Se presenta una guía completa del procedimiento experimental para la determinación del valor umbral ΔKth intrínseco y caracterización de curvas FCGR sobre el acero de alta resistencia S960QL empleando probetas de pequeña escala. El análisis se realiza comparando probetas SENB de diferentes geometrías, distinguiendo entre “convencionales” y “miniatura”. Las primeras con espesor (B) de 6 mm y anchura (W) de 19, y las segundas con B = 3 y dos valores distintos de W, 4 y 6 mm. A fin de identificar el valor umbral ΔKth intrínseco, la relación de cargas (R) aplicada en todos los ensayos fue 0,8. La técnica de caída de potencial (DCPD) se utilizó para la monitorización del tamaño de grieta a lo largo de los ensayos.
La evaluación de los resultados experimentales confirma la equiparación de resultados entre las distintas geometrías y configuraciones de ensayo practicadas, así como la viabilidad de emplear probetas de pequeña escala en la determinación del valor umbral ΔKth intrínseco. Esto abre nuevas perspectivas en lo relativo al empleo de probetas de reducidas dimensiones en la caracterización de materiales a fatiga. A su vez, permite ofrecer una serie de recomendaciones para reproducir con éxito ensayos semejantes utilizando probetas de pequeñas dimensiones de acero y otros materiales metálicos.
Directing nanoparticles to the nucleus by attachment of nuclear localization sequences (NLS) is an aim in many applications. Gold nanoparticles modified with two different NLS were studied while crossing barriers of intact cells, including uptake, endosomal escape, and nuclear translocation. By imaging of the nanoparticles and by characterization of their molecular interactions with surface-enhanced Raman scattering (SERS), it is shown that nuclear translocation strongly depends on the particular incubation conditions. After an 1 h of incubation followed by a 24 h chase time, 14 nm gold particles carrying an adenoviral NLS are localized in endosomes, in the cytoplasm, and in the nucleus of fibroblast cells. In contrast, the cells display no nanoparticles in the cytoplasm or nucleus when continuously incubated with the nanoparticles for 24 h. The ultrastructural and spectroscopic data indicate different processing of NLS-functionalized particles in endosomes compared to unmodified particles. NLS functionalized nanoparticles form larger intraendosomal aggregates than unmodified gold nanoparticles. SERS spectra of cells with NLS-functionalized gold nanoparticles contain bands assigned to DNA and were clearly different from those with unmodified gold nanoparticles. The different processing in the presence of an NLS is influenced by a continuous exposure of the cells to nanoparticles and an ongoing nanoparticle uptake. This is supported by mass-spectrometry-based quantification that indicates enhanced uptake of NLS-functionalized nanoparticles compared to unmodified particles under the same conditions. The results contribute to the optimization of nanoparticle analysis in cells in a variety of applications, e.g., in theranostics, biotechnology, and bioanalytics.
Accurate and reproducible measurement of the structure and properties of high-value nanoparticles is extremely important for their commercialization. A significant proportion of engineered nanoparticle systems consist of some form of nominally core-shell structure, whether by design or unintentionally. Often, these do not form an ideal core-shell structure, with typical deviations including polydispersity of the core or shell, uneven or incomplete shells, noncentral cores, and others. Such systems may be created with or without intent, and in either case an understanding of the conditions for formation of such particles is desirable. Precise determination of the structure, composition, size, and shell thickness of such particles can prove challenging without the use of a suitable range of characterization techniques. Here, the authors present two such polymer core-shell nanoparticle systems, consisting of polytetrafluoroethylene cores coated with a range of thicknesses of either polymethylmethacrylate or polystyrene. By consideration of surface energy, it is shown that these particles are expected to possess distinctly differing coating structures, with the polystyrene coating being incomplete. A comprehensive characterization of these systems is demonstrated, using a selection of complementary techniques including scanning electron microscopy, scanning transmission electron microscopy, thermogravimetric analysis, dynamic light scattering, differential centrifugal sedimentation, and X-ray photoelectron spectroscopy. By combining the results provided by these techniques, it is possible to achieve superior characterization and understanding of the particle structure than could be obtained by considering results separately.
The new ligand 4´-(4´´´-pyridyl-N-oxide)-2,2´:6´,2´´-terpyridine (pyNoxterpy) and its homoleptic iron(II) complex have been synthesised, and structural and spectroscopic studies have been carried out. The obtained results have been compared with the reported data for the parent ligand 4´-(4´´´-pyridyl)-2,2´:6´,2´´-terpyridine (pyterpy) and its homoleptic iron(II) complex. Significant differences between the spectral and electrochemical properties of the metal complexes have been found, derived from the changes in the electronic properties of the coordinated ligands.
