Unter Berücksichtigung praktischer und wissenschaftlich-technischer Erkenntnisse der letzten 15 Jahre - insbesondere auch auf der Basis verschiedener BAM-Forschungsergebnisse und Zulassungsverfahren - erfolgte eine Überarbeitung der Grundlagen, Bauprinzipien, Ausführungs- und Einbaukriterien sowie der Zulassungs- und Gütesicherungsmaßnahmen für insgesamt 4 Bauarten von Fugenfüllsystemen für hochbeanspruchte Verkehrsflächen. In diesem - für den Bundesfernstraßenbereich verbindlichen bautechnischen Regelwerk - finden insbesondere auch die Ergebnisse mehrerer wissenschaftlicher Vorhaben der BAM bezüglich labortechnischer Untersuchungen und Felduntersuchungen zum Funktionsverhalten und zur Dauerhaftigkeit erste baurechtliche Konsequenzen. Auf der Basis von Untersuchungen u.a. im Brückenbau wurden Regelungen für die Fugenausbildung in Asphalt und bei Befestigungskombinationen erarbeitet und umgesetzt. Damit konnten umfassende Regelungen für die maßgebenden Fahrbahnbeläge im Bundesfernstraßenbereich der Straßenbauverwaltung und ausführenden Industrieunternehmungen zur Verfügung gestellt werden. Die Neufassung dieser Regelungen dient darüber hinaus dem Ziel, den von der BAM vertretenen Wechsel hin zu einer performance-bezogenen Bewertungsmethodik, vorzubereiten.
A composite material is a combination of two or more materials with very different mechanical, thermal and electrical properties. The various forms of composite materials, due to their high material properties, are widely used as structural materials in the aviation, space, marine, automobile, and sports industries. However, some defects like voids, delamination, or inhomogeneous fiber distribution that form during the fabricating processes of composites can seriously affect the mechanical properties of the composite material. In this study, several imaging NDE techniques such as: thermography, high frequency eddy current, ultrasonic, x-ray radiography, x-ray laminography, and high resolution x-ray CT were conducted to characterize the microstructure of carbon fiber composites. Then, a 3D analysis was implemented by the destructive technique of serial sectioning for the same sample tested by the NDE methods. To better analyze the results of this work and extract a clear volume image for all features and defects contained in the composite material, an intensive comparison was conducted among hundreds of 3D-NDE and multi serial sections’ scan images showing the microstructure variation.
The solid lubricant molybdenum disulfide is a well known material for extreme conditions where conventional oil or grease lubrication is not feasible (vacuum, high pressures). However, under reactive environments e.g. high humidity or hydrogen MoS2 is prone to premature failure. By using a PVD process, deposition parameters can be optimized to gain coatings with a more textured structure orienting the basal planes parallel to the Substrate surface, A similar concept has been used by Koch et al. for coatings developed to withstand the extreme operational conditions of the WENDELSTEIN W7-X fusion experiment. Therefore, an improved tribological behavior even under adverse conditions can be achieved.
Nebivolol (NEB), a β-blocker frequently used to treat cardiovascular diseases, has been widely detected in aquatic environments, and can be degraded under exposure to UV radiation, leading to the formation of certain transformation products (UV-TPs). Thus, the toxic effects of NEB and its UV-TPs on aquatic organisms are of great importance for aquatic ecosystems. In the present study, the degradation pathway of NEB under UV radiation was investigated. Subsequently, zebrafish embryos/larvae were used to assess the median lethal concentration (LC50) of NEB, and to clarify the sub-lethal effects of NEB and its UV-TPs for the first time. It was found that UV radiation could reduce the toxic effects of NEB on the early development of zebrafish. Transcriptomic analysis identified the top 20 enriched Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways in zebrafish larvae exposed to NEB, most of which were associated with the antioxidant, nervous, and immune systems. The number of differentially expressed genes (DEGs) in the pathways were reduced after UV radiation. Furthermore, the analysis of protein biomarkers, including CAT and GST (antioxidant response), AChE and ACh (neurotoxicity), CRP and LYS (immune response), revealed that NEB exposure reduced the activity of these biomarkers, whereas UV radiation could alleviate the effects. The present study provides initial insights into the mechanisms underlying toxic effects of NEB and the detoxification effects of UV radiation on the early development of zebrafish. It highlights the necessity of considering the toxicity of UV-TPs when evaluating the toxicity of emerging pollutants in aquatic systems.
For the vast majority of ellipsometric measurements, the application of planar substrates is mandatory and requires a proper sample alignment prior to the measurement. Here, the authors present a generalized approach of how to extract the isotropic ellipsometric sample parameters from an imaging Mueller-matrix measurement even if the sample is significantly misaligned. They validate the method by layer-thickness calculations based on imaging Mueller-matrix measurements of flat crystalline silicon samples that were misaligned on purpose. Furthermore, they also exploit this method’s capabilities to perform spatially resolved layer-thickness measurements of a single-layer indium-tin-oxide coating on a fused-silica microlens without the need of realignment or repositioning of the sample during the measurement.
Nach mehr als 15-jähriger Anwendung und mit der zwischenzeitlichen Einführung europäisch harmonisierter Normen für Fugenmaterialien war eine Anpassung Qualitäts- und Gütesicherungsanforderungen für Fugenfüllungen im Bundesfernstraßenbereich erforderlich. Hierbei ging es insbesondere darum, Maßnahmen und Verfahren zu entwickeln, um das nationale Qualitäts- und Gütesicherungsniveau weiter zu entwickeln und weiterhin sicher stellen zu können. Nach den Untersuchungserfahrungen im Labor und im Feld ging darum, neuartige Material- und Stoffkombinationen qualitätssicher dem Bauprozess zur Verfügung stellen zu können. Erforderliche Anforderungskriterien und zulässige Toleranzen der Lieferqualität wurden neu gefasst. Es wurde ein Verfahren entwickelt, europäische Bauprodukte qualitätsgesichert in den Bauprozess im Bundesfernstraßenbericht zu überführen.
Zeolitic imidazolate framework (ZIF) hybrid fluorescent nanoparticles and ZIF antibody conjugates have been synthesized, characterized, and employed in lateral-flow immunoassay (LFIA). The bright fluorescence of the conjugates and the possibility to tailor their mobility gives a huge potential for diagnostic assays. An enzyme-linked immunosorbent assay (ELISA) with horseradish peroxidase (HRP) as label, proved the integrity, stability, and dispersibility of the antibody conjugates, LC-MS/MS provided evidence that a covalent link was established between these metal-organic frameworks and lysine residues in IgG antibodies.
