T1 - Application of an ELISA to the quantification of carbamazepine in ground, surface and wastewaters and validation with LC-MS/MS N2 - Carbamazepine is a psychiatric pharmaceutical widely detected in aquatic environments. Due to its generalized occurrence and environmental persistence it might be considered as an anthropogenic pollution indicator. In this research, a previously developed enzyme-linked immunosorbent assay (ELISA), based on a commercial monoclonal antibody, was applied to the quantification of carbamazepine in ground, surface and wastewaters and results were validated by liquid chromatography–tandem mass spectrometry (LC–MS/MS). The performance of the applied ELISA methodology was tested in the presence of high concentrations of sodium chloride and dissolved organic matter. The method was not significantly affected by matrix effects, being adequate for the quantification of carbamazepine in environmental samples, even without sample pre-treatment. This method allows the quantification of carbamazepine in the range of 0.03–10 µg L-1, with a relative error lower than 30%. Due to a pH dependent cross-reactivity with cetirizine, an antihistaminic drug, the assay also enabled the quantification of cetirizine in the samples. The application of the developed method to the quantification of carbamazepine was performed by using environmental samples with very different matrices, collected in the geographical area of Ria de Aveiro, an estuarine system located in the North of Portugal. Carbamazepine was detected in all analyzed wastewater samples and in one surface water with concentrations between 0.1 and 0.7 µg L-1. Validation with LC–MS/MS revealed that results obtained by ELISA are 2–28% overestimated, which was considered highly satisfactory due to the absence of sample pre-treatments. T1 - Phase equilibria and partitioning of L-histidine and three pharmaceuticals to pH-adjusted high-pressure liquid phases of the ternary system (ethene + water + 2-propanol) N2 - Adding some salt to a homogeneous aqueous liquid solution of an organic solvent often results in a liquid–liquid phase split. However, such a phase split can also be achieved by charging such a liquid with a gas, in particular when the temperature is close to the critical temperature of that specific gas. This phenomenon is called 'salting out by a near-critical gas'. It might be applied in a high-pressure extraction process, for example, to separate and recover valuable biomolecules from aqueous phases. Using a neutral gas like, for example, ethene for pressurizing additionally allows to adjust the pH of the coexisting liquid phases and to influence the partitioning of biomolecules when they change their electric net charge with the pH of the solution. The design of such separation processes requires not only reliable information on the phase forming system, that is, the ternary system (near-critical gas + water + organic solvent), but also on the partitioning of typical solutes to the coexisting phases. The present publication reports data (from an experimental study with a static-analytical device) for the partitioning of four biomolecules, that is, L-histidine, Aspirin, cimetidine, and 4-dimethylaminoantipyrine (at nearly infinite dilution) to coexisting liquid phases of the high-pressure three-phase liquid–liquid–vapor (L1L2V) equilibrium of the ternary system (ethene + water + 2-propanol) at (293 and 333) K and pressures from about (5.5 to 17) MPa. The coexisting liquid phases are characterized by distinctly different compositions, the aqueous phase being more hydrophilic than the alkanol-rich phase. Moreover, electrolytes were additionally added to adjust the pH conditions in the liquid phases. The pH-dependent dissociation equilibrium and the related net charge of the biomolecules primarily determine the partitioning behavior: The pH effect is stronger than the impact of varying pressure or temperature. For example, a switch from basic to acidic conditions can invert the partitioning, if that switch at the same time effects a change in the net charge