There have been an increasing number of publications on flow chemistry applications of compact NMR. Despite this, there is so far no comprehensive workflow for the technical design of flow cells. Here, we present an approach that is suitable for the design of an NMR flow cell with an integrated static mixing unit. This design moves the mixing of reactants to the active NMR detection region within the NMR instrument, presenting a feature that analyses chemical reactions faster (5–120 s region) than other common setups. During the design phase, the targeted mixing homogeneity of the components was evaluated for different types of mixing units based on CFD simulation. Subsequently, the flow cell was additively manufactured from ceramic material and metal tubing. Within the targeted working mass flow range, excellent mixing properties as well as narrow line widths were confirmed in validation experiments, comparable to common glass tubes.

This key comparison aims to assess the capabilities of the participants to determine the amount-of-substance fraction oxygen in nitrogen. The GAWG has classified this as a track B comparison, due to the unexpected 50 μmol/mol argon mole fraction content of the transfer standards, which effects the achievable performance of some measurement techniques such a GC-TCD. The separation of oxygen and argon is challenging, and not all systems in use are equally well designed for it. As this analytical challenge due to a substantial fraction of argon in the transfer standards became a reality, the Gas Analysis Working Group (GAWG) decided to qualify this key comparison as a regular key comparison and not as a core comparison, which may be used to support calibration and measurement capabilities (CMCs) for oxygen in nitrogen, or for oxygen in nitrogen mixtures containing argon only (see also the section on support to CMCs).
Optimizing the Green Synthesis of ZIF-8 by Reactive Extrusion Using In Situ Raman Spectroscopy

We report the scale-up of a batch solid synthesis of zeolitic imidazolate framework-8 (ZIF-8) for reactive extrusion. The crystalline product forms in the extruder directly under the mixture of solid 2-methylimidazole and basic zinc carbonate in the presence of a catalytic amount of liquid. The process parameters such as temperature, liquid type, feeding rate, and linker excess were optimized using the setup specifically designed for in situ Raman spectroscopy. Highly crystalline ZIF-8 with a Brunauer–Emmett–Teller (BET) surface area of 1816 m2 g–1 was quantitatively prepared at mild temperature using a catalytic amount of ethanol and a small excess of the linker. Finally, we developed a simple and comprehensive approach to evaluating the environmental friendliness and scalability of metal–organic framework (MOF) syntheses in view of their large-scale production.
Reaction monitoring in disperse systems, such as emulsions, is of significant technical importance in various disciplines like biotechnological engineering, chemical industry, food science, and a growing number other technical fields. These systems pose several challenges when it comes to process analytics, such as heterogeneity of mixtures, changes in optical behavior, and low optical activity. Concerning this, online nuclear magnetic resonance (NMR) spectroscopy is a powerful technique for process monitoring in complex reaction mixtures due to its unique direct comparison abilities, while at the same time being non-invasive and independent of optical properties of the sample. In this study the applicability of online-spectroscopic methods on the homogeneously catalyzed hydroformylation system of 1-dodecene to tridecanal is investigated, which is operated in a mini-plant scale at Technische Universität Berlin. The design of a laboratory setup for process-like calibration experiments is presented, including a 500 MHz online NMR spectrometer, a benchtop NMR device with 43 MHz proton frequency as well as two Raman probes and a flow cell assembly for an ultraviolet and visible light (UV/VIS) spectrometer. Results of high-resolution online NMR spectroscopy are shown and technical as well as process-specific problems observed during the measurements are discussed.
