Raw data from metabolomics experiments are initially subjected to peak identification and signal deconvolution to generate raw data matrices m × n, where m are samples and n are metabolites. We describe here simple statistical procedures on such multivariate data matrices, all provided as functions in the programming environment R, useful to normalize data, detect biomarkers, and perform sample classification.
Das Ziel der Arbeit war die Entwicklung einer magnetischen Anreicherungsmethode für eine Bead-basierte Substanzbibliothek zur Identifizierung von Proteinbindern. Im Ramen des Forschungsbelegs wurde bereits eine Anreicherungsmethode für ein Modellsystem einer Peptidbibliothek entwickelt. Das Modellsystem bestand dabei aus zwei verschiedenen Peptiden. Als Positivkontrolle wurde das FLAG-Peptid gewählt, welches selektiv gegen den Anti-FLAG-Antikörper bindet, und ein weiteres Peptid wurde als Negativkontrolle gewählt. Anhand dieses Modellsystems konnte die zuvor entwickelte Anreicherungsmethode von 20.000 auf 1.000.000 Beads vergrößert werden. Hierbei wurde ein Anreicherungsfaktor von 818 und eine gute Wiederfindungsrate von 82% erreicht.
In dieser Arbeit wurden drei verschiedene Nanomaterialien auf ihre Bindungsfähigkeit zu Proteinen untersucht. Zu Beginn standen dabei die Herstellung stabiler Dispersionen der einzelnen Nanopartikel und die Stabilität der gebildeten Konjugate im Vordergrund. Der Nachweis einer erfolgreichen Konjugatbildung, sprich der Beschichtung von Nanopartikel mit Proteinen, wurde sowohl qualitativ mittels DLS-Messungen als auch über quantitative Protein-Bestimmungen erbracht. Für die Quantifizierung konnten verschiedene Methoden eingesetzt werden. Neben der klassischen Vorgehensweise, welche indirekt über die Quantifizierung von ungebundenem Protein im Überstand erfolgt, konnten ihm Rahmen dieser Arbeit verschiedene direkte Bestimmungsmethoden entwickelt werden. So wurden mittels kolorimetrischer Tests, wie dem BCA-Assay und dem Bradford-Assay, Nanodiamantdispersionen mit Hilfe einer Korrekturwellenlänge vermessen und quantifiziert. Ebenso zum Einsatz kam die Methode der Aminosäureanalytik, welche aufgrund ihrer guten Rückführbarkeit auf Aminosäurestandards Ergebnisse mit hoher Richtigkeit generieren kann und ebenso die Detektion kleiner Proteinmengen möglich macht.
Nach den erfolgten quantitativen Betrachtungen wurden die Protein-beschichteten Nanopartikel auf ihre Anwendbarkeit als Analoga von Virus-like Particles (VLP) bei einer Immunisierung zur Gewinnung von polyklonalen Antikörpern gegen humanes Ceruloplasmin in Kaninchen überprüft. Es konnte mittels ELISA gezeigt werden, dass die Konjugate erfolgreich für die Herstellung von Antikörpern eingesetzt werden können und im zeitlichen Verlauf einer Immunisierung eine Steigerung des Antikörper-Titers zu erreichen ist.
This Special Issue of Fire and Materials brings together a Collection of papers that were presented at the 2016 Interflam conference in the Fire & Facades session along with recent submissions to the Fire and Materials Journal that are very relevant to the Fire and Facades subject. Having attended the Interflam conference, all Fire and Materials Journal editors who attended realised the importance of this topic. We felt it important that this session content would both be reviewed and made available to a wider audience and that it was timely that we featured this increasing important topic in our journal. Accordingly, authors of selected papers were invited to update and extend their papers for journal submission, and other appropriate peer review papers subsequently submitted to the journal were included in the collection.
We propose a method based on artificial neural networks to extract strain information from wavelength-scanning coherent optical time domain reflectometry (C-OTDR) data. Our neural network algorithm performs more than two orders of magnitude faster than the conventional approach.
This is due to the highly parallel evaluation of the neural networks on a GPU accelerated computer and the fact that conventional correlation and interpolation analysis needs many Iteration steps. This opens the way for real-time C-OTDR strain sensing because the neural Network strain predictions require less time than the measurements themselves. Real-time data Analysis enables long-term sensing e.g. in structural health monitoring, because the large amount of raw data does not have to be stored but can immediately be reduced to the strain data of interest.
Monitoring chemical reactions is the key to chemical process control. Today, mainly optical online methods are applied, which require excessive calibration effort. NMR spectroscopy has a high potential for direct loop process control while exhibiting short set-up times. Compact NMR instruments make NMR spectroscopy accessible in industrial and harsh environ¬ments for advanced process monitoring and control, as demonstrated within the European Union’s Horizon 2020 project CONSENS.
We present a range of approaches for the automated spectra analysis moving from conventional multivariate statistical approach, (i.e., Partial Least Squares Regression) to physically motivated spectral models (i.e., Indirect Hard Modelling and Quantum Mechanical calculations). By using the benefits of traditional qNMR experiments data analysis models can meet the demands of the PAT community (Process Analytical Technology) regarding low calibration effort/calibration free methods, fast adaptions for new reactants or derivatives and robust automation schemes.
