This presentation discusses industry 4.0 from the perspective of human machine interaction (HMI). Based on available literature the changes, new demands and possible problems related to people’s jobs as a result of increased digitisation and use of artificial intelligence are addressed. Combining literature with own experience in non-destructive testing (NDT), the author addresses the possible challenges and chances for the NDT personnel in the era of NDT 4.0.
The spatial heterodyne spectrometer (SHS) concept, which is based on an interferometricoptical setup, boasts both the Fellgett and Jacquinot advantages. Theoretically it can provideboosted sensitivity and spectral resolution with respect to dispersion spectrometers in acompact, reasonably cheap arrangement without any moving components – this set ofcharacteristics can be attractive to a number of industrial, space and other field applications. The potential of SHS has already been demonstrated in IR and Raman spectroscopies(e.g.), and more recently also in LIBS. The scientific goal of our present project is toapply the SHS concept to the development of an optimized, but practical dual-grating,tunable SH-LIBS setup, which would possess appealing spectroscopic characteristics.During this development, we extensively rely on the computer-based simulation of theoptical setup, which is an efficient approach that we found to have been missing from earlierSHS efforts published. It can provide application-specific optimization of the SHS systemand predict the performance of the final system. In particular, we use optical simulation tostudy the effect of various important parameters on the relevant spectroscopic figures ofmerits of the system. We used the non-sequential ray-tracing mode of the Zemax/OpticStudio software for optical modelling of the SH-LIBS setup (Optical distortions were studiedin sequential mode). Characteristics of the setup and interferograms were calculated with atleast one million rays. All calculations were carried out for the visible spectral range(400-700 nm), using stepwise extension of monochromatic simulations with 5 nm steps.Wherever applicable, characteristic discrete visible wavelengths from the emissionspectrum of Hg discharge lamps (404.7 nm, 435.8 nm, 546.1 nm, 579.0 nm) were used for thecalculation of spectroscopy figures of merit and for the validation of simulation.
Asymmetrical laser-induced plasmas were investigated by a tomography approach based onthe inverse Radon transform. Two distinct sources of asymmetricity were investigated:double-pulsed laser-induced plasmas in the orthogonal configuration and single-pulsedlaser-induced plasmas under an inclined incidence angle. Both cases were observed atvarious delay times. The optical thinness of the laser-induced plasmas was achieved byappropriately adjusting the pulse energies. High temporal resolution was achieved by agated intensified charge-coupled-device camera. The asymmetrical laser-induced plasmaswere investigated in terms of their total emissivity, spectrally resolved emissivity, andtemperature. The latter was obtained by the Saha–Boltzmann plot method. The imagesrequired for the inverse Radon transform technique were obtained with a high angularaccuracy and reproducibility provided by mounting the spectrometer on a high-precisionnano-positioning rotary stage. The plasmas were induced in the center of rotation of thestage. This arrangement enabled the reconstruction of emissivity which was integrated overthe full spectral range (200–800 nm) or over a desired spectral range selected by a bandpassfilter (~10 nm). It also allowed for the reconstruction of spectrally-resolved emissivity ineach cross-sectional plasma slice by scanning the plasma across a spectrometer slit. The 3Dmaps of the temperature and electron density were thus obtained for different types ofasymmetric plasmas. The work will provide a more detailed description of the twoasymmetrical laser-induced plasmas. This might help with the development of LIBSinstrumentation using the orthogonal double-pulse geometry, or remote LIBS applicationswhich inherently rely on inclined-angle ablation.
Rapid cooling rates and steep temperature gradients are characteristic of additively manufactured (AM) parts and important factors for residual stress formation which have implications on structural integrity. This study examined the influence of heat input on the distribution of residual stresses in two prisms produced by laser powder bed fusion (L-PBF) of austenitic stainless steel 316L.
The layers of the prisms were exposed using two distinct helix scanning strategies: one scanned from the centre to the perimeter and the other from the perimeter to the centre. Residual stresses were characterised at one plane perpendicular to the building direction at half of its build height using neutron diffraction. In addition, the defect distribution was analysed via micro X-ray computed tomography (µCT) in a twin specimen.
Both scanning strategies reveal residual stress distributions typical for AM: compressive stresses in the bulk and tensile stresses at the surface. However, temperature gradients and maximum stress levels differ due to the different heat input. Regarding the X-ray µCT results, they show an accumulation of defects at the corners where the laser direction turned through 90°.
The results demonstrate that neutron diffraction and X-ray µCT can be successfully used as non-destructive methods to analyse through-thickness residual stress and defect distribution in AM parts, and in the presented case, illustrate the influence of scanning strategies. This approach contributes to deeper assessment of structural integrity of AM materials and components.
Laser based Powder Bed Fusion (L-PBF) is an additive manufacturing technique that has been continuously developed in the past years. It offers unparalleled design freedom and the resulting mechanical properties match, in some cases even exceed, those of materials processed by conventional manufacturing techniques. Nonetheless the process is prone to create Residual Stresses (RS) resulting from the sequential melting and solidification of the material. RS can reduce load bearing capacity and generate unwanted distortions thus diminishing the potential of L-PBF. This research activity aimed at characterizing the RS state in Inconel 718 L-PBF specimens using multiple diffraction methods. The microstructure as well as the surface and bulk residual stresses were investigated. The RS analysis was performed using X-ray, synchrotron and neutron diffraction methods to provide information at different depths within the specimen. The measurements were performed at the Bundesanstalt für Materialforschung und –prüfung (BAM), the EDDI beamline at BESSY II synchrotron and E3 line at BER II neutron reactor of the Helmholtz-Zentrum für Materialien und Energie (HZB) Berlin. The results revealed a depth depending RS state. The longitudinal and transverse stress components measured by X-ray and synchrotron at the surface agree well, exhibiting stress values around the yield strength of the material. In addition, synchrotron mapping showed gradients along the width and length of the sample for the longitudinal and transverse stress components. Lower RS values compared to surface RS were measured in the bulk of the material using neutron diffraction. The longitudinal stress component in the bulk was tensile and gradually decreased towards the edge of the specimen. The normal component however did not change significantly along the specimen dimensions and was of compressive nature. The transversal component was almost negligible. The results indicate that a stress re-distribution takes place during the deposition of the consecutive layers, which has to be further investigated.
The key point of structural reliability analysis is the estimation of the failure probability. This probability is defined as the integral over the failure domain which is given by a limit state function. Usually, this function is only implicit given by an underlying finite element simulation of the structure. It is generally not possible to solve the integral analytically. For that reason, numerical methods based on sampling and surrogates have been developed. Nevertheless, these sampling methods still require a few thousand calculations of the underlying finite element model, making reliability analysis computationally expensive for relevant applications.
Coupling a reduced order model (proper generalized decomposition) with an efficient variance reducing sampling algorithm can reduce the computational cost of reliability analysis drastically. In the proposed method, an importance sampling technique is coupled with a reduced structural model by means of PGD to estimate the failure probability. Instead of calculating the design point e.g. with optimization algorithms, the design point is adaptively estimated by using the idea of subset simulation. The failure probability is estimated in an iterative scheme based on adaptively computing the design point and refining the PGD model.
In addition to previously reported results on the accelerated weathering of polystyrene samples (PS) containing 1 wt.% hexabromocyclododecane (HBCD), we present the first results of our investigations of polypropylene samples (PP) containing 0.1 wt.% BDE-209. All studied polymer samples were exposed to a defined weathering schedule in a climate chamber in accordance to regulation EN ISO 4892-3:2006. For the determination of BDE-209 in the collected raining water the samples were prepared in accordance to a validated protocol. Before the analyses each sample was spiked with isotopically labeled BDE-209. Subsequently the samples were extracted with isooctane. The obtained extracts were concentrated, and the resulting solutions were analyzed by GC/MS. Additionally, the total bromine content was monitored for the weathered and untreated samples using laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) and X-ray fluorescence analysis (XRF) as a non-destructive and rapid method. In general, the resulting data from the accelerated weathering will be compared to those from the natural weathering experiments. Here, the surfaces of the test pieces were analyzed by LA-ICP-MS and XRF as well. Moreover, soil bed tests were conducted in a well characterized model soil. This soil (boulder-clay, sand with 12 % loam, particle size in total 0.2-4 mm) is filled in a free-draining basin inside of an air-conditioned cellar. In this manner, TOC, water capacity and humidity are recorded parameters. To induce a leaching process from the samples by the raining water, the target water content is calculated to 8%. The actual humidity is measured by a tensiometer, assuring the duration of the raining period. A defined humidity is a fundamental parameter for a biological activity in the soil. The test polymers were placed up to the half in the soil. Microbial activity of the soil is monitored by a reference polymer (polyurethane) and should induce the release of HBCD and BDE-209 out of the test materials. These released analytes will be captured by passive samplers (silicone tubes) placed in a distinct distance to the polymer samples in the soil. The soil bed experiments are complementary to the weathering experiments due to the biological activity in the soil.

