Raw data from metabolomics experiments are initially subjected to peak identification and signal deconvolution to generate raw data matrices m × n, where m are samples and n are metabolites. We describe here simple statistical procedures on such multivariate data matrices, all provided as functions in the programming environment R, useful to normalize data, detect biomarkers, and perform sample classification.
Monitoring chemical reactions is the key to chemical process control. Today, mainly optical online methods are applied, which require excessive calibration effort. NMR spectroscopy has a high potential for direct loop process control while exhibiting short set-up times. Compact NMR instruments make NMR spectroscopy accessible in industrial and harsh environ¬ments for advanced process monitoring and control, as demonstrated within the European Union’s Horizon 2020 project CONSENS.
We present a range of approaches for the automated spectra analysis moving from conventional multivariate statistical approach, (i.e., Partial Least Squares Regression) to physically motivated spectral models (i.e., Indirect Hard Modelling and Quantum Mechanical calculations). By using the benefits of traditional qNMR experiments data analysis models can meet the demands of the PAT community (Process Analytical Technology) regarding low calibration effort/calibration free methods, fast adaptions for new reactants or derivatives and robust automation schemes.
A systematic study of the luminescence properties of monodisperse β-NaYF4: 20% Yb3+, 2% Er3+ upconversion nanoparticles (UCNPs) with sizes ranging from 12–43 nm is presented utilizing steady-state and time-resolved fluorometry.
Special emphasis was dedicated to the absolute quantification of size- and environment-induced quenching of upconversion luminescence (UCL) by highenergy O–H and C–H vibrations from solvent and ligand molecules at different excitation power densities (P). In this context, the still-debated Population pathways of the 4F9/2 energy level of Er3+ were examined. Our results highlight the potential of particle size and P value for color tuning based on the pronounced near-infrared emission of 12 nm UCNPs, which outweighs the red Er3+ emission under “strongly quenched” conditions and accounts for over 50% of total UCL in water. Because current rate equation models do not include such emissions, the suitability of these models for accurately simulating all (de)population pathways of small UCNPs must be critically assessed. Furthermore, we postulate population pathways for the 4F9/2 energy level of Er3+, which correlate with the size-, environment-, and P-dependent quenching states of the higher Er3+ energy levels.
Cerro do Jarau is a conspicuous, circular morpho‐structural feature in Rio Grande do Sul State (Brazil), with a central elevated core in the otherwise flat “Pampas” terrain typical for the border regions between Brazil and Uruguay. The structure has a diameter of approximately 13.5 km. It is centered at 30o12′S and 56o32′W and was formed on basaltic flows of the Cretaceous Serra Geral Formation, which is part of the Paraná‐Etendeka Large Igneous Province (LIP), and in sandstones of the Botucatu and Guará formations. The structure was first spotted on aerial photographs in the 1960s. Ever since, its origin has been debated, sometimes in terms of an endogenous (igneous) origin, sometimes as the result of an exogenous (meteorite impact) event. In recent years, a number of studies have been conducted in order to investigate its nature and origin. Although the results have indicated a possible impact origin, no conclusive evidence could be produced. The interpretation of an impact origin was mostly based on the morphological characteristics of the structure; geophysical data; as well as the occurrence of different breccia types; extensive deformation/silicification of the rocks within the structure, in particular the sandstones; and also on the widespread occurrence of low‐pressure deformation features, including some planar fractures (PFs). A detailed optical microscopic analysis of samples collected during a number of field campaigns since 2007 resulted in the disclosure of a large number of quartz grains from sandstone and monomict arenite breccia from the central part of the structure with PFs and feather features (FFs), as well as a number of quartz grains exhibiting planar deformation features (PDFs). While most of these latter grains only carry a single set of PDFs, we have observed several with two sets, and one grain with three sets of PDFs. Consequently, we here propose Cerro do Jarau as the seventh confirmed impact structure in Brazil. Cerro do Jarau, together with Vargeão Dome (Santa Catalina state) and Vista Alegre (Paraná State), is one of very few impact structures on Earth formed in basaltic rocks.
The main drawback in radiometric imaging is the degradation of the spatial resolution with increasing depth, which results in blurred images for deeper lying structures. We circumvent this degradation with blind structured illumination, combined with a non-linear joint sparsity reconstruction algorithm. We demonstrate this by imaging a line pattern and a star-shaped structure through a metal sheet with a resolution four times better than the width of the thermal point-spread-function. The ground-breaking concept of super-resolution can be transferred from optics to diffusive imaging by defining a thermal point-spread-function similar to the Abbe limit for a certain optical wavelength.
The detection of cracks before the failure is highly significant when it comes to safety-relevant structures. Crack detection in metallic samples at high surface temperature is one of the challenging situation in manufacturing industries.
Laser thermography has already proved its detection capability of surface cracks in metallic samples at room temperature. In this work a continuous wave (CW) laser use to generate a laser, which is using to scan the metal surface with notch.
The corresponding heat distribution on the surface monitored using infrared thermal (IR) camera. A simplified 3D model for laser thermography is developed and validated with experimental results. A dedicated image processing algorithm developed to improve the detectability of the cracks. To understand the dependency of surface temperature, laser power, laser scanning speed etc. in defect detection, we carried out parametric studies with our validated model. Here we Report the capability of laser thermography in crack detection at elevated temperature.
X-ray fluorescence analysis, due to its non-destructive nature and ist suitability to work with historic objects in situ, quickly became one of the most important methods for the evaluation of iron-gall ink. The main advantage of this qualitative and semi-quantitative method is that it makes it easy to differentiate between inks, based on the assumption that the differences result from the manufacture of the ink.
This work explores the question whether the ink ‘fingerprint’ results strictly from the elemental composition of the basic ink ingredients even if it is stored in vessels made of metals or metal alloys. In addition, we tested and compared the performance of three different XRF spectrometers.
We prepared various lab-grade inks according to historical ink recipes and measured the metal content of the ink deposited on sized cotton Linters paper with three types of XRF spectrometers: a simple hand-held device with an interaction spot of 4 mm and two devices equipped with poly-capillary Xray optics for line scanning and imaging. Since the exact elemental mass composition of the non-aged ink samples was known, we were able to evaluate the accuracy of the research procedure. Lab-grade inks were then aged in the metal jars imitating inkwells. The aging of the inks in the metal containers resulted in the significant change of the primary inks fingerprint as opposed to that of the control inks stored in glass containers. This effect was independently confirmed by the measurements conducted with every instrument we used. We will present a brief comparison of the results 43 achieved when using different spectrometers and a possible hypothesis explaining the processes that occurred.