A major industrial reaction based on homogeneous catalysis is hydroformylation for the production of aldehydes from alkenes and syngas. Hydroformylation in microemulsions, which is currently under investigation at Technische Universität Berlin on a mini-plant scale, was identified as a cost efficient approach which also enhances product selectivity. Herein, we present the application of online Raman spectroscopy on the reaction of 1-dodecene to 1-tridecanal within a microemulsion. To achieve a good representation of the operation range in the mini-plant with regard to concentrations of the reactants a design of experiments was used. Based on initial Raman spectra partial least squares regression (PLSR) models were calibrated for the prediction of 1-dodecene and 1-tridecanal. Limits of predictions arise from nonlinear correlations between Raman intensity and mass fractions of compounds in the microemulsion system. Furthermore, the prediction power of PLSR models becomes limited due to unexpected by-product formation. Application of the lab-scale derived calibration spectra and PLSR models on online spectra from a mini-plant operation yielded promising estimations of 1-tridecanal and acceptable predictions of 1-dodecene mass fractions suggesting Raman spectroscopy as a suitable technique for process analytics in microemulsions.
Der Artikel ist ein Editorial, der für die Herstellung eines grundlegenden Vertrauens in medizinische Produkte - insbesondere Impfstoffe - wirbt, um Skeptiker zu Unterstützern zu machen. Er wirbt nicht nur für die Impfstoffe, sondern auch für die Kultur der Pharmazie und Chemie und die Wissenschaft ganz allgemein und regt an, mehr erklärende Formate wie z. B. im Stil der Sendung mit der Maus für diese Dinge zu werben. Der Artikel geht auch auf den ungeheuren Reiz der seit gut zwanzig Jahren diskutierten risikobasierten Ansätze für pharmazeutische Entwicklung und Herstellung von Arzneimitteln auf Basis der Quality Guidelines ein.
Compact nuclear magnetic resonance (NMR) instruments make NMR spectroscopy and relaxometry accessible in industrial and harsh environments for reaction and process control. An increasing number of applications are reported. To build an interdisciplinary bridge between “process control” and “compact NMR”,we give a short overviewon current developments in the field of process Engineering such as modern process design, integrated processes, intensified processes along with requirements to process control, model based control, or soft sensing. Finally, robust field integration of NMR systems into processes environments, facing explosion protection or Integration into process control systems, are briefly discussed.
Driven mostly by the search for chemical syntheses under biocompatible conditions, so called "click" chemistry rapidly became a growing field of research. The resulting simple one-pot reactions are so far only scarcely accompanied by an adequate optimization via comparably straightforward and robust analysis techniques possessing short set-up times. Here, we report on a fast and reliable calibration-free online NMR monitoring approach for technical mixtures. It combines a versatile fluidic system, continuous-flow measurement of 1H spectra with a time interval of 20 s per spectrum, and a robust, fully automated algorithm to interpret the obtained data. As a proof-of-concept, the thiol-ene coupling between N-boc cysteine methyl ester and allyl alcohol was conducted in a variety of non-deuterated solvents while its time-resolved behaviour was characterized with step tracer experiments. Overlapping signals in online spectra during thiol-ene coupling could be deconvoluted with a spectral model using indirect hard modeling and were subsequently converted to either molar ratios (using a calibrationfree approach) or absolute concentrations (using 1-point calibration). For various solvents the kinetic constant k for pseudo-first order reaction was estimated to be 3.9 h-1 at 25 °C. The obtained results were compared with direct integration of non-overlapping signals and showed good agreement with the implemented mass balance.

Eine gleich bleibend effektive Nutzung von Anlagen, Rohstoffen und Energie in Verbindung mit der optimalen Prozessführung sind die wichtigsten Voraussetzungen für globale Wettbewerbsvorteile der Prozessindustrie.
Unternehmen aus den Bereichen Chemie, Pharmazie, Petrochemie, Gasverarbeitung, Lebensmittel-, Zellstoff-, Papier-, Glas-, Stahl- und Zementherstellung sowie ihren Zulieferern haben den Einsatz zuverlässiger Prozessautomation für eine gute Prozessführung erkannt. Überlegene Verfahren stützen sich zunehmend auf prozessanalytische Messtechnik zur Kontrolle und präventiven Sicherstellung der geforderten Produktqualität. Gleichzeitig ermöglicht sie eine ständige Verbesserung des Wissens über den Prozess und erhöhen die Prozess- und Anlagensicherheit.
