Preactivation Crosslinking - An Efficient Method for the Oriented Immobilization of Antibodies

Crosslinking of proteins for their irreversible immobilization on surfaces is a proven and popular method. However, many protocols lead to random orientation and the formation of undefined or even inactive by-products. Most concepts to obtain a more targeted conjugation or immobilization requires the recombinant modification of at least one binding partner, which is often impractical or prohibitively expensive. Here a novel method is presented, which is based on the chemical preactivation of Protein A or G with selected conventional crosslinkers. In a second step, the antibody is added, which is subsequently crosslinked in the Fc part. This leads to an oriented and covalent immobilization of the immunoglobulin with a very high yield. Protocols for Protein A and Protein G with murine and human IgG are presented. This method may be useful for the preparation of columns for affinity chromatography, immunoprecipitation, antibodies conjugated to magnetic particles, permanent and oriented immobilization of antibodies in biosensor systems, microarrays, microtitration plates or any other system, where the loss of antibodies needs to be avoided, and maximum binding capacity is desired. This method is directly applicable even to antibodies in crude cell culture supernatants, raw sera or protein-stabilized antibody preparations without any purification nor enrichment of the IgG. This new method delivered much higher signals as a traditional method and, hence, seems to be preferable in many applications.
Wirtschaftliche und ökologische Aspekte führten in den letzten Jahren zu deutlich gesteigerten Anforderungen an die Effizienz und die Flexibilität petrochemischer Anlagen. Die heutzutage geforderten Prozesstemperaturen und -drücke lassen sich nur durch den Einsatz neuer warmfester Stahlgüten erreichen. Der mit Vanadium modifizierte Stahl 13CrMoV9-10 weist eine bessere Kriech- und Druckwasserstoffbeständigkeit auf und wird seit Mitte der 90er Jahre im petrochemischen Reaktorbau eingesetzt. Aufgrund der niedrigen Zähigkeit und hohen Festigkeit des Schweißgutes im geschweißten, nicht spannungsarm geglühten Zustand sowie einer erhöhten Sensitivität gegenüber Spannungsrelaxationsrissen bedarf dieser Stahl allerdings einer äußerst sorgfältigen schweißtechnischen Verarbeitung. Bisherige Untersuchungen zur Rissentstehung in warmfesten Stählen konzentrierten sich vorrangig auf thermische und metallurgische Einflussfaktoren, bieten jedoch nur wenige Erkenntnisse zum Einfluss des Schweißprozesses auf die Rissbildung beim Spannungsarmglühen unter Berücksichtigung realitätsnaher Fertigungsbedingungen.
Im ersten Teil wurde zunächst der Einfluss der Wärmeführung auf die mechanischen Eigenschaften anhand von frei schrumpfenden Laborproben untersucht. Während sich die Wärmeführung wesentlich auf die Schweißnahtstruktur auswirkte, war ein signifikanter Effekt auf die mechanischen Eigenschaften nicht nachweisbar. Auch traten infolge der Wärmenachbehandlung der frei schrumpfend geschweißten Proben keine Spannungsrelaxationsrisse auf.
Der zweite Teil umfasste die realitätsnahe Abbildung der Fertigungsbedingungen im petrochemischen Reaktorbau. Zu diesem Zweck wurden die konstruktiven Randbedingungen während des Vorwärmens, Schweißens, des Wasserstoffarmglühens und der abschließenden Wärmenachbehandlung realitätsnah in einer speziellen 3-D-Prüfanlage zur Simulation von Bauteilschweißungen abgebildet. Unter konstruktiver Schrumpfbehinderung gelang der Nachweis der unterschiedlichen Wirkung von Vorwärm- / Zwischenlagentemperatur und Streckenenergie auf die resultierenden Kräfte, Momente und Spannungen. Die Gesamtreaktionsspannung nach dem Schweißen wurde innerhalb des untersuchten Parameterfeldes maßgeblich durch die Streckenenergie beeinflusst.
Ein möglichst geringer Gesamtwärmeeintrag hat die niedrigste Bauteilbeanspruchung zur Folge. Während der Wärmenachbehandlung kam es in allen Versuchen zu Spannungsrelaxationsrissen. Deren kumulierte Länge korrelierte mit den ermittelten schweißbedingten Reaktionsspannungen.
Die Detektion der Risse während der Wärmenachbehandlung im Bauteilversuch erfolgte erstmalig in-situ mittels Schallemissionsanalyse. Die Rissinitiierung fand nachweislich im Temperaturbereich von 300 °C bis 500 °C statt. Die Reaktionsspannungen nach der Wärmenachbehandlung lagen unabhängig von der Ausgangsbelastung auf einem vergleichbaren Niveau. Die Zähigkeit der Schweißverbindung nahm klar gegenüber den frei schrumpfend geschweißten Proben ab.
Mittels REM- und TEM-Analysen an vergleichbaren belasteten und unbelasteten Proben wurde eine beschleunigte Alterung, durch das frühzeitige Ausscheiden von Sonderkarbiden während der Wärmenachbehandlung unter definierter Einspannung (d. h. unter Belastung), nachgewiesen. Die verstärkte Korngrenzensegregation korrelierte mit der signifikanten Abnahme der Kerbschlagarbeit der unter Einspannung geschweißten Proben.
If the particle size decreases, the ratio of surface area to volume increases considerably. This provides benefits for all surface-driven processes that run faster or at lower temperatures than larger particles. However, handling and characterization of the nanopowders are much more difficult. Particularly polydisperse powders with irregular shape, as grinding products, represent a challenge. Granulometry in the submicron and nanoscale often leads to incorrect results without knowledge of particle morphology.
This presentation demonstrates potentials of using the volume-specific surface area (SV or VSSA) in the granulometric characterization of nanopowders, for instance, correlations between the volume-specific surface area and the median particle size are discussed considering the particle morphology and the model of the logarithmic normal distribution.
Moreover, the presentation deals with the optimal dispersion of nanopowders during sample preparation. Indirect ultrasound device with defined cooling was developed to prevent both contamination by sonotrode abrasion and sample changes by heat.
Successful granulometric characterization of nanopowders demands both improved dispersion technology and very often an effective combination of two or more measurement methods.
