The reassessment of bridges is becoming increasingly important. The basic requirement for analyses of structural safety is reliable knowledge about individual structures. This paper introduces the new approach to evaluate the quality of measured data gained from non-destructive testing (NDT) to provide reliable, objective, and relevant information about existing bridges. The purpose is to relate this validated knowledge to probabilistic analyses. Bridging the gap between NDT and numerical reassessments indicates reduced numerical uncertainties and residual service time extensions. This paper deals with an application of this approach using measurement data collected by ultrasonic technique at a prestressed concrete bridge.
The determination of microscopic dose-damage relations in aqueous environment is of fundamental interest for dosimetry and its application in radiation-therapy and protection. We present a combined experimental and simulational approach to quantify the microscopic energy deposit at biomolecules in liquid environment which is applicable to a wide range of primary radiation sources, e.g. photons, electrons or ions, and targets, such as DNA, proteins or cells. Therefore, we combine Geant4 particle-scattering simulations in water with calculations concerning the movement of biomolecules to obtain the energy deposit in the biologically relevant nanoscopic volume. We present, how to combine these simulational results and experimental data via a generalised damage model to determine the microscopic dose-damage relation at a molecular level.
To show the viability of this approach, we apply this method to an experimentally challenging system, the direct irradiation of plasmid DNA (pUC19) in water with electrons as primary particles. Here we combine electron-scattering simulations with calculations concerning the diffusion and convection induced movement of the DNA, within a coarse-grained model of the irradiated liquid. Additionally a microscopic target model for DNA molecules based on the relation of lineal energy and radiation quality is used to calculate their effective target volume.
It was found that on average fewer than two ionisations within a 7.5\,nm radius around the sugar-phosphate backbone are sufficient to cause a single strand break, with a corresponding median lethal energy deposit being E\textsubscript{1/2}=6+-4\,eV. The presented method is applicable for all types of ionising radiation and a broad variety of biological targets.
High performance polymers of intrinsic microporosity (PIMs) have emerged as novel materials with broad applications from gas separation to electronic devices. Sufficiently rigid, even contorted polymer chains show only limited molecular mobility, therefore undergo inefficient packing and give rise to intrinsic microporosity with pore size generally smaller than 1 nm and BET surface areas larger than 700 m2/g. Further performance optimization and long-term stability of devices incorporating PIMs rely on our understanding of structure-processing-property relationships and physical aging, in which glass transition plays a key role. Up to now no glass transition temperature (Tg) of PIMs could be detected with conventional thermal analysis techniques before degradation. Decoupling the time scales responsible for the glass transition and the thermal decomposition is a reliable strategy to overcome this. This was achieved by employing fast scanning calorimetry (FSC) based on a chip sensor, which is capable to heat and cool a small sample (ng-range) with ultrafast rates of several ten thousand K/s. FSC provides definitive evidence of glass transition of a series of PIMs with a special consideration on the chain rigidity. The determined glass transition temperature of these PIMs follows the order of the rigidity of their backbone structures. FSC provides the first clear-cut experimental evidence of the glass transition of PIM-EA-TB with a Tg of 663 K, PIM-1 of 644 K and PIM-DMDPH-TB of 630 K at a heating rate of 1Χ104 K/s. Local fluctuations are featured in glass transition of highly rigid PIMs. As conformational changes are prevented by the backbone rigidity, the glass transition must rather be assigned to local small scale fluctuations.
Polymeric membranes represent a low-cost, energy efficient solution for gas separation. Recently polymers of intrinsic microporosity (PIMs) have emerged as prestigious membrane materials featuring a large concentration of pores smaller than 1 nm, a BET surface area larger than 700 m2/g and high gas permeability and selectivity. Unusual chain structure combining rigid segments with sites of contortion gives rise to the intrinsic microporosity. However, this novel class of glassy polymers are prone to pronounced physical aging. The initial microporous structures approach a denser state via local small scale fluctuataions, leading to a dramatic reduction in the gas permeabilities. For the first time, dielectric relaxation spectroscopy with state-of-the-art high-resolution analyzers was employed to investigate three representative PIMs with a systematic change in chain rigidity: PIM-EA-TB 〉 PIM-1 〉 PIM-MDPH-TB. The molecular mobility, the charge transport and their response upon heating (aging) in the polymers were measured in a broad temperature range through isothermal frequency scans during different heating / cooling cycles. Multiple dielectric processes following Arrhenius behavior were observed for the investigated polymers. Local fluctuations, Maxwell-Wagner-Sillars (MWS) polarization and structural relaxation phenomena were discussed and attempted to be correlated with the structural features of PIMs. Moreover, all PIMs showed conductivity in the glassy state. The significant increase in the conductivity with increasing temperature far below the glass transition temperature of PIMs is explained in terms of the loosely packed microporous structure and the formation of local intermolecular agglomerates due to interaction of π-electrons in aromatic moieties of the polymer backbone.
An equation for wind vector estimation using a multirotor aircraft as a flying anemometer is shown. To compute the wind vector an estimate of the thrust of the aircraft is required, which is related to the rotational speed of the rotors. Hence, a sensing system for the rotational speed using phototransistors is presented.
In this paper, we present an autonomous aerial robot to reconstruct tomographic 2D slices of gas plumes in outdoor environments. Our platform, the so-called Unmanned Aerial Vehicle for Remote Gas Sensing (UAV-REGAS) combines a lightweight Tunable Diode Laser Absorption Spectroscopy (TDLAS) sensor with a 3-axis aerial stabilization gimbal for aiming on a versatile octocopter. The TDLAS sensor provides integral gas concentration measurements but no information regarding the distance traveled by the laser diode's beam or the distribution of the gas along the optical path. We complemented the set-up with a laser rangefinder and apply principles of Computed Tomography (CT) to create a model of the spatial gas distribution from these integral concentration measurements. To allow for a rudimentary ground truth evaluation of the applied gas tomography algorithm, we set up a unique outdoor test environment based on two 3D ultrasonic anemometers and a distributed array of 10 infrared gas transmitters. We present first results showing the 2D plume reconstruction capabilities of the system under realistic conditions.