We examine the behavior of reinforced concrete components subjected to impact induced loading conditions which might be caused by vessels collisions such as aircraft fuel tanks The concrete plates were impact damaged at TU Dresden and shipped to BAM At BAM laminar tomography as the imaging method is used to determine and quantify the damage state An automatic crack detection method based on template matching is applied to find the cracks and we aim to develop a new method using machine learning Algorithms In addition numerical models are developed to understand the experiment and to predict the damage structures based on failure mechanisms.
X-ray scatter imaging is a well-established NDT technique to inspect complex objects using only a single-sided access. We present a specially designed multi-slit scatter camera consisting of several twisted slits which are parallelly arranged in a metal block. The camera projects one image per slit to the digital detector array, where the images are overlaying each other. The aperture is corrected based on a de-convolution algorithm to focus the overlaying projections into a single representation of the object. To achieve high scatter intensities from an object, it is necessary to optimize the parameters of the scatter system by simulation.

In case study one of the CONSENS project, two aromatic substances were coupled by a lithiation reaction, which is a prominent example in pharmaceutical industry. The two aromatic reactants (Aniline and o-FNB) were mixed with Lithium-base (LiHMDS) in a continuous modular plant to produce the desired product (Li-NDPA) and a salt (LiF). The salt precipitates which leads to the formation of particles. The feed streams were subject to variation to drive the plant to its optimum.

A novel approach for rating fatigue-initiating inclusions in highly demanding steel (INCAFAT)
(2018)
INCAFAT project aimed to improve existing fatigue damage models by establishing the most suitable combination of measurement techniques to characterise harmful inclusion populations in highly demanding steels. The different inclusion assessments carried out confirm that, chemical composition, secondary metallurgy and manufacturing route affect content, nature, size and shape of inclusions. According to the FEM model, inclusions produce an alteration of the stress field in their surrounding region, which can promote a fatigue failure. Experimental work on fatigue testing has demonstrated that depending on the stressed direction fatigue failures in highly demanding steels could be produced by different types of inclusions. Fractography analyses confirmed that meso-inclusions harmful in fatigue cannot be rated by standard methods, nor 10 MHz ultrasonic testing (macro) or micro-cleanness assessments. The necessity of rating these meso-inclusions has led to critical evaluation of Extreme Value Analysis according to ASTM E2283-08 and the development of high frequency immersion ultrasonic testing. EVA methodology based on inclusion width can be applied reliably when principal stress is parallel to the rolling direction. On the contrary, if inclusions are testing in the elongated directions its fails. On the other hand, the guidelines and recommendations for high frequency ultrasonic testing have been compiled in a new European standard draft. This method based on focal beam probes and high-resolution devices is able to provide information on meso-inclusion distribution.

Surface analysis is still gaining importance, as most modern technology is in essence surface and thin film technology. In microelectronics and optics but also in many fields of energy storage and conversion, thin surface layers or structured surfaces are key components. Optical surface measurements are useful because of their reliability, robustness, and scalability – properties, which are especially desirable in a production environment. However, many of these methods take require considerable effort in data analysis. Spectroscopic ellipsometry (SE) is maybe the prime example of this class of methods. To combine the advantages of SE with metrological trueness and traceability, we combine it in a group of other methods, such as electron microscopy, instrumental surface analysis, X-ray diffraction.
This presentation will give an overview on the principles of SE, its available quantities and the general properties of this measurement technique. Several examples from current and past projects of BAM-6.7 will show how a measurement community involving ellipsometry can benefit many fields of interest, from polymer science to energy applications. In the context of creating a “measurement landscape”, standardisation and standard compliance play an important role. The efforts of BAM in standardisation and accrediting of ellipsometry will be shown demonstrating the possible impact on quality assurance in different fields of technology.
While today, engineers can choose from a wide range of rheology modifying admixtures, in some parts of the world, these are difficult to access, due to their complex processing. However, alternatives can be bio‐based polymers such as polysaccharides from various sources. These are easily accessible all over the world, do not demand for complicated processing, and typically they are more sustainable than many established materials, which are crude oil‐based.
The paper presents the effects of acacia gum, cassava starch and the gum of triumfetta pendrata A. Rich on the rheological performance of cementitious systems. It is shown that acacia gum can be as efficient as polycarboxylate based superplasticisers, cassava starch can reduce the yield stress slightly with little effect on the plastic viscosity, and the gum of triumfet ta pendrata A. Rich increases the thixotropy of cement pastes with plasticizing polymers significantly.
Structural design and application have always been linked to the compressive strength of concrete as the main relevant criterion. This was justifiable in the past, where concrete consisted of water, ordinary Portland cement and aggregates, but this concept is no longer relevant for modern and more sustainable cement and concrete.
Despite these new developments, existing standards, guidelines and academic curricula have not been much updated and are still used worldwide. There is a need to change this situation by proper education of the users.
This overview describes the challenges that arise at a user Level from the higher complexity of modern concrete, and defines needs and requirements for enhanced applicability of sustainable concrete concepts.
Furthermore, recommendations are given on how better concrete practice can be communicated to all the involved parties, from civil and design engineers to constructors and site-appliers on the construction site.

Nanomaterials are used in many different applications in the material and life sciences. Examples are optical reporters, barcodes, and nanosensors, magnetic and optical contrast agents, and catalysts. Due to their small size and large surface area, there are also concerns about their interaction with and uptake by biological systems. This has initiated an ever increasing number of cyctoxicity studies of nanomaterials of different chemical composition and surface chemistry, but until now, the toxicological results presented by different research groups often do not address or differ regarding a potential genotoxicity of these nanomaterials. This underlines the need for a standardized test procedure to detect genotoxicity.1,2
Aiming at the development of fast, easy to use, and automatable microscopic methods for the determination of the genotoxicity of different types of nanoparticles, we assess the potential of the fluorometric γH2AX assay for this purpose. This assay, which can be run on an automated microscopic detection system, relies on the determination of DNA double strand breaks as a sign for genotoxicity.3 Here, we present first results obtained with broadly used nanomaterials like CdSe/CdS and InP/ZnS quantum dots as well as iron oxide, gold, and polymer particles of different surface chemistry with previously tested colloidal stability. These studies will be also used to establish nanomaterials as positive and negative genotoxicity controls or standards for assay performance validation for users of this fluorometric genotoxicity assay. In the future, after proper validation, this microscopic platform technology will be expanded to other typical toxicity assays.

