The existence of microbial activity hotspots in temperate regions of Earth is driven by soil heterogeneities, especially the temporal and spatial availability of nutrients. Here we investigate whether microbial activity hotspots also exist in lithic microhabitats in one of the most arid regions of the world, the Atacama Desert in Chile. While previous studies evaluated the total DNA fraction to elucidate the microbial communities, we here for the first time use a DNA separation approach on lithic microhabitats, together with metagenomics and other analysis methods (i.e., ATP, PLFA, and metabolite analysis) to specifically gain insights on the living and potentially active microbial community. Our results show that hypolith colonized rocks are microbial hotspots in the desert environment. In contrast, our data do not support such a conclusion for gypsum crust and salt rock environments, because only limited microbial activity could be observed. The hypolith community is dominated by phototrophs, mostly Cyanobacteria and Chloroflexi, at both study sites. The gypsum crusts are dominated by methylotrophs and heterotrophic phototrophs, mostly Chloroflexi, and the salt rocks (halite nodules) by phototrophic and halotolerant endoliths, mostly Cyanobacteria and Archaea. The major environmental constraints in the organic-poor arid and hyperarid Atacama Desert are water availability and UV irradiation, allowing phototrophs and other extremophiles to play a key role in desert ecology.
Ziel des Vorhabens ist die hochgenaue und vollständige Erfassung innerer und äußerer Geometrien von Mikrobauteilen mittels Mikro-Computertomographie (µCT). Industriell wichtige Mikrobauteile sind z.B. Einspritzsysteme, Festplattenkomponenten oder Instrumente für die minimal-invasive Chirurgie. Typische Bauteilgrößen liegen hier im Bereich 0,1 mm - 100mm; typische zu messende Strukturen haben Größen von 0,5 µm - 500 µm. Mit klassischen Methoden der Messtechnik ist eine schnelle und hochgenaue Erfassung der gesamten Oberfläche von Mikrobauteilen bisher kaum möglich. Innenstrukturen von Mikrobauteilen können mit keiner optischen oder taktilen Messtechnik zerstörungsfrei gemessen werden. Diese Einschränkungen sollen mittels hochgenauer µCT überwunden werden. Hierzu soll der Messprozess simuliert werden, um die Anwendung von Korrekturverfahren zur Genauigkeitssteigerung zu ermöglichen. CT-Messungen an kalibrierten Mikroprüfkörpern sollen die Korrekturverfahren verifizieren. Für die Kalibrierung der Prüfkörper sollen Multisensor-Verfahren entwickelt und getestet werden. Diese sollen es erlauben, Prüfkörper gleichzeitig hochgenau und mit großer Messpunktdichte zu kalibrieren. Es ist geplant, die an Prüfkörpern erreichten Messunsicherheiten und Auflösungen zum Projektabschluss an realen, industriellen Mikrobauteilen (z.B. Mikrozahnrädern) messtechnisch nachzuweisen.
Die industrielle Computertomographie (CT) ist ein gut bekanntes Verfahren zur zerstörungsfreien Prüfung von Werkstücken. In den letzten Jahren wird die CT aber zunehmend auch für dimensioneile Messungen von Werkstücken eingesetzt. Der wesentliche Vorteil der CT liegt in der Kompletterfassung mit hoher Messpunktdichte, die auch für andere Messverfahren unzugängliche, innere Geometrien einschließt. An kleinen Bauteilen können Strukturen bis herunter zu wenigen Mikrometern gemessen werden, wodurch sich das Verfahren gut für die Messung von Mikroteilen eignet. Nachteilig ist, dass viele Einflussgrößen das Messergebnis beeinflussen, was die Rückführung der Messergebnisse, die Bestimmung der Messunsicherheit und die Definition von Spezifikationen für CT-Anlagen erschwert.
Der Beitrag stellt nach einer Einführung in das Verfahren Anwendungsbeispiele der CT an Mikroteilen vor. Darüber hinaus geht der Beitrag auf Prüfkörper und -verfahren zur Einmessung und Überprüfung von CT-Anlagen ein und berichtet über aktuelle Normungsaktivitäten. Abschließend werden Möglichkeiten zur weiteren Steigerung der Auflösung aufgezeigt, um zukünftig möglicherweise auch Nanogeometrien messen zu können.
Recently, non-destructive testing in civil engineering (NDT-CE), in particular of concrete components, has successfully mastered the leap from research to practice. Several methods have been established for field inspections to determine the concrete cover of reinforcement or to estimate the compressive strength as well as other parameters related to the concrete material. In addition, the application of nondestructive testing is indispensable, if information about the inner structure - such as the location of rebars and tendon ducts or the damage-related condition assessment to detect grouting defects, honeycombs, delamination, or corrosion - is required. Besides the selection of a suitable NDT method and an appropriate inspection system, the reliability of the results depends largely on the person who applies the non-destructive inspection technique and evaluates the inspection results. To ensure a high quality of non-destructive concrete evaluation as well as to keep the uncertainty caused by the inspection personnel to a minimum, structured, consistent and regulated theoretical as well as practical training of inspection personnel is essential. To close this gap, the subcommittee of education (UA-A) within the committee for NDT-CE of the German Society for Nondestructive Testing (DGZfP) has been reactivated in 2018 to establish uniform training standards for nondestructive concrete inspections in the long term. The subcommittee consists of scientists, practitioners, authorities, and clients. So far, the national standard DIN 4871 “Non-destructive testing - Qualification and Certification of NDT personnel in Civil Engineering (NDT-CE)” was developed and is currently under review. This standard considers the civil-industry-specifics, for example, that standards for NDT of concrete, as well as related product standards with a few exceptions, still do not exist at the moment. Within this presentation, the concept, the connection to ISO 9712 and other standards as well as an overview of the developed German standard DIN 4871 will be presented.