Warfarin, also known as Coumadin is one of the most popular anticoagulant drugs used as a therapeutic in humans to prevent thrombosis, atrial fibrosis, and fibrillation since the 1950s. Because of its ability to hinder blood from clotting by blocking vitamin K-depending carboxylation of blood clotting precursors, it is also used as a rodenticide worldwide. Until today it has been partially substituted by far more potent anticoagulant rodenticides (ARs), so-called superwarfarins. There are numerous references confirming secondary and tertiary poisoning in non-target-animals and wildlife like mammals, birds, and fish associated with ARs. Up to now relatively little is known about persistence and toxicity of transformation products of those AR in the environment and food chain. Herein, warfarin was chosen as a model compound to determine andelucidate TPs which are generated technically and under environment-like conditions. Such as the simulation of the oxidative phase I metabolism by implementing electrochemical flow cells coupled to mass spectrometry (MS). Obtained results are being compared to TPs generated by UVB-irradiation and ozonation.
Analysis and separation of degradation products and TPs were achieved by a variety of gas- and liquid chromatographic techniques coupled to (high resolution) MS.
In this contribution, we present three models to capture singularities in combination with the Spectral Element Method. The first model, the continued-fraction-based Scaled Boundary Finite Element Method, the second model, a new approach based on enrichment with static modes, and the third model, which uses an hp-refinement near the singularity, are compared among each other and evaluated in terms of their respective efficiency and accuracy.

