A surface-labeled lyophilized lymphocyte (sLL) preparation has been developed using human peripheral blood mononuclear cells prelabeled with a fluorescein isothiocyanate conjugated anti-CD4 monoclonal antibody. The sLL preparation is intended to be used as a reference material for CD4+ cell counting including the development of higher order reference measurement procedures and has been evaluated in the pilot study CCQM-P102. This study was conducted across 16 laboratories from eight countries to assess the ability of participants to quantify the CD4+ cell count of this reference material and to document cross-laboratory variability plus associated measurement uncertainties. Twelve different flow cytometer platforms were evaluated using a standard protocol that included calibration beads used to obtain quantitative measurements of CD4+ T cell counts. There was good overall cross-platform and counting method agreement with a grand mean of the laboratory calculated means of (301.7 ± 4.9) µL-1 CD4+ cells. Excluding outliers, greater than 90% of participant data agreed within ±15%. A major contribution to variation of sLL CD4+ cell counts was tube to tube variation of the calibration beads, amounting to an uncertainty of 3.6%. Variation due to preparative steps equated to an uncertainty of 2.6%. There was no reduction in variability when data files were centrally reanalyzed. Remaining variation was attributed to instrument specific differences. CD4+ cell counts obtained in CCQM-P102 are in excellent agreement and show the robustness of both the measurements and the data analysis and hence the suitability of sLL as a reference material for interlaboratory comparisons and external quality assessment.
This report focuses on the characterization of CD4 expression level in terms of equivalent number of reference fluorophores (ERF). Twelve different flow cytometer platforms across sixteen laboratories were utilized in this study. As a first step the participants were asked to calibrate the fluorescein isothiocyanate (FITC) channel of each flow cytometer using commercially available calibration standard consisting of five populations of microspheres. Each population had an assigned value of equivalent fluorescein fluorophores (EFF denotes a special case of the generic term ERF with FITC as the reference fluorophore). The EFF values were assigned at the National Institute of Standards and Technology (NIST). A surface-labelled lyophilized cell preparation was provided by the National Institute of Biological Standards and Control (NIBSC), using human peripheral blood mononuclear cells (PBMC) pre-labeled with a FITC conjugated anti-CD4 monoclonal antibody. Three PBMC sample vials, provided to each participant, were used for the CD4 expression analysis. The PBMC are purported to have a fixed number of surface CD4 receptors. On the basis of the microsphere calibration, the EFF value of the PBMC samples was measured to characterize the population average CD4 expression level of the PBMC preparations. Both the results of data analysis performed by each participant and the results of centralized analysis of all participants' raw data are reported. Centralized analysis gave a mean EFF value of 22,300 and an uncertainty of 750, corresponding to 3.3% (level of confidence 68%) of the mean EFF value. The next step will entail the measurement of the ERF values of the lyophilized PBMC stained with labels for other fluorescence channels. The ultimate goal is to show that lyophilized PBMC is a suitable biological reference cell material for multicolor flow cytometry and that it can be used to present multicolor flow cytometry measurements in terms of ABC (antibodies bound per cell) units.
A comparative evaluation of systematically tailored chemical structures of various phosphorus-containing aminic hardeners for epoxy resins was carried out. In particular, the effect of the oxidation state of the phosphorus in the hardener molecule on the curing behavior, the mechanical, thermomechanical, and hot-wet properties of a cured bifunctional bisphenol-A based thermoset is discussed. Particular attention is paid to the comparative pyrolysis of neat cured epoxy resins containing phosphine oxide, phosphinate, phosphonate, and phosphate (with a phosphorus content of about 2.6 wt %) and of the fire behavior of their corresponding carbon fiber-reinforced composites. Comparatively faster curing thermosetting system with an enhanced flame retardancy and adequate processing behavior can be formulated by taking advantage of the higher reactivity of the phosphorus-modified hardeners. For example, a combination of the high reactivity and of induced secondary crosslinking reactions leads to a comparatively high Tg when curing the epoxy using a substoichiometric amount of the phosphinate-based hardener. The overall mechanical performance of the materials cured with the phosphorus-containing hardeners is comparable to that of a 4,4-DDS-cured reference system. While the various phosphorus-containing hardeners in general provide the epoxy-based matrix with enhanced flame retardancy properties, it is the flame inhibition in the gas phase especially that determines the improvement in fire retardancy of carbon fiber-reinforced composites. In summary, the present study provides an important contribution towards developing a better understanding of the potential use of such phosphorus-containing compounds to provide the composite matrix with sufficient flame retardancy while simultaneously maintaining its overall mechanical performance on a suitable level.