In industrial environments, airborne by-products such as dust and (toxic) gases, constitute a major risk for the worker’s health. Major changes in automated processes in the industry lead to an increasing demand for solutions in air quality management. Thus, occupational health experts are highly interested in precise dust and gas distribution models for working environments. For practical and economic reasons, high-quality, costly measurements are often available for short time-intervals only. Therefore, current monitoring procedures are carried out sparsely, both in time and space, i.e., measurement data are collected in single day campaigns at selected locations only. Real-time knowledge of contaminant distributions inside the working environment would also provide means for better and more economic control of air impurities. For example, the possibility to regulate the workspace’s ventilation exhaust locations can reduce the concentration of airborne contaminants by 50%.
To improve the occupational health and safety of (industrial) workplaces, this work aims for developing a swarm of gas-sensitive aerial nano robots for monitoring indoor air quality and for localizing potential emission sources.
Luminescence techniques are amongst the most commonly used analytical methods in life and material sciences due to their sensitivity and their nondestructive and multiparametric character. Photoluminescence signals are, however, affected by wavelength, polarization and time dependent instrument specific effects, and provide only relative intensities. This hampers the comparability of fluorescence measure-ments and calls for simple tools for instrument characterization and the quantification of measured fluorescence intensities. Well characterized fluorescence standards for instrument calibration and performance validation (IPV) can be used as references for fluorescence signals. Of special importance is the correct determination of photoluminescence quantum yields (QF) (number of emitted per absorbed photons) that provides a direct comparison of the fluorescence efficiency of emitters. Such well characterized standards have been successfully developed by BAM for the relative determination of f values of transparent solutions of molecular and nanoscale emitters in the wavelength range from 350 and 1100 nm and will be soon certified. These standards can also be used to evaluate integrating sphere setups, which are increasingly being used for absolute measurements of QF values.
Digital holographic cytometry (DHC) is a state-of-the-art quantitative Phase imaging (QPI) method that permits time-lapse imaging of cells without induced cellular toxicity. DHC platforms equipped with semi-automated image segmentation and analysis software packages for assessing cell behavior are commercially available. In this study we investigate the possible uptake of nanoprobes in macrophages in vitro over time.
The European building sector is moving towards more complex and high-tech building approaches. While focusing on energy efficiency, aspects e.g. occupant health, sustainability and life cycle costing are often neglected. This study highlights the potential of earthen plasters in combination with natural ventilation for low-tech solutions.
The EU funded project [H]house established the outstanding performance of earthen materials in light of hygrothermal and air purifying properties, which were further supported by experimental data from monitoring of naturally ventilated pilot buildings in Berlin. Additionally, [H]house demonstrated through LCC an increased cost efficiency of earth based low-tech solutions in comparison to conventional constructions relying on mechanical ventilation.
New Focus On Boehmite-Reinforced Nanocomposites Molecular Approach With Advanced FTIR-Techniques
(2019)
By FTIR-study it was possible to proof a chemical reaction between boehmite and the hardener of anhydride cured epoxy resins. Future studies can assume that the chemical environment of the resin system is changed in the surrounding of boehmite nanoparticles. This highly affects especially localized properties.
Fibreoptic sensors (FOS) represent sensing technology with small footprint, low invasiveness, electromagnetic passivity and immunity, plus potential for remote and real-time monitoring. Modern FOS techniques allow truly temporally- and spatially-continuous monitoring over extended distances; a feature not attainable with any other sensing technology. Moreover, depending on their particular material composition and design, optical fibres can be made resistant to high temperatures, chemicals and ionizing radiation. Due to this unique combination of advantageous properties, ever since their emergence, FOS have been attracting considerable attention for monitoring tasks in harsh, hazardous and difficult-to-access locations. The potential of FOS has been recognized also in the field of radioactive waste management and fibreoptic sensors belong to the most promising technologies for nuclear waste repositories (NWR) monitoring.
Vast majority of distributed fibreoptic sensor applications rely on use of silica-based optical fibres as sensing elements. At the same time, distributed measurement of local temperature and strain along the fibre are the most common monitoring tasks addressed by fibreoptic sensors. Nevertheless, FOS offer much larger flexibility both in terms of utilized sensing fibre as well as targeted measurand. In this contribution, we will review some of more alternative implementations of FOS that are being explored at “Fibre Optic Sensors” division of Federal Institute for Material Research and Testing (BAM), in Berlin. The main focus will be twofold. On one side, we will address FOS applications with polymer optical fibres (POF), that may enable monitoring of large strains (>100%) and high-sensitivity radiation detection. On the other side, we will present our activities in the area of distributed acoustic sensing (DAS); one of the most recent developments in the fibreoptic sensing field enabling highly-dynamic vibration sensing with nanostrain sensitivity. We will introduce the principles of the addressed FOS technologies, present application examples from our case studies, discuss advantages and limitations of the techniques and highlight their potential for NWR monitoring.
Ultrashort laser pulse micromachining features a high precision. By increasing the repetition rate of the applied laser to several 100 kHz, laser processing becomes quick and cost-effective and make this method attractive for industrial applications. Upon exceeding a critical laser intensity, hard X-ray radiation is generated as a side effect. Even if the emitted X-ray dose per pulse is low, the accumulated X-ray dose becomes significant for high-repetition-rate laser systems so that radiation safety must be considered.

The closure of underground nuclear waste disposal facilities requires reliable gas- and water-tight engineered barriers. In Germany, barriers made from salt concrete have been evaluated in full scale. While the barriers seem to fulfill the requirements regarding permeability, some unexpected cracks have been detected at the surface and at depth. In cooperation between the Federal Company for Radioactive Waste Disposal (BGE) and the Federal Institute for Materials Research and Testing (BAM), several experiments have been carried out to evaluate the applicability for ultrasonic measurements in crack detection and general quality assurance. Both commercial instruments and specially developed devices have been tested on site.
Using commercial ultrasonic echo devices designed for concrete inspection it was possible to detect cracks and objects in salt concrete up to a depth of 2 m. The check for delamination in shotcrete is another field of application. A unique device available at BAM, the wide aperture, deep penetration instrument LAUS, was able to locate cracks and objects up to a depth of 8 m so far, which is thought to be a record for ultrasonic echo measurements in concrete. Adapted imaging procedures, partly adopted from geophysics, helped to reveal 3D structure at depth.
