As a result of continuous and prolific use, mainly in aviation firefighting foams, thousands of industrial and military installations have been found to contain contaminated soil, groundwater and surface water. As a result of the perpetual use of PFAS containing products, effluents and sewage sludge from wastewater treatment plants (WWTPs) have been observed to be an important pathway for PFAS into the environment. In Germany, phosphorus and other nutrients from sewage sludge and wastewater should be recycled in WWTPs of cities with a large population. However, it is not clear if PFAS contamination from wastewater and sewage sludge end up in novel wastewater-based fertilizers. Normally, PFAS are analyzed using PFAS protocols typically with liquid chromatography tandem mass spectrometry (LC-MS/MS) quantification. To get a better overview of the amount of “total PFAS,” we applied sum parameter methods based on combustion ion chromatography (CIC) to screen the PFAS contaminations in various sewage sludge and wastewater-based fertilizers. Furthermore, current decontamination strategies of PFAS-burdened soils mainly consist of adsorption methods using adsorbents for fixation of PFAS in the ground. A second option is the utilization of a “pump and treat” process, cycling polluted soils through a washing plant leading to the concentration of the pollutants in the fine fraction. Only a subsequent, high-energy consuming pyrolysis process guarantees the total destruction of all fluorinated organic contaminants. Both approaches are cost-intensive and not intended for the direct decomposition of all PFAS contaminants. Interaction pathways between cells and the metal surface remain unclear. The development of this novel single cell-inductively coupled plasma-time of flight-mass spectrometry analytical method and a MIC-specific staining procedure facilitate the investigation of steel-MIC interactions. With this it is possible to analyze the multi-elemental fingerprint of individual cells. The detection method revealed elemental selectivity for the corrosive methanogenic archaeal strain Methanobacterium-affiliated IM1. The interface between material and environmental analysis thus receives special attention, e.g., when considering MIC on solid steel. Hence, the possible uptake of individual elements from different steel samples is investigated. Results showed the cells responded at a single-cell level to the different types of supplemented elements and displayed the abilities to interact with chromium, vanadium, titanium, cobalt, and molybdenum from solid metal surfaces. The high versatility of this nanomaterial is based on their unique size-tunable photoluminescence properties, which can be adjusted from the visible to the near-infrared range. In contrast to other nanomaterials, QDs made the transition from a laboratory curiosity to the utilization in commercial products, like the QLED television screen or in smartphone displays. The best investigated QDs are composed of heavy metals like cadmium or lead, which is not the best choice in terms of toxicity and environmental pollution. A more promising material is Indium Phosphide (InP), which is also currently used by Samsung, Sony and co. in the QLED displays. In addition, the European Commission has re-emphasised the necessity to significantly improve the effectiveness of the current Ecodesign framework for energy-related products. With this paper, we present an initial outline of a policy approach which we call the “EU frontrunner approach”. The approach aims at installing a regulatory framework which enables a semi-automated, progressive adaptation of ecodesign minimum requirements for products. It builds on performance levels of the best products available on the market by aggregating information in a database. The “front-runner approach” could first be applied to progressively adapt product-related minimum energy-efficiency requirements. This way it would serve as a starting point to introduce this conceptto the EU policy arena. While the approach can be applied for energy efficiency, it is neither limited to energy-related products nor to energy-related requirements. N2 - The global COVID-19 pandemic has brought far-reaching changes for our society. Suddenly, millions of people spent the majority of their time at home - either due to a lock down or by switching from office to working remotely. This paper offers a literature review regarding pandemic-related behavioural changes and a first mapping of likely consequences for EU Ecodesign regulations. These impacts are considered for product groups that are already regulated by EU Ecodesign or are under discussion for future regulation. Furthermore, potential for smart home applications are analysed exemplarily. We identified which behavioural changes have the potential to become established in the long-term. This might need to result in adaptions for regulations under the Ecodesign Directive and EU-Energy Labelling regulations to better reflect the recently appeared usage profiles. It might require different priority settings regarding the product groups to be regulated and communicated to users, due to online shopping proliferation, to be updated. Changed behaviour can be observed in product groups such as ICT, consumer electronics and household appliances. Examples include the increased use of laptops and tablets for digital teaching, use of gaming devices and office equipment in private households (IT, printing and network devices) and changed cooking behaviour. This resulted in increased sales of some product types. Online sales became even more important than in-store sales, which also means that consumers are increasingly informed online about energy efficiency. More do-it-yourself (DIY) home projects result in more frequent - not yet regulated - DIY power tool use and over the long run might stimulate the need for (semi-)professional tools. However, it is not yet possible to predict the removal of individual pollutants. An appropriate material characterization, matching adsorption processes in water, might be the missing piece in the puzzle. To this end, this study examined 25 diferent commercially available ACs to evaluate their material properties. Frequently reported analyses, including N2 adsorption/desorption, CHNS(O), point of zero charge (PZC) analysis, and X-ray photoelectron spectroscopy, were conducted on a selected subset of powdered ACs. Inorganic elements examined using X-ray fuorescence and X-ray difraction spectroscopy revealed that relative elemental contents were distinctive to the individual AC’s raw material and activation procedure. This study also is the frst to use thermogravimetric analysis (TGA) coupled to Fourier-transform infrared spectroscopy (FTIR) to conduct quantitative analyses of functional surface oxygen groups (SOGs: carboxylic acid, anhydride, lactone, phenol, carbonyl, and pyrone groups) on such a large number of ACs. The comparably economical TGA method was found to provide good surrogates for the PZC by pyrolytic mass loss up to 600 ◦C (ML600), for the oxygen content by ML1000 and for the carbon content by oxidation. Mass