Biofuels including ethanol and biodiesel (fatty acid methyl ester, FAME) represent an important renewable fuel alternative to petroleum-derived transport fuels. Increasing biofuels use would bring some benefits, such as a reduction in oil demands and greenhouse gas emissions, and an improvement in air quality. Materials compatibility is a major concern whenever the fuel composition is changed in a fuel system.
The objective of this research was to determine the resistance of frequently used sealing materials such as CR (chloroprene rubber), CSM (chlorosulfonated polyethylene), EPDM (ethylene-propylene-diene rubber), FKM (fluorocarbon rubber), FVMQ (methyl-fluorosilicone rubber), IIR (butyl rubber), NBR (acrylonitrile-butadiene rubber), PA (polyamides), PUR (polyester urethane rubber) and VMQ (methyl-vinyl-silicone rubber), in heating oil with admixtures of biogenic sources such as E10 (fuel with max. 10 % ethanol), E85 (fuel with 85 % ethanol), non-aged and aged biodiesel, diesel fuel with 5 % biodiesel, non-aged and aged B10 (heating oil with 10 % biodiesel) at 20 °C, 40 °C and 70 °C. Mass, tensile strength and breaking elongation of the test specimens were determined before and after the exposure for 84 days in the fuels. The visual examination of some elastomer test specimens clearly showed the great volume increase until break or partial dissolution. Shore hardness A and D (for PA) were determined before and after exposure of the test specimens in the biofuels for 42 days.
There is not determined a threshold for the reduction in tensile properties and Shore hardness in the international standards. Therefore, a threshold of 15 % was set for the evaluation of the compatibility. The sealing materials CR, CSM, EPDM, IIR and NBR were generally not resistant to biodiesel and B10. In summary, it can be therefore stated that the chemical resistance of the fluoropolymers FKM and FVMQ in fuels and biofuels is the best one.
Mit QI-FoKuS / Forschung für Konformitätsbewertung und Sicherheit streben wir auf Basis einer wiederkehrenden Befragung von Unternehmen und Konformitätsbewertungsstellen in Deutschland die Schaffung einer besseren Datengrundlage für die Forschung an. Diese soll es erleichtern, wichtige Zusammenhänge evidenzbasiert zu analysieren, um das systemische Zusammenwirken der Elemente der Qualitätsinfrastruktur besser zu verstehen. Dabei sollen zunächst die Elemente Konformitätsbewertung und Akkreditierung besonders im Vordergrund stehen.
Dose enhancement by gold nanoparticles (AuNP) was shown to increase the biological effectiveness of radiation damage in biomolecules and tissue. Most of the current studies focus on external beam therapy on combination with AuNP. Here we present a Monte-Carlo study (Geant4) to characterise radioactive AuNP. Radioactive 198 Au emits beta and gamma rays and is considered for applications with solid tumours. To effectively apply 198 AuNP their energy deposit characteristics have to be determined in terms of intrinsic and extrinsic properties e.g. AuNP diameter, AuNP density, and their clustering behaviour. After each decay process, the energy deposit, inelastic scattering events, kinetic energy spectrum of secondary particles within the AuNP themselves and in a spherical target volume of water up to 1 μm radius were determined. Simulations were performed for AuNP radii ranging from 2.5 nm to 20 nm radius, different cluster sizes and densities. The results show an increase of the energy deposit in the vicinity of the AuNP up to 150 nm. This effect nearly vanishes for distances up to one micron. For the case of AuNP clusters and the same activity, the enhancement of the energy deposit increases with the relative gold mass percentage and therefore can be adjusted by changing AuNP radius or clustering behaviour.
Die Untersuchungen sind durch das das DFG Schwerpunktprogramm 1374 für Biodiversitätsforschung motiviert. Anhand des überwachten Abbaus von Totholz soll untersucht werden, welche Auswirkung unterschiedliche Formen und Intensitäten der Landnutzung auf die Biodiversität innerhalb von drei Exploratorien zeigen. Bei den vorangegangenen Studien wurden Nahrungs¬netzte auf organismischer Ebene sowie Klima- und Umgebungsdaten untersucht und die Veränderung des ausgebrachten Totholzes als Masseverlust erfasst. Es wird jedoch erwartet, dass in Anhängigkeit der beteiligten Organismen und Umgebungsparameter verschiedene Mechanismen des Holzabbaus vorliegen, die in unterschiedlichen Abbauraten der Holzkomponenten Lignin, Cellulose und Hemicellulose resultieren. Aus diesem Grund ist die quantitative Untersuchung des Ligningehalts an Hölzern ein wesentlicher Schritt für ein verbessertes Verständnis der Biodiversität in den untersuchten Exploratorien. Um der hohen Probenanzahl, die sich aus der Kontrolle von jeweils 100 Proben (Plots) in den drei Exploratorien für Hölzer aus 13 Baumarten ergibt, gerecht zu werden, wird eine schnelle und effiziente Untersuchungsmethode benötigt. Diese wird derzeit in Form einer NIR-spektroskopischen Bestimmung des Ligningehalts entwickelt. Um basierend auf den NIR Spektren vermahlener Holzproben Lignin und weitere Holzkomponenten korrekt vorherzusagen, muss zunächst basierend auf nasschemischen Analysen ein chemometrisches Modell kalibriert werden. Dazu ist die Charakterisierung von mindestens 20 – 30 Proben, die eine hohe Varianz des Ligningehalts aufweisen, notwendig. Im Vortrag werden erste Untersuchungs¬ergebnisse für die Vorhersage des Ligningehalts von Kiefernhölzern, die für ein Jahr in dem Exploratorium Schorfheide exponiert wurden, vorgestellt. Die anschließende explorative Datenanalyse (PCA) unter Verwendung von insgesamt 28 Variablen zur Beschreibung klimatischer und Umgebungsparameter weist eine Reihe interessanter Korrelationen auf. Demnach ist der Ligningehalt auf der ersten Hauptkomponente (29% erklärte Varianz) positiv mit dem C/N-Verhältnis und negativ mit Landnutzungsindex korreliert. Masseverlust und Temperatur hingegen dominieren die zweite Hauptkomponente (12% erklärte Varianz).
