Aufgrund aktueller Leichtbaubestrebungen sowie Forderungen nach Energie- und Ressourceneffizienz werden zunehmend hochfeste Feinkornbaustähle mit Streckgrenzen ≥ 690 MPa in modernen Stahlkonstruktionen eingesetzt. Vorrangig werden diese mit Streckgrenzen bis 960 MPa verarbeitet und mittels MAG-Schweißprozessen gefügt. Moderne Invertertechnik ermöglichte jüngst zahlreiche Firmenentwicklungen (bspw. „DeepARC“, „forceArc“), die zu modifizierten Sprühlichtbögen (mod. SLB) mit ähnlichen Eigenschaften führten. Neben den bekannten wirtschaftlichen Vorteilen dieser mod. SLB, lassen gerade die reduzierten möglichen Nahtöffnungswinkel und die geringere Wärmeeinbringung eine positive Wirkung auf schweißbedingte Beanspruchungen erwarten. Erste Eigenspannungsanalysen bei vorangegangenen Schweißeignungsuntersuchen mit mod. SLB zeigten bereits bei freischrumpfenden Laborproben geringere Zugeigenspannungen in der Schweißnaht im Vergleich zu konventionell geschweißten Proben. Die betriebsfeste Auslegung von hochfesten Schweißkonstruktionen und die Ausnutzung hoher Streckgrenzen erfordern jedoch eine ausreichende Quantifizierung schweißbedingter Beanspruchungen der mod. SLB unter definierter, bauteilrelevanter Schrumpfbehinderung. Dies betrifft sowohl die Kräfte und Spannungen im lokalen Schweißnahtbereich als auch die Gesamtbeanspruchung sowie den Einfluss von Wärmeführung, Bauteilgeometrie, Einspanngrad und Nahtöffnungswinkel. Hierzu lagen in der Industrie, besonders bei den KMU, kaum Kenntnisse vor und sollten deshalb als Ziel dieses Forschungsprojektes erarbeitet werden.
Die vergleichenden Untersuchungen mit konventionellen Lichtbogen und mod. SLB mit angepasster Nahtkonfiguration erfolgten mithilfe von speziellen Prüfanlagen, mit der reale Bauteilsteifigkeiten abgebildet werden können. Bei beiden Prozessvarianten waren mit den gewählten Parametern Schweißnähte unter Erfüllung der Anforderungen an die mechanisch-technologischen Eigenschaften realisierbar. Bei den Analysen konnte eine signifikante Reduzierung der Eigenbeanspruchungen durch eine verminderte Wärmeeinbringung bei dem Einsatz einer Nahtkonfiguration mit abgesenktem Nahtöffnungswinkel beobachtet werden. Insbesondere erwiesen sich zur Absenkung mehrachsiger Beanspruchungszustände aufgrund von Biegemomenten in den bauteilnahen Schweißungen die engeren Nahtspalte als zielführend. Eine Vorstellung der erarbeiteten Kenntnisse und Empfehlungen in entsprechenden Gremien zur Normung erfolgten, um mittelfristig vor allem den KMU eine einfachere Konstruktionsauslegung und sichere Verarbeitung hochfester Feinkornbaustähle zu ermöglichen.
Mit einer normativen Absicherung kann Qualifizierungs- und Zulassungsaufwand eingespart werden, der aktuell beim Einsatz hochfester Stähle vielfach notwendig ist. Ferner werden die Betriebe durch die Ergebnisse in die Lage versetzt, die technischen und wirtschaftlichen Vorteile der mod. SLB-Prozesse auszunutzen und signifikante Einsparungen bei Schweißzusatz, Schweiß- und Rüstzeit sowie unter Berücksichtigung der Eigenbeanspruchung optimierte Ergebnisse hinsichtlich der Tragfähigkeit von Bauteilen zu erreichen und die Produktivität zu erhöhen.