Active IR Thermography

Ternary semiconductors Quantum Dots (t-QD) are Cd-free semiconductors nanocrystals made from I-III-VI group elements like CIS or AIS. They are interesting alternatives for Cd-based QDs for applications as optically active components in solar concentrators or solar cells, light emitting diodes (LED) or in the life sciences. To enhance the PL quantum yield (PL QY) and prevent material deterioration and oxidation, these QDs are commonly surface-passivated with by ZnS shell.
AIS QDs exhibit broad photoluminescence (PL) spectra in the visible and near infrared, which are tunable by size, chemical composition (ratio of components or doping), and surface ligand. They show a relatively high absorption coefficient, high PL QY (up to 70%), and long luminescence lifetimes in the order of a few hundred nanoseconds that make them promising materials for a broad variety of applications. Moreover, they can be simply prepared even in water in high quality which avoids further ligand exchange steps for all water-based applications know to decrease QY.
Here we present a systematic spectroscopic study of differently colored AIS/ZnS QDs synthesized in water, size selected by precipitation, transferred to organic solvents via ligand exchange, and embedded in different polymers by a photochemically initiated polymerization. The PL properties including PL QY and the PL decay kinetics measured in different matrices are shown and discussed.
One of the most important goals in civil engineering is to guaranty the safety of constructions. National standards prescribe a required failure probability in the order of 10−6 (e.g. DIN EN 199:2010-12). The estimation of these failure probabilities is the key point of structural reliability analysis. Generally, it is not possible to compute the failure probability analytically.
Therefore, simulation-based methods as well as methods based on surrogate modeling or response surface methods have been developed. Nevertheless, these methods still require a few thousand evaluations of the structure, usually with finite element (FE) simulations, making reliability analysis computationally expensive for relevant applications.
The aim of this contribution is to increase the efficiency of structural reliability analysis by using the advantages of model reduction techniques. Model reduction is a popular concept to decrease the computational effort of complex numerical simulations while maintaining a reasonable accuracy. Coupling a reduced model with an efficient variance reducing sampling algorithm significantly reduces the computational cost of the reliability analysis without a relevant loss of accuracy.
Ternary semiconductors quantum dots (t-QD) are Cd-free core-only or core-shell semiconductor nanocrystals with a core made from I-III-VI group elements like Cu-In-S (CIS) or Ag-In-S (AIS). Aldehyde-functionalized materials have found broad use in bioconjugation applications. For example, coupling of aldehyde surface groups with to proteins or amine-functionalized oligonucleotides can readily produce biomolecule-covered chip microarray and bead surfaces for multiplex analyses. Additionally, aldehyde-modified nanoparticles can possess bioadhesive properties that can extend their retention time in biological compartments. These emerging novel bioanalytical applications call for reliable tools and methods to detect and quantify accessible aldehyde functionalities.
We present here a straightforward concept to quantify the amount of accessible aldehyde moieties on the surface of polymethylmethacrylate (PMMA) particles through the specific binding and subsequent release of small reporter molecules such as absorbing and fluorescent dyes utilizing hydrazone formation as a reversible covalent labeling strategy. Unbound reporter molecules can be easily removed by washing steps, eliminating inaccuracies caused by unspecific adsorption to hydrophobic surfaces. Cleavage of the hydrazones at acidic pH assisted by a carbonyl trap releases the optical reporters rapidly and quantitatively and allows for their optical detection at low concentration. Importantly, this strategy separates the signal-generating molecules from the bead surface, thereby circumventing light scattering and signal distortions due to binding-induced changes in reporter fluorescence and quenching dye-dye interactions on crowded material surfaces. The potential of this catch-and-release strategy for surface group quantification is representatively demonstrated for a set of microparticles functionalized with different aldehyde densities. This concept is validated by a colorimetric assay with a different optical probe, which contains a reductively cleavable disulfide bond and a reporter that can be quantified photometrically in solution after its cleavage. The excellent match of the results of both optical assays confirms their suitability for the rapid and sensitive quantification of aldehydes on microbead surfaces. These simple catch-and-release assays are excellent tools for process control during bead fabrication and the comparison of different bead batches.

