Here, we describe a fast and simple extraction method for the determination of per- and polyfluorinated alkyl substances (PFASs) utilizing extractable organic fluorine (EOF) sum parameter analysis and high resolution-continuum source-graphite furnace molecular absorption spectrometry (HR-CS-GFMAS) in soil samples. After extraction and separation of inorganic and organic fluorinated compounds, organically bound fluorinated compounds were indirectly determined via the molecular absorption of gallium mono-fluoride at 211.248 nm. The implementation of the decisions of the “Stockholm Convention on persistent organic pollutants” as well as the “Protocol on Persistent Organic Pollutants of the Convention on Long-Range Transboundary Air Pollution” of the UNECE include the reduction respectively the elimination of PFASs in the environment. Currently, regulations aim to target single compounds (mostly C8-PFAS). But the PFAS group includes over 4700 substances, which are potentially persistent and toxic, thus target analytical approaches are not suitable for a holistic approach investigating the PFAS pollutant situation. Furthermore, forbidden PFASs are substituted by short-chain PFASs, thus the number of unknow substances is steadily increasing. For this reason, sum parameter approaches are more suitable to investigate and assess the pollution situation as well as derive exposure limits. Our developed analytical method was successfully applied to determine PFASs in soil samples from a polluted site. In view of steadily increasing numbers of PFAS substances, our method will make an important contribution in assessing the pollution situation as well as support policy makers in deriving exposure limits for PFAS in the future.
Per- and polyfluoroalkyl substances (PFAS) have been used extensively in the past because of their inert chemical character and resistance to degradation by environmental influences. Since the beginning of their commercial use, PFAS have been widely exposed to the environment by application of PFAS in consumer products or as foaming agent in firefighting foams, thus several cases of contaminated soils sites have been reported. Since the number of known PFAS already exceeds 4700, their characterization and direct analysis is challenging given the current available techniques. Here, we introduce the novel fluorine (F) K-edge X-ray absorption near-edge structure (XANES) spectroscopy as a tool to analyze PFAS and inorganic fluorine compounds in contaminated soils and sewage sludges. While F K-edge bulk-XANES spectroscopy provide us information on the overall fluorine bonding in a sample micro X-ray fluorescence (XRF) in combination with F K-edge micro-XANES spectroscopy can also detect minor fluorine compounds and PFAS hotspots in investigated soils and sludges. Additionally, we used the combustion ion chromatography (CIC) to analyze the total amount of all PFAS as a sum parameter (extractable organic fluoride: EOF) in soils and sewage sludges. During combustion in the CIC, the PFAS in the sample get destroyed at temperatures of approx. 1000 °C and converted in inorganic fluorides that subsequently gets quantified by ion chromatography. Thus, for the first time we successfully combined F K-edge XANES spectroscopy and CIC as analytical tools to detect and quantify PFAS contaminants in soils and sewage sludges.
Rock-EATING FUNGI - Genetic Dive into the Biology of the Microcolonial Black Fungus Knufia petricola

Microcolonial black fungi are a group of ascomycetes that exhibit high stress tolerance, yeast‑like growth and constitutive melanin formation. They dominate a range of hostile natural and man‑made environments, from desert rocks and salterns to dishwashers, roofs and solar panels. Due to their slow growth and a lack of genetic tools, the underlying mechanisms of black fungi’s phenotypic traits have remained largely unexplored. We chose to address this gap by genetically engineering the rock‑inhabiting fungus Knufia petricola (Eurotiomycetes, Chaetothyriales), a species that exhibits all characteristics of black fungi. The state‑of‑the‑art genetic toolkit, together with the annotated genome sequence of strain A95, firmly established K. petricola as a model for exploring microcolonial black fungi.
Antimon (Sb) wird in Industrieprodukten hauptsächlich als Flammschutzmittel in Kunststoff verwendet. Aufgrund solcher Zusätze in Kunststoffen landet etwa die Hälfte von Sb am Ende der Lebensdauer in der Verbrennung fester Siedlungsabfälle und folglich in relevanten Mengen in der erzeugten Bodenasche. Bei Kontakt mit Wasser ist die anfängliche Auslaugbarkeit gering, da Antimonat mit Ca2+ schwerlösliche Verbindungen bilden. Nach der Carnonatisierung der Verbrennungsasche (IBA) während der fortschreitenden Alterung nimmt der pH-Wert in den Sickerwasser ab. Mit abnehmender Ca-Konzentration im Eluat ändert sich das Lösungsgleichgewicht und Antimonate lösen sich auf.
In Deutschland ist beabsichtigt, Sb in IBA mit der geplanten Umsetzung der sogenannten Mantelverordnung (MantelVO, die Bestimmungen zur Verwertung von Mineralabfällen enthält) in naher Zukunft zu regulieren. Die im Entwurf festgelegten Grenzwerte können für IBA kritisch sein und stellen daher ein Risiko für die Verwendung der Mineralfraktion von IBA in den gut etablierten Recyclingwegen dar.
Geokunststoffe werden im Wasserbau häufig eingesetzt, beispielsweise bei Maßnahmen für den Küstenschutz oder in Ballastschichten für Windenergieanlagen. Eine zunehmende Anwendung von Geokunststoffen wird aufgrund der Ausweitung der Offshore-Windenergieerzeugung und des Schutzes auf steigende Wasserspiegel und extremere Wetterbedingungen infolge des Klimawandels beobachtet. Die Anwendung von Geokunststoffen in Wasserbauprojekten bietet zwar verschiedene wirtschaftliche und technische Vorteile, wurde jedoch kürzlich in Frage gestellt, da sich diese Materialien während ihrer Lebensdauer verschlechtern und langfristig gefährliche Auswirkungen auf die aquatische Umwelt haben können, insbesondere als Ursprung von Kunststoffabfällen oder als Quelle / Senke für Chemikalien wie Weichmacher und Stabilisatoren, die bei der Herstellung von Geokunststoffen verwendet werden, um deren Leistung zu verbessern. Der Vortrag geht auf diese Bedenken ein.
A brief perspective of BAM on nanocarriers is presented including examples with special emphasis on the characterization of such materials and underlying challenges. In this respect, also ongoing activities at BAM on different types of core/shell nanomaterials and related systems are briefly summarized.
Formaldehyde is considered as carcinogenic and is emitted from particleboards and plywood used in toy manufacturing. Currently, the flask method is frequently used in Europe for market surveillance purposes to assess formaldehyde release from toys, but its concordance to levels measured in emission test chambers is poor. Surveillance laboratories are unable to afford laborious and expensive emission chamber testing to comply with a new amendment of the European Toy Directive; they need an alternative method that can provide reliable results. Therefore, the application of miniaturised emission test chambers was tested. Comparisons between a 1 m3 emission test chamber and 44 mL microchambers with two particleboards over 28 days and between a 24 L desiccator chamber and the microchambers with three puzzle samples over 10 days resulted in a correlation coefficient r2 of 0.834 for formaldehyde at steady state. The correlation between the results obtained in microchambers vs. flask showed a high variability over 10 samples (r2: 0.145), thereby demonstrating the error-proneness of the flask method in comparison to methods carried out under ambient parameters. An exposure assessment was also performed for three toy puzzles: indoor formaldehyde concentrations caused by puzzles were not negligible (up to 8 μg/m3), especially when more conservative exposure scenarios were considered.
Test chamber measurements are an important tool to improve indoor air quality and occupational safety. Test chamber measurements are possible for a wide range of materials, products and technologies. Determination of concerning contaminants is important to ensure good indoor air quality. The detection of concerning contaminants depends on the approriate sampling and analysis.
Recycled fertilizers produced using processes for the recovery of phosphate from residual materials such as wastewater, sewage sludge and sewage sludge ashes show very good bioavailability, but it is still a product largely unknown to the market. The aim of CLOOP is therefore to document the properties and effects of such fertilizers through chemical, mineralogical and ecological analyses as well as analytical method development and pot and field plant growth trials.
Outotec focused on 3 points in CLOOP: Experimental campaigns, process simulations as well as economic analyses with focus on the AshDec process, respectively the design of a large-scale AshDec plant for phosphorus recovery. The laboratory scale trials as well as the semi-industrial scale campaign provided valuable insights into the operating parameters of the process and the plant. As a result, for example, the temperature range could be significantly lowered, and the additive addition reduced by approx. 20 %. They have further shown that the AshDec process is not susceptible to operating fluctuations and that the product can be consistently produced at high quality. With the selection of suitable operating parameters, heavy metals (As, Pb, Cd, (Zn)) can be removed. Within the campaign, about 1.5 t of fertilizer for the plant trials in CLOOP could be produced. An AshDec plant process was digitally created in simulation software, allowing valuable process parameters to be simulated at various operating parameters. On this basis, a full-scale plant was designed. The data obtained in the project were used for a detailed economic analysis including a sensitivity analysis. It was possible to show under which conditions this plant can be operated economically.
At BAM, the AshDec fertilizer was synthesized with different additives and then applied to plant experiments at Uni Bonn. The goal hereby was to check differences in plant uptake. Because of the promising results of AshDec synthesized with sodium-carbonate and because this AshDec version does not require special off gas treatment for sulfur recovery (compared to AshDec synthesized with sodium-sulfate), all project partners agreed on continuing working with this AshDec variation. It was then used as a raw P-source for formulating it into NPK-fertilizers, by granulation with ammonium-sulfate and straw ash as potassium source. These recycling fertilizers were applied to plant- and field experiments by project partners in Brazil (University of Sao Paulo) and Australia (University of Queensland). Furthermore, in leaching experiments, the solubility of phosphorus in AshDec was compared to triple super phosphate. The experiments were carried out on soils with a varying phosphorus buffering index. Results show, that the phosphorus form in AshDec is way less soluble in water. This indicates that AshDec has the potential for a so-called next generation fertilizer – a fertilizer which’s nutrients remain in the soil and supply the plant according to its needs. At the moment, this behavior gets examined more in depth in lysimeter experiments in cooperation with University of Technology Berlin.
The focus of KWB is the Life Cycle Assessment (LCA) of different NextGen fertilizers to evaluate the entire process chain from recovery to fertilizer application. The NextGen fertilizer is credited by the amount of plant available nutrients in the product, which replace nutrients from conventional fertilizer. The LCA covers N-struvite precipitation from municipal wastewater, K-struvite precipitation from industrial wastewater and the AshDec-product from sewage sludge. The LCA task is almost complete. It could be shown that struvite precipitation has comprehensive environmental benefits, mainly since positive side effects occur in sewage sludge treatment (e.g. reduced sludge volume). In principle, the energetic and ecological profile of the AshDec process compared to direct use of sewage sludge ash cannot be assessed as being generally beneficial or negative. Regarding the global warming potential, the Ashdec process shows that the expenses (e.g. energy, chemicals) cannot be covered by the P fertilizer credit. In contrast, the impact categories “terrestrial acidification potential”, and “freshwater eutrophication potential” show positive results as the credits for conventional fertilizer are higher than the burdens for the process.
At University Bonn, the standardized pot experiments were conducted with several AshDec variations, using ryegrass, soybean, and spinach on a slightly acidic sandy soil and an organic-free standard substrate. P-uptake and biomass production of different AshDec variations were generally like those of triple super phosphate and struvite, and clearly outperformed untreated sewage sludge ash and rock phosphate. Field trials in Australia and Brazil with sugarcane on acidic soils are still ongoing and results are expected by the end of 2021.
Das Jahr 2020 war vor allen Dingen von der Pandemie geprägt, die alle Bereiche des Lebens und Arbeitens beeinflusst hat und immer noch beeinflusst. Dies hat sich auch auf verschiedene Maßnahmen im Zusammenhang mit zugelassenen Produkten ausgewirkt. In diesem Zusammenhang sollen die Punkte kurz aufgeführt werden. Des Weiteren soll aufgezeigt werden, wie bei der Bearbeitung der Richtlinien, der Zulassungstätigkeit und Beurteilung der Produkte an der BAM und im Fachbeirat vorgegangen wird. Darüber hinaus soll noch einmal kurz auf die Entlassung aus der Nachsorgephase eingegangen werden. Am Ende sollen verschiedene aktuelle Fragestellungen des Fachbeirat aufgezeigt werden.
This study shows that formaldehyde Emission results in micro-scale emission chambers are comparable with results in bigger chambers both for particle boards and wooden toys. On the contrary, the WKI flask method was leading to more variable results depending on sample geometry. Thus, microchambers could be used for reliable routine market surveillance. An exposure assessment led to noticeable formaldehyde indoor air concentration values, pointing out the need for an effective Surveillance of such samples.
New developments in legislation and technology development for P-recovery were presented with focus on the situation in Germany.
Innovation Network PerFluSan

