T1 - Evidence for an allosteric mechanism of substrate release from membrane-transporter accessory binding proteins N2 - Numerous membrane importers rely on accessory water-soluble proteins to capture their substrates. These substrate-binding Proteins (SBP) have a strong affinity for their ligands; yet, Substrate release onto the low-affinity membrane transporter must occur for uptake to proceed. It is generally accepted that release is facilitated by the association of SBP and transporter, upon which the SBP adopts a conformation similar to the unliganded state, whose affinity is sufficiently reduced. Despite the appeal of this mechanism, however, direct supporting evidence is lacking. Here, we use experimental and theoretical methods to demonstrate that an allosteric mechanism of enhanced substrate release is indeed plausible. First, we report the atomic-resolution structure of APO TeaA, the SBP of the Na!-coupled ectoine TRAP transporter TeaBC from Halomonas elongata DSM2581T, and compare it with the substrate-bound structure previously reported. Conformational freeenergy landscape calculations based upon molecular Dynamics simulations are then used to dissect the mechanism that couples ectoine binding to structural change in TeaA. These insights allow us to design a triple mutation that biases TeaA toward apo-like conformations without directly perturbing the binding cleft, thus mimicking the influence of the membrane transporter. Calorimetric measurements demonstrate that the ectoine affinity of the conformationally biased triple mutant is 100-fold weaker than that of the wild type. By contrast, a control mutant predicted to be conformationally unbiased displays wild-type affinity. This work thus demonstrates that substrate release from SBPs onto their Membrane transporters can be facilitated by the latter through a mechanism of allosteric modulation of the former. Im derzeitigen Erkenntnisstand zum Materialverhalten des Betons gibt es gerade bei dessen Ermüdungsverhalten noch viele nicht oder nicht ausreichend geklärte Fragestellungen. Deshalb wurde in Laborversuchen das Ermüdungsverhalten von Beton unter Druckschwellbeanspruchungen untersucht. Im Fokus dieser Untersuchungen standen dabei die während der Lebensdauer im Materialgefüge ablaufenden Prozesse, die zur Schädigung des Gefüges und schließlich auch zum Ermüdungsversagen führen können. In den Ermüdungsversuchen wurden versuchsbegleitend Dehnungsmessungen, Ultraschall-Laufzeitmessungen und Schallemissionsmessungen an den Proben zur Erfassung der Schädigungsevolution eingesetzt. Die Schallemissionsmessung ist dabei ein wichtiges Instrument, da sie die Rissentwicklungsprozesse detektiert, die die Ursache für die Materialschädigung sind. Mit ihr konnten die unterschiedlichen Phasen der Rissentwicklungen über die gesamte Lebensdauer der Betonproben festgestellt werden. Diese Phasen sind gekennzeichnet durch sich stabilisierende, stabile und instabile Rissprozesse, die entsprechende Auswirkungen auf das Materialverhalten haben. In einem Forschungsprojekt werden derzeit Möglichkeiten zur Verwertung von heterogenen mineralischen Bau- und Abbruchabfällen durch Herstellung von Blähgranulaten mit maßgeschneiderten Eigenschaften (Schüttdichte, Rohdichte, Korngröße und Kornfestigkeit) untersucht. Dies eröffnet die Möglichkeit einer rohstofflichen Verwertung solcher Abfälle. Im Mittelpunkt der Untersuchungen steht der Mauerwerkbruch. Er fällt mit über 20 Tonnen pro Jahr an und weist im Vergleich zum Betonbruch größere Verwertungsdefizite auf hl. Die erzeugten Granulate sollen als leichte Gesteinskörnung im Beton, Schüttung oder Pflanzsubstrat Anwendung finden. Die ersten Ergebnisse der Granulatherstellung mit einer thermischen Behandlung zeigen, dass sich Mauerwerkbruch als Rohstoff zur Herstellung leichter Gesteinskörnungen eignet 121. Die so erzeugten Blähgranulate weisen ähnliche Eigenschaften wie herkömmliche Blähtöne auf 131. Das Hauptziel der in diesem Beitrag vorgestellten weiteren Untersuchungen war, die Rohdichte der Granulate bei gleichbleibender oder steigender Kornfestigkeit zu senken. Aus energetischer Sicht des Blähprozesses wurde neben dem thermischen Verfahren auch die Möglichkeit einer hydrothermalen Erhärtung der Granulate untersucht. The Variation of the friction coefficient with sliding speed depends on the structure and Constitution of the base oil, the viscosity index, the surface topography of the liner and ring, and the friction modifiers used. Novel organic friction modifiers of low molecular mass not only reduce the friction coefficients under mixed / boundary lubrication, but also in the fluid film regime. In addition, the influence of surface roughness on the wettability of surfaces can be significant for both static and dynamic wetting. For this reason, the interaction of the lubricant with the surfaces was also investigated to enable a correlation between the surface properties of the liquid and solid phases and the tribological behavior. With new upcoming techniques and research topics in the last decades, such as provenance and authenticity of food, the number of published isotope data strongly increased. The development of inductively coupled plasma mass spectrometers (ICPMS) from an instrument for simple quantitative analysis to highly sophisticated isotope abundance ratio machines influenced this process significantly. While in former times only experts in mass spectrometry were able to