Understanding the mechanism of interactions of nanomaterials at biointerfaces is a crucial issue to develop new antimicrobial vectors. In this work, a series of water-soluble fullerene-polyglycerol sulfates (FPS) with different fullerene/polymer weight ratios and varying numbers of polyglycerol sulfate branches are synthesized, characterized, and their interactions with two distinct surfaces displaying proteins involved in target cell recognition are investigated. The combination of polyanionic branches with a solvent exposed variable hydrophobic core in FPS proves to be superior to analogs possessing only one of these features in preventing interaction of vesicular Stomatitis virus coat glycoprotein (VSV-G) with baby hamster kidney cells serving as a model of host cell. Interference with L-selectin-ligand binding is dominated by the negative charge, which is studied by two assays: a competitive surface plasmon resonance (SPR)-based inhibition assay and the leukocyte cell (NALM-6) rolling on ligands under flow conditions. Due to possible intrinsic hydrophobic and electrostatic effects of synthesized compounds, pico- to nanomolar half maximal inhibitory concentrations (IC50) are achieved. With their highly antiviral and anti-inflammatory properties, together with good biocompatibility, FPS are promising candidates for the future development towards biomedical applications.
The construction sector has traditionally been a source of employment in African regions. However, due to policy changes, cut backs in public investments as well as a fall in the number of employments in the private sector have been observed in many countries. Furthermore a shift from registered companies to informal contractors could be noticed. Many contractors offer services they do not provide permanently or for which the required equipment has to be borrowed. A study that was published in 2000 about the situation in Tanzania pointed out that only ca. 15% of the enterprises that offer concrete works own a concrete mixer. The building practice with concrete varies widely over the African continent. Whereas the level of technology and the infrastructure for cement and concrete in South Africa is elaborate, in East Africa and particularly those that were heavily suffering from civil war during the nineties of the last century, the availability of cement and concrete is poor and these materials are far from being everyday building practice. Most of the population relies on indigenous forms of construction requiring wood obtained from felling of forest trees and ultimately contributing to de-forestation. Informal settlements in South-Africa and Kenya are literal evidence where originally green natural vegetation has been turned into artificial desert-like landscapes.
Baupraxis und Infrastrukturentwicklung im Hinblick auf die Betonindustrie in Ost- und Südafrika

Der Bausektor stellt in Afrika traditionell eine der stabileren Quellen für den Arbeitsmarkt dar. Durch ständige Wechsel in der Politik wurden jedoch in zahlreichen Ländern Einschnitte bei den öffentlichen Investitionen sowie ein Rückgang der Beschäftigtenzahlen im Privatsektor beobachtet. Darüber hinaus konnte eine Verlagerung von offiziell tätigen und registrierten hin zu nicht registrierten Bauunternehmen festgestellt werden. Zahlreiche Bauunternehmen bieten Dienstleistungen an, die sie nicht ständig ausführen oder für die sie sich das erforderliche Gerät leihen müssen. Einer im Jahre 2000 hinsichtlich der Situation in Tansania veröffentlichten Studie zufolge verfügen nur etwa 15 % der Unternehmen, die Betonbauarbeiten anbieten, über einen eigenen Betonmischer. Die auf dem afrikanischen Kontinent angewandten Baupraktiken im Betonbau weichen erheblich voneinander ab. Während Südafrika über eine fortgeschrittene Technologie und eine hoch entwickelte Infrastruktur im Hinblick auf die Zement- und Betonindustrie verfügt, sind Zement und Beton in den ostafrikanischen Ländern, insbesondere in jenen, die in den 90er Jahren des vergangenen Jahrhunderts von schweren Bürgerkriegen erschüttert wurden, kaum verfügbar und zählen keinesfalls zu den in der täglichen Baupraxis verwendeten Baustoffen. Hier wird auf einheimische Baumethoden, bei denen vorwiegend Holz verwendet wird, zurückgegriffen. Dieses wird durch Abholzung gewonnen, was schließlich zur Rodung ganzer Waldgebiete führt. Die ohne entsprechende Planung, Genehmigung und Erschließung errichteten Siedlungen in Südafrika und Kenia veranschaulichen erschreckend, wie einstmals von grüner Vegetation bedeckte Landstriche in künstliche, wüstenartige Landschaften verwandelt wurden.
Errichtet aus Glas und Beton prägen heute unzählige Sakralbauten der 1950er- und 60er-Jahre unsere Stadtbilder. Nicht selten sind in diesen Kirchen erhebliche Schäden an den Betonglasfenstern zu verzeichnen. Diese fachgemäß zu erhalten, erfordert spezielles technisches und denkmalpflegerisches Know-how. An der Entwicklung geeigneter Instandsetzungsstrategien arbeitet derzeit ein interdisziplinäres Projektteam.
Die Kaiser-Wilhelm-Gedächtniskirche (KWG) gilt als eine der wichtigsten Sehenswürdigkeiten Berlins in der City West und als ein herausragendes Beispiel für die „Dalle de Verre“ Verglasungen in sakralen Gebäuden. Der vorliegende Beitrag widmet sich der Fassade und den Glasfenstern des neuen Glockenturms, wobei die beschriebenen Szenarien aufgrund der annähernd gleichen Bauweise der restlichen neuen Baukörper in weiten Teilen starke Analogien aufweisen und insofern übertragbar sind.
Main topics of the research project BEGLARES are the restoration of "dalle de verre" concrete Windows and the development of repair techniques and mortars. This project is executed in Cooperation between Glasmalerei Peters GmbH, the University of York and the BAM Federal Institute for Materials Research and Testing. It is funded by the Federal Ministry of Economics and Technology. The main mechanism of damages, the development of repair mortar and special test specimens are presented.
The aim of an ongoing research project is to develop a design approach for typical offshore driven piles (e.g. Jacket piles) based on the application of injections by compaction grouting directly at the pile shaft. The paper aims to present the results of laboratory and in-situ tests, which reveal the efficiency and the promising potential of the optimised foundation concept for a more economic dimensioning of pile foundations and to increase their bearing capacity in non-cohesive soil at any moment after installation.
The conventional hybridoma screening and subcloning process is generally considered to be one of the most critical steps in hapten-specific antibody production. It is time-consuming, monoclonality is not guaranteed, and the number of clones that can be screened is limited. Our approach employs a novel hapten-specific labeling technique of hybridoma cells. This allows for fluorescence-activated cell sorting (FACS) and single-cell deposition and thereby eliminates the above-mentioned problems. A two-step staining approach is used to detect antigen specificity and antibody expression: in order to detect antigen specificity, hybridoma cells are incubated with a hapten−horseradish peroxidase conjugate (hapten−HRP), which is subsequently incubated with a fluorophore-labeled polyclonal anti-peroxidase antibody (anti-HRP−Alexa Fluor 488). To characterize the expression of membrane-bound immunoglobulin G (IgG), a fluorophore-labeled anti-mouse IgG antibody (anti-IgG−Alexa Fluor 647) is used. Hundreds of labeled hybridoma cells producing monoclonal antibodies (mAbs) specific for a hapten were rapidly isolated and deposited from a fusion mixture as single-cell clones via FACS. Enzyme-linked immunosorbent assay (ELISA) measurements of the supernatants of the sorted hybridoma clones revealed that all hapten-specific hybridoma clones secrete antibodies against the target. There are significant improvements using this high-throughput technique for the generation of mAbs including increased yield of antibody-producing hybridoma clones, ensured monoclonality of sorted cells, and reduced development times.