Per- and polyfluorinated alkyl substances (PFAS) are transferred into the environment as part of their broad industrial use, where they pose a problem due to their critical properties. The sustainable protection of people and the environment from this group of substances is an important task for the future. However, available PFAS removal and remediation processes are expensive and often inadequate. PerFluSan would like to make a relevant contribution to solve this problem by developing new PFAS remediation methods for soils and water, as well as innovative analytical methods for PFAS detection.
The long-term vision is to develop PerFluSan into a national contact point for the PFAS topic, which uses synergies between the PerFluSan actors to address all aspects of the problem holistically, sustainably and effectively.
As one of the most energy and carbon consuming industries, steelmakers have an intrinsic interest in the development of technologies, reducing their carbon footprint as well as their carbon input. Besides generating new feedstocks from biomass or waste polymers, the valorization of carbon fiber reinforced polymer (CFRP) waste streams seems to be extremely promising based on the high carbon content of carbon fibers (CF), chars from CFRP and even unprocessed CFRP waste of >92%, 98% and 79%, respectively. Although, reduction of environmental impact from those high-performance materials was achieved by the development of smart recycling solutions, the continuous truncation by mechanical treatment of CF over multiple cycles, analogous to paper fibers, and the large global CFRP waste stream of 62 kt/a demand for a sustainable management of end-of-life (EOL) CFRP. Recently it was demonstrated that CF can be used safely as reductant in pyrometallurgical processes in a downhole electric furnace without significant WHO fiber emission. Therefore we anticipate a huge potential of CF based materials as sustainable reductant for steelmaking in an electric arc furnace (EAF). To facilitate the applicability of EOL CFRP in steelmaking, we investigated the interaction between CF based materials and liquid metal phases, appearing during the process, on different scales. Particularly, we examined the reactivity, wettability and dissolution behavior of CF and CFRP concerning liquid slag and steel. The presented results indicate that understanding the impact of the microstructure of CF on their behavior is crucial for their application in EAF steelmaking without risking potential hazards by WHO fiber emission. Besides the detailed elucidation of structure reactivity relationships of CF, the broader impact on circular economy will be presented.
In Bayern wird Klärschlamm aus der kommunalen Abwasserreinigung weitgehend verbrannt. Monoverbrennungsanlagen - ausschließlich für kommunalen Klärschlamm - gibt es in Altenstadt, München, Gendorf, Straubing, Neu-Ulm und zukünftig auch in Augsburg. Die Anlage in Altenstadt verbrennt jährlich ca. 55.000 t Klärschlamm-Trockensubstanz und produziert dabei rund 15.000 t phosphatreiche Aschen.
Die Asche aus Altenstadt wird zurzeit überwiegend direkt landwirtschaftlich genutzt. Aufgrund der niedrigen Schadstoffgehalte ist diese Nutzung nach den düngemittelrechtlichen Regelungen gesetzlich zulässig. Um der Intention der neuen Düngeverordnung (26. Mai 2017) nach hocheffizienten, d.h. gut pflanzenverfügbaren Düngemitteln zu entsprechen, ist eine Aufbereitung der Klärschlammasche geplant. Im Rahmen des R-Rhenania Projektes errichten der Betreiber der Klärschlammverbrennungsanlage Altenstadt in Bayern Emter GmbH - und der Düngemittelhersteller sePura GmbH gemeinsam eine AshDec®-Demonstrationsanlage, die im Jahr 2023 den Betrieb aufnehmen soll. Das angewandte AshDec®-Verfahren schließt die Phosphate in der Klärschlammasche thermisch im Drehrohrofen auf und macht diese für Pflanzen vollständig verfügbar. Gleichzeitig werden Schadstoffe wie Arsen, Blei und Cadmium entfernt. Den thermischen Aufschluss kennt man bereits vom erfolgreichen, aber nicht mehr produzierten Düngemittel „Rhenania-Phosphat“, das auf der Basis von Rohphosphaten hergestellt wurde.
Die geplante Anlage wird neben den Aschen aus Altenstadt zusätzlich die Aschen aus weiteren bayerischen Verbrennungsanlagen verwerten und für eine Kapazität von 30.000 Jahrestonnen Asche ausgelegt sein. Der Projektpartner sePura plant den produzierten Dünger vollständig in Bayern zu verwerten. Dies verringert die Umweltbelastung durch lange Transportwege und fördert die Regionalität des Vorhabens.
Bei der Stahlproduktion fallen verfahrensbedingt große Mengen an Nebenprodukten und Abfällen wie Stahlwerksschlacken, Filterstäube und Gichtgasschlämme an. Hohe Anteile dieser Stoffströme werden aufgrund ihrer Zusammensetzung immer noch deponiert, obwohl sie ein hohes Wertstoffpotential aufweisen. An der Bundesanstalt für Materialforschung und -prüfung (BAM) werden Verfahren untersucht und entwickelt, welche eine Verwertung und Kreislaufführung dieser Materialströme ermöglichen. Aus sogenannten Linz-Donawitz-(LD)-Schlacken können durch carbothermische Nachbehandlung Roheisen und ein dem Portlandzementklinker (PZK) ähnliches Bindemittel erzeugt werden. Aus Elektroofenstäuben und Gichtgasschlämmen kann Zink gewonnen und die Anteile an Eisen und Kohlenstoff in den Stahlproduktionsprozess zurückgeführt werden. Durch diese technischen Maßnahmen kann es gelingen die Deponierung von Abfällen der Stahlproduktion zukünftig deutlich zu reduzieren.
In recent times, as the need for new high-tech materials is growing steadily to fulfil future sustainability goals, more and more looks turn towards industrial waste streams as potential sources of valuable metals. Bauxite residue (BR), accumulating during alumina production, is one of the major waste streams in the metal industry. Suitable re-use possibilities for BR are intensively investigated including approaches for the recovery of scandium (Sc). This transition metal, even though rarely recognized by the broader audience yet, has recently gained the interest of e.g. aerospace industries and SOFC-producers since its use is strongly improving the performance and durability of the respective materials.
Even though many sources classify Sc as one of the REE, there are many aspects, such as its ionic radius, which significantly differentiate Sc from the REE and cause its behaviour to be distinct as well. The large ionic potential (radius/charge) causes Sc to be incompatible in most rock forming minerals, which results in generally low concentrations in the earth’s crust and rare occurrences of natural Sc-deposits.
The general occurrence of enriched concentrations of Sc in some BRs compared to the earth’s crust is known for some time now and has especially been investigated for Greek materials. However, the (geo)chemical and mineralogical behaviour, the Sc speciation, the association and the variation of Sc occurrences in BRs of different origin is still not fully understood. This study aims to gain better knowledge on the different kinds of Sc-occurrences and their possible impact on future recovery schemes. We therefore apply a diverse set of analytical methods including novel measurement techniques such as X-ray absorption near edge structure (XANES) spectroscopy as well as standard techniques such as electron microprobe analyses, and Raman spectroscopy on BRs from Germany, Hungary and Greece. Information on the local distribution of Sc is determined by high-resolution laser ablation – inductively coupled plasma – mass spectrometry (LA-ICP-MS) mappings.
Sc in Greek BR seems to have an affinity towards Fe-Phases such as Goethite and Hematite. However, whereas some of these phases show distinct enrichment of Sc, others are particularly barren, which might be a result of the different input materials. By means of XANES measurements it was possible to show that Sc in the Greek samples occurs adsorbed on the surface of those Fe-Phases, likely in the form of amorphous Sc-hydroxide or -oxyhydroxide or can be incorporated into the crystal lattice as well. The general affinity of Sc to those kinds of phases has been reported for natural rocks such as laterites before, where similar investigations were made using the XANES method [8]. Likewise, the Fe-association has been observed for Greek BR by Vind et al. [7]. Hungarian BRs show similar results as the Greek samples and LA-ICP-MS mappings show distinct Sc enrichment rims surrounding mineral particles. In German BR, Fe as well as Ti-phases occur to be partially enriched in Sc. Depending on the original primary bauxite, the source rock of the bauxite and the bauxite processing route, it is likely that many different parameters influence the occurrence and species of Sc in the BR. Theoretically, it either can therefore remain within its original carrier or can be partially or fully redistributed within the BR. Since the processing of the bauxite involves elevated temperatures, pressure and dissolving reagents, it is however very likely that some of the Sc will be redistributed and adsorb on mineral surfaces of smaller and larger particles, making beneficiation steps rather ineffective.
The study shows that there is no general rule defining how Sc is associated mineralogically or chemically within BRs. Therefore, the effective development of a recovery method needs a case specific background knowledge on the Sc-species present in the BR of interest. This research has received funding from the European Community’s Horizon 2020 Programme SCALE (H2020/2014-2020) under grant agreement n° 730105.
Microbiologically influenced corrosion (MIC) is a highly unpredictable process involving a complex group of microorganisms, including sulfate-reducing bacteria and methanogens. Standard laboratory MIC testing using static serum bottle enrichments is an easy but limited method, offering poor resolution on the biomineralization process of corrosion products. An example of this is the presumed corrosion product siderite by corrosive methanogens (Mi-MIC). Previous publications reported siderite was the sole corrosion product of M. maripaludis using metal coupons incubated under stationary conditions.
However, the formation of siderite is closely related to the surrounding environmental conditions, i.e. pH, CO2 concentration, flow and temperature. Thus, siderite as the sole corrosion product of Mi-MIC remain inconclusive and questionable. To study Mi-MIC effectively, a novel versatile multiport flow-column corrosion monitoring system (MFC) was developed. MFC allows sectional corrosion rate determination under flow conditions using different types of material, inoculum and packing material. MFC offers great flexibility, ease of operation and accurate corrosion measurements that can be combined with many other techniques. Using MFC, we studied multiple strains of methanogens and compared it with sulfate-reducing bacteria under neutral and low pH conditions. It was revealed by MFC that corrosive methanogens have equally high corrosion potential as sulfate-reducing bacteria. Additionally, siderite is not the dominant nor sole corrosion product of Mi-MIC. Thus, effective corrosion monitoring and establishing standard laboratory practices, i.e. incorporating MFC as part of regular testing process, will provide deeper understanding of MIC. This will allow further microbial electrophysiology understandings, contributing to effective mitigation strategy development.
The intention of the presentation is to inspire a discussion on the needs and challenges for the digitalisation of SOPs. SOPs are available for the disciplines of physical-chemical characterisation, for toxicology, for environmental applications and for exposure applications. Furthermore the SOPs need to be integrated in the regulatory framework and need to be detailed enough for the digital processing. This is highlighted in this presentation.
Designing the performance of geogrids in reinforce soil constructions usually does not consider long-term behavior and possible failure modes of junctions between longitudinal and transverse ribs. How could behavior of junctions be included? To which extend is it necessary? On the conference Geo-Chicago 2016, Swan and Yuan presented an ultimate limit state equation for the short-term material resistance of junctions. We discuss limitations and suggest improvements to include long-term behavior of junctions. Nevertheless, the approach applies only to a specific mode of shear-tensile failure of junctions and only to strictly rigid geogrids. A new design concept presented in the same year by Jacobs on the conference GeoAmerica for the special case of the anchorage of geogrids, which reinforce capping system on long and steep slopes of landfills, tried to overcome these drawbacks. We discuss the aspects of this concept related to the long-term behavior of junctions and the interplay between the load applied to junctions and the flexibility of longitudinal ribs. This interplay and the long-term junction strength determine the long-term behavior of geogrids.
Museums worldwide are equipped with different display cases. Exhibit display cases should protect cultural objects from dust as well as from mechanical and physical damage. To ensure a stable climate inside the display cases, a low air exchange rate is maintained. Typically, air exchange rates are often smaller than 0.1 d-1, which can result in rising concentrations of potential harmful VOC inside of the display cases due to emissions from materials. Especially organic acids, e. g. formic or acetic acid which can emit from e.g. sealing materials or wood-based materials, can produce damage of cultural objects.
In 2012 BAM introduced a procedure which is called: BEMMA-Scheme (Bewertung von Emissionen aus Materialien für Museums¬ausstattungen) which stands for: “Assessment of Emissions from Materials for Museum Equipment”. Regarding the testing procedure only construction materials were evaluated, not the display cases their self. Micro chambers are used for VOC emission tests of display case construction materials, e.g. plastics, sealing materials, coatings, textiles and others. Each sampling procedure is carried out in duplicate. Emissions like formic acid, acetic acid, formaldehyde, piperidine-derivates and oximes are excluded and the sum of emissions of VVOCs, VOCs and SVOCs is limited. For a positive assessment all listed criteria must be fulfilled; otherwise the display construction material fails the BEMMA scheme.
Biocides are in contact with soils through direct application and passive leaching from protected materials. Consequently, soil microorganisms are exposed to biocides even though they are not their primary targets. Soil is a large reservoir of microbial diversity and has been hypothesized to be a crucial factor for the evolution and spread of antimicrobial resistance.
Currently, there is little knowledge on how biocides used to protect materials affect the evolution and spread of resistance. Thus, our aim is to investigate the risk for the evolution of biocide resistance and cross-resistance to antibiotics. In addition, we aim to elucidate the affect of biocides on the spread of resistance via horizontal gene transfer (HGT).
In adaptive laboratory evolution experiments we cultured selected model soil microorganism with representative biocides under selection regimes with increasing and stable biocide concentrations followed by antibiotic and biocide cross-resistance determination. Moreover, we investigate if the selected biocides affect the rates of de novo mutations and HGT of plasmids that carry resistance genes among soil microorganism.
Our results show only small increases of biocide resistance during serial transfers under increasing biocide concentrations. One reason for this might be the narrow selective window for biocide resistance due to steep dose-response relationships. Furthermore, our results indicate that a stable low-level biocide regime did not select for high level cross-resistance to antibiotics and other biocides.
Moreover, material preservatives affected the rates of HGT via conjugation and the mutation rates at sub-inhibitory concentrations. The results will enable future risk assessment regarding resistance evolution for biocides used as material preservatives.
Dem Fachbeirat Bodenuntersuchungen des BMBF/UBA wurden die abschließenden Ergebnisse zum UFOPLAN-Forschungsvorhaben „Monitoring zur Verwertung von Bodenmaterial“ vorgestellt. Die potentielle Auswirkung der neuen Regularien in der geplanten Novelle der BBodSchV (4. Referentenentwurf der Mantelverordnung) auf die Erweiterung der Verwertungsmöglichkeiten von Bodenmaterialien wurden aufgezeigt.
After their discovery on rock surfaces in cold and hot deserts, a polyphyletic group of ascomycetous black fungi was found to dominate a range of hostile environments – natural and man-made, from salterns to dishwashers, roofs and solar panels. Together with bacteria and algae they may establish subaerial biofilms and cause weathering of the surfaces they grow on. Their impressive survival abilities as well as their constitutive protective pigmentation and cluster-like microcolony organisation are similar in environmental isolates as well as in heat-tolerant opportunistic pathogens of animals and humans. We selected the rock-inhabiting fungus Knufia petricola (class Eurotiomycetes, order Chaetothyriales) that exhibits all the characteristics of microcolonial black fungi such as yeast-like cell growth, absence of reproductive structures and constitutive dihydroxynaphthalene (DHN) melanogenesis (Nai et al. 2013, Fungal Genet Biol). We developed protocols to efficiently generate and transform protoplasts resulting in stable homokaryotic transformants by targeting genes involved in pigment synthesis. The differences between the wild-type fungus and its melanin-deficient mutants were tested in geomicrobiological experiments and measured are now discussed in detail, with special accent on the possible effects of the mutation on EPS and other exuded substances.