Die BAM ist nahezu über die gesamte Wertschöpfungskette hinweg wissenschaftlich tätig. Von der sicheren und effizienten Wasserstofferzeugung (POWER-to-GAS), über die (Zwischen-)Speicherung von Wasserstoff in Druckgasspeichern bis hin zum Transport bspw. mittels Trailerfahrzeug zum Endverbraucher.
Komplettiert werden die Aktivitäten der BAM durch die sicherheitstechnische Beurteilung von wasserstoffhaltigen Gasgemischen, die Verträglichkeitsbewertung von Werkstoffen bis hin zur Detektion von Wasserstoffkonzentrationen über geeignete Sensorik, auch mittels ferngesteuerter Messdrohnen (sog. UAV-Drohnen).
Zudem untersucht die BAM proaktiv Schadensrisiken und Unfallszenarien für die Sicherheitsbetrachtung, um mögliche Schwachstellen aufzeigen und potenzielle Gefährdungen erkennen zu können.
Degradation of polyurethanes in various environments – Effects on molecular mass and crosslinking

The increasing application of polyurethanes (PU) in safety relevant sectors (fire protection, insulation, medicine technique) requires detailed knowledge of the stability and reliability of these materials. Different climate factors are supposed to induce diverse and overlapping degradation reactions. The knowledge of these degradation mechanisms is necessary for an estimation of the period of application depending on usage of the material. An essential property of a polymeric system is represented by the molecular weight. Since a change of the molecular weight is a measure for the chemical stability of a polymer, size-exclusion chromatography (SEC) was used to monitor changes of the molecular weight of thermoplastic polyether- and polyester urethane (TPU) exposed to thermal, hydrolytic and photo-oxidative (UV) degradation conditions for several days. Thermal treatments were performed at elevated temperatures (100 - 200 °C) under oxidative (air) as well as non-oxidative (nitrogen) conditions to evaluate the specific influence of oxygen on the degradation. At higher temperatures (≥ 175 °C) a fast decrease of the molecular masses of both PU accompanied by a high degree of crosslinking was found. At lower temperatures (≤ 150 °C) the polymers remained widely unaffected by thermal degradation within the investigated degradation interval of up to two weeks, which was already known from FTIR spectroscopy[1]. In contrast to that, UV treatment at 25 °C at less than 10 % rel. humidity (RH) resulted in a fast crosslinking, whereas the molecular masses of both PU decreased slower than during the thermal treatments. The depth of penetration of the UV radiation was determined using 3D printed PU samples with different thicknesses. Hydrolysis based degradation effects were less significant. Only slight molecular mass changes were detected at temperatures ≤ 80 °C within a time span of 14 days, while no crosslinking could be measured. Considering the degradation results at the investigated exposure parameters, it could be shown that ester-based PU in general exhibits a significant higher stability compared to ether-based materials.
Kurzfassung. Die meisten Faserverbundbehälter, die in Europa für den Transport gefährlicher Stoffe oder als Speicherbehälter für Wasserstoff oder Erdgas in Fahrzeugen Verwendung finden, werden für eine nicht begrenzte Lebensdauer ausgelegt. In regelmäßigen Abständen sind für diese Druckbehälter wiederkehrende Prüfungen vorgeschrieben, die in der Regel aus visuellen Inspektionen und hydraulischen Innendruckprüfungen bestehen. Die wiederkehrenden Prüfungen sind jedoch weder geeignet, die sichere Betriebsdauer festzulegen noch den Alterungszustand der Druckbehälter zu beurteilen. Aufgrund des weiter steigenden Drucks zur Kosten- und Gewichtsreduktion von Faserverbundbehältern und der damit einhergehenden Ausnutzung der existierenden Sicherheitsmargen gewinnen Methoden zur Beurteilung des Alterungsverhaltens dieser Druckbehälter immer mehr an Bedeutung.
Vor diesem Hintergrund wurde in der BAM das Vorhaben „Alterungsverhalten von Composite-Druckgefäßen mit Carbonfaser-Compositen unter gekoppelten Beanspruchungen (COD-AGE)“ gestartet, um die Alterung von Faserverbundbehältern mittels verschiedener zerstörungsfreier Prüfverfahren zu untersuchen. Die hier vorgestellten Ergebnisse wurden an einem Typ 4 Hybridbehälter mit Kunststoffliner gewonnen, in den im Rahmen des Herstellungsprozesses optische Glasfasern in verschiedenen Schichten integriert wurden. Der Behälter wurde mittels hydraulischem Innendruck und erhöhter Umgebungstemperatur zyklisch bis zum Lebensende belastet. Zwischen den einzelnen Lastsequenzen erfolgte eine an die wiederkehrende Prüfung im Betrieb angelehnte hydraulische Druckbelastung.
Sowohl die hydraulische Alterung als auch die Druckrampen wurden mittels Schallemissionsprüfung (SEP) und verteilter faseroptischer Sensorik mit dem Ziel einer frühzeitigen Versagensvorhersage überwacht. Durch die Schallemissionsanalyse sollte untersucht werden, ob zum Lebensende hin ein signifikanter Anstieg der Schallemissionsaktivität und -intensität, unterteilt in verschiedene Lastbereiche, beobachtet werden kann. Zusätzlich zum kontinuierlichen Monitoring der Zyklierung sollten die Druckrampen separat untersucht werden, um hier Potenziale für die wiederkehrende Prüfung zu erkennen.
Bei Geräten und Maschinen zur bestimmungsgemäßen Verwendung in explosionsgefährdeten Bereichen muss in der europäischen Union eine Zündgefahrenbewertung durchgeführt werden. Dabei müssen unter anderem die Gefahren von nichtelektrischen Zündquellen betrachtet werden, zu denen auch die mechanischen Schlagvorgänge gehören. Bei Schlagvorgängen kommt es infolge des Zusammenstoßes zweier Werkstücke zu einer Umwandlung der kinetischen Energie. Dabei erhöht sich die Temperatur der Werkstoffe an der Kontaktstelle und es kommt unter Umständen zu einem Abtrennvorgang kleiner Partikel erhöhter Temperatur. Sowohl die heißen Kontaktstellen als auch die abgetrennten Partikel können eine wirksame Zündquelle für ein explosionsfähiges Gasgemisch darstellen. Zur Festlegung von Grenzwerten wurden in der Norm EN 13463-1:2009 die Gasgemische anhand ihrer Explosionsgruppe klassifiziert und zu jeder Gruppe die maximale Energie des Schlagvorgangs festgelegt, unter derer die Entstehung einer wirksamen Zündquelle als unwahrscheinlich angenommen werden kann.