In addition, we have developed ultrasonic probes to be deployed in boreholes, currently at up to 20 m depth. They can collect information on cracks and other features in a radius of about 1.5 m around the borehole in the current version and might be used in echo or transmission mode. Evaluation experiments have been performed at an experimental barrier at the ERAM site in Morsleben, Germany. The results showed several empty and injected cracks as well as built-in instrumentation. The results have been verified using borehole endoscopy as well as core examination and will be used to set up a reliable quality assurance system for engineered barriers. All instruments are based on ultrasonic shear wave transducers with a frequency range between 25 kHz and 100 kHz. Current research focuses on the improvement of the hardware (e. g. optimization of array characteristics) and imaging techniques such as Reverse Time Migration, both aiming at the improvement of depth of penetration, resolution and probability of detection.
Evolution of a friction material with braking sequence: properties related to the microstructure
(2019)
The microstructural complexity (multi-component and multi-scale) of friction materials, such as those used in braking applications, usually limits the characterization to the pristine state, even when they are to be submitted to high loading brake sequences. The understanding of properties ageing as a function of load history and microstructure is an important step to gain further knowledge on the tribological mechanisms occurring at the contact surface, as well as on the braking performances. To do so, the bulk mechanical and microscopical properties of a semi-metallic friction material are investigated before and after a braking sequence. Uniaxial compression tests combined with DIC are used to study the axial evolution of the mechanical behaviour (i.e., from the contact surface to the backplate). A significant mechanical evolution is observed in the after-braking material, which develops a layered behaviour along the depth. The layers (three) exhibit distinct mechanical/microstructural features that can be related to the braking thermal gradients and the occurrence of local damage.
The proposed BAM project SealWasteSafe will advance the state of the art for the construction and monitoring of safe sealing systems for underground repositories of radioactive or toxic waste. During this project, a novel salt concrete exhibiting neither significant cracking nor shrinkage will be optimized for use in the sealing systems. The composition of this material will be based on alkali-activated materials, which are characterized by particularly small thermal deformations during the hardening reaction. Quality assurance and continuous monitoring systems developed during this project will be demonstrated not only for high reliability, but also for resistance to highly alkaline environments and to water intrusion along cables or at sensor locations. A variety of sensors will be used in combination with wireless Radio Frequency Identification (RFID) technology to record moisture, temperature, and, if necessary, corrosion activity within the sealing system. Distributed Fibre Optic Sensor (FOS) technology will also be used for strain, temperature, and moisture content measurement. Ultrasound-based measuring methods will be utilized for the detection of cracks and delaminations. Additionally, digital image correlation and acoustic emission analysis will be used for deformation measurements and crack detection. A novel borehole probe and advanced ultrasound imaging techniques will be further developed to track cracks and delaminations within the host rock in 3D. The surface-based Large Aperture Ultrasound System (LAUS) will also be utilized to detect cracks and delaminations deep below the exterior surface of the sealing system. Although the focus of this project will be on the host rock salt, the resulting technologies will be intentionally developed in a way that facilitates their adaptation to other host rocks.
In this work, the structural integrity of LBM fabricated IN625 small cylinders (d = 1 mm, h = 6 mm) was investigated regarding the porosity and the surface roughness by means of computed tomography. The measurements were carried out on a GE v|tome|x L 300/180 with a reconstructed voxel size of 2 µm. The pores were analyzed for size, shape and spatial distribution. The correlation between compactness C and spatial distribution showed that elongated pores (C < 0.2) appear exclusively within a distance of 80 µm to the sample surface. The reconstructed surface was digitally meshed and unwrapped to evaluate the mean roughness Ra. Since the gravity correlates linearly with the sine of the build angle, the influence of gravity on porosity and surface roughness was determined.

Nanocomposites are extremely versatile due to their physicochemical properties, which differ significantly from bulk homopolymers. One of the inorganic nanomaterials which are increasingly used as a filler in polymer matrices is boehmite, typically used as an inexpensive flame retardant. Here, it is used as a nanofiller in polycarbonate and polyamide, expecting to improve their mechanical properties. For industrial use boehmite is obtained by the solvothermal method, resulting in a layered nanomaterial, whereas naturally it occurs as single crystals with the size of <100µm. In this work we are obtaining and isolating boehmite crystals by a bottom-up method, in which a reaction between aluminum nitride and sodium hydroxide. Obtaining boehmite as microcrystals is necessary for its analysis and characterization, as well as to investigate its interaction with polymer matrices at the polymer/particle interface. Here, the obtained particles in polymer matrices are characterized with differential scanning calorimetry and thermogravimetry analysis.

In well-annealed thin polymer films, with non-repulsive polymer/substrate interactions, an irreversibly adsorbed layer is formed. These adsorbed layers have shown enormous potential for technological applications. The growth kinetics and molecular dynamics of these buried layers in thin films are still not fully investigated due to the hard accessibility. Here, the irreversibly adsorbed layers of homopolymer thin films are revealed by solvent-leaching experiments. The growth kinetics of these layers is investigated as a function of original film thickness and annealing times. The thickness, topography and quality of the adsorbed layer is determined with Atomic Force Microscopy (AFM) and spectroscopic ellipsometry. Additionally, the molecular mobility of the adsorbed layer is investigated with Broadband Dielectric Spectroscopy (BDS). A recently developed nanostructured capacitor (NSC) is employed to measure the adsorbed layers with a free surface layer depending on annealing and solvent-leaching time. The results are quantitatively compared and discussed with respect to recently published work.