Der Mangel an Anwendungsrichtlinien und Validierungsverfahren für struktur-eingebettete faseroptische Dehnungssensoren, insbesondere auf Faser-Bragg-Gitter (FBG)-Basis, führte bislang dazu, dass diese Sensoren trotz ihrer hervorragenden Eigenschaften im Bereich der kommerziellen Material- und Strukturüberwachung nur in geringem Umfange eingesetzt wurden. Fragen zur Degradation der Sensoren unter Beanspruchung, Alterung durch klimatische Einflüsse und Enthaftung infolge Belastung der Struktur konnten bisher nur theoretisch simuliert oder anhand exemplarischer Proben für einzelne einflussnehmende Parameter im Labor untersucht werden. Die Erfassung des Sensorverhaltens im Bauteil unter komplexen Umwelteinflüssen während des Bauteilbetriebes, um damit eine Aussage zur Zuverlässigkeit der Sensormessdaten zu gewinnen, war bisher nicht möglich.
Im Forschungsprojekt FAMOS² wurde deshalb von 2014 bis 2018 die Realisierung eines Autodiagnoseverfahrens für faseroptische Dehnungssensoren erarbeitet, um die Funktionszuverlässigkeit und Langzeitstabilität der Sensoren während des Bauteilbetriebes innerhalb einer Bauteilstruktur bewerten zu können. Im Rahmen des Forschungsprojektes wurde gezeigt, dass ein FBG-Sensor mit einer speziell angepassten magnetostriktiven Schicht magnetisch so angeregt werden kann, dass mit einer zugeschnittenen Messmethodik zu jedem Zeitpunkt eine Bewertung der Sensorzuverlässigkeit unter Betriebsbedingungen möglich ist. Die auf den Sensor aufgebrachte magnetostriktive Schicht erzeugt bei Anregung mit einem passenden Magnetfeld eine gepulste, synthetische Dehnung. Diese Dehnung steht in einem definierten Verhältnis zum anregenden Magnetfeld und wird optisch über die Verschiebung der Bragg-Wellenlänge ausgelesen. Die Konstanz dieses Verhältnisses stellt dann ein Maß für die korrekte Funktionsfähigkeit des Sensors dar. Das Beschichtungs-verfahren, das magnetische Anregungs- und optische Ausleseverfahren, wie auch die Validierung des Sensors unter Berücksichtigung faseroptischer Effekte wurden während des Projekts konzeptionell entwickelt, aufgebaut und charakterisiert. Qualitativ kann mit diesem Verfahren zunächst eine Gut-/Schlecht-Beurteilung des Sensors vorgenommen werden. Perspektivisch bietet das Verfahren jedoch auch die Möglichkeit, verschiedene Versagensmechanismen bestimmen und klassifizieren zu können. Es zeichnete sich schon während des Projektverlaufs ab, dass sich aus den erzielten Ergebnissen über das Autodiagnoseverfahren hinaus weitere Anwendungsmöglichkeiten für den Einsatz des kombinierten magnetostriktiv-faseroptischen Sensorprinzips, beispielsweise als reiner Magnetfeldsensor, ergeben.
Nach Abschluss dieses Forschungsprojekts besteht nun die Möglichkeit, den Funktionszustand faseroptischer Dehnungssensoren jederzeit unabhängig von Alter, Belastung oder Kenntnis eines Referenzzustands zu einem bestimmten Zeitpunkt bestimmen zu können. Aufbauend auf diesen Ergebnissen können weiterführende Forschungen die Messmethodik vertiefend charakterisieren und weitere Anwendungsfälle erschließen.
Degradation of AISI 630 exposed to CO2-saturated saline aquifer at ambient pressure and 100 bar

In general high alloyed steels are suitable as pipe steels for carbon capture and storage technology (CCS), because they provide sufficient resistance against the corrosive environment of CO2-saturated saline aquifer which serves as potential CCS-site in Germany. High alloyed martensitic steel AISI 630 has been proven to be sufficient resistant in corrosive environments, e.g. regarding heat, pressure, salinity of the aquifer, CO2-partial pressure), but reveals a distinct corrosion pattern in CCS environment. Therefore coupons of AISI 630 heat treated using usual protocols were kept at T=60 °C and ambient pressure as well as p=100 bar up to 8000 h in an a) water saturated supercritical CO2 and b) CO2-saturated synthetic aquifer environment similar to on-shore CCS-sites in the Northern German Basin. AISI 630 precipitates a discontinuous ellipsoidal corrosion layer after being exposed for more than 4000 hours. Best corrosion resistance in the CO2-saturated synthetic aquifer environment phase is achieved via normalizing prior to exposure. In water saturated supercritical CO2 tempering at medium temperatures after hardening gives lowest corrosion rates. Corrosion fatigue via push-pull tests with a series of 30 specimens was evaluated at stress amplitudes between 150 MPa and 500 MPa (sinusoidal dynamic test loads, R=-1; resonant frequency ~ 30 Hz). The endurance limit of AISI 630 is reduced by more than 50% when exposed to CCS environment (maximum number of cycles (10 x 106) at a stress amplitude of 150 MPa).