Vor 100 Jahren, am 22. Mai 1913, erhielt Dr. Paul Gmelin, Mitarbeiter im physikalischen Laboratorium der Badischen Anilin und Soda Fabrik in Ludwigshafen das erste Patent auf ein Prozessanalysenmessgerät. Dieses Jubiläum würdigt CITplus mit diesem Beitrag, in dem die Autoren nicht nur die vergangene sondern auch eine mögliche künftige Entwicklung der Prozessanalysentechnik (PAT) Revue passieren lassen.
Medium resolution nuclear magnetic resonance (MR-NMR) spectroscopy is currently a fast developing field, which has an enormous potential to become an important analytical tool for reaction monitoring, in hyphenated techniques, and for systematic investigations of complex mixtures. The recent developments of innovative MR-NMR spectrometers are therefore remarkable due to their possible applications in quality control, education, and process monitoring. MR-NMR spectroscopy can beneficially be applied for fast, non-invasive, and volume integrating analyses under rough environmental conditions.
Within this study, a simple 1/16'' fluorinated ethylene propylene (FEP) tube with an ID of 0.04'' (1.02 mm) was used as a flow cell in combination with a 5 mm glass Dewar tube inserted into a benchtop MR-NMR spectrometer with a 1H Larmor frequency of 43.32 MHz and 40.68 MHz for 19F. For the first time, quasi-simultaneous proton and fluorine NMR spectra were recorded with a series of alternating 19F and 1H single scan spectra along the reaction time coordinate of a homogeneously catalysed esterification model reaction containing fluorinated compounds. The results were compared to quantitative NMR spectra from a hyphenated 500 MHz online NMR instrument for validation. Automation of handling, pre-processing, and analysis of NMR data becomes increasingly important for process monitoring applications of online NMR spectroscopy and for its technical and practical acceptance. Thus, NMR spectra were automatically baseline corrected and phased using the minimum entropy method. Data analysis schemes were designed such that they are based on simple direct integration or first principle line fitting, with the aim that the analysis directly revealed molar concentrations from the spectra.
Finally, the performance of 1/16'' FEP tube set-up with an ID of 1.02 mm was characterised regarding the limit of detection (LOQ (1H) = 0.335 mol L-1 and LOQ (19F) = 0.130 mol L-1 for trifluoroethanol in D2O (single scan)) and maximum quantitative flow rates up to 0.3 mL min-1. Thus, a series of single scan 19F and 1H NMR spectra acquired with this simple set-up already presents a valuable basis for quantitative reaction monitoring.
In der Forschung wird die Kernspinresonanzspektroskopie (NMR-Spektroskopie von englisch: nuclear magnetic resonance) für die Strukturaufklärung und Quantifizierung genutzt. Eine Arbeitsgruppe an der Berliner Bundesanstalt für Materialforschung und -prüfung (BAM) und am Karlsruher Institut für Technologie (KIT) hat die Möglichkeiten untersucht, die NMR-Spektroskopie auch als Online-Methode für das Monitoring chemischer Reaktionen in der Prozesskontrolle einsetzbar zu machen.
Die Entwicklung innovativer Medium-Resolution-NMR-Spektrometer eröffnet eine Reihe bemerkenswerter Applikationen für die Qualitätskontrolle, die akademische Ausbildung und die Prozessanalytik. Die Verwendung von Permanentmagneten erlaubt den Einsatz der NMR-Spektroskopie in industriellen Prozessumgebungen.
This project concerns the purity analysis of nitrogen as used in reference gas mixture preparation. This project was carried out without adding impurities to the gas used for this comparison, and is therefore more representative to evaluate the analysis of CO, CO2, CH4, O2, Ar and H2O impurities in high purity nitrogen. The analysis of the amount–of–substance fraction water was optional.