We report on a versatile concept to quantify the accessible aldehyde moieties on particle surfaces through the specific binding and subsequent release of small reporter molecules such as fluorescent dyes and non-fluorescent chromophores utilizing acylhydrazone formation as a reversible covalent labeling strategy. This is representatively demonstrated for a set of polymer microparticles with different aldehyde labeling densities. Importantly, this strategy separates the signal-generating molecule from the bead surface, thereby circumventing common issues associated with light scattering and signal distortions due to binding-induced changes in reporter fluorescence as well as quenching dye-dye interactions on crowded material surfaces. In addition, we demonstrate that the release of a non-fluorescent chromophore via disulfide cleavage and subsequent quantification by absorption spectroscopy gives comparable results, verifying that both assays are capable of rapid and sensitive quantification of aldehydes on microbead surfaces. These strategies enable a quantitative comparison of bead batches with different functionalization densities, and a qualitative prediction of their coupling efficiencies in bioconjugations, as demonstrated in reductive amination reactions with Streptavidin.
A goal of this work is to apply the model, which was initially developed for laser induced plasmas, to plasmas used in chemical reactors, in particular, the inductively-coupled-RF discharge plasma. The model predicts equilibrium chemical compositions of reaction mixtures as functions of plasma temperature and stoichiometry of reactants. The mixtures investigated are BCl3/H2/Ar and BF3/H2/Ar where Ar serves as the plasma-forming gas and H2 as a binding agent which binds the active species Cl and F and Cl- and F-containing intermediates to produce gaseous B and its condensate. An additional goal is to obtain information about intermediate reaction products for different ratios of BCl3/H2 and BF3/H2 and at different temperatures and different Ar flow rates. Also, chemical reactions in laser induced plasmas (LIPs) created on calcium hydrate and calcium carbonate targets in argon are modeled. The results are compared with those obtained by means of the equilibrium model based on the minimization of Gibbs free energy.
Molecule formation in calcium carbonate and calcium hydroxide libs plasmas: model and experiment
(2019)
Analysis of calcium hydrate and calcium carbonate samples and their mixtures is important for archeology, anthropology, and geology. Laser-induced plasma spectroscopy (LIBS) is a suitable tool for such the analysis as it allows for in- and on-line real time chemical assays. LIBS is inherently a technique for atomic analysis; however, since recently, it is also used for molecular analysis. The information attained by the latter is mainly related to “secondary” chemistry that deals with re-association of atoms and ions into molecules at long delay times (≥10 μs) after the initial breakdown. Even though the direct information about the initial molecular content in the target may be lost, the molecular analysis by LIBS can still be useful to assess the composition of samples.
In this work, chemical reactions in laser induced plasmas (LIPs) created on calcium hydrate and calcium carbonate targets in argon are modeled and compared to experiment. The model is based on the assumption that all ionization processes and chemical reactions are at local thermodynamic equilibrium. A chemical composition of argon-calcium-oxygen and argon-calcium-hydrogen plasmas is studied as a function of plasma temperature and pressure. It is established that more than twenty simple and composite molecules and ions can be formed in the course of chemical reactions. The results are compared with those obtained by means of the equilibrium model based on the minimization of Gibbs free energy.
The elemental analysis of seawater is often critical to the understanding of marinechemistry, marine geochemistry, and the deep-sea ecosystems. Laser-induced breakdownspectroscopy (LIBS) with the advantage of rapid multi-elements detection, has a greatpotential for in-situ elemental analysis of seawater. In practice, it is crucial to create acompact, low cost and power saving instrument for the long-term deep-sea observation. Arecently appeared diode-pumped solid-state (DPSS) laser seems to be a promising candidateas it is both compact and robust. Additionally, its high repetition rate up to hundreds of kHzcan provide a considerable throughput for LIBS analysis. However, the DPSS lasers operateat moderate pulse energies, usually less than one mJ, which cannot sustain stablebreakdowns in bulk water. To ensure stable laser-induced plasmas underwater with such aμJ-DPSS laser, we introduced an ultrasound source to assist the breakdown process. Thephase interface and mass flow generated by the near-field ultrasound can greatly reduce thebreakdown threshold and enhance element-specific emissions. Meanwhile, the highrepetition-rate pulses can also improve the breakdown probability and generate uniqueemission lines originated from the water molecule. We further demonstrate that the highrepetition-rate DPSS laser combined with the Echelle spectrometer can provide effectivequantitative analysis for metal elements in bulk water.
Welding residual stress engineering by means of an adjusted martensite phase transformation would be highly attractive as detrimental tensile residual stresses may be prevented already during welding without time and cost intensive post processing. The present study shows a synchrotron diffraction analysis of a martensitic steel subjected to thermo-mechanical load cycles. Experiments were conducted regarding the microstructural strain response during the austenite to martensite transformation. The strains are a function of the temperature and the specific loads applied during cooling. The relation between the transformation plasticity of the material, the amount of martensite formed and the arising strains can thus be assessed. The lattice plane specific strains were compared to experimental findings from (macro) dilatation tests. It is shown that the microscopic material behavior differs remarkably from the one observed on the macroscopic scale, what leads to characteristic residual stresses in the material.
Bacterial biofilms have the capacity to develop and thrive in virtually all circumstances and surfaces, even in the most challenging environmental conditions. The pervasive and recalcitrant existence of biofilms renders them to be a significant safety risk and economical encumbrance in a wide array of industries and technologies, and therefore is a priority area of research.
It is essential to develop an improved understanding of the mechanisms implicated during biofilm formation, such as in the case of the diffusion of bacterial-secreted extracellular electron transporters, which are purported to play an important role during biocorrosion by exoelectrogenic bacteria. Hence, a fundamental understanding of electron transfer mechanisms between bacteria and extracellular electron acceptors will contribute insight to our understanding of charge transport and chemistry at the biofilm – external insoluble electron acceptor interface.
In the present work, Attenuated Total Reflection - Fourier transform-infrared (ATR-FTIR) spectroscopy has been coupled to electrochemical techniques for the nondestructive, in situ spectro-electrochemical monitoring of biofilms in real-time. Shewanella sp. have been selected for this investigation due to their adaptable exoelectrogenic respiratory capacities and their notable ability to reduce metals via several different mechanisms of extracellular electron transfer mechanisms, including self-secreted flavin shuttles. Gold-thin film model substrates have been used due to their inert nature and for their ability to permit precise manipulation of the substrate surface polarization. Additionally, hydrogels comprised of calcium cross-linked alginate have been used to mimic the architectural features of extracellular polymeric substances which are integral to a bacterial biofilm, to allow the study of electron-transporting flavin molecules in an artificial biofilm. The results will demonstrate patterns of diffusion, akin to how flavins would diffuse in a naturally occurring biofilm, and how polarization affects this process. Furthermore, insight will be gained on how the redox behavior of flavins can influence the development and evolution of a biofilm.
This interdisciplinary approach should shed light on bacterial electron transfer mechanisms which could contribute towards emerging technologies which seek to better understand such mechanisms for novel antifouling strategies, renewable energies, and bioremediation.
Residual stresses are crucial when assessing the performance of welded components. The present work deals with the possibilities of transferring the real-life boundary conditions of welding, which influence the residual stress, into the laboratory. The possibilities of a test system with a load capacity of 2 MN specifically developed for online monitoring of stress formation and cracking are shown. Due to the structural design, global process, geometry and material-dependent stresses are induced, which can be quantified in-situ during welding and post weld heat treatment. Examples are presented how the conditions to be found during production are simulated in the laboratory. It is shown how welding residual stresses in high-strength steels are affected by the heat control. Elevated working temperatures significantly increase the tensile residual stresses in the heat affected zone (HAZ). The effect of mechanical stresses resulting from welding on stress relief cracking is demonstrated by the example of a creep resistant steel. Reheat cracks were monitored online during post weld heat treatment.

The main flame retardant modes of action are known, nevertheless in practise the detailed scientific understanding usually falls short, when it comes to modern multicomponent systems, the important tiny optimizations, or quantifying in terms of specific fire properties. The description of the flame retardant modes of action remains usually vague and fragmentary. This talk tries to deliver thought-provoking impulses how the understanding of the fire behaviour and flame retardancy can be utilized to direct the development of future flame retardant polymer products. Some overseen details are picked up as well as rethinking of concepts memorised long ago is encouraged to discover something new. Furthermore, the talk tries to fill the gap between flame retardant modes of action and fire performance constituting a product. This talk promotes the evidence-based development of flame retardant polymers.