In CCS environment (carbon capture and storage) pipes are loaded statically and/or cyclically and at the same time exposed constantly to the highly corrosive hot thermal water. Experimental procedures such as ambient pressure immersions tests, in-situ corrosion fatigue experiments using a flexibly designed corrosion chamber at ambient pressure and a specially designed corrosion chamber at high pressure. Experimental set-ups for push/pull and rotation bending load are introduced. The corrosion behavior and lifetime reduction of high alloyed steels (X46Cr13, 1.4043), (X5CrNiCuNb16-4, 1.4542) and (X2CrNiMoN22-5-3, 1.4462) is demonstrated (T=60 °C, geothermal brine: Stuttgart Aquifer flow rate: 9 l/h, CO2 ).
The commercial usage of latent thermal energy storages primarily depends on the development of a suitable phase change material (PCM). For industrial high temperature applications above 400 °C multicomponent chloride eutectics are promising and therefore discussed seriously. The profound thermodynamic investigation of such eutectics requires a much greater amount of specimen material than conventional calorimeter can handle. Therefore, a special adiabatic calorimeter was developed and designed. With a specimen mass of > 100 g the typical thermodynamic measurements with a commercial calorimeter can be extended by cycle stability measurements, which are often decisive for practical application of PCM. Furthermore, by implementing corrosion specimens inside the calorimeter high temperature corrosion experiments according to ISO 21608 can be performed inside the calorimeter. Adiabatic measuring conditions can be provided by using two separate heating systems. Therefore, the outer “protective system” follows the temperature curve of the inner “measuring system” minimizing the temperature difference between the heating systems and simultaneously preventing heat losses from the measuring systems.
The vulnerability of low-quality concrete of some bridges to ageing and deterioration while they are exposed to environmental conditions during their service life, influence of growing population and climate changes, is a well-known effect and requires survey. For survey, the development of a low cost, cost-efficient sensor network to be embedded for continuous or attached to the surface for periodic monitoring is essential.
Changing ground water conditions, flood or even leakage in fresh or disposal of waste water can lead to in-creased moisture content in building materials and structural elements as walls or floors. Moisture allows the transport of other adverse chemicals that might reduce the durability of structures. The paper presents a feasibility study about the capability of microwave modules as bluetooth® or ZigBee with frequency range at 2,4 GHz to create advanced Wireless Sensor Networks (WSN) for estimation and visualization of moisture distribution in infrastructure assets. It was found that the value of the received signal strength indication (RSSI) depends on the moisture content and can be the basis for the moisture distribution inside a massive structure.
The microwave modules may be used to compose networks outside and inside of building materials or structural elements. The higher the number of connections between BLE modules, the more precise and the more distant information can be obtained from the network. Mesh networking was applied to increase the quantitative information.
A systematic study of the luminescence properties of monodisperse β-NaYF4: 20% Yb3+, 2% Er3+ upconversion nanoparticles (UCNPs) with sizes ranging from 12–43 nm is presented utilizing steady-state and time-resolved fluorometry.
Special emphasis was dedicated to the absolute quantification of size- and environment-induced quenching of upconversion luminescence (UCL) by highenergy O–H and C–H vibrations from solvent and ligand molecules at different excitation power densities (P). In this context, the still-debated Population pathways of the 4F9/2 energy level of Er3+ were examined. Our results highlight the potential of particle size and P value for color tuning based on the pronounced near-infrared emission of 12 nm UCNPs, which outweighs the red Er3+ emission under “strongly quenched” conditions and accounts for over 50% of total UCL in water. Because current rate equation models do not include such emissions, the suitability of these models for accurately simulating all (de)population pathways of small UCNPs must be critically assessed. Furthermore, we postulate population pathways for the 4F9/2 energy level of Er3+, which correlate with the size-, environment-, and P-dependent quenching states of the higher Er3+ energy levels.
A shell test bench was developed at BAM 5.3 which allows for static and fatigue testing of curved fiber-reinforced plastic (FRP) structures, during which in-situ the damage state can be non-destructively inspected by thermography and strain-field measurement techniques. Sandwich shell specimens with typical wind turbine blade manufacturing defects were designed and tested. The tested imperfections show a fairly significant reduction (up to 90%) of the shell test specimens‘ lifetime, depending on the type of imperfection. Using the in-situ NDT methods incorporated in the shell test bench, the location and cycle time of the initial defects and the damage evolution was investigated.
Due to rapid, localized heating and cooling, distortions accumulate in additive manufactured laser metal deposition (LMD) components, leading to a loss of dimensional accuracy or even cracking. Numerical welding simulations allow the prediction of these deviations and their optimization before conducting experiments. To assess the viability of the simulation tool for the use in a predictive manner, comprehensive systems as well as to choose the optimal product matches, product analysis methods are needed. Indeed, most of the known methods aim to analyze a product or one product family on the physical level. Different product families, however, may differ largely in terms of the number and nature of components. This fact impedes an efficient comparison and choice of appropriate product family combinations for the production system. A new methodology is proposed to analyze existing products in view of their functional and physical architecture. The aim is to Cluster these products in new assembly oriented product families for the optimization.