loss profles depict the AC’s chemistry like fngerprints. Furthermore, we found that SOG contents determined by TGA-FTIR covered a wide individual range and depended on the raw material and production process of the AC. TGA and TGA-FTIR might therefore be used to identify the suitability of a particular AC for a variety of target substances in diferent target waters. However, concerns have been raised about the evolution and selection of resistance against disinfectants and AMCs. In turn, resistance against disinfectants and AMCs can be associated to antibiotic resistance due to cross-resistance and co-resistance. We need to understand the mechanisms and risks of disinfectants and AMCs for resistance and cross-resistance evolution to optimize their application and safeguard their long-term efficacy. We used adaptive laboratory evolution (ALE) experiments based on repeated exposure of bacteria to disinfectants. Our results show that repeated disinfection of E. coli with benzalkonium chloride in suspension results in a 2000-fold increase in survival within 5 exposure cycles. Adaption is linked to the initial presence of persister cells highly tolerant to benzalkonium chloride. We used the same approach to develop standardizable ALE experiments to determine resistance evolution to AMCs. The results highlight rapid adaptation of E. coli and P. aeruginosa towards copper surfaces. Moreover, there are multiple situations in the clinic or in the environment in which biocides and antibiotics co-occur and in which combination effects can shape their antimicrobial activity or their selective effects. Our work with P. aeruginosa shows prevalent combination effects of biocides and antibiotics, ranging from synergy to antagonism and resulting in the selection for or against antibiotic resistant strains. The combination effects are dependent on the biofilm mode-of-growth, manifesting in apparent differences in the structural arrangement of antibiotic sensitive and resistant strains in biofilms exposed to combinations. Furthermore, biocides affect rates of mutation and horizontal gene transfer, thereby having a potential facilitating effect on resistance evolution. Taken together, our work shows that the role of biocides as potential drivers of resistance evolution and selection deserves further study and regulative action. Especially for filter dusts and sludges containing zinc and lead from gas purification, there are still considerable recycling problems. However, the high heavy metal contents make landfilling of these dusts and sludges cost-intensive and ecologically problematic. In addition, the relatively high zinc and iron loads represent a valuable resource, which can make reprocessing of the material with recovery of the zinc and iron loads as secondary raw materials ecologically as well as economically attractive. Nevertheless, established recycling methods like the Waelz process fall short in regaining more than one valuable metal from the dusts and operate only under narrow ranges of material compositions, which makes it problematic to adapt these processes to new dust compositions as are to be formed in future steel work operations. To tackle these problems, the German Federal Institute for Materials Research and Testing (BAM) and Ferro Duo GmbH investigate innovative recycling methods for steelwork dusts and sludges in a joint project. This mainly focusses on regaining valuable metals such as iron as well as zinc from blast furnace sludge (BFS) and electric arc furnace dust (EAFD) by thermochemical treatment. Therefore, mixtures of these materials are heated under inert atmosphere employing the carbon present in BFS as internal reducing agent. During the process, metallic iron is formed in the solid reaction product while zinc and other volatile heavy metals are evaporated. This paves the way for the generation of zinc as well as sponge iron as two separate products valuable for metallurgical processes. The process basically consists of heating a mixture of BFS and EAFD to temperatures between 700 and 1100 °C under inert (e.g., nitrogen) or reducing (e.g., CO) gas flow. When a suitable mixing ratio is applied, the carbon present in the BFS can be effectively used as reducing agent for iron and zinc present in both starting materials generating a solid residue with high metallic iron and low carbon content as well as a gas stream laden with zinc and other volatile heavy metals, that can be regained by off-gas treatment. The project aims at testing this concept as a continuous process in a pilot plant established by Ferro Duo GmbH with BAM supplying comprehensive scientific support by lab- and small scale-experiments. Additionally, considering the necessity to mitigate CO2 emissions and the resulting shift in compositions of steel work dusts and sludges, our research also investigates replacement of carbon present in the sludges by hydrogen as potentially carbon-neutral reducing agent. Biocides are antimicrobials used as disinfectants, antiseptics and preservatives. They find application on a large scale in the industrial and medical sector, but also in private households. In terms of mass, the worldwide use of biocides exceeds that of antibiotics. Thus, despite their important role in preventing the spread of pathogens, due to their ubiquity, biocides are suspected to be drivers of the antimicrobial resistance crisis. In our work we try to understand how biocides contribute to the emergence of AMR, what the underlying adaptation principles and mechanisms are and how they compare to those found for antibiotics. Here, I present results which demonstrate that phenotypic heterogeneity in E. coli, namely the formation of tolerant persister cells, can enable the survival of disinfection and consequentially facilitate the evolution of AMR. Using experimental evolution that selects for tolerance, we find that mutations in lipid A biosynthesis arise following periodic treatment with benzalkonium chloride (BAC), a widely used disinfectant. BAC tolerance is associated with additional, diverse mutations as well as changes in the susceptibility to antibiotics from different classes and changes in motility and biofilm formation, suggesting extensive regulatory re-wiring in the evolved clones. Remarkably, we find that fitness costs (growth rate reduction) incurred by BAC tolerance are alleviated in the presence of antibiotics. These findings demonstrate the complexity underlying the adaptation to antimicrobials and highlight the links between persistence to disinfectants and resistance evolution to antimicrobials. As a result of continuous and prolific use, mainly in aviation firefighting foams, thousands of industrial and military installations have been found to contain contaminated soil, groundwater and surface water. Furthermore, because of the continuous contamination through PFAS containing commercial products, effluents and sewage sludge from WWTPs have been shown to be an important source of PFAS discharge into the aquatic environment. In the last few years, legacy PFAS (≥C4) have been found in various environments, including soil, water and wastewater, and