Over the past decades research on the molecular dynamics of miscible polymer blends is of topical interest in the literature, to understand the segmental mobility of individual components, as it is affected by blending. In general, miscible polymer blends exhibit a complex dynamic behavior. For an A/B blend the relaxation times of component A and component B are affected by the spatial local compositional heterogeneity, present in binary systems on a microscopic level, regardless of the macroscopic homogeneity. Here, a combination of broadband dielectric and specific heat spectroscopy was employed to study the dynamically asymmetric PVME/PS blend with seven different compositions, focusing on samples with high PS contents. Considering that PS is dielectrically invisible, BDS is a powerful technique to study the response of PVME, as it is affected by PS segments. Here, three separate relaxation processes were found by dielectric investigations, related to confined or constrained PVME segments due to the spatial local compositional heterogeneities, which is in contrary to the previous literature findings [1]. Moreover, the dielectric data was compared with results obtained by specific heat spectroscopy, where a fourth relaxation process was found, due to the cooperative fluctuations of PVME and PS.

For inorganic/polymer nanocomposites a so-called Rigid Amorphous Phase (RAF) is formed in the interfacial region by adsorption of polymer segments onto the nanoparticles. The segmental dynamics of RAF is expected to be altered, as compared to the pure matrix, which might percolate to the entire system, affecting the overall nanocomposite properties. A combination of two relaxation spectroscopy techniques (Broadband Dielectric Spectroscopy (BDS) and Temperature Modulated DSC (TMDSC)) as well as Fast Scanning Calorimetry (FSC) was employed to investigate the structure and molecular mobility of nanocomposites based on Epoxy and Layered Doubled Hydroxides with different nanoparticle content. First, BDS investigations proved the existence of a process, which is present only for nanocomposites, assigned to the dynamics of polymer segments within RAF. Second, the amount of RAF was quantified by analyzing the change of specific heat capacity step of nanocomposites, comparing to the pure material. Thirdly, the glass transition of nanocomposites was studied with FSC, applying high heating rates (0.5-10 kK/s). Considering that all techniques probe essentially the same molecular process, an activation plot was constructed, delivering a complete picture of the molecular mobility and structure of the polymer nanocomposites including RAF.
The Spectral Element Method (SEM) has been proven to be an efficient numerical method for solving the wave equation in linear elastic bodies. This efficiency is reduced if a stress singularity is present in the body. For example, re-entrant corners, material interfaces, fixed boundaries, and especially crack tips can cause stress singularities. To preserve the efficiency of the SEM, special solution strategies are required. There are many approaches which consider stress singularities, but comparisons are rare for dynamic problems. Finding an efficient model is an important step for many applications. In particular, applications for structural health monitoring and non-destructive evaluation rely on accurate and efficient crack models.
In this contribution, we present several models to capture singularities in combination with the SEM. The theory behind the models are shortly summarized and we show results for different benchmark problems in two dimensions. The first model deploys a new class of singular elements. For these singular elements, the crack tip approximation is computed based on the static Scaled Boundary Finite Element Method (SBFEM). The second model uses the continued‐fraction‐based SBFEM, while the third model uses an hp-refinement near the singularity. The models are compared among each other and evaluated in terms of their respective efficiency and accuracy.
Laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) is nowadays an established multi-elemental analysis and mapping technique. It was shown that LA-ICP-MS can visualize the elemental distribution within tissue thin sections or cell samples. Quantification is possible by using appropriate matrix-matched calibration samples. Besides naturally occurring elements and metals from contrast agents, biomolecules using metal-tagged antibodies were detected in different bio-medical samples. By combining the results with findings from histology, magnetic resonance imaging (MRI) and other techniques disease related changes like alterations of the extracellular matrix can be investigated.
The successful off-line coupling of asymmetrical flow field flow fractionation (AF4) and capillary electrophoresis (CE) for Separation of nanoparticles (NPs) with different surface coatings was shown.
Two mixtures of polystyrene nanoparticles (PS-NPs) with comparable core sizes (20 nm and 50 nm) but different coatings (no coating/carboxyl-coated) were studied. Separation in either method resulted in non-baseline resolved or non-separated peaks. In contrast, two-dimensional off-line coupling of AF4 and CE resulted in clearly separated regions in their 2 D plots and can obviouly improve separation resolution.
An affinity column, which removed up to 99 % of high affinity Anti-TNT antibody at high antibody concentrations, was manufactured based on a monolithic glass core and a trinitroaniline-BSA conjugate. To detect the label Dy654 in the nM range an epi-fluorescence microscope setup with a CMOS camera was established to serve as online fluorescence detector with multiplexing capabilities.
Screening of one-bead-one-peptide libraries is a powerful analytical tool for the identification of protein ligands. However, the traditional peptide screening procedure involves tedious steps such as manual selection, sequencing, and characterization. We present a high-throughput “all in one chip” system, allowing the screening of a high number of resin beads in short time. Here, beads of a combinatorial one-bead one compound peptide library are immobilized on an in-house produced chip, on which every bead has a well-defined position. The chip is then incubated with a fluorophore-labeled protein, identifying suitable peptides by a high-resolution fluorescence scan. The screening is followed by MALDI-MS experiments directly on the respective glass chip. To circumvent the need for peptide fragmentation normally used for peptide de novo sequencing, which can result in incomplete sequence information, an approach based on ladder sequencing has been used. This allows the peptide sequence identification by fragmentation-free MS with almost 100 % accuracy. For this purpose, a software tool was developed automatically translating MALDI-MS spectra into the corresponding peptide sequences.
We compare four different sensing solutions suitable for distributed fiber optic humidity sensing in per uorinated graded-index polymer optical fibers (PFGI-POFs). Compared to silica fbers, polymer optical fibers over advantageous beneffits including signifficantly higher break down strain, fracture toughness and humidity sensitivity. Various humidity-related effects in PFGI-POFs have been reported in the last years including measured attenuation and length changes as well as Brillouin frequency and Bragg wavelength shifts. The four aforementioned methods could serve as a basis for distributed and quasi-distributed humidity sensing and are described here closely with an emphasis on plausible cross effects to temperature and strain. The main focus of this paper lies on the comparison of four approaches with regard to method complexity, sensitivity to humidity, spatial resolution, real-time capability and effort to compensate for cross effects.