Two 50 litre high purity nitrogen cylinders were purchased from a well-qualified supplier of specialty gases. The listed components were expected to be present in the pure nitrogen at the target levels as a result of the purification of the nitrogen. From the start of this comparison it was clear that the comparison may not lead to reference values for the constituents analysed.
The results indicate that analyses of high purity gases are often limited by the limits of detection of analytical equipment used. The reports of the participating laboratories also indicate that there is no agreed method of determination of the uncertainty on a detection Limit value. The results provide useful information on the Performance of participants. For all analysed components there is reasonable agreement in results for LNE, VSL, Metas and NPL.
For BAM only the Argon result is in agreement.
Medium-resolution nuclear magnetic resonance spectroscopy (MR-NMR) currently develops to an important analytical tool for both quality control and processmonitoring. In contrast to high-resolution onlineNMR (HR-NMR),MR-NMRcan be operated under rough environmental conditions. A continuous re-circulating stream of reaction mixture fromthe reaction vessel to the NMR spectrometer enables a non-invasive, volume integrating online analysis of reactants and products. Here, we investigate the esterification of 2,2,2-trifluoroethanol with acetic acid to 2,2,2-trifluoroethyl acetate both by 1H HR-NMR (500MHz) and 1H and 19F MRNMR (43MHz) as amodel system. The parallel online measurement is realised by splitting the flow,which allows the adjustment of quantitative and independent flow rates, both in the HR-NMR probe as well as in the MR-NMR probe, in addition to a fast bypass line back to the reactor. One of the fundamental acceptance criteria for online MR-MNR spectroscopy is a robust data treatment and evaluation strategy with the potential for automation. The MR-NMR spectra are treated by an automated baseline and phase correction using the minimum entropy method. The evaluation strategies comprise (i) direct integration, (ii) automated line fitting, (iii) indirect hard modelling (IHM) and (iv) partial least squares regression (PLS-R). To assess the potential of these evaluation strategies for MR-NMR, prediction results are compared with the line fitting data derived from the quantitative HR-NMR spectroscopy. Although, superior results are obtained from both IHM and PLS-R for 1H MR-NMR, especially the latter demands for elaborate data pretreatment, whereas IHM models needed no previous alignment.
Die gravierenden Veränderungen im Umfeld der Informations- und Kommunikationstechnik beeinflussen auch die Prozessautomation und sind zugleich eine Chance. Denn die Automatisierungskonzepte gemäß Industrie 4.0 sichern den Wettbewerbsvorteil einer Volkswirtschaft. Ein zentrales Element bilden dabei smarte Sensoren, die Dienste innerhalb eines Netzwerks bereitstellen und Information daraus nutzen. Die im November 2015 vorgestellte Technologieroadmap Prozess-Sensoren 4.0 [1] zeigt Anforderungen an smarte Prozess-Sensoren auf. Der Beitrag befasst sich mit den Fragen „Was kann der Prozess-Sensor zu Industrie 4.0 beitragen?“ und „Welchen Nutzen bringt Industrie 4.0 für den Prozess-Sensor?“
Heute wie morgen müssen Prozess-Sensoren zuerst die Aufgabe des Messens und der Generierung von Prozessinformationen erfüllen. Die Mess-Signale gehen in der Regel auf physikalische oder chemische Größen zurück, werden durch eine Kennlinie repräsentiert und lassen sich kalibrieren. Heute muss man dazu einen großen Aufwand betreiben, denn die physikalischen oder chemischen Informationen werden in vielen Fällen nicht direkt gemessen, sondern verbergen sich in Rohdaten, z. B. in einem optischen Spektrum. Diese Kalibrierungen können für komplexe Sensoren teilweise mehr Kostenaufwand bedeuten, als das Messgerät selbst.