their environmental pathways have been partly described. Several long-chain PFAS species, and their respective salts are considered as persistent organic pollutants by the United Nations Stockholm Convention. These pollutants have been linked to altered immune and thyroid function, liver disease, lipid and insulin dysregulation, kidney disease, adverse reproductive and developmental outcomes, and cancer. A significant shift in the chemical industry towards production of short (C4-C7) and ultrashort (C1-C3) alternatives was observed in response to recently intensified regulations and restrictions on the use of long-chain (≥C8) PFAS. PFAS analysis in environmental samples is currently mainly done by liquid chromatography tandem mass spectrometry (LC-MS/MS). This efficient method is conducted in a targeted fashion analyzing a small subset of PFAS. The US EPA method for analysis of PFAS using LC-MS/MS for example currently lists 40 PFAS (≥C4). However, to get a better overview of the amount of “total PFAS,” sum parameter methods like total oxidizable precursor (TOP) assay and methods based on combustion ion chromatography (CIC) are in development. CIC results in data regarding the sum of absorbable organic fluorine (AOF) or extractable organic fluorine (EOF), which can also quantify other organically bound fluorine compounds such as fluorinated pesticides and pharmaceutical. Because PFAS have been extensively used in a variety of AFFF products they can be found in soils from industrial and military installations. Current decontamination strategies of PFAS-burdened soils mainly consist of adsorption methods using adsorbents for fixation of PFAS in the ground. A second option is the utilization of a “pump and treat” process, cycling polluted soils through a washing plant leading to the concentration of the pollutants in the fine fraction. Only a subsequent, high-energy consuming pyrolysis process guarantees the total destruction of all fluorinated organic contaminants. Both approaches are cost-intensive and not intended for the direct decomposition of all PFAS contaminants. Hence, there is a great demand for innovative developments and chemical treatment technologies, dealing with new strategies of tackling the PFAS problem. Previously, mechanochemical treatment of polychlorinated organic compounds in soils showed an efficient dechlorination. Biocides are antimicrobials used as disinfectants, antiseptics and preservatives. They find application on a large scale in the industrial and medical sector, but also in private households. In terms of mass, the worldwide use of biocides exceeds that of antibiotics. Thus, despite their important role in preventing the spread of pathogens, due to their ubiquity, biocides are suspected to be drivers of the antimicrobial resistance crisis. In our work we try to understand how biocides contribute to the emergence of AMR, what the underlying adaptation principles and mechanisms are and how they compare to those found for antibiotics. Here, I present results which demonstrate that phenotypic heterogeneity in E. coli, namely the formation of tolerant persister cells, can enable the survival of disinfection and consequentially facilitate the evolution of AMR. Using experimental evolution that selects for tolerance, we find that mutations in lipid A biosynthesis arise following periodic treatment with benzalkonium chloride (BAC), a widely used disinfectant. BAC tolerance is associated with additional, diverse mutations as well as changes in the susceptibility to antibiotics from different classes and changes in motility and biofilm formation, suggesting extensive regulatory re-wiring in the evolved clones. Remarkably, we find that fitness costs (growth rate reduction) incurred by BAC tolerance are alleviated in the presence of antibiotics. These findings demonstrate the complexity underlying the adaptation to antimicrobials and highlight the links between persistence to disinfectants and resistance evolution to antimicrobials. Research so far has focused mainly on the impacts on organisms by fresh plastic fragments or beats. However, plastic materials found in the environment are mainly a mixture of different polymers and additives and particularly degraded during aging processes. Although especially oxidized degradation products might escape from plastic materials during aging, there is a lack of information on potentially adverse effects on aquatic biota. The latter is of particular interest as oxidized chemicals become more water soluble due to higher polarity and are more bio-available, therefore. The present study focused on plastic leachates of polystyrene (PS) and polylactic acid (PLA), which were derived from alternating stress by hydrolysis and UV radiation. Test specimen of PS, PLA or PS/PLA blends (each 50 %) were alternatingly maintained in water at 45 °C for five days and UV radiated at 45 °C for two days, for in total six weeks. Ecotoxicological effects of potentially generated degradation products of plastic materials in the storage water (mineral water, pH: 7.2) were detected by algae growth inhibition tests with Desmodesmus suspicatus and photosystem II inhibition tests with Raphidocelis subcapitata. Results clearly indicate inhibitory effects on algae growth by contaminants in the storage water of stressed plastic materials with increasing growth inhibition of proceeding hydrolysis and UV stress times. Here, different plastics cause variable responses of algal growth. First chemical analyses indicate dissolved monomers and their oxidation products of plastic materials as possible driver of detected ecotoxicological effects, since detected microplastic particles do not seem to harm algae. T1 - Detection of plastics in soil N2 - Soil acts as a final sink for pollutants. Microplastics from different sources such as plastic mulching, littering, compost, sewage sludge, sedimentary deposition, and tyre abrasion are expected to be found in soil. However, representative and comprehensive information is missing on the sources, transport, and fate. Therefore, a reliable analysis method for microplastics in soils needs to be developed. The presented work describes the development of a procedure for microplastics analysis in soils. A representative sampling based on the on-site conditions and a sample preparation method was established and comprised of a drying step, the separation of microplastic particles > 1 mm, and a density separation for particles < 1 mm. The detection of the large microplastic particles (> 1 mm) was conducted with Attenuated Total Reflection - Fourier Transform infrared Spectroscopy (ATR-FTIR) , while Thermal Extraction Desorption - Gas Chromatography / Mass Spectrometry (TED-GC/MS) was applied for particles < 1 mm, gaining information on the type of polymer and mass fraction. Based on the established method, 14 environmental soil samples ? with different exposure of microplastics from agriculture, industrial sites, roads, and floods were investigated. Due to the arbitrary microplastic particle distribution among the samples for large microplastics, it was reasoned that the found particles were