Microbiologically influenced corrosion (MIC) of iron is usually attributed to sulfate-reducing microorganisms (SRM) in offshore industries, such as the oil and gas pipelines, due to the high concentrations of sulfate in the seawater. SRM act upon the metal by the reactiveness of hydrogen sulfide (HS-), and by withdrawal of the available electrons (Fe --> Fe2+ + 2e-; E° = -0.47 V) in electrical contact with the metal (EMIC). However, methanogenic archaea can also cause MIC. Because they do not produce HS-, withdrawal of electrons may be their main corrosive mechanism; however, mechanistic details and kinetics of the overall process are poorly understood.
To investigate the corrosion potential of methanogens, we studied the EMIC methanogenic strains isolated from marine sediments (Methanobacterium-affiliated strain IM1) and crude oil tanks (Methanococcus maripaludis Mic1c10), in an in-house developed flow-through cell to simulate a fluctuating environment. A co-culture of M. maripaludis and D. alaskensis was also established to study the effect of syntrophic growth on metal corrosion that may occur in industrial pipelines. Results indicate that the rates of iron corrosion due to coupled methanogenesis (up to 0.4 mm/yr) are higher to that caused by the marine SRM Desulfovibrio alaskensis (0.15 mm/yr). Surface analyses of the metal showed severe pitting with high methane production. Genomic analysis of the EMIC methanogen M. maripaludis Mic1c10 will provide an insight on the mechanisms of MIC. Such knowledge and deeper understanding also from an electrokinetic point of view may not only provide further models in microbial electrophysiology, but also contribute to mitigation strategies in MIC.
Digital Analytical Sciences
(2019)
Based on inductive reasoning scientific inquiry in Analytical Sciences has a long tradition. The talk addresses in which way the digital transformation will change the scientific paradigm in Analytical Sciences to a data-driven discipline. Three main areas, integration, instruments, and methods, are discussed in their transformation through machine learning and other digital tools. Finally, some shortcomings and pitfalls in the deployments of algorithmic shortcuts are illustrated.
Permitted explosives have a tradition of many decades in Germany and had been designed to the needs of the underground coal mining in the Ruhr. In neighboring countries of Germany similar provisions were made to prevent hazards from fire damp and coal dust explosions in underground coal mines. Due to the termination of coal mining in Germany and most other European countries, also the testing facilities are disappearing. This presentation outlines the current situation of testing regimes and facilities.
During a European convention of the authorities for market surveillance (AdCO) for explosives the activities of the notified bodies are presented, putting a focus on matters concerning market surveillance.
The presentation outlined in detail, which verifications are possible
with explosives, while practical tests involving initiation of
explosives are excluded. Elements of labeling and the conclusions in
view of quality control were explained, and typical failures by
manufacturers to comply were orally described.
Modular plants using intensified continuous processes represent an appealing concept to produce pharmaceuticals. It can improve quality, safety, sustainability, and profitability compared to batch processes, and it enables plug-and-produce reconfiguration for fast product changes. To facilitate this flexibility by real-time quality control, we developed a solution that can be adapted quickly to new processes and includes a compact Nuclear Magnetic Resonance (NMR) spectrometer for online quality monitoring as well as a new model-based control approach. The NMR sensor is a benchtop device enhanced to the requirements of automated chemical production including ro-bust evaluation of sensor data.
Here, we present alternatives for the quantitative determination of the analytes using modular, physically motivated models. These models can be adapted to new substances solely by the use of their corresponding pure component spectra, which can either be derived from experimental spectra as well as from quantum mechanical models or NMR predictors. Modular means that spec-tral models can simply be exchanged together with alternate reagents and products. Beyond that, we comprehensively calibrated an NIR spectrometer based on online NMR process data for the first time within an industrial plant. The integrated solution was developed for a metal organic reac-tion running on a commercial-scale modular pilot plant and it was tested under industrial conditions.
In the past, δ26/24Mg measurements were referenced to NIST SRM 980, the initial zero of the δ26/24Mg scale. With the development of MC-ICPMS, the detection of small but measurable isotopic differences in different chips of SRM 980 became apparent. To solve this problem a suite of magnesium isotope reference materials, ERM-AE143, -AE144 and -AE145, has been certified in a first study by applying an ab initio calibration for absolute Mg isotope ratios without any a priori assumptions, a procedure which fulfils all requirements of a primary method of measurement. We could achieve for the first time measurement uncertainties for isotope amount ratios close to the typical precision of magnesium delta values, δ26/24Mg, which are at the 0.1 ‰ level (2SD). In addition, it was demonstrated that commonly used fractionation laws are invalid for correcting Mg isotope ratios in multi-collector ICPMS as they result in a bias which is not covered by its associated uncertainty. Depending on their type, fractionation laws create a bias up to several per mil, with the exponential law showing the smallest bias between 0.1 ‰ to 0.7 ‰.
With these isotope reference materials, it is possible to establish SI-traceability for magnesium delta measurements. To realize this, we organized a second study within which five expert laboratories participated to cross-calibrate all available magnesium isotope standards, which are NIST SRM 980, IRMM-009, ERM-AE143, ERM-AE144, ERM-AE145 and the standards DSM3 and Cambridge-1. The mean δ26/24Mg values for the individual iRMs, calculated from the laboratory means show 2 SD reproducibilities varying between 0.025 and 0.093 ‰. Propagated measurement uncertainties suggest a standard uncertainty of about 0.1‰ for δ26/24Mg determinations (2SD). Thus, SI traceability for magnesium isotope amount ratios and delta values is demonstrated to be established.
