Metaproteomics has matured into a powerful tool to assess functional interactions in microbial communities. While many metaproteomic workflows are available, the impact of method choice on results remains unclear. Here, we carry out a community-driven, multi-laboratory comparison in metaproteomics: the critical assessment of metaproteome investigation study (CAMPI). Based on well-established workflows, we evaluate the effect of sample preparation, mass spectrometry, and bioinformatic analysis using two samples: a simplified, laboratory-assembled human intestinal model and a human fecal sample. We observe that variability at the peptide level is predominantly due to sample processing workflows, with a smaller contribution of bioinformatic pipelines. These peptide-level differences largely disappear at the protein group level. While differences are observed for predicted community composition, similar functional profiles are obtained across workflows. CAMPI demonstrates the robustness of present-day metaproteomics research, serves as a template for multi-laboratory studies in metaproteomics, and provides publicly available data sets for benchmarking future developments.
This study aimed to assess the measurement capabilities of participating National Metrology Institutes/ Designated Institutes (NMIs/DIs) and expert laboratories in determining of low-polarity organics in surface water. This comparison was organized by Government Laboratory, Hong Kong (GLHK).
At the CCQM Organic Analysis Working Group (OAWG) Meeting held in November 2012 in Hong Kong, GLHK initially proposed a CCQM key comparison and a parallel pilot study programme on pharmaceuticals in surface water. Further discussion at the CCQM Meeting held in April 2014 in Paris, the OAWG approved a CCQM Track C comparison (CCQM-K126) on low polarity pharmaceuticals in surface water. In the meeting, the programme was supported by more than six National Metrology Institutes/ Designated Institutes (NMIs/ DIs).
CCQM-K126 officially commenced in July 2014 and had registration from nine NMIs/DIs. Participants were provided two bottles (40 mL each) of surface water and were requested to determine the mass fraction of spiked carbamazepinein in surface water. The coordinator received nine sets of results from eight NMIs/DIs in February 2015. Apart from one using an immunoassay technique, all participants applied isotope dilution liquid chromatography-tandem mass spectrometry (ID-LCMS/MS) technique as their determination technique.
Gußeisen mit Kugelgraphit und ferritischer bzw. ferritisch-perlitischer Matrix gehören zu einer Werkstoffgruppe, die ausreichende Festigkeiten mit hohen Duktilitäten verbindet. Mit der vorliegenden Arbeit wurden die Kenntnisse über die Zusammenhänge zwischen der chemischen Zusammensetzung, der Gefügeausbildung und den mechanischen Eigenschaften erweitert. Dabei wurden sowohl unterschiedliche Gießverfahren (Sand- und Kokillenguß) in der Herstellung, als auch die Auswirkungen von Umwandlungsprozessen im Gefüge infolge von Wärmebehandlungen diskutiert. Die statistische Auswertung von Einzelergebnissen aus der qualitätssichernden Begutachtung von Lager- und Transportbehältern für radioaktive Stoffe wurden durch mechanische und bruchmechanische Untersuchungen an 24 Werkstoffsorten aus Gußeisen mit Kugelgraphit ergänzt.
In the past decade, nanoindentation has become commonly used experimental technique for obtaining quantitative data on elastic and inelastic material properties in small volumes, typically in submicron length scale. The methodology was developed mainly for homogeneous materials or, at least, the homogeneity is assumed far enough from the tested volume in evaluation procedures1. Measurement on a heterogeneous system, in which the material phases are intermixed in small volume, is still a challenging task due to several circumstances.
First, the preparation of a flat surface on the heterogeneous sample with high differences in stiffness needs Special care. Second, the mechanical interaction between the material phases during surface loading is often unavoidable and therefore the extraction of intrinsic properties of individual phases from the heterogeneous system may not be possible. If differences between the stiffnesses of the interacting phases are low, finding of intrinsic properties can be successful using conventional Evaluation. In other cases, the error in estimating intrinsic properties can reach tens of percent.
Für die Entfernung von VOC (volatile organic compounds) aus der Luft von Fertigungsstätten werden vielfach mikroporöse Adsorbentien eingesetzt. Die gereinigte Luft wird rezyklisiert. Für die mit VOC beladenen Adsorbentien wird ein Regenerierungsweg für den mehrfachen Gebrauch gesucht. Bei der Regenerierung sollen die VOC möglichst vollständig desorbiert werden und mit einer geringen Gasmenge in einer katalytischen Verbrennung zu CO2 und H2O umgewandelt werden. Die Wirtschaftlichkeit der Luftreinigung wird wesentlich bestimmt durch die Sorptionskapazität des Adsorbens, die erforderliche Energie für die thermische Regenerierung und die Menge des Spülgases bei der Regenerierung. In dieser Arbeit wird der Einsatz von Typ X-Zeolithpulver in der Luftreinigung einer Lackiererei beschrieben und dessen Regenerierung im Labor mittels DTA/TG und Isothermenmessung der Schlüsselkomponenten untersucht. Ferner wurde die Regenerierung des beladenen Zeoliths in einem Labor-Drehrohrofen unter Variation der Betriebsparameter untersucht und anschließend mit ausgewählten Parametern in einem kleintechnischen Drehrohrofen durchgeführt. Die Verfahrensvarianten werden diskutiert.
