results obtained by conventional ICP-IDMS after analyte-matrix separation. Plotting the mass fraction of sulphur in copper obtained by LA-ICP-IDMS versus those obtained by ICP-IDMS yields a linear curve with a correlation coefficient of 0.9999 showing a strong agreement between both techniques.
The metrological traceability to the SI from the kg down to the sulphur mass fraction in copper is established by an unbroken chain of comparisons, each accompanied by an uncertainty budget. Thus, the measurement results are considered reliable, acceptable and comparable within the stated measurement uncertainty.

Humidified gas flow at adjusted gas pressure was split into two flows, one passed a coulometric sensor and the other one passed a reference hygrometer after decompression. Both signals were recorded and then compared after calculation of resulting frost point temperature. Calculation is based on a calibration function obtained at ambient pressure. Comparison showed that an increasing pressure resulted in a higher derivation between sensor signal (calculated frost point temperature) and reference frost point temperature. At an absolute pressure of 2 bar the differences were minor in consideration of the uncertainty, however at 15 bar the differences were 6.77 K. Nevertheless, it was possible to measure the gas humidity at higher pressure with coulometric trace humidity sensors.

Diclofenac (DCF) is a widely used drug against fever, inflammation, pain, and rheumatic diseases. An average of 70 % of the ingested diclofenac is excreted in the urine. Thus, 63 tons per year are introduced into the water cycle in Germany. Due to insufficient removal of diclofenac in wastewater treatment plants, residues of diclofenac can be found in surface water and sometimes in drinking water. In order to establish an immunoassay, an anti-DCF antibody was required. Some antibody developments have been reported, based on an immunogen resulting from direct coupling of diclofenac to proteins via the carboxylic function. Finally, we used a monoclonal antibody, clone F01G21.
Due to their toxicity and widespread application for mining and industrial purposes, cyanides are ranking among the most important inorganic pollutants which should be tested and monitored not only in the aquatic environment, but also in soils and soil-like materials. Reference materials of soils with relevant contents of cyanide to ensure reliable test results of laboratories are rare today.
New certified reference material (CRM) BAM-U116/CGL306 “Cyanide in soil” was produced within a framework of cooperation between Central Geological Laboratory (CGL) of Mongolia and Federal Institute for Materials Research and Testing (BAM) of Germany in 2013-2017.
The CRM BAM-U116/CGL306 represents a mixture of a sandy soil collected from a contaminated former gasworks area in the Berlin region (Germany) and an unpolluted sandy soil from Nalaikh region (Mongolia). The bulk candidate material for this reference material was prepared at CGL CRM Laboratory exclusively destined to the preparation of reference materials and equipped with modern technical equipment. Homogeneity, stability and shelf life were studied in full compliance with ISO Guide 35. The CRM was evaluated as sufficiently homogeneous. Statistical evaluation of certification analysis was performed using software packages SoftCRM and ProLab Plus. Certified value of total cyanide of the CRM is 12.0 mg/kg and expanded uncertainty was assigned as 0.8 mg/kg.
The intended purpose of this material is the verification of analytical results obtained for the mass fraction of total cyanide in soils and soil-like materials applying the standardized procedure ISO 11262:2011. As any reference material, it can also be used for routine performance checks (quality control charts) or validation studies.

Die generierte Gasfeuchte für die Untersuchungen betrug bezogen auf die Frostpunkttemperatur -80 °C bis -30 °C, was gleichbedeutend mit einem Wasserdampf-Stoffmengenanteil von 0,5 μmol·mol-1 bis 376 μmol·mol-1 ist. Zusätzlich zu der Messung mit coulometrischen Sensoren erfolgte eine Referenzmessung der Gasfeuchte mittels eines Taupunkthygrometers. Erste Ergebnisse haben gezeigt, dass es möglich ist mit dieser Art der Beschichtung unterschiedliche Gasfeuchten zu messen. Aber es ergaben sich trotz gleicher Feuchte unterschiedliche Signalintensitäten bei den benutzen Sensoren und es zeigte sich eine Drift des Sensorsignales.