of the solute, for example, from an ionic to a neutral molecule (or vice versa). The ionic solute is more hydrophilic (and thus prefers the aqueous phase), whereas the neutral or zwitterionic solute is less hydrophilic (i.e., more lipophilic) and consequently prefers the propanol-rich liquid phase. Through the consumption of contaminated foods, they can cause acute or chronic intoxications in humans. Here, it is demonstrated how covalent hydrazine chemistry can be used to improve the performance of instrumental methods for the quantification of trace level food mycotoxins. In the case of the Alternaria mycotoxin tenuazonic acid, pre-column derivatisation with 2,4-dinitrophenylhydrazine resolved chromatographic issues due to the chemical properties of the analyte and allowed for its rapid, sensitive and selective quantification in cereals and beer by high performance liquid chromatography- ion-trap two stage mass spectrometry (HPLC-IT-MS2). Tenuazonic acid could be detected for the first time in beer and buckwheat flour. Although the encountered levels were too low to cause acute intoxications, the frequency of contamination indicated possible health risks due to chronic exposure. In a second scenario, dynamic covalent hydrazine chemistry (DCHC) was exploited for a novel extraction and cleanup method applicable to the Fusarium mycotoxin zearalenone occurring in edible oils. Zearalenone was extracted by hydrazone formation on a hydrazinefunctionalised polymer resin and subsequently released hydrolytically for quantification by HPLC-fluorescence detection (HPLC- FLD). The high selectivity of the approach allowed for the omission of MS detection and immunoaffinity cleanup. The DCHC method was superior to previously published methods in terms of handling efforts, cost, precision and selectivity and is well suited for the monitoring of the current European maximum level for zearalenone in refined maize oil. In the second part of the dissertation, possible degradation routes of Alternaria mycotoxins upon storage and bread baking are discussed. In the frame of a kinetic study, it was shown that tenuazonic acid is degraded by two parallel processes, deacetylation and epimerisation, when stored in aqueous solution (half-life at 25 °C ~ 74 days). The primary degradation product deacetyl tenuazonic acid was less stable than its parent compound and degraded rapidly in beverage matrices. In model baking experiments it was furthermore revealed that alternariol, alternariol monomethyl ether and altenuene are stable under typical baking conditions. A newly identified degradation route, which is based on a sequence of hydrolysis and decarboxylation, caused only minor substance losses (< 1 %). Still, the degradation products could be detected in commercial rusk and crispbread by HPLC-tandem mass spectrometry (HPLC-MS/MS). N2 - Mykotoxine sind sekundäre Stoffwechselprodukte ubiquitärer Schimmelpilze. Über den Konsum belasteter Nahrungsmittel können sie beim Menschen, je nach Art und Ausmaß der Exposition, akute bzw. chronische Vergiftungen hervorrufen. Ziel der Arbeiten war es, durch chemische Methoden zwei Probleme aus dem Bereich der quantitativen Mykotoxinanalytik zu lösen. Hierbei ging es erstens um die Inkompatibilität des Alternaria Mykotoxins Tenuazonsäure mit üblichen Hochleistungsflüssigkeitschromatographie (HPLC) Säulen und zweitens um die selektive analytische Extraktion des in der Europäischen Union regulierten Fusarium Mykotoxins Zearalenon aus Speiseölen. Für beide Problemstellungen wurden Lösungen erarbeitet, die auf dem Einsatz kovalenter Hydrazinchemie im Rahmen der Probenvorbereitung beruhen. Die Verfahren wurden validiert und auf Verbraucherprodukte angewendet. Die Bestimmung der Tenuazonsäure erfolgte dabei nach Derivatisierung mit 2,4-Dinitrophenylhydrazin mittels HPLC-Ion-trap Massenspektrometrie (HPLC-IT-MS2). Für Zearalenon wurde eine neuartige Festphasenextraktionsmethode basierend auf dynamischer kovalenter Hydrazinchemie (DCHC) entwickelt und in Verbindung mit HPLC-Fluoreszenzdetektion (HPLC-FLD) eingesetzt. Beide Ansätze