produce reproducible isotope data, nowadays many laboratories, never been in touch with mass spectrometry before, produce isotope data with an ICPMS. Especially for such user isotope reference materials (IRM) are indispensible to enable a reliable method validation. The fast development and the broad availability of ICPMS also lead to an expansion of the classical research areas and new elements are under investigation. Here all users require IRM to correct for mass fractionation or mass discrimination or at least to enable isotope data related to a common accepted basis. Part 2: Influence of crossing angle N2 - The wear behaviour of thin steel wires has been analyzed under oscillating sliding conditions in crossed cylinders contact geometry. The focus of this analysis was the influence of the crossing angle between the wires on the wear. The wires used had 0.45 mm in diameter and the material was cold-drawn eutectoid carbon steel (0.8% C) with a tensile strength higher than 2800 MPa. Two different types of tests were carried out, the first one representing the influence of the crossing angle for a constant load and the second one representing the influence of the crossing angle with constant contact pressure. In the first type of tests it was seen that as the contact angle decreases the contact pressure decreases too and hence less energy specific wear resistance is observed. As a consequence less wear is produced, thus increasing the life of the wires. In the second type of tests it was seen that with constant contact pressure but different crossing angles, nearly the same energy specific wear resistance was observed. This points at an identical wear behaviour in both type of tests but with a running-in and a steady state period as two different wear periods. The tests showed that the running in period may play an important role in the overall wear particle generation and hence the wear occurring in the steady state period is rather mild. Das konventionellen SAFT oder FT-SAFT-Verfahren liefern in Verbindung mit einer Phasenauswertung einen schnellen und sicheren Hinweis darauf, dass ein Lufteinschluss/Defekt vorliegt. Darüber hinaus können jedoch Rückausbreitungs-Verfahren, wenn ein gutes Geschwindigkeitsprofil vorliegt, eine wesentlich bessere Lokalisierung des Defekts bieten als SAFT-Verfahren, jedoch mit dem Nachteil, dass sie sehr zeit- und rechenintensiv sind, vor allem im Dreidimensionalen. Als Mittelweg kann hierzu die "One-Way"-Methode dienen. Diese Methode verwendet Pseudo-Differential-Operatoren und ist sehr robust auch bei relativ groben Geschwindigkeitsprofilen. Sie wird an Beispielen aus der Bauwerkdiagnose mit den konventionellen Verfahren verglichen. This goes beyond the regular application of conventional radar systems offering only qualitative structural information. The new approach provides a fast NDT method for the classification of ballast fouling. A main advantage of the multi-offset radar is that regular ballast digging for velocity evaluation can be avoided and travel time can be directly transformed into an equivalent depth by using the automatically evaluated velocity. Fast monitoring of ballast fouling condition can reduce maintenance cost significantly by supporting the works necessary for ballast cleaning and furthermore this method reduces interruption time for a better railway service. T1 - Mapping local microstructure and mechanical performance around carbon nanotube grafted silica fibres: Methodologies for hierarchical composites N2 - The introduction of carbon nanotubes (CNTs) modifies bulk polymer properties, depending on intrinsic quality, dispersion, alignment, interfacial chemistry and mechanical properties of the nanofiller. These effects can be exploited to enhance the matrices of conventional microscale fibre-reinforced polymer composites, by using primary reinforcing fibres grafted with CNTs. This paper presents a methodology that combines atomic force microscopy, polarised Raman spectroscopy, and nanoindentation techniques, to study the distribution, alignment and orientation of CNTs in the vicinity of epoxy-embedded micrometre-scale silica fibres, as well as, the resulting local mechanical properties of the matrix. Raman maps of key features in the CNT spectra clearly show the CNT distribution and orientation, including a ‘parted’ morphology associated with long grafted CNTs. The hardness and indentation modulus of the epoxy matrix were improved locally by 28% and 24%, respectively, due to the reinforcing effects of CNTs. Moreover, a slower stress relaxation was observed in the epoxy region containing CNTs, which may be due to restricted molecular mobility of the matrix. The proposed methodology is likely to be relevant to further studies of nanocomposites and hierarchical composites. The dielectric constant and the temperature stability of the dielectric constant are strongly correlated with the composition of the ZnO-B2O3-Li2O additives. A high dielectric constant up to 3000 and a dielectric loss less than 0.024 were measured on multilayer capacitors sintered at 900 °C with silver inner electrodes. Due to its high infrared refractive index, average pore sizes up to 1 µm for optimal Mie scattering cross sections and over 35% porosity the material is a promising candidate for elevated temperature insulations. The essentially in large pore growth is achieved by graphitic nano and micro particle additives. The microstructure