An Fallbeispielen aus der gutachterlichen Praxis und Forschungsprojekten der Vergangenheit sollen die Möglichkeiten der Papier- und Wasserzeichenforschung bei Datierungen und Authentizitätsprüfungen demonstriert werden.
Im Jahr 2019 jährt sich die Inbetriebnahme der ersten Papiermaschine in Deutschland zum 200. Male. Mit dem Vortrag wird eine Einführung in die Technologie der Maschinenpapierherstellung, samt entwicklungsgeschichtlicher Meilensteine gegeben. Es gilt die Produktvielfalt von Papier und die Gründe für die enormen Produktionssteigerungen bei der Papierherstellung aufzuzeigen.
Wasserzeichen und Papierstrukturen können einen wesentlichen Beitrag bei der Analyse von Kunstobjekten auf Papier darstellen. Ausgangspunkt hierfür bildet die korrekte Dokumentation mit entsprechender Bildaufnahme und der richtigen Erfassung und Interpretation der Phänomene. An Fallbeispielen aus Forschungsprojekten zu niederländischen Zeichnungen am Kupferstich-Kabinett Dresden und der Grafischen Sammlung Weimar sollen die Möglichkeiten der Papier- und Wasserzeichenforschung als historische Hilfswissenschaft demonstriert werden. Dieser Abendvortrag dient zugleich als Einführung in die Technologie der (Hand-)Papierherstellung, Wasserzeichenerfassung und –recherche.
Dihydroxynaphthalene (DHN) melanin is produced by different Ascomycetes via slightly differing biosynthetic routes. The polyketide synthases (PKS) release the heptaketide YWA1, the hexaketide AT4HN or the pentaketide T4HN. The first two products are deacetylated by ‘yellowish-green’ hydrolases to T4HN, and T4HN is further converted by a core set of enzymes to DHN. Final polymerization steps are accomplished by multicopper oxidases. DHN melanogenesis is often regulated in a spatial and temporal fashion resulting e.g. in melanized reproduction, survival and/or infection structures. Remarkable is the regulation of the DHN melanogenesis in the foliar plant pathogen Botrytis cinerea: it involves two differently expressed PKSs providing the precursor in conidia and sclerotia, respectively (Schumacher 2016, Mol Microbiol). In contrast, a polyphyletic group of Ascomycetes (microcolonial fungi/ black yeast) dwelling in hostile habitats such as bare rock surfaces in hot and cold deserts, exhibits constitutive melanogenesis. Here, DHN melanin builds a protective layer around all vegetative cells thus contributing to the survival of diverse environmental stresses even without specialized reproduction structures. As part of our continuing research on microcolonial rock-inhabiting fungi, we chose the genetically amenable Knufia petricola strain A95 (Nai et al. 2013, Fungal Genet Biol; Noack-Schönmann et al. 2014, AMB Express) for detailed studies. DHN-deficient mutants generated by targeted mutation of biosynthetic genes were studied with regard to the architecture of the cell wall and the EPS (extracellular polymeric substances) matrix, attachment to and weathering of olivine, as well as the tolerance to abiotic and biotic stresses. We will discuss the critical role of the outer cell surface (DHN melanin and EPS) in adhesion to the substrate and subsequent damage of the colonized surface.
Microcolonial black fungi, a polyphyletic group of ascomycetes, exhibit constitutive melanin formation, yeast-like growth and high stress tolerances. They dominate – often together with bacteria and algae in sub-aerial biofilms – a range of hostile environments including natural and man-made ones, from salterns to dishwashers, roofs and solar panels. Because of lacking genetic tools and the slow growth of most isolates, the genetic bases for these specific properties are largely unknown. The rock-inhabiting fungus Knufia petricola (Eurotiomycetes, Chaetothyriales) exhibits all characteristics of microcolonial black fungi and was selected as recipient for genetic engineering to study gene functions and genetic interactions. Different variants of green and red fluorescent proteins were successfully expressed indicating that fluorescence microscopy using genetically encoded fluorescent proteins and fluorescent dyes enables various cell biology approaches. Furthermore, genes of biosynthetic pathways (DHN melanin, carotenoids, uracil, adenine) were successfully mutated by applying traditional gene replacement and plasmid-based or ribonucleoprotein (RNP)-based CRISPR/Cas9 or silenced by RNA interference (RNAi). The availability of this advanced and efficient genetic toolbox and the annotated genome sequence of strain A95 makes K. petricola an excellent model for exploring the secrets of microcolonial black fungi.

So wurde in einer Sondersitzung des Fachbeirats über die Entlassung aus der Nachsorgephase beim Einsatz von Geokunststoffprodukten in Böschungen diskutiert. Die Unterarbeitsgruppe Fremdüberwachung in der Produktion hat getagt. Darüber hinaus werden Änderungen in den Zulassungsrichtlinien für Kunststoffdichtungsbahnen, Kunststoff-Dränelemente und für die Fremdüberwachung kurz vorgestellt.
Scientific analysis based on spectroscopic methods provide essential information on the composition of colourants and binders in paintings. These results can be set in a historical context and help to confirm art historical interpretations. Proofs of certain pigments can be used for dating purposes and may reveal if the artist used not only local but also imported materials. A pilot study of two Chinese reverse glass paintings from the late 19th (Yingying and Hongniang) and early 20th centuries (The Archer) was performed using a multi-analytical approach including X-ray fluorescence (XRF), diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS) and Raman spectroscopy. This approach allowed the identification of the pigments and the classification of the binding media. The results reveal a combined use of traditional Chinese and imported European materials. Several pigments like cinnabar, lead white, orpiment, carbon black and copper-arsenic green (probably emerald green) were found in both paintings; red lead, artificial ultramarine blue, Prussian blue and ochre appear in at least one of the paintings. The proof of limewash (calcite and small amounts of portlandite) as a backing layer in “Yingying and Hongniang” indicates that clamshell white was also used for reverse glass paintings. Drying oil was classified as a binding media in most areas of both paintings. However, the orange background of The Archer yielded prominent bands of both proteinaceous and fatty binder.
The aim of the lecture is to present the productive interplay of connoisseurs and material analysis using the example of selected drawings from the Rembrandt collection of the Klassik Stiftung Weimar. We try to determine the point at which the question of the materiality of the drawings makes sense in the traditional discourse of connoisseurship. On the one hand, this involves the question of authorship – Rembrandt – or not? – and the locating of attributed drawings in the context of the work. On the other hand, it is about making drawing processes visible, characterizing ways of drawing and workshop practices as well as distinguishing different forms of reworking and copying within the Rembrandt workshop. The material analyses culminate in the question of whether Rembrandt’s inks can be distinguished at all.
It becomes apparent that the newly gained knowledge about the materiality of the drawing no longer reveals itself to the “mere” view of the connoisseur. Rather, it is only generated by the digital image. The rhetoric of connoisseurs must therefore be contrasted with the necessity of a hermeneutics of the digital image. The envisaged “material turn” of drawing science is thus simultaneously able to take up the critique of the question of authorship, which has been growing ever more recently, and to transfer it to a more comprehensive perspective of aesthetics of production.
Es wurden erste Ergebnisse des Projekts extraWERT vorgestellt. Im Projekt werden monatlich Klärschlammproben von 10 kommunalen Kläranlagen untersucht, deren P-Gehalt im Grenzbereich der AbfKlärV liegt zwischen 1,7 und 2,3%. Die ersten Ergebnisse lagen zu fünf Kläranlagen vor, es wurden Daten zum TS-Gehalt, zur oTS und zu den Nährstoffgehalten präsentiert.
The Weimar research project has set the goal of placing style-critical research on Old Master drawings on a new, methodologically proven foundation. The starting point is the stock of Dutch drawings possessed by the Klassik Stiftung Weimar, whose extent (ca. 1,400 items) and character make it one of the most significant outside the Netherlands. The focus here is on drawings that were executed with red chalk. This drawing material has different functions - one of which is the use as a transmission material. The methodological approach is double: first, the scientific indexing of this stock. The foundations for this, along with classic analysis of style, are innovative material-scientific methods of investigation.
Interface between the atmosphere and mineral substrates is the oldest terrestrial habitat. Morphologically simple microbial biofilms were the first settlers on these inhospitable surfaces at times when the Earth was inhabited only by microorganisms and the solid substrates represented only by natural rock surfaces i.e. lithosphere. Miniature, self-sufficient microbial ecosystems continue to develop on subaerial (i.e. air-exposed) solid surfaces at all altitudes and latitudes where direct contact with the atmosphere and solar radiation occurs – on rocks, mountains, buildings, monuments, solar panels. All these sub-aerial biofilms develop under fluctuating and hostile conditions – and thus frequently harbour stress-tolerant black fungi inherently able to cope with the stresses of bright sunlight and constantly changing atmospheric conditions. Black fungi – a polyphyletic group of Ascomycetes– accumulate the dark pigment DHN melanin, diverse carotenoids and mycosporines in their cells and thus successfully colonise sunlight-flooded habitats from phyllosphere to rock surfaces. Various chemical and physical extremes and fluctuating environments belong to the challenges effectively mastered by black fungi. In our laboratory we isolate novel black fungi from man-made habitats like building materials and solar panels. Using Knufia petricola A95 as a model we conduct experiments to clarify interactions of black fungi with inorganic substrates. We use available mutants to determine the functional consequences of changes in the outer cell wall envelopes – from excreted EPS to layers of protective pigments. A genetic toolbox to manipulate this Chaetothyriales representative is in further development. Our long-term goal is to understand the fundamental mechanisms how black fungi are able (i) to adhere to dry atmosphere-exposed surfaces, (ii) to survive multiple stresses and (iii) to change the underlying substrates including rocks.
In the modern world there is an increased understanding that design and performance monitoring of materials have to be tested in connection to chemical, physical and (micro)biological challenges. A systematic study on how biofilms interact with materials and what could be done to engineer biofilms and/or materials in order to maximize the resistance of the material (surface) or the resistance the biofilm-modified material (bulk) is in strong need. In the Department “Materials and the Environment” of the BAM new experimental platform is being developed. With the help of different type of device for high throughput and microbiologically-controlled environment simulation we establish a new approach to clarify the mechanisms of biofilm/material interactions. Despite the focus on fundamental research, the main results of this project proposal will be transferable into material technology and construction chemistry and will influence the development of standardization in this topic. As the interactions of biofilms and materials have implications for most constructions as well as climate change, the results of the research generates additional value.
In the 15th century, Persian manuscripts underwent significant and lasting changes. Possibly as a response to the import of tinted and gold-decorated paper from China, which was used for manuscript production in Greater Iran, Persian papers were also tinted and embellished with gold-decorated and marbled margins or frames. In a joint project between CSMC and the University of St Andrews, and in co-operation with the Golestan Palace Museum, we aim to analyse papers and their decorations in manuscripts in the holdings of the Golestan Palace Library. The analyses aim to determine the consistencies of papers, pigments and metals and the techniques used for their production and application. The support will be first described in its codicological characteristics such as dimensions, thickness, colour (expressed in colour coordinates), pattern left from the sieve (observed in transmitted light). The plant used as raw material in paper production will be studied by fibres analysis, while the sizing agents, the pigments, dyes and metals will be analysed by the application of several methods: in particular Reflectography, X-Ray Fluorescence and InfraRed, Raman and Visible Reflectance Spectroscopy. The results of the analyses will be published in an online database that will form an important resource for the material aspects of Persian manuscripts and be a starting point for establishing a first chronology of the materials and techniques used in Persian manuscript production.
This talk will be divided into two parts. In the first part I will present the analytical techniques and the equipment used by the Centre for the Study of Manuscript Cultures that can be applied for the study of decorated papers. The analyses aim to determine the constituent materials of papers, pigments, dyes and metals and the techniques used for their production. I will focus in particular on the kind of information these techniques allow us to gather: through optical microscopy we conduct fibre analysis in order to identify the plant used as raw material in paper production while Reflectography, X-Ray Fluorescence and InfraRed, Raman and Visible Reflectance Spectroscopy are used to study the sizing agents, the pigments, dyes and metals. In the second part I will show some results obtained by the application of these techniques on a few Persianate manuscripts, dating between the 15th and 17th centuries, belonging to the corpus of Ilse Sturkenboom.
Ink recipes from the islamic era: Texts, manuscripts, ink reproductions and scientific analyses