Ausgehend von dem in der Literatur beschriebenen Verfahren zur Bestimmung aromatischer Aminosäuren konnte eine Analysemethode zur Proteinbestimmung über die optische Detektion von Tyrosin und Phenylalanin nach chromatographischer Trennung der Protein-Hydrolysate entwickelt werden, die gegenüber dem Verfahren aus der Literatur erhebliche Vorteile besitzt. Der Einsatz von Rundbodenvials stellt eine Neuerung und kostengünstige Alternative zu der konventionellen Hydrolyse in Vakuumhydrolyseröhrchen wie auch zu den Mikrowellenvials der Mikrowellenhydrolyse dar, die das Potential für Miniaturisierungen der Proben hat und sich zudem für große Probendurchsätze eignet. Anstelle der Mikrowellenheizungen wurde ein konventionelles Ölbad als Heizquelle verwendet. Somit ergibt sich eine Verbesserung auch darin, dass diese Methode geringere Geräteinvestitionen erfordert. Wie die Ergebnisse nahelegen, sind sowohl Salzsäure als auch Bromwasserstoffsäure geeignete Reagenzien, die bei der sauren Hydrolyse von Proteinen verwendet werden können. Mit Hilfe antioxidativer Zusätze können Nebenreaktionen erfolgreich vermieden werden. Hier haben sich L-Cystein und Oxalsäure als wirkungsvolle Additive herausgestellt. Wichtige Verbesserungen betreffen vor allem die Zeitersparnis bei der Hydrolyse, die Aufarbeitung der Hydrolysate und die chromatographischen Separationen. Erstere konnte gegenüber der konventionellen Methode von 22 Stunden auf 45 Minuten reduziert werden. Die chromatographische Trennung wurde so weit optimiert, dass die Laufzeit von 64 auf 35 Minuten verkürzt werden konnte. Zudem konnte auf das zeitraubende und kontaminationsanfällige Eindampfen der Hydrolysate verzichtet werden. Während der Aufarbeitung der Hydrolysate konnten die Menge eingesetzter Chemikalien reduziert und damit mögliche Störungen während der optischen Detektion verringert werden. Letztere wurde zum einen durch das Einbeziehen der UV-Absorption bei 260 nm erweitert, sodass eine Alternative zur Absorption bei 215 nm vorliegt, die ebenere Basislinien im Chromatogramm ermöglicht. Zum anderen wurde gezeigt, dass mit Hilfe der Fluoreszenz die Empfindlichkeit der Methode gegenüber der UV-Absorption erheblich verbessert werden kann. Anhand der Probe eines Birkenpollenextraktes ließ sich die Anwendbarkeit der optimierten Methode auf Realproben demonstrieren.
In this thesis a protocol based on a direct competitive and non-competitive immunoassay using the microarray format was developed to screen emulated hybridoma supernatants against the drug carbamazepine (CBZ). As support, epoxy slides were manufactured by coating microscope glass slides with epoxy silanes. The manufactured epoxy slides showed similar capacity for protein immobilization as commercially available NEXTERION slides, but showed a higher autofluorescence background. On the epoxy slides, protein A, Cys-A, G and Cys-G were immobilized as antibody capturing coating. It could be shown that the Cys-tag considerably increased the immobilization of all compared proteins, especially of protein G. Protein Cys-G immobilized at pH 8-9 was considered the most suitable protein for antibody immobilization due to good spot uniformity and a high binding capacity for IgG. Additionally, in immobilization experiments it could be shown, that 10-500 fold excess of bovine IgG, introduced by two different fetal bovine serum (FBS) products, showed no significant competition on the immobilization of mice or goat IgG on protein Cys-G. To examine the captured antibodies for affinity to CBZ, the CBZ-TOTA-DY654 tracer was custom synthesized. It could be demonstrated, that the tracer antibody interaction was competitive with the tracer precursor, CBZ-TOTA-NH2 and CBZ. Furthermore, the synthesized tracer was also successfully used by externals to evaluate CBZ affine hybriodoma cells in flow cytometry. In the simulated screening, five antibodies with known affinity to CBZ were diluted in a cell culture medium. The screening method allowed to differentiate between affine antibodies (IC50 < 20 µg/ml) and unaffine antibodies (IC50 > 20 µg/ml), when concentrations of 0.1 µg/ml IgG or greater have been used. Of five applied antibodies, all highly affine clones (CE2 and B3212M) were reported as positives while no false-positive samples were observed. The screening included competitive, with around 89500 fold excess of CBZ, and non-competitive tracer incubation on different segments of the same chip, which resulted in total signal suppression on the competitive segment. The screening was performed in 20 hours, only a few nanoliters, of each simulated supernatant, were consumed in the screening process, furthermore in the method standard 96 well MTPs were used and no cleanroom facilities were required.