unrepresentative. In contrast, microplastic particles < 1 mm were discovered in a high mass in soil samples exposed to plastic mulching or fertilization with sewage sludge or compost (0 – 115 mg/kg). On average, microplastic contents detected in soil samples taken from a construction site and an inner-city lake were higher (13 – 238 mg/kg). As expected, microplastic content in soil sampled in proximity to roads was more pronounced in the upper soil layers. In contrast, very remote sampling sites, expectably uncontaminated, did not lead to the detection of microplastic regarding to thermoanalytical detection method. In a proof of concept experiment several in vivo and in vitro ecotoxicological tests were applied to evaluate the effect of microplastics (tyre abrasion, polystyrene containing hexabromocyclododecane) in natural soils. In summary, while no effects of the examined probes could be detected on higher levels of biological organization after exposures to earthworm E. andrei, significant changes in several oxidative stress related biomarkers were observed. Ziel dieser Branchen ist perspektivisch die vollständige Vermeidung klimaschädlicher Emissionen – mit besonderem Fokus auf Kohlendioxid. Neben der Umstellung auf regenerative Energieträger, sind im Speziellen für die Zementindustrie die erheblichen Anteile an CO2-Emissionen durch die Entsäuerung von Kalkstein zukünftig zu vermeiden. Da die hervorragenden Bindemitteleigenschaften des bisher hauptsächlich in Zementen eingesetzten Portlandklinkers jedoch auf Calciumverbindungen wie Tri- und Di-calciumsilikaten basieren, stellt die Substitution des Kalks derzeit für die Zementindustrie die größte Herausforderung dar. Neben vollständig neuen Bindemittelsystemen ohne Verwendung von Kalk und CCS/CCU-Technologien zur Vermeidung der CO2-Emissionen bei Verwendung von Kalk, können Abfälle und Nebenprodukte wie etwa Betonbrechsand, verschiedene Schlacken und weitere Massenreststoffe sinnvoll zur Einsparung von Rohstoffen und zur Vermeidung von CO2-Emissionen eingesetzt werden. Es gilt diese Potentiale von Sekundärrohstoffen branchenübergreifend zu identifizieren und Synergien zwischen den Branchen nutzbar zu machen. Hierbei werden neben der chemischen Zusammensetzung der Abfälle und Nebenprodukte auch die Verfügbarkeit und der Massenstrom relevante Kriterien sein. Die potenziellen Sekundärrohstoffe und deren Gemische mit chemisch ähnlicher Zusammensetzung wie die bisher verwendeten Bindemittel, können durch verschiedene technische Ansätze wieder in reaktive Bindemittel umgewandelt und damit nutzbar gemacht werden. Eine gezielte Umwandlung der nicht reaktiven mineralischen Phasen in neue mineralische Phasen mit guten Bindemitteleigenschaften, kann u. a. durch Schmelzprozesse erzielt werden. In Schmelzen können bei hohen Temperaturen die bestehenden, nicht reaktiven mineralischen Phasen zerstört und neue mineralische Phasen mit guten Bindemitteleigenschaften erzeugt werden. Hohe Freiheitsgrade durch gezielte Einstellung der stofflichen Zusammensetzung der Schmelze, durch Stoffseparationen und durch geregelte Abkühlregimes können eine maßgeschneiderte Erzeugung neuer Bindemitteltypen ermöglichen. Aufgrund der hohen erforderlichen Temperaturen von Schmelzprozessen ist die Umsetzung dieser neuen Verfahren nach Möglichkeit in bestehenden pyrometallurgischen Produktionsrouten wie etwa der Stahlproduktion vorzunehmen. So kann etwa der Einsatz von Betonbrechsand als Schlackebildner in der Stahlproduktion zielführend sein, wenn die Schlacke wieder zu einem Bindemittel prozessiert wird und in Zementen Verwendung findet. Eine stoffliche Kreislaufführung wäre hiermit gegeben, wobei der Einsatz von Kalkstein als Schlackebildner oder Rohstoff für die Zementproduktion, und damit die entsprechenden CO2-Emissionen, vermieden werden. Die Entwicklung neuartiger Bindemittelsysteme auf Basis von Sekundärrohstoffen sollte von erforderlichen Anpassungen und neuen Entwicklungen in der Regelsetzung und Normung begleitet werden. Vor allem muss gewährleistet sein, dass die mit neuen Bindemitteln hergestellten Betone den hohen Anforderungen an Dauerhaftigkeit und Umweltverträglichkeit genügen. T1 - A Novel Dry Treatment for Municipal Solid Waste Incineration Bottom Ash for the Reduction of Salts and Potential Toxic Elements N2 - The main obstacle to bottom ash (BA) being used as a recycling aggregate is the content of salts and potential toxic elements (PTEs), concentrated in a layer that coats BA particles. This work presents a dry treatment for the removal of salts and PTEs from BA particles. Two pilotscale abrasion units (with/without the removal of the fine particles) were fed with different BA samples. The performance of the abrasion tests was assessed through the analyses of particle size and moisture, and that of the column leaching tests at solid-to-liquid ratios between 0.3 and 4. The results were: the particle-size distribution of the treated materials was homogeneous (25 wt % had dimensions <6.3 mm) and their moisture halved, as well as the electrical conductivity of the leachates. A significant decrease was observed in the leachates of the treated BA for sulphates (44%), chlorides (26%), and PTEs (53% Cr, 60% Cu and 8% Mo). The statistical analysis revealed good correlations between chloride and sulphate concentrations in the leachates with Ba, Cu, Mo, and Sr, illustrating the consistent behavior of the major and minor components of the layer surrounding BA particles. In conclusion, the tested process could be considered as promising for the improvement of BA valorization. The European biocidal products regulation requires environmental risk assessments for biocidal products under service conditions. This is only possible, if processes that lead to biocide emissions into the environment are understood and can be reasonably predicted. Actual emissions due to leaching result from different processes that are affected by material properties and environmental conditions. Transformation of biocides can affect emissions considerably. Leaching and possible transformation of the film preservatives carbendazim, diuron, OIT and terbutryn in paints was observed under laboratory and field conditons in order to investigate the influence of different factors, e.g. water contact and radiation on biocide emissions. The paints contained either white titanium dioxide or a red iron oxide pigment to check whether pigments affect leaching processes. Methods. Test specimens of a water-based styrene acrylic dispersion paint on glass were exposed to UVA-radiation in a UV weathering device and/or exposed to water contact by immersion events on the basis of the European standard EN 16105. Plywood panels were coated and exposed to natural weather conditions in a semi-field experiment. Concentrations of biocides and transformation products were analysed in leachates from laboratory tests, runoff samples and remaining coatings. Results. The impact of weathering conditions on transformation processes differs for the investigated active substances. Water contact, radiation as well as pigments in the paints determine the pattern and amount of transformation