The additional element from the filler wire in the laser beam welding is usually distributed inhomogeneously in the final weld due to the high solidification rate of weld pool. It has been found that the electromagnetic stirring produced by an external oscillating magnetic field can enhance the material mixing in the weld pool to achieve a more uniform element distribution. However, the magnetic field has a highly non-linear and multi-coupled interaction with the weld pool behavior, which makes the quantitative explanation of the physical mechanism difficult. In this study, the effect of electromagnetic stirring on the transport phenomena in the wire feed laser beam welding is investigated by a numerical modelling. A 3D transient multi-physical model considering the magnetohydrodynamics, heat transfer, fluid flow, keyhole dynamics and element transport is developed. The multiple reflections and the Fresnel absorption of the laser on the keyhole wall are calculated using the ray tracing method. The numerical results show that a Lorentz force produced by the oscillating magnetic field and its induced eddy current gives significant influence on the transport phenomena in the molten pool. The forward and downward flow is enhanced by the electromagnetic stirring, which homogenizes the distribution of the additional elements from a nickel-based filler wire in a steel weld pool. The numerical results show a good agreement with the high-speed images of the molten pool, the fusion line from the optical micrograph and the element distribution from the energy dispersive X-ray spectroscopy. This work provides a physical base for the electromagnetic-controlled laser beam welding and some guidance for the selection of electromagnetic parameters.
In (aquatischen) Umweltmatrizes (z.B.
Gängige Testorganismen sind u.a. Kieselalgen (Diatomeen).
Rail inspection performed by ultrasonic rail inspection trains is a complex and challenging process. A large number of variables and parameters given by the environment, the track and the testing-system have an influence on the overall performance of the inspection and the inspection result. Typically the parameter vary in a combination depending on the track condition.
To evaluate the individual influence of each relevant variable, simulation tools can be used. Therefore the entire inspection process has to be transferred into a model using combined modelling techniques.
The goal of this work is to model an instrumented ultrasonic test run with a rail inspection train with the parameters varied by a script over a virtually driven distance.
For more than 60 years ultrasonic rail inspection is used as non-destructive testing method to ensure the safe operation of rail tracks. Constantly increasing traffic density and heavy loads have been the motor for the development of new test equipment from handheld devices to rail inspection trains. (Krull 2003)Up to the present most of the system solutions feature conventional ultrasonic transducers housed in wheel-type and slide-type probes. Different tasks have to be carried out during an in-service inspection for flaws in the rail head, rail web and rail foot as well as rolling contact fatigue (Heckel 2018). The more tasks the inspection system has to perform, the more probes are needed. Compared against standard ultrasonic testing methods the application of array probes offers advantages and flexibility by the electronic steering possibilities to control the transmitted and received sound fields. This allows to increase functionality by software while decreasing the number of probes needed in hardware in parallel. One drawback in application of phased array probes is that the repetition frequency of the subsequent measurements will be reduced by the number of virtual probe functions each phased array probe has to perform. This may limit the range of use for phased array probes in high speed applications. To overcome these limits special designs for array probes and signal processing are necessary.

Daraus resultieren unterschiedliche optimale Bearbeitbarkeitsparameter.
Monitoring programs should generate high-quality data on the concentrations of substances and other pollutants in the aquatic environment to enable reliable risk assessment. Furthermore, the need for comparability over space and time is critical for analysis of trends and evaluation of restoration of natural environment. Additionally, research work and exercises at the European level have highlighted that reliable measurements of estrogenic substances at the PNEC level are still challenging to achieve. The project EDC-WFD “Metrology for monitoring endocrine disrupting compounds under the EU Water Framework Directive” aims to develop traceable analytical methods for determining endocrine disrupting compounds and their effects, with a specific focus on three estrogens of the first watch list (17-beta-estradiol (17βE2), 17-alpha-ethinylestradiol (EE2), and estrone (E1)). Estrogens 17-alpha-estradiol (17aE2) and estriol (E3) will be included to demonstrate the reliability of the developed methods - Mass Spectrometry based method and effect-based methods (EBM) - and to support the requirements of Directive 2013/39/EC, Directive 2009/90/EC and Commission Implementation Decision (EU) 2018/840, hence improving the comparability and compatibility of measurement results within Europe. During the EDC-WFD project four EBM will be deeply investigated in order to improve their rationale use and their support in water quality assessment.

Acoustic-structure interaction in the Scaled Boundary Finite Element Method for primsatic geometries
(2019)
Due to the short wavelength compared to the dimensions of the structure, the simulation of ultrasonic waves is still a challenging task. A numerical method well suited for this purpose is the semi-analytical Scaled Boundary Finite Element Method (SBFEM). When applying this method, only the boundary of a computational domain is discretized using finite elements, while the interior is described by an analytical ansatz. Hence, the number of degrees of freedom is reduced significantly compared to the classical Finite Element Method (FEM).
In recent years, a particular formulation of the SBFEM for the simulation of ultrasonic guided waves was developed. The method constitutes an efficient algorithm for prismatic structures of arbitrary length, such as plates, pipes, or beams. Wave propagation phenomena in such structures can be modeled for isotropic and anisotropic inhomogeneous waveguides. Even though the method is an efficient tool for the simulation of guided waves in solid media, a reliable model for the simulation of acoustic wave propagation in fluids as well as acoustic-structure interaction in terms of SBFEM is still missing. In principle, the fluid can be described by a displacement-based formulation and thus be implemented in existing SBFEM algorithms for solid bodies. However, due to the discretization with classical finite elements, spurious modes occur, which cannot be separated from the physical modes straightforwardly. The spurious modes can be suppressed using a penalty parameter. Although very accurate results were achieved for some problems, this procedure has been proven unreliable for certain cases.
For this reason, we propose a different approach in this contribution. We employ a pressure model to simulate the acoustic behavior of fluids. The implementation of the pressure model results in a higher effort due to the necessity of incorporating coupling terms, but it presents a stable alternative without spurious modes. The accuracy of the method is demonstrated in comparison with analytical solutions and results obtained using the FEM.
Non-destructive testing (NDT) is a major contributor to the safe railway operation. Even though NDT reliability in railway maintenance is affected by human factors, there are only just a few studies published in this field so far. Education and training of the NDT personnel are some of the most important drivers of safe and reliable NDT. Continuously improving current practices and tools used for educational purposes can be achieved not only through technical con¬tent, but also through the attention to human factors. The aim of this study was to deepen the understanding of possible human-related risks in the manual ultrasonic inspection of the hollow railway axles and to suggest measures to improve the education and training of the NDT person¬nel. This was achieved by means of Failure Modes and Effects Analysis (FMEA) carried out with eight NDT experts and by a survey of 27 experienced inspectors. The results show that failures can happen throughout the entire NDT process. Prevention of those failures could be improved through the optimization of the organization, technology, documentation and regulations, working conditions and the general process, and through the optimization of the formal education and training. Specialized training of the executives, extended training of the supervisors and the inspectors and improvement of the inspection documentation have been suggested. The study also showed potential for the improvements of the inspection in the field.