A systematic and comparative evaluation of the pyrolysis of halogen-free flame-retarded epoxy resins containing phosphine oxide, phosphinate, phosphonate, and phosphate (phosphorus contents around 2.6 wt.%) and the fire behaviour of their carbon fibre composites is presented. Decomposition pathways are proposed based on the thermal analysis (TG), TG coupled with evolved gas analysis (TG-FTIR), kinetics and analysis of the residue with FTIR and XPS. All organophosphorus-modified hardeners containing phenoxy groups lead to a reduced decomposition temperature and mass loss step for the main decomposition of the cured epoxy resin. With increasing oxidation state of the phosphorus the thermally stable residue increases, whereas the release of phosphorus-containing volatiles decreases. The flammability of the composites was investigated with LOI and UL 94 and the fire behaviour for forced-flaming conditions with cone calorimeter tests performed using different irradiations. The flame retardancy mechanisms are discussed. With increasing oxidation state of the phosphorus additional charring is observed, whereas the flame inhibition, which plays the more important role for the performance of the composites, decreases. The processing and the mechanical performance (delamination resistance, flexural properties and interlaminar bonding strength) of the fibre-reinforced composites containing phosphorus were maintained at a high level and, in some cases, even improved. The potential for optimising flame retardancy while maintaining mechanical properties is highlighted in this study.
Highly soluble 9,10-dihydro-9-oxa-10-phosphaphenanthrene-10-oxide-(DOPO)-based diamino hardener (2), bearing its amino groups directly on the DOPO framework, is investigated with respect to its use as a reactive flame retardant in thermosets. A mechanism for decomposition of the corresponding phosphorus-modified epoxy resin system based on a diglycidylether of bisphenol A DGEBA and 2 (DGEBA/2) is proposed and compared to the systems using DGEBA and 4,4'-diaminodiphenylsulfon (DGEBA/DDS) and to a similar system based on the structurally comparable non-reactive DOPO-based compound (DGEBA/DDS/1). Additive 1 changed the decomposition characteristics of the epoxy resin only slightly and phosphorus was released. Incorporating 2 induces two-step decomposition and most of the phosphorus remains in the residue. Furthermore, the fire behaviour of neat epoxy resin systems and a representative carbon fibre-reinforced composite based on DGEBA, DDS and 2 (DGEBA/DDS/2) were examined and compared to that of the analogous composite systems based on DGEBA/DDS and DGEBA/DDS/1. Based on different flame retardancy mechanisms both the reactive compound 2 and the additive compound 1 improve flammability (increase in LOI >13% and achieving V-1 behaviour) of the epoxy resin and composites. Under forced flaming only the flame inhibition of the additive compound 1 acts sufficiently. Lastly, the superior key mechanical properties of the epoxy resin and composite based on 2 are sketched.
The pyrolysis of an epoxy resin and the fire behavior of corresponding carbon fiber-reinforced composites, both flame-retarded with either 10-ethyl-9,10-dihydro-9-oxa-10-phosphaphenanthrene 10-oxide or 1,3,5-tris[2-(9,10-dihydro-9-oxa-10-phosphaphenanthrene 10-oxide-10-)ethyl]1, 3,5-triazine-2,4,6(1H,3H,5H)-trione, are investigated. The different fire retardancy mechanisms are discussed, and their influence on the fire properties assessed, in particular for flammability (limiting oxygen index, UL 94) and developing fires (cone calorimeter with different external heat fluxes of 35, 50, and 70 kW m-2). Adding the flame retardants containing 9,10-dihydro-9-oxa-10-phosphaphenanthrene-10-oxide affects the fire behavior by both condensed phase and gas phase mechanisms. Interactions between the additives and the epoxy resin result in a change in the decomposition pathways and an increased char formation. The release of phosphorous products results in significant flame inhibition. The fire properties achieved are thus interesting with respect to industrial exploration.
Summary: The decomposition of novel phosphorus-containing poly(oxyphenylene-sulfonyl-phenylene-oxy-diphenyl phenylene phosphine oxide) (PSU_I), 2,5-dihydroxy-1-biphenylene-phosphine oxide based polysulfone (PSU_II), poly(sulfonyl-diphenylphenylene phosphonate) (PSU_P) and bisphenol A-based polysulfone (PSU) is studied. The influence of the chemical structure, charring and phosphorus release is discussed based on the mass loss, kinetics and products. The pyrolysis and fire behaviour of blends with epoxy resin (EP) are studied. For EP-PSU_II, phosphorus initiates water elimination and changes the decomposition pathway of EP. The fire behaviour of EP-PSU shows some improvements, whereas the heat release rate is crucially reduced for EP-PSU_II due to simultaneous char formation and flame inhibition.