The application of implants e.g. for osteosynthesis or substitution of worn out joints is common practice since decades. Successes in surgery and orthopedics are highly dependent on the use of artificial parts for implanting into the human body. Over the years the number of surgical procedures and the number of different implants were growing rapidly. Implants, however, are exposed to complex mechanical, corrosive and tribological loads along with restriction in geometric dimension. While being essentially successful, the sheer number of cases leads to a rising number of implant failures. Such failures are painful for the affected patients and are very costly.
The failure cases presented in this paper occurred from 1981 to 2016. The predominant failure mechanism of all kinds of implants is fatigue fracture. Fatigue cracks are initiated due to a whole bunch of reasons: Material defects and fabrication defects are rare events. The main factors are cyclic overload due to improper fitting with delayed bone healing, corrosion and unauthorized modification of the implant during surgery
A shell test bench was developed at BAM 5.3 which allows for static and fatigue testing of curved fiber-reinforced plastic (FRP) structures, during which in-situ the damage state can be non-destructively inspected by thermography and strain-field measurement techniques. The described investigation of carbon-fibre-reinforced plastics (CFRP) documents that damage evolution can be observed by means of X-ray refractography. Comparative investigations with synchrotron technique on CFRP and grey-scale analysis on glass fibre-reinforced-plastics (GFRP) confirm these results. Moreover it was found that the fracture mechanical properties of the matrix system influence damage nucleation and propagation in the laminate during static and fatigue loads. Single-step fatigue tests were carried out on Laminates with RIM135 and LY556 matrix systems made from non-crimped fabric (NCF) or twill weave in different fibre orientations. The damage to the LY556 laminates was characterized by laminate cracks growing rapidly over the whole specimen width, whereas the damage on the RIM135 laminates was characterized by an earlier onset of micro-cracking followed by laminate cracks. The specimens were fatigued up to 108 (very high cycle fatigue (VHCF) regime) load cycles. S-N-curves of damage initiation were drawn and boundaries were identified for endurance within the VHCF regime. A phenomenology based model focusing on matrix stress was applied to reproduce the first inter-fibre failure (IFF) under static and fatigue loads.
Cerro do Jarau is a conspicuous, circular morpho‐structural feature in Rio Grande do Sul State (Brazil), with a central elevated core in the otherwise flat “Pampas” terrain typical for the border regions between Brazil and Uruguay. The structure has a diameter of approximately 13.5 km. It is centered at 30o12′S and 56o32′W and was formed on basaltic flows of the Cretaceous Serra Geral Formation, which is part of the Paraná‐Etendeka Large Igneous Province (LIP), and in sandstones of the Botucatu and Guará formations. The structure was first spotted on aerial photographs in the 1960s. Ever since, its origin has been debated, sometimes in terms of an endogenous (igneous) origin, sometimes as the result of an exogenous (meteorite impact) event. In recent years, a number of studies have been conducted in order to investigate its nature and origin. Although the results have indicated a possible impact origin, no conclusive evidence could be produced. The interpretation of an impact origin was mostly based on the morphological characteristics of the structure; geophysical data; as well as the occurrence of different breccia types; extensive deformation/silicification of the rocks within the structure, in particular the sandstones; and also on the widespread occurrence of low‐pressure deformation features, including some planar fractures (PFs). A detailed optical microscopic analysis of samples collected during a number of field campaigns since 2007 resulted in the disclosure of a large number of quartz grains from sandstone and monomict arenite breccia from the central part of the structure with PFs and feather features (FFs), as well as a number of quartz grains exhibiting planar deformation features (PDFs). While most of these latter grains only carry a single set of PDFs, we have observed several with two sets, and one grain with three sets of PDFs. Consequently, we here propose Cerro do Jarau as the seventh confirmed impact structure in Brazil. Cerro do Jarau, together with Vargeão Dome (Santa Catalina state) and Vista Alegre (Paraná State), is one of very few impact structures on Earth formed in basaltic rocks.
Mycotoxins are toxic secondary metabolites produced by filamentous fungi. The contamination of mycotoxins in food and feed products has been recognized to be an important safety issue for human and animal health. In the context of food safety, medical herbs nowadays are also considered as potential source of mycotoxins. Considering that mycotoxins are generally stable compounds and could not be destroyed completely during food-processing operations, prevention of contamination is identified as main issue.
Currently, biochemical, and immunochemical methods based on the use of antibodies as a specific recognition bioreagents are increasingly found to be applied in screening. Along with good common enzyme linked immunosorbent assay (ELISA), more and more widely used method is fluorescence polarization immunoassay (FPIA). These methods help to monitor of many toxic substances in large number of samples and carried out quickly, easily, cheaply, and give good results in quantifying the one or more substances. That is why the use of immunochemical methods of analysis, such as ELISA and FPIA, is the most promising for solving this problem.
Thus, the aim of our work is to optimize the determination of mycotoxins by ELISA and FPIA in medical herbs. We selected antibodies and a tracer for the FPIA, then we constructed obtained a calibration curve and determined the sensitivity of this method for the samples in aqueous media. Then, the optimal sample preparation was elaborated: for spiked nuts samples were selected and prepared and a calibration curve was obtained with respect to the matrix effect. After that, the analysis of spiked nuts samples was performed by ELISA and FPIA and the sensitivity of both methods was compared.