Microbiologically induced corrosion due to bacterial biofilms causes several problems in industrial systems, technical applications and in medicine. Prior to the formation of a biofilm on a substrate, planktonic cells attach on the surface. Hence, the properties of the surface play a key role in biofilm formation and are of great importance for the development of strategies to prevent bacterial attachment and biofilm formation.
This project aims at clarifying to which extent surface micro-/nanostructuring and chemical functionalization affects bacterial attachment and whether a synergistic combination of the two can be used to control bacterial adhesion. To answer these questions, model surfaces with regular patterns of 5-10 micrometers in size have been prepared, which provide distinct zones differing in terms of their chemistry or nano-roughness. This was achieved by micro contact printing of self-assembled monolayers with different functional groups and deposition of patterned ZnO nanorod arrays for studying the effect of surface chemistry and morphology, respectively. Typical contrasts studied were combinations of positively/negatively charged, hydrophobic/hydrophilic or flat/rough.
The attachment behavior of bacteria on tailored surfaces were studied in a flow chamber as a function of time. The strain Pseudomonas fluorescens SBW25 was chosen as a model organism. DNA-intercalating dyes such as Syto9 have a high affinity to adsorb on ZnO nanorods. To overcome this limitation a genetic modification was performed by introducing a gene which expresses a green fluorescent protein in P. fluorescens SBW25 enabling the quantitative evaluation of the flow chamber studies by means of fluorescence microscopy. Further analysis of the attachment behavior was performed by means of scanning electron microscopy.
The presentation will summarize the results of our systematic study on the role of individual parameters on bacterial attachment and highlight synergistic combinations, showing an inhibition or enhancing effect. As the investigations with model substrates enable a precise control of the surface parameters, this approach can be applied to different microorganisms and material systems to achieve a correlative description of bacterial adhesion on solid surfaces.
In this study, we developed a new immunoprecipitation method based on protein-coated nanodiamonds. Performing SDS-PAGE and using the SDS-PAGE buffer as an eluent showed that CER could be successfully enriched from human serum. Based on the copper determination with ICP-MS, the amount of bound CER on the nanodiamonds can be calculated. We could show the fulfilled mass balance of bound CER and CER in the supernatant after incubation with a known amount of CER. For isotope ratio analysis this method can be applied to compare ratios of the total copper content in human serum to copper ratios from CER enrichment.

Multiplexed encoding schemes of nano- and micrometer sized particles with fluorescent dyes or quantum dots (QDs) and their optical detection, are of increasing interest for applications in the life sciences, for example in flow cytometry. Almost all strategies utilizing fluorescence focus on spectrally distinguishable emission bands or colors and different intensity levels as fluorescence codes. The fluorescence parameter lifetime has been, however, barely exploited. In this work the goal is to perform multiplexing with encoding fluorophores with different fluorescence lifetimes (LTs). In comparison to the spectral multiplexing strategies this has the advantage, that the different fluorescence LT codes can be measured with the excitation and emission wavelength, thus reducing instrument costs. Moreover, LTs should not depend on emitter concentration. Unlike organic dyes, the LTs of which are typically < 10 ns, the fluorescence LTs of ternary semiconductor QDs that represent a “green” alternative to conventional Cd-containing QDs are in the range of several hundred ns, independent of oxygen concentration, and can be tuned to a certain extent by chemical composition and surface chemistry. This present a time region that can be barely covered by other emitters that have either much shorter or longer lifetimes. In this project, different encoding strategies will be assessed and the encoded particles will be then used for fluorescence assays for the analysis of several targets in parallel. Therefor the encoded particles will be functionalized with different target-specific bioligands and read out with a specifically designed flow cytometer enabling time-resolved fluorescence detection. With this instrument, the particles will be discriminated by their fluorescence LTs In one detection channel while the analytes will be quantified by fluorescence labels in a second channel in the intensity domain.