parameters of porosity, specific surface area, median and average pore sizes are determined by mercury intrusion porosimetry. Beyond this classical technique supplementary measurements by X-ray Absorption and X-ray refractometry reveal considerably larger average pore sizes due to closed pores not accessible for mercury. An additional advantage of the X-ray techniques is their non-destructive application, which allows other treatments afterwards. The combined application of both methods provides additional information and reliability in case of complex ceramic microstructures and may serve for improved developments of porous ceramics with optimised thermal scattering for energy savings in high temperature applications. T1 - Selenium speciation in different organs of African catfish (Clarias gariepinus) enriched through a selenium-enriched garlic based diet N2 - Speciation of Se in fish is needed to elucidate the metabolism of this element in living organisms in the marine environment. In this paper, selenium concentration and its species distribution in several organs and tissues (liver, gills, kidney, muscle and gastrointestinal tract) of African catfish fed with a selenium-enriched garlic based diet was studied. The intention of this paper is focused on both the investigation of selenium distribution in the soluble protein fraction and the detection of selenoaminoacids. Thus, two different procedures have been developed. In the first procedure, screening of selenium in proteins in the Tris-buffer soluble fraction of different tissues was carried out by size exclusion chromatography-inductively coupled plasma-mass spectrometry (SEC-ICP-MS) and by laser ablation-inductively coupled plasma-mass spectrometry (LA-ICP-MS) after sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) separation and electroblotting onto membranes. For the amino acid analysis, several sample treatments for Se-species extraction, based on enzymatic hydrolysis, were compared. The best results were obtained for incubation at controlled temperature. Application of several sample treatments in conjunction with different chromatographic techniques (reverse phase, anion exchange and ion exchange/size exclusion) was crucial to unambiguous Se-species identification. In Se-enriched African catfish a noticeable increase in the content of selenium in different organs was observed, except for the liver, where the Se level remained unaltered. The kidney was the Se-target organ in animals fed with enriched Se food. Selenomethionine (SeMet) was the main Se species identified in fillet extracts, whereas the presence of selenocysteine (SeCys) was detected in the liver and both SeMet and SeCys were present in the kidney. Para-hexaphenylene (p6P) nanofibers emit polarized light with a highly anisotropic emission profile, act a sub-wavelength optical waveguides, and form the basis tor a new type of miniature OLEDs. However, p6P molecules suffer reduction of luminescence intensity during photoexcitation under UV light. Such photoreaction is understood as a consequence of the interaction of the fibers with oxygen. We have developed a bilayer coating that does not change significantly the p6P spectrum but strongly reduces bleaching. This bilayer coating consists of a first layer of a stable polymer (PMMA) on top of the organic nanofibers as a protecting layer tor avoiding modifications of the p6P luminescence spectrum and as a second layer SiOx tor blocking the oxygen penetration. In this work, we show the applicability of this bilayer coating of PMMA/SiOx in prolonging the operationallifetime of field-effect transistor (FET) devices based on p6P nanofibers. The coating prevents strang degradation of transferred nanofibers on FET platforms during electrical biasing as is observed tor uncoated devices. A novel phosphorous compound (DOPI) was used. Its action is dependent on the epoxy matrix. DGEBA/DMC and DOPI decompose independently of each other. Only flame inhibition occurs in the gas phase. RTM6 shows flame inhibition and a condensed phase interaction increasing charring. Both mechanisms decrease with increasing irradiance, whereas in RTM6-CF charring is suppressed at low ones. RTM6+DOPI shows a higher LOI (34.2%) than DGEBA/DMC+DOPI and a V-0 classification in UL 94. Adding CF only enhances the LOI, DOPI+CF leads to a superposition in LOI for DGEBA/DMC-CF+DOPI (31.8%, V-0) and a synergism for RTM6-CF+DOPI (47.7%, V-0). T1 - Investigation of process-induced strains development by fibre Bragg grating sensors in resin transfer moulded composites N2 - A comprehensive understanding of the development of residual strains in composite processing is essential to manufacture high quality composite parts. In this paper, the use of fibre Bragg grating (FBG) optical sensors was investigated to measure in situ the build-up of the process-induced strains in composite panels manufactured by resin transfer moulding. The FBG sensors, embedded in the composite laminate, successfully measured the evolution of the composite in-plane strains due to the temperature history. The sensors also captured a strain discontinuity during the cool-down related to the debonding of the composite from the mould. Finite element models were then proposed to simulate the strain development measured by the FBG sensors. Three different contact interactions between the tool and the laminate were investigated: no bonding, perfect bonding and frictional contact.