Die P-Rückgewinnung aus Teilströmen mit erhöhten P-Gehalten kann direkt in den Kläranlagenbetrieb integriert werden und führt dort in vielen Fällen zu betrieblichen und ökonomischen Vorteilen. Sie ist in der Regel auf Bio-P-Kläranlagen beschränkt, da hier die anaerobe P-Rücklösung initiiert und genutzt werden kann. Durch die reine P-Rücklösung durch Schaffung anaerober Bedingungen ist allerdings das Potential der Rückgewinnung auf 5 bis 15 Prozent der Zulauffracht limitiert.
Die meisten Kläranlagen in Deutschland werden mit einer chemischen Phosphor-Fällung betrieben. Diese Anlagen können aus dem Klärschlamm Phosphat in ausreichend hoher Konzentration nur zurückgewinnen, indem zusätzlich anorganische Säuren wie Schwefelsäure zugeführt werden oder ein Aufschluss mit flüssigem Kohlendioxid stattfindet. Es wird angenommen, dass sich mit diesen Verfahren mehr als 50 Prozent der Zulauffracht einer Kläranlage zurückgewinnen lassen. Der Aufschluss von Klärschlamm mit Schwefelsäure geht aber mit vergleichsweise hohen Umweltwirkungen sowie mit höheren Kosten im Vergleich zu anderen Phosphor-Rückgewinnungsverfahren einher. Ein sehr hohes Potential der Rückgewinnung von Phosphor (P) ist bei allen Verfahren gegeben, die als Rohstoff die Asche aus der Monoverbrennung verwerten. Der Aufschluss mit anorganischen Säuren ermöglicht die sichere Phosphor-Rückgewinnung von mehr als 80% Prozent des Kläranlagenzulaufs. Die Verfahren sind zum Teil großtechnisch umgesetzt oder es befinden sich großtechnische Anlagen im Bau. Auch thermochemische Verfahren wurden entwickelt und deren Einsatzbereitschaft im großtechnischen Maßstab erfolgreich demonstriert. In dem nachfolgenden Beitrag werden unterschiedliche praxiserprobte Verfahren zur Phosphor-Rückgewinnung unter technischen und wirtschaftlichen Aspekten vorgestellt.
Die agronomische Effizienz von Düngemitteln wird üblicherweise in Vegetationsversuchen geprüft. Diese erlauben die es, unterschiedliche Kulturpflanzen und deren Strategien der Nährstoffmobilisierung und -aneignung zu betrachten und verschiedene Substrattypen und deren Eigenschaften im Hinblick auf Nährstofffixierung und -freisetzung zu berücksichtigen. Solche Versuche, selbst wenn sie nicht im Feld, sondern nur im Gefäß durchgeführt werden, sind jedoch sowohl zeit- als auch kostenintensiv. In erster Näherung wird die Pflanzenverfügbarkeit insbesondere von Phosphor (P)-Düngemitteln daher in der Regel mit Hilfe standardisierter chemischer Extraktionsmethoden abgeschätzt, deren Durchführung in der deutschen ebenso wie in der europäischen Düngemittelverordnung normiert ist. Diese Methoden werden auch bei der staatlichen Düngemittelverkehrskontrolle genutzt, um zu überprüfen, ob die düngemittelrechtlichen Vorgaben eingehalten werden. Wie jüngere Arbeiten gezeigt haben, ist es jedoch schwierig, herkömmliche Standardextraktionsmethoden auf neue Dünger aus Recyclingmaterialien wie Klärschlamm zu übertragen. Korrelationen zwischen P-Aufnahme im Vegetationsversuch und chemischer Löslichkeit im Standardextrakt sind oft nur mäßig bis schwach oder überhaupt nicht signifikant. Die vorliegende Literaturübersicht gibt einen Überblick zum aktuellen Stand des Wissens über die chemische Löslichkeit von klärschlammbasierten P-Recyclingdüngern, deren Effizienz im Vegetationsversuch und statistische Relationen zwischen beiden Parametern. Anschließend werden alternative Methoden zur Abschätzung der P-Verfügbarkeit von Recyclingdüngern vorgestellt, die derzeit Gegenstand der wissenschaftlichen Diskussion sind.
Durch das alkalische Porenwassermilieu ist Stahl im Beton vor Korrosion geschützt. Unter ungünstigen Umgebungsbedingungen (Karbonatisierung, Chlorideintrag) kann die passive Deckschicht auf der Stahloberfläche zerstört werden. Im weiteren Verlauf können sich korrosionsbedingte Folgeschäden, wie Risse und Abplatzungen am Bauwerk ergeben. Um notwendige Sanierungsmaßnahmen sind frühzeitige und weitgehend zerstörungsfrei ermittelte Informationen über das aktuelle Korrosionsverhalten der Stahlbewehrung von großer Bedeutung. Die Potentialfeldmessung ist ein etabliertes und weit verbreitetes Verfahren zur Beurteilung des Korrosionszustandes der Bewehrung in Stahlbetonbauwerken. Mit Hilfe dieses Verfahrens können Bereiche aktiv korrodierender Bewehrung zerstörungsfrei lokalisiert werden. In der Regel kommt diese Messmethode bei der Detektion chloridinduzierter Korrosion zum Einsatz.