Many ambient desorption/ionization mass spectrometry (ADI-MS) techniques rely critically on thermal desorption. Meanwhile, the analyte classes that are successfully studied by any particular ADI-MS methods are strongly dependent on the type of ionization source. Generally, spraybased ionization sources favor polar analytes, whereas plasmabased sources can be used for more hydrophobic analytes and are more suitable for molecules with small molar masses. In the present work, classic atmospheric-pressure chemical ionization (APCI) is used. To provide improved desorption performance for APCI, a surface acoustic wave nebulization (SAWN) device was implemented to convert liquid analytes into fine airborne particles. Compared to conventional SAWN that is used solely as an ionization source for liquid samples, the coupling of SAWN and APCI significantly improves ion signal by up to 4 orders of magnitude, reaching comparable ion abundances to those of electrospray ionization (ESI). Additionally, this coupling also extends the applicable mass range of an APCI source, conventionally known for the ionization of small molecules <500 Da. Herein, we discuss cursory evidence of this applicability to a variety of analytes including both polar and nonpolar small molecules and novel peptides that mimic biomolecules upward of 1000 Da. Observed species are similar to ESI-derived ions including doubly charged analyte ions despite presumably different charging mechanisms. SAWN−APCI coupling may thus involve more nuanced ionization pathways in comparison to other ADI approaches.
Raman microspectra combine information on chemical composition of plant tissues with spatial information. The contributions from the building blocks of the cell walls in the Raman spectra of plant tissues can vary in the microscopic sub-structures of the tissue. Here, we discuss the analysis of 55 Raman maps of root, stem, and leaf tissues of Cucumis sativus, using different spectral contributions from cellulose and lignin in both univariate and multivariate imaging methods. Imaging based on hierarchical cluster analysis (HCA) and principal component analysis (PCA) indicates different substructures in the xylem cell walls of the different tissues. Using specific signals from the cell wall spectra, analysis of the whole set of different tissue sections based on the Raman images reveals differences in xylem tissue morphology. Due to the specifics of excitation of the Raman spectra in the visible wavelength range (532 nm), which is, e.g., in resonance with carotenoid species, effects of photobleaching and the possibility of exploiting depletion difference spectra for molecular characterization in Raman imaging of plants are discussed. The reported results provide both, specific information on the molecular composition of cucumber tissue Raman spectra, and general directions for future imaging studies in plant tissues.

Dislocations are the carriers of plastic deformation. As such, their characterization offers important information on the properties they affect. In this contribution, a new tool is presented, which is incorporated in Amira ZIB Edition and allows for three-dimensional (3D) imaging and quantification of dislocations substructures from thick regions an electron-transparent specimen. In the tool, the dislocation segments are traced on diffraction contrast images that are obtained in the transmission electron microscope (TEM). The uncertainties related to the experimental setup and to the proposed method are discussed on the base of a tilt series.
The transition from X-ray film to digital detectors in radiography is accompanied by an increase of unsharpness due to the larger inherent digital detector unsharpness in comparison to film. The basic spatial resolution of digital detectors (see EN ISO 17636-2) is used today to describe this unsharpness. The geometrical unsharpness of the radiographic projection of object structures onto the detector plane is determined by the focal spot size of the X-ray tube and the magnification. The focal spot size is measured today (see ASTM E 1165) from pin hole camera exposures or edge unsharpness (see ASTM E 2903). The final image unsharpness is a result of a convolution of the geometrical and inherent detector unsharpness function, divided by the magnification factor of the object onto the detector plane. Different approximations of this convolution result in ASTM E 1000 and ISO 17636-2 in different optimum values for the magnification factor for a given focal spot size of a X—ray tube and the basic spatial resolution of the detector. The higher contrast sensitivity, an advantage of digital radiography, compared to film radiography is furthermore improved when using higher X-ray voltages as used with film and smaller focal spots of the X-ray tubes. This allows a higher distance between object and detector resulting in reduced object scatter in the image. The interactions between all these parameters will be discussed and simple rules for practitioners will be derived in this contribution.
Digital radiographic images were analysed to predict the visibility of image quality indicators (IQI), based on normalized noise power spectra (NNPP) and modulation transfer function (MTF) measurements. The fixed pattern noise of some digital detectors result in different noise spectra, which influence the visibility of different IQIs, depending on the hole diameter. Studies, based on measurement of basic spatial resolution and contrast to noise ratio were performed together with presampled MTF measurements and the NNPS in dependence on the spatial frequency. Plate hole IQIs, step hole IQIs, and equivalent penetrameter sensitivity (EPS) IQIs based on ASTM E 746 were measured to verify the influence of the different parameters. Modelling of digital images was used to verify the applied numeric tools. A study has been performed for imaging plates and digital detector arrays to analyse differences. Formulas for the prediction of the visibility functions for hole type IQIs are derived. In consequence the standards for characterization and classification of computed radiography (ASTM E 2446) and radiography with DDAs (ASTM E 2597) need to be revised.
Investigation on Wall Thickness Ranges Using Digital Radiography for Tangential Projection Technique
(2018)
X-ray testing is based on the attenuation of X-rays when passing through matter. Image detectors acquire the X-ray information which is defined by the local penetrated wall thickness of the tested sample. By X-ray absorption in the detector and following read-out and digitization steps a digital image is generated. As detectors a radiographic film and film digitization, a storage phosphor imaging plate and a special Laser scanner (Computer Radiography - CR) or a digital detector array (DDA) can be used. The digital image in the computer can then be further analyzed using many types of image processing. In the presented work the automated evaluation of wall thickness profiles are investigated using a test steel pipe with 9 different wall thicknesses and various X-ray voltages and different filter materials at the tube port and intermediate between object and detector. In this way the influence of different radiation qualities on the accuracy of the automated wall thickness evaluation depending on the penetrated wall thickness of the steel pipe was investigated.

We describe a lattice-based crystallographic approximation for the analysis of distorted crystal structures via Electron Backscatter Diffraction (EBSD) in the scanning electron microscope. EBSD patterns are closely linked to local lattice parameter ratios via Kikuchi bands that indicate geometrical lattice plane projections. Based on the transformation properties of points and lines in the real projective plane, we can obtain continuous estimations of the local lattice distortion based on projectively transformed Kikuchi diffraction simulations for a reference structure. By quantitative image matching to a projective transformation model of the lattice distortion in the full solid angle of possible scattering directions, we enforce a crystallographically consistent approximation in the fitting procedure of distorted simulations to the experimentally observed diffraction patterns. As an application example, we map the locally varying tetragonality in martensite grains of steel.
What is the future of Analytical Sciences? The talk starts with a definition, comparing the current view with that from 1968. How do wie set trends? How do we get Analytics inside? Some examples of "Big Science" are given and discussed in relation to a definition of AS. How does AS face the current Grand Challenges?
As exaples for something significant, several exaples are presented, such as Climate Change of Hydrogen Storage. Another important trand are eScience and automation concepts for AS, which are highlighted.
But (Analytical) Science has to be politcal in our times to face Fake News and to breake barriers!