zeichnen sich durch hohe Selektivität, einfaches Handling und einen geringen Lösungsmittelverbrauch aus; alle Reaktionen erfolgen bei Raumtemperatur. So konnte Tenuazonsäure erstmals in Getränken (Bier) und Buchweizenmehl nachgewiesen werden. Der zweite Teil der Arbeit beschäftigt sich mit möglichen Abbaureaktionen der Alternaria Mykotoxine bei der Lagerung und Zubereitung von Lebensmitteln. Im Rahmen einer kinetischen Studie wurde gezeigt, dass Tenuazonsäure in wässriger Lösung über zwei parallele Prozesse, hydrolytische Deacetylierung und Epimerisierung, abgebaut wird (Halbwertszeit bei 25 °C ~ 74 Tage). Während das vornehmlich gebildete Produkt, Deacetyl-Tenuazonsäure, in wässriger Lösung stabil war, wurde es in Getränkematrices innerhalb weniger Tage abgebaut. In Modell-Backversuchen wurde weiterhin die Stabilität der Dibenzo- -pyron Derivate Alternariol, Alternariol-monomethylether und Altenuen untersucht. Unter typischen Backbedingungen erwiesen sich die Verbindungen als stabil. Ein erstmals belegter Abbaumechanismus für Alternariol und Alternariol-monomethylether, der auf einer Sequenz aus Hydrolyse und Decarboxylierung beruht, führte nur zu geringen Substanzverlusten (< 1 %). Dennoch konnten die Abbauprodukte mittels HPLC-Tandem-Massenspektrometrie (HPLCMS/MS) in extrudierten Produkten (Knäckebrot und Zwieback) nachgewiesen werden. The aim of this work was the construction of novel SERS hybrid probes. The hybrid probes consisted of Au and Ag nanoparticles and reporter molecules, as well as a targeting unit. The concept for the SERS hybrid probe design was followed by experiments comprising characterization techniques such as UV/Vis- spectroscopy (UV/Vis), Transmission electron microscopy (TEM) and Dynamic Light Scattering (DLS), respectively. SERS experiments were per- formed for studying and optimizing the plasmonic properties of nanoparticles with respect to their enhancement capabilities. The SERS-probes had to meet following requirements: biocompatibility, stability in physiological media, and enhancement of Raman-signals from Raman reporter molecules enabling the identification of different probes even in a complex biological environment. Au and Ag nanoaggregates were found to be the most appropriate SERS substrates for the hybrid probe design. The utilization of Raman reporters enabled the identification of different SERS probes in multiplexing experiments. In particular, the multiplexing capability of ten various reporter molecules para-aminobenzenethiol, 2-naphthalenethiol, crystal violet, rhodamine (B) isothiocyanate, fiuorescein isothiocyanate, 5,5'dithiobis(2-nitrobenzoic acid), para- mercaptobenzoic acid, acridine orange, safranine O und nile blue was studied using NIR-SERS excitation. As demonstrated by the results the reporters could be identified through their specific Raman signature even in the case of high structural similarity. Chemical separation analysis of the reporter signatures was performed in a trivariate approach, enabling the discrimination through an automated calculation of specific band ratios. The trivariate identification could be a promising method for SERS-multiplexing in analytical applications. Multivariate methods such as Principal Components Analysis (PCA) and Hierarchical Cluster Analysis (HCA) were as well applied for discrimination and imaging of the reporter signatures. With the help of multivariate imaging methods based on cluster analysis, it could be for the first time demonstrated that such methods provided a fast identification of various SERS hybrid probes inside the biological matrix, this was demonstrated using living 3T3 cells. Further, in a duplex imaging approach, the probes fulfill the requirements for the sensitive detection of both the specific reporter signatures and intrinsic information coming from eukaryotic cells. The results of cluster methods and principal components approaches for discrimination indicate that fast, multivariate evaluation of whole sets of multiple probes is feasible, beyond the visual inspection of individual spectra that has been practiced