Ink recipes can often be found in Arabic manuscripts. They may be included in treatises of diverse subjects – from handbooks for secretaries or calligraphers to books on arts and crafts, to alchemical and astrological essays. Recipes can also appear, with or without relation to the main text in the manuscript, in the form of lists or collections or even added in empty spaces as single entries. Why were these recipes written down? Were they used by the many professionals dealing with inks in their every-day work? Or were they part of the literary genre of adab with little or no practical application? To answer these questions a research project was set up: ink recipes have been collected from written Arabic sources, their feasibility has been assessed and some of them have been reproduced. These samples were artificially aged and analysed through an array of analytical techniques, most non-invasive and non-destructive, and the results compared to the data obtained by the application of the same analyses on concrete manuscripts. The codicological characteristics of the manuscripts have also been studied. My aim in this talk is to show how such an interdisciplinary approach could be beneficial for the study of material culture.
Fifty shades of black: Typologies and terminology of black inks in the light of new discoveries

The identification of the materials constituting an artefact is the basis of any correct conservation project. For this reason (among others) in the last 50 years analytical techniques have been applied to the study of manuscripts. Unfortunately, the scientific investigation of Arabic books is still uncommon, focusing mainly on single case studies of illuminated manuscripts where the black media used to write the text is largely overlooked. This lack of knowledge prompted a recent research in which black ink recipes have been collected from written Arabic sources, their feasibility has been assessed, and some of them have been reproduced. These samples were artificially aged and analysed through an array of analytical techniques, mostly non-invasive and non-destructive, in order to build a database of Arabic inks and their ingredients and to verify the detection limits of the equipment employed. Finally, the results were compared to the data collected from historical manuscripts.
In this talk I will focus on the ink typologies and the terminology (ancient and modern) used to describe them. In particular I will highlight how even the modern classification fails to capture the variety encountered in both recipes and manuscripts, particularly in the light of recent discoveries. I will address especially the terminological and conceptual issues of mixed inks – in the form of carbon inks and tannins, of carbon and iron gall inks, and of carbon and metallic salts –, iron gall inks made without vitriol and iron gall inks made with sources of tannins different from gall nuts.
Papyrus was in use in the Islamic world until the 11th century and few ink recipes collected from treatises about the art of the books are dedicated to this support. But papyrus was also burnt in order to obtain soot used as an ingredient for a particular type of ink (midād al-qarāṭīs). In this talk I will give an overview of the recipes – in particular those by ar-Rāzī –, their context and transmission in order to evaluate this double nature of papyrus, which is considered at the same time a writing support and a material to be recycled. Moreover, I will discuss what kinds of ink are most likely to be found, according to these literary sources, on Arabic manuscripts and documents written on papyrus and compare them to the typologies mentioned for paper and parchment. These results are the premises of a research project starting at the beginning of 2019, in which a number of dated fragments from several European institutions will be analysed with non-destructive techniques in order to establish a profile of the main writing media used in the early centuries of the Islamic Era.

The story of how black fungi survive harsh conditions on sun-exposed desert rocks and material surfaces is fascinating. In the presentation examples of how knowledge of these organisms can be of practical value (e.g., in biodeterioration studies, such as on stone markers in cemeteries) would be given. Among other stories, the example of how roof tiles amended with a titanium oxide layers as a biocide actually selected for the black fungi will be told. A hypothesis about how these organisms would likely be found on solar panels and some early BAM work in that area will be presented. This then brings the connection to a study ripe for investigation in North Carolina. The talk presents the development of a study aiming at deciphering the influence of microbial biofilm formation on the energy conversion efficiency of solar photovoltaic panels or modules at two facilities (one facility under the impact of high intensity of animal agriculture and high deposition of ammonia from atmosphere and the other with low animal agriculture and lower atmospheric ammonia deposition) in North Carolina. The main hypothesis of the study is that microbial biofilm formation on solar photovoltaic panels will lead to significant decreases in energy conversion efficiency of solar photovoltaic modules and biofilm formation will also be accelerated by high ammonia concentration in the ambient atmosphere and high nitrogen deposition.
The Interdisciplinary Alchemist: Reproduction and Study of Black Inks from the Islamicate World

Unfortunately, the scientific investigation of Arabic books is still uncommon, focusing mainly on single case studies, often producing unclear results, especially for black inks. This lack of knowledge prompted a recent research in which black ink recipes have been collected from written Arabic sources on bookmaking, their feasibility has been assessed, and finally some of them have been reproduced. These samples were artificially aged and analysed through an array of analytical techniques, mostly non-invasive and non-destructive, in order to build a database of Arabic inks and their ingredients and also to verify the detection limits of the portable equipment employed. In this talk I will present some instances of how this approach can help textual criticism, for example in evaluating variants and determining the competence of authors and compilers. At the same time the research highlights how much this scientific work depends on textual studies, especially concerning the identification of ingredients. Moreover, I will focus on some of the problematics I faced both during the textual interpretation and the actual reproduction of the recipes and how they influenced the results of the scientific analyses. In particular I’ll look into the role played by the degree of purity of the components and the function (real and perceived) of some of the ingredients.
Boden und Steine sind mit jährlich etwa 140 Mio t der mengenmäßig größte Abfallstrom in Deutschland. Bodenmaterial kann in Abhängigkeit seiner vorherigen Nutzung oder Herkunft mit Schadstoffen belastet sein. Seine weitere Verwertungsmöglichkeit hängt nicht nur vom Schadstoffgehalt im Feststoff, sondern auch von der Mobilisierbarkeit dieser Stoffe ab.
Die Paragraphen 6 bis 8 der vorgesehenen Novelle der BBodSchV enthalten neue Vorgaben hinsichtlich des Auf- und Einbringens von Bodenmaterial unterhalb oder außerhalb einer durchwurzelbaren Bodenschicht (4. Referentenentwurf Mantelverordnung, Stand: 06.02.2017). Es soll die Möglichkeit eingeräumt werden, Bodenmaterialien mit Feststoffgehalten von regulierten Stoffen bei Überschreitung des Vorsorgewertes einer weiteren Verwertung zuzuführen unter der Voraussetzung, dass die Eluatwerte eingehalten werden. Für die Auswirkung dieser neuen Regelungen auf die Verwertung von Bodenmaterialien bestehen bisher noch Datendefizite besonders für organische Stoffe.
Durch die Umstellung des Elutionsverfahrens von einem W/F von 10 l/kg (bisherige Regelung in der LAGA M 20) auf ein W/F von 2 l/kg bestehen zusätzliche Unsicherheiten im Hinblick auf die Einstufung von Bodenmaterialien zu Verwertungszwecken.
Im Rahmen eines UFOPLAN-Projektes (FKZ 3716 74 203 0) wurden unterschiedliche Bodenmaterialien verschiedenen Ursprungs (z.B. Baggergut, Bankettschälgut, Stadt- und Auenböden, Bergematerial) mit besonderem Fokus auf PAK untersucht. Von den unter die Kriterien fallenden, ausgewählten Bodenmaterialien wurden Feststoffgehalte, TOC-Gehalte sowie die Eluatwerte aus Säulenversuchen nach DIN 19528 und Schüttelversuchen nach DIN 19529 bei einem Wasser-/Feststoffverhältnis (W/F) von 2 l/kg und 10 l/kg ermittelt und gegenübergestellt.
Mit den neuen und auch mit Auswertung vorhandener Daten kann hinsichtlich PAK gezeigt werden, dass mit Verabschiedung der neuen Regelungen sich zusätzliche Verwertungsmöglichkeiten von Bodenmaterialien außerhalb der durchwurzelbaren Bodenschicht eröffnen und deren Deponierung vermieden werden kann. Analysen zu anorganischen Schadstoffen wurden parallel miterfasst und ausgewertet.
Paragraphs 6 to 8 of the proposed amendment to the German Federal Soil Protection and Contaminated Sites Ordinance contain new requirements regarding the application of soil-like material outside of a rootable soil layer. For example, it is intended to enable reusing of soil-like materials containing regulated substances in concentrations between the single and double precautionary values (PV), provided that the limit values for the eluate are complied with. The aim of a running project (funding code UFOPLAN 3716 74 203 0, German Federal Environmental Agency) is to fill data deficits for soil-like materials which meet the above-mentioned criteria to promote circular economy and conservation of natural resources. Different soil-like materials of various origin (for example dredged material, banquet peeling material, urban and meadow soils) are considered with a special focus on PAHs. A major challenge in sample acquisition is the relatively narrow target range of the PAH content between 3 and 6 mg/kg dm considering a high measurement uncertainty at this concentration level.
The contents of PAHs, heavy metals and other inorganic elements in solid matter are determined following established standards. In addition, the soil-like materials are characterized concerning particle size, pH, conductivity, SOM, TOC and carbonate content. To determine the eluate concentration, column tests according to DIN 19528 and batch tests according to DIN 19529 at a liquid/solid ratio (L/S) of 2 l/kg and 10 l/kg are performed comparatively.
The results show that the new requirements of the planned amendment to the Federal Soil Protection and Contaminated Sites Ordinance certainly opens additional possibilities for reuse of soil-like materials. Even most soil-like materials within 2-3 x PV comply with the eluate value for PAH (0.2 μg/l) both in the batch test and in the column test at L/S 2 l/kg. Selected inorganic substances lead to similar evaluations. Therefore, the application of such soil-like materials outside of rootable soil layers would be possible. Previously, those materials were often used for landfill construction. In the future, a utilization in the adjacent environment by backfilling, recultivation, renaturation, composting or landscaping would be conceivable.
Dem Fachbeirat Bodenuntersuchungen des BMBF/UBA wurden Ergebnisse aus BISp-Forschungsvorhaben zur Untersuchung der Stofffreisetzung aus Sportböden auf Kunststoffbasis sowie zur Modellierung des Schadstofftransfers auf dem Pfad Boden-Grundwasser vorgestellt. Dabei wurde auch auf die derzeitigen gesetzlichen Regelungen auf EU Ebene und die vorhandenen Anforderungen in der Normung eingegangen. Der verbliebene Forschungsbedarf wurde aufgezeigt und dabei auch die Problematik der Bildung von Mikroplastik im Zusammenhang mit Sportböden thematisiert.
Verwertung von Bodenmaterialien - Ergebnisse aus dem UFOPLAN-Vorhaben und Transfer der Ergebnisse