Zur Bedarfsplanung von Feuerwehren in Städten in der Bundesrepublik Deutschland wurden im Jahr 1998 die Qualitätskriterien Hilfsfrist, Funktionsstärke und Erreichungsgrad für ein standardisiertes Schadensereignis erarbeitet. Das standardisierte Schadensereignis basiert auf Ergebnissen der O.R.B.I.T.-Studie aus dem Jahr 1978. Der dort dargestellte Rauchgas-temperaturverlauf für einen standardisierten Wohnungsbrand stellt Ergebnisse von Brandversuchen in einer Wohnung aus dem Jahr 1939 dar. Die zur Untersuchung herangezogenen Einrichtungsgegenstände bestanden dabei überwiegend aus cellulosehaltigen Materialien wie Holz und Papier. Die zunehmende Verwendung von Werkstoffen auf der Basis von Holz und Polymeren führt jedoch zu einer veränderten stofflichen Zusammensetzung von Einrichtungsgegenständen in Wohnungen. Im Rahmen der vorliegenden Dissertation wurde der Einfluss dieser veränderten stofflichen Zusammensetzung von gegenwärtigen Einrichtungsgegenständen auf den Verlauf von Raumbränden sowie auf die damit verbundene Stoff- und Energiefreisetzung während der Brandentstehungs- und Brandausbreitungsphase untersucht. Im Fokus der Untersuchung stand der Raum der Brandentstehung sowie ein angrenzender Raum.
Die Ergebnisse zeigen, dass gegenwärtige Einrichtungsgegenstände sowohl im Raum der Brandentstehung als auch in dem angrenzenden Raum zu höheren Rauchgastemperaturen und -konzentrationen führen. Grund hierfür sind neben den höheren Wärmefreisetzungsraten auch die höheren Stoffausbeuten der in den gegenwärtige Einrichtungsgegenständen verwendeten Materialzusammensetzungen. Vor diesem Hintergrund zeigt sich, dass sich die auf Basis der O.R.B.I.T.-Studie definierten Standards nicht mehr als Grundlage zur Bedarfsplanung von Feuerwehren eignen. Diese können mit Hilfe der vorliegenden Ergebnisse einer Prüfung unterzogen werden, um den veränderten Brandverläufen im Rahmen einer zukünftigen Bedarfsplanung gerecht zu werden.
Laserhybridschweißen von dickwandigen Stählen mit elektromagnetischer Schmelzbadunterstützung

Die steigenden Anforderungen in Hinsicht auf Sicherheitsfaktoren von gefügten Bauteilen führen zu einer Zunahme der zu schweißenden Bauteildicken. Das Laserstrahl-Lichtbogen-Hybridschweißverfahren – verbreitet im industriellen Einsatz vor allem im Schiffs- und Windkraftanlagenbau – ermöglicht das einlagige Fügen von dickwandigen Strukturen. Eine Herausforderung stellt das Schweißen von dickwandigen Bauteilen mit reduzierter Geschwindigkeit in Wannenlage (PA-Position) da. Sie ist aufgrund des erhöhten hydrostatischen Druckes und die daraus resultierenden Tropfenbildung an der Wurzelseite bedingt realisierbar. Die im Rahmen dieser Studie eingesetzte elektromagnetische Schmelzbadunterstützung wirkt dem gravitationsbedingten Austropfen der Schmelze entgegen und kompensiert den hydrostatischen Druck. Dabei werden unterhalb der Schweißzone mit Hilfe eines extern angelegten oszillierenden Magnetfeldes Wirbelströme im Werkstück induziert, die eine nach oben gerichtete Lorentzkraft erzeugt. Die Lorentzkraft wirkt dem hydrostatischen Druck entgegen und stellt einen sicheren Schweißprozess ohne Tropfenbildung dar. Mit dem Hybridschweißverfahren mithilfe der elektromagnetischen Schmelzbadunterstützung gelingt es mit einem 20-kW Faserlaser bis zu 30 mm dicke Bleche in einer Lage zu schweißen. Bei 25 mm dicken einlagig geschweißten Platten aus S355 konnte ein Spalt bis 1 mm und ein Kantenversatz bis zu 2 mm sicher überbrückt werden. Die Reduzierung der Schweißgeschwindigkeit hat eine Verringerung der notwendigen Laserleistung zur Folge und begünstigt außerdem die mechanisch-technologischen Eigenschaften, infolge der reduzierten Abkühlgeschwindigkeit. Durch die geringe Martensitbildung führt dies zu einer Verbesserung der Kerbschlagzähigkeit.
Optimization of protein quantification via isotope dilution ICP-MS of a standard reference protein

Quantitative proteomics are nowadays one of the key tasks in life sciences. A multitude of methods for protein quantification are established and more techniques are developed each year, but there still is a lack of well characterized and quantified protein standards. We aim to develop an ICP-MS based method to quantify pure proteins reliably and traceable to SI. Here, we employ isotope Dilution analysis for the quantification of proteins of known stoichiometry via their sulfur content.
The quantification of the exact amount of sulphur is a big challenge due to a lack of SI-traceability and inconsistent results, when different methods are compared. Therefore, a reference procedure is required which allows SI-traceable values. In this work three procedures were developed for the quantification of the total sulphur amount in biodiesel by using inductively coupled plasma-isotope dilution mass spectrometry (ICP-IDMS), pure copper metals and copper alloys by ICP-IDMS and external calibration for GDMS and LA-ICP-MS at low concentration levels.
The most critical parts of the sulphur quantification were sulphur purification and pre-concentration. Sulphur-matrix separation procedures were developed to serve both sample types. For biodiesel samples the sulphur was purified and matrix separated by an anion exchange chromatographic procedure. The analytical procedure was fully validated by the use of a certified reference material, a step-by-step validation and an inter-laboratory comparison at CCQM key comparison level.
In the case of copper samples, the copper matrix was separated from sulphur by adding ammonia which forms a complex with the copper while releasing the sulphur prior to a chromatographic separation using a weak cation resin. After that the sulphur fraction was further purified by chromatographic means using first an anion ion exchange method and second a chelating resin. The method was validated by appropriate certified reference materials. The developed procedures enable sulphur measurements at the low g·g-1 level with sufficiently low measurement uncertainties (< 2 %, Urel).
The external calibration was performed to produce reliable measurement results for the routine analytical techniques GDMS and LA-ICP-MS. Matrix-matched reference materials whith exactly known amount of sulphur obtained by ICP-IDMS beforehand, were used as calibrators to quantify sulphur in copper samples. The metrological traceability to the SI for the mass fraction of sulphur is established for all presented procedures by an unbroken chain of comparisons, each accompanied by an uncertainty budget.