so far. This suggests multiplexing applications with SERS hybrid nanoprobes and SERS tags in very high density sensing and biological imaging applications, where fast read-out is required. The pH-sensitivity of SERS-Tags that consisted of different reporter molecules attached to aggregated Au and Ag nanoparticles in the range between pH∼3-10 was studied. It could be demonstrated that the reporter molecules provided pH-dependent SERS signatures and could therefore be suitable for the sensitive pH-detection, e.g., inside cellular compartments. TFE belongs to the group of chemically instable gases which are able to decompose under specific conditions releasing a huge amount of heat comparable to a gas explosion of flammable gases. Due to several incidents in PTFE – production plants the investigation of the safety related properties of TFE at elevated condition was necessary and resulted in a research project at BAM which is subsidized by the PlasticsEurope association. In a first stage the pressure dependence of the Minimum Ignition Temperature for Decomposition (MITD) was determined in the range of 0.5 MPa - 3 MPa in small autoclaves with volumes of 0.2 dm³ and 3 dm³. These results were used to validate the numerical model and were found to correlate quite well with the simulated values. Nevertheless the applicability had to be confirmed for larger volumes as well. Therefore a heat able 100-dm³-autoclave was constructed and build to determine the MITD in the range of 0.5 MPa to 1.1 MPa. The ballasted track is modelled in detail by the finite element method. The infinite soil is modelled by the boundary element method as a homogeneous or layered half-space. The track-soil system is coupled with a simple rigid mass model of the vehicle so that the vehicle-track interaction is completely included. Transfer functions are calculated in frequency domain without and with vehicle-track interaction, the compliance of the track and the mobilities of the soil at different distances from the track. Finally, the ratios between the ground vibration amplitudes with and without mitigation measure are calculated to quantify the effectiveness of the mitigation measure. Tracks with under sleeper pads have been investigated in a wide parameter study. The main parameters that influence the reduction of ground vibration are the stiffness of the under sleeper pad, the mass and the width of the sleeper. The softest sleeper pad yields the best reduction of the ground vibration. The influence of the sleeper mass is not as strong as the characteristic frequency is ruled by the mass of the sleeper and the mass of the wheelset as well. The influence of other parameters has been examined. The stiffness of the rail pads, the bending stiffness of the track, the stiffness of the ballast, the sub-soil, and the soil, and the layering of the soil. All these parameters show no or only a minor influence on the mitigation effect. As the standard isolated track, a track with an under sleeper pad of a stiffness of kS = 5 107 N/m has been chosen, which can also be expressed as a stiffness per area of kS ’’ = 7.4 107 N/m3 = 0.074 N/mm3. The resonance frequency for this pad stiffness is observed between 32 and 40 Hz. The reduction of the ground vibration is about vi,I /vi,U = 0.2 and 0.1 at 100 Hz. The reduction can be improved by softer sleeper pads. But the compliance of the track gets higher than tolerable. A possibility to use softer sleeper pads without increasing the static compliance of the track is their combination with wide sleepers. The softer sleeper pads under wider sleepers yield the same compliance of the track and a better reduction of the ground vibration. Durch den anhaltenden Trend zum Leichtbau und den immer höheren Anforderungen an die passive Sicherheit kommt es hier zu einem verstärkten Einsatz innovativer hochfester Stähle. Obwohl es sich beim Punktschweißen um ein lange erprobtes Verfahren handelt, das sich anhand der Prozessparameter und regelmäßiger zerstörender Prüfung überwachen lässt, ist durch diese Entwicklung die Suche nach geeigneten