Ferritic high temperature alloys are widely used as boiler tube and heat exchanger materials in coal, biomass and co-fired power plants. All technologies have in common that the applied materials are exposed to different temperatures, process pressures and reactive atmospheres that lead to a change of the material properties and a further degradation of the material. Material changes caused by aging in highly corrosive and toxic gases such as SO2 are mainly studied ex situ after the reaction is finished. The solid material is deposited in the atmosphere for a certain period of time, and material changes are then examined by various microscopic techniques such as optical microscopy (OM), electron microprobe analysis (EMPA), scanning electron microscopy (SEM and TEM) and X-ray diffraction (XRD). Nevertheless, extensive efforts were made to study material changes of high temperature alloys under oxidizing and reducing atmospheres by environmental scanning electron microscopy or in situ TEM techniques However, the possibilities of microscopic in situ techniques are very limited for the use of highly corrosive and toxic gases such as SO2. Since Sulfur induced corrosion at temperatures relevant for coal and biomass fired power plants, which is causing breakaway oxidation and sulfide precipitation at grain boundaries, is still of scientific interest, the current work focuses on the effect of SO2 in an initial stage of corrosion of ferritic alloys. For the analysis of early stages of combined oxidation and sulphidation processes of Fe-Cr model alloys the usage of a light furnace to conduct a rapid reactive annealing experiment is feasible. Previous studies presented distinct results of the influence of chromium on early high temperature corrosion by SO2 by this technique and subsequent classical metallographic analyses. However, it is still not possible to trace the corrosion mechanism in real time by conducting single aging experiments. The current work introduces two different approaches to study the initial stages of high temperature oxidation processes by applying above state of the art X-ray diffraction and spectroscopy methods. One part focuses on the real time observation of the formation of corrosion products such as oxides and sulfides by energy dispersive X-ray diffraction (EDXRD). The potential of this technique to study crystallization and growth processes of thin films in a reactive environment in real time was previously shown for different compound semiconductors. This approach was now applied to follow oxidation and sulphidation processes of ferritic model alloys in SO2 and SO2/H2O environments. The diffraction signals of the X-rays were detected during the corrosion process and the peak area and positions were analyzed as a function of time. This procedure enables monitoring external oxide growth and material loss in real time in an early stage of corrosion.
The other part of the current work presents the possibilities of X-ray absorption near edge structure spectroscopy (XANES) to characterize oxide scales and their growth mechanisms. Precise phase identification and quantification of corrosion products in a multi-phase oxide/sulfide scale is a pre-requisite to understand diffusion paths of metal ions and gas components. It is a challenging task to distinguish structurally similar reaction products such as Fe3O4 and FeCr2O4 especially in thin films with texture effects by diffraction. To illustrate for example Cr-out diffusion of an alloy throughout an inner and external oxide scale the differentiation of Fe3O4 and FeCr2O4 is indispensable. XANES uses the photoionization effect at the metal absorption edge in an aging product and accesses by this structural and chemical information. The current work uses XANES at the Fe-K and Cr-K absorption edge to identify various aging products grown as thin layers on alloys after short time aging experiments. A reaction chamber for combining high temperature oxidation experiments with surface sensitive X-ray absorption near edge structure spectroscopy will be introduced and first results of XANES on scales at high temperatures will be presented.
Ferritic-martensitic alloys with 12-16 % Cr in weight are subject to devices for various energy systems, such as coal power plants and waste incineration plants. These materials are exposed to a highly corrosive environment which lead to a degradation of the material. Especially the simultaneous oxidation and sulfidation is thereby of special interest. Proper spatially resolved measurements that determine not only chemical compositions but phases are rare. However, precise phase identification and quantification of corrosion products within the multi-phase corrosion scales is a key aspect to understand diffusion paths of metal ions and gas ions/molecules. This study investigated Fe-Cr model alloys with Cr contents from 0 to 13 % in weight in 0.5 % SO2 and 99.5 % Ar atmosphere to aim in a fundamental and systematic analysis. Samples were aged at 650 °C for time scales from 12 h to 250 h. The results presented here correspond to depth dependent phase identification of oxide and sulphide phases in the corrosion scales by using X-ray absorption near edge spectroscopy (XANES). Per sample a series of ca. 20 spots (1-5 µm spot size) from scale-gas to scale-metal interface were measured. XANES spectroscopy was performed at the Fe-K edge (7.11 keV) on polished cross sections. The collected spectra were fitted to a combination of reference materials to quantify the present phases at different positions within the scale. The phase distribution differs with Cr content and the Cr diffusion through pure Fe-oxide and mixed Fe-Cr-oxide phases is discussed.
Up to now THz-TDS-systems aren’t considered to be nondestructive testing facilities for large scale industrial applications, despite it was proven that they provide a comprehensive set of quality parameters. A practical approach to bring THz-TDS in addition to already existing testing systems into the industrial mainstream is systematic development of future test procedures and test facilities for dielectrics. For this purpose polyeth-ylene test specimen with introduced artefacts were designed, to evaluate the detection sensitivity of Time of Flight measurements based on dielectrics. SAFT reconstructed tomograms are presented which visualize the sizes and location of artificially introduced flaws.
Increased speed, heavier loads, altered material and modern drive systems result in an increasing number of rail flaws. The appearance of these flaws also changes continually due to the rapid change in damage mechanisms of modern rolling stock. Hence, interpretation has become difficult when evaluating non-destructive rail testing results. Due to the changed interplay between detection methods and flaws, the recorded signals may result in unclassified types of rail flaws. Methods for automatic rail inspection (according to defect detection and classification) undergo continual development. Signal processing is a key technology to master the challenge of classification and maintain resolution and detection quality, independent of operation speed. The basic ideas of signal processing, based on the Glassy-Rail-Diagram for classification purposes, are presented herein. Examples for the detection of damages caused by rolling contact fatigue also are given, and synergetic effects of combined evaluation of diverse inspection methods are shown.
In the last twenty years, components made of fibre-reinforced plastic became a prominently used material in safety-relevant structures. Periodic in-service inspection of these structures using reliable non-destructive testing methods became a relevant issue in the field. The goal of the EMRP-funded project VITCEA (Validated Inspection Techniques for Composites in Energy Applications) aims on the evaluation of the performance of various non-destructive testing methods on the inspection of fibre-reinforced plastics.
In this talk the results of the comparison of different ultrasonic testing methods will be discussed. There are two challenges for the ultrasonic inspection of fibre-reinforced plastics. Firstly, different material properties of the fibre and the resin cause anisotropic acoustical behaviour of the material. Secondly, the physical layout of fibre layers leads to complex structures of fibre-reinforced plastic parts, and thus to a complex acoustical response. Both result in a reduction of the signal to noise ratio and make interpretation of measurement results rather extensive.
During the VITCEA project, the acoustical behaviour of fibre-reinforced plate materials has been simulated. Specimens with artificial flaws for the evaluation of the detection thresholds have been designed and manufactured.
Various mechanized ultrasonic testing methods including phased-array sensors, air-coupled transducers, immersion tank testing and contact technique have been evaluated on the specially designed specimen. Laboratory scale tests and a round robin test have been carried out. The probability of detection and the detection thresholds for each method have been estimated.