In isotope ratio applications metrological principles such as measurement uncertainty and SI traceability often are not considered or realized. This is also well-documented by the traceability exception related to delta scale isotope ratio measurements CCQM requested from the CIPM. Generally, delta scale isotope ratio measurements can be performed on a precision level that is significantly lower than the uncertainty level of isotope amount ratio measurements. In the case of magnesium, we demonstrated for the first time that isotope amount ratios can be measured with uncertainties close to the typical precision of magnesium delta values, δ26/24Mg, which are at the 0.1 ‰ level.
In the past, δ26/24Mg measurements were referenced to NIST SRM 980, the initial zero of the δ26/24Mg scale. With the development of MC-ICPMS, the detection of small but measurable isotopic differences in different chips of NIST SRM 980 became apparent and NIST SRM 980 was replaced by an intermediate artefact, DSM3. To solve this problem a suite of magnesium isotope reference materials, ERM-AE143, -AE144, and -AE145, has been certified in a first study by applying an ab initio calibration for absolute Mg isotope ratios without any a priori assumptions, a procedure which fulfils all requirements of a primary method of measurement. Thus, these materials qualified to establish SI-traceability for magnesium delta measurements. In a second study five expert laboratories participated to cross-calibrate all available magnesium isotope standards, which are NIST SRM 980, IRMM-009, ERM-AE143, ERM-AE144, ERM-AE145 and the standards DSM3 and Cambridge-1. The mean δ26/24Mg values for the individual iRMs, calculated from the laboratory means show 2 SD reproducibilities varying between 0.025 and 0.093 ‰. Propagated measurement uncertainties suggest a standard uncertainty of about 0.1 ‰ for δ26/24Mg determinations. Thus, SI traceability for magnesium isotope amount ratios and delta values is demonstrated to be established.
The development of new strategies for the sensing of phosphorylated and carboxylate-containing biomolecules such as phosphorylated tyrosine and histidine or sialic acid is currently of strong interest because those molecules are often involved in cancerous processes. Molecularly Imprinted Polymers (MIPs) are formed through the polymerization of a set of functional monomers and cross-linkers in the presence of a target molecule or an analogue of it. The target molecule is incorporated into the polymer network due to non-covalent interactions established with one or more functional monomers. Because those interactions are weak, the target molecule can be desorbed a posteriori from the polymer matrix, leaving imprinted cavities of complementary size, shape and electronic nature. Resembling the antigen-binding site of an antibody, MIPs can then be highly selective towards the target molecule. Besides these features, our approach involves also the use of functional fluorescent monomers which undergo fluorescence changes upon binding of the target molecule. This allows us to investigate the sensing process using fluorescence as a highly sensitive read-out. To that aim we present here the use of silica particles which contain a MIP shell formed by thiourea or guanidinium functionalized dyes for the recognition of phosphorylated and carboxylate-containing molecules of biological relevance in polar, protic solvents.
This work examined the factors that influence the droplet corrosion of CO2 pipeline steels caused by oxyfuel flue gases in dense phase CO2 at 278 K, simulating the underground transport conditions. The wetting properties were studied by contact angle measurement, revealing pH and time dependency on the reactive wetting behaviors of carbon steel X70. Exposure tests with CO2 saturated water droplet on steel surface showed that the impurities (220 ppmv SO2 and 6700 ppmv O2) diffused into the droplet and then reacted with metal surfaces in dense phase condition, forming the corrosion products. The corrosion rate was confirmed strongly depending on the droplet volume as well as the SO2 concentration. Condensation experiments carried out on freshly polished coupons in CO2 with 200 ppmv H2O, 220 ppmv SO2 and 6700 ppmv O2, showed that the formation and aggregation of droplets is time and temperature dependent. At 278 K, condensation happened stronger and the corrosion products, mainly consisted of dense hydrated FeSO3/FeSO4. While at 288 K, more fluffy corrosion products consisting of iron oxide/hydroxide and hydrated FeSO3/FeSO4 were found. Further exposure tests on carbon steel coupons with different surface roughness did not reveal the difference in weight loss/gain and therefore the corrosion rate.
Recent studies have shown that even at a very low concentration of impurities (less than 100 ppmv of SO2, NO2, O2 and H2O) the droplet formation and condensation of sulfuric and nitric acids in dense phase CO2 are possible and observable. To reveal the mechanism of droplet corrosion in dense phase CO2 at high pressure and low temperature, further studies on factors that affect wettability and resulting corrosion behaviors of transport pipeline steels are needed. In this study, effects of surface morphology were investigated by varying surface roughness of carbon steel coupons exposed to CO2 stream containing impurities to measure the wettability by contact angle and to observe the condensation as well as possible droplet corrosion that followed. Other considered factors were: pH of the droplet, temperature, droplet volume, and exposure time.
According to the results of the experiments carried out, the following points can be concluded:
1. Exposure tests showed that specimens with the addition of SiO2 particles and polyaniline particles exhibit better corrosion resistance than specimens with the addition of only polyaniline particles or binder only.
2. Open circuit potential measurements showed that in the beginning of the experiments, both coatings had an open circuit potential 100 mV higher than carbon steel. Within one day of immersion, the potential of carbon steel increased significantly, indicating the formation of oxide layer. After extended exposure the carbon steel potential sat between PS1 (which had higher potential) and PS2 (which had a lower potential) and all three were in the range –600 mV to –500 mV vs Ag/AgCl.
3. Both coatings PS-1 and PS-2 were degraded with increasing exposure time, shown by the decrease of absolute impedance value at low frequency range after 2, 4, and 6 days exposure.
4. Potentiodynamic test was performed after 3 hours and 6 days exposure, and it was shown that the coating in the initial stage exhibited more passive behavior than the specimens exposed for 6 days.
By exposure and electrochemical tests in the laboratory the Cu-effect on corrosion behavior of carbon steel, high-alloyed steels and Ti-alloy can be assessed.
Critical materials specific properties were determined by static exposure and electrochemical tests in an artificial geothermal water with high salinity and low pH, containing Cu. Conclusions were drawn using characteristic potential values.