Straßenverkehr stellt in Deutschland die stärkste Quelle für Lärmbelastungen dar. In 2014 wurde durch den Forschungsverbund Leiser Verkehr ein Bericht zur Situation der Lärmwirkungsforschung in Deutschland veröffentlicht, der den aktuellen Wissensstand zum Einfluss des Verkehrslärms auf die Lebensqualität und die Gesundheit der Bevölkerung dokumentiert. Verschiedene Studien belegen, dass Dauerbelastungen über 65 dB(A) die Betroffenen stark beeinträchtigen. So mindern die hohen Lärmbelastungen nicht nur die Lebensqualität, sondern sie können auch Herz-Kreislauf-Erkrankungen, Schlafstörungen, kognitive Leistungseinschränkungen und Tinnitus verursachen. Gerade wegen des stetig wachsenden Verkehrsaufkommens sind deshalb Strategien zu entwickeln, die die Bevölkerung noch wirksamer vor Straßenverkehrslärm schützen.
Aus dem Verkehrsforschungsprogramm der Bundesregierung wurden seit 2001 die Verbundprojekte Leiser Straßenverkehr gefördert. In 2014 wurde das dritte und letzte Verbundprojekt erfolgreich abgeschlossen.
Das Verbundprojekt Leiser Straßenverkehr 3 (LeiStra3) hatte sich als zentrales Ziel gesetzt, Maßnahmen zur Minderung des Straßenverkehrslärms in Ballungsräumen zu entwickeln, die dort aufgrund der hohen Bevölkerungsdichte besonders wirkungsvoll sind. Es wurden verschiedene Forschungsansätze verfolgt, die die Geräuschemission an der Lärmquelle nachhaltig reduzieren. Da die Geräuschemission des fließenden Verkehrs maßgeblich durch Wechselwirkungen an der Kontaktstelle von rollendem Reifen und Fahrbahn bestimmt wird, standen die Reifen-Fahrbahn-Geräusche im Fokus der Untersuchungen.
Ein Schlüsselelement in LeiStra3 war die Erweiterung des Simulationsmodells SPERoN zur quantitativen Analyse und Vorhersage der Reifen-Fahrbahn-Geräusche. Systematische Analysen von experimentell aufgenommenen Geräuschspektren mit dem Modell zeigten in LeiStra3, dass neben der Anregung von Reifenschwingungen aerodynamische Effekte im Reifen-Fahrbahn-Kontakt maßgeblich an der Geräuschentstehung beteiligt sein müssen. Um diese aerodynamischen Effekte eingehend charakterisieren und kritische Größen identifizieren zu können, mussten auf einem angemieteten Testgelände Modelloberflächen mit speziellen Texturmustern hergestellt werden, die die Entstehungsmechanismen des Rollgeräusches weitgehend getrennt und unabhängig voneinander anregen. Diese Projektstudien bildeten die Grundlage für die Entwicklung eines tragfähigen physikalischen Modells zur Beschreibung der aerodynamischen Effekte.
Im Ergebnis von LeiStra3 lassen sich mit SPERoN 2020 nun reale Fahrbahnoberflächen hinsichtlich ihres Einflusses auf die Reifen-Fahrbahn-Geräusche quantitativ analysieren und Designs für neuartige geräuschmindernde Fahrbahnbeläge zuverlässig synthetisieren. Darüber hinaus stellt das Modell ein leistungsfähiges Modul zur Reifenoptimierung zur Verfügung, mit dem der Einfluss der Profilierung und des Reifenaufbaus auf die Schallabstrahlung und den Rollwiderstand untersucht werden kann. Im Fokus stand dabei die Auswahl und Anwendung geeigneter numerischer Verfahren zur Verkürzung der Rechenzeiten, um ein praxistaugliches Optimierungstool bereit zu stellen. Dies ermöglicht eine effiziente Bestimmung von Schallabstrahlung und Rollwiderstand ohne aufwendigen Prototypenbau.