The main drawback in radiometric imaging is the degradation of the spatial resolution with increasing depth, which results in blurred images for deeper lying structures. We circumvent this degradation with blind structured illumination, combined with a non-linear joint sparsity reconstruction algorithm. We demonstrate this by imaging a line pattern and a star-shaped structure through a metal sheet with a resolution four times better than the width of the thermal point-spread-function. The ground-breaking concept of super-resolution can be transferred from optics to diffusive imaging by defining a thermal point-spread-function similar to the Abbe limit for a certain optical wavelength.
The detection of cracks before the failure is highly significant when it comes to safety-relevant structures. Crack detection in metallic samples at high surface temperature is one of the challenging situation in manufacturing industries.
Laser thermography has already proved its detection capability of surface cracks in metallic samples at room temperature. In this work a continuous wave (CW) laser use to generate a laser, which is using to scan the metal surface with notch.
The corresponding heat distribution on the surface monitored using infrared thermal (IR) camera. A simplified 3D model for laser thermography is developed and validated with experimental results. A dedicated image processing algorithm developed to improve the detectability of the cracks. To understand the dependency of surface temperature, laser power, laser scanning speed etc. in defect detection, we carried out parametric studies with our validated model. Here we Report the capability of laser thermography in crack detection at elevated temperature.
Gegenstand der vorzustellenden Arbeiten ist die Prüfung der Umwelt-beständigkeit und -verträglichkeit von Materialien und Produkten hinsichtlich der Emission von potenziellen Schadstoffen in die Umwelt. Hierzu werden chemisch-physikalische Einflüsse (Bewitterung) und mikrobielle Beanspruchungen an Modellmaterialien evaluiert. So werden die Freisetzungsraten von Schadstoffen in Abhängigkeit der Beanspruchung beschrieben. Als Modellmaterialien kommen die Polymere Polystyrol (PS) und Polypropylen (PP) zum Einsatz. Synergistische Effekte der Bewitterungsparameter und der mikrobiologischen Beanspruchung sollen dabei ebenso betrachtet werden, wie die gezielte Alterung. Auch findet eine Beschreibung des Verhaltens der ausgetragenen Schadstoffe in den Umweltkompartimenten Boden oder Wasser statt. Hier sind mit Hilfe der zu entwickelnden Screening- und non-Target-Analyseverfahren die Transformation und der Metabolismus durch Mikroorganismen zu beschreiben. Aus den Ergebnissen sollen Korrelationen zwischen den künstlichen Alterungsverfahren und realen Szenarien abgeleitet werden.
The work presents the microstructure, chemical composition and mechanical properties of Inconel 686 coatings after high - temperature corrosion in environment of aggressive gases and ashes. To produce the Ni - based coatings the QS Nd:YAG laser cladding process was carried out. As the substrate used 13CrMo4-5 boilers plate steel. Ni - base alloys characterize the excellent high-temperature corrosion resistance, good strength and ability to work in aggressive environments. Formed clad were characterized by high quality of metallurgical bonding with the substrate material and sufficiently low amount of the iron close to the clad layer surface. After corrosion experiment the oxide scale on the substrate and clad created. The scale on 13CrMo4-5 steel had 70 μm thickness while the scale of the clad had less than 10 μm. The microstructure, chemical composition of the obtained clad and scales were investigated by scanning electron microscope (SEM) and electron probe microanalyzer (EPMA) equipped with the EDS detectors.
Anoxische Objektbehandlungen gegen Schädlingsbefall stellen eine besonders materialverträgliche und umweltschonende Alternative zu chemischen Bekämpfungsverfahren dar. Die Wirksamkeit hängt jedoch von der Dauer, dem Restsauerstoffgehalt sowie der Temperatur und Feuchtigkeit ab. Unterschiedliche Arten von Schadinsekten sind verschieden tolerant gegenüber sauerstoffarmen Bedingungen. Insbesondere bei Behandlungsbedingungen von unter 24° C und mehr als 0,5 % Restsauerstoffgehalt stößt das Verfahren an die Grenze der Zuverlässigkeit. In Versuchsreihen unter standardisierten Bedingungen erwiesen sich insgesamt 12 von 34 im Projekt untersuchte Insektenarten hierbei als widerstandsfähig. Davon als besonders tolerante Arten sind Hylotrupes bajulus, Lasioderma serricorne, Trogoderma parabile und Mezium affine zu nennen.
Mit dem entwickelten UWB-Sensoriksystem können durch elektromagnetische Wellen Bewegungen von Schädlingen innerhalb geschlossener Holzobjekte berührungslos erfasst werden. Voraussetzung für die eindeutige Detektion der Schädlingsaktivität ist neben der digitalen Signalverarbeitung zur Filterung und Extraktion des Nutzsignals, die genügend hohe Eindringtiefe, die ausreichend ist, wenn Schädlingsvorkommen häufig dicht unter der Holzoberfläche gelegen sind. Die Versuchsergebnisse haben gezeigt, dass bereits eine einzelne Trockenholztermite der Art Incisitermes marginipennis hinter einer 5 cm dicken Holzplatte aufgespürt werden kann. Somit erscheint das Messsystem potentiell für die Erfolgskontrolle einer Anoxia-Behandlung an Museumsgütern geeignet.