zerstörungsfreien Prüfverfahren wieder aktuell. Wegen der hohen Produktionsgeschwindigkeiten in der automatisierten Fertigung muss sich das Prüfverfahren für eine hundertprozentige Prüfung vollautomatisch mit der typischen Taktrate von etwa einem Hertz durchführen lassen. In dieser Arbeit wird zuerst die prinzipielle Durchführbarkeit der thermografischen Prüfung von Punktschweißverbindungen an speziell präparierten Proben überprüft. Auf dieser Grundlage wird eine Serienuntersuchung an 75 Proben, die sich auf drei Gruppen von Schweißparametern aufteilen und neben optimal geschweißten Proben typische Fehlerbilder repräsentieren, eine Serienuntersuchung gezeigt. Hierzu wird eine Blitzlichtanregung in Transmissionsanordnung verwendet. Es kann gezeigt werden, dass sich mit einer geeigneten Datenaufbereitung die einzelnen Gruppen sicher trennen lassen. Durch eine statistische Bewertung der Ergebnisse kann unter Verwendung eines angepassten Gütekriteriums die Trennschärfe noch deutlich erhöht werden. Die Anregung wird über einen Diodenlaser mit einem optischen Scanner realisiert. Bei der Auswertung kommt ein effizienter und robuster Algorithmus zum Einsatz, der direkt auf die mit der Kamera aufgenommene Thermografiesequenz angewendet werden kann, ohne auf eine Synchronisation zwischen Kamera und Lasersystem angewiesen zu sein. Daher eignet sich diese Herangehensweise besonders zur Nachrüstung bestehender Thermografiesysteme. Die prinzipielle Eignung dieser Methode wird anhand einer Messung an einem Schienenstück mit Rollkontaktermüdungsrissen gezeigt. Als Referenzverfahren werden die Magnetpulverprüfung und eine auf GMR (Giant Magnetoresistance)- Sensoren basierende Streufeldmessung herangezogen. Mit dem thermografischen Verfahren konnten sämtliche enthaltenen Fehler erkannt werden. Dabei wurden Risse im mm-Abstand getrennt nachgewiesen, was etwa der geometrischen Auflösung der eingesetzten Kamera entspricht. The selectivity of this reaction and the yield in primary amino groups was found to be low. Here, a prominent side-reactions of this process was investigated, the hydrogenation by NH3 plasma. For identification of hydrogenation ammonia (NH3) and deuterated ammonia (ND3) were exposed to Polyethylene (h-PE) and fully deuterated polyethylene (d-PE) as well as hexatriacontane (h-HTC) and fully deuterated hexatriacontane (d-HTC) as low-molecular weight model for PE. H–D exchange was assumed and detected by Time-of-Flight Secondary Ion Mass Spectrometry (ToF–SIMS), X-ray Photoelectron Spectroscopy (XPS), Attenuated Total Reflectance (ATR-FTIR) and 1H and 2H Nuclear magnetic Resonance. Results show a significant H–D exchange within the sampling depth of ATR (2.5 µm). However, N- and NH2 introduction was limited to the topmost surface as shown by SIMS and XPS (a few nanometers). A1 - Unger, Wolfgang T1 - Ultra-shallow chemical characterization of organic thin films deposited by plasma and vacuum-ultraviolet, using angle- and excitation energy-resolved XPS N2 - Nitrogen (N)-rich organic thin films were deposited using both low-pressure plasma- and vacuum-ultraviolet-based techniques, from mixtures of ammonia (NH3) and ethylene (C2H4). These films were investigated using angle-resolved and excitation energy resolved X-ray photoelectron spectroscopy (ARXPS and ERXPS, respectively) in order to determine their sub-surface chemical profiles. These two techniques enable one to tune the “XPS 95%” information depth, z 95%, by varying either the angle or the excitation energy. Using a combination of both techniques, z 95% can be varied continuously from 0.7 to 11 nm. The surface-near chemistry is investigated using both high-resolution C 1s spectra and elemental concentrations derived from elemental peak intensities. Results show that while laboratory XPS, and even ARXPS, suggest homogenous surface chemistries, the novel combination of ARXPS and ERXPS points to the existence of a compositional profile in the extreme outer surface layer. Our conclusions are supported by simulations using SESSA software. T1 - Alloys for gas sensors investigated using