The library of the remote monastery of Qäqäma Maryam Kidanä Mehrät in Däg'a Tämben (Tagray, North Ethiopia) was investigated by the team of the project Ethio-SPaRe in June and November 2012. A preliminary study of the library showed that the collection was extended and profoundly renovated in the nineteenth Century, in the course of the revival the monastery had during the reign of King Yohannas IV (1872-89). The shaping of the collection was apparently linked to the figure of the powerful abbot, Gäbrä Giyorgis, who donated dozens of books of diverse provenance, which he himself gathered or received as gifts.1 As the result, the extensive library contains a huge amount of information, but its heterogeneity poses a very serious challenge to the researcher.
A novel qPCR protocol to the specific detection and quantification of Hormoconis resinae in fuel systems

Introduction. Microbial contamination of fuels is a phenomenon widely reported in the literature causing dramatic problems in fuel systems such as blockage of pipelines and filters, and corrosion of their metal components. A variety of microorganisms, fungi, yeasts and bacteria, can contaminate the distillate fuels. Between them, the filamentous fungus Hormoconis resinae, previously described as Cladosporium resinae and also known as "kerosene fungus" or "diesel bug", has traditionally been considered the main responsible of such troubles due to its great capacity to produce biomass and biofilms.
Aim. The overarching goal of this study was to develop a novel real-time quantitative PCR (qPCR) method to detect and quantify the fungus H. resinae in samples from fuel systems.
Materials and methods. Four specific primer sets targeting on two molecular markers, RNA polymerase II second largest subunit (RPB2) and internal transcribed spacers of rDNA (ITS), were designed using Primer Blast. The specificity of primer sets was evaluated by both conventional PCR and qPCR, analyzing the DNA extracts from H. resinae strains and other fungi and yeasts previously described in fuels as well as four taxonomically closest related species. The efficiency and sensitivity of these protocols were assessed by constructing the corresponding standard curves for each primer sets.
Results.
Two primer sets, Hr556F/Hr814R (RPB2) and Hr101F/Hr408R (ITS), were selected according to their remarkable specificity. Their standard curves showed a good correlation coefficient and efficiency. Additionally, the selected primer pairs showed a high sensitivity detecting a tiny amount of H. resinae DNA, 10 pg for RPB2 primers and 0.1 pg for ITS primers.
Conclusions.
The established qPCR protocol is a reliable and useful tool to the specific detection and quantification of H. resinae. Its subsequent implementation to analyze samples of fuels or biofilms covering fuel systems will allow the early detection of fungal outbreaks in order to apply the appropriated control procedures. Moreover, these analyses will determine the current incidence of this species in the cases of biological fuel contamination.

Auch Kunststoffdränelemente sind wie alle Kunststoffprodukte für Kriechen und Stabilitätsversagen anfällig. Ein Kunststoffdränelement verformt sich unter langanhaltender Druck- und Scherbelastung allmählich. Dabei werden die Dicke und damit auch das Wasserableitvermögen reduziert. Wenn die Verformung parallel zur Ebene im Laufe der Zeit einen kritischen Wert überschreitet, kann es zum Scherversagen kommen. Das Kunststoffdränelement kann auch Zusammenbrechen, wenn die Dicke des Produktes einen kritischen Wert unterschreitet. Diese Effekte werden an Hand von Daten von Herstellern von vier verschiedenen Produkten beschrieben. Das Langzeit-Wasserableitvermögen und die akzeptable Grenzen für die Scher- und Normalspannung vor dem Hintergrund von Scherversagen und Dränkemstabilität und die Lebensdauer in Bezug auf diese Versagensmechanismen werden durch Zuhilfenahme standardisierter Prüfverfahren bestimmt. Diese Eigenschaften von Kunststoffdränelementen müssen bei der Planung dauerhafter geotechnischer Bauwerke berücksichtigt werden. Zudem wird kurz auf das Qualitätsmanagement bei dem Einbau und der Lagerung der Produkte auf der Deponiebaustelle eingegangen.
Odours might cause offensive nuisance and are under suspicion to be directly harmful to health. Surprisingly their assessment is by definition no part of the determination of indoor air quality (yet). As odours emit from building materials as well as from common consumer goods the users awareness increases continuously and demand for improved consumer protection raises. In comparison to well established methods for the determination of volatile organic compounds (VOC) in the air, there is little experience regarding the applicability, comparability and reproducibility of odour measurements. Considering this, the international and the German standardisation take into account odour assessment, e.g. ISO 16000-28 and VDI 4302-1.
In Germany the AgBB-Scheme is a mandatory tool for the evaluation of building products and it is intended to implement odour measurement into it. Initially and essentially the newly developed method had to be put to test involving institutes that deal with VOC measurements. To test the applicability of the method an interlaboratory test was conducted. The results and identified potential improvements are presented in this study (Brosig, 2013).

Die neue VDI Richtlinie 3925 stellt Bewertungswerkzeuge vor, die es erlauben, ökologische, ökonomische und inzwischen auch soziale Aspekte und Auswirkungen verschiedener Abfallbehandlungsverfahren miteinander zu vergleichen. Die Richtlinie verfolgt das Ziel, die Anwendung von geeigneten wissenschaftlich-technischen Bewertungsmethoden auf abfallwirtschaftliche Fragen zu beschreiben. The new VDI guideline 3925 presents assessment tools for the comparison of economic, ecological and also social aspects of different waste treatment processes. Objective of the guideline is to describe the application of suitable scientific and technical assessment methods to waste management strategies. The methods, their applicability, complexity, opportunities and limits are illustrated using practical examples.
