We report on the use of a sprayable and thermogelating biomaterial (Poloxamer; a.k.a. Pluronic) in optical imaging of pH values, local oxygen and temperature. The material is highly biocompatible and easy to handle. We also show that the material is well permeable to oxygen (thus making it a good choice for use in oxygen sensors), and is stable in liquid solution and at elevated temperature. We demonstrate its applicability in optical sensors for oxygen, pH and temperature. This was accomplished by incorporating appropriate luminescent probes in various kinds of microparticles (which act as hosts for the probes and prevent dye leaching and aggregation), and then dispersing the microparticles in the thermogelating polymer. The resulting sensor gels were deposited on the surface of interest via spraying at temperatures of <20 °C. At these temperatures, the gels adhere well to the target, even on uneven surfaces such as skin, wounds, and bacterial cultures. If temperature is risen to above 25 °C, the gels form a thin and soft but solid sensing layer which, however, can be simply removed from surface of interest by cooling and wiping it off, or by washing with water. Sprayable thermogelating sensors present obvious advantages over other sensors by not causing damage to the surface of interest. In our perception, the sensing materials also have wide further applicability in sensors for other species including clinically relevant gases, enzyme substrates (such as glucose or lactate) and ions.
The effect of the presence of Fe(III) during the cultivation on the electrochemical activity and corrosion behaviour of dissimilatory iron reducing bacteria Shewanella putrefaciens was studied by means of ex situ and in situ X-ray absorption near-edge spectroscopy (XANES). Stainless steel AISI 304 and thin iron films were studied as substrates. XANES analysis indicated an accelerated iron dissolution and growth of an oxide/hydroxide film for the culture grown with Fe(III) in comparison to the culture grown in absence of Fe(III). Electrochemical Analysis indicated that the biofilm resulted in acceleration of the general corrosion but provides protection against local corrosion.
Acetylenflaschen im Feuer

Acetylen ist eines der wichtigsten technischen Gase und findet insbesondere Anwendung beim Schweißen und Schneiden verschiedener Werkstoffe. Damit Acetylen in entsprechenden Mengen vor Ort zur Verfügung gestellt werden kann, wird es in Druckgasbehälter abgefüllt. Im Falle eines Brandes stehen Acetylenflaschen im Verdacht, jederzeit – auch nach Löschen des Feuers – aufgrund einer möglicherweise initiierten Zerfallsreaktion zu bersten. Um insbesondere Personenschäden zu vermeiden, evakuieren und sperren die Rettungskräfte einen Sicherheitsbereich mit einem Radius von mindestens 200 m um die verdächtige Flasche ab. Gleichzeitig wird umgehend mit der Kühlung der Flasche begonnen. Wie lange muss aber eine Acetylenflasche gekühlt werden, bis sie sicher abtransportiert werden kann, und wie lange muss der Sicherheitsbereich mit dem großen Radius aufrecht erhalten werden? Diesen Fragen ist die BAM in den vergangenen Jahren im Rahmen verschiedener Untersuchungen und Projekte nachgegangen.
Cementitious materials are frequently applied in environments in which they are exposed to acid attack, e.g., in sewer systems, biogas plants, and agricultural/food-related industries. Alkali-activated materials (AAMs) have repeatedly been shown to exhibit a remarkably high resistance against attack by organic and inorganic acids and, thus, are promising candidates for the construction and the repair of acid-exposed structures. However, the reaction mechanisms and processes affecting the acid resistance of AAMs have just recently begun to be understood in more detail. The present contribution synthesises these advances and outlines potentially fruitful avenues of research. The interaction between AAMs and acids proceeds in a multistep process wherein different aspects of deterioration extend to different depths, complicating the overall determination of acid resistance. Partly due to this indistinct definition of the ‘depth of corrosion’, the effects of the composition of AAMs on their acid resistance cannot be unambiguously identified to date. Important parallels exist between the deterioration of low-Ca AAMs and the weathering/corrosion of minerals and glasses (dissolution-reprecipitation mechanism). Additional research requirements relate to the deterioration mechanism of high-Ca AAMs; how the character of the corroded layer influences the rate of deterioration; the effects of shrinkage and the bond between AAMs and substrates.
