Several procedures have been developed to recovery these technological important elements. Nevertheless, actual REE recycling from fluorescence lamps is scarce so far (recovery rate of less than 1 %), with current recycling approaches concentrating on glass recovery. Since most recycling processes include several, also wet-chemical steps, a complete knowledge of the actual elemental composition of the respective mass flows is necessary for an efficient REE recovery. We tested seven different reagent mixtures for microwave-assisted digestion of fluorescent lamp shredder, including HF, HClO4, and H2O2. We determined the concentrations of 25 of the most relevant rare earth and other trace elements in the respective dilutions. Two independent digestions, one a mixture of perchlorid/nitric/hydrofluoric acid and the other aqua regia, showed the highest concentrations of 23 of these elements, excluding only Sn and Tb. The REE concentrations in the tested lamp shredder sample (stated in g/kg) were 10.2 (Y), 12.1 (La), 7.77 (Ce), 6.91 (Eu), 1.90 (Gd), and 4.11 (Tb). Since then various other environmental systems all over the world, like rivers, lakes and biota have been found to be polluted by plastics. Lately, agriculturally used land, waste-water treatment and biogas facilities come into focus as possibly relevant distribution vectors. To monitor the situation and investigate distribution pathways, the need for fast and reliable analytical procedures is high. Up to now, there is no standard procedure for sampling, concentrating and analyzing plastic particles in environmental samples. Our group recently reported the development of a new analyzing method for microscale plastic particles, thermal extraction desorption gas chromatography mass spectrometry (TED-GC-MS). This method consists of thermal extraction of the sample via thermogravimetric analysis, collection of the evolved gaseous decomposition products on a solid-phase adsorber and analysis of the loaded adsorber using Thermal-Desorption-GC-MS. This technique utilizes the characteristic decomposition products of polymers for identification and even quantification of plastics in environmental samples. In the present work we investigated samples of fermentation residues taken from a biogas plant. The results of sample preparation, concentration and analysis are presented as well as a comparison with alternative methods. Therefore, the interest in development of new materials with improved surfaces to reduce bacterial colonization rises. In order to evaluate the quality and safety of these new materials, it is highly important to ensure world-wide comparable tests that are relying on statistical evidence. The only way to reach this statistical safety is through a high-throughput Screening under standardized test conditions. We developed a flow through system for cultivation of biofilm-forming bacteria under controlled conditions with a total capacity for testing up to 32 samples in parallel. Quantification of the surface colonization was done by staining the bacterial cells with a fluorescence marker, followed by epifluorescence microscopy. More than 100 images of each sample were automatically taken and the surface coverage was estimated with the free open source software gmic (http://gmic.eu), followed by a precise statistical evaluation. Overview images of all gathered pictures of the whole material coupon were generated to illuminate the colonization characteristics of the selected bacteria on certain materials. With this method, differences in bacterial colonization on different materials can be quantified in a statistically validated manner. Here, we present SBA-15-type mesoporous silica materials containing boron–dipyrromethene (BODIPY) probes in their pore systems that allow for the sensitive and selective detection of the two target analytes CWAs and Hg2+ in natural waters via a change in the optical properties. In case of Hg2+, the BODIPY dye is directly adsorbed on the (inner and outer) surface of the highly porous material and coordinates the metal ion, leading to an enhancement of fluorescence with a limit of detection (LOD) of 15 ppt. Using the same silica material but incorporating a covalently attached BODIPY dye, the CWAs GA, GD and GB can be detected via fluorescence quenching induced by an intraannular cyclization. The obtained LOD in natural waters is in the pM range and hence below the Maximum Permissible Concentrations for CWAs in drinking water. However, the use of large quantities of the treated plants as animal feed leads to residues in meat, milk and eggs. Furthermore, the herbicide can drain away and contaminate ground and drinking water. The ingestion of 2,4-D-contaminated food and water causes damage to the inner organs of humans and animals, e.g., the kidneys and the liver. Analytical assays based on molecularly imprinted polymers (MIPs) have emerged as a valuable tool in the field of environmental analysis due to the low production costs, stability, format adaptability and the possibility to imprint and thus their ability to recognize a wide variety of target analytes.5 With regard to optical sensing technologies, however, MIPs have only been used in considerably few applications, especially in fluorescence sensors. This limitation is basically due to the fact that the incorporation of a fluorescently responding moiety into a polymer matrix is challenging. One way to overcome this limitation is to use tailor-made fluorescent indicator monomers for direct transfer of the binding event into an optical signal and coat the MIP via reversible addition-fragmentation chain transfer (RAFT) polymerization as a thin layer onto the surface of silica nanoparticles as primary sensing element. Although several countries already recycle high amounts of CDW, the use as secondary raw materials is often limited by inferior building material properties. Specific characteristics, like high porosity and low density are caused by hardened cement paste in crushed concrete and the content of mortar and plaster in brick debris. Impurities like wood or gypsum and also harmful substances like organic pollutants may be a major problem for a reuse and should be minimized. Therefore unwanted materials and impairing substances have to be separated from the secondary building material stream. This can be done during the demolition process by using techniques for selective dismantling or during the subsequent treatment of the resulting rubble. Since almost all processing steps are associated with environmental impacts, the benefits of saving natural resources by applying secondary building materials should be weighed carefully. Compatible solutes are of diverse chemical structure (sugars, polyols, amino acid derivatives) and are beneficial for bacterial cells not only as osmoregulatory solutes, but also as protectants of proteins by mitigating detrimental effects of freezing, drying and high temperatures. The aspartate derivative ectoine is a wide spread compatible solute in Bacteria and possesses additional protective properties compared with other compatible solutes