Online NMR spectroscopy is an excellent tool to study complex reacting multicomponent mixtures and gain process insight and understanding. For online studies under process conditions, flow NMR probes can be used in a wide range of temperature and pressure. This paper compiles the most important aspects towards quantitative process NMR spectroscopy in complex multicomponent mixtures and provides examples. After NMR spectroscopy is introduced as an online method and for technical samples without sample preparation in deuterated solvents, influences of the residence time distribution, pre-magnetization, and cell design are discussed. NMR acquisition and processing parameters as well as data preparation methods are presented and the most practical data analysis strategies are introduced.
Supernatants from a fermentation process of Pichia pastoris were investigated by Raman spectroscopy. Using partial least squares regression, the principal substrates glycerol and methanol could be predicted, however not the expressed protein. To gain further insight, a priori prepared calibration samples were studied by vibrational-, UV/Vis-, and fluorescence spectroscopy. For the quantification of glycerol and methanol, Raman spectroscopy was identified as the most sensitive technique, and superior to near-infrared spectroscopy, but not for protein contents below 1 g L–1. Both UV/Vis absorption and fluorescence spectroscopy are well suited for the quantification of protein, however, best results were obtained with UV/Vis absorption.
Die Bundesanstalt für Materialforschung und -prüfung (BAM) ist eine wissenschaftlich technische Bundesoberbehörde im Geschäftsbereich des Bundesministeriums für Wirtschaft und Energie (BMWi), die über einen gesetzlichen Auftrag für die Sicherheit in Technik und Chemie verfügt und auf nahezu 150 Jahre zurückblicken kann. Vor diesem Hintergrund forscht, entwickelt, und prüft die BAM in den fünf Themenfeldern Energie, Infrastruktur, Umwelt, Material und Analytical Sciences mit einer unikalen Vielfalt an analytischen Methoden.
Die auf der NAMUR HS 2015 vorgestellte Technologie-Roadmap „Prozess-Sensoren 4.0“ zeigt die nötigen Anforderungen an Prozess-Sensoren sowie an deren Kommunikations-fähigkeiten auf.
Eine Topologie für smarte Sensoren, das Zusammenwirken mit daten- und modellbasierte Steuerungen bis hin zur Softsensorik sowie weitere Anforderungen an Sensoren sind heute noch nicht angemessen beschrieben. Wir müssen jetzt schnell die Weichen für eine smarte und sichere Kommunikationsarchitektur stellen, um zu einer störungsfreien Kommunikation aller Sensoren auf Basis eines einheitlichen Protokolls zu kommen, welches alle Sensoren ausgeben und verstehen.
Im Rahmen der Herstellung und Überwachung von Arzneimitteln käme wohl niemand – außer dem alten Paracelsus – auf die Idee, diese Aussage in irgendeiner Form anzuzweifeln. Zeit, der Pharmazeutischen Industrie endlich einmal ein Kompliment zu machen. Danke zu sagen, für die technisch, wissenschaftliche Kultur, die fast für jede Krankheit etwas parat hat und uns, unserer Familie, unseren Freunden ein unbeschwertes und langes Leben ermöglicht. Leute, verkauft Euch doch nicht immer so schlecht!
Aktuell wird von der Prozessindustrie die Weiterentwicklung einiger Messaufgaben und Technologien als sehr bedeutend eingeschätzt. Dazu zählen Infrarot-Kaskadenlaser, die Online-Analyse von Partikeln und Emulsionen, neuartige NIR/MIR-Spektroskopiemodule und deren Miniaturisierung sowie die Weiterentwicklung und Kostenvergünstigung der Raman-Spektroskopie. Heute wie morgen müssen Prozess-Sensoren zuerst die Aufgabe des robusten Messens und der Generierung von Prozessinformationen erfüllen.
Quantitative NMR spectroscopy for gas analysis for production of primary reference gas mixtures

Due to its direct correlation to the number of spins within a sample quantitative NMR spectroscopy (qNMR) is a promising method with absolute comparison abilities in complex systems in technical, as well as metrological applications. Most of the samples studied with qNMR are in liquid state in diluted solutions, while gas-phase applications represent a rarely applied case. Commercially available NMR equipment was used for purity assessment of liquid and liquefied hydrocarbons serving as raw materials for production of primary reference gas standards. Additionally, gas-phase studies were performed within an online NMR flow probe, as well as in a high-pressure NMR setup to check feasibility as verification method for the composition of gas mixtures.