Compared to standard ultrasonic testing methods the application of phased arrays offers advantages and flexibility by the electronic steering possibilities to control the transmitted and received sound fields as well as the image based evaluation of the recorded scans. For more than one decade phased array systems have been introduced and used in the railway sector for the inspection of wheelsets in the workshops. Up to the present field applications of phased array systems in rail inspection are rather rare. On the other hand a lot of scientific work on rail testing using phased arrays has been carried out in the recent past.
Mechanized ultrasonic rail inspection poses strong challenges e.g. due to the harsh environment, complex flaw types and high inspection speeds. Different tasks have to be carried out by non-destructive testing ranging from manufacturing inspection of thermite welds to in-service inspection for operational induced flaws in the rail head, rail web and rail foot as well as rolling contact fatigue. This work will give an overview and categorization of different present approaches for rail testing using phased array.
Phased array systems typically offer digital image based inspection results. This make these systems become the perfect candidates to feed inspection data into a modern big data based workflow, using e.g. cloud computing, signal processing and data fusion for recording, positioning and tracking of flaws and artefacts in rails and to gain additional benefit.
Evaluation of promising innovative approaches and solutions for phased array based ultrasonic rail inspection in the frame of “Industry 4.0” will be discussed.
Increased use of high performance steel (HPS) for demanding applications calls for non-destructive ultrasonic testing methods which is covered by standards like SEP 1927 (1) and ASTM E588 (2). Here, an adaption of standards like increasing sensitivity and resolution of ultrasonic testing may be needed in order to improve determination of steel cleanliness.
Modern requirements on standardization in that field have been evaluated during the INCAFAT project “A novel approach for rating fatigue-initiating inclusions in highly demanding steel” which was funded by the European Commission.
Main results of this research show that an adapted standard should be open to different type of specimen due to the diversity in application. This includes a wide range from bars, rings and plates as well as different inspection sensitivities. An adapted standard should be open to the definition of the volume to be investigated and open to be selected by the user. Furthermore, for enhanced resolution the frequency range should be extended to higher frequencies. The use of focal beam probes shall be mandatory and probes should be selected according to the geometry of the specimen. Therefore, calibration procedures have to be improved significantly to allow proper sizing of indications. Indication height shall be directly transferred into an equivalent disc shaped reflector taking distance amplitude curves as well as signal amplitude distribution and signal travel time into account.
A new standard draft proposal taking all these requirements into account has been filed in the frame of the INCAFAT project and is now on discussion.
The presence of polycarboxylate ether (PCE) based superplasticizers (SPs) has an enormous influence on the early hydration of cement. The hydration is retarded and the timing of formation and the morphology of hydrates is affected. This short paper presents experimental results about the influence of delayed addition time of PCE SPs on hydration of cement and alite pastes, investigated by isothermal heat flow calorimetry. For cement as well as for alite pastes the hydration is retarded with SP, whereby the high charge PCE has a stronger retarding effect than the low charge PCE. The retardation caused by PCE is much more pronounced for alite than for cement mixes. If PCE is added later to the mix, the induction period is shortened and the hydration is accelerated compared to simultaneous addition. This applies for cement and alite pastes. With delayed PCE addition the alite shows a clearly less retarded setting and main hydration than after simultaneous addition. It is obvious that for alite pastes there is less retardation the later the addition of SP.

Nowadays, different nanoparticles have been developed for commercial applications. However, since data on toxicity are barely available, their increasing application in cosmetic products, food and their release in the environment might cause severe problems. An accurate separation, identification and characterization of nanoparticles becomes increasingly important1,2.
In this presentation, a two-dimensional separation approach based on AF4 and CE was showed and used to separate NPs with similar sizes but different coatings. Standard reference polystyrene NPs having comparable core sizes but different coatings were investigated. Different migration time and profiles were compared. Separation in either method resulted in non-baseline resolved or non-separated peaks. In contrast, two-dimensional coupling of AF4 and CE resulted in clearly separated regions in their 2 D plots in case of 20 and 50 nm particle mixtures, whereas the 100 nm NP mixture could not be separated. Various factors affecting the separation like hydrodynamic diameter or SDS concentration were discussed.
Future investigations will be focussed on inorganic NPs with differently charged coatings. There, AF4-CE coupling can be coupled with inductively coupled plasma mass spectrometry (ICP-MS) to enhance the sensitivity of this method.

However, since data on toxicity are barely available, their increasing application in cosmetic products, food and their release in the environment might cause severe problems. An accurate separation, identification and characterization of nanoparticles becomes increasingly important.
A common method for nanoparticle separation, which was introduced in 1976 by Giddings, represents asymmetrical field-flow fractionation (AF4). It is a flow based separation method, which can be theoretically used to separate particles range from 1 nm to 50 µm. However, when the particles are smaller than 10 nm, separation with AF4 will become difficult to perform. Because in this case strong separation force, which induces aggregation of particles, should be applied. This will decrease recoveries of analytes and limit its application in accurate quantitative analysis. Capillary electrophoresis (CE) is another well-developed separation technique, in which samples will be separated in relation to their electrophoretic mobility. In recent years, CE has been used to separate different kinds of nanoparticles like, gold colloids or CdSe Quantum dots. However, till now only separation of particles smaller than 50 nm was reported. Because large size distribution of bigger particles will result in strong peak broadening and long separation time.
A two-dimensional coupling of AF4 and CE might provide us a new separation method, which can extend the separation ranges of both methods and be a way to characterise particles with large size distributions.

The accumulation of plastics leaked into the environment achieve a high perception in the last years. It is uncontroversial that often an uncontrolled, high consumption of plastic products and a poor waste management resulted in plastic findings all over in the environment. However, less is known about the relevant pathways and fate of plastic in the environment. Especially plastic fragments smaller 5 mmm, so called microplastics, achieve a high attention, because their amount and risks arising from these fragments are still unknown. The basic mechanisms of plastic or more precise, of different polymer degradation processes are often not considered, resulting in unrealistic statements.
For generation of fundamental understanding of this problem we developed in the last years a fast thermoanalytical method for identification and mass quantification of microplastics in complex samples. The presentation will also give a short introduction about polymer degradation.