Polyurethane versus silicone catheters for central venous port devices implanted at the forearm
(2016)
Purpose: We aimed to analyse short and long-term complications of polyurethane (PU) versus silicone catheters used in totally implantable venous-access ports (TIVAPs) implanted at the forearm.
Methods: Retrospective analysis of 698 consecutively implanted TIVAPs was performed. Primary end-points were defined as rates of major complications associated with either type of central venous port catheter. Technical success rate, device service interval as well as minor complications not requiring port explantation were defined as secondary end-points.
Results: A total of 698 port devices were implanted in 681 patients, 396 equipped with a PU catheter, 302 with a silicone catheter. The technical success rate was 99.9% with no major periprocedural complications. During follow-up a total of 211 complications in 146 patients were observed (1.0/1000 catheter days), 183 occurred associated with PU catheters (1.8/100 catheter days), 28 (0.3/1000 catheter days) with silicone catheters (log rank test p < 0.0001). Catheter-related bloodstream infections as well as thrombotic complications occurred significantly more frequently with PU catheters, while silicone catheters exhibited a trend towards a higher rate of mechanical failure such as disconnection or catheter rupture. Major complications requiring explantation of the device occurred more frequently with PU-based catheters (10.6%) compared to silicone catheter carrying ports (4.6%, log rank test p < 0.001).
Central venous port devices made of two different polymeric materials, thermoplastic polyurethane (TPU)and silicone rubber (SiR), were compared due their material properties. Both naïve catheters as well as catheters after removal from patients were investigated. In lab experiments the influence of various chemo-therapeutic solutions on material properties was investigated, where as the samples after removal were compared according to the implanted time inpatient. The macroscopic, mechanical performance was assessed with dynamic, specially adapted tests for elasticity. The degradation status of the materials was determined with common tools of polymer characterisation, such as infrared spectroscopy, molecular weight measurements and various methods of thermal analysis. The surface morphology was an alysed using scanning electron microscopy. A correlation between material properties and clinical performance was proposed. The surface morphology and chemical composition of the polyurethane catheter materials can potentially result in increased susceptibility of the catheter to bloodstream infections and thrombotic complications. The higher mechanic failure, especially with increasing implantation time of the silicone catheters is related to the lower mechanical performance compared to the polyurethane material as well as loss of barium sulphate filler particles near the surface of the catheter. This results in preformed microscopic notches, which act as predetermined sites of fracture.
Welded joints show large variation of the weld toe geometry along the weld seam, which is one important reason for the comparably large scatter in fatigue life. Therefore, it is crucial to take the local geometry at the weld toe into account, to reduce the conservatism in fatigue assessment of welded joints. This study is based on the IBESS procedure for the calculation of the fatigue strength, whereby the evaluation of local geometrical parameters is carried out by means of 3D surface scans. The approach is validated against 26 fatigue test series. The fatigue life is in general overpredicted, whereas good agreement is achieved for high stress ratio (R = 0.5). A sensitivity analysis conducted with IBESS shows that weld toe radii ρ < 2 mm and flank angle α < 30° have a significant influence on the calculated fatigue strength. In contrast to this, no strong correlation between ρ and the fatigue strength was determined experimentally in this study.
In this study, carried out within the Joint Danube Survey 4, a comprehensive microplastic screening in the water column within a large European river basin from its source to estuary, including major tributaries, was realized. The objective was to develop principles of a systematic and practicable microplastic monitoring strategy using sedimentation boxes for collection of suspended particulate matter followed by its subsequent analysis using thermal extraction desorption-gas chromatography/mass spectrometry. In total, 18 sampling sites in the Danube River Basin were investigated. The obtained suspended particulate matter samples were subdivided into the fractions of >100 μm and <100 μm and subsequently analyzed for microplastic mass contents. The results showed that microplastics were detected in all samples, with polyethylene being the predominant polymer with maximum contents of 22.24 μg/mg, 3.23 μg/mg for polystyrene, 1.03 μg/mg for styrene-butadiene-rubber, and 0.45 μg/mg for polypropylene. Further, polymers such as different sorts of polyester, polyacrylates, polylactide, and natural rubber were not detected or below the detection limit. Additional investigations on possible interference of polyethylene signals by algae-derived fatty acids were assessed. In the context of targeted monitoring, repeated measurements provide more certainty in the interpretation of the results for the individual sites. Nevertheless, it can be stated that the chosen approach using an integrative sampling and determination of total plastic content proved to be successful.