Mechanochemistry is increasingly used as a ‘green alternative’ for synthesizing various materials including pharmaceutical cocrystals. Herein, we present the mechanochemical synthesis of three new cocrystals containing the API carbamazepine (cocrystals CBZ:Indometacin 1:1, CBZ:Benzamide 1:1, and CBZ:Nifedipine 1:1). The mechanochemical reaction was investigated in situ documenting a fast and complete reaction within one minute. Online NMR spectroscopy proved the direct influence of the dissolution behaviour of the coformers to the dissolution behaviour of the API carbamazepine. The dissolution behaviour of the organic cocrystals is compared to the behaviour of the pure drug indicating a general applicability of this approach for detailed cocrystal dissolution studies.
Flexible, modulare Produktionsanlagen auf der Basis von verfahrenstechnischen Teilmodulen stellen einen vielversprechenden Ansatz für die kontinuierliche Produktion von Fein- und Spezialchemikalien dar. Der Schwerpunkt des Horizont-2020-Projekts der Europäischen Kommission CONSENS (Integrated Control and Sensing) liegt in der Implementierung von innovativen Sensorkonzepten zur Prozessüberwachung und -regelung innerhalb von containerbasierten modularen Produktionsan lagen. In diesem Artikel wird die Feldintegration eines Online-NMR-Sensors als smartes Modul für die Prozesskontrolle beschrieben. Dieses Modul basiert auf einem kommerziell erhältlichen Niederfeld-NMRSpektrometer, das zurzeit für die Anwendung im Laborbereich erhältlich ist. Für die Feldintegration wurde ein ATEX-zertifiziertes, explosionsgeschütztes Gehäuse entwickelt sowie Automationsschemen für den unbeaufsichtigten Betrieb und für die spektrale Datenauswertung erstellt. Nachdem die Machbarkeit und Leistungsfähigkeit des Sensorkonzeptes in Laborexperimente an einer aromatischen Substitutionsreaktion bereits erfolgreich demonstriert wurde, ist die Inbetriebnahme des NMR-Sensormoduls in
industrieller Umgebung für 2017 geplant.
This key comparison aims to assess the core capabilities of the participants in gas analysis. Such competences include, among others, the capabilities to prepare Primary Standard gas Mixtures (PSMs), perform the necessary purity analysis on the materials used in the gas mixture preparation, the verification of the composition of newly prepared PSMs against existing ones, and the capability of calibrating the composition of a gas mixture. According to the Strategy for Key Comparisons of the Gas Analysis Working Group, this key comparison is classified as an RMO track A key comparison.
The artefacts were binary mixtures of propane in nitrogen at a nominal amount-of-substance fraction level of 1000 μmol/mol. The values and uncertainties from the gravimetric gas mixture preparation were used as key comparison reference values (KCRVs). Each transfer standard had its own KCRV. The results are generally good. All results are within ± 1 % of the KCRV.
In dieser Arbeit werden die Grundlagen der Normung, die Entstehung einer Norm und die für die Gasanalytik relevanten Normungsgremien kurz dargestellt. Dazu werden die wichtigsten Normen zur Gasanalytik entlang des analytischen Prozesses gruppiert und aufgeführt. Schließlich wird als Anwendungsbeispiel aus der eigenen Praxis der Prozess von der Herstellung bis zur Zertifizierung eines Kalibriergases beschrieben.