Dem Fachbeirat Bodenuntersuchungen des BMBF/UBA wurden Ergebnisse zum UFOPLAN-Forschungsvorhaben „Monitoring zur Verwertung von Bodenmaterial“ vorgestellt. Die potentielle Auswirkung der neuen Regularien in der geplanten Novelle der BBodSchV (4. Referentenentwurf der Mantelverordnung) auf die Erweiterung der Verwertungsmöglichkeiten von Bodenmaterialien wird aufgezeigt. Probleme des Umgangs mit der analytischen Messunsicherheit im Grenzwertbereich und mit der Vergleichbarkeit der zugelassenen Elutionsverfahren werden diskutiert.
Gegenstand der vorzustellenden Arbeiten ist die Prüfung der Umwelt-beständigkeit und -verträglichkeit von Materialien und Produkten hinsichtlich der Emission von potenziellen Schadstoffen in die Umwelt. Hierzu werden chemisch-physikalische Einflüsse (Bewitterung) und mikrobielle Beanspruchungen an Modellmaterialien evaluiert. So werden die Freisetzungsraten von Schadstoffen in Abhängigkeit der Beanspruchung beschrieben. Als Modellmaterialien kommen die Polymere Polystyrol (PS) und Polypropylen (PP) zum Einsatz. Synergistische Effekte der Bewitterungsparameter und der mikrobiologischen Beanspruchung sollen dabei ebenso betrachtet werden, wie die gezielte Alterung. Auch findet eine Beschreibung des Verhaltens der ausgetragenen Schadstoffe (polybromierte Flammschutzmittel) in den Umweltkompartimenten Boden oder Wasser statt. Hier sind mit Hilfe der zu entwickelnden Screening- und non-Target-Analyseverfahren die Transformation und der Metabolismus durch Mikroorganismen zu beschreiben. Aus den Ergebnissen sollen Korrelationen zwischen den künstlichen Alterungsverfahren und realen Szenarien abgeleitet werden.
Das Prozessmonitoring stellt einen wichtigen Baustein für eine Qualitätssicherung additiv gefertigter Bauteile dar, insbesondere bei kleinen Losgrößen. Der Vortrag zeigt einen Überblick aktueller Entwicklungen der BAM im Bereich des Prozessmonitorings für das Laserstrahlschmelzen (laser powder bed fusion) metallischer Werkstoffe. Durch Vergleich mit der Referenzmethode Computertomografie wird am Beispiel der thermografischen Verfahren gezeigt, wie relevante Defekte in Bauteilen detektiert werden können und Einsatzpotenziale sowie Limitierungen aufgezeigt.
Biocide mitigation strategies of microbiologically influenced corrosion (MIC) in the oil and gas industry have been primarily used to eliminate the growths of sulfate-reducing microorganisms (SRM). However, methanogenic Archaea (MA) can also be highly corrosive by using iron as an electron source for methanogenesis. Because of the fundamental physiological differences between archaea and bacteria, responses of MA towards SRM-specific biocides cannot be deduced using SRM. Due to the lack of information available on the effect of biocides on corrosive MA, we selected THPS, glutaraldehyde, nitrate and perchlorate to compare against corrosive SRM.
Preliminary results showed that at low concentrations of THPS (0-10 ppm), growth of MA was not affected, methane production and corrosion rates (0.1 mm/yr) were comparable between the different THPS concentrations. On the contrary, the SRM strain showed decreased corrosion rates (0.18 mm/yr to 0.03 mm/yr) with increasing THPS concentrations. Further corrosion tests including electrochemical measurements of different biocides on the growth of MA and SRM will be conducted. Such knowledge not only provide important insights on the physiological response of MA to biocides but also contribute to more effective mitigation strategies that can be both economic and environmentally beneficial.
Environmental sustainability and –stability of materials concerning the migration of pollutants

In addition to previously reported results on the accelerated weathering of polystyrene samples (PS) containing 1 wt. % hexabromocyclododecane (HBCD), we present the first results of our investigations of polypropylene samples (PP) containing 0.1 wt. % BDE-209. All studied polymer samples were exposed to a defined weathering schedule in a climate chamber in accordance to regulation EN ISO 4892-3:2006. For the determination of BDE-209 in the collected raining water the samples were prepared in accordance to a validated protocol. Before the analyses each sample was spiked with isotopically labeled BDE-209. Subsequently the samples were extracted with isooctane. The obtained extracts were concentrated, and the resulting solutions were analyzed by GC/MS. Additionally, the total bromine content was monitored for the weathered and untreated samples using laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) and X-ray fluorescence analysis (XRF) as a non-destructive and rapid method. In general, the resulting data from the accelerated weathering will be compared to those from the natural weathering experiments. Here, the surfaces of the test pieces were analyzed by LA-ICP-MS and XRF as well. Moreover, soil bed tests were conducted in a well characterized model soil. This soil (boulder-clay, sand with 12 % loam, particle size in total 0.2-4 mm) is filled in a free-draining basin inside of an air-conditioned cellar. In this manner, TOC, water capacity and humidity are recorded parameters. To induce a leaching process from the samples by the raining water, the target water content is calculated to 8%. The actual humidity is measured by a tensiometer, assuring the duration of the raining period. A defined humidity is a fundamental parameter for a biological activity in the soil. The test polymers were placed up to the half in the soil. Microbial activity of the soil is monitored by a reference polymer (polyurethane) and should induce the release of HBCD and BDE-209 out of the test materials. These released analytes will be captured by passive samplers (silicone tubes) placed in a distinct distance to the polymer samples in the soil. The soil bed experiments are complementary to the weathering experiments due to the biological activity in the soil.
Eine Vielzahl unterschiedlicher Materialien findet für die Gestaltung von Zeichnungen Verwendung. So sind es insbesondere „braune Tinten“, welche die Zeichnungen dominieren. Der Vortrag möchte zunächst einen kurzen Überblick über die Terminologie und die Kunsttechnologie farbiger Tinten geben, um den Kosmos dieser Vielfältigkeit und auch deren Vergänglichkeit zu illustrieren.
Der zweite Teil des Vortrags ist dann den sogenannten Bistertinten und deren analytischen Nachweisen gewidmet. Es handelt sich um Zeichentinten, die aus den wasserlöslichen Bestandteilen von Kaminruß gewonnen werden. Die farbliche Bandbreite reicht von gelb-orange bis braun. Zuweilen wurden diese Tinten mit weiteren Pigmenten versetzt, um eine bestimmte Farbtiefe zu erzielen.
Microbiologically induced corrosion due to bacterial biofilms causes several problems in industrial systems, technical applications and in medicine. Prior to the formation of a biofilm on a substrate, planktonic cells attach on the surface. Hence, the properties of the surface play a key role in biofilm formation and are of great importance for the development of strategies to prevent bacterial attachment and biofilm formation.
This project aims at clarifying to which extent surface micro-/nanostructuring and chemical functionalization affects bacterial attachment and whether a synergistic combination of the two can be used to control bacterial adhesion. To answer these questions, model surfaces with regular patterns of 5-10 micrometers in size have been prepared, which provide distinct zones differing in terms of their chemistry or nano-roughness. This was achieved by micro contact printing of self-assembled monolayers with different functional groups and deposition of patterned ZnO nanorod arrays for studying the effect of surface chemistry and morphology, respectively. Typical contrasts studied were combinations of positively/negatively charged, hydrophobic/hydrophilic or flat/rough.
The attachment behavior of bacteria on tailored surfaces were studied in a flow chamber as a function of time. The strain Pseudomonas fluorescens SBW25 was chosen as a model organism. DNA-intercalating dyes such as Syto9 have a high affinity to adsorb on ZnO nanorods. To overcome this limitation a genetic modification was performed by introducing a gene which expresses a green fluorescent protein in P. fluorescens SBW25 enabling the quantitative evaluation of the flow chamber studies by means of fluorescence microscopy. Further analysis of the attachment behavior was performed by means of scanning electron microscopy.
The presentation will summarize the results of our systematic study on the role of individual parameters on bacterial attachment and highlight synergistic combinations, showing an inhibition or enhancing effect. As the investigations with model substrates enable a precise control of the surface parameters, this approach can be applied to different microorganisms and material systems to achieve a correlative description of bacterial adhesion on solid surfaces.
Normativ ist der Holzschutz in Deutschland durch die DIN 68800-Reihe geregelt. Die Normungsarbeit in CEN/TC 38 bildet dafür den Rahmen, der national angepasst und fachspezifisch erweitert wurde. Zusammen mit dem Kommentar ergeben beide ein in Europa einzigartiges Regelkompendium im Holzschutz. In Zukunft wird die Regelsetzung noch stärker durch internationale Zusammenarbeit bestimmt sein. Der interkontinentalen Marktöffnung sollten Verständigungen auf gemeinsame Qualitätsstandards vorausgehen.

To test the hypothesis that light is an environmental cue that Ascomycota use to coordinate growth, stress responses as well as to establish pathogenic or symbiotic relationships, the photoreceptor (PR) distribution in species from different ecological niches was analysed. The genomes of black fungi from phyllosphere and exposed solid surfaces contain multiple photoreceptors (PRs). The filamentous foliar plant pathogen Botrytis cinerea (Leotiomycetes) has a highly sophisticated photosensory and signalling system that helps to avoid light and to locate susceptible hosts. Rock-inhabiting Dothideo- and Eurotiomycetes including Knufia petricola possess equal numbers of PRs along with the same set of protective pigments. This similarity between black fungi from plant and rock surfaces suggests that photoperception and photoregulation are important for fungi that avoid loss of energy and nutrients through cooperation with phototrophs.

