A new dual purpose cask design was developed for the safe transport and interim storage of spent fuel elements of German research reactors. In the framework of the safety assessment within the package approval procedure the Bundesanstalt für Materialforschung und –prüfung (BAM) as competent authority performed a series of drop tests according with the IAEA Transport Regulations. The package consists of a cylindrical thick-walled ductile cast iron cask body closed by a bolted lid system with metallic seals. A lid and bottom sided impact limiter consisting of a wood/steel construction limit the mechanical impact loading. The full-scale test specimen was equipped with a basket and assembled with dummy-fuel elements. The package and test specimen, respectively have a total mass of approximately 24 metric tons. The mechanical drop test program included three 9m free drop tests, in horizontal, vertical and oblique cask orientation onto the lid system. Additionally, a 1m-puncture bar drop test followed the horizontal drop test to consider an IAEA-drop test sequence. The horizontal and vertical drop tests were performed at a temperature of minus 40°C. During the oblique drop test the upper impact limiter was heated up to +80°C. The tests were conducted onto an unyielding target, fulfilling the IAEA requirements. The test specimen was considerably instrumented with strain gauges and accelerometers. Transient strains at selected locations of the inner and outer container walls, of the primary and secondary lid, as well as of the corresponding lid bolts were measured during the drop tests. Furthermore, decelerations in different locations at the cask body and the lids were measured. The complex geometrical deformation of the impact limiters due to the impact were determined by optical 3D- measurements. Before and after the drop tests the leakage rate of the lid system was determined by helium leakage testing. The experimental results contribute to the evaluation of the package response to mechanical tests, demonstrating safety under normal and accident conditions of transport and especially to the verification of the dynamic finite-element model of the package used in the package design safety report.

Wasserzeichen und Papierstrukturen können einen wesentlichen Beitrag bei der Analyse von Kunstobjekten auf Papier darstellen. An Fallbeispielen aus der gutachterlichen Praxis und Forschungsprojekten der Vergangenheit sollen die Möglichkeiten der Papier- und Wasserzeichenforschung bei Datierungen und Authentizitätsprüfungen demonstriert werden.
Im Jahr 2019 jährt sich die Inbetriebnahme der ersten Papiermaschine in Deutschland zum 200. Male. Mit dem Vortrag wird eine Einführung in die Technologie der Maschinenpapierherstellung, samt entwicklungsgeschichtlicher Meilensteine gegeben. Es gilt die Produktvielfalt von Papier und die Gründe für die enormen Produktionssteigerungen bei der Papierherstellung aufzuzeigen.
The primary screening of hybridoma cells is a time-critical and laborious step during the development of monoclonal antibodies. Often critical errors occur in this phase, which supports the notion that the generation of monoclonal antibodies with hybridoma technology is difficult to control and hence a risky venture. We think that it is crucial to improve the screening process to eliminate most of the immanent deficits of the conventional approach. With this new microarray-based procedure, several advances could be achieved: Selectivity for excellent binders, high throughput, reproducible signals, avoidance of misleading avidity (multivalency) effects, and simultaneous performance of competition experiments. The latter can directly be used to select clones of desired cross-reactivity properties. In this paper, a model system with two excellent clones against carbamazepine, two weak clones and blank supernatant has been designed to examine the effectiveness of the new system. The excellent clones could be detected largely independent of the IgG concentration, which is unknown during the clone screening since the determination and subsequent adjustment of the antibody concentration is not possible in most cases. Furthermore, in this approach, the enrichment, isolation, and purification of IgG for characterization is not necessary. Raw cell culture supernatant can be used directly, even when fetal calf serum (FCS) or other complex media had been used. In addition, an improved method for the oriented antibody-immobilization on epoxy-silanized slides is presented. Based on the results of this model system, we conclude that this approach should be preferable to most other protocols leading to many of false positives, causing expensive and lengthy confirmation steps to weed out the poor clones.

Entwicklung eines luftgekoppelten Ultra-schall-Echo-Prüfverfahrens mittels fluidischer Anregung

In vielen technischen Bereichen werden Ultraschallverfahren zur zerstörungsfreien Werkstoffprüfung eingesetzt. Dabei wird ein Schallpuls von einem Prüfkopf in ein Prüfobjekt eingebracht. Der Puls kann unter anderem durch Membranen oder Pie-zoelemente erzeugt werden und wird in der Regel durch direkten Kontakt oder über ein Koppelmittel an das Objekt übertragen. Luftgekoppelter Ultraschall spielt in kommerziellen Anwendungen bisher eine untergeordnete Rolle, da die Differenz der akustischen Impedanzen von Luft und Festkörpern immense Verluste beim Über-gang des Schallsignals hervorruft.
In diesem Beitrag soll ein neuartiges Anregungsprinzip vorgestellt werden, mit dem ein Großteil dieser Verluste vermieden wird. Anstelle eines Festkörpers soll mit Hilfe einer fluidischen Düse Druckluft zur Signalerzeugung eingesetzt werden. In dieser Düse wird eine selbsterhaltende Strömungsinstabilität erzeugt, die einen Schallpuls von bis zu 100 kHz hervorruft, sodass der Impedanzverlust in die Umge-bungsluft entfällt. Da die charakteristische Frequenz eines fluidisch generierten Pul-ses maßgeblich von der Bauform der Düse und dem anliegenden Druck abhängt, lässt sich mittels einer geeigneten Strömungsregelung ein breiter Frequenzbereich zur Abtastung nutzen. Die so emittierten Pulse werden auf das Prüfobjekt gerichtet und die reflektierten Signale mit einem Laservibrometer an der Oberfläche des Objekts im Echo-Verfahren abgetastet. Von drei signalmindernden Materialübergängen bei gewöhnlichem luftgekoppeltem Ultraschall bleibt in dem hier vorgeschlagenen Mes-system lediglich die Grenzfläche von Luft zu Prüfkörper, sodass eine höhere Signal-ausbeute als bisher erwartet werden kann.
Zur Versiegelung von Endlagern für radioaktive oder chemische Abfälle in ehemaligen Salzbergwerken werden Verschlussbauwerke aus Salzbeton auf ihre Eignung geprüft. Seit einigen Jahren werden durch die Bundesgesellschaft Endlagerung (BGE) und der Bundesanstalt für Materialforschung und -prüfung (BAM) an Versuchsbauwerken Erfahrungen mit der Ultraschalltechnik zur Qualitätssicherung unter Tage gesammelt. Hierbei kommen sowohl kommerzielle und experimentelle Geräte zur Echoprüfung als auch eigens entwickelte Bohrlochsonden zum Einsatz. Alle Geräte basieren auf niederfrequenten s-Wellen-Punktkontaktprüfköpfen (25 kHz– 50 kHz), die in unterschiedlichen Arrays kombiniert werden. In mehreren Versuchsreihen gelang es, unverfüllte und teilverfüllte Risse über mehrere Meter zu verfolgen und in ihrer Lage abzubilden. Technische Einbauten konnten bis in ca. 8 m Tiefe im Bauwerk detektiert werden. Mit Hilfe aus der Geophysik adaptierte Abbildungsverfahren gelang es, einige bisher nicht detektierbare Features sichtbar zu machen.
Die Ergebnisse dienen sowohl der Analyse des Versuchsbauwerks als auch der Entwicklung eines Qualitätssicherungssystems für zukünftige reale Verschlussbauwerke.