The fluorescence lifetime is a key property of fluorophores that can be utilized for microenvironment probing, analyte sensing, and multiplexing as well as barcoding applications. For the rational design of lifetime probes and barcodes, theoretical methods have been developed to enable the ab initio prediction of this parameter, which depends strongly on interactions with solvent molecules and other chemical species in the emitters' immediate environment. In this work, we investigate how a conductor-like screening model (COSMO) can account for variations in fluorescence lifetimes that are caused by such fluorophore−solvent interactions. Therefore, we calculate vibrationally broadened fluorescence spectra using the nuclear ensemble method to obtain distorted molecular geometries to sample the electronic transitions with time-dependent density functional theory (TDDFT). The influence of the solvent on fluorescence lifetimes is accounted for with COSMO. For example, for 4-hydroxythiazole fluorophore containing different heteroatoms and acidic and basic moieties in aprotic and protic solvents of varying polarity, this approach was compared to experimentally determined lifetimes in the same solvents. Our results demonstrate a good correlation between theoretically predicted and experimentally measured fluorescence lifetimes except for the polar solvents Ethanol and acetonitrile that can specifically interact with the heteroatoms and the carboxylic acid of the thiazole derivative.
Recent developments of photon counting and energy discriminating detectors for radiographic imaging
(2018)
The direct detection of X-ray photons into electrical signals is enabled by usage of highly absorbing photo conducting materials (e.g. CdTe) as detection layer of an underlying CMOS semiconductor X-ray image detector. If the read-out speed is high enough (ca. 50 - 100 ns dead time) single X-ray photons can be counted and their energy measured. Read-out noise and dark image correction can be diminished. By setting energy thresholds, selected energy ranges of the X-ray spectrum can be detected or suppressed. This allows material discrimination or reduction of scattered radiation, which results in an enhanced contrast sensitivity.
The presented development of a damping model is a research component of an experimental feasibility study about moisture in building materials measured with Bluetooth® Low Energy (BLE) signals. This study may be part of a structural health- and long-term monitoring aiming at early damage detection in the built infrastructure and is increasingly focusing on wireless sensor network technology. It is investigated, how the Received Signal Strength Indicator (RSSI) of a BLE signal, transmitted from the BLE-module embedded in building materials with changing moisture content is damped. The aim of the modelling is the derivation of a damping equation for the formal model to determine the moisture damping exponent to finding a correlation.

This presentation gives a short overview of isotope ratio measurements being carried out in the past few years at BAM in different fields such as plant metabolism, food web structures and archaeology. The corresponding isotope reference materials which have been certified at BAM in parallel are presented as well. Additionally an outlook is being provided on future iRM projects.
Hepcidin-25 concentrations measured by various methods differ considerably, complicating interpretation. Here, a previously identified plasma-based candidate secondary reference material (csRM) was modified into a serum-based two-leveled sRM. We validated its functionality to increase the equivalence between methods for international standardization. We applied technical procedures developed by the International Consortium for Harmonization of Clinical Laboratory Results. The sRM, consisting of lyophilized serum with cryolyoprotectant, appeared commutable among nine different measurement procedures using 16 native human serum samples in a first round robin (RR1). Harmonization potential of the sRM was simulated in RR1 and evaluated in practice in RR2 among 11 measurement procedures using three native human plasma samples. Comprehensive purity analysis of a candidate primary RM (cpRM) was performed by state-of-the-art procedures. The sRM was value assigned with an isotope dilution mass spectrometry-based candidate reference method calibrated using the certified pRM. The inter-assay CV without harmonization was 42.1% and 52.8% in RR1 and RR2, respectively. In RR1, simulation of harmonization with sRM resulted in an inter-assay CV of 11.0%, whereas in RR2 calibration with the material resulted in an inter-assay CV of 19.1%. Both the sRM and pRM passed international homogeneity criteria and showed long-term stability. We assigned values to the low (0.95 ± 0.11 nmol/L) and middle concentration (3.75 ± 0.17 nmol/L) calibrators of the sRM. Standardization of hepcidin is possible with our sRM, which value is assigned by a pRM. We propose the implementation of this material as an international calibrator for hepcidin-25.
Luminescence techniques are amongst the most commonly used analytical methods in life and material sciences due to their high sensitivity, nondestructive character, and easy instrumentation suitable for miniaturization. Photoluminescence signals are, however, affected by wavelength-, polarization-, and time-dependent instrument-related effects. Thus, at the core of standardization approaches for all fluorescence-based techniques are evaluated fluorescence standards for the consideration of instrument-specific spectral and intensity distortions of measured signals and for instrument performance validation (IPV).
Here, we summarize the research of BAM division 1.2 on development of liquid and solid fluorescence standards for various application-relevant fluorescence parameters and techniques. The portfolio of BAM fluorescence reference materials presently consists of:
i) a Spectral Fluorescence Standard Kit, i.e., a set of liquid fluorescence standards with certified normalized corrected emission spectra, for the determination of a broad variety of fluorescence parameters
ii) a ready-to-use, glass-based multi-emitter fluorescence standard for IPV and the determination of instrument-to-instrument variations
iii) specially adapted calibration tools and validation concepts for microarray-based platforms used in molecular diagnostics and food safety control
iv) quantum yield (Φf) standards for relative determination of the key performance parameter Φf of fluorescent materials, which can be also used for the evaluation of the performance of absolute, standard-free methods utilizing integrating sphere setups or spectrometer accessories. These materials, that will eventually cover the ultraviolet, visible, and near infrared spectral region, are currently under certification.
This toolbox of method-adapted reference materials can perfectly complement existing fluorescence standards. These easy-to-use, reference materials can pave the way to traceable fluorescence measurements to a radiometric scale like the spectral radiance or spectral photon radiance for all users of fluorescence techniques in material sciences and analysis, as well as environmental monitoring and biotechnology. They are particularly useful for customers working in strongly regulated areas like medical diagnostics or pharmaceutical research, where certified standards in conjunction with validated standard operating procedures are mandatory.
Data fusion for multi-sensor nondestructive detection of surface cracks in ferromagnetic materials
(2018)
Fatigue cracking is a dangerous and cost-intensive phenomenon that requires early detection. But at high test sensitivity, the abundance of false indications limits the reliability of conventional materials testing. This thesis exploits the diversity of physical principles that different nondestructive surface inspection methods offer, by applying data fusion techniques to increase the reliability of defect detection. The first main contribution are novel approaches for the fusion of NDT images. These surface scans are obtained from state-of-the-art inspection procedures in Eddy Current Testing, Thermal Testing and Magnetic Flux Leakage Testing. The implemented image fusion strategy demonstrates that simple algebraic fusion rules are sufficient for high performance, given adequate signal normalization. Data fusion reduces the rate of false positives is reduced by a factor of six over the best individual sensor at a 10 μm deep groove. Moreover, the utility of state-of-the-art image representations, like the Shearlet domain, are explored. However, the theoretical advantages of such directional transforms are not attained in practice with the given data. Nevertheless, the benefit of fusion over single-sensor inspection is confirmed a second time. Furthermore, this work proposes novel techniques for fusion at a high level of signal abstraction. A kernel-based approach is introduced to integrate spatially scattered detection hypotheses. This method explicitly deals with registration errors that are unavoidable in practice. Surface discontinuities as shallow as 30 μm are reliably found by fusion, whereas the best individual sensor requires depths of 40–50 μm for successful detection. The experiment is replicated on a similar second test specimen. Practical guidelines are given at the end of the thesis, and the need for a data sharing initiative is stressed to promote future research on this topic.