Si isotope fractionation during BIF formation – inferences from a modern Archean ocean analogue
(2019)
Silica-rich sedimentary rocks like cherts and BIFS, typical for the Archean, have been used to reconstruct temperatures and other properties of the early oceans through the study of their Si isotope variations. Precambrian cherts and BIFS span a δ30Si range of ~7‰, with BIFs being about 2‰ lower in δ30Si than cherts. These lower δ30Si signatures have been attributed to represent contributions from different input sources such as hydrothermal fluids, variable continental weathering regimes or sorption onto Fe oxides/hydroxides [e.g. 2 and references therein]. In this study, fluids and BIF-like sediments have been investigated for their Si isotope compositions in Paulina Lake (PL), a hydrothermally-influenced crater lake in the Newberry Caldera, Oregon, USA. PL lake sediments are rich in silica (~65wt% SiO2) and are composed of up to 22.5wt% Fe2O3, which is comparable to Archean BIFs and thus serve as a modern Archean ocean analogue. We compared our analyses with East Lake (EL), the twin Newberry crater lake without hydrothermal input. Dissolved Si in EL has an average δ30Si signature of +1.55±0.16‰ (1sd) and sediments an average δ30Si signature of +0.18±0.28‰ (1sd). Dissolved Si in PL has an average δ30Si signature of +2.02±0.15‰ (1sd), whereas the sediments show a large range in δ30Si values between +0.59‰ and -1.24‰. PL sediments show a trend towards more negative δ30Si with increasing Fe2O3 contents. The magnitude of Si isotope fractionation thus appears to depend on the presence of Fe. This fractionation induced by interaction with Fe precipitation is defined here as the offset in δ30Si between PL and EL sediments at comparable depths (Δ30SiPL-EL). The resulting Δ30SiPL-EL values range between +0.69 and -1.42‰ and increase with increasing Fe2O3 content in the sediments. Our results are the first to quantify the magnitude of Fe-induced δ30Si fractionation observed in a natural analogue of the Archean ocean and can explain the lighter δ30Si signatures found in BIFs.
Time-resolved flow cytometry
(2019)
The fast identification of a large number of analytes or events is increasingly required in bioanalytical, diagnostic, and security applications. The versatility and straightforward use make multiparametric fluorescence techniques particularly interesting as detection techniques. An established method for high-throughput single-cell and single-particle measurements is flow cytometry (FCM). Using only spectral encoding without further intensity information, state-of-the-art instruments equipped with several light sources and detectors can resolve almost 20 different color codes. However, this is not sufficient to answer complex research questions, e.g. in cell biology and immunology. In contrast, routine applications demand low-cost and sometimes even portable instruments and thus a minimum number of instrument components. Thus, there are currently two main research directions in FCM: the development of methods that can either address increasingly complex analytical challenges or provide low-cost and robust approaches for routine multiplex analyses. Common spectral multiplexing approaches face limitations in both directions. On the one hand, spectral overlap of labels restricts the number of codes and makes elaborate correction schemes necessary. On the other hand, even for lower degrees of multiplexing often a sophisticated optical setup is needed. An alternative to spectral multiplexing and intensity encoding is to exploit the luminescence lifetime (LT) as an encoding parameter. This can allow for extending the parameter space in combination with spectral encoding or result in more simple and compact devices due to fewer optical components. The availability of fast electronics enables miniaturized and portable lifetime measurement setups at relatively low cost. LT-FCM requires to master LT determination with a limited number of detected photons due to the short interaction time of the encoded objects with the laser spot. In this study, we address this issue for time-domain cytometry and present a novel lifetime flow cytometry (LT-FCM) platform based on a compact setup and straightforward time-domain measurements utilizing LT-encoded luminescent beads. Moreover, we present the realization of a first bioanalytical assay with LT-encoded beads.
Mechanochemistry paves the way to simple, fast, and green syntheses. Despite considerable effort, there remains a lack in understanding of the underlying mechanisms. In situ investigations help to understand these mechanisms, which occur during a mechanochemical reaction. Here we present a universal strategy for simultaneous real-time in situ analysis, combining X-ray diffraction, Raman spectroscopy, and thermography. The potential of our approach is shown for different model reactions.
While today, engineers can choose from a wide range of rheology modifying admixtures, in some parts of the world, these are difficult to access, due to their complex processing. However, alternatives can be bio-based polymers such as polysaccharides from various sources. These are easily accessible all over the world, do not demand for complicated processing, and typically they are more sustainable than many established materials, which are crude oil-based.
The paper presents the effects of acacia gum, cassava starch and the gum of triumfetta pendrata A. Rich on the rheological performance of cementitious systems. It is shown that acacia gum can be as efficient as polycarboxylate based superplasticisers, cassava starch can reduce the yield stress slightly with little effect on the plastic viscosity, and the gum of triumfetta pendrata A. Rich increases the thixotropy of cement pastes with plasticizing polymers significantly.
Two nanosensors for simultaneous optical measurements of temperature (“T”), oxygen (“O”), and pH (“P”) have been designed. These “TOP” nanosensors are based on 100 nm-sized silica-coated polystyrene nanoparticles (PS-NPs) doped with the near infrared emissive oxygen- and temperature-sensitive chromium(III) complex ([Cr(ddpd)2][BPh4]3 CrBPh4)[1][2] and an inert reference fluorescence dye (Nile Red NR or 5,10,15,20-tetrakis-(pentafluorophenyl) porphyrin TFPP) and are covalently labeled with the pH-sensitive fluorophore fluorescein isothiocyanate (FITC). These emitters can be excited at the same wavelength and reveal distinguishable emission spectra suitable for ratiometric intensity-based and time-resolved studies in the visible and near infrared spectral region. The core-shell nanostructure of these sensors reveals high colloidal stability in various aqueous media. Studies in PBS buffer solutions and in a model body liquid demonstrate the applicability of the TOP nanosensors for optically detecting the three bioanalytically and biologically relevant analytes temperature, oxygen and pH simultaneously at the same position.

The necessity and demand for nondestructive testing of wood-based materials which can automatically scan huge areas of wood is increasing. Air-coupled ultrasound (ACU) is used to detect defects and damage without altering the structure permanently. Using through transmission it is possible to detect even small holes and missing adhesive. If only one side of an object is accessible the reflection mode is preferred at the expense of a reduced resolution and penetration depth. Novel ferroelectret transducers with a high signal-to-noise ratio (SNR) enable a high-precision structure recognition. The transducers made of cellular polypropylene (PP) are quite suitable for ACU testing due to their extremely low Young’s modulus and low density which result in a favorable acoustic impedance for the transmission of ultrasonic waves between the transducer and air. Thus, defects such as delamination, rot, and cracks can be detected. Promising results were obtained under laboratory conditions with frequencies from 90 kHz to 200 kHz. The advantage of these ACU transducers is that they do not require contact to the sample, are accurate, and cost effective. Ultrasonic quality assurance for Wood is an important attempt to increase the acceptance of wooden structures and towards sustainability in civil engineering in general.