Since materials play an important role when designing and servicing industrial facilities. Also, for energy systems there is no way around. Selection of appropriate materials is an essential aspect for successful application and reliable service life. Future engineers must be made aware on the importance of this fact. So, they may prevail in techno‐economic discussions.
The book fulfills its intension of providing to students in science and engineering a comprehensive understanding of different energy processes and what role materials play in this conjunction. Also, it gives interested engineers and scientists an insight in this matter. After being introduced the user should dig deeper in the existing detailed knowledge available for materials selection within these special conditions to assure a reliable operation of the energy production facility.
Waste, Evaluation Methods
(2019)
It is difficult to decide which waste management system fulfills best predefined sustainability goals such as maximum materials and energy recovery, least environmental impact, and lowest societal cost. Such decisions are based on many parameters. However, several evaluation methods exist with different scope and output, and the application of one or more evaluation methods provides an objective comparison of alternatives.
Depending on the specific plastic’s ageing sensitivities, the durations which plastic components remain in the environment can be very long. As, in the past, the advantages in ageing resistance and durability were highly evaluated, we now face the problem of quite stable plastic waste within the environment.
However, there is only little knowledge on the real timescales until macroscopic fragmentation for the different kinds of plastic under various environmental conditions.
Here, weathering methods are presented, which have been used for the failure prediction in specific outdoor conditions. Issues of uncertainty, reproducibility, or validation are discussed.
For the prediction of the plastic’s fragmentation, much longer time scales have to be considered. To do this within a limited project life span, various processes as well as their acceleration potential have to be evaluated separately, such as temperature increase or spectral shift.
The possibilities and limitations of such controlled acceleration will be discussed.
Weathering reference materials are used to characterize the harshness of an exposure, aiming on either reproducibility of a specific exposure or on the comparability between various kinds of weathering exposure.
The materials that are used as weathering reference materials differ in their sensitivities (as well as in interactions and interferences of the latter), conditioned by the different processes which lead to the respective property change. It is also essential to take into account the necessary measurement equipment for the respective property change, in order to allow timely intervention.
What are the key issues on choosing a weathering reference material?
What can be learned from the weathering reference materials, investigated so far?
Possibilities and limitations are discussed on the basis of existing weathering reference materials. Conclusions are drawn, for establishing new weathering reference materials.
Fungi that share light-flooded habitats with phototrophs may profit from excess photosynthetic products. But to cope with sunlight-associated stresses it is important for fungi to accurately sense and respond to changes in light. To test the hypothesis that light is an environmental cue that Ascomycota use to coordinate growth, stress responses as well as to establish pathogenic or symbiotic relationships, the photoreceptor (PR) distribution in species from different ecological niches was analysed. The genomes of black fungi from phyllosphere and exposed solid surfaces contain multiple photoreceptors (PRs). The filamentous foliar plant pathogen Botrytis cinerea (Leotiomycetes) has a highly sophisticated photosensory and signalling system that helps to avoid light and to locate susceptible hosts. Rock-inhabiting Dothideo- and Eurotiomycetes including Knufia petricola possess equal numbers of PRs along with the same set of protective pigments. This similarity between black fungi from plant and rock surfaces suggests that photoperception and photoregulation are important for fungi that avoid loss of energy and nutrients through cooperation with phototrophs.

Specific amounts of oxidizing and reductive impurities as well as some moisture were added to dense phase CO2 to replicate CO2 streams from sources in a CCS pipeline network. Due to the moisture content being only 50 ppmV no visible acid condensation took place. To simulate stress conditions at the inside pipeline surface due to fluid pressure (10 MPa) specimens were preloaded using a load frame. Experiments conducted at 278 K and at 313 K revealed the highest corrosion rate at lower temperature. Corrosive effect of impurities was strongest applying mixed atmosphere, containing oxidizing and reductive components, closely followed by CO2 streams with pure oxidizing character. By far, the lowest corrosion rate (10x lower) resulted from reductive atmosphere. In general, at constant temperature and pressure the CO2 stream composition strongly influences the morphology, thickness and composition of the corrosion products. Applying oxidizing or mixed impurities, iron hydroxides or oxides (e.g. goethite, hematite) occur as dominating corrosion products, capable to incorporate different amounts of sulfur. In contrast, using reductive atmosphere very thin corrosion layers with low crystallinity were developed, and phase identification by XRD was unfeasible. SEM/EDX analysis revealed the formation of Fe-O compounds, most likely attributed to the oxygen partial pressure in the system induced by CO2 (≥0.985 volume fraction) and volatile H2O. In addition to the surface covering corrosion layer, secondary phases had grown locally distributed on top of the layer. These compounds are characteristic for the applied atmosphere and vary in number, shape and chemical composition.

Oxidation effects on the functionality and formation of laser-induced periodic surface structures
(2019)
In the last decades, the formation of laser-induced periodic surface structures (LIPSS) has been widely studied, not only to stablish a general understanding of the main mechanisms present during the irradiation with ultrashort laser pulses, but also to identify the main processing parameters for its fabrication on various technical materials. However, an important and almost always present effect that is typically set aside is the material oxidation during or after the laser processing. Usually, this leads to optical, chemical, structural and mechanical alterations that are seldomly studied in detail. Therefore, in this work we present a study that includes the formation of LIPSS on Ti6Al4V alloy for tribological applications were different oxidations layers are involved.
The material characterization using different techniques, such as energy dispersive X-ray analyses (EDX), X-ray photoelectron spectroscopy (XPS), micro Raman spectroscopy (µ-RS), and depth-profiling Auger electron microscopy (AEM), and other techniques, confirms the formation of a non-negligible superficial oxide layer – strongly dependent on the specific processing parameters. We perform tribological experiments on laser irradiated samples oxidized by alternative ways to have a comparison on its friction performance. The obtained results show significant differences that depend directly on the thickness and the quality of the oxidized layer. This may open a fertile research area until now barely explored with clear impact on LIPSS based applications.
In this study an approach for dressing metallic bonded honing stones with hard cutting grains on the basis of electrolysis is investigated. By a combination of concepts from electrochemical machining (ECM) and electropolishing a test rig was designed and put into operation. In general, it can be stated that the Approach investigated in this paper has proved to be a suitable dressing method for honing stones. However, the dressing result is highly dependent on bond components, cutting grain size and concentration which lead to local differences in the material removal and irregular topographies. This could be overcome, for example, by setting the dressing parameters more precisely based on the best results presented in this paper.
In recent years, elemental imaging of biological samples like tissue thin sections using laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) is gaining more and more importance. Improvements concerning spatial resolution as well as signal-to-background ratio due to low-dispersion sample chambers make LA-ICP-MS also interesting for single cell analysis.
To evaluate the interaction of nanoparticles (NPs) with cells LA-ICP-MS was applied for the imaging of individual cells. Our findings show, that NP aggregates can be localized within cellular compartments. The uptake efficiency depends strongly on the physico-chemical properties of the nanostructures (size, chemical composition, surface modification), as well as on the incubation conditions (concentration, time).
Moreover, LA-ICP-MS is increasingly becoming an important complementary technique in bioanalysis by using element-tagging strategies to determine biomolecules indirectly. Based on the specific binding between antibodies and their corresponding antigens, proteins and peptides can be detected in tissue or cells using tagged antibodies. As artificial tags metal chelates loaded with lanthanides, polymer-based elemental tags or metal-containing nanoparticles can be used. Thereby LA-ICP-MS is a sensitive detection tool for multiplexed immuno-histochemistry of tissue and cell samples.
Our results demonstrate the potential of LA-ICP-MS to investigate the distribution of naturally occurring elements, administered agents as well as biomolecules by using metal-tagged antibodies.
Non-linear optical emitters are promising materials for energy applications and biotechnologies. Solid-state multi-band emitters like lanthanide doped up-conversion nanoparticles (UCNPs) show excellent photostability, are excitable in the near infrared (NIR), and show emission bands from the UV to SWIR spectral regions. The optical properties of these materials strongly depend on the excitation power density, i.e., the number of photons absorbed per time interval. The upconversion (ΦUC) and downshifting quantum efficiencies (ΦDS) of these materials, the excitation power dependent population, and the deactivation dynamics are influenced by nanoparticle architecture, doping concentration, and the microenvironment. We studied the fundamental changes of the luminescence properties of ß-NaYF4 UCNPs doped with Yb3+ and Er3+ depending on size, different surroundings such as aqueous and organic media, and different surface chemistries. We obtained further insights into shelling procedures, FRET optimization, influence of doping concentration, and advantages of different sensitizer ions.

