Currently research in chemical manufacturing moves towards flexible plug-and-play approaches focusing on modular plants, capable of producing small scales on-demand with short down-times between individual cam-paigns. This approach allows for efficient use of hardware, a faster optimization of the process conditions, and thus, an accelerated introduction of new products to the market. Driven mostly by the search for chemical syntheses under biocompatible conditions, so-called “click” chemistry rapidly became a growing field of research. The re-sulting simple one-pot reactions are so far only scarcely accompanied by an adequate optimization via compara-bly straightforward and robust analysis techniques. Here we report on a fast and reliable calibration-free online high field NMR monitoring approach for technical mixtures. It combines a versatile fluidic system, continuous-flow measurement with a time interval of 20 s per spectrum, and a robust, automated algo-rithm to interpret the ob-tained data. All spectra were acquired using a 500 MHz NMR spectrometer (Varian) with a dual band flow probe having a 1/16 inch polymer tubing working as a flow cell. Single scan 1H spectra were recorded with an acquisition time of 5 s, relaxation delay of 15 s.

With the introduction of advanced process analytical technology, the closeness of key process variables to their limits can be directly controlled and the product can be classified or even released in real time.
Compact NMR instruments can make NMR spectroscopy accessible in industrial and harsh environments for process control.
Aktuell wird von der Prozessindustrie die Weiterentwicklung einiger Messaufgaben und Technologien als sehr bedeutend eingeschätzt. Dazu zählen Infrarot-Kaskadenlaser, die Online-Analyse von Partikeln und Emulsionen, neuartige NIR/MIR-Spektroskopiemodule und deren Miniaturisierung sowie die Weiterentwicklung und Kostenvergünstigung der Raman-Spektroskopie. Heute wie morgen müssen Prozess-Sensoren zuerst die Aufgabe des robusten Messens und der Generierung von Prozessinformationen erfüllen.
Supernatants from a fermentation process of Pichia pastoris were investigated by Raman spectroscopy. Using partial least squares regression, the principal substrates glycerol and methanol could be predicted, however not the expressed protein. To gain further insight, a priori prepared calibration samples were studied by vibrational-, UV/Vis-, and fluorescence spectroscopy. For the quantification of glycerol and methanol, Raman spectroscopy was identified as the most sensitive technique, and superior to near-infrared spectroscopy, but not for protein contents below 1 g L–1. Both UV/Vis absorption and fluorescence spectroscopy are well suited for the quantification of protein, however, best results were obtained with UV/Vis absorption.
Smart sensors and smart reference materials – an approach to the industrial internet of things

The BAM targets within Bonares I4S are adaption of two online sensors being optimised for mobile applications: A LIBS spectrometer (Laser Induced Breakdown Spectroscopy) as well an a XRF spectrometer (X-Ray Fluorescence
Spectroscopy). Beyond , the certification of soil reference materials is scope of I4S at BAM. Therefore, a managable relational database structure is needed, based on a modular Architecture, which is dedicated to spectroscopy. The requirements to such a database are discussed.
Online NMR spectroscopy is an excellent tool to study complex reacting multicomponent mixtures and gain process insight and understanding. For online studies under process conditions, flow NMR probes can be used in a wide range of temperature and pressure. This paper compiles the most important aspects towards quantitative process NMR spectroscopy in complex multicomponent mixtures and provides examples. After NMR spectroscopy is introduced as an online method and for technical samples without sample preparation in deuterated solvents, influences of the residence time distribution, pre-magnetization, and cell design are discussed. NMR acquisition and processing parameters as well as data preparation methods are presented and the most practical data analysis strategies are introduced.
qNMR provides the most universally applicable form of direct purity determination without need for reference materials of impurities or the calculation of response factors but only exhibiting suitable NMR properties. The development of CRMs addressing qNMR specific measurement issues will give analysts compounds ideally suited for the analytical method and also provide full characterisation of qNMR related parameters to enable more realistic uncertainty budgets. These materials will give users the tools to exploit qNMR more easily and enable them to speed up analytical method development and reduce the time and financial burden of multiple analytical testing.
Forderungen nach höherer Geschwindigkeit und die Komplexität der Fragen geben der analytischen Chemie neue Impulse. So nutzt die Prozessanalytik gepulste Ramanspektrometer, die Elemente Arsen und Quecksilber dominieren die Forschung in der Speziesanalytik, und die Omics-Techniken entwickeln sich zu Multi-Omics-Ansätzen. Ambiente MS-Techniken benötigen keine aufwendige Probenvorbereitung, multidimensionale Trenntechniken werden verstärkt in der Routine eingesetzt, und Chip-basierte Trennungen fallen durch Schnelligkeit auf. Molekülspektroskopie und Massenspektrometrie dominieren die bildgebenden Verfahren, und die Lateralauflösung der ToF-Sekundärionenmassenspektrometrie hat sich bei Oberflächenuntersuchungen verbessert.
Homogen katalysierte Reaktionen sind wichtige Werkzeuge der chemischen Industrie. Milde Reaktionsbedingungen und hohe Selektivitäten führen zu einer energie- und ressourcenschonenden Produktion. Ein bedeutender Prozessschritt ist die Hydroformylierung. Hier kommen Kobalt- und Rhodiumkomplexe mit mehrzähnigen Liganden zum Einsatz, die zumeist in wässriger Lösung vorliegen. Die Anwendbarkeit beschränkt sich demnach auf kurzkettige Edukte mit hinreichender Wasserlöslichkeit.
Process monitoring of an intensified continuous production unit with compact NMR spectroscopy

Monitoring chemical reactions is the key to chemical process control. Today, mainly optical online methods are applied. NMR spectroscopy has a high potential for direct loop process control. Compact NMR instruments based on permanent magnets are robust and relatively inexpensive analyzers, which feature advantages like low cost, low maintenance, ease of use, and cryogen-free operation. Instruments for online NMR measurements equipped with a flow-through cell, possessing a good signal-to-noise-ratio, sufficient robustness, and meeting the requirements for integration into industrial plants (i.e., explosion safety and fully automated data analysis) are cur-rently not available off the rack. Recently, promising benchtop NMR instruments with acceptable performance came to market and process integrated sensors developed on basis of such laboratory instruments are on their way.