Invited for this month’s cover picture is the group of Dr. Knut Rurack at the Department of Analytical Chemistry; Reference Materials at the Bundesanstalt fuer Materialforschung und -pruefung (BAM) in Berlin (Germany). The cover picture shows how differences in color and fluorescence on a test strip can be easily read out with a mobile device. Two reference spots Frame the sensitive spot that indicates the presence of trace amounts of HgII below the threshold in a natural water sample. This dipstick contains a hybrid material that combines boron-dipyrromethene (BODIPY) probes sterically loaded into specifically tailored mesoporous silica particles, allowing for ultrasensitive HgII detection through enhanced fluorescence in a few seconds. The applicability in real water samples and fish extracts are also studied.
Type-I pyrethroids are frequently used for disinfection purposes against insects such as adult mosquitoes, or diseases carried by insects, like Malaria or Zika in cabins of airplanes on long-distance flights especially from tropical destinations. This treatment is mandatory at various airports but compliance with the rules is difficult to test for. Moreover, if improperly used, these compounds can entail negative health effects for crews and passengers.
The detection of the pyrethroids will be achieved thanks to an antibody-gated indicator delivery system (gAID) utilizing monoclonal antibodies and hybrid sensory nanoparticles. After the interaction of the pyrethroid with the gAID, the liberated indicator (dye) will be detected. Since only few analyte molecules are necessary for pore opening yet release a large number of dyes, the system shows intrinsic signal amplification.
The device system to be developed has to be so simple that chemically untrained personnel, such as ground or cabin crew, can use it and obtain a result in a reasonably short period of time, e.g., ≤5 min. The need for high accuracy and sufficient sensitivity, established at 0.001 g m–2, is a critical requirement and imposes another significant challenge since this value is beyond current LFTs reported in the literature for pesticide detection to date.
In order to achieve the selectivity and sensitivity required by the test itself, and to avoid cross reactivity with other type I pyrethroids, the production of a monoclonal antibody for both Permethrin and Phenontrin is necessary. The synthesis of the two hapten molecules and the subsequent immunization with different immunogens represent the first goal of the work.

The paper presents rheometric results and hydration curves for cementitious systems based on cement mixed with different aqueous phases at solid volume fraction of = 0.45. The varied aqueous phases were deionized water, limewater, the filter residue of a cement paste mixed at w/c = 2.0, and the filtrate of cement paste at solid volume fraction of 0.45. Each cementitious system provided different amounts and sizes of particles formed in the aqueous phase. The pastes were observed with and without polycarboxylate ether based superplasticizer. It was observed that the presence of particles causes higher PCE saturation dosages to achieve a minimum value, but the minimum value is only affected in the case of large particles. In all cases with and without PCE, the presence of small particles causes increased plastic viscosity. In addition, with decreased number and size of hydrates in the aqueous phase the hydration was retarded, which was specifically pronounced in the presence of PCE
Pulse and flash thermography are experimental techniques which are widely used in the field of non-destructive testing for materials characterization and defect detection. We recently showed that it is possible to determine quantitatively the thickness of semitransparent polymeric solids by fitting of results of an analytical model to experimental flash thermography data, for both transmission and reflection configuration. However, depending on the chosen experimental configuration, different effective optical absorption coefficients had to be used in the model to properly fit the respective experimental data, although the material was always the same. Here, we show that this effect can be explained by the wavelength dependency of the absorption coefficient of the sample material if a polychromatic light source, such as a flash lamp, is used. We present an extension of the analytical model to describe the decay of the heating irradiance by two instead of only one effective absorption coefficient, greatly extending its applicability. We show that using this extended model, the experimental results from both measurement configurations and for different sample thicknesses can be fitted by a single set of parameters. Additionally, the deviations between experimental and modeled surface temperatures are reduced compared to a single optimized effective absorption coefficient.
The Cairo Geniza is an “archive” discovered in the 19th century in Ben Ezra Synagogue in Fustat, a district in Old Cairo (Egypt). The collection of mostly Jewish documents that vary in genres, languages and writing supports contains a large number of early medieval Hebrew manuscripts.
The Geniza provides sources for the literary, linguistic, historical studies of the various aspects Jewish life. As the documents attest, at least two Jewish communities co-existed in Fustat up to 11th century: a so-called Babylonian and Palestinian. These communities had different leaders, different traditions and lived independently. The differences seem to manifest themselves also in the paleographical, codicological and some material properties of the manuscripts produced by each community.
The aim of this project is to compare the inks used in the Jewish documents depending on different variables: support (paper, parchment), purpose of the manuscript (legal, private, religious), provenance of the scribe. For the determination of the inks type and composition we had to choose non-invasive, non-destructive and portable techniques to analyze the corpus directly on-situ.
The analyses were carried out with a micro-X-ray spectrometer ArtTAX® (Bruker GmbH) and a digital stereomicroscope features built-in LED illumination at 395 nm and 940nm, Dino Lite.
We would like to stress that though the methods of material analysis listed above have been successfully employed in the field of cultural heritage and conservation including ancient and medieval manuscripts they have not yet been used to study fragments from the Cairo Genizah. Therefore, we believe that this research project is a pioneering study that will provide new insights into the history of Hebrew writing materials, their production techniques and materials and, thus, contribute new data to the field of Hebrew paleography.
We report a new method for top-down one-pot, gram-scale production of high-quality nanographene from graphite under mild conditions. The nanographene sheets had a very low oxygen content, comparable to that of nanographene grown by chemical vapor deposition. This method is easily scalable and comparatively mild, opening the door to an environmentally friendly process to produce high-quality nanographenes.
Nanographene sheets were further covalently functionalized using a non-destructive [2+1] cycloaddition reaction that left the π-conjugated system intact.