Im Regionalverkehr treten vielfach Beschleunigungs- und Abbremsvorgänge auf, zudem werden häufig enge Kurven durchfahren. Um die nötige Traktion zu gewährleisten, müssen die Antriebsachsreifen Profile mit einem hohen Anteil an Querrillen aufweisen. Diese Blockprofile neigen zu einer starken Schwingungsanregung auf der Fahrbahn und damit zu einer höheren Geräuschemission. In LeiStra3 wurden technologische Ansätze erarbeitet, um Rollgeräusche blockprofilierter Lkw-Reifen über die gesetzlichen Grenzwerte hinaus zu reduzieren. Im Straßenversuch hat eine Entwicklungsvariante einen Schalldruckpegel von 74,4 dB(A) erreicht. Gegenüber den aktuellen Produkten wurde damit eine Geräuschreduzierung von etwa 4 bis 5 dB(A) erzielt. Diese Variante wurde etwa ein Jahr nach Projektabschluss auf dem Markt eingeführt.
Darüber hinaus wurden in LeiStra3 schallabsorbierende Unterbodenverkleidungen realisiert, die das Rollgeräusch effizient absorbieren und damit den Vorbeifahrtpegel um bis zu 3 dB(A) mindern können. Mit Blick auf Elektrofahrzeuge wurden grundlegende Untersuchungen zur Reifen-Fahrbahn-Wechselwirkung unter Antriebsmoment und zur psychoakustischen Charakterisierung durchgeführt.
Einen weiteren Schwerpunkt in LeiStra3 stellte die Entwicklung leiserer Fahrbahnoberflächen dar. Modellrechnungen belegen, dass die Oberflächengestalt einen starken Einfluss auf die Schwingungsanregung des Reifens und die aerodynamischen Vorgänge in der Kontaktfläche zwischen Reifen und Fahrbahn hat. Aus akustischer Sicht ist es günstig, wenn alle Profilspitzen auf einer Ebene liegen und sich plateauartige Texturen mit schluchtenförmigen Vertiefungen ausbilden.
Die lärmtechnisch optimierte Asphaltdeckschicht AC D LOA stellte zu Projektbeginn eine neue Entwicklung in der Straßenbautechnik dar und greift diesen Ansatz auf. Die Deckschicht wurde auf Basis einer dichten Asphaltzusammensetzung konzipiert und zeichnet sich durch eine Korngrößenverteilung aus, die auf eine lärmtechnisch wirksame Oberflächentextur abgestimmt ist. Der erste Einbau von AC D LOA erfolgte 2007 in Düsseldorf im Bereich der kommunalen Straßen. In LeiStra3 wurde daran gearbeitet, die Verdichtungswilligkeit des Asphaltmischgutes zu verbessern. Auf diese Weise lassen sich die Dauerhaftigkeit der Deckschicht und damit ihr lärmtechnischer Nutzen erhöhen. Die Erkenntnisse flossen in die neuen „Empfehlungen für die Planung und Ausführung von lärmtechnisch optimierten Asphaltdeckschichten“ ein, die in 2015 von der Forschungsgesellschaft für Straßen- und Verkehrswesen herausgegeben wurden.
Der Gussasphalt mit offenporiger Oberfläche (engl.: Porous Mastic Asphalt, PMA) zeigt ebenfalls lärmmindernde Effekte. Im Vergleich zum herkömmlichen Gussasphalt enthält der PMA einen wesentlich höheren Anteil grober Gesteinskörnung. Es entsteht eine Deckschicht, die im unteren Bereich dicht und im oberen Bereich offenporig strukturiert ist. Die akustische Wirksamkeit ergibt sich aus der günstigen Oberflächentextur und einem hohen Hohlraumgehalt. In 2009 wurden erste Erprobungsstrecken gebaut, bei denen Schallpegelmessungen nach der Statistischen Vorbeifahrtmethode eine Lärmminderungen von bis zu 3 dB(A) gegenüber dem Referenzwert zeigten.