Our research focused on Inconel 625 weld overlays on 16Mo3 steel boiler pipes. The Investigation focused on the characterization of changes in the microstructure and chemical composition after annealing. The annealing was performed for ten hours at temperatures from 600 to 1000°C. Changes in the microstructure were observed with a scanning and transmission electron microscope (SEM and TEM). The investigation was supplemented by hardness measurements.
This work presents the non-destructive spectroscopic characterisation of original Dead Sea Scrolls (DSS)parchment fragments from Ronald Reed collection. The fragments are of paramount importance becausethey have never been subjected to any treatment of preservation and restoration, this allows to investi-gate the manufacturing method of real original Jewish parchments. The manufacture of “sacred” Jewishparchments, in fact, is traditionally supposed to use a superficial tannin treatment. To study the DSS frag-ments, it was necessary both to analyse mock-up samples, especially manufactured in order to reproduceancient Oriental Jewish ritual parchments, and to compare the results with those obtained in the analysisof modern and ancient Western Jewish ritual parchments, in order to test the effectiveness of the selectedspectroscopic techniques. Traditionally, the main difference between Oriental and Western traditionalparchment preparation consisted in the dehairing method: enzymatic for Oriental and lime-based forWestern. Moreover, a finishing treatment with tannin was supposed to be applied on ritual Jewish parch-ments. The need of reference samples derives from the knowledge that each parchment preparation,treatment and degradation can induce structural modifications that affect the spectral features. FourierTransform Infrared Spectroscopy by Attenuated Total Reflection (ATR-FTIR), FT-Raman and m-Ramanwere used in this study. The experimental results allowed us to recognise, with different sensitivity, thepresence of tannin by using m-Raman and IR spectroscopies and to prove that not all the archaeologicalsamples were manufactured in the same way with vegetal extracts. Many salts (tschermigite, dolomite,calcite, gypsum and iron carbonate) were found on the surface of DSS fragments. They can derive fromthe degradation processes and storage environment before the discovery or from the manufacture. More-over, the different sensitivities and instrumental characteristics of the used techniques permitted us toestablish an analytical protocol, useful for further studies of similar materials.
Organic/inorganic mixtures were prepared from ordinary Portland cement (OPC), water (w/c 0.22), a fluorescent dye in aqueous solution (stable at alkaline pH; BAM-I), and two different comb shape polycarboxylates (PCEs), i.e., high charge (PCE-HC) and low charge (PCE-LC), respectively. Rheological and calorimetric measurements were performed prior to optical studies in order to select PCE concentrations. Absorption and fluorescence spectroscopy of the system OPC + BAM-I (CBAM-I) revealed maxima of dye BAM-I located at 645 nm and 663 nm, respectively. In presence of PCE-HC and PCE-LC, these mixtures displayed a small red shift in reflectance and a faster decrease in intensity compared to studies with CBAM-I; however, only slight differences were observed between the different PCEs. With time, all systems exhibited a decrease in intensity of BAM-I in absorption/reflectance and emission. This could be caused by dye adsorption and possibly decomposition when in contact with cement particles or hydration products.
X-ray fluorescence analysis, due to its non-destructive nature and ist suitability to work with historic objects in situ, quickly became one of the most important methods for the evaluation of iron-gall ink. The main advantage of this qualitative and semi-quantitative method is that it makes it easy to differentiate between inks, based on the assumption that the differences result from the manufacture of the ink.
This work explores the question whether the ink ‘fingerprint’ results strictly from the elemental composition of the basic ink ingredients even if it is stored in vessels made of metals or metal alloys. In addition, we tested and compared the performance of three different XRF spectrometers.
We prepared various lab-grade inks according to historical ink recipes and measured the metal content of the ink deposited on sized cotton Linters paper with three types of XRF spectrometers: a simple hand-held device with an interaction spot of 4 mm and two devices equipped with poly-capillary Xray optics for line scanning and imaging. Since the exact elemental mass composition of the non-aged ink samples was known, we were able to evaluate the accuracy of the research procedure. Lab-grade inks were then aged in the metal jars imitating inkwells. The aging of the inks in the metal containers resulted in the significant change of the primary inks fingerprint as opposed to that of the control inks stored in glass containers. This effect was independently confirmed by the measurements conducted with every instrument we used. We will present a brief comparison of the results 43 achieved when using different spectrometers and a possible hypothesis explaining the processes that occurred.
In this work, a probabilistic framework for identification of traffic loads on concrete Bridge structures is presented using data from a FE structural model in combination with a finite volume approach for traffic load modelling. The identification approach uses Bayesian Inference to identify traffic loads from measured sensor data from travelling load experiments performed at BAM. The work focuses on the load identification part of the Framework utilizing global structural response measurements only. The obtained information on traffic loads can be forwarded to further analysis such as fatigue and structure state estimation or model updating.