products on coatings, leachates and runoff water. Observations from laboratory and field experiments were compared. It proved that results from laboratory tests support understanding of data from field experiments. Detailed knowledge on the fate of active substances under environmental conditions supports manufacturers to develop safe applications of film preservatives and authorities to evaluate its environmental impact. All organisms must protect themselves from the harmful effects of light such as UV radiation, ROS accumulation, heat, and desiccation. Finally, light qualities and quantities can be used for decision making, timing and as guide for directed growth when they are sensed and transduced into intracellular signals. Botrytis cinerea and other plant pathogens infecting the sun-exposed parts of the plant must cope with the high light conditions the host plant seeks. Further they experience an altered light spectrum (‘green gap’) when they colonize shaded parts of the plant; it is depleted for blue and red light that is absorbed by the plant chlorophyll and enriched for green and far-red light that is reflected or transmitted by the plant tissue. As these ambient light conditions trigger the shade avoidance response in the plant, the pathogens may trigger their own ‘shading response’ such as the upregulation of virulence determinants and inoculum production. B. cinerea maintains a highly sophisticated light signaling machinery that senses different light qualities to trigger a variety of responses, that are protection, morphogenesis, positive and negative tropisms, and entrainment. These characteristics render B. cinerea a valuable model to enlighten the role of light in parasitic fungus-plant interactions and beyond. The vegetative mycelium – the core of all infection and developmental programs – is not visibly pigmented and thus considered to be sensitive to biotic and abiotic stresses. However, the vegetative hyphae have a very limited half-life and are usually restricted to the invasive growth phase in which they are protected from light by the plant tissue. Fast colonization of host tissues and by this proper nutrient acquisition enables the rapid formation of long-lasting reproduction structures (melanized conidiophores with conidia, sclerotia) on the surfaces of rotted plant tissues. Depending on the light and temperature conditions, conidiation or sclerotial development is initiated. In theory, the uptake into the passive sampler is only dependent on the diffusion coefficient of the analyte in air and the geometry of the sampler (ideal adsorption). For characterization, the uptake rate (UR) is used, defined as the ratio of the uptaken mass and the product of ambient concentration and exposure time. Various reported effective uptake rates (UR,eff) differ considerably from ideal values (UR,id). Literature values in national and international standards, are essentially sorted by the applicable exposure time, while detailed information about the corresponding concentration range is lacking. Moreover the number of itemized substances is limited. The aim of this study was to review and expand this set of uptake rates by determination of own quality assured laboratory values.The determined uptake rates are considered accurate in terms of RSD and comparability to literature values and can be recommended for exposure times of seven days at 50 100 µg/m³ (approximately 100 300 ppm min). Seven day uptake rates in ISO16017 2 and ASTM D6196 are not generally suited for this purpose, as has been exemplarily shown for benzene. Those natural microbial communities are frequently exposed to biocides used as material preservatives. Previous studies have shown that antibiotics, metals and pesticides affect the underlying processes of resistance evolution and spread; namely de novo mutagenesis and horizontal gene transfer by conjugation and transformation in microbial communities. However, it is unknown if active substances used in material preservatives are involved in these processes. We show that biocides used as material preservatives affect rates of mutation and conjugation in microorganism in a species- and substance-dependent manner, while rates of transformation are not directly affected. However, concerns have been raised about the possibility for the evolution and selection of resistance against disinfectants and AMCs. In turn, resistance against disinfectants and AMCs can be associated to antibiotic resistance due to cross-resistance - a single mechanism conferring resistance to a disinfectant and an antibiotic- and co-resistance - two distinct mechanisms physically linked on e.g. a plasmid. The risk for resistance and cross-resistance during use of biocides (including disinfectants and AMCs) must be evaluated during authorization according to the EU biocidal product regulation. However, to date there is a lack of standardized methods that support risk assessment during the authorization process. Methods We used adaptive laboratory evolution (ALE) experiments which are based on repeated exposure of bacteria to disinfectants or AMCs. The experiments are followed by phenotypic (antimicrobial susceptibility testing) and genotypic (whole genome sequencing) characterization of the evolved strains. The basic idea of these experiments is to expose bacteria to lethal conditions and select for mutants with increased survival. This approach is fundamentally different to other ALE experiments, which commonly select for increased growth at subinhibitory concentrations. However, selection for increased survival represents a selective pressure that more realistically reflects selection under in-use conditions of disinfectants and AMCs. Results First, we studied adaptation of E. coli during repeated disinfection with benzalkonium chloride in a suspension assay. The experiments showed a 2000-fold increase in survival within 5 exposure cycles. The adaptive changes are linked to highly parallel mutations in genes related to lipid A biosynthesis, less negative cell surface charge, reduced growth rate and increased competitive ability in the presence of certain antibiotics. We use the same approach to develop standardizable ALE experiments based upon accepted standards that are used to determine the efficacy of disinfectants (EN 13697) and antimicrobial surfaces (ISO 22196). The results highlight pronounced adaptation of different test strains towards surface disinfection (benzalkonium chloride and isopropanol) and AMCs (copper). Conclusion Bacteria can adapt with increased survival towards lethal stress imposed by disinfectants and AMCs. Biocides play a major role in the prevention of microbial infections in healthcare and animal husbandry. The use of biocides often leads to the discharge of active biocidal substances into wastewater streams, causing the exposure of wastewater microbial communities to subinhibitory concentrations. In turn, it is known that wastewater treatment plants (WWTP) are hotspots for antibiotic resistant bacteria. Since similar mechanisms confer resistance to biocides and antibiotics, exposure to biocides can result in co-selection of