Zusammen mit der Genauigkeit von Messungen ist die ihnen zugeordnete Messunsicherheit Ausdruck und Maß von Vergleichbarkeit und Richtigkeit. Die Messunsicherheit spielt ebenso die Schlüsselrolle im Aufbau einer Rückführbarkeitskette, wie in diesem Beitrag beschrieben. Es ist der vierte und letzte Teil einer Artikelserie (siehe Infokasten), die Einblicke gibt in die Arbeit des DIN-Normenausschusses „Gasanalyse und Gasbeschaffenheit“ (DIN NA 062-05-73 AA) sowie der international tätigen Technical Committees 158 und 193 der ISO.
In vielen industriellen High-Tech Verfahren und Prozessen, Forschung und Wissenschaft, Medizin und Umwelttechnik werden Sondergase eingesetzt.
Als Gattungsbegriff umfasst diese Bezeichnung ein ansehnliches Spektrum höchster Qualitäten an Gasen. Dazu gehören u. a. Reinstgase mit besonders hohen Anforderungen an ihre Reinheit, Gasgemische genau definierter Zusammensetzung und Isotopengemische. Wie genau kann man Gaszusammensetzungen heute messen? Wie schafft man es, dass alle dasselbe messen? Welche Techniken kommen dabei zum Einsatz?
Mass Spectrometry (MS) and Nuclear Magnetic Resonance Spectroscopy (NMR) are valuable analytical and quality control methods for most industrial chemical processes as they provide information on the concentrations of individual compounds and by-products. These processes are traditionally carried out manually and by a specialist, which takes a substantial amount of time and prevents their utilization for real-time closed-loop process control. This paper presents recent advances from two projects that use Artificial Neural Networks (ANNs) to address the challenges of automation and performance-efficient realizations of MS and NMR. In the first part, a complete toolchain has been realized to develop simulated spectra and train ANNs to identify compounds in MS. In the second part, a limited number of experimental NMR spectra have been augmented by simulated spectra, to train an ANN with better prediction performance and speed than state-of-the-art analysis. These results suggest that, in the context of the digital transformation of the process industry, we are now on the threshold of a strongly simplified use of MS and MRS and the accompanying data evaluation by machine-supported procedures, and can utilize both methods much wider for reaction and process monitoring or quality control.
Die Wettbewerbsfähigkeit der Chemischen und Pharmazeutischen Industrie basiert auf der Sicherung der geforderten Produktqualität bei einer optimalen Nutzung von Anlagen, Rohstoffen und Energie [1]. Eine gute Prozessführung unter Einsatz zuverlässiger Prozessanalytik sichert hier den globalen Wettbewerbsvorteil gegenüber Niedriglohnländern, die mit weniger effizienten Verfahren produzieren. Prozessanalytik trägt zur Erhöhung der Prozess- und Anlagensicherheit bei. In Kombination mit geeigneten Informationsmanagementsystemen verbessert dieser Ansatz ständig das Wissen über den Prozess und führt damit zu einer präventiven Sicherstellung der geforderten Qualität.
The design of sample flow cells, commonly used in on-line analytics and especially for medium resolution NMR spectroscopy (MR-NMR) in low magnetic fields, was experimentally and theoretically investigated by 1H NMR and numerical simulations. The flow pattern was characterised to gain information about the residence time distribution and mixing effects. Both 1H NMR imaging and spectroscopy were used to determine the characteristics of flow cells and their significance for on-line measurements such as reaction monitoring or hyphenated separation spectroscopy. The volume flow rates investigated were in the range from 0.1 to 10 ml/min, typically applied in the above mentioned applications. The special characteristics of flow cells for MR-NMR were revealed by various NMR experiments and compared with CFD simulations and to flow cells commonly used in high-field NMR. The influence of the design of the inlet and outlet on the flow pattern was investigated as well as the effect of the length of the cell. For practical use, a numerical estimation of the inflow length was given. In addition, it was shown how experiments on the polarisation build-up revealed insight into the flow characteristics in MR-NMR.