The halophilic γ-proteobacterium Halomonas elongata DSM 2581T thrives at salt concentrations well above 10 % NaCl (1.7 M NaCl). A well-known osmoregulatory mechanism is the accumulation of the compatible solute ectoine within the cell in response to osmotic stress. While ectoine accumulation is central to osmoregulation and promotes resistance to high salinity in halophilic bacteria, ectoine has this effect only to a much lesser extent in non-halophiles. We carried out transcriptome analysis of H. elongata grown on two different carbon sources (acetate or glucose), and low (0.17 M NaCl), medium (1 M), and high salinity (2 M) to identify additional mechanisms for adaptation to high saline environments. To avoid a methodological bias, the transcripts were evaluated by applying two methods, DESeq2 and Transcripts Per Million (TPM). The differentially transcribed genes in response to the available carbon sources and salt stress were then compared to the transcriptome profile of Chromohalobacter salexigens, a closely related moderate halophilic bacterium. Transcriptome profiling supports the notion that glucose is degraded via the cytoplasmic Entner-Doudoroff pathway, whereas the Embden-Meyerhoff-Parnas pathway is employed for gluconeogenesis. The machinery of oxidative phosphorylation in H. elongata and C. salexigens differs greatly from that of non-halophilic organisms, and electron flow can occur from quinone to oxygen along four alternative routes. Two of these pathways via cytochrome bo' and cytochrome bd quinol oxidases seem to be upregulated in salt stressed cells. Among the most highly regulated genes in H. elongata and C. salexigens are those encoding chemotaxis and motility proteins, with genes for chemotaxis and flagellar assembly severely downregulated at low salt concentrations. We also compared transcripts at low and high-salt stress (low growth rate) with transcripts at optimal salt concentration and found that the majority of regulated genes were down-regulated in stressed cells, including many genes involved in carbohydrate metabolism, while ribosome synthesis was up-regulated, which is in contrast to what is known from non-halophiles at slow growth. Finally, comparing the acidity of the cytoplasmic proteomes of non-halophiles, extreme halophiles and moderate halophiles suggests adaptation to an increased cytoplasmic ion concentration of H. elongata. Taken together, these results lead us to propose a model for salt tolerance in H. elongata where ion accumulation plays a greater role in salt tolerance than previously assumed.
For decades, the low-strain impact integrity testing using a hammer blow is well established as a method of quality assurance for various pile types. However, this method has its limitations. Our research and development focuses on improving the excitation signal using a shaker system in contrast to the standard hammer method. Another approach is to increase the amount of sensors used during testing. The purpose is to identify the direction of wave propagation which gives advantages under difficult conditions, such as piles below structures.
Pile integrity testing (PIT) using a shaker system was performed on two 11 m long piles of 90 cm in diameter. While one pile was intact, the other one showed a flaw at approx. 3.5 m below pile top, which was confirmed by standard PIT in 2012. A logarithmic sweep between 500 Hz and 1 KHz of 0.1 s was used as the input signal, being vertically injected into the pile. Prior to that, simulations on similar pile geometries showed that the depth of the pile toe as well as flaws within the pile can be extracted by applying regularized deconvolution. The result is the impulse response in the time domain.
The application of deconvolution on the measured signals shows that it is possible to identify the pile length but it is more difficult to clearly extract the flaw’s position in the pile. Additional digital signal processing techniques and the improvement of the regularized deconvolution method as well as the experimental setup need to be investigated.
Another way to improve the PIT method is to use a multichannel sensor arrangement. By arranging several accelerometers vertically along the accessible part of the pile shaft, it is possible to distinguish between downward and upward traveling waves. Furthermore, it is possible to estimate the unknown wave speed, which gives the possibility of more accurate pile length calculations. The method was evaluated successfully during a measurement campaign of a slab foundation with subjacent piles. In 20 of 28 cases the pile length could be detected accurately.
Four donor-acceptor boron difluoride complexes based on the carbazole electron donor and the [1,3,5,2]oxadiazaborinino[3,4-a][1,8]naphthyridine acceptor were designed, synthesized, and systematically spectroscopically investigated in solutions, in the solid states, and dye-doped polymer films. The dyes exhibit an intense blue to red solid-state emission with photoluminescence quantum yields of up to 56% in pure dye samples and 86% in poly(methyl methacrylate) films. All boron complexes show aggregation-induced emission and reversible mechanofluorochromism. The optical properties of these dyes and their solid state luminescence can be tuned by substitution pattern, i.e., the substituents at the naphthyridine unit. Exchange of CH3- for CF3-groups does not only increase the intramolecular charge transfer character, but also provides a crystallization-induced emission enhancement.
This chapter aims to give a brief introduction into the field of alkali activated materials (AAM) and their physicochemical material behavior when exposed to sulfuric acid and in biochemically aggressive sewer environments. Additionally, the differences in material behavior within chemical (abiotic) and microbially induced acid environments are highlighted. Finally, existing data on overall performance of AAM, compared to conventional concrete-based building materials in sewer systems will be discussed.