antibiotic resistant bacteria in WWTP due to biocides. Here, we investigate the magnitude and the drivers of co-selection of antibiotic resistance in natural wastewater microbial communities upon biocide exposure. Microbial communities will be sampled at the WWTP Ruhleben in Berlin and characterized regarding their biocide and antibiotic resistance. Changes in the resistance level after exposure to different biocides will be determined by enumerating resistant and non-resistant E. coli and heterotrophic bacteria on selective plates with and without several biocides and antibiotics. Moreover, we are establishing a synthetic community comprising about 100 environmental E. coli isolates each with different antimicrobial resistance traits. Each isolate will be tagged with a unique DNA-barcode. All isolates will be pooled and exposed to different biocides at various concentrations. The barcode labeling enables us to determine the abundance of each isolate at the beginning and end of the experiment by transposon-tag sequencing. The project results will inform risk assessment of the effects of biocidal residues on antimicrobial resistance selection in WWTP. N2 - In urban waters, a multitude of organic micropollutants, often termed emerging pollutants, has been found over the last decades. Analytical methods suitable for trace analysis are needed that are desirably also fast, inexpensive and, if possible, robust and portable. Immunoanalytical, i.e., antibody-based, methods which are available in a broad range of formats, can be profitably used here to screen for the distribution and to monitor the trends of concentration levels of contaminants of emerging concern in the environment. Some of these formats are single-analyte but high-throughput methods. To use them wisely, indicator substances, sometimes called anthropogenic markers, should be selected and used in screening approaches, i.e., as indicators for contamination and the pre-selection of samples at which to have a closer look by multiplex methods like LC-MS/MS. Other methods are suitable to be performed on portable instrumentation in the field (on-site) or in facilities such as wastewater treatment plants for on-line monitoring of the treatment and elimination process. Furthermore, array technologies have been established that allow for parallel (multiplex) analysis of several analytes of interest. The microtiter-plate based ELISA (Enzyme-linked Immunosorbent Assay) is the method of choice for the analysis of a large number of samples [1]. ELISAs are available to monitor for anthropogenic markers such as the antiepileptic carbamazepine, the analgesic diclofenac, the antihistaminic cetirizine, the steroid hormone estrone, the antimicrobial sulfamethoxazole, psychoactive caffeine and cocaine, the priority pollutant bisphenol A, and the bile acid isolithocholic acid. For on-site screening and monitoring, simpler formats, like mix-and-read assays, e.g., the Fluorescence Polarization Immunoassay (FPIA) [2] or Lateral-flow Immunoassays (LFIA) [3] are more suitable tools, the latter based on dipsticks or little cassettes, with which users have become very familiar during the COVID-19 pandemic via rapid antigen tests. The suitability of multi-analyte formats such as immunomicroarrays depends on the choice of a signal-producing system that provides small uncertainties and good reproducibility of the measurements. Bead-based (“suspension”) arrays, read out in flow cytometers, are a powerful platform for multiplex assays [4]. Electrochemical formats, run on portable devices, provide additional advantages as no light source is required. They are most promising for stand-alone analysers and biosensors [5]. The speed, low cost and on-site capabilities of these methods allow to gather a lot more data on anthropogenic compounds which enables to quantify inputs, differences in degradation power of elimination processes, dilution phenomena and a more precise image of individual water cycles which is demonstrated by several examples. As a result of the perpetual use of PFAS containing products, effluents and sewage sludge from wastewater treatment plants (WWTPs) have been observed to be an important pathway for PFAS into the environment. In Germany, phosphorus and other nutrients from sewage sludge and wastewater should be recycled in WWTPs of cities with a large population. However, it is not clear if PFAS contamination from wastewater and sewage sludge end up in novel wastewater-based fertilizers. Normally, PFAS are analyzed using PFAS protocols typically with time-consuming extraction steps and LC-MS-MS quantification. However, for screening of PFAS contaminations in sewage sludge or wastewater-based fertilizers also passive sampler based on the Diffusive Gradients in Thin-films (DGT) technique can be used for the PFAS extraction. Afterwards, combustion ion chromatography (CIC) can be applied to analyse the “total” amount of PFAS on the passive sampler. Here, we show results from the DGT method in comparison to those of the extractable organic fluorine (EOF) method for a variety of wastewater-based fertilizers. Additionally, we analysed the adsorption of PFAS on the weak anion exchanger (WAX) based DGT passive sampler binding layer by infrared and fluorine K-edge X-ray adsorption near-edge structure (XANES) spectroscopy. Altogether, there are currently more than 4700 known but only partly characterized fluorinated compounds identified by the Organization for Economic Cooperation and Development (OECD), as shown by a recently published classification proposal. The ongoing production of new, yet unrestricted PFAS alternatives has become a major challenge for environmental routine analytics, since the state-of-the-art method LC-MS/MS relies on structural information and availability of isotope standards of the targeted compound. First reported by Miyake et al., fluorine sum parameters like adsorbable organic fluorine (AOF), extractable organic fluorine (EOF) and total fluorine (TF) can be applied to survey and detect the presence of large amounts of unidentified organofluorine compounds in environmental matrices. Today fluorine sum parameters have been established as a useful supplement to classic target-analytical approaches of PFAS and were implemented for the first time as a sum value “PFAS-total” in the recently revised Drinking Water Directive (2020/2184) by the European Commission. In contrast, X-ray absorption near-edge structure (XANES) spectroscopy has been widely applied to identify low concentration of element-specific contamination without pre-treatment in environmental samples in the past. The XANES approach enables a penetration depth of approx. 1 µm (at fluorine K-edge energy; depending on the matrix) which is significantly deeper than for X-ray photoelectron spectroscopy (XPS; penetration depth approx. 