Ergot alkaloids are a group of toxic compounds, formed by fungi on infested grasses. In 2022, the European Commission set into effect maximum levels for the sum of the twelve major ergot alkaloids in multiple foods. To facilitate the laborious and costly individual quantification of the twelve major ergot alkaloids by HPLC–MS/MS or -FLD, we recently reported a sum parameter method (SPM) for ergot alkaloid quantification. Here, derivatization to lysergic acid hydrazide—a derivative of the mutual ergoline backbone in all ergot alkaloids—allowed simplified determination of all ergot alkaloids in flour via HPLC-FLD. For the measurement of more complex matrices like processed foods, we now developed a MS/MS-based SPM. Two internal standards (IS), isotopically labelled at different positions of the molecule, were synthesized and employed in the MS/MS-measurements. Method performance using either the 13CD3-labelled or the 15N2-labelled IS was evaluated on naturally contaminated rye and wheat flour samples as well as on processed food matrices. Employing the 13CD3-labelled IS leads to lower variances and better consistency with the reference data (obtained by the FLD-based SPM) in flour samples compared to the 15N2-labelled IS. The novel method significantly improves the measurement of ergot alkaloids in complex food matrices, due to their increased selectivity and thus lower interferences. Furthermore, the application of isotope labelled IS obviates the need for time-consuming steps like the determination of recovery rate based, matrix specific correction factors as described in the MS/MS-based European standard method for ergot alkaloid quantification (EN 17425).
Leguminosen wie z.B. Bohnen, Erbsen, Soja und zunehmend auch die Samen der Süßlupine liefern aufgrund ihres hohen Proteingehaltes einen wertvollen Beitrag zu einer veganen oder vegetarischen Ernährung. Jedoch sind Lupinen anfällig für Infektionen mit dem Pilz Diaporthe toxica, der u.a. Phomopsin A als hepatotoxischen Sekundärmetaboliten bildet. Ziel der Arbeiten waren die Gewinnung und Anwendung eines 15N-isotopenmarkierten internen Standards für die HPLC-MS/MS-Analyse dieses Toxins sowie die Quantifizierung der Toxinbildung in einem „worst case“-Szenario.
Fungal secondary metabolites such as phomopsin A (PHO-A) produced by the fungus Diaporthe toxica are referred to as “emerging” mycotoxins. Recently, we developed a biosynthetic approach to gain access to 15N-isotopically labelled PHO-A as internal standard (IS) for PHO-A matrix independent quantification of PHO-A in various commodities. Aim of the present work was to elucidate the applicability of the new IS for accurate quantification and to investigate the phomopsin formation on various legume seeds as well as plant material. In summary, the applicability of the SIDA-HPLC-MS/MS method could be proven and the potential of D. toxica to produce high amounts of phomopsins under unfavorable conditions on lupins and other legume seeds could be shown. Moreover, a novel toxic methylated PHO-A derivative was unveiled in natural samples infested with D. toxica.
Zur Bestimmung von Arzneimittelrückständen in Oberflächengewässern wird häufig die LC-MS/MS als Methode der Wahl eingesetzt. Obwohl sich dieses Nachweisverfahren durch eine hohe Empfindlichkeit und Selektivität auszeichnet, gibt es vor allem bei geringen Substanzkonzentrationen häufig störende Einflüsse durch Matrixeffekte. Diese Matrixeffekte können unterschiedliche Ursachen haben.
Einige Matrixeffekte werden am Beispiel des Wirkstoffs Carbamazepin erläutert. Carbamazepin wird durch Kläranlagen nur partiell aus dem Abwasser entfernt und ist im Spurenbereich auch noch im Trinkwasser nachzuweisen. Wenn für die LC-MS/MS wasserbasierte Anteile in der mobilen Phase genutzt werden, befinden sich trotz Verwendung einer Labor-Reinstwasseranlage in der mobilen Phase noch kleinste Konzentrationen von Carbamazepin, welche einen Blindwert im unteren ppt-Bereich verursachen.
Weitere Matrixeffekte treten durch die unterschiedliche Qualität der Oberflächenwasserproben auf. Koelutionen von weiteren Inhaltstoffen zeitgleich mit dem Zielanalyten führen zu einer Diskriminierung des Meßsignals. Verbindungen, die den gleichen Massenübergang bei nahezu identischer Retentionszeit zeigen, erfordern eine reproduzierbare chromatographische Vortrennung oder die Wahl einer alternativen HPLC-Säule.
Three strategies to sample volatile organic compounds (VOC) from lung cancer cell lines cultured in vitro were compared. Headspace solid phase microextraction was applied in situ to culture flasks and alternatively to subsamples of headspace gas or to nutrient solution subsamples followed by gas chromatography–mass spectrometry. The direct quantification of 55 VOC in the headspace of cell cultures was validated and is discussed with respect to reproducibility and system-related interferences. The role of the VOC background from culture media and usually employed polystyrene culture vessels is examined and was seen to invoke potentially misleading conclusions. The commercial A549 and two further adenocarcinoma cell lines displayed largely similar VOC profiles with distinct differences regarding certain individual substances. There is evidence for the inappropriateness of the standard cell culturing methods in the search for volatile cancer markers.