Auf Grundlage dieser Erkenntnisse wurden in LeiStra3 weiterführende Untersuchungen durchgeführt, um die Ausführungssicherheit des PMA zu erhöhen. Zu diesem Zweck wurde die Mischgutzusammensetzung optimiert und Laborverfahren zur Herstellung von praxisnahen Probekörpern für die Mischgutkonzeption entwickelt. Auf den Bundesautobahnen A 553 bei Brühl, A 3 bei Lohmar und A 5 bei Friedberg konnten Erprobungsstrecken mit verschiedenen PMA-Varianten hergestellt werden. Diese wurden hinsichtlich der Mischgut- und Einbauparameter untersucht.
Außerdem wurde in LeiStra3 untersucht, inwieweit der Herstellungsprozess von Elektroofenschlacke (EOS) verbessert werden kann, um bestimmte Kornformen und Korngrößen gezielt zu erzeugen und damit einen hochwertigen Ausgangswerkstoff für den Asphaltstraßenbau zu gewinnen. Im ersten Schritt wurden dazu EOS-Körnungen hinsichtlich ihrer bautechnischen Eignung geprüft und darauf aufbauend das Potenzial verschiedener Prozessrouten bewertet. Mit einer ausgewählten EOSKörnung (EloMinit®) konnten zwei Asphaltdeckschichtvarianten konzipiert und im Mai 2014 auf einem Testfeld eingebaut werden.
Die Nachgiebigkeit bestimmt neben der Oberflächentextur und der Offenporigkeit die akustischen Eigenschaften eines Fahrbahnbelags. Ein Ziel in LeiStra3 war es, Asphaltkonstruktionen durch die Nutzung der schwingungsdämpfenden Eigenschaften des Straßenoberbaus geräuschmindernd zu gestalten. Dabei wurden verschiedene Ansätze wie die Integration einer elastischen Dämpfungsschicht oder die Konzeption eines Asphaltmischgutes mit schwingungsdämpfenden Eigenschaften verfolgt.
Im Bereich Betonbauweisen stand die Herstellung einer Betondecke mit offenporiger Oberfläche (COPS) im Vordergrund. Die Grundidee des Systems COPS greift die im Asphaltstraßenbau bereits erfolgreich eingesetzte Mischgutkonzipierung mit hohem Grobkornanteil und einer feinkörnigen standfesten Bindemittel-/Mörtelphase auf. Während des Einbauvorganges sinkt der feinkörnige Mörtel nach unten ab, so dass innerhalb der oberen 10 mm der Deckschicht eine offenporige Oberfläche entsteht. In Laborversuchen wurde die Betonrezeptur so eingestellt, dass ein kontrolliertes Absetzverhalten des Zementleimes erreicht wird. Die Betonrezeptur muss möglichst stabil gegenüber Einflüssen von Witterung und Einbau sein, um die vorteilhaften akustischen Eigenschaften der fertigen Betondeckschicht sicherzustellen.
Im Verbundprojekt LeiStra3 haben Partner aus Wirtschaft und Wissenschaft in einer interdisziplinär angelegten Forschungsarbeit gemeinsam Lösungen erarbeitet, mit denen das Lärmminderungspotenzial von Reifen, Fahrzeug und Fahrbahn weiter ausgeschöpft werden kann. In allen Arbeitspaketen wurden zahlreiche Ergebnisse und Erkenntnisse gewonnen, die dazu beigetragen haben, die bestehende Technik zu verbessern, die Impulse zur Entwicklung neuer Technologien gesetzt haben und auf deren Basis das Technische Regelwerk fortgeschrieben wurde.