For many heterogeneous sensor applications as well as the synthesis of hapten antigens to produce antibodies, protein conjugates of the target substance are essential. A requirement is that the target substance already offers or is modified to contain a functionality that allows for coupling to a protein, that is, an amino acid residue. Ideally, to avoid shielding of the compound by the carrier protein, a sufficient distance to the protein surface should be provided. With its carboxyl function diclofenac (DCF) allows for direct binding to lysine residues after in situ synthesis of the NHS ester. One problem is that diclofenac as free acid tends to autocondensation, which results in low yields. Here we describe the 'insertion' of a C6 spacer via synthesis of the amide with 6-aminohexanoic acid. To carry out the reaction in solution, first the methyl ester of the amino acid had to be produced. Due to otherwise low yields and large cleaning efforts, solid-phase synthesis on Fmoc Ahx Wang resin is recommended. The crude product is mainly contaminated by cleavage products from the resin which were removed by chromatography. The structure of the highly pure hapten was completely determined by nuclear magnetic resonance (NMR) spectroscopy.
In Deutschland fallen mittelfristig erhebliche Mengen von Mauerwerkbruch an. Aus dem jährlichen Bauschuttanfall und den Produktionszahlen von Wandbausteinen lassen sich die Mengen an Mauerwerksrestmassen ableiten, die als Rückbaumaterial bei Abriss-, Umbau und Sanierungsarbeiten in Deutschland anfallen werden. Hiernach ist pro Jahr mit bis zu 10 Millionen Tonnen heute vornehmlich heterogen anfallendem Mauerwerkbruch zu rechnen [1a]. Im Sinne einer nachhaltigen Kreislaufwirtschaft sind diese Restmassen zukünftig einem möglichst hochwertigen Recycling zuzuführen. In diesem Spannungsfeld führt die deutsche Mauersteinindustrie das breit aufgestellte gemeinschaftliche Forschungsprojekt "SIM Stoffkreislauf im Mauerwerksbau" durch. In insgesamt acht mauersteinübergreifenden IGFForschungsvorhaben wurden und werden technisch orientierte Lösungsmöglichkeiten für das Recycling von Mauerwerk erarbeitet und Anwendungsbereiche für Mauerwerksrestmassen aufgezeigt. Eine grundsätzliche Entscheidung über die Eignung von Mauerwerksrestmassen für bestimmte Recyclingwege kann es letztlich nicht geben. Es wird immer eine Einzelfallentscheidung bleiben, die wesentlich durch komplexe technische sowie ökologische und ökonomische Randbedingungen geprägt wird.
Vor diesem Hintergrund wurde im Rahmen des IGF-Forschungsvorhabens 17022 N der Forschungsvereinigung Porenbetonindustrie e.V., Berlin die Nachhaltigkeit verschiedener Recyclingoptionen für Mauerwerkbruch ganzheitlich untersucht und bewertet. Unter Einbeziehung der Hauptwandbausteinarten Ziegel, Kalksandstein, Porenbeton und Leichtbeton wurden dabei material- und verfahrenstechnische sowie ökologische, ökonomische und soziale Aspekte berücksichtigt. Auf Grundlage der gewonnenen Erkenntnisse wurde ein EDV-gestütztes Analysetool (Web-Tool) zur Nachhaltigkeitsbewertung für das Mauerwerksrecycling entwickelt. Mit dem Web-Tool sollen Anwender (Planer, Architekten, Recycler) die Möglichkeit erhalten, für verschiedene Untersuchungsfälle unter Einbeziehung regional und sachlich bedingter Parameter, wie Materialqualität, Stoffmengen, Logistik, Aufbereitungstechnik, Anwendungsbereiche, Kosten und Erlöse, objektspezifische Aussagen zur Nachhaltigkeit treffen zu können. Auf diesem Wege können im Sinne der aktuellen gesetzlichen Vorgaben nachhaltige Stoffkreisläufe im Mauerwerksbau geschlossen und somit letztlich die Deponierung von Mauerwerksrestmassen vermieden werden.
Alle vier Wandbaustoffarten haben sich im Rahmen des Forschungsvorhabens als recyclingfähig und hinsichtlich der Nachhaltigkeit als gleichwertig erwiesen. Das Ziel des Vorhabens wurde damit erreicht.
Single Femtosecond Laser-Pulse-Induced Superficial Amorphization and Re-Crystallization of Silicon

Superficial amorphization and re-crystallization of silicon in <111> and <100> orientation after irradiation by femtosecond laser pulses (790 nm, 30 fs) are studied using optical imaging and transmission electron microscopy. Spectroscopic imaging ellipsometry (SIE) allows fast data acquisition at multiple wavelengths and provides experimental data for calculating nanometric amorphous layer thickness profiles with micrometric lateral resolution based on a thin-film layer model. For a radially Gaussian laser beam and at moderate peak fluences above the melting and below the ablation thresholds, laterally parabolic amorphous layer profiles with maximum thicknesses of several tens of nanometers were quantitatively attained. The accuracy of the calculations is verified experimentally by high-resolution transmission electron microscopy (HRTEM) and energy dispersive X-ray spectroscopy (STEM-EDX). Along with topographic information obtained by atomic force microscopy (AFM), a comprehensive picture of the superficial re-solidification of silicon after local melting by femtosecond laser pulses is drawn.
This paper addresses the quantification of the value of structural health monitoring (SHM) before its implementation for structural systems on the basis of its Value of Information (VoI). The value of SHM is calculated utilizing the Bayesian pre-posterior decision analysis modelling the structural life cycle performance, the integrity management and the structural risks. The relevance and precision of SHM information for the reduction of the structural system risks and the expected cost of the structural integrity management throughout the life cycle constitutes the value of SHM and is quantified with this framework. The approach is focused on fatigue deteriorating structural steel systems for which a continuous resistance deterioration formulation is introduced. In a case study, the value of SHM for load monitoring is calculated for a Daniels system subjected to fatigue deterioration. The influence of and the value of SHM in regard to the structural system risks and the integrity management is explicated and explained. The results are pointing to the importance of the consideration of the structural system risks for the quantification of the value of SHM.
Quantification of nanoparticle (NP) uptake in cells or tissues is very important for safety assessment. Often, electron microscopy based approaches are used for this purpose, which allow imaging at very high resolution. However, precise quantification of NP numbers in cells and tissues remains challenging. The aim of this study was to present a novel approach, that combines precise quantification of NPs in individual cells together with high resolution imaging of their intracellular distribution based on focused ion beam/ scanning electron microscopy (FIB/SEM) slice and view approaches.
This report focuses on the characterization of CD4 expression level in terms of equivalent number of reference fluorophores (ERF). Twelve different flow cytometer platforms across sixteen laboratories were utilized in this study. As a first step the participants were asked to calibrate the fluorescein isothiocyanate (FITC) channel of each flow cytometer using commercially available calibration standard consisting of five populations of microspheres. Each population had an assigned value of equivalent fluorescein fluorophores (EFF denotes a special case of the generic term ERF with FITC as the reference fluorophore). The EFF values were assigned at the National Institute of Standards and Technology (NIST). A surface-labelled lyophilized cell preparation was provided by the National Institute of Biological Standards and Control (NIBSC), using human peripheral blood mononuclear cells (PBMC) pre-labeled with a FITC conjugated anti-CD4 monoclonal antibody. Three PBMC sample vials, provided to each participant, were used for the CD4 expression analysis. The PBMC are purported to have a fixed number of surface CD4 receptors. On the basis of the microsphere calibration, the EFF value of the PBMC samples was measured to characterize the population average CD4 expression level of the PBMC preparations. Both the results of data analysis performed by each participant and the results of centralized analysis of all participants' raw data are reported. Centralized analysis gave a mean EFF value of 22,300 and an uncertainty of 750, corresponding to 3.3% (level of confidence 68%) of the mean EFF value. The next step will entail the measurement of the ERF values of the lyophilized PBMC stained with labels for other fluorescence channels. The ultimate goal is to show that lyophilized PBMC is a suitable biological reference cell material for multicolor flow cytometry and that it can be used to present multicolor flow cytometry measurements in terms of ABC (antibodies bound per cell) units.
A surface-labeled lyophilized lymphocyte (sLL) preparation has been developed using human peripheral blood mononuclear cells prelabeled with a fluorescein isothiocyanate conjugated anti-CD4 monoclonal antibody. The sLL preparation is intended to be used as a reference material for CD4+ cell counting including the development of higher order reference measurement procedures and has been evaluated in the pilot study CCQM-P102. This study was conducted across 16 laboratories from eight countries to assess the ability of participants to quantify the CD4+ cell count of this reference material and to document cross-laboratory variability plus associated measurement uncertainties. Twelve different flow cytometer platforms were evaluated using a standard protocol that included calibration beads used to obtain quantitative measurements of CD4+ T cell counts. There was good overall cross-platform and counting method agreement with a grand mean of the laboratory calculated means of (301.7 ± 4.9) µL-1 CD4+ cells. Excluding outliers, greater than 90% of participant data agreed within ±15%. A major contribution to variation of sLL CD4+ cell counts was tube to tube variation of the calibration beads, amounting to an uncertainty of 3.6%. Variation due to preparative steps equated to an uncertainty of 2.6%. There was no reduction in variability when data files were centrally reanalyzed. Remaining variation was attributed to instrument specific differences. CD4+ cell counts obtained in CCQM-P102 are in excellent agreement and show the robustness of both the measurements and the data analysis and hence the suitability of sLL as a reference material for interlaboratory comparisons and external quality assessment.
Gastric cancer (GC) is the 3rd deadliest cancer worldwide, due to limited treatment options and late diagnosis. Human epidermal growth factor receptor-2 (HER2) is overexpressed in similar to 20% of GC cases and anti-HER2 antibody trastuzumab in combination with conventional chemotherapy, is recognized as standard therapy for HER2-positive metastatic GC. This strategy improves GC patients' survival by 2-3 months, however its optimal results in breast cancer indicate that GC survival may be improved. A new photoimmunoconjugate was developed by conjugating a porphyrin with trastuzumab (Trast: Porph) for targeted photodynamic therapy in HER2-positive GC. Using mass spectrometry analysis, the lysine residues in the trastuzumab structure most prone for porphyrin conjugation were mapped. The in vitro data demonstrates that Trast: Porph specifically binds to HER2-positive cells, accumulates intracellularly, co-localizes with lysosomal marker LAMP1, and induces massive HER2-positive cell death upon cellular irradiation. The high selectivity and cytotoxicity of Trast: Porph based photoimmunotherapy is confirmed in vivo in comparison with trastuzumab alone, using nude mice xenografted with a HER2-positive GC cell line. In the setting of human disease, these data suggest that repetitive cycles of Trast: Porph photoimmunotherapy may be used as an improved treatment strategy in HER2-positive GC patients.
The synthesis of a novel PS conjugated with bovine and human serum albumin (BSA and HSA) and a monoclonal antibody anti-CD104 is reported, as well as their biological potential against the human bladder cancer cell line UM-UC-3. No photodynamic effect was detected when the non-conjugated porphyrin was used. Yet, when it was coupled covalently with the mAb anti-CD104, BSA and HSA, the resulting photosensitizer conjugates demonstrated high efficacy in destroying the cancer cells, the mAb anti-CD104 efficacy overruling the albumins.
The optical properties of semiconductor nanocrystals (SC NCs) are largely controlled by their size and surface chemistry, i.e., the chemical composition and thickness of inorganic passivation shells and the chemical nature and number of surface ligands as well as the strength of their bonds to surface atoms.
The latter is particularly important for CdTe NCs, which – together with alloyed CdₓHg₁₋ₓTe – are the only SC NCs that can be prepared in water in high quality without the need for an additional inorganic passivation shell. Aiming at a better understanding of the role of stabilizing ligands for the control of the application-relevant fluorescence features of SC NCs, we assessed the influence of two of the most commonly used monodentate thiol ligands, thioglycolic acid (TGA) and mercaptopropionic acid (MPA),
on the colloidal stability, photoluminescence (PL) quantum yield (QY), and PL decay behavior of a set of CdTe NC colloids. As an indirect measure for the strength of the coordinative bond of the ligands to SC NC surface atoms, the influence of the pH (pD) and the concentration on the PL properties of these colloids was examined in water and D₂O and compared to the results from previous dilution studies with a set of thiol-capped Cd₁₋ₓHgₓTe SC NCs in D₂O. As a prerequisite for these studies, the number of surface ligands was determined photometrically at different steps of purification after SC NC synthesis with Ellman’s test.
Our results demonstrate ligand control of the pH-dependent PL of these SC NCs, with MPA-stabilized CdTe NCs being less prone to luminescence quenching than TGA-capped ones. For both types of CdTe colloids, ligand desorption is more pronounced in H₂O compared to D₂O, underlining also the role of hydrogen bonding and solvent molecules.
Under the auspices of the Protein Analysis Working Group (PAWG) of the Comité Consultatif pour la Quantité de Matière (CCQM) a pilot study, CCQM-P216, was coordinated by the Chinese National Institute of Metrology (NIM), National Research Council of Canada (NRC) and the Bureau International des Poids et Mesures (BIPM). Eleven Metrology Institutes or Designated Institutes and the BIPM participated in the first phase of the pilot study (Part 1). The purpose of this pilot study was to develop measurement capabilities for larger proteins using a recombinant humanized IgG monoclonal antibody against Spike glycoprotein of SARS-CoV-2 (Anti-S IgG mAb) in solution. The first phase of the study was designed to employ established methods that had been previously studies by the CCQM Protein Analysis Working Group, involving the digestion of protein down to the peptide or amino acid level. The global coronavirus pandemic has also led to increased focus on antibody quantitation methods. IgG are among the immunoglobulins produced by the immune system to provide protection against SARS-CoV-2. Anti-SARS-CoV-2 IgG can therefore be detected in samples from affected patients. Antibody tests can show whether a person has been exposed to the SARS-CoV-2, and whether or not they potentially show lasting immunity to the disease. With the constant spread of the virus and the high pressure of re-opening economies, antibody testing plays a critical role in the fight against COVID-19 by helping healthcare professionals to identify individuals who have developed an immune response, either via vaccination or exposure to the virus. Many countries have launched large-scale antibody testing for COVID-19. The development of measurement standards for the antibody detection of SARS-CoV-2 is critically important to deal with the challenges of the COVID-19 pandemic. In this study, the SARS-CoV-2 monoclonal antibody is being used as a model system to build capacity in methods that can be used in antibody quantification. Amino acid reference values with corresponding expanded uncertainty of 36.10 ± 1.55 mg/kg, 38.75 ± 1.45 mg/kg, 18.46 ± 0.78 mg/kg, 16.20 ± 0.67 mg/kg and 30.61 ± 1.30 mg/kg have been established for leucine, valine, phenylalanine, isoleucine and proline, respectively. Agreement between nearly all laboratories was achieved for the amino acid analysis within 2 to 2.5 %, with one participant achieving markedly higher results due to a technical issue found in their procedure; this result was thus excluded from the reference value calculations. The relatively good agreement within a laboratory between different amino acids was not dissimilar to previous results for peptides or small proteins, indicating that factors such as hydrolysis conditions and calibration procedures could be the largest sources of variability. Peptide reference values with corresponding expanded uncertainty of 4.99 ± 0.28 mg/kg and 6.83 ± 0.65 mg/kg have been established for ALPAPIEK and GPSVFPLAPSSK, respectively. Not surprisingly due to prior knowledge from previous studies on peptide quantitation, agreement between laboratories for the peptide-based analysis was slightly poorer at 3 to 5 %, with one laboratory's result excluded for the peptide GPSVFPLAPSSK. Der Festschmierstoff Molybdändisulfid ist insbesondere für Extremanwendungen ein gerne eingesetzter Werkstoff, problematisch ist jedoch der negative Einfluss von Feuchtigkeit. Bei mittels PVD-Verfahren abgeschiedenen MoS2-Schichten ergibt sich jedoch die Möglichkeit, durch Anpassung der Beschichtungsparameter vorwiegend basal ausgerichtete Schichten herzustellen, die auch an Luft ein verbessertes tribologisches Verhalten zeigen.
Embedding of information on surfaces is state of the art for identification testing in which public, hidden and forensic features are used. In many instances, the legal authentication of a product, a material or a document is required. Among the surface-based encoded labels, bar codes and data matrices are most frequently applied. They are publicly visible. The material itself is irrelevant, only a sufficient optical contrast is required.
However, a strong material dependence of the label can be achieved by means of Fabry-Perot layer stacks. Stack designs are described with regard to all three security levels: public features (e.g. color and tilt effect) perceptible by the human eye, hidden features (e.g. optical response in a given spectral range) detectable by commonly available instruments and forensic features (ellipsometric quantities Ψ and Δ as a function of wavelength λ and angle of incidence AOI) only detectable by sophisticated instruments.
Regarding material-correlated authentication, ellipsometric quantities Ψ and Δ are used as encoded forensic features for the first time. Hence, Fabry-Perot layer stacks as information carriers in combination with imaging ellipsometry as optical read-out system provide all-in-one anti-counterfeiting capabilities.
In Deutschland fällt mittelfristig eine nicht unerhebliche Menge an Mauerwerksbruch an. Im Sinne einer nachhaltigen Kreislaufwirtschaft sind diese Restmassen zukünftig einem möglichst hochwertigen Recycling zuzuführen. Im Rahmen des IGF-Forschungsvorhabens 17022 N der Forschungsvereinigung Porenbetonindustrie e.V., Berlin, wird im Gesamtkontext SIM Stoffkreislauf im Mauerwerksbau die Nachhaltigkeit verschiedener Recyclingoptionen für Mauerwerksbruch unter Einbeziehung der Hauptwandbausteinarten Ziegel, Kalksandstein, Porenbeton und Leichtbeton sowie unter Beachtung material- und verfahrenstechnischer sowie ökologischer, ökonomischer und sozialer Aspekte ganzheitlich untersucht und bewertet. Auf Grundlage der dabei gewonnenen Erkenntnisse wird ein rechnergestütztes Analysetool zur Nachhaltigkeitsbewertung für das Mauerwerksrecycling entwickelt. Auf diesem Wege sollen im Sinne der aktuellen gesetzlichen Vorgaben nachhaltige Stoffkreisläufe im Mauerwerksbau geschlossen und somit letztlich die Deponierung von Mauerwerksrestmassen vermieden werden.-------------------------------------------------------------------------------------------------------------------------------------------------
In Germany, a not unimportant quantity of masonry rubble is generated in the medium term. With regard to a sustainable closed substance cycle waste management, these rest masses have to be recycled if possible. Within the IGF research project Sustainability Analysis for the Recycling of Masonry, different recycling options will be assessed with regard to their sustainability performance. In doing so, the main kinds of masonry units (clay units, calcium silicate units, autoclaved aerated concrete units, aggregate concrete units), their material properties, the processing technologies as well as ecological, economic and social aspects will be considered. Based on the results, a web-based tool will be developed to assess the sustainability of different recycling options for masonry rubble.
Inconel 718 is a precipitation strengthened, nickel-based super alloy of interest for the Additive Manufacturing (AM) of low volume, complex parts to reduce production time and cost compared to conventional subtractive processes. The AM process involves repeated rapid melting, solidification and reheating, which exposes the material to non-equilibrium conditions that affect elemental segregation and the subsequent formation of solidification phases, either beneficial or detrimental. These variations are difficult to characterize due to the small length scale within the micron sized melt pool. To understand how the non-equilibrium conditions affect the initial solidification phases and their critical temperatures, a multi-length scale, multi modal approach has been taken to evaluate various methods for identifying the initial phases formed in the as-built Inconel 718 produced by laser-powder bed fusion (L-PBF) additive manufacturing (AM). Using a range of characterization tools from the bulk differential thermal analysis (DTA) and x-ray diffraction (XRD) to spatially resolved images using a variety of electron microscopy tools, a better understanding is obtained of how these minor phases can be properly identified regarding the amount and size, morphology and distribution. Using the most promising characterization techniques for investigation of the as-built specimens, those techniques were used to evaluate the specimens after various heat treatments. During the sequence of heat treatments, the initial as-built dendritic structures recrystallized into well-defined grains whose size was dependent on the temperature. Although the resulting strength was similar in all heat treated specimens, the elongation increased as the grain size was refined due to differences in the precipitated phase distribution and morphology.
The existence of microbial activity hotspots in temperate regions of Earth is driven by soil heterogeneities, especially the temporal and spatial availability of nutrients. Here we investigate whether microbial activity hotspots also exist in lithic microhabitats in one of the most arid regions of the world, the Atacama Desert in Chile. While previous studies evaluated the total DNA fraction to elucidate the microbial communities, we here for the first time use a DNA separation approach on lithic microhabitats, together with metagenomics and other analysis methods (i.e., ATP, PLFA, and metabolite analysis) to specifically gain insights on the living and potentially active microbial community. Our results show that hypolith colonized rocks are microbial hotspots in the desert environment. In contrast, our data do not support such a conclusion for gypsum crust and salt rock environments, because only limited microbial activity could be observed. The hypolith community is dominated by phototrophs, mostly Cyanobacteria and Chloroflexi, at both study sites. The gypsum crusts are dominated by methylotrophs and heterotrophic phototrophs, mostly Chloroflexi, and the salt rocks (halite nodules) by phototrophic and halotolerant endoliths, mostly Cyanobacteria and Archaea. The major environmental constraints in the organic-poor arid and hyperarid Atacama Desert are water availability and UV irradiation, allowing phototrophs and other extremophiles to play a key role in desert ecology.
Die Infrastruktursysteme der Industriestaaten erfordern heute und in Zukunft ein effizientes Management bei alternder Bausubstanz, steigenden Lasten und gleichbleibend hohem Sicherheitsniveau. Digitale Technologien bieten ein großes Potenzial zur Bewältigung der aktuellen und künftigen Herausforderungen im Infrastrukturmanagement. Im BMBF-geförderten Projekt Bewertung alternder Infrastrukturbauwerke mit digitalen Technologien (AISTEC) wird untersucht, wie unterschiedliche Technologien und deren Verknüpfung gewinnbringend eingesetzt werden können. Am Beispiel der Maintalbrücke Gemünden werden ein sensorbasiertes Bauwerksmonitoring, bildbasierte Inspektion mit durch Kameras ausgestatteten Drohnen (UAS) und die Verknüpfung digitaler Bauwerksmodelle umgesetzt. Die aufgenommenen Bilder dienen u. a. als Grundlage für spätere visuelle Anomaliedetektionen und eine 3D-Rekonstruktion, welche wiederum für die Kalibrierung und Aktualisierung digitaler Tragwerksmodelle genutzt werden. Kontinuierlich erfasste Sensordaten werden ebenfalls zur Kalibrierung und Aktualisierung der Tragwerksmodelle herangezogen. Diese Modelle werden als Grundlage für Anomaliedetektionen und perspektivisch zur Umsetzung von Konzepten der prädiktiven Instandhaltung verwendet. Belastungsfahrten und historische Daten dienen in diesem Beitrag der Validierung von kalibrierten Tragwerksmodellen.
Dieser Bericht beschreibt ein Systemmodell für eine integrale Ermittlung und Prognose der Schadens- und Zustandsentwicklung der Elemente eines Brückensystems unter Berücksichtigung von Ergebnissen aus Inspektionen und Überwachung. Das Systemmodell wurde anhand eines ausgesuchten Spannbetonüberbaus in einzelliger Kastenbauweise entwickelt. Es besteht aus zwei integralen Teilmodellen: ein Modell zur Beschreibung des Systemschädigungszustandes und ein Modell zur Beschreibung der Standsicherheit. Für die Modellierung des stochastischen Systemschädigungszustandes eines Brückensystems werden dynamische Bayes'sche Netze (DBN) vorgeschlagen. Dieser Ansatz ermöglicht es, alle relevanten Schädigungsprozesse und deren stochastische Abhängigkeiten zu berücksichtigen. Ein wesentlicher Vorteil dieses Ansatzes ist es, dass DBN ideal dafür geeignet sind, Bayes'sche Aktualisierungen auf Grundlage von Informationen aus Inspektionen und Überwachungsmaßnahme auf eine effiziente und robuste Art und Weise durchzuführen. Der DBN-Ansatz ist deshalb für die Entwicklung von Software für das Erhaltungsmanagement von alternden Brückenbauwerken, die vom Benutzer keine vertieften Kenntnisse der Zuverlässigkeitstheorie verlangt, ideal geeignet. Für die Modellierung der Standsicherheit eines alternden Kastenträgers wird vereinfachend Biegeversagen des globalen Längssystems betrachtet. Zur Berechnung der maximalen Traglast eines Kastenträgers infolge des Systemschädigungszustandes wird ein plastisch-plastisches Verfahren eingesetzt, wobei die Beanspruchungen mittels der Fließgelenktheorie unter Ausnutzung der plastischen Beanspruchbarkeit der Querschnitte des Kastenträgers ermittelt werden. Ein Kastenträger versagt, wenn sich durch die Ausbildung einer ausreichend großen Anzahl von Fließgelenken eine kinematische Kette ausbildet. Dieser Modellierungsansatz berücksichtigt Redundanzen, die sich aus der plastischen Beanspruchbarkeit der Querschnitte und der statischen Unbestimmtheit eines Kastenträgers ergeben. Zum Nachweis der praktischen Einsetzbarkeit des entwickelten Systemmodells wurde ein Software-Prototyp entwickelt, der eine intuitiv benutzbare graphische Benutzeroberfläche (Front-End) mit einem Berechnungskern (Back-End) koppelt. Die aktuelle Version des Software-Prototyps implementiert ein Modell der chloridinduzierten Bewehrungskorrosion und ein Tragwerksmodell, welches das Verfahrens der stetigen Laststeigerung zur Bestimmung der maximalen Traglast des Kastenträgers auf der Grundlage eines Finite-Elemente-Modells umsetzt. Zur Durchführung von Bayes'schen Aktualisierungen des Systemschädigungszustandes auf der Grundlage des DBN-Modells implementiert der Prototyp den Likelihood-Weighting-Algorithmus. Die entwickelte Architektur des Prototyps ermöglicht eine Erweiterung der Software um weitere Schädigungsprozesse. Der entwickelte Software-Prototyp ermöglicht Benutzern ohne vertiefte Kenntnisse der Zuverlässigkeitstheorie eine Berechnung des Einflusses von Bauwerksinformationen auf den Systemschädigungszustand und die Tragsicherheit eines Kastenträgers. Auf dieser Grundlage können effiziente Inspektions- und Überwachungsmaßnahmen identifiziert und das Erhaltungsmanagement optimiert werden.
Bright and stable near-infrared (NIR) and infrared (IR) emitting chromophores are in high demand for applications in telecommunication, solar cells, security barcodes, and as fluorescent reporters in bioimaging studies. The best choice for wavelengths >750 nm are semiconductor nanocrystals, especially ternary or alloy nanocrystals like CdHgTe, which enable size and composition control of their optical properties. Here, we report on the influence of growth time and surface chemistry on the composition and optical properties of colloidal CdHgTe. Up to now, these are the only NIR and IR emissive quantum dots, which can be synthesized in high quality in water, using a simple one-pot reaction. For this study we utilized and compared three different thiol ligands, thioglycolic acid (TGA), 3-mercaptopropionic acid (MPA), and glutathione (GSH). Aiming at the rational design of bright NIR- and IR-emissive alloy materials, special emphasis was dedicated to a better understanding of the role of the surface ligand and adsorptiondesorption equilibria on the photoluminescence quantum yield and stability. In this respect, dilution and protonation studies were performed. Our results show that with this simple synthetic procedure, strongly fluorescent CdHgTe colloids can be obtained with MPA as stabilizing ligand revealing quantum yields as high as 45% independent of particle concentration.
The effects of long-term application of different organic amendments, as compared to mineral fertilizer, on Zn, Cu and Pb content and leachability in a luvisol derived from loess were assessed. The organic fertilizers, applied since 1962, were compost (COM) – from green organic household waste, sewage sludge (SLU) – from municipal water treatment facilities, farmyard manure (FYM) and the doses applied since 1997 were 90 t ha-1, 10 t ha-1 and 9 t ha-1, once in 3 years, respectively. The kinetics of metals extraction with 0.05 mol dm-3 EDTA at pH 6.0 has been studied. The two first-order reactions model was fitted to the kinetic data and allowed to distinguish two pools for each metal: a 'labile' fraction (Q1), quickly extracted with a rate constant k1, and a 'moderately labile' fraction (Q2), more slowly extracted, with a rate constant k2. Simultaneously, the pseudo-total metal contents in the soil samples were determined after digestion with aqua regia (3:1 HCl + HNO3). The obtained parameters Q1, k1, Q2, k2, for the kinetics of extraction of each metal in the three replicates of each fertilization mode, as well as the pseudo-total metal contents, were statistically analysed. COM and SLU application resulted in an increase of the total contents of Pb, Zn and Cu in soil. Further, the percentage of labile Zn and Pb also increased in consequence of the application of those amendments, particularly COM. The increase was more noticeable for Zn. FYM, despite not increasing the total content of Pb, Zn or Cu, did also have an effect on the leachability of Zn and Pb, increasing their labile fraction in soil. These results point to a potential risk of increasing metals mobility in soil, mainly Zn, associated to the use of organic amendments, particularly COM or SLU.
Amphibians are undergoing a global decline. One poorly investigated reason could be the pollution of aquatic habitats by endocrine disrupting compounds (EDCs). We tested the susceptibility to the synthetically stabilized estrogen 17α-ethinylestradiol (EE2) in three deeply diverged anuran species, differing in sex determination systems, types of gonadogenesis and larval ecologies. To understand whether data from the amphibian model Xenopus laevis (Pipidae) are analogous and applicable to only distantly related non-model amphibians, tadpoles of X. laevis, Hyla arborea (Hylidae) and Bufo viridis (Bufonidae) were simultaneously exposed to 50, 500 and 5000 ng/L EE2 from hatching until completion of metamorphosis, using a flow-through-system under identical experimental conditions. Comparing molecularly established genetic with histologically assessed phenotypic sex in all species, we have recently shown that EE2 provoked numerous genetic-male-to-phenotypic-female sex reversals and mixed sex individuals, confirming overall its expected feminizing effect. In the present study, we focus on the influence of EE2 on gonadal and somatic development. Anatomy and histology revealed several species-specific effects. In both non-model species, H. arborea and B. viridis, high numbers of anatomically impaired gonads were observed. In H. arborea, exposed to 5000 ng/L EE2, numerous underdeveloped gonads were detected. Whereas EE2 did not alter snout-to-vent length and body weight of X. laevis metamorphs, H. arborea showed a treatment-dependent decrease, while B. viridis exhibited an increase in body weight and snout-to-vent length. Apart from a concentration-dependent occurrence of yellowish skin color in several H. arborea, no organ-specific effects were detected. Since EE2 ubiquitously occurs in many aquatic ecosystems and affects sexual and somatic development, among EDCs, it may indeed contribute to amphibian decline. The inter-species variation in developmental EE2-effects corroborates species-specific vulnerability differences towards EDCs between deeply diverged amphibian groups
In this work, we report an electrochemical immunosensor to detect ethinylestradiol in water samples, using electrochemical impedance spectroscopy (EIS) as a detection technique. For the development of this immunosensor, the direct modification of the working electrode of a screen-printed carbon electrode was carried out. First, to reduce the resistance of the electrode, electroreduced graphene was incorporated on the surface.
Second, a porous gold structure was electrodeposited on reduced graphene by electrodeposition and the dynamic hydrogen bubble template assisted method. Thus, a marked increase in surface area was obtained for anti-EE2 antibodies immobilization. Subsequently, the specific anti-EE2 antibodies were covalently immobilized using α-lipoic acid for attaching them to the gold surface.
The electrode modified with the antibodies was incubated for 30 min in the samples containing EE2, producing the specific Antigen antibody binding. As the charge transfer resistance of a redox probe in the electrode surface is governed by the surface blocking effects, the charge transfer resistance was related to the amount of EE2 captured to realize a quantitative determination. For this, the EIS measurements were performed in a 4 mM [Fe(CN)6]4−/3− solution in 0.1 M KCl. The obtained Nyquist diagrams were adjusted using the Randles circuit as an equivalent circuit to obtain the corresponding resistances. The developed methodology showed good selectivity, precision, and sensitivity; although the LOD obtained was higher than those presented in other published articles, it turned out to be an alternative that allows the determination of ethinylestradiol using a simple disposable electrode.
Hydroliquefaction is a possible pathway to produce liquid transportation fuels from solid feedstocks like coal or biomass. Though much effort has been put into the investigation of maximizing the oil yield using expensive catalysts and pasting oils in batch setups, little is known about how to commercialize the process. This work aims at the demonstration of lignin hydroliquefaction under conditions interesting for commercial operation. The results from hydroliquefaction experiments of two different lignin types using a cheap iron-based catalyst and anthracene oil as the pasting oil in a semibatch system are presented here. Oil yields of above 50% are reached without observing coke formation. Extensive analyses of the feedstocks and product oils were performed. The process supplies high-quality oil, while differences in the decomposition path of both lignin types are observed. An high heating value of 39 400 J/g and H/C and O/C ratios of up to 1.6 and 0.1, respectively, are detected for the produced bio-oils.
MoS2 is a suitable solid lubricant for environments free of oxygen or water vapor (i.e. vacuum). Humid air degrades film properties due to oxidation accompanied by high wear and increasing coefficients of friction. The present study aims at the further development of sputtered pure MoS2 coatings, extending their applicability to varying environmental conditions by increasing the resistance against humidity. The systematic coating development process is supported by using an experimental Box–Behnken design with variations of the deposition parameters cathode voltage, target/substrate distance, temperature and argon gas pressure. In contrast to common one-factor-at-a-time (OFAT) studies, this approach enables a determination of interactions between deposition process parameters and tribological–mechanical MoS2 film properties. The tribological improvement focuses on a maximization of wear resistance in air and vacuum measured in ball-on-disk experiments. The evaluated mechanical properties are hardness, elastic modulus and residual stresses. These stresses were determined by the substrate curvature method. The study reveals that the residual stress state in the films and the hardness-to-modulus ratio are crucial for their tribological performance in humid air and vacuum environments. After a detailed determination of the relationships between deposition conditions and film properties, some selected microstructural analyses are presented which show that a substantially basal orientation of the lattice has positive effects on wear but also causes anisotropic film properties which result in fissile fracture of the coating if strong shock or point loads occur.
Diese Hinweisbroschüre erläutert allgemeine Planungsvoraussetzungen und Dimensionierungsregelungen, Baugrundsätze und Zulassungsbedingungen zur Anwendung innovativer Belagsdehnfugensysteme (Fahrbahnübergangskonstruktionen) in hochbeanspruchten Verkehrsflächen des Bundesfernstraßenbereiches. Alle planungsrelevanten, baustoffrelevanten, versuchstechnische, qualitätssichernden und baupraktischen Aspekte werden zur Unterstützung der Straßenbauverwaltungen, der Lieferfirmen und Ausführungsbetriebe erläutert. Die Veröffentlichung stellt eine technische Analyse aus Erfahrungssammlungen und Auswertung praktischer Erkenntnisse dar.
Im Vorhaben SNELLIUS wurden in enger Zusammenarbeit der BAM mit den Firmen Accurion und SENTECH Instruments aufbauend auf der Norm DIN 50989-1 „Ellipsometrie - Teil 1: Grundlagen; Ellipsometry – Part 1: Principles“ insgesamt fünf Normenentwürfe erarbeitet, die fünf Modell-basierten Anwendungsfällen der Ellipsometrie entsprechen. Unter Berücksichtigung des neuartigen Modell-basierten Normungskonzepts wurde in den Normenentwürfen erstmals eine GUM- und DIN EN ISO/IEC 17025:2018 konforme Bestimmung bzw. Abschätzung der Messunsicherheit umgesetzt. Dies erforderte neben der textlichen Erstellung auch umfangreiche experimentelle Arbeiten bei den Firmen, Vergleichsversuche und komplementäre Modell- und Datenevaluierungen, da sowohl die verwendete ellipsometrische Hardware (unterschiedliche ellipsometrische Funktionsprinzipien) als auch die ellipsometrische Software (Modell-Randbedingungen, Fitalgorithmus) firmenspezifisch zum Teil sehr unterschiedlich sind. Hierbei mussten in einem Konsensprozess Lösungen gefunden werden, die die beiden deutschen Ellipsometer-Anbieter Accurion (Imaging-Ellipsometrie) und SENTECH Instruments (Mapping Ellipsometrie) gleichermaßen akzeptieren konnten und die zudem einen Transfer auf die Internationale Ebene (ISO Norm) ermöglichen, also vom Grundsatz her auch auf weitere Ellipsometerhersteller wie (J. A. Woollam Co., USA; SEMILAB, Ungarn; HORIBA, Japan) mit wieder anderen ellipsometrischen Funktionsprinzipien, Hard- und Softwarelösungen anwendbar sind. Die weitgehende Unabhängigkeit von ellipsometrischen Funktionsprinzipien, Hard- und Softwarelösungen wurde dadurch erreicht, dass normativ festgelegt wurde, dass die ellipsometrischen Größen PSI und DELTA im Sinne von DIN ENISO/IEC 17025 als Rohdaten fungieren.