Two different hair reference materials, one produced from authentic hair displaying an ethyl glucuronide (EtG) content of about 25 pg/mg and one obtained by fortification of blank hair to an EtG level of 85 pg/mg were submitted to accelerated aging between 4 degrees C and 60 degrees C for periods between one and 24 months. Subsequently, the EtG content was determined in the aged samples and untreated reference samples stored at -22 degrees C under repeatability conditions following the so-called isochronous approach. The EtG content remained stable even at 40 degrees C for 24 months and at 60 degrees C over six months. This is in contrast to many organic analytes contained in trace concentrations in diverse matrices. A slight but significant increase of the recovered EtG in case of authentic hair samples having been exposed for 24 months between 4 degrees C and 60 degrees C may be due to a temperature-driven process that allows increased recoveries of the physiologically embedded EtG.
Cylindrospermopsin (CYN) is a cyanobacterial toxin associated with human and animal poisonings. Due to its toxicity in combination with its widespread occurrence, the development of reliable methods for selective, sensitive detection and accurate quantification is mandatory. Liquid chromatography tandem mass spectrometry (LC-MS/MS) analysis using stable isotope dilution analysis (SIDA) represents an ideal tool for this purpose. U-[15N5]-CYN was synthesized by culturing Aphanizomenon flos-aquae in Na15NO3-containing cyanobacteria growth medium followed by a cleanup using graphitized carbon black columns and mass spectrometric characterization. Subsequently, a SIDA-LC-MS/MS method for the quantification of CYN in freshwater and Brassica matrices was developed showing satisfactory performance data. The recovery ranged between 98 and 103 %; the limit of quantification was 15 ng/L in freshwater and 50 µg/kg dry weight in Brassica samples. The novel SIDA was applied for CYN determination in real freshwater samples as well as in kale and in vegetable mustard exposed to toxin-containing irrigation water. Two of the freshwater samples taken from German lakes were found to be CYN-contaminated above limit of quantification (17.9 and 60.8 ng/L). CYN is systemically available to the examined vegetable species after exposure of the rootstock leading to CYN mass fractions in kale and vegetable mustard leaves of 15.0 µg/kg fresh weight and 23.9 µg/kg fresh weight, respectively. CYN measurements in both matrices are exemplary for the versatile applicability of the developed method in environmental analysis.
Pure U-[13C18]-labelled cis-zearalenone (cis-ZEA) has been prepared and characterised as internal standard (ISTD) for a reliable quantification of cis-ZEA in contaminated food and feed products. The cis-isomer of the naturally trans-configurated Fusarium mycotoxin zearalenone is often neglected. However, isomerisation easily occurs by exposure of ZEA to (UV-)light. Thus, the applicability of the new cis-ZEA ISTD was demonstrated in a long-term isomerisation study comparing naturally trans-ZEA-contaminated edible oil with spiked edible oil. To estimate the benefits of the newly prepared cis-ZEA ISTD, various approaches to quantify cis-ZEA by high performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS) were compared. As a result, a significant bias was revealed if no appropriate cis-ZEA standards are used. Furthermore, the new ISTD was applied to the analysis of 15 edible oils by stable isotope dilution analysis in combination with HPLC-electrospray ionisation-MS/MS. One of the maize germ oils showed the presence of cis-ZEA above LOD (>0.3 µg/kg), whereas two out of 15 maize germ oils were found to be contaminated with trans-ZEA (range 17.0-31.0 µg/kg).
The fungus Claviceps purpurea grows on grasses
and cereal grains and produces six predominant ergot
alkaloids. These toxic substances undergo different transformation
reactions during storage and cereal processing. One
of these reactions is the addition of water to a double bond in
the ergoline skeleton. Since light is required for this process,
the substances formed were named lumi-ergot alkaloids.
From these, a new asymmetric carbon and consequently two
epimers with different polarities are formed. For investigations
of lumi-ergot alkaloids, ergometrine was used exemplarily
as it represents one of the six ergot alkaloids
predominantly formed by Claviceps purpurea. The main
reaction product, the less polar compound of the two lumiergometrine
epimers, was separated by HPLC and unambiguously
identified as 10-(S)-lumi-ergometrine using X-ray
structural analysis. A HPLC-MS/MS method was developed
for the detection of this substance in sclerotia extracts. Using
this method, the existence of both epimeric forms of lumiergometrine
could be proved in the sclerotia. This is the first
time that the existence of a lumi-transformation product of
ergot alkaloids was proved in naturally grown samples.
Hexabromocyclododecane enantiomers: microsomal degradation and patterns of hydroxylated metabolites

The degradation of the enantiomers of α-, β-, and γ-hexabromocyclododecane (HBCD) by phase I metabolism was investigated using induced rat liver microsomes. HBCD isomers were quantified using HPLC-MS/MS (ESI-) after separation on a combination of a reversed phase and a chiral analytical column. The degradation of all six isomers followed first-order kinetics and the estimated half-lives ranged from 6.3 min for both β-HBCD enantiomers to 32.3 min in case of (+)-γ-HBCD. (+)-α- and (–)-γ-HBCD displayed significantly shorter half-lives than their corresponding antipodes. It could be shown that this degradation led to a significant enrichment of the first eluting enantiomers (–)-α- and (+)-γ-HBCD. Individual patterns of mono- and dihydroxylated derivatives obtained from each α- and γ-HBCD enantiomer were seen to be distinctly characteristic. The patterns of monohydroxylated HBCD derivatives detected in liver and muscle tissues of pollack, mackerel and in herring gull eggs were largely similar to those observed in the in vitro experiments with rat liver microsomes. This enabled individual hydroxy-HBCDs to be assigned to their respective parent HBCD enantiomers.