In earthquake-prone regions such as the Pacific Northwest, damage assessment tools are needed to enable safety evaluations to support recovery. Currently, damage assessment is performed primarily by visual inspection and is often impossible for structural members that are inaccessible, such as deep foundations or interior members hidden by cladding. This study explores the possibility of using embedded ultrasonic transducers to monitor reinforced concrete members for damage progression under earthquake loading. A novel methodology is proposed where changes in the member condition due to an increase in the earthquake-type loading of a full-scale column-foundation specimen are correlated with changes in the recorded ultrasonic waveforms. The discussed preliminary analysis of the ultrasonic signals is based on wave propagation velocity, changes in the coda wave portion, and maximum amplitude of the signals. Three embedded transducers were used to continuously monitor the laboratory specimen during destructive testing. This paper provides an overview of the proposed methodology, outlines the laboratory experiment, and discusses some preliminary observations.
Ultrasonic transmission measurements are used to monitor concrete elements mostly on a laboratory scale since decades. Recently, coda wave interferometry, a technique adapted from seismology, has been introduced to civil engineering experiments. It can be used to reveal subtle changes in concrete samples and even large construction elements without having a transducer directly at the location where the change is taking place. The methodology works best with embedded transducers to avoid coupling issues or excessive environmental influence. These transducers can be used for newly built and existing structures. Recently, large concrete beams have been equipped with a network of transducers and loaded until failure. Using code wave interferometry, it was possible to visualize stress fields and damaged areas. This paper gives an overview of the state of the art, recent results achieved at BAM and a task list for further results and development.

Spent fuel storage periods well beyond those originally foreseen are a reality. This publication offers several ideas and approaches that may be considered to address the increasingly longer storage times. The aim is to raise awareness, encourage dialogue and provide ideas on how to manage spent fuel. Key messages include the following:
- Delays in reprocessing or disposal could result in spent fuel being stored for 100 years or longer. Safe, secure and effective storage of spent fuel manages fuel degradation while preserving future fuel cycle options.
- Ageing management programmes apply engineering, operations and maintenance actions to ensure safety is maintained during storage, future handling and transport.
- Site selection and facility and equipment design can significantly reduce the risks and costs of spent fuel storage over longer periods.
- Spent fuel storage configurations can be selected to accommodate uncertain storage periods, to facilitate ageing management and to provide flexibility needed to accommodate the uncertainty of future end points, such as reprocessing or disposal.
- By considering multiple licence renewals, regulatory frameworks can be designed to ensure safe storage until an acceptable end point is achieved.
- Safety can be assured by maintaining shielding, containment, decay heat removal and criticality control. Navigating the complexity of societal beliefs and values, as well as political systems, has proven to be a greater challenge for the management of spent fuel than maintaining its safety and security or addressing the technical and economic aspects.
- Sustainable spent fuel management requires policies and strategies to provide a clear, consistent and stable direction because they drive the need for spent fuel storage as well as the available options and timing for achieving an acceptable end point. Unless States address spent fuel reprocessing and disposal on a sufficient scale to accommodate their spent fuel discharges, then storage for longer and longer periods becomes the de facto end point — which is not considered to be consistent with the responsibility to protect human health and the environment.
An effective, periodic licence renewal process can ensure effective ageing management and strong institutional control. Hence, spent fuel can be safely and securely stored for as long as it may be necessary until transport for reprocessing or disposal. However, the risks and costs of storing the growing inventory of spent fuel will continue to increase; and in the absence of an end point, it will eventually become a significant societal burden.

The knowledge of transformation pathways and identification of transformation products (TPs) of veterinary drugs is important for health, food and environmental matters. Monensin (MON) is an ionophore antibiotic widely used to cure and prevent coccidiosis by chicken especially in broiler farming. Residues are not only found in food products (chicken and eggs) but also in the environment (manure, soil or water). Several transformation processes can alter the parent compound MON, ranging from biotransformation in living organism to biotic/abiotic and microbial processes in environmental matters.
The main objective of this work was to investigate the potential of electrochemistry (EC) to simulate oxidative transformation processes and to predict TPs of MON. An electrochemical reactor was used consisting of a flow-through cell with a glassy carbon working electrode. Derived TPs were analyzed by online coupling of EC and high-resolution mass spectrometry (HRMS) and LC-HRMS offline measurements. Among the generated TPs already known as well as unknown TPs of MON could be found.
Additionally, MON was subjected also to other transformation methods such as Fenton reaction, photochemical and hydrolysis experiments as well as metabolism tests with microsomes. As a result, different targeted and suspected TPs could be identified by analysis with LC-HRMS.
An overview of detected/identified TPs from this study will be presented in comparison to literature known metabolites and TPs.

Fresh cement paste is a suspension consisting of a hydraulic binder (cement), water, and numerous minor components – admixtures. Addition of admixtures aims at specific modification of properties of the fresh cement paste or hardened cementitious building material. Specific admixtures, so-called superplasticizers (SP), are used to improve the flowability of the fresh cement paste with reduced water content. The latter is the starting material for the high-strength concrete. Thus, SPs are essential for the ambitious construction projects.
However, uncontrollable retardation of the setting time in presence of SPs is occasionally observed. Obviously, SPs influence early products of the cement hydration leading to changes in the microstructure development. The hardening is thus delayed, and the quality of the resulting building material suffers. The mechanisms of the admixture action during the hydration process are still intensively investigated [1-7]. A detailed understanding of the admixture effects during the early hydration stage is the key to control and individual adjustment of the cement-based construction materials.
We use the unique combination of the wall-free sample holder and the time-resolved X-ray scattering analysis to achieve the full information about the hydrate phases formed under the influence of admixtures. We use ultrasonic levitator to start the cement hydration in levitated cement pellets [8, 9]. The sample levitation allows collection of the unimpaired information about cement hydrate phases. The most beneficial is the avoiding of the contributions of the sample holder material to the data signal.