the continuous gradient-high throughput screening macroscope CG-HTSM N2 - Ternary alloys have been studied as the gate metal of chemical sensors based on the field effect in silicon. The investigation of reduced poisoning of hydrogen sensors by H2S is presented in the alloy system PdxNiyCo1-x-y. A new method in high throughput screening of alloys is applied. Not singular sample compositions but a continuous gradient in alloy concentrations was produced on a semiconductor sample. A laser scanning system produces a photo current in the semiconductor to map the sensor signal and provide a quasi continuous alloy characterization. The new system is therefore called Continuous Gradient—High Throughput Screening Macroscope (CG-HTSM). Typically 625 alloys are characterized in 15 minutes. The resolution of our set-up allows distinguishing 100.000 alloy compositions. Static secondary ion mass spectrometry is an appropriate tool to determine hydrogen isotopes as elements and in fragment ions with high sensitivity and selectivity. Specifically the exchange of hydrogen isotopes and the incorporation of N-containing moieties in the near-surface layer of the polyolefins have been studied in ND3 plasma experiments with conventional polypropylene (h-PP) and polyethylene (h-PE) and NH3 plasma experiments with deuterated polyethylene (d-PE). Considering the exchange of hydrogen between the plasma and the polymer surface studied by using deuterated ammonia and polyolefin samples the conclusion has been derived that polypropylene with its side chain methyl groups is more susceptible to hydrogen exchange reactions. For ND3 plasma treatment of polyethylene and polypropylene similar N-containing fragments were obtained and the measured semi-quantitative ToF-SIMS N-uptake data are rather similar. The observation of a wide range of characteristic mixed CkNlHmDn+ secondary fragment ions suggests complex and manifold reaction pathways at the polymer–plasma interface besides simple grafting of –ND2 or –NH2 moieties formed by ammonia fragmentation in the plasma. Finally, indications of an isotopic effect for hydrogen isotopes in the plasma process have been observed by comparison of ND3/h-PE results with those of NH3/d-PE. A1 - Unger, Wolfgang T1 - Adlayers of dimannoside thiols on gold: surface chemical analysis N2 - Carbohydrate films on gold based on dimannoside thiols (DMT) were prepared, and a complementary surface chemical analysis was performed in detail by X-ray photoelectron spectroscopy (XPS), time-of-flight secondary ion mass spectrometry (ToF-SIMS), near-edge X-ray absorption fine structure (NEXAFS), FT-IR, and contact angle measurements in order to verify formation of ω-carbohydrate-functionalized alkylthiol films. XPS (C 1s, O 1s, and S 2p) reveals information on carbohydrate specific alkoxy (C–O) and acetal moieties (O–C–O) as well as thiolate species attached to gold. Angle-resolved synchrotron XPS was used for chemical speciation at ultimate surface sensitivity. Angle-resolved XPS analysis suggests the presence of an excess top layer composed of unbound sulfur components combined with alkyl moieties. Further support for DMT attachment on Au is given by ToF-SIMS and FT-IR analysis. Carbon and oxygen K-edge NEXAFS spectra were interpreted by applying the building block model supported by comparison to data of 1-undecanethiol, poly(vinyl alcohol), and polyoxymethylene. No linear dichroism effect was observed in the angle-resolved C K-edge NEXAFS. Eine uantitative Beurteilung der nicht immer zugänglichen Rückwand eines Bauteils (Minderdicken) ist durch Inversion thermografischer Messdaten möglich. Diese quantitativen Informationen können durch iterative Methoden vom Gauß-Newton-Typ (Inversion) gewonnen werden. Als reale Prüfobjekte werden ca. 2 cm dicke PVC-Platten mit örtlich variierender Rückwandgeometrie untersucht. Die Erwärmung erfolgt mit Blitzlampen, welche die PVC-Platte kurzzeitig um mehrere K erwärmt. Ein wichtiger Teil der Inversion ist die Simulation des Experiments, welche mit Finite-Elemente-Methoden (FEM) in 2D realisiert wird.