Untersuchungen von Zeichnungen und Manuskripten mit zerstörungsfreien Methoden haben in den letzten Jahren enorm zugenommen. Die nicht-invasiven Analysen ermöglichen Informationen zur Herkunft, zum Herstellungsprozess und möglicherweise zum Alter (direkte Dating nicht möglich, Ausnahme: Papierstruktur, Wasserzeichen). Die zerstörungsfreien Analysen können Rückschlüsse auf mögliche Fälschungen geben, sind neben den offensichtlichen Vorteilen jedoch auch mit einzelnen Limitierungen behaftet. Der Vortrag soll anhand einiger Fallbeispiele die Möglichkeiten aber auch Grenzen bei der zerstörungsfreien (mobilen) Auffindung von Fälschungen aufzeigen.
A broad suite of immune adaptations have evolved in social insects which hold close parallels with the immune systems of multicellular individuals. However, comparatively little is known about the evolutionary origins of immunity in social insects. We tackle this by identifying immune genes from 18 cockroach and termite species, spanning a gradient of social lifestyles. Termites have undergone contractions of major immune gene families during the early origin of the group, particularly in antimicrobial effector and receptor proteins, followed by later re-expansions in some lineages. In a comparative gene expression analysis, we find that reproductive individuals of a termite invest more in innate immune regulation than other castes. When colonies encounter immune-challenged nestmates, gene expression responses are weak in reproductives but this pattern is reversed when colony members are immune-challenged individually, with reproductives eliciting a greater response to treatment than other castes. Finally, responses to immune challenge were more comprehensive in both subsocial and solitary cockroaches compared to termites, indicating a reduced overall ability to respond to infection in termites. Our study indicates that the emergence of termite sociality was associated with the evolution of a tapered yet caste-adapted immune system.
Results suggest a reduction in immune gene repertoires in termites and possible complementary expression between termite castes. With comparative genomics we will investigate the evolution of gene families related to immunity, try to understand where reductions and expansions take place and relate these changes to shifts in sociality and ecology. The role of TEs in expansions and contractions of immune gene families will be investigated. For these analyses, we propose to generate high quality, highly contiguous genomes of species from different levels of sociality, covering all major termite families. With comparative transcriptomics we will investigate the expression of immune genes in different castes. Via network analyses we will identify pathways indicated in differential immunity between castes and between species of different sociality levels. We will investigate how these pathways have been rewired along the transitions to higher levels of sociality and how, intra-specifically, they change between castes.
Social insects nesting in soil environments are in constant contact with entomopathogens and have evolved disease resistance mechanisms within a colony to prevent the occurrence and spread of infectious diseases. Among these mechanisms: mutual grooming reduces the cuticular load of pathogens, and burial of cadavers and cannibalism can prevent pathogens from replicating within the group. We explored how the rate and type of collective behavioural response is determined by stepwise infection dynamics operating at the level of the individual. Specifically, we infected the eastern subterranean termite Reticulitermes flavipes with different types of infectious particle and infection route of the entomopathogenic fungus Metarhizium anisopliae and recorded behavioural responses of nestmates to individuals at different times during the progression of infections. As expected, termites groomed conidia-exposed individuals significantly more than controls. Interestingly, grooming was significantly elevated after fungal germination than before, suggesting that pathogen growth cues act as strong stimulators of allogrooming. Conidia-exposed termites were cannibalized, but only after they became visibly ill. By contrast, termites did not groom blastospore-injected individuals more than controls at any time-point following infection. Instead, we found that blastospore-injected individuals were continually cannibalized at a low-level following injection with either viable or heat-killed blastospores, with a marked increase in cannibalism after termites injected with viable blastospores became visibly ill and were close to death. Together, these findings point to the importance of host condition as a cue for social hygienic behavior, and that the host itself appears to emit essential sickness cues that act as targets for its own sacrifice. This demonstrates that termites have independently evolved to both identify and destructively respond to sickness.
The termites are a derived eusocial lineage of otherwise non-social cockroaches. Understanding the proximal and ultimate drivers of this major evolutionary transition represents an important goal in biology. One outstanding question concerns the evolution of termite immunity, which is thought to have undergone broad-sweeping adaptations in order to enhance group-level immune protection. To understand the evolutionary origins of termite immunity, we conducted qualitative and quantitative transcriptome analyses along a gradient of sociality. Firstly, we aimed to identify large-scale genetic shifts in immune traits linked to eusociality by comparing immune gene repertoires in solitary and subsocial cockroaches and across a range of eusocial termite lineages. Secondly, we compared the responses of a solitary cockroach, a subsocial wood-roach and different castes of a lower termite species to a non-specific immune challenge, in order to understand how sociality may have influenced the evolution of immune gene regulation. Firstly, we found that termites have a broadly representative repertoire of canonical immune genes as compared to gregarious cockroaches and subsocial wood-roaches. Secondly, with respect to immune challenge, the solitary cockroach and the subsocial wood-roach displayed a similarly comprehensive induced response, while the termite response was considerably dampened by comparison and strongly influenced by caste; with reproductives displaying a generally higher constitutive level of immune-gene expression compared to sterile castes. In summary we did not find termite eusociality to be associated with significant changes in immune gene diversity, but rather to be linked with significant modifications to the regulation of immunity following the origin of division of labour.
In arid sun-exposed environments, rock-inhabiting fungi dominate microbial communities on rock surfaces. Fungi are known to enhance rock weathering, although the exact mechanisms are not completely understood. These natural processes can be simulated by reproducible experimental systems. Particularly attractive are genetically amenable rock-weathering fungi, where certain traits can be knocked-out to identify weathering-relevant functions. Here, we studied the effect of the rock-inhabiting fungus Knufia petricola A95 and its melanin-deficient mutant (ΔKppks) on the weathering of a Fe-bearing olivine. We examined the olivine dissolution kinetics and looked at the olivine-fungus interface. For the former we inoculated olivine powder with the fungus in batch and mixed flow reactors and analysed the medium solution by ICP-OES, for the latter biofilm-covered olivine sections from flow-through reactors were analysed by FIB-TEM.
In general olivine dissolution was lower when olivine was incubated without fungi: the abiotic olivine dissolution rates were lowest and the surface of the abiotically reacted olivine sections showed no etching. Various chemical analyses show Fe (oxyhydr)oxide precipitation on top of the abiotically reacted olivine, indicating its role in inhibiting olivine dissolution. Both the wild type (WT) and ΔKppks solubilised and bound significant amounts of Fe released by olivine dissolution. Fe (oxyhydr)oxide precipitation was thus reduced, explaining the enhanced olivine dissolution in their presence. Particularly efficient at preventing Fe precipitation were attached WT cells: the inhibition of olivine dissolution almost completely disappeared. The attachment capacity of the WT is most likely caused by the production of WT-specific extracellular polymeric substances. Our presented experimental systems allowed the precipitation of Fe (oxyhydr)oxides and included a rock-inhabiting fungus and thus simulated weathering processes relevant to natural ecosystems.
Vorstellung des REFOPLAN-Projekts extraWERT Erhebung zusätzlicher Wertstoffpotenziale in Klärschlamm

Auf dem Regionalgruppentreffen wurden erste Projektergebnisse vorgestellt und die Probenahmestrategie diskutiert. Vorgestellt wurde eine Rückberechnung der mittleren Wertstoffgehalte aus dem Klärschlamm-Monitoring 2014 (Krüger und Adam) und erste Auswertungen der Klärschlammbericht-Rohdaten aus NRW und Hessen.
Im Projekt extraWERT geht es um die Ermittlung von Nährstoffpotenzialen in Abwasser und Klärschlamm. Neben kommunalen Kläranlagen soll auch die Industrie als Quelle für nährstoffreiche Abfallströme betrachtet werden. Mit dem Vortrag sollten Mitglieder des MIV zu einer Projektbeteiligung bewegt werden. Es sollen rohes und gereinigtes Abwasser von direkt einleitenden Betrieben auf Makronährstoffe untersucht werden, neue Erkenntnisse werden insbesondere für die Nährstoffe Calcium und Magnesium erwartet.
Die landwirtschaftliche Klärschlammverwertung ist in der Bundesrepublik Deutschland seit Jahren rückläufig. Neben den Risiken durch Schadstoffeinträge gibt es in Deutschland Regionen mit einem stark erhöhten Nährstoffüberschuss. Um die Grundwasserbelastung zu verhindern wurde u.a. das Düngerecht verschärft, was die Flächenkonkurrenz zwischen Klärschlamm und Wirtschaftsdünger vergrößern wird.
Es ist davon auszugehen, dass sich die neuen Regelungen im Düngerecht und der Klärschlammverordnung massiv auf die landwirtschaftliche Verwertung von Klärschlamm und damit auf die Kreislaufwirtschaft verschiedener Nährstoffe auswirken werden. In diesem Beitrag werden erste Ergebnisse des Projekts extraWERT vorgestellt.
Der mitteleuropäische Mensch hält sich 80 – 90 % der Zeit in Innenräumen auf. Je nach Aktivität atmet er 10 – 20 m³ bzw. 12 - 24 kg Luft täglich, das ist weit mehr als das Doppelte der Summe an Essen und Trinken. Die technischen Voraussetzungen für die Bestimmung von Emissionen aus Bauprodukten in die Luft liegen vor – harmonisierte horizontale Prüfnorm EN 16516. Es dürften mittlerweile zig-tausende Emissionsdatensätze vorliegen. Defizite gibt es bei der Kennzeichnung zur Unterscheidung emissionsarmer und emissionsreicher Produkte, die bestehende CE-Kennzeichnung ist unvollständig, ersatzweise gibt es nationale Regelungen.
Gerade bei modernen, dichten Bauten mit geringem natürlichen Luftwechsel sind hohe Schadstoffkonzentrationen zu erwarten, wenn nicht emissionsarme Materialien und Produkte zum Einsatz kommen.
The initial focus of this research was on the development of a general workflow for the documentation and monitoring of historical stained glass windows using structured light scanning.
Therefore windows from different churches, time periods and with different corrosion and damage phenomena were scanned before and after conservation measures.
Neuartige Materialien, die bekannte (Werk-)Stoffe mit neuen Funktionalitäten ausstatten, spielen eine zunehmend wichtige Rolle im Bereich der Materialforschung und -prüfung. Das Spektrum neuartiger Materialien reicht von der gezielten Oberflächenfunktionalisierung und -strukturierung makroskopischer Materialien, dünnen Beschichtungen bis hin zu mikro- und nanoskaligen Kompositmaterialien und funktionalen Materialien an der Schnittstelle zur Biologie, Biotechnologie, nachhaltige Energiespeicherung und Sensorik. Dabei bieten neuartige Materialien die Chance, Werkstoffe und Produkte mit erweiterter oder verbesserter Funktionalität zu erhalten und Sicherheit bereits im Designprozess zu berücksichtigen. Durch dieses breite Anwendungsspektrum und die Herausforderungen, die solche Materialien für die Sicherheit in Chemie und Technik mit sich bringen, sind diese in allen Themenfeldern der BAM repräsentiert (Material, Analytical Sciences, Energie, Infrastruktur und Umwelt).
Die Aufgaben der BAM erstrecken sich dabei von der Herstellung von Referenzmaterialien für Industrie, Forschung und Regulation, über die Erstellung von standardisierten Referenzverfahren für nachhaltige Messungen im Umwelt- und Lebenswissenschaftsbereich bis hin zur Bereitstellung von belastbaren und zitierbaren Referenzdaten. Durch die genaue Charakterisierung neuartiger Materialien können potentiell problematische Substanzen identifiziert und deren Risiken besser abgeschätzt werden. In diesem Beitrag werden einige aktuelle Beispiele aus diesen Bereichen vorgestellt.
Museums worldwide are equipped with different display cases. Exhibit display cases should protect cultural objects from dust as well as from mechanical and physical damage. To ensure a stable climate inside the display cases, a low air exchange rate is maintained. Typically air exchange rates are often smaller than 0.1 d 1, which can result in rising concentrations of potential harmful immissions inside of the display cases due to emissions from materials. Especially high concentrations of organic acids, which can emit from e.g. sealing materials, can produce damage of cultural objects. In 2012 BAM introduced a procedure witch is called: BEMMA-Scheme (Bewertung von Emissionen aus Materialien für Museumsausstattungen) which stands for: “Assessment of Emissions from Materials for Museum Equipment”. Micro chambers are used for VOC emission tests of display case construction materials, e.g. textiles, plastics, sealing material, coatings and others. Each sampling procedure is carried out in duplicate. Emissions like formic acid, acetic acid, formaldehyde and oximes are excluded and the sum of emissions of VVOCs, VOCs and SVOCs is limited. For a positive assessment all listed criteria must be fulfilled; otherwise the display construction material fails the BEMMA scheme. The BEMMA scheme is not a guarantee for an emission free display case, but a necessary requirement for the choice of suitable materials for emission and immission reduced display cases.
Die gemeinsame Forschungsstrategie der Bundesoberbehörden zur Nanotechnologie wurde 2016 veröffentlicht. Die darin enthaltenen Aufgaben wurden von den Bundesoberbehörden vielfältig bearbeitet. Diese Präsentation gibt einen Überblick über die Projekte, die von der BAM bis 2019 bearbeitet wurden/werden und sich in den Rahmen der Forschungsstrategie einordnen.
Traceable and reliable chemical element analysis of aerosols by X-ray spectrometry was investigated using aerosol samples from field campaigns which have been measured in the GIXRF-beamline at BESSY.
The reference-free XRF approach allows for a traceable analysis of the mass deposition.

Measurements of aerosol particles are vital for enforcing EU air quality regulations to protect human health, and for research on climate change effects. Although metrics such as PM10 and PM2.5 are currently in use, the level of uncertainty of aerosol metrics is too high and the traceability is insufficient. The project AEROMET, which has been started in June 2017 aims at implementing improvements in a) the uncertainty of particle mass, size and number concentration measurements and b) in the characterization of regulated components in airborne particles. Both are demanded by existing networks within the EU as well as by global atmospheric research.
On-site measurement campaigns
One of the objects is the application of mobile x-ray spectroscopy techniques combined with aerosol sampling techniques for quantifying particle compositions in the field for real time analysis. During two in-field measurement campaigns in Budapest, Hungary in May 2018 and Cassino, Italy in September 2018 the size dependent mass concentrations of specific elements in ambient aerosols were monitored under dynamic conditions.
Typically, airborne particles are sampled on filter substrates. During this project new sampling methods with specially designed substrate holders for an in-situ TXRF analysis were developed and applied for the first time. This approach allows a direct time and size resolved analysis without laborious digestion steps and a reduced risk of contamination.
Aerosol particles were sampled in a 13-stage DLPI impactor - size range from 0,03 µm to 10 µm - which was equipped with special adapters for acrylic discs of 30 mm diameter, serving as substrates. TXRF analysis was performed on site with the transportable spectrometer S2 PICOFOX (Bruker Nano GmbH) equipped with a Mo X-ray tube and a 30 mm² Silicon Drift Detector (SDD). Excitation conditions were 50 kV, 600 µA, measurement time 1000 s. Quantification was based on internal standardization using 50 ng of Y in solution, which was pipetted into the centre of the discs prior to sampling.
At moderate air pollution levels, i.e. PM10 ~ 20 µg/m³, sampling times of less than 2 hours were enough for the detection of elements in different particle size bins. The in-situ approach and the high sensitivity of TXRF enables the observation of rather quick changes in the quantity and distribution of elements in an ambient aerosol on the day of sampling, as the below example from the Cassino field campaign on 11 Sept. 2018 shows: The analysis of the morning and afternoon sampling shifts reveals the occurrence of the elements Fe, Ca and Si in different size bins as well as their significant temporal change in respective mass concentrations over the day while the distributions of several other elements in the aerosol remain unchanged.
The validation of these results by backup measurements is planned.
Simple particulate matter sensors are gaining popularity due to their low price, easy handling and good temporal resolution. In this presentation, we report on the performance of a Sharp optical PM sensor GP2Y1010AU0F, which costs less than 15 €.
The sensor is built around an infrared emitting diode (ILED) and a phototransistor detecting the light scattered from the aerosol particle. An electronic circuit shapes the detected light in a pulsed signal. The manufacturer advises sampling the output signal 280 microseconds after the ILED pulse. The measured output voltage is an indicator of dust concentration.
We have built two identical simple PM monitoring devices using Raspberry Pi 3 computer interfacing the PM sensor with Microchip’s MCP3002 ADC via SPI. The ADC is capable of more than 100 ksamples/s at 10-bit resolution. The Rpi3 was pulsing the sensor at 10Hz, digitizing and saving the data and sending the results wirelessly.
Sensor’s output pulse shape was sampled with 10 microsecond time steps and saved, thus making offline analysis possible. A time jitter of output pulses can be observed and suggests a peak fitting as a better approach to the signal readout compared to the single sampling at a fixed time after pulse triggering We compared both methods.