Computertomographische Aufnahmen von Objekten mit stark
voneinander abweichenden Absorptionseigenschaften stellen sowohl für den
Anwendungsbereich der zerstörungsfreien Prüfung als auch für den der
dimensionellen Messtechnik eine besondere Herausforderung dar. Die
Aufnahmekonfiguration kann bei Objekten dieser Art häufig nicht so gewählt
werden, dass die unterschiedlichen Materialeigenschaften mit ausreichender Qualität
abgebildet werden können. Aus diesem Grund ist eine objektive Bewertung der
Oberflächenmessung mit Hilfe eines in dieser Untersuchung eingeführten
Qualitätsmaßes erforderlich. Anhand der in dieser Arbeit vorgestellten
Prüfkörperserie soll der Einfluss der Materialzusammensetzung auf das
dimensionelle Messen mit CT erfasst und charakterisiert werden. Unter
Berücksichtigung der Qualitätsbewertung der Oberflächenpunkte erfolgt die
Berechnung der Form- und Abstandsabweichungen. Ziel ist es, die Genauigkeit der
Oberflächenmessung hinsichtlich unterschiedlicher Aufnahmeparameter und
Materialzusammensetzungen darzustellen. Anhand dieser Ergebnisse ist es möglich,
den computertomographischen Aufnahme- und Auswertungsprozess quantitativ zu
bewerten und Ursachen der Abweichungen zu diskutieren.
Anhand des hier vorgestellten zylindrischen Prüfkörpers, an dem
Kugeln aus einem Material mit einem hohen Absorptionskoeffizienten und sehr
geringen Formabweichungen angebracht wurden, können die geometrischen
Eigenschaften von CT-Anlagen untersucht werden. Die Basis der Auswertung
bilden die radiometrischen Aufnahmen. Mit Hilfe eines angepassten
Ellipsenoperators werden die Mittelpunkte der in den radiometrischen Aufnahmen
überlagerungsfrei abgebildeten Kugeln ermittelt. Anhand dieser Mittelpunkte
können die geometrischen Eigenschaften des Gesamtsystems zunächst rein an den
sich daraus ergebenen Spuren diskutiert und in einem weiteren Schritt
photogrammetrisch mit Hilfe der Bündelblockausgleichung ausgewertet werden.
Die Untersuchungen zeigen, dass der Prüfkörper geeignet ist, die geometrischen
Eigenschaften von CT-Anlagen zu überwachen. In einem weiteren Schritt könnte
das vorgestellte Verfahren benutzt werden, um z.B. Abweichungen von Drehtischen
zu charakterisieren und ggf. zu korrigieren.
Phasenkontrastmessungen haben sich als hilfreich bei der
Kantenfindung von Strukturen mit nur geringem Absorptionskontrast erwiesen.
Solche Messungen können besonders gut mit monochromatischer Röntgenstrahlung
hoher räumlicher Kohärenz durchgeführt werden, wie sie bei der Synchrotron-
Computertomographie (SY-CT) benutzt wird. Nachteilig wirken sich allerdings die
Phasenverschiebung des Synchrotronstrahls über das Strahlprofil und damit die
Kantenverbreiterung in den zu untersuchenden Strukturen aus. Daher ist ein
dimensionelles Messen mit normalen Rekonstruktionsverfahren nicht möglich. In
der vorliegenden Arbeit werden neue Filterverfahren zur Korrektur der
Phasenverschiebung unter Beibehaltung des erhöhten Kontrastes bei der
Rekonstruktion angewandt, um ausgewählte einkomponentige Referenzobjekte
hinsichtlich der dimensionellen Messeigenschaften zu untersuchen. Als Referenz
werden rückgeführte Messungen mit taktilen Koordinatenmessmaschinen
herangezogen. Die Untersuchungen wurden im freien Strahl durch Variation des
Abstandes zwischen Objekt und Detektor am Strahlrohr der BAM am Helmholtz-
Zentrum Berlin für Materialien und Energie - Speicherring BESSY II durchgeführt.
Information about an existing structure can be collected at certain costs to evaluate the reliability and condition as realistically as necessary. This information can be relevant or irrelevant, true or biased, precise or imprecise. The incorporation of relevant and quality-assessed measured information into reliability reassessment offers the chance to extend remaining lifetimes and support decision making about optimal actions or maintenance strategies. This paper shows recent developments in a national research project that aims to produce a guideline on the NDT-based, structure-specific modification of partial safety factors. The general methodology, results from recalculations according to the Eurocodes and metrologically solvable testing tasks relevant in the recalculation of the concrete bridges are shown and compared with the non-destructive testing methods applicable to concrete bridges. A case study is used to demonstrate that as-built drawings, in this case of the positions of tendons and shear reinforcement, can be verified using the radar method.