In summary, by combining the NIR-emissive [Cr(ddpd)2][BPh4]3 complex CrBPh4 with its extremely large energy gap between the longest wavelength absorption and emission maxima with a pH-responsive fluorescein derivative (FITC) and an inert reference dye like Nile Red (NR) and 5,10,15,20tetrakis(pentafluorophenyl) porphyrin (TFPP), we developed nanosensors for simultaneously sensing temperature, O2 partial pressure, and pH. These novel TOP nanosensors (temperature, oxygen, pH) cover the biologically and physiologically relevant concentration ranges of these parameters/analytes with single wavelength excitation in PBS buffer and in a cell culture medium containing bovine serum albumin (BSA). The response of both nanosensors to all parameters is fully reversible and only minimally affected by the presence of BSA, the most common serum albumin. Moreover, comparative studies with nanosensors containing only a single type of stimuli-responsive molecule and with the respective molecular systems revealed that the different sensor components do not interfere with each other. Future research will include the testing of these nanosensors in cellular uptake studies and, after surface modification with targeted bioligands, eventually in in vivo experiments as previously done by some of us with other polystyrene nanoparticle reporters and nanosensors.46,47 Moreover, this concept of multianalyte sensing will be expanded to nanosensors derived from differently sized premanufactured biocompatible polymer particles and different stimuli-responsive dyes like fluorescent indicators for biologically and bioanalytically relevant metal ions.
This presentation reviews current and expected developments in the field of laser-induced periodic surface structures (LIPSS, ripples). These surface nanostructures are a universal phenomenon and can be generated on almost any material by irradiation with intense linearly polarized radiation. LIPSS are formed in a “self-ordered” way and are often accompanying laser material processing applications. The structures can be produced in a single-step process and enable surface functionalization through the adaption of optical, mechanical and chemical surface properties. Their spatial periods typically range from several micrometers down to less than 100 nanometers, exhibiting a clear correlation with the polarization direction of the laser radiation used. Various types of LIPSS have been classified, relevant control parameters were identified, and material specific formation mechanisms are analyzed for different types of inorganic solids, i.e., metals, semiconductors, and dielectrics, through time-resolved optical experiments and theoretical simulations. Special attention will be paid to a comparison of the currently available formation theories for LIPSS with a discussion of their respective strengths and weaknesses. Currrently explored applications featuring surface functionalization in the fields of optics, surface wetting, medicine, and tribology will be discussed.
Luminescence techniques are amongst the most commonly used analytical methods in the life and material sciences due to their high sensitivity and their nondestructive and multiparametric character. Photoluminescence signals are, however, affected by wavelength-, polarization- and time-dependent instrument specific effects. This hampers the comparability of fluorescence measurements and calls for simple tools for instrument characterization and the quantification of measured fluorescence intensities. Well characterized fluorescence standards for instrument calibration and performance validation (IPV) can be used also to reference fluorescence signals. Of special importance is the reliable and accurate determination of photoluminescence quantum yields (Ф f), that equals the number of emitted per absorbed photons and presents the key performance parameter for emitter efficiency and the comparison of different luminophores. The determination of Ф f is typically done with the aid of so-called quantum yield standards with well-known Ф f values. These standards can also be applied to evaluate integrating sphere setups, which are increasingly being used for absolute measurements of Ф f values. In this respect, division biophotonics of BAM has certificated a set of Ф f standards, which absorb and fluorescence in the wavelength range from 350 to 1100 nm. In the following, the route to Ф f standards with reliable and traceable Ф f values with a complete uncertainty budget will be presented.

Our aim is to develop a new simple and inexpensive method for full field X-ray fluorescence imaging . We combine an energy dispersive array detector with a coded aperture to obtain high resolution images . To obtain the information from the recorded image a reconstruction step is necessary . The reconstruction methods we have developed , were tested on simulated data and then applied to experimental data . The first tests were carried out at the BAM line @ BESSY II. This method enables the simultaneous detection of multiple elements , which is important e.g. in the field of catalysis.
Due to the many possible applications, the uncomplicated production and the high application range, reinforced concrete is a widely used building material. This large range of physical material properties still poses an engineering challenge in determining all necessary requirements for predicting dynamic effects under impact load.
Many aspects of impact have already been examined and some correlations have been studied intensively. Examples are the work of Lastunen and Booker, who studied the influence of projectile properties on the impact form. Li has also investigated the local effects of an impact. The field of detailed damage analysis has not been in focus so far.
This presentation shows some studies in medium-velocity impact with the focus on post-impact damage evaluation. The impactor is modified so that the test plates show low penetration on the top and scabbing on the bottom. With the unique tomography lab test stand at BAM the plate is scanned after the impact and the damage is analyzed. Cracks and scabbing are made visible with a reconstruction. The comparison of simulation and tomography allows to create prognosis models for damage characterization.