Defining a Reduced Volume Zone for the Simulation of Burst Test on a Composite Pressure Vessels

The random nature of fibre break in composite materials has to be modelled in all part of the structure as it may contain millions of fibres. The reduced volume method was introduced to determine a smaller zone where we could characterise the strength based on the accumulation of fibre break, hence increasing the computation time
The developed model has certain limitations of the element size to be used in the simulation to characterise the strength of composite materials. A reduced volume method is proposed in order to reduce the number of degree of freedom of the finite element simulation.This study has revealed certain configuration to be followed to speed up the computation time.
During dynamic processes, a certain range of strain rates is often observed along loaded structures and components. For precise numerical simulations, it is necessary to determine rate-dependent properties in dynamic tests and to describe the material behavior correctly within an appropriate domain of strain rates including adiabatic heating effects at higher strain rates, typically higher than 10 1/s. In principle, numerical simulations are compared to experimental results to verify the applied material models. For dynamic tensile tests considering ductile materials and large plastic deformation beyond uniform elongation, it is challenging to obtain comparable results due to plastic instability and necking of the specimen, e.g..
Based on the strain gradient in a general tensile specimen, a theoretical criterion was derived describing the plastic instability in rate-dependent materials under isothermal conditions in. It was applied to different multiplicative and additive constitutive relations and the analytical onset of necking was compared to results from numerical calculations of quasi-static and dynamic tensile tests. The simulations of a sheet-metal specimen with rectangular cross-section were carried out using the Finite Element Method and it was found that the numerical calculated and the theoretical predicted onset of plastic instability agree very good. The analytical criterion for instability holds even for specimens without geometrical or material imperfections and confirms that the onset of plastic instability must be considered a material characteristic.
However, real dynamic problems with higher strain rates are not isothermal, the heat generated by plastic work is not dissipated to the surrounding and the temperature of the material increases significantly. Adiabatic heating and thermal softening must be considered within the constitutive relations of rate-dependent materials and the discussion of plastic instability. In this paper, an enhanced and more generalized approach for the description of the condition for stability is discussed and applied to phenomenological as well as more physical constitutive relations from the literature. This allows an individual assessment of the accuracy and verification of rate-dependent material models with respect to plastic instability.

The subject of this Special Issue is the fracture mechanics-based determination of the fatigue strength of weldments. Except for one, all papers were written in closer or wider relation to a methodology developed within the framework of the German Project cluster IBESS. Some of them provide background or supplementary information needed in that context but which is also relevant in a wider frame of research activities. The acronym IBESS stands for the topic of this Special Issue (in German: „Integrale Methode zu Bruchmechanischen Ermittlung der Schwingfestigkeit von Schweißverbindungen). Eight partners were involved. The cluster was cooperatively founded by the German Research Foundation (Deutsche Forschungsgemeinschaft) and by the German AiF Network (Arbeitsgemeinschaft industrieller Forschungsvereinigungen) for industrial research.
We examine the behavior of reinforced concrete components subjected to impact induced loading conditions which might be caused by vessels collisions such as aircraft fuel tanks The concrete plates were impact damaged at TU Dresden and shipped to BAM At BAM laminar tomography as the imaging method is used to determine and quantify the damage state An automatic crack detection method based on template matching is applied to find the cracks and we aim to develop a new method using machine learning Algorithms In addition numerical models are developed to understand the experiment and to predict the damage structures based on failure mechanisms.
Defining a reduced volume zone for the simulation of burst test on composite pressure vessels

The developed model uses a Monte-Carlo simulation to evaluate the accumulation of fibre breaks on composite structures. As the micromechanical aspects affecting the accumulation process is evaluated within a particular finite element size (0.1 mm x 0.1 mm x 8 mm), the industries would not be able to use the model on a real-size structure. Therefore, the reduced volume method is used to provide the statistical information between the required number of elements and simulations.
Fiber-reinforced-polymers (FRPs) are in current research focus in the lightweight construction industry, because of their extraordinary characteristics (stiffness and strength-to-density relation). The structure of polymer matrix and the interaction with reinforcement are crucial for optimization of the mechanical and thermal properties of FRPs. Due to the macromolecular chain structure, the mechanical properties of a polymer strongly vary with temperature: Below the glass transition, the chain segments of a polymer are “frozen”. Regarding fracture, the total changed energy during fracture if only dissipates for the generation of the new surfaces. However, in the region of the glass transition, the polymer chain segments start to get “unfrozen”, and the energy is not only required for generating new surfaces, but also for irreversibly deformation. This irreversible deformation is affected by the global temperature and the local temperature near the crack tip, which is affected by the local strain rate and crack propagation velocity.
Hence, in this research project, the irreversible deformation of neat and reinforced polymers will be controlled by changing the global temperature as well as the local temperature. With using different fracture experiments, the amount of energy required for creating new surfaces and for the irreversible deformation will be separated. The fracture tests include the conventional tensile test, the macroscopic peel test and the single fiber peel – off test.