Statistic process control as well as process capability demand for calibrated determination of layer thicknesses in various industries, e.g. automotive, aerospace, microelectronics manufacturing. Calibration requires well know and well characterized samples. A calibration laboratory accredited according to DIN EN ISO 17025 has the objective to distribute standards traceable to SI units to industrial laboratories for quality control of manufacturing of various products. Especially, the thickness determination of thin metallic coatings e.g. from galvanic processes or layer deposition using X-Ray Fluorescence can be significantly improved by customized calibration samples. This is essential as the measurement uncertainty directly correlates to the capability performing reliable control of processes with high yield. For calibration laboratories, the validation of results using round robins and the direct comparison to national metrology institutes is a prerequisite to demonstrate the competence to perform calibration services.
In this paper a strategy to obtain traceability and validation for thin alloy layers as well as first results are presented. The combined use of the accredited method for determination of mass per area from measurement of mass and area combined with standard free X-Ray Fluorescence as well as chemical analysis of dissolved samples with thin layers is deployed for material systems as NixZn1-x as well as NixP1-x. The obtained results are compared to reference free X-Ray Fluorescence at the BESSY II laboratories of Physikalisch-Technische Bundesanstalt. An excellent agreement of the obtained measured values as mass per unit area and alloy concentrations from the different applied methods within the measurement uncertainty was observed for NixP1-x showing the successful performed traceability of the calibration samples to SI units in combination with a validation of results by national metrology institutes and the round robin approach.
Due to the increasing consumption of cereals worldwide, monitoring of growth, storage and processing is becoming more crucial. Particularly when stored, infested grains breed fungal clusters (“hot spots”) in which mycotoxins greatly exceed allowed maximum levels. Because of their unpredictable presence, current sampling and procedures for mycotoxin analysis are complex and contain invasive and cost intensive steps.
A possible approach to avoid sampling inaccuracies is the non-invasive analysis of the homogeneous gas phase above the crops, instead of analyzing random samples. However, this procedure requires microbial volatile organic compounds (MVOC´s) being released by the samples. Previous investigations revealed trichodiene to be a precursor in trichothecenes biosynthesis – one of the largest mycotoxin groups with more than 180 compounds. Due to its non-functionalized sesquiterpene structure, quantification of trichodiene using Headspace GC-MS methods is possible (e.g. [1]). Thereby, it could be used as biomarker for trichothecene contamination in foodstuff.
However, further studies are necessary. The correlation between trichodiene concentration in the gas phase and trichothecenes mass fraction in the sample must be examined closely to draw conclusions about the trichothecene content in samples. Realizing this idea, would widely extend the applicability of trichodiene and enormously simplify trichothecene quantification. Hence, this first step of an ongoing study aims to develop a laboratory reference method using trichodiene as volatile biomarker to quantify trichothecenes in cereals. Static headspace and SPME-enrichment coupled to gas chromatography with mass spectrometry (GC-MS) were employed. In a second step, this reference method is intended to validate new approaches for fast on-site screening of trichodiene in cereals.
Alternariol (AOH) and Alternariol monomethylether (AME) are two secondary metabolites of Alternaria fungi which can be found in various foodstuffs like tomatoes, nuts and grains. Due to their toxicity and potential mutagenic activity the need for the development of high-throughput methods for the supervision of AOH- and AME-levels is of increasing interest. As the availability of both native and labelled AOH and AME analytical standards is very limited we herein wish to present a novel concise approach towards their synthesis employing a ruthenium-catalyzed ortho-arylation4 as the key step.
Multivariate analysis of MALDI imaging mass spectrometry data of mixtures of single pollen grains

Here, we present an advanced approach to identify pollen grains in mixtures based on:
- a simplified sample preparation procedure
- MALDI imaging mass spectrometry
- chemometric tools
Our goals are to:
- explore the biomolecular variations in pollen
- determine species-specific peak patterns
- discriminate and identify single pollen grains in mixtures using MSI
Die ehemals wegen komplexer Technik und vergleichsweise hohen Wartungskosten beim Anwender ungeliebte Prozessanalysentechnik (PAT) erfährt sich mittlerweile immer mehr als etablierender Bereich mit einem großen Zuwachs und Dynamik. Die Prozesskontrolle und -steuerung über physikalische Kenngrößen wie Druck und Temperatur lässt eine weitere Optimierung der Anlagen kaum mehr zu. Nur mittels stoffspezifischer Analysen lassen sich Rohstoffschwankungen, Ausbeuten und Energieeinsatz konsequent optimieren.
Der systematische Einsatz der Prozessanalysentechnik verändert Prozesse und Produktionsumgebungen und hat damit die Chance, Kernstück dezentral automatisierter Produktionseinheiten zu werden. Ein neuer Arbeitskreis der NAMUR AK 3.7 „Smarte Sensorik, Aktorik und Kommunikation“ wird diesem verstärkt Rechnung tragen. Es werden reale Anwendungsbeispiele aufgezeigt, die eine schnelle Amortisation von PAT im Prozess untermauern.
To keep up with emerging mycotoxins and their transformation products fast and reliable toxicity tests are needed. Toxicity testing of mycotoxins is carried out usually by performing in vitro assays or is evaluated by using laboratory animals like mice, rats or chicken in in vivo studies.
Settled between classical in vitro approaches and in vivo studies with higher animals are tests with the nematode Caenorhabditis elegans. Since Sydney Brenner described 1974 the cultivation and handling of C. elegans, this worm is widely used as model organism in developmental biology and neurology. Due to many benefits like easy and cheap cultivation, a completely sequenced genome and short generation time, it also plays an important role in toxicological research. Finally, the high number of conserved genes between human and C. elegans make the worm an ideal candidate for toxicological investigations.
In this study we used C. elegans to assess the toxic effects of the relevant food mycotoxin citrinin (CIT), the mycoestrogen zearalenone (ZEN) and the modified mycotoxin ZEN-14-sulfate (ZEN-14-S) on different lifetable parameters including reproduction, thermal and oxidative stress tolerance and lifespan. All tested mycotoxins significantly decreased the amount of offspring. In case of ZEN and CIT also significant negative effects on stress tolerance and lifespan were observed compared to the control group.
Moreover, metabolization of mycotoxins in the worms was investigated by using LC MS/MS. Extraction of the worms treated 5 days with mycotoxin-containing and UVC-killed bacteria showed metabolization of ZEN to α-ZEL and β-ZEL (ZEL = zearalenol, ratio about 3:2). ZEN 14-S was reduced to ZEL 14-S and CIT was metabolized to mono hydroxylated CIT.
An overview about different analytical approaches will be presented of how to detect metals in individual biological cells by use of ICP-MS. For this purpose, we are using different sample introduction systems for ICP-MS for detection, imaging and quantification of metals at cellular levels.
By use of laser ablation, we have studied the up-take by and distribution of nanoparticles in single cells. Recently we have developed staining techniques to measure protein and DNA content of cells and identifying the cell status by immunoassays using metal-tagging of antibodies. New research based on cell arrays will be shortly discussed.
Using pneumatic nebulization and microdroplet generation, we have also studied the up-take of nanoparticles and toxic metals as well as essential elements in single cells using different ICP-MS mass spectrometric concepts (sector field instrument, triple-quad instrument, time of flight (CyTOF) instrument).
The different ICP-MS based methods will be compared concerning their analytical figures of merit and their strengths and weaknesses will be evaluated.