Es werden Methoden der Verwertung carbonfaserhaltiger Reststoffe in der Pyrometallurgie diskutiert.
Conventional fertilisers have their disadvantages as nutrient release is not synchronised with the demand of plants. Several approaches of "next generation fertilisers" are aiming at controlled nutrient release. Recycling fertilisers are often not soluble in water but fully plant available. They are suitable raw materials for the production of new types of controlled release fertilisers.
EN 16516 sets the test method and requirements for the determination of emissions of Volatile Organic Compounds (VOCs) from building materials into indoor air. To address the quality control requirements for the class of semi-VOCs (SVOCs), VSL developed gaseous reference materials. A novel home-made dynamic gas mixture preparation system, operating according to ISO 6145-4 (continuous injection method), has recently been developed and validated. Thanks to the stable temperature control up to 100 oC, the system can prevent condensation of the SVOCs in air at indoor air concentration levels. The in-situ obtained SVOC gas standards can be sampled in sorbent tubes to obtain SVOC transfer standards. A study was performed to determine the optimal sorbent material and storage conditions. This study will be presented together with the results of the 2018 Round Robin test for emission test chamber measurements organised by BAM. Using the novel system, VSL prepared transfer standards with known amounts of VOCs and SVOCs for participants to evaluate their analytical performance.
The chemical emissions from products are tested by means of emission test chambers under defined conditions (climate, loading, air change rate). The standard method for the determination of volatile organic compounds (VOC) is the sampling onto Tenax-tubes followed by thermal desorption (TDS) and gas chromatography-mass spectrometry (GC-MS) analysis. The EU-LCI list includes some very volatile organic compounds (VVOC) and some VOC for which there are limitations when using the standard method. For VVOC additional sampling is required using stronger absorbers like Carbotrap or multi-bed adsorption tubes. The analysis of VVOC also requires a different GC oven program and a different column for the separation. For the determination of formaldehyde and other low boiling aldehydes (e.g. acetaldehyde, acetone, propanal, propenal) DNPH-cartridges are used which are extracted with acetonitrile followed by liquid chromatography (HPLC-UV) analysis. The derivatisation of propenal and other unsaturated aldehydes (e.g. 2-butenal) with DNPH might lead to lower findings due to incomplete derivatization and forming of by-products. For a better quantification of acetic acid the use of ion chromatography (IC) is recommended because the analysis of acetic acid with the standard method (TDS) leads to lower findings due to break through during sampling. The use of ion chromatography for the analysis of organic acids requires a third sampling technique. The acids are adsorbed onto silica-gel and extracted with water.
Microbiologically influenced corrosion (MIC) of iron is usually attributed to sulfate-reducing microorganisms (SRM) in offshore industries, such as the oil and gas pipelines, due to the high concentrations of sulfate in the seawater. SRM act upon the metal by the reactiveness of hydrogen sulfide (HS-), and by withdrawal of the available electrons (Fe --> Fe2+ + 2e-; E° = -0.47 V) in electrical contact with the metal (EMIC). However, methanogenic archaea can also cause MIC. Because they do not produce HS-, withdrawal of electrons may be their main corrosive mechanism; however, mechanistic details and kinetics of the overall process are poorly understood.
To investigate the corrosion potential of methanogens, we studied the EMIC methanogenic strains isolated from marine sediments (Methanobacterium-affiliated strain IM1) and crude oil tanks (Methanococcus maripaludis Mic1c10), in an in-house developed flow-through cell to simulate a fluctuating environment. A co-culture of M. maripaludis and D. alaskensis was also established to study the effect of syntrophic growth on metal corrosion that may occur in industrial pipelines. Results indicate that the rates of iron corrosion due to coupled methanogenesis (up to 0.4 mm/yr) are higher to that caused by the marine SRM Desulfovibrio alaskensis (0.15 mm/yr). Surface analyses of the metal showed severe pitting with high methane production. Genomic analysis of the EMIC methanogen M. maripaludis Mic1c10 will provide an insight on the mechanisms of MIC. Such knowledge and deeper understanding also from an electrokinetic point of view may not only provide further models in microbial electrophysiology, but also contribute to mitigation strategies in MIC