A continuum damage model for concrete is developed with a focus on fatigue under compressive stresses. This includes the possibility to model stress redistributions and capture size effects. In contrast to cycle based approaches, where damage is accumulated based on the number of full stress cycles, a strain based approach is developed that can capture cyclic degradation under variable loading cycles including different amplitudes and loading frequencies. The model is designed to represent failure under static loading as a particular case of fatigue failure after a single loading cycle. As a consequence, most of the material parameters can be deduced from statictests. Only a limit set of additional constitutive parameters is required to accurately describe the evolution under fatigue loading. Another advantage of the proposed model is the possibility to directly incorporate other multi-physics effects such as creep and shrinkage or thermal loading on the constitutive level. A multiscale approach in time is presented to enable structural computations of fatigue failure with a reduced computational effort. The damage rate within the short time scale corresponding to a single cycle is computed based on a Fourier based approach. This evolution equation is then solved on the long time scale using different implicit and explicit time integration schemes. Their performance and some limitations for specific loading regimes is discussed.

X-ray scatter imaging is a well-established NDT technique to inspect complex objects using only a single-sided access. We present a specially designed multi-slit scatter camera consisting of several twisted slits which are parallelly arranged in a metal block. The camera projects one image per slit to the digital detector array, where the images are overlaying each other. The aperture is corrected based on a de-convolution algorithm to focus the overlaying projections into a single representation of the object. To achieve high scatter intensities from an object, it is necessary to optimize the parameters of the scatter system by simulation.
Microfluidic electrochemical immunosensor for the trace analysis of cocaine in water and body fluids

Quick but accurate testing and on‐the‐spot monitoring of cocaine in oral fluids and urine continues to be an important toxicological issue. In terms of drug testing, a number of devices have been introduced into the market in recent decades, notably for workplace inspection or roadside testing. However, these systems do not always fulfill the requirements in terms of reliability, especially when low cut‐off levels are required. With respect to surface water, the presence of anthropogenic small organic molecules such as prescription and over‐the‐counter pharmaceuticals as well as illicit drugs like cannabinoids, heroin, or cocaine, has become a challenge for scientists to develop new analytical tools for screening and on‐site analysis because many of them serve as markers for anthropogenic input and consumer behavior. Here, a modular approach for the detection of cocaine is presented, integrating an electrochemical enzyme‐linked immunosorbent assay (ELISA) performed on antibody‐grafted magnetic beads in a hybrid microfluidic sensor utilizing flexible tubing, static chip and screen‐printed electrode (SPE) elements for incubation, recognition, and cyclic voltammetry measurements. A linear response of the sensor vs. the logarithm of cocaine concentration was obtained with a limit of detection of 0.15 ng/L. Within an overall assay time of 25 minutes, concentrations down to 1 ng/L could be reliably determined in water, oral fluids, and urine, the system possessing a dynamic working range up to 1 mg/L.
Development of a robust calibration model for determination of nutrients in soils using EDXRF

As part of the BonaRes research initiative, funded by the BMBF, strategies are being developed to use soil as a sustainable resource in the bio economy. The interdisciplinary subproject I4S - “Intelligence for soil” - is responsible for the development of an integrated system for site-specific management of soil fertility. For this purpose, a platform is constructed and various sensors are installed. Real-time data will be summarised in models and decision-making algorithms will be used to control fertilisation and accordingly improve soil functions. Aim of the BAM in the frame of I4S is the characterisation of an energy-dispersive X-ray fluorescence (EDXRF) based sensor for robust determination of plant essential nutrients in soil.
First a principal component analysis (PCA) was used to identify outliers and to observe the largest variance within the German soil samples. It could be monitored that splitting of the samples was due to their iron content. Given that clay samples contain high amounts of iron and sandy samples low amounts, a classification of the samples by their soil texture according to VD LUFA was possible. Considering the complex composition of soil, a matrix-specific calibration was carried out by univariate and multivariate data analysis. The figures of merit demonstrated that a more robust calibration model with negligible matrix effects can be obtained by a multivariate approach using partial least squares regression (PLSR). A better correlation between predicted values compared to reference values for German soil samples was observed for the chemometric calibration model than for the univariate one.
Different factors can affect the received calibration models such as moisture and particle size distribution which is especially important due to later online Analysis.
In first studies the influence of moisture on the detection of plant essential nutrients was investigated. With increasing water content, the characteristic fluorescence peaks decrease and start to increase again at a water content of 15 %. With lower moisture content the soil agglomerates which leads to lower packing of the sample, resulting in a rougher surface which negatively influence the signals. Whereas, agglomerates are not formed at higher water content. This allows the sample to be packed more tightly thus a smoother surface and a better homogeneity is obtained.
Furthermore, particle size distribution leads to significantly higher uncertainties and lower signals when comparing grounded (< 500 μm) and not grounded (< 2 mm) samples. This can be explained by amplifying of the already known inhomogeneity of soils. Both factors must be included in the chemometric PLSR to obtain robust calibration models for each macro and micro nutrient.
Component and material investigations on metal seals for high level radioactive waste containers

Long-term investigations performed at BAM look to extend the state of knowledge on safety-related components of interim storage containers. Metal seals act as the primary sealing barrier in the bolted double lid closure system of the containers. The behaviour of metal seals has been investigated for ageing times up to 8.5 years and for various temperatures. The main cause for reduction in useable resilience overtime was due to creep deformation of the outer jacket of the seal.