The aim of this work was the preparation of a novel stationary monolithic phase for affinity chromatography and HPLC-applications. Therefore, we have chosen porous glass filters that are available with different pore sizes as raw materials to prepare monolithic columns. We purchased VitraPOR 4 (10-16 μm pore size) and VitraPOR 5 (1.0-1.6 μm pore size) monolithic glass filters. The physical properties of these glass filters were characterized. The surface area, pore size distribution and the porosity were determined using mercury intrusion porosimetry and BET. These glass filters only exhibit flow through pores and therefore show no bimodal pore size distribution in the mercury intrusion curves. Due to their low permeability, the applied filters that exhibit an inner diameter of 8.0 mm and a length of 15.0±0.1 mm could be operated at flow rates more than 10 ml/min. High flow rates are favorable for fast separation experiments.
Das Ziel dieser Arbeit war es, die Vernetzung (Crosslinking) zwischen Protein G und einem Antikörper so durchzuführen, dass es zu keiner Modifikation der Antigen-Bindungsstelle des Antikörpers kommt. Zudem wurde im Rahmen dieser Arbeit ein Immunoassay entwickelt, mithilfe dessen zwanzig verschiedene Crosslinker in Hinblick auf ihre Fähigkeit Protein G zu funktionalisieren, untersucht werden konnten. Es konnten fünf Reagenzien identifiziert werden, die eine Derivatisierung von Protein G sowie eine anschließende Immobilisierung des Antikörpers auf dem Protein ermöglichen. Des Weiteren wurde gezeigt, dass mithilfe dieses Verfahrens sowohl die Immunpräzipitation als auch die massenspektrometrische Identifizierung und Charakterisierung von Proteinkomplexen erleichtert und verbessert werden kann.

The fine fraction of bottom ash from municipal solid waste incineration is characterized of both a high pollution and relevant resource potential. Methods for studying the environmental behaviour of solid residues are leaching tests. Various leaching test are adopted all over Europe with liquid to solid ratio of 2 or 10 l/kg. In Germany three different leaching Tests are valid with the commencement of the draft of the ordinance of reuse of mineral waste as alternative building ordinance. Comparing two batch tests, only the concentration for chloride is comparable. To evaluate the metal resource potential in the sand fraction wet chemical Methods have been applied. Methodical problems were caused mainly by the material heterogeneity and especially for precious metals due to their low concentration around 1 mg/kg. Other Methods have been available, but hardly applicable.
Charakterisierung von Glas-Polymer Verbundmaterialien fur die Hochdruckspeicherung von Wasserstoff

In der vorliegenden Arbeit werden Glaskapillaren und Strukturen aus Borosilikatglas (Simax) f¨ur die Hochdruckspeicherung von Wasserstoff betrachtet. Um die mechanischen Belastungen zu analysieren und anschließend ein optimiertes Speicherdesign abzuleiten, findet die Finite Element Methode (FEM) Einsatz.
Die notwendigen Materialkennwerte werden zuvor experimentell ermittelt. Dazu geh¨oren die Festigkeit, die Schubfestigkeit der Grenzfl¨ache Klebung/Glas, der Wasserstoffpermeationskoeffizient, das Temperatur-Zeit-abh¨angige Verhalten, sowie die obere Temperatureinsatzgrenze.
Die Glasfestigkeit ist stark abh¨angig von der Oberfl¨acheng¨ute. Daher werden in dieser Arbeit Kapillaren selber gezogen und beschichtet. Der Einfluss auf die Festigkeit wird mit Hilfe von Berstversuchen untersucht. Die eingesetzten Polymere werden zus¨atzlich mit dem Push-out Versuch, der dynamisch mechanischen Analyse und der Permeationsmessung charakterisiert.
Dar¨uber hinaus wird aus den DMA Versuchen ein viskoelastisches Materialmodell abgeleitet, womit der Temperatur- und Zeiteinfluss auf die mechanische Belastung simuliert wird.
Unter Beachtung der experimentell gewonnenen Erkenntnisse, sowie der durchgef¨uhrten Simulation ist es m¨oglich, ein optimiertes Speicherdesign zu erstellen und Aussagen ¨uber die Einsatzgrenzen zu treffen.