Substrate-induced self-assembly of donor-acceptor type compounds with terminal thiocarbonyl groups

Two types of conjugated thiocarbonyl-terminated compounds have been synthesized and their ability to be adsorbed on surfaces of the different nature, namely, glass, polytetrafluoroethylene (PTFE), and gold has been studied. Different morphology of the films prepared by thermal vacuum evaporation and drop-casting from solutions has been observed depending on the surface used. It has been found that gold surface has a unique property to influence self-assembly of both monolayer and larger aggregates or crystals of the compounds, in contrast to glass and PTFE substrates. It was found that thiocarbonyl group is able to be chemisorbed to the gold surface. However, it was concluded that in spite of the fact that the thiocarbonyl groups are important for the chemical interaction with the gold surface, physical adsorption on the substrate surface, compoundcompound and compound–solvent (when using solution) interactions are of great significance to drive self-assembly of the final film.
To investigate the imaging performance of an elastin-specific molecular magnetic resonance imaging (MRI) probe with respect to the extracellular matrix (ECM) in an experimental hepatic cancer model. Twelve rabbits with hepatic VX2 tumors were examined using 3 T MRI 14, 21, and 28 days after tumor implantation for two subsequent days (gadobutrol, day 1; elastin-specific probe, day 2). The relative enhancement (RE) of segmented tumor regions (central and margin) and the peritumoral matrix was calculated using pre-contrast and delayed-phase T1w sequences. MRI measurements were correlated to histopathology and element-specific and spatially resolved mass spectrometry (MS). Mixed-model analysis was performed to assess the performance of the elastin-specific probe. In comparison to gadobutrol, the elastin probe showed significantly stronger RE, which was pronounced in the tumor margin (day 14–28: P ≤ 0.007). In addition, the elastin probe was superior in discriminating between tumor regions (χ2(4) = 65.87; P < 0.001). MRI-based measurements of the elastin probe significantly correlated with the ex vivo elastinstain (R = .84; P <0 .001) and absolute gadolinium concentrations (ICP-MS: R = .73, P <0 .01). LA-ICP-MS imaging confirmed the colocalization of the elastin-specific probe with elastic fibers. Elastin-specific molecular MRI is superior to non-specific gadolinium-based contrast agents in imaging the ECM of hepatic tumors and the peritumoral tissue.
Terrestrial mud volcanoes are unique structures driven by tectonic pressure and fluids from the deep subsurface. These structures are mainly found in active tectonic zones, such as the area near the Los Bajos Fault in Trinidad. Here we report a chemical and microbiological characterization of three mud volcanoes, which included analyses of multiple liquid and solid samples from the mud volcanoes. Our study confirms previous suggestions that at least some of the mud volcano fluids are a mixture of deeper salt-rich water and surficial/precipitation water. No apparent water quality differences were found between sampling sites north and south of a major geological fault line. Microbiological analyses revealed diverse communities, both aerobic and anaerobic, including sulfate reducers, methanogens, carbon dioxide fixing and denitrifying bacteria. Several identified species were halophilic and likely derived from the deeper salt-rich subsurface water, while we also cultivated pathogenic species from the Vibrionaceae, Enterobacteriaceae, Shewanellaceae, and Clostridiaceae. These microorganisms were likely introduced into the mud volcano fluids both from surface water or shallow ground-water, and perhaps to a more minor degree by rain water. The identified pathogens are a major health concern that needs to be addressed.
Among new types of plasma processes, the underwater plasma is one of the most attractive methods for functionalization of polymer surfaces. The interesting features of plasma solution system are that the material surfaces to be modified remain in contact with the plasma-moderated solution. The role of plasma-moderated liquids, allows the reach of the reactive species through solution onto the geometrically hindered sites. The UV radiation produced in plasma formation helps in generating additional excited, ionized, and dissociated molecules and species in the reaction solution. An interesting feature of the technique is its flexibility to use a wide variety of additives as or in solution system. This allows us to create a selective or monotype functionalization of material surfaces. Such system was studied for the selective hydroxyl functionalization of polypropylene surface. The oxidation of polymer surfaces and the introduction of O-containing functional groups by underwater plasma was found to exceed concentrations typically achieved in oxygen low-pressure gas discharge plasmas up-to two-folds (maximal 56 O/100 C). The fraction of OH groups among all O-containing moieties amounts from 25 to 40% in comparison to that in the gas plasma of about 10% OH groups. Addition of hydrogen peroxide into this same system increases the fraction of C—O bonds up to 75% (27-OH/100 O). A study was focused to optimize the role of hydrogen peroxide on the efficiency of oxidation and selectivity with chemical derivatization with respect to the formation of mono-sort hydroxyl functionalities, calculated using a chemical derivatization technique.