The application of Raman spectroscopy as a monitoring technique for bioprocesses is severely limited by a large background signal originating from fluorescing compounds in the culture media. Here, we compare time-gated Raman (TG-Raman)-, continuous wave NIRprocess Raman (NIR-Raman), and continuous wave micro-Raman (micro-Raman) approaches in combination with surface enhanced Raman spectroscopy (SERS) for their potential to overcome this limit. For that purpose, we monitored metabolite concentrations of Escherichia coli bioreactor cultivations in cell-free supernatant samples. We investigated concentration transients of glucose, acetate, AMP, and cAMP at alternating substrate availability, from deficiency to excess. Raman and SERS signals were compared to off-line metabolite analysis of carbohydrates, carboxylic acids, and nucleotides. Results demonstrate that SERS, in almost all cases, led to a higher number of identifiable signals and better resolved spectra. Spectra derived from the TG-Raman were comparable to those of micro-Raman resulting in well-discernable Raman peaks, which allowed for the identification of a higher number of compounds. In contrast, NIR-Raman provided a superior performance for the quantitative evaluation of analytes, both with and without SERS nanoparticles when using multivariate data analysis.
Digitalisierung und Industrie 4.0 verändern komplette Geschäftsmodelle, heben neue Effizienzpotenziale und stärken die Wettbewerbsfähigkeit. Auf dem 57. Tutzing-Symposion vom 15.–18.04.2018 wurde mit Vorträgen und Kreativworkshops erkundet, welche speziellen Anforderungen die Prozessindustrie hat, welche digitalen Innovationen bereits umgesetzt wurden und wo noch Handlungsbedarf besteht. Ein Workshop befasste sich mit den Themenfeldern Datenkonzepte, Datenanalyse, Big Data und künstliche Intelligenz. Es geht nicht um die Digitalisierung von heute. Im Angesicht der wachsenden Digitalisierung unserer Prozesse stellt sich die Frage, ob wir den Prozess wirklich gut kennen. Ob alle Verfahrensschritte detailliert hinterlegt wurden. Nur mit einem heuristischen Ansatz kann das vorhandene Wissen nicht digitalisiert werden.
Sehr schnell werden die Mechanismen eines Massenmarktes mit denen einer Nische verwechselt. Nicht jeder Mechanismus, den wir von großen Suchmaschinen oder Einkaufsportalen kennen, gibt uns einen Hinweis auf Nutzen und Verfügbarkeit für die Prozess- oder pharmazeutische Industrie. Eine gute Analyse der Anforderungen in der Zukunft mit einem Abgleich der derzeitigen technischen Möglichkeiten ist Voraussetzung für eine Verbesserung der derzeitigen digitalen Umsetzung. Dabei ist es sinnvoll unkonventionelle Methoden einzusetzen.
Für die Rückführung von aus Rückbauprojekten anfallender künstlicher Mineralwolle in ihren Herstellungsprozess ist es notwendig, Stein- und Glaswolle zu unterscheiden und voneinander getrennt zu halten. Zu diesem Zweck wurde das Potential von NIR-Spektroskopie (NIRS) für einen werks- oder baustellenseitigen Einsatz getestet. NIRS wird aufgrund kurzer Messzeiten, minimaler Probenvorbereitung und hoher Robustheit der Spektrometer häufig in der Prozessanalytik eingesetzt. Untersucht wurden 70 verschiedene Mineralwollen, wobei sich der Probenumfang sowohl aus werksneuen Proben mit Herstellerangaben als auch unbekannten Proben ohne Spezifikation zusammensetzen, die auf verschiedenen Baustellen in den Jahren 2016–2017 entnommen oder von Deponien zur Verfügung gestellt worden waren. Als Referenzmethode wurde die Röntgenfluoreszenzanalyse verwendet, um den Mineralwolle-Typ über den Elementgehalt nach VDI 3492 zu identifizieren. Mit Hilfe eines multivariaten Datenanalyseverfahrens konnte schließlich eine Methode etabliert werden, die eine zuverlässige Identifikation von unbekannten Mineralwollen anhand deren NIR-Spektren als Stein- bzw. Glaswolle ermöglicht.