Inflammatory bowel diseases (IBD) comprise mainly ulcerative colitis (UC) and Crohn´s disease (CD). Both forms present with a chronic inflammation of the (gastro) intestinal tract, which induces excessive changes in the composition of the associated extracellular matrix (ECM). In UC, the inflammation is limited to the colon, whereas it can occur throughout the entire gastrointestinal tract in CD. Tools for early diagnosis of IBD are still very limited and highly invasive and measures for standardized evaluation of structural changes are scarce. To investigate an efficient non-invasive way of diagnosing intestinal inflammation and early changes of the ECM, very small superparamagnetic iron oxide nanoparticles (VSOPs) in magnetic resonance imaging (MRI) were applied in two mouse models of experimental colitis: the dextran sulfate sodium (DSS)-induced colitis and the transfer model of colitis. For further validation of ECM changes and inflammation, tissue sections were analyzed by immunohistochemistry. For in depth ex-vivo investigation of VSOPs localization within the tissue, Europium-doped VSOPs served to visualize the contrast agent by imaging mass cytometry (IMC). VSOPs accumulation in the inflamed colon wall of DSS-induced colitis mice was visualized in T2* weighted MRI scans. Components of the ECM, especially the hyaluronic acid content, were found to influence VSOPs binding. Using IMC, colocalization of VSOPs with macrophages and endothelial cells in colon tissue was shown. In contrast to the DSS model, colonic inflammation could not be visualized with VSOP-enhanced MRI in transfer colitis. VSOPs present a potential contrast agent for contrast-enhanced MRI to detect intestinal inflammation in mice at an early stage and in a less invasive manner depending on hyaluronic acid content.
Concrete technology results more and more in the development of materiale with enhanced properties, such as high strength, higher durability and increased ecological compatibility. One exceptional example is Ultra-High Performance Concrete (UHPC). Its advantages are very high compressive strength and an improved durability, caused by their dense micro structure. Curing under water vapor Saturation pressure (autoclaving) can improve these properties by affecting the micro structure and better the mechanical properties of UHPC. Autoclaving leads to an enhanced cement hydration and accelerated reaction rate of supplementary cementitious materials (SCM) and mineral additions, Used at the production of aerated autoclaved concrete (AAC) and sand-lime bricks, the process of autoclaving is well known, especially in highly porous Systems. One well known requirement to autoclave effectively is a porosity of at least 10%. There is no sufficient heat transfer and also not enough water available if the porosity is lower, Nevertheless, autoclaving also works in the dense structure of UHPC with porosities lower than 2%, as former investigations have shown. There is only a visible difference between the outer parts of the samples compared to their core. Investigations on the chemistry, the micro structure as well as the mechanical properties where performed to find an explanation to this effect and to explain the functionality of autoclaving UHPC in spite of the low porosity.
Pharmaceuticals, certain food ingredients, and mammalian endogenous metabolic products in wastewater are mostly of human origin. They are anthropogenic markers.
Proper knowledge of their levels in wastewater helps to track sources of pollutants in natural waters and allows for calculation of removal efficiencies in wastewater Treatment plants. Here, we describe the development and application of an indirect competitive, multiplexing suspension Array fluorescence immunoassay (SAFIA) for the detection of carbamazepine (CBZ), diclofenac (DCF), caffeine (CAF), and isolithocholic acid (ILA) in wastewater, covering those classes of anthropogenic markers. The assay consists of haptens covalently conjugated to fluorescence-encoded polystyrene core/silica shell microparticles to create a site for competitive binding of the antibodies (Abs). Bound Abs are then stained with fluorophore-labeled Abs. Encoding and signaling fluorescence of the particles are determined by an automated flow cytometer.
For compatibility of the immunoassay with the 96-well microtiter plate format, a stop reagent, containing formaldehyde, is used. This enables a wash-free procedure while decreasing time-to-result. Detection limits of 140 ± 40 ng/L for CBZ, 180 ± 110 ng/L for CAF, 4 ± 3 ng/L for DCF, and 310 ± 70 ng/L for ILA are achieved, which meet the sensitivity criteria of wastewater analysis. We demonstrate the applicability of SAFIA to real wastewater samples from three different wastewater Treatment plants, finding the results in good agreement with LC-MS/MS. Moreover, the accuracy in general exceeded that from classical ELISAs. We therefore propose SAFIA as a quick and reliable approach for wastewater analysis meeting the requirements for process analytical technology.