In line with the industrial trend of additive manufacturing, cold spray as a non-laser-based process is becoming increasingly important for many fields of application. For the evaluation of additive manufacturing of winding components made of copper for large electrical high-voltage machines, material and component properties such as electrical conductivity, mechanical load capacity and the component size that can be produced are of particular importance. In this context, the cold spray process offers advantages over laser-based additive manufacturing processes such as laser powder bed fusion (LPBF) or laser cladding by using the kinetic energy of the copper powder particles to generate particle cohesion. To investigate the electrical conductivity as well as the mechanical load capacity of cold spray parts, specimens were machined out of cold sprayed bulk copper deposits. The characteristic values were obtained with regard to the direction of deposition, which is defined by the direction of the robot’s movement. Thus, for the investigation of the component properties, specimens were provided that had been produced both longitudinally and transversely as well as orthogonally to the direction of deposition. The results of the investigations show that both the electrical conductivity and the mechanical load capacity of the specimen have a strong preferential direction of the specimen orientation with respect to the direction of deposition. Furthermore, it could be shown that by increasing the deposition height, there is an increasing oxygen content in the sample material, combined with increasingly significant defect networks. These effects have a negative impact on the electrical conductivity as well as on the mechanical load capacity. As a conclusion, further need for investigation is identified in the optimization of the process parameters as well as in the deposition strategy for the additive manufacturing of large-volume components with cold spray.
Microplastic detection in water samples becomes important for tracing microplastic sources. Microplastic may harm desalination facilities by blocking filters and disturbing the marine food chain. Thermo analytical methods such as pyrolysis gas chromatography mass spectroscopy, and spectroscopic methods like (micro) Raman spectroscopy or (micro) Fouriertransform infrared spectroscopy in combination with appropriate filters and sample preparation are suitable for analyzing microplastics on a scale from 1 µm to 1000 µm fast and unambiguous. While the thermo analytical methods are suitable for larger sample volumes, Raman spectroscopy and Fouriertransform infrared spectroscopy are able to detect and analyze single microplastic particles for instance in bottled water. Machine learning algorithms ensure a reliable classification of different plastic materials.
Microplastic (MP) contamination in natural water circulation is a concern for environmental issues and human health. Various types of polymer materials have been identified and were detected in MP analytic test procedures. Beyond MP polymer type, particle size and form play a major role in water analysis due to possible negative toxicologic effects on flora and fauna. However, the correct quantitative measurement of MP size distribution over several orders of magnitude is strongly influenced by sample preparation, filtration materials and processes, and microanalytical techniques, as well as data acquisition and analysis. In this paper, a reference methodology is presented aiming at an improved quantitative analysis of MP particles. An MP analysis workflow is demonstrated including all steps from reference materials to sample preparation, filtration handling, and MP particle size distribution analysis. Background-corrected particle size distributions (1–1000 µm) have been determined for defined polyethylene (PE) and polyethylene terephthalate (PET) reference samples. Microscopically measured particle numbers and errors have been cross-checked with the total initial mass. In particular, defined reference MP samples (PE, PET) are initially characterized and applied to filtration experiments. Optical microscopy imaging on full-area Si filters with subsequent image analysis algorithms is used for statistical particle size distribution analysis. To quantify the effects of handling and filtration, several blind tests with distilled water are carried out to determine the particle background for data evaluation. Particle size distributions of PE and PET reference samples are qualitatively and quantitatively reproduced with respect to symmetry, and maximum and cut-off diameter of the distribution. It is shown that especially MP particles with a radius of >50 µm can be detected and retrieved with high reliability. For particle sizes <50 µm, a significant interference with background contamination is observed. Data from blank samples allows a correction of background contaminations. Furthermore, for enhanced sampling statistics, the recovery of the initial amount of MP will be qualitatively shown. The results are intended as an initial benchmark for MP analytics quality. This quality is based on statistical MP particle distributions and covers the complete analytic workflow starting from sample preparation to filtration and detection. Microscopic particle analysis provides an important supplement for the evaluation of established spectroscopic methods such as Fourier-transform infrared spectroscopy or Raman spectroscopy.