In a rapidly evolving world, the demand on raw materials is increasing steadily and many technologies are dependent on a secure supply of hi-tech metals. Recently, scandium (Sc) has attracted attention since its use in high strength Al-alloys and solid-oxide-fuel-cells strongly improves the performance of those materials.
The element Sc is not exceptionally rare but quite resistant to geochemical enrichment processes, it is scarcely found enriched to high concentrations and is recovered as a by-product. Since Sc enrichment in Greek bauxite residues was shown by Ochsenkühn-Petropoulou et. al (1994), intensive research on this material and development of efficient Sc-recovery methods is ongoing.
This study investigates Sc-bearing species in bauxite residues from alumina production. It aims to provide direct evidence about the Sc-speciation’s in those secondary resources and tries to find the link to speciation’s in primary resources, e.g. bauxites and laterites.
Therefore, Sc K edge XANES (X-ray absorption near edge structure) spectroscopy is performed using synchrotron radiation to determine the presence of certain Sc-components and distinguish between adsorbed and chemically bonded Sc as was shown for lateritic deposits in Australia by Chassé et al. 2016. For comparison, reference standards of Sc-bearing and Sc-adsorbed species are synthesized. Indirect inferences from leaching behavior of bauxites, in cases supported by analyses with LA-ICP-MS, suggest Sc to be associated with either iron- or aluminum phases (Vind et al. 2017); (Suss et al.). It remains unclear how different primary materials influence Sc-speciation in the bauxite residue. Therefore, a comparison between different European bauxite residues is made in this study. The investigations should help to understand Sc chemistry and
behavior in different primary and secondary materials and provide fundamentals for metallurgical processing. The research is incorporated in the SCALE project (GA No. 730105) funded by EU Horizon 2020 research and innovation program.
Laser Powder Bed Fusion (L-PBF) is a promising additive manufacturing (AM) technology for metal part production especially for complex and lightweight structures or functional designs. In L PBF processes several by-products including welding plume and its condensates, spatter and ejected powder are generated during laser exposure. Investigations of micro- and nano-sized by-products have received little attention in literature. This study focuses on the analysis of particle emissions in L PBF of 316L stainless steel using a scattered light aerosol spectrometer and a fast mobility particle sizer spectrometer during the process which allows for in-situ analysis of particle sizes in the range of 6 nm to 100 µm. A distinct correlation of emission signals to part position can be revealed. In addition, a significant influence of laser scanning vector directions on emission signals is presented. Furthermore, differing powder layer thicknesses can be recognised by deviations in emission signals.
Moderne Vitrinen sollen gleichzeitig eine hohe Dichtigkeit und die Abwesenheit jeglicher Fremdstoffe in den Vitrinen gewährleisten. Diese Anforderungen sind zeitgleich kaum erfüllbar, da es emissionsfreie Materialien (vor allem Kleb- und Beschichtungsstoffe) nicht gibt. Um die potentiellen Emissionen zu minimieren wurde ein Messverfahren entwickelt, welches inzwischen als BEMMA-Schema (Bewertung von Emissionen aus Materialien für Museumsausstattungen) in die Museumswelt Einzug genommen hat.
Die erfolgreiche Bewertung der für den Bau verwendeten Materialien gemäß BEMMA-Schema resultiert nicht in einer emissionsfreien Vitrine, sondern soll bei der Auswahl geeigneter Materialien zur Herstellung möglichst emissionsarmer Vitrinen helfen. Konkrete handwerkliche Durchführungen beim Bau, Dichtheits-anforderungen, wie auch die Aufstellbedingungen (Umgebung der Vitrine) beeinflussen die Emissionscharakteristik der fertigen Vitrine in der Praxis. Eine saubere Vitrine hilft nichts in einer belasteten Umgebung oder auch bei emittierenden Exponaten. Inzwischen liegen Erfahrungen mit dem Schema seit 2012 vor.
Der Messablauf sieht vor, dass von den Materialien kleine Muster mit der Mikrokammer (Markes) innerhalb von 2 Tagen untersucht werden. Dabei wird mit Silikagelkartuschen auf Ameisen- und Essigsäure, mit DNPH auf Aldehyde und Ketone und mit Tenax® sowohl auf VOC als auch für die spezifische Oxim-Analytik die Abluft der Mikrokammern untersucht. Die Bewertungskriterien basieren auf den Ergebnissen einer Vorstudie mit ca. 150 verschiedenen Produkten im Zeitraum 2010 bis 2012.
Vor nun 30 Jahren, im Jahre 1989, wurde die erste Zulassung für eine Kunststoffdichtungsbahn (KDB) von der BAM auf der Grundlage des „Niedersächsischen Dichtungserlasses“ erteilt. Vor ungefähr 10 Jahren wurde die BAM mit dem Erscheinen der Deponieverordnung (DepV) mit der Zulassung von Geokunststoffen, Polymeren und Dichtungskontrollsystemen beauftragt. Wir möchten dies zum Anlass nehmen, an dieser Stelle den viele ehrenamtlichen Experten des Fachbeirats und seiner Arbeitsgruppen für ihren Einsatz und die Unterstützung zu danken.
Darüber hinaus soll über aktuelle Arbeitsthemen und Entwicklungen im Fachbeirat berichtet werden. So wurde in einer Sondersitzung des Fachbeirats über die Entlassung aus der Nachsorgephase beim Einsatz von Geokunststoffprodukten in Böschungen diskutiert. Die Unterarbeitsgruppe Fremdüberwachung in der Produktion hat getagt. Darüber hinaus werden Änderungen in den Zulassungsrichtlinien für Kunststoffdichtungsbahnen, Kunststoff-Dränelemente und Geogitter kurz vorgestellt.
Im Rahmen des Projektes „Hinterglasmalerei als Technik der Klassischen Moderne von 1905 – 1955“, welches vom Museum Penzberg – Sammlung Campendonk, Penzberg (Gisela Geiger und Diana Oesterle) geleitet wurde und in Zusammenarbeit mit der freischaffenden Restauratorin Simone Bretz, der Bundesanstalt für Materialforschung und -prüfung (BAM) und dem Doerner Institut erfolgte, konnte im Rahmen einer Promotion erstmals ein Konvolut von mehr als 60 Hinterglasbildern mit nicht-invasiven Analysemethoden untersucht werden. Die eingesetzten Verfahren ermöglichten die Identifizierung der Pigmente und eine Klassifizierung der verwendeten Bindemittel.
Die materialwissenschaftliche Beschäftigung mit der Hinterglaskunst in ihrer Vielfalt ist ein relativ junges Forschungsfeld. Lange wurde ihr mit Unkenntnis, Missverständnis und sogar Geringschätzung begegnet. Im kunsthistorischen Kontext wurde diese Art der Kaltmalerei zumeist mit dem Begriff der Glasmalerei belegt und nicht als eigenständige Kunstform anerkannt, da in beiden Fällen Glas als Bildträger seinen Einsatz findet. In der Hinterglasmalerei wird die Tafelrückseite mit organisch gebundenen Malfarben verziert und das Werk ausschließlich bei auffallendem Licht betrachtet. Die künstlerische Herausforderung liegt immer in der technischen Notwendigkeit eines umgekehrten Malvorganges: die oberste, durch das Glas sichtbare Schicht muss zuerst auf den Bildträger Glas aufgetragen werden, der optische Hintergrund wird zuletzt aufgebracht. Im Vergleich zum Tafelbild, bei dem die Malerei erst durch den Firnisauftrag Tiefenlicht erhält, entfalten die Farben in der Hinterglasmalerei bereits beim Malen ihre Intensität und erhalten gleichzeitig Schutz durch den gläsernen Bildträger (Bretz et al. 2016).
Neben der kunstgeschichtlichen und kunsttechnologischen Bearbeitung der Hinterglasbilder spielten materialwissenschaftliche Untersuchungen an einer Auswahl von 66 Bildern im Rahmen des von der VolkswagenStiftung finanzierten Forschungsprojektes eine zentrale Rolle. Grundsätzlich wurde ein nicht-invasiver Ansatz gewählt, um die fragilen Kunstwerke in-situ zu messen und somit einen Transport zu vermeiden. Die Methodik umfasst neben VIS-Spektrometrie in Reflexion und energiedispersiver Röntgenfluoreszenzanalyse (RFA) auch Infrarotspektroskopie in diffuser Reflexion (DRIFTS) und Raman-Spektroskopie. Mit Hilfe des gewählten methodischen Ansatzes lassen sich sowohl anorganische als auch organische Pigmente identifizieren und die Zusammensetzung von Metallfarben und Folien sowie die Glaszusammensetzung qualitativ bestimmen. Ferner ermöglicht die DRIFT-Spektroskopie eine Klassifizierung der Bindemittel.
Die im Rahmen des Forschungsprojektes analysierten Hinterglasbilder können folgendermaßen chronologisch gruppiert werden: 1910-1919 (17 Bilder), 1920-1929 (18 Bilder), 1930-1939 (6 Bilder), 1940-1949 (7 Bilder), 1950-1955 (13 Bilder) und >1955 (5 Bilder). Die Auswertung der Ergebnisse ermöglicht grundsätzliche Aussagen über die verwendeten Malmaterialien in Hinterglasbildern in der ersten Hälfte des 20. Jahrhunderts und zeigt Änderungen der verwendeten Pigmente im zeitlichen Kontext auf.
Während der ersten Hälfte des 20. Jahrhunderts kamen viele neue Pigmente und Malmaterialien auf den Markt, wobei speziell die synthetischen organischen Pigmente (SOP) zu nennen sind. Eine Vielzahl an SOP konnte in Hinterglasbildern aus allen Zeitgruppen identifiziert werden, wobei hier vor allem Farblacke aus synthetischem Alizarin (PR83), Vertreter der roten und orangen β-Naphthol Pigmente [(z.B. PR3, PR4, PO5 (in Nächtliche Fahrt, 1921 von L. Hildebrandt nachgewiesen)], die blauen und grünen Kupferphthalocyanine [z.B. PG7, PB15 (taucht ab 1949 in Hinterglasbildern auf)] und einige gelbe Azopigmente [z.B. PY1, PY3, PY12 (in Geneigter Mädchenkopf, 1941 von Oskar Schlemmer gefunden)] aufzuführen sind (Steger et al. 2018; Steger et al. 2019a). Einige seltene SOP der Triarylcarboniumgruppe konnten ebenso nachgewiesen werden. Darunter fallen neben der Identifizierung von PR81 (Ohne Titel, 1954, M. Uhlenhuth) auch die erstmaligen Nachweise von PV2 (Hahn, um 1945, L.-G. Buchheim) und PG1 (Exzellente und einmalige Dressuren, 1945/46, L.-G. Buchheim) in Kunstwerken überhaupt (Steger et al. 2018; Steger et al. 2019a). Ferner konnten mit PB52 aus der Gruppe der Anthrachinone und PR60 aus der Gruppe der verlackten Naphthalin-Sulfonsäure-Pigmente weitere seltene SOP in den Bildern Rudern (um 1912) beziehungsweise Apokalyptischer Reiter II (1914) von Wassily Kandinsky nachgewiesen werden (Steger et al. 2019b).
Neben den Entwicklungen der SOP können auch eindeutige Trends in den anorganischen Pigmenten veranschaulicht werden. Seltene anorganische Pigmente wie zum Beispiel Strontiumweiß (SrSO4) in der Kreuzabnahme (1914/15) von Carlo Mense (Steger et al. 2019c) wurden identifiziert. Neu entwickelte anorganische Pigmente wie zum Beispiel Cadmiumrot (Cd(S,Se); ab 1928 in 11 Hinterglasbildern nachgewiesen) oder Titanweiß (TiO2; Anatas kommt ab 1922 vor, Rutil konnte nur in Kissen der Träume, 1976 von Werner Schriefers identifiziert werden) tauchen in einigen Hinterglasbildern auf. Schweinfurter Grün (3Cu(AsO2)2·Cu(CH3COO)2) oder Strontiumgelb (SrCrO4) wurden in einigen Bildern bis etwa um 1925 gefunden; diese Pigmente konnten dagegen in späteren Bildern nicht mehr nachgewiesen werden. Als Beispiel sind hier vier frühe Hinterglasbilder (1909–1914) von Wassily Kandinsky zu nennen, wobei Strontiumgelb in vielen Bildpartien zu finden war (Steger et al. 2019b). Publizierte materialwissenschaftliche Ergebnisse von Kandinskys Leinwandgemälden aus späteren Werksperioden zeigen, dass Strontiumgelb zu dieser Zeit vollkommen aus seiner Palette verschwunden ist und vor allem durch ein anderes Gelbpigment, nämlich Cadmiumgelb, ersetzt wurde (McMillan et al. 2013).
Eine materialtechnische Besonderheit in der Hinterglaskunst ist die Verwendung von Metallfolien und Metalleffektpigmenten. Metallfolien aus Silber, Zinn, Aluminium oder Messing wurden vielfach als künstlerisches Element in die Komposition integriert, wobei sie als finale Schicht auf, die bereits gestaltete, leicht transparente Malerei geklebt wurden. Durch die Betrachtung im auffallenden Licht erzeugten die Folien bemerkenswerte Glitzereffekte. Zinn- und Aluminiumfolien wurden zum Beispiel in Rudern (um 1912, W. Kandinsky) nachgewiesen, während Messingfolien häufig in Bildern von L.-G. Buchheim gefunden wurden. Die Zinnfolien wurden im 19. und frühen 20. Jahrhundert für Verpackungszwecke (z.B. Zigaretten, Schokolade) verwendet. Aluminiumfolie wurde erst ab 1910 industriell in der Schweiz hergestellt und zu Beginn vor allem für Schokoladenverpackungen eingesetzt (Skrabec 2016). Die frühe Verwendung solche Folien in Hinterglasbildern (z.B. Rudern, um 1912) zeigen eine rasche Verbreitung dieses Materials bei den Künstlern an. Unter Metalleffektpigmenten versteht man feingemahlene Pulver aus Metall (z.B. Zinn, Nickel, Kupfer) und Legierungen (z.B. Messing, Neusilber) mit unterschiedlichen Tönungen, welche bei seitlichem Lichteinfall auf Hinterglasbildern ein merkbares Funkeln erzeugen. Solche Metalleffektpigmente wurden vielfach bei Werken von Heinrich Campendonk nachgewiesen (Geiger und Bretz 2017) und konnten in allen chronologischen Gruppen gefunden werden.
Grundsätzlich ermöglichen die Ergebnisse ein besseres Verständnis für die Technik der Hinterglasmalerei und helfen insbesondere, diese eigenständige Kunst im Hinblick auf die „klassische“ Tafel- bzw. Leinwandmalerei abzugrenzen. So zeigen Vergleiche mit Gemälden auf Leinwand von Wassily Kandinsky deutliche Unterschiede in den verwendeten Farbmitteln für beide Techniken. Die Anzahl von 66 gemessenen Hinterglasbildern von 39 Künstlern erscheint im Kontext von über 1200 Arbeiten, welche im Rahmen des Projektes lokalisiert werden konnten, gering. Allerdings können die durchgeführten Untersuchungen als Startpunkt für zukünftige Projekte in diesem Forschungsfeld, beispielsweise für einzelne Werkkomplexe verstanden werden.
Die Forschungsergebnisse werden auf die Webseite www.hinterglas-klassischemoderne.de eingestellt. Im Sommer 2020 präsentiert das Museum Penzberg – Sammlung Campendonk Hinterglasbilder von 1910 bis 1960 als Abschlussausstellung des Forschungsprojektes. Die Ausstellung wird von einem ausführlichen Katalog über die fächerübergreifende Forschung begleitet. Für Herbst 2020 ist ein Hinterglas-Symposium an der Bundesanstalt für Materialforschung und -prüfung (BAM) Berlin im Rahmen eines N.i.Ke. Workshops geplant.
A reliable analysis of aerosol particle is curial for enforcing EU air quality regulations to protect human health, and for research on climate change effects [1]. Although metrics such as PM10 and PM2.5 are currently in use, the level of uncertainty of aerosol metrics is too high and the traceability is insufficient. Within the AEROMET project [2] procedures are developed aiming at reducing the uncertainties of particle mass, size, and number concentration measurements including the characterization of regulated components in airborne particles. Here, we present an approach how to improve the uncertainties of the particle mass by mobile total reflection x-ray fluorescence (TXRF) analysis. The combination of TXRF and aerosols sampling techniques supported by reference-free synchrotron radiation-based XRF enables a quantitative real-time analysis of particle mass. During in-field campaigns, the procedure was tested, monitoring the size dependent mass concentrations of specific elements in ambient aerosols under dynamic conditions. This approach allows a direct time and size-resolved analysis without laborious digestion steps and a reduced risk of contamination.
Aerosol particles were sampled in a 13-stage DLPI impactor on acrylic discs. TXRF analysis was performed on-site with the transportable spectrometer S2 PICOFOX (Bruker Nano GmbH). The TXRF quantification was based on internal standardization. At moderate air pollution levels (PM10 20 µg/m³) sampling times of less than 2 hours were enough to detect elements in different particle size bins. The on-site approach and the high sensitivity of TXRF enables the observation of rather quick changes in the quantity and distribution of elements in an ambient aerosol on the day of sampling. The analysis of the morning and afternoon sampling shifts reveals the occurrence of the elements Fe, Ca and Si in different size bins as well as their temporal change in respective mass concentrations over the day while the distributions of several other elements remain unchanged.
Ein Ringversuch ist eine Methode der externen Qualitätssicherung für sowohl Messverfahren als auch die Eignung von Mess- und Prüflaboratorien. Dabei werden möglichst identische Proben mit identischen Verfahren oder mit unterschiedlichen Verfahren untersucht. Die Teilnahme an Ringversuchen ist für alle akkreditierten Laboratorien ein wesentlicher Baustein der institutsinternen und -externen Qualitätssicherung. Je mehr Faktoren Einfluss auf das Endergebnis haben können, desto wichtiger ist es diese Faktoren zu kennen und deren Einflüsse durch weitestgehende Vergleichbarkeit der Messverfahren zu minimieren.
Für die Bewertung von Emissionen aus Bauprodukten haben sich Emissionsmess-kammer-Untersuchungen als Messverfahren etabliert. Um den Geruch auch für Innenraumanwendungen bewertbar zu machen sind in den letzten Jahren erste Normen entstanden (z. B. DIN ISO 16000-28 und VDI 4302-1). Um die Eignung dieser Normen und deren Anwendung zu untersuchen sind seit 2012 nunmehr 3 Ringversuche der BAM auch zum Thema Geruch durchgeführt worden.
Die bislang durchgeführten Ringversuche mit dem Ziel der Geruchsbewertung wiesen Standardabweichungen von ca. 24 bis 30 % um den jeweiligen Mittelwert auf und wiesen somit ähnliche Werte auf, die auch bei den VOC-Vergleichen ermittelt wurden. Diese Entwicklungen werden hier aufgezeigt.
Bacterial samples are typically freeze dried or cryo-prepared prior to XPS analysis to allow for measurements in ultra-high vacuum (UHV). The sample environment in the near-ambient pressure (NAP) XPS instrument EnviroESCA allows for measurements in up to 15 mbar water vapor, thus, sample preparation is no longer restricted to UHV-compatible techniques. For instance, biofilms grown in medium can be transferred directly from the medium to the measurements chamber, maintaining a humid environment throughout the measurements. Considering the complexity of bacterial samples, sample preparation must be carefully considered in order to obtain meaningful and reproducible results.
In this talk, various strategies for sample preparation of bacteria and biofilms for NAP-XPS measurements will be discussed. Model systems of planktonic bacteria, artificial biofilms resembling the exopolysaccharide matrix and biofilms have been characterised in various conditions. The stability and homogeneity of the samples was assessed by monitoring the C1s core level peak at different sample locations. The quality of the XPS-spectra is also influenced by the gas environment, which will be exemplified by core level spectra of P. Fluorescens acquired in air, water vapor and ultra-high vacuum.