10 nm). Furthermore, the method is fast, non-destructive and only simple preparation of the samples is required. In our work, we combined both F-XANES spectroscopy and combustion ion chromatography (CIC) in order to present a new analytical perspective on the investigation of PFAS in environmental media and material samples. Biocides are antimicrobials used as disinfectants, antiseptics and preservatives. They find application on a large scale in the industrial and medical sector, but also in private households. In terms of mass, the worldwide use of biocides exceeds that of antibiotics. Thus, despite their important role in preventing the spread of pathogens, due to their ubiquity, biocides are suspected to be drivers of the antimicrobial resistance crisis. In our work at BAM we try to understand how biocides contribute to the emergence of AMR, what the underlying adaptation principles and mechanisms are and how they compare to those found for antibiotics. Within our group, I mainly focus on the following two questions: How does phenotypic heterogeneity in bacteria affect the ability to survive treatment with biocides? And what are the consequences of phenotypic heterogeneity for the evolution of resistance to biocides and antibiotics? I will share published and unpublished results which demonstrate that phenotypic heterogeneity can enable the survival of biocide treatment and, through this, facilitate the evolution of AMR. On the other hand, we find that adaptation to a biocide can unexpectedly impair the ability to evolve resistance against an antibiotic. An der BAM wurden die unterschiedlichen Methodenkombinationen aus Aufschluss- und Messmethode an 15 Klärschlämmen überprüft und in einem Ringversuch mit einem Klärschlamm getestet. A low SO2 content within the CO2-stream might be more important than a low water content. Cr13-steels showed a general susceptibility to shallow pitting and pitting. So, they seem to be not suitable for CCUS applications. Low alloyed steels showed better corrosion behavior (predictable uniform corrosion). For direct contact with saline aquifer fluids only high alloyed steels shall be used. A challenging task however, is the generation of three-dimensional distribution maps from these spatially sparse gas measurements. To obtain high-quality reconstructions, the choice of optimal measuring poses is of great importance. Remote gas sensors, that are commonly used in Robot Assisted Gas Tomography (RAGT), require reflecting surfaces within the sensor’s range, limiting the possible sensing geometries, regardless of whether the robots are ground-based or airborne. By combining ground and aerial robots into a heterogeneous swarm whose agents are equipped with reflectors and remote gas sensors, remote inter-robot gas measurements become available, taking RAGT to the next dimension – releasing those constraints. In this paper, we demonstrate the feasibility of drone-to-drone measurements under realistic conditions and highlight the resulting opportunities. The objective is to create a flame retarded epoxy resin from a commercial bio epoxy resin with halogen free inorganic and organic flame retardants, respectively. Alumina trihydrate, aluminum diethyl phosphinate, a DOPO-based phosphonamidate and ammonium polyphosphate seem to have promising performances. Properties are enhanced with different bio fillers: pyrolyzed cocoa shells and plant waste (provided by Otto A. Müller Recycling GmbH, thanks!), short fibers and nonwovens of the natural fiber kenaf, hydroxypropyl-ß-cyclodextrin and sulfobutylether-ß-cyclodextrin. Fire performance is investigated by cone calorimeter examinations, LOI and UL-94 ratings. Thermal analysis is given by TG-FTIR and DSC measurements. Combinations of flame retardants and bio-fillers lead to reduced PHRR and THR, reach V0 in UL-94 and have a significant increase in LOI of up to 37 vol.-%. 10 % ammonium polyphosphate with 10 % pyrolyzed cocoa shell performs best, builds a magnificent protective layer, and shows good intumescence. A1 - Schwibbert, Karin T1 - Reducing Escherichia coli adhesion to PET by modulating spatial periods of laser-induced surface nanoripples N2 - Using nanofiber-like cell appendages, secreted proteins and sugars, bacteria can establish initial surface contact followed by irreversible adhesion and the formation of multicellular biofilms. Here, the stabilizing extracellular biofilm matrix together with physiological changes on the single cell level leads to an increased resilience towards harsh environmental conditions, antimicrobials, the host immune response and established cleaning procedures. Persistent microbial adhesion on e.g., medical implants, in water supply networks or food-processing industry is often associated with chronic inflammation, nosocomial and foodborne infections, enhanced biofouling and product contamination. To prevent persistent microbial colonization, antibacterial surface strategies often target the initial steps of biofilm formation and impede adhesion of single cells before a mature biofilm is being formed. While chemical coatings have been widely used, their restricted biocompatibility for eukaryotic cells and attenuated antibacterial-effects due to compound release limit their areas of application and alternative strategies focus on modified surfaces topographies to impede bacterial adhesion. In this work, we used ns-UV laser treatment (wavelength 248 nm and a pulse duration of 20 ns) to generate laser-induced periodic surface structures (LIPSS) with different submicrometric periods ranging from ~210 to ~610 nm on commercial poly(ethylene terephthalate) (PET) foils. Following structurally and chemically analyses, PET samples were subjected to bacterial colonization studies with Escherichia coli TG1, a bacterial test strain with a strong biofilm formation capacity due to the formation of nanofiber-like cell-appendages (pili). Bacterial adhesion tests revealed that E. coli repellence decisively depends on the spatial periods of the LIPSS with the strongest reduction (~91%) in cell adhesion observed for LIPSS periods of 214 nm. Scanning electron microscopy and additional biofilm studies using a pili-deficient E. coli TG1 strain revealed the importance of extracellular appendages in the bacterial repellence observed here, thus, pointing out new antibiotics-free strategies for antibacterial surfaces by impeding nanofiber-mediated bacterial adhesion. The wild type was selected over the BAC-tolerant strain in the absence of antibiotics, while the opposite was observed at all ciprofloxacin concentrations investigated.Furthermore, we assessed the evolvability of resistance of the two strains to inhibitory concentrations of ciprofloxacin by performing a serial dilution evolution experiment with gradually increasing ciprofloxacin concentrations. The wild type had a higher probability to develop resistance to ciprofloxacin than the tolerant strain. By the end of the evolution experiment both strains evolved to grow at the highest ciprofloxacin concentration investigated, which was 2048 ×MIC of the wild type. Problematisch ist, dass viele der wirtschaftlich relevantesten Kunststoffe sehr lange brauchen, um in der Umwelt abzubauen, sich dort akkumulieren und in kritisch betrachtetes Makro- und Mikroplastik zu zerfallen. Auf der Grundlage des Vorsorgeprinzips fordert die Europäische Kommission daher die Entwicklung neuer polymerer Materialien, die diesem Problem der Persistenz entgegenwirken und unter natürlichen Umweltbedingungen schneller abgebaut werden können. 