For the antiepileptic drug and anthropogenic marker carbamazepine (CBZ), a fast and cost-effective immunoassay based on fluorescence polarization (FPIA) was developed. The required fluorophore conjugates were synthesized from different fluorescein and CBZ derivatives. The most suitable tracer was CBZ–triglycine–5-(aminoacetamido)fluorescein. Additionally, the applicability of the assay in tubes and on microtiter plates was tested. The first format can be performed in a portable instrument and therefore can be applied in field measurements. The measurement of an individual sample can be carried out within 4 min. This assay shows a measurement range of 2.5–1000 µg L-1 and a test midpoint (or IC50) of 36 µg L-1. The FPIA performed on microtiter plates is useful for the assay development and is suitable for a very high throughput (up to 24 samples in 20 min). The test midpoint of this assay is 13 µg L-1 and the measurement range is 1.5–300 µg L-1. Furthermore, this assay requires smaller sample volumes and less reagents, including the crucial amount of antibody. The applicability of both assays to spiked surface water samples was evaluated. The recovery rates vary between 66–110% on microtiter plates and 81–140% in tubes.
Pharmacologically active compounds are often detected in wastewater and surface waters. The nonsteroidal anti-inflammatory drug diclofenac (DCF) was included in the European watch list of substances that requires its environmental monitoring in the member states. DCF may harmfully influence the ecosystem already at concentrations ≤ 1 μg L−1. The fast and easy quantification of DCF is becoming a subject of global importance. Fluorescence polarization immunoassay (FPIA) is a homogeneous mix-and-read method which does not require the immobilization of reagents. FPIA can be performed in one phase within 20–30 min, making it possible to analyse wastewater without any complicated pre-treatment. In this study, new tracermolecules with different structures, linking fluorophores to derivatives of the analyte, were synthesized, three homologous tracers based on DCF, two including a C6 spacer, and one heterologous tracer derived from 5-hydroxy-DCF. The tracer molecules were thoroughly assessed for performance. Regarding sensitivity of the FPIA, the lowest limit of detection reached was 2.0 μg L−1 with a working range up to 870 μg L−1. The method was validated for real wastewater samples against LC-MS/MS as reference method with good agreement of both methods.
Kunststoff- und Kunststoffrasenflächen kommt bei intensiv genutzten Flächen als Allwetterbeläge eine große Bedeutung zu. Sie leisten einen wichtigen Beitrag zur Absicherung eines ganzjähriger Sportangebote.
Gerade Kunststoffrasen ist aber in den letzten Jahren häufig in die Kritik geraten, insbesondere durch gefährliche Inhaltsstoffe und andere negative Einflüsse auf die Umwelt. Dies hat bei Bauherren zu einer Verunsicherung geführt und Fragen aufgeworfen, die erst teilweise beantwortet sind. Die Diskussionen rund um diesen Allwetterbelag haben bereits dazu geführt, dass von Seiten der Industrie viel verändert wurde und neue innovative Lösungen gefunden werden.
Der Fachbericht stellt eine Handreichung für Fachleute und interessierte Laien dar, um mehr Klarheit besonders hinsichtlich der Umweltanforderungen zu erzielen.
Public, hidden, and forensic features either encoded or directly legible are used for authentication. Fabry-Perot layer stacks as information carriers in combination with imaging ellipsometry as optical read-out system provide all-in-one anti-counterfeiting capability that may establish the new security level 'encoded forensic'. Different layer designs are described with regard to all three security levels: public features (e.g. color and tilt effect) perceptible by the human eye, hidden features (e.g. spectroscopic response in the UV or IR), and forensic features (ellipsometric quantities Ψ and Δ as a function of wavelength λ and angle of incidence AOI). Physically uncloneable functions (PUF) could be realized as a result of a multi-material and a multi-parameter deposition approach as well as by means of specific design features of the Fabry-Perot layer stack. Hence, they are not subject to any reverse engineering strategies. Examples of stratified, micro-structured, and laser-modified Fabry-Perot layer systems are considered that may be used at all perception levels (e.g. human eye, bar code reader, and imaging ellipsometry) for authentication against product counterfeiting and related areas. --------------------------------------------------------------------------------------------------------------------
Zur Authentifizierung werden offene, versteckte und forensische Merkmale entweder codiert oder direkt auslesbar verwendet. Mit Fabry-Perot Schichtsystemen als Informationsträger in Kombination mit der abbildenden Ellipsometrie als optischem Auslesesystem steht ein 'all-in-one' Fälschungsschutz-System zur Verfügung, das ein neues Sicherheitsniveau 'kodiert-forensisch' darstellt. Es werden unterschiedliche Schichtdesigns beschrieben, mit denen alle drei Sicherheitsniveaus realisiert werden können: offene Merkmale (z.B. Diclofenac (DCF) wird als nicht-steroidales, antientzündliches Arzneimittel gegen Fieber, Entzündungen, rheumatische Erkrankungen und Schmerzen eingesetzt. 70 % werden nach Einnahme wieder ausgeschieden. Aufgrund unvollständiger Elimination in den Kläranlagen lassen sich Rückstände im Trinkwasser und im Oberflächengewässer nachweisen. Diclofenac wird in der Umweltanalytik v.a. mittels GC-MS oder LCMS nachgewiesen. Bioanalytische Methoden machen sich die spezifische Antigen-Antikörper-Wechselwirkung zunutze, so z.B. Immunoassays. Die hierfür erforderlichen Antikörper werden durch Immunisierung mit einem Konjugat aus dem Analyten und einem Trägerprotein gewonnen. Die Kopplung kann direkt über die Carboxylfunktion von Diclofenac erfolgen. Um Abschirmungseffekte zu vermeiden, kann zwischen Protein und Analy1 ein Spacer sinnvoll sein.