Exploratory analysis of hyperspectral FTIR data obtained from environmental microplastics samples
(2020)
Hyperspectral imaging of environmental samples with infrared microscopes is one of the preferred methods to find and characterize microplastics. Particles can be quantified in terms of number, size and size distribution. Their shape can be studied and the substances can be identified. Interpretation of the collected spectra is a typical problem encountered during the analysis. The image datasets are large and contain spectra of countless particles of natural and synthetic origin. To supplement existing Analysis pipelines, exploratory multivariate data analysis was tested on two independent datasets. Dimensionality reduction with principal component analysis (PCA) and uniform manifold approximation and projection (UMAP) was used as a core concept. It allowed for improved visual accessibility of the data and created a chemical two-dimensional image of the sample. Spectra belonging to particles could be separated from blank spectra, reducing the amount of data significantly. Selected spectra were further studied, also applying PCA and UMAP. Groups of similar spectra were identified by cluster analysis using k-means, density based, and interactive manual clustering. Most clusters could be assigned to chemical species based on reference spectra. While the results support findings obtained with a ‘targeted analysis’ based on automated library search, exploratory analysis points the attention towards the group of unidientified spectra that remained and are otherwise easily overlooked.
Reversible light-modulation of fulgimide based monolayers on Si(1 1 1) and Si(1 0 0) was investigated using ATR-FTIR spectroscopy. Fulgimide monolayers were prepared from neat COOH-terminated SAMs on Si(1 1 1) obtained from methyl undec-10-enoate, (1:1)-diluted COOH-terminated monolayers on Si(1 1 1), and GPTMS monolayers on Si(1 0 0). The epoxy-terminated monolayer on oxidized Si(1 0 0) was characterized with ellipsometry, XPS, as well as contact angle measurements, and ATR-FTIR spectroscopy revealed a strong influence of toluene water content on reproducible high-quality monolayer formation. The results of this study show that environmental polarity has a strong influence on fulgimide imide IR band locations and read-out options for the two photostationary states PSS(365 nm), containing E/Z- and C-isomers, and PSS(545 nm), with solely the E/Zisomers.
Neat COOH-terminated monolayers on flat Si(1 1 1) have the Advantage of high functional Group concentration, orientation and stability, and an upright arrangement of fulgimide head groups.
Reliable simulation of polymers on an atomistic length scale requires a realistic representation of the cured material. A molecular modelling method for the curing of epoxy systems is presented, which is developed with respect to efficiency while maintaining a well equilibrated system. The main criterion for bond formation is the distance between reactive groups and no specific reaction probability is prescribed. The molecular modelling is studied for three different mixing ratios with respect to the curing evolution of reactive Groups and the final curing stage. For the first time, the evolution of reactive groups during the curing process predicted by the molecular modelling is validated with near-infrared spectroscopy data, showing a good agreement between simulation results and experimental measurements. With the proposed method, deeper insights into the curing mechanism of epoxy systems can be gained and it allows us to provide reliable input data for molecular Dynamics simulations of material properties.
The aim of the ZIKA research project, funded by the BMBF funding program FORKA (FKZ:15S9446 A-C), is the automated detection of internal corrosion of radioactive drums using non-destructive testing (NDT). The newly gained findings will be combined with research results from the previous project EMOS (FKZ:15S9420), which dealt with the external damage of drums. Using NDT, internal corrosion and possible internal sources of damage can be identified before they become a safety-relevant issue. However, if internally sourced damages can be seen externally, the integrity of the damaged drum is no longer guaranteed, which has significant consequences. Therefore, early detection before integrity failure is of particular importance for interim storage facilities with low- and medium-level radioactive waste drums.
A novel phosphorus-modified polysulfone (P-PSu) was employed as a combined toughness modifier and a source of flame retardancy for a DGEBA/DDS thermosetting system. In comparison to the results of a commercially available polysulfone (PSu), commonly used as a toughness modifier, the chemorheological changes during curing measured by means of temperature-modulated DSC revealed an earlier occurrence of mobility restrictions in the P-PSu-modified epoxy. A higher viscosity and secondary epoxy-modifier reactions induced a sooner vitrification of the reacting mixture; effects that effectively prevented any phase separation and morphology development in the resulting material during cure. Thus, only about a 20% increase in fracture toughness was observed in the epoxy modified with 20 wt.% of P-PSu, cured under standard conditions at 180 °C for 2 h. Blends of the phosphorus-modified and the standard polysulfone (PSu) were also prepared in various mixing ratios and were used to modify the same thermosetting system. Again, no evidence for phase separation of the P-PSu was found in the epoxy modified with the P-PSu/PSu blends cured under the selected experimental conditions. The particular microstructures formed upon curing these novel materials are attributed to a separation of PSu from a miscible P-PSuepoxy mixture. Nevertheless, the blends of P-PSu/PSu were found to be effective toughness/flame retardancy enhancers owing to the simultaneous microstructure development and polymer interpenetration.