1,2 Um dieses Problem anzusprechen, wurden mittels additiver Fertigung Polymerblends aus photooxidationsempfindlichem Polystyrol (PS) und hydrolyseempfindlicher Polymilchsäure (PLA) hergestellt. Die Hypothese war, dass sich die beiden Thermoplaste aufgrund ihrer unterschiedlichen Abbauprozesse gegenseitig in ihrem Abbau unterstützen, wenn sie unter moderaten, realitätsnahen Umweltalterungsbedingungen (45 °C) abwechselnd der Photooxidation und Hydrolyse ausgesetzt werden. In der Praxis zeigten die Ergebnisse, dass die Degradation von PLA durch die Anwesenheit von PS deutlich beschleunigt worden ist – wohingegen die Degradation von PS gehemmt wurde. The BAM departments 7.1 (building materials) and 4.0 (Material and the environment) try to improve the sustainability of concrete panels by establishing a biofilm on the weather protection layer. Using an ultra-high-performance-concrete allows the production of very thin facade tiles with a small carbon footprint. Furthermore, the durability of these tiles is expected to surpass the one of “normal” concrete facades and offer the possibility of modular renewing of defective tiles instead of a wall, or even remodeling a whole building. The controlled application of biofilms on facades aims to improve biodiversity, microclimate, and air quality in cities. Furthermore, the greening of typically gray surfaces could improve the mental health of the residents. Typically, airborne particles are sampled over long time periods on filters, followed by lab-based analysis, e.g., with inductively coupled plasma mass spectrometry (ICP-MS). Within the EURAMET EMPIR AEROMET project, cascade impactor aerosol sampling was combined for the first time with on-site total reflection X-ray fluorescence (TXRF) spectroscopy to develop a tool for quantifying particle element compositions within short time intervals and even on-site. This makes variations of aerosol chemistry observable with time resolution of only a few hours and with good size resolution in the PM10 range. A proof of principles of this methodological approach and the comparison to standard methods within the scope of a field campaign will be presented. Secondly, aerosol sampling and TXRF analysis seems suitable for the quantification of elements in indoor aerosols as well and may provide an important enhancement of existing methods for the analysis of organic species in aerosols (such as sampling and TD-GC/MS). T1 - Nitrogen Gain and Loss Along an Ecosystem Sequence: From Semi-desert to Rainforest N2 - Plants and microorganisms, besides the climate, drive nitrogen (N) cycling in ecosystems. Our objective was to investigate N losses and N acquisition strategies along a unique ecosystem-sequence (ecosequence) ranging from arid shrubland through Mediterranean woodland to temperate rainforest. These ecosystems differ in mean annual precipitation, mean annual temperate, and vegetation cover, but developed on similar granitoid soil parent material, were addressed using a combination of molecular biology and soil biogeochemical tools. Soil N and carbon (C) contents, δ15N signatures, activities of N acquiring extracellular enzymes as well as the abundance of soil bacteria and fungi, and diazotrophs in bulk topsoil and rhizosphere were determined. Relative fungal abundance in the rhizosphere was higher under woodland and forest than under shrubland. This indicates toward plants' higher C investment into fungi in the Mediterranean and temperate rainforest sites than in the arid site. Fungi are likely to decompose lignified forest litter for efficient recycling of litter-derived N and further nutrients. Rhizosphere—a hotspot for the N fixation—was enriched in diazotrophs (factor 8 to 16 in comparison to bulk topsoil) emphasizing the general importance of root/microbe association in N cycle. These results show that the temperate rainforest is an N acquiring ecosystem, whereas N in the arid shrubland is strongly recycled. Simultaneously, the strongest 15N enrichment with decreasing N content with depth was detected in the Mediterranean woodland, indicating that N mineralization and loss is highest (and likely the fastest) in the woodland across the continental transect. Higher relative aminopeptidase activities in the woodland than in the forest enabled a fast N mineralization. Relative aminopeptidase activities were highest in the arid shrubland. The highest absolute chitinase activities were observed in the forest. This likely demonstrates that (a) plants and microorganisms in the arid shrubland invest largely into mobilization and reutilization of organically bound N by exoenzymes, and (b) that the ecosystem N nutrition shifts from a peptide-based N in the arid shrubland to a peptide- and chitin-based N nutrition in the temperate rainforest, where the high N demand is complemented by intensive N fixation in the rhizosphere. In theory, the uptake into the passive sampler is only dependent on the diffusion coefficient of the analyte in air and the geometry of the sampler (ideal adsorption). For characterization, the uptake rate (UR) is used defined as the ratio of the mass adsorbed and the product of ambient concentration and exposure time. Various reported effective uptake rates (UR,eff) differ to an increasing degree from ideal values (UR,id) with increasing exposure doses (denominator of the given definition of UR), (Tolnai, 2001). In national and international standards, uptake rates are essentially sorted by the applicable exposure time, while detailed information about the corresponding concentration range is lacking. Moreover, especially for long exposure periods as applied in indoor air monitoring, the number of itemized substances is limited. Therefore it is the aim of this contribution to review and expand uptake rate data by comparison of literature and own laboratory values of assured quality. Passive samplers were exposed to nine single compound atmospheres of known concentrations for seven days. Concentrations were checked twice a day via active sampling.The determined uptake rates are considered accurate in terms of RSD and comparability to literature values and can be recommended for exposure times of seven days at 50 – 100 μg m-3 (approximately 100 – 300 ppm min). Seven-day uptake rates in ISO16017-2 and ASTM D6196 are not generally suited for this purpose, as has been exemplarily shown for benzene. Thus, it is crucial for optimization of the method to produce more reliable uptake rate data, including specific information about applicable exposure times and concentrations, which will be promoted in the course of this ongoing study. Magnetic bead-based immunoassays (MBBAs) allow for implementation into an immunosensor for online testing. Particles are prepared by either coupling the analyte molecule (diclofenac) or the respective antibody (amoxicillin) to the surface. For miniaturization, detection is performed electrochemically (chronoamperometry) on a microfluidic chip. The developed immunosensor will enable detection of pharmaceuticals directly in water supply pipes.