It could be shown that significant Cu-deposition and -precipitation only occurred in combination with carbon steel. High-alloyed materials (S31603, S31653, S31700, S31703, S31803 and N08904) prevent the disturbing Cu-agglomeration. Therefore, they are suitable to be chosen for future design of the piping system, either in massive or in cladded form, if formation of crevices with non-metallic materials can be excluded.
From the interactions and pitting corrosion point of view, R50400 seems to be most favorable.

Chemical companies must find new paths to successfully survive in a changing environment. The potential of digital technologies belongs to these. Flexible and modular chemical plants can produce various high-quality products using multi-purpose equipment with short down-times between campaigns and reduce time to market for new products. Intensified continuous production plants allow for difficult to produce compounds. Therefore, fully automated “chemical” process control along with real-time quality control are prerequisites to such concepts while being based on “chemical” information.
As an example, a fully automated NMR sensor is introduced, using a given pharmaceutical lithiation reaction as an example process within a modular pilot plant. Therefore, a commercially available benchtop NMR spectrometer was adapted to the full requirements of an automated chemical production environment such as, e.g., explosion safety, field communication, and robust evaluation of sensor data. It was thereof used for direct loop advanced process control and real-time optimization of the process. NMR appeared as preeminent online analytical tool and allowed using a modular data analysis tool, which even served as reliable reference method for further PAT applications.
A full integration and intelligent interconnection of such systems and processes progresses only hesitantly. The talk should encourage to re-think digitization of process industry based on smart sensors, actuators, and communication more comprehensively and informs about current technical perspectives such as the “one-network paradigm”, edge computing, or virtual machines. These give smart sensors, actuators, and communication a new perspective.
Die Kreislaufwirtschaftsinitiative der Europäischen Kommission fordert eine grundlegende Erweiterung bestehender Rechtsrahmen. Anforderungen zu einer gesetzlich vorgeschriebenen Lebensdauer, Reparierbarkeit, Recyclingfähigkeit, etc. von Produkten sollen unter der Ökodesign-Richtlinie geregelt werden. Bei diesem Vorhaben steht der Normungsauftrag M/543 im Zentrum. Es wurde 2015 von der Europäischen Kommission vergeben und bislang sind acht horizontale bzw. generische EN-Normen geplant. Trotz der zu erwartenden hohen Reichweite dieses Vorhabens scheinen die Inhalte und der eigentliche Zweck der zu erarbeitenden Normen vielen Marktakteuren noch nicht vollständig bekannt zu sein, daher sollen im Rahmen des Webinars diese Inhalte kurz vorgestellt werden.
Bei der Bestrahlung und Bewitterung von polymeren Werkstoffen wird die Degradation durch eine Absorption von Strahlung initiiert und meistens durch weitere Klimagrößen, wie z.B. Wärme, Feuchte und atmosphärische Gase beeinflusst. Die Messung einer effektiven Bestrahlungsstärke und ihre zeitliche Integration sind deshalb von großer Bedeutung und zwar sowohl für die Kontrolle der Funktion des Bewitterungs- oder Bestrahlungsgerätes als auch als zusätzliche Angabe der wirkenden Beanspruchung.
Photochemische Prozesse werden beim Sehen oder allgemeiner bei Farbstoffen schon durch sichtbare (VIS) Strahlung, dagegen bei der Erythembildung der Haut und der Photodegradation von Polymeren erst durch die energiereicheren Photonen der ultravioletten (UV) Strahlung initiiert. Die spektrale Empfindlichkeit des Auges und die Erythembildung sind im VIS- bzw. UV-Bereich sehr stark wellenlängenabhängig, was durch die Angabe der mit der spektralen Hellempfindlichkeit V(λ) des Auges bewerteten VIS-Strahlung in Lux (lx) bzw. die mit der Erythem-Wirkfunktion Serythem bewertete UV Strahlung als dimensionsloser UV-Index berücksichtigt wird. Spektrale Empfindlichkeiten.
Die Photodegradation von Polymeren ist im UV-Bereich ebenfalls stark wellenlängenabhängig, was bisher bei der radiometrischen UV-Messung nicht berücksichtigt wurde. Seit Mitte der 70er Jahre, als die lx- bzw lxh- Messung bei der Strahlungsbeanspruchung langsam durch die Ermittlung der radiometrischen UV-Bestrahlungsstärke bzw. der UV-Bestrahlung im gesamten UV-Bereich abgelöst wurde, gab es hier bei der Bewitterungsprüfung keine weiteren Fortschritte auf dem Gebiet der Charakterisierung der Strahlungsbeanspruchung im UV-Bereich.
Seit Jahren liegen zahlreiche Ergebnisse über die spektrale Empfindlichkeit von Photo-degradationsprozessen vor, die zeigen, dass die spektrale Empfindlichkeit im Bereich der Globalstrahlung an der kurzwelligen Grenze bei ca. 300 nm, am größten ist und im langwelligen UV-Bereich bei ca. 400 nm gegen Null geht. Die Messungen der radiometrischen Größen UV-Gesamtbestrahlungsstärke und UV-Gesamtbestrahlung erfasst deshalb über weite Wellenlängenbereiche Strahlungsanteile, die für die Photodegradation bedeutungslos sind und mittelt über potenziell bedeutsame Änderungen im kurzwelligen UV-Bereich hinweg.
Es wird das Konzept eines neuen UV-Radiometers vorgestellt, dessen spektrale Empfindlichkeit einer mittleren spektralen Empfindlichkeit der Photodegradation von Polymeren entspricht. Die effektive UV-Bestrahlungsstärke und UV-Bestrahlung kann dadurch wesentlich genauer erfasst werden.

Methods for the rapid and sensitive detection of target analytes are gaining importance in medical diagnostics and environmental monitoring, in the security, occupational health and safety as well as food sectors. Among all of the methods employed for rapid tests, lateral flow assays (LFAs) are the most commonly used, and hundreds of test kits based on this technique are available on the market. A major drawback is that most of these capture agents either indicate the analyte only indirectly, and in most cases a second binding agent able to bind directly or indirectly to the analytes is necessary (e.g., a secondary labeled antibody). Furthermore, in certain cases in which the (ultra)trace detection of an analyte is required, the traditional approach of a certain number of probe molecules being conjugated to a particular support is not sufficient. Therefore, novel concepts implementing steps of effective signal amplification are urgently required.