The Scaled Boundary Finite Element Method is known as an efficient method for the simulation of ultrasonic wave propagation. As to investigate acoustic wave behavior in case of fluid‐structure interaction, a fluid model is implemented in the SBFEM for prismatic structures. To omit coupling terms a displacement‐based formulation is used. Spurious modes, which occur in the solution, are suppressed using a penalty parameter. To verify this formulation dispersion curves obtained with Comsol Multiphysics are compared to results of SBFEM. The results of both methods are in very good agreement
The technique of painting on the reverse side of a glass panel was rediscovered by German artists at the beginning of the 20th century. They appreciated the impressive gloss, luminosity, and depth of colours in this genre. Compared to stained glass, the distinctive properties of this technique are: (1) the paint is applied “cold”, hence, it doesn’t involve a firing step, (2) reverse paintings on glass are framed and always viewed in reflected light and (3) the reverse paint stratigraphy is different from canvas paintings, starting with the front most layer and ending with the backing layer. In 1908 several artists, including Gabriele Münter, Wassily Kandinsky, Heinrich Campendonk, August Macke and Franz Marc of the “Der Blaue Reiter” (the Blue Rider) collective took an interest in this technique and started to share their interest with other colleagues in Europe. Our pioneering project is tracing this transfer of knowledge by a multidisciplinary approach in terms of art history, paint technology and material science. More than 100 artists and >1000 reverse paintings on glass were identified during the project. This high number of objects clearly points out that this technique was by far more important for modern art than previously assumed. In-situ, non-invasive measurements (XRF, Raman, VIS, DRIFTS) on a well-considered selection of 67 paintings reveal the broad palette of colorants ranging from traditional to experimental. Special attention is paid to the impact of synthetic organic pigments (SOP) in artists palette. Demonstrative examples by W. Kandinsky, L. G. Buchheim and F. Jespers are used to discuss analytical challenges and highlights.
Near-ambient pressure XPS makes it possible to characterise samples not compatible to ultra-high vacuum, and enables the study of liquid-solid, gas-liquid and gas-solid interfaces. NAP-XPS meas-urements of biofilms, suspended nanoparticles and metal-organic frameworks were performed with EnviroESCA developed by SPECS.
An interesting application is surface characterisation of biofilms, which are bacterial communities embedded in a self-produced polysaccharide matrix. Various model systems ranging from pure polysaccharides of alginate to biofilms harvested directly from the growth medium have been char-acterised in humid conditions[1].
NAP-XPS also makes it possible to characterise nanoparticles in solution. Silver nanoparticles in aqueous solution were characterised and the Ag 3d-spectrum compared to spectra obtained of dried nanoparticles in UHV-conditions[2]. The binding energy of the Ag 3d-core level peak was shifted by 0,6 eV towards higher binding energy for suspended nanoparticles compared to the dried sample measured in UHV. This can be assigned to a change in surface potential at the water-nanoparticle interface.
Metal-organic frameworks (MOFs) are suitable materials for gas storage of small molecules due to their nanoporous, crystalline structure. However, instability in humidity remains an issue for many types of MOFs. XPS-measurements of the MOF-structure HKUST-1 were performed in various NAP-conditions to assess the stability of the sample and its interaction with the gas molecules as water, methanol and pyridine.
Synthetic peptide pools are used in antigen-specific T-cell assays, which are an important part in vaccine and immunotherapeutic clinical trials. As the analytical characterization is challenging due to the similarity of the single peptides or is expensive due to isotope labeled standards, usually only a pre-characterization of the single peptides is performed. However, a regular quality control of the peptide mix would be highly desirable. Therefore, a cost-efficient high performance liquid chromatography-high resolution mass spectrometry (HPLC-HRMS) method for quality control of a model peptide pool is developed.
Peptides were synthesized using peptides&elephants proprietary libraries of individual peptides (LIPS) technology and purified by reversed-phase chromatography to > 90% each. The lyophilized single peptides were combined to a model peptide pool and analyzed by reversed-phase high-performance capillary liquid chromatography coupled to an orbitrap mass spectrometer. Separation was performed on a capillary reversed phase column (2 μm, ID x L 300 μm x 150 mm) with a linear gradient of acetonitrile + 0,05% trifluoroacetic acid.
For the separation of a model peptide pool the additive, additive concentration, the gradient elution and the temperature were optimized. Different quantification approaches were tested. Identification was performed by high resolution mass spectrometry in which extracted ion chromatograms (XIC) were used to confirm exact masses.
In line with the development of a new cost-efficient quality control method for the separation and identification of complex synthetic peptide pools, varied HPLC parameters highlighted their influence on chromatographic resolution and peptides were identified with high mass accuracy.
Most factors acting on concrete rheology work at an extremely small-scale level. Influencing factors in the millimetre or centimetre area are essentially restricted to sand and aggregates. The latter, however, make up 50 to 70% of the total volume of most concretes – a fact often ignored in research on controlling concrete processing properties.
Whereas suitably chosen concrete admixtures and additives can influence rheology in a very targeted manner, sand and aggregates are less suitable for controlling rheology but nonetheless contribute to the rheology of the Overall system. The actions of sand and aggregate can impose themselves upon the actions of admixtures and additives
and, in unfavourable circumstances, even render them redundant. For this reason, any results concerning the processability of binding agent systems can only be transferred to concrete with great care. It is important to better understand the action of sand and aggregates in order to be able to harmonise them in such a way that they complement the action of superplasticisers positively, instead of
working against them. Savings on costs can also be made by this targeted fine-tuning.
Temperaturindikatoren in mittelalterlichen Hochbrandgipsen: Eine Ramanmikrospektroskopische Studie
(2018)
Z02 is one of the three technically supporting projects at the Centre for the Study of Manuscript Cultures (CSMC). In collaboration with the other two service projects, Z01 and Z03, it aims at bridging the gap between humanities and natural sciences and technology. To that purpose, we set up a laboratory with a range of high-end instruments, most of them mobile, allowing thorough non-destructive analysis of manuscripts. In addition to working on constantly improving the laboratory and the methods of analysis, a substantial part of our activities is dedicated to service, by supporting different research projects conducted at the centre. In this talk, we will present our equipment and the possibilities offered by the different techniques available regarding the different kinds of missions: typology and classification of inks, provenance studies, recovery of faded inscriptions and palimpsests, reconstruction of the history of manuscripts, authentication and dating. We will give a brief overview of our past and ongoing activities in the frame of the second phase of the CSMC. Finally, a selection of projects will be presented in greater detail to highlight the possibilities of our laboratory and the diversity of missions which can be carried out.
The sensitive detection of the mycotoxin citrinin (CIT) utilizing ist fluorescence requires approaches to enhance the emission. In this respect, we studied the complexation of CIT and ochratoxin A (OTA) with Al3+ in methanol using absorption and fluorescence spectroscopy. In this context, an isocratic high performance liquid chromatography (HPLC) method using a polymer column and a fluorescence detector was also developed that enables the separation of the metal ion complexes from the free ligands and non-complexed Al3+. CIT and OTA showed distinct changes in their absorption and fluorescence properties upon Al3+-coordination, and the fluorescence of CIT was considerably enhanced. Analysis of the photometrically assessed titration of CIT and OTA with Al3+ using the Job plot method revealed 1:2 and 1:1 stoichiometries for the Al3+ complexes of CIT (Al:CIT) and OTA (Al:OTA), respectively. In the case of CIT, only one -diketone moiety participates in Al3+ coordination. These findings can be elegantly exploited for signal amplification and provide the base to reduce the limit of detection for CIT quantification by about an order of magnitude, as revealed by HPLC measurements using a fluorescence detector.
