Over decades, the German Federal Institute for Materials Research and Testing (BAM) has established a sophisticated calibration laboratory for force, temperature and electrical quantities. Since more than 15 years it is accredited, currently by the national accreditation body (DAkkS), and offers its service also to external entities on a global scale. As a public provider, we are furthermore committed to research and development activities that demand measurements with highest quality and low level of uncertainties. Two R&D examples are highlighted within this contribution.
To examine the capability to detect and localise damage using the Measurement- and Model-based Structural Analysis (MeMoS), a small-scale truss bridge (1520 mm × 720 mm × 720 mm) made of aluminium profiles is built as a test specimen for this purpose. The truss frame of the test bridge is made of aluminium profiles with a sophisticated design of the cross-sectional area. In comparison, with solid profiles, only a fraction of the material is needed to produce the profiles, while their bending resistance decreases slightly. The profiles are built into a truss frame by connecting them by means of fastening sets made of steel. The bridge model is mounted on four steel bearings which each of them consist of a cylinder arranged between two plates. Fixed bearings are made by holding onto one end of the bridge. The bridge is subjected by an external load by placing a heavy object beneath it. At the same time, measurements can be conducted below the bridge. Therefore, the bridge specimen is elevated by attaching it on a pedestal with four columns. Damages can be induced by loosening the fastening pieces.
In Kalibrierscheinen werden oft aus messtechnisch ermittelten Daten Kennwerte berechnet, die mit Referenzwerten verglichen werden. Meist mehrere davon bilden festgelegte Klassengrenzen und dienen der Zuweisung der Kalibrierobjekte in verschiedene Güteklassen innerhalb ihrer Vergleichsgruppe. Der Vortrag geht auf Grenzfälle ein, die nahe an einer Klassengrenze liegen und deren Verquickung mit der Verwendung von Kennwerten und der Rundung bei Berechnungen.
Poly(butadiene)-b-poly(ethylene oxide) vesicles were successfully loaded with hydrophilic dye Phloxine B. Dye addition during vesicle formation leads to Phloxine B encapsulated inside the water filled vesicle core as well as to freely diffusing dye molecules. The removal of uncapsulated substrate involves time consuming methods like dialysis or harsher methods like ultra filtration or selective precipitation, posing the risk of irreversible sample manipulation. Here used Phloxine B as pH sensitive fluorescence indicator allows the characterization of hydrophilic loading without separation procedure by adjusting the pH value. Additionally membrane blocking efficiency can be studied by time dependent fluorescence measurements. Cryogenic TEM studies showed that the self-assembled structure remained unchanged when the hydrophilic dye was incorporated within the vesicles. Fluorescence microscopy imaging proved the encapsulation of the hydrophilic dye inside the core volume. The combination of fluorescence correlation spectroscopy (FCS) and dynamic light scattering (DLS) measurements as ensemble methods confirmed those results additionally.

The Measurement- and Model-based Structural Analysis (MeMoS) integrates a finite element model into least squares adjustment and thus allows to evaluate a mechanical model and measurements in a combined analysis. To examine the capability to detect and localise damage using this integrated analysis MeMoS, a small-scale truss bridge made of aluminium profiles is built as a test specimen for this purpose.