Keeping in mind these limitations, we thought that the sensitivity of these systems should be improvable through employment of gated reporter molecule-releasing hybrid nanoparticle materials on novel lateral flow devices. On one hand, the gated sensor material can produce a massive signal amplification, by releasing many reporter molecules only after chemical recognition of a few analyte molecules has taken place in an independent and separate step at the pore openings. On the other hand, the employment of tailored capture materials for the selective interaction with the released reporter molecules in a second arbitrary zone on the strip allows to concentrate or focus the latter for more efficient detection or to create selective multi-spot detection zones, which renders the simultaneous detection of several reporter molecules at the same time in multiplexed detection of various analytes possible.
For that purpose, we have prepared several stimuli-responsive materials for small-molecule sensing based on specific interactions between biomolecules such as antibodies with the corresponding analytes for the detection of certain explosives. In order to prepare these bio-capped materials, we have selected silica mesoporous nanoparticles (MSNs) as inorganic support due to their unique properties such as defined void structure, high inner surface area and flexible functionalization chemistry. These MSNs are loaded with a brightly fluorescent indicator dye, and the external surface is subsequently functionalized with suitable molecules able to interact with antibodies, efficiently inhibiting dye release. The opening protocol and delivery of the entrapped dye is reminiscent of a displacement reaction involving the presence of the target analyte, producing a displacement of the biomolecule and allowing the detection of the target analyte.
The presentation discusses general aspects of system design as well as analytical performance and highlights the integration into a lateral-flow assay, showing as an example the determination of the explosives TATP, TNT and PETN with fluorescence readout, in single-substance and multiplexing modes.
Es lässt sich feststellen, dass eine Vielzahl unterschiedlicher flüchtiger organischer Verbindungen aus Bauprodukten freigesetzt werden kann. Hierbei gibt es in den meisten Bauproduktgruppen sowohl emissionsarme als auch emissionsreiche Produkte.
Manche Verbindungen sind produktgruppenspezifisch; so werden beispielsweise Siloxane aus Silikondichtmassen freigesetzt, andere Verbindungen sind eher unspezifisch wie Formaldehyd und Essigsäure, die aber in unterschiedlichen Konzentrationen aus einer Vielzahl von Produkten abgegeben werden und zu den am häufigsten emittierten Verbindungen zählen.
Es ist darauf hinzuweisen, dass VOC-Analytik und Gerüche im Allgemeinen keinen Zusammenhang zeigen.
Gerade bei modernen, dichten Bauten mit geringem natürlichem Luftwechsel sind hohe Schadstoffkonzentrationen zu erwarten, wenn nicht emissionsarme Materialien und Produkte zum Einsatz kommen.

Deflagrationssicherungen sind Sicherheitseinrichtungen, die den Durchfluss von brennbaren Gasen und Dämpfen brennbarer Flüssigkeiten ermöglichen, den Flammendurchschlag und weitere Explosionsausbreitung nach der Zündung von explosionsfähigen Gas-/Dampf-Luftgemischen jedoch verhindern sollen.
Von besonderer Bedeutung für die Wirksamkeit der Deflagrationsrohrsicherung ist der Einbauort. Diesbezügliche Abweichungen von den normativen Anforderungen, die Grundlage der Prüfung und Zulassung von Deflagrationsrohrsicherungen sind, können zum Versagen der Sicherheitseinrichtung führen. Oft lassen sich allerdings aus betrieblichen bzw. verfahrenstechnischen Gründen der Norm entsprechende Einbaubedingungen nur schwer oder nicht realisieren.
Deshalb besteht das Ziel der Untersuchungen eines F&E-Vorhabens mit der Braunschweiger Flammenfilter GmbH darin, einen Beitrag zur Schaffung einer belastbaren Datenbasis zu leisten, um den Einfluss von Rohrleitungsverzweigungen auf deflagrative Gasexplosionen systematisch zu bewerten.
Considerations for nanomaterial identification of powders using volume-specific surface area method

The EC’s recommendation for a definition of nanomaterial (2011/696/EU) should allow the identification of a particulate nanomaterial based on the number-based metric criterion according to which at least 50% of the constituent particles have the smallest dimension between 1 and 100 nm. However, it has been recently demonstrated that the implementation of this definition for regulatory purposes is conditioned by the large deviations between the results obtained by different sizing methods or due to practical reasons such as high costs and time-consuming.
For most measurement methods for particle size determination it is necessary to initially disperse the particles in a suitable liquid. However, as the particle size decreases, the adhesion forces increase strongly, making it more difficult to deagglomerate the particles and to assess accurately the result of this process. Therefore, the success of the deagglomeration process substantially determines the measurement uncertainty and hence, the comparability between different methods.
Many common methods such as dynamic light scattering (DLS), centrifugal liquid sedimentation (CLS) or ultrasound attenuation spectroscopy (US) can give good comparable results for the size of nanoparticles, if they are properly separated and stabilized (e.g. in reference suspensions).
In order to avoid the use of hardly available and expensive methods such as SEM / TEM for all powders, an agglomeration-tolerant screening method is useful.
One of the measurement methods well suited to probe the size of particulate powder is the determination of the volume-specific surface area (VSSA) by means of gas adsorption as well as skeletal density. The value of 60 m2/cm3 corresponding to spherical, monodisperse particles with a diameter of 100 nm constitutes the threshold for decisioning if the material is a nano- or non-nanomaterial. The identification of a nanomaterial by VSSA method is accepted by the EU recommendation.
However, the application of the VSSA method was associated also with some limitations. The threshold of 60 m2/cm3 is dependent on the particle shape, so that it changes considerably with the number of nano-dimensions, but also with the degree of sphericity of the particles. For particles containing micro-pores or having a microporous coating, false positive results are induced. Furthermore, broad particle size distributions made necessary to additionally correct the threshold. Based on examples of commercially available ceramic powders, the applicability of the VSSA approach was tested in relation with SEM and TEM measurements. The introduction of a correction term for deviations from sphericity and further additions improved the applicability of VSSA as a screening method.