We induce the cement hydration by adding water to unhydrated Portland cement during the data acquisition. The full phase composition of the hydrating cementitious system can be gathered in situ using wide angle X-ray scattering (WAXS). During the hydration of cement both crystalline and amorphous hydrate phases are formed. WAXS data contain the information about crystalline phases behind the Bragg reflections, whereas the amorphous hydrates influence the appearance of the background. Application of the data analysis specific for crystalline or amorphous phases is needed. The data quantification by the Rietveld method allows to conclude about the changes of the phase amounts due to the presence of admixture. The calculation of the pair distribution functions allows analysis of the amorphous hydrates. Based on this information, the SP effects and the extent of their involvement into the ongoing reactions can be concluded. A detailed understanding of the complex cement hydration process is envisioned.
Due to its advantages of being a direct comparison method, quantitative NMR spectroscopy (qNMR) becomes more and more popular in industry. While conventional high-field NMR systems are often associated with high investment and operational costs, the upcoming market of permanent-magnet based benchtop NMR systems show a considerable option for a lot of applications. The mobility of these systems allows to bring them more closely to the real production environment, e.g. for at-line quality control.
In this work we present an interlaboratory comparison study investigating the qNMR performance of state-of-the-art benchtop NMR spectrometers. Therefore, BAM prepared two samples of a mixture of NMR reference standards tetramethylbenzene (TMB) and tetrachloronitrobenzene (TCNB) at concentration levels of 200 mM and 10 mM. These “ready-to-use” samples were sent to participant laboratories, which performed analysis on their benchtop NMR equipment of different vendors and fields from 43 to 80 MHz. Raw data was reported back and further investigated by using different data analysis methods at BAM.
After this very first qNMR comparison study of benchtop NMR spectrometers show promising results, following studies are planned to cover more parts of the qNMR process, e.g. sample preparation and weighing, but also data analysis, as commonly done in similar studies for high-field NMR spectroscopy in industry and metrology.
X-ray absorption fine structure (XAFS) spectroscopy is a powerful and widely used tool for material characterization. It is non-destructive, element-sensitive and requires no long-range order of the sample, making it suitable for the investigation of amorphous phases. XAFS includes the X-ray absorption near edge structure (XANES) and the extended X-ray absorption fine structure (EXAFS), providing information about the electronical state of the absorbing atom and the spatial order of the neighboring atoms respectively.
Our new approach presented here is derived from the classical dispersive XAFS setup. The aim is to have a scanningless, stable and reproducible setup for applications, in which dynamic processes occur in the second time scale.
A broad polychromatic beam passes through the sample and is dispersed afterwards by a convexly bent Si (111) crystal. All energies are reflected under different angles and thus spatially separated, allowing a position sensitive detector to record a spectrum over the energy range of the incoming beam in a single shot.
Tests of this setup were performed at the BAMline @ BESSY-II (Berlin, Germany) with different metal foils. First in situ measurements comprised the investigation of the early stages of the zinc(II)2-methylimidazolate (ZIF-8) crystallization. This topic of metal-organic-frameworks (MOF) research is of interest for medical applications. Structural changes with a time resolution of 1 s could be followed. The second dimension of the broad beam can be used for a lateral resolution.
Working towards a comprehensive understanding of introduction pathways, number, and fate of micro¬plastics in the environment, suitable analytical methods are a precondition. Micro-spectroscopic methods are probably the most widely used techniques. Besides their ability to measure single spectra of a particle or fiber, most modern FTIR- and Raman microscopes are also capable of two-dimensional imaging. This is very appealing to microplastics research because it allows to simultaneously characterize the analytes chemically as well as their size (distribution) and shape.
Two-dimensional imaging on extensive sample areas with FTIR-micros¬copes is facilitated by focal plane array (FPA) detectors resulting in large data sets comprised of up to several million spectra. With numbers too large for manual inspection of each individual spectrum, automated data evaluation is inevitable. Identifying different polymers based on the comparison with known reference spectra (library search) has proven to be a suitable approach. For that purpose, FTIR-spectra of common plastics can be collected to create an individual reference library.
To Supplement this ‘targeted analysis’, looking for known substances via library search, an exploratory approach was tested. Principal component analysis (PCA) proved to be a helpful tool to drastically reduce the size of the data set while maintaining the significant information. Subsequently, cluster analysis was used to find groups of similar spectra. Spectra found in different clusters could be assigned to different polymer types. The variation observed within clusters gives a hint on chemical variability of microplastics of the same polymer found in the sample. Spectra labeled according to the respective cluster/polymer type were used to build a classification model which allowed to quickly predict the polymer type based on the FTIR spectrum. Classification was tested on a second, independent data set and results were compared to the spectral library search procedure.
Over the last few years, there has been a growing interest to apply spectroscopic methods to the agricultural field for better understanding of soil properties and for efficient, sustainable management of arable land. Within the project I4S (intelligence for soil), funded by the BMBF, an integrated system for site-specific soil fertility management is developed, consisting of different sensors like X-Ray fluorescence analysis (XRF), near-infrared spectroscopy (NIR) and laser-induced breakdown spectroscopy (LIBS). LIBS provides a fast and simultaneous multi-element analysis with little to no sample preparation, which makes it a suitable method for real-time analysis on the field.
The quantification of macro and micro nutrients in soils with LIBS is challenging due to matrix effects, different levels of moisture content and varying grain sizes. First studies revealed that the problems with matrix effects can be overcome by using well characterised soils as reference materials and chemometric tools like Partial Least Squares Regression (PLSR) for calibration.
The next step was to investigate the influence of moisture and grain sizes on the LIBS signal, which is a big issue when measuring directly on the field. The results showed that the LIBS signal decreases exponentially with increasing moisture content, as most of the laser energy is used for vaporising the water. With moisture contents of 30 % or higher almost no signal can be detected. This decrease is more severe for sandy soils than for clay soils. First tests of different grain size distributions indicate that the variation of the LIBS signal increases with growing amounts of larger grains. This results in a higher standard deviation, because of a poorer reproducibility of the plasma formation and plasma characteristic. With the help of chemometric tools the influence of moisture and grain sizes should be implemented in the calibration model for accurate analysis of nutrient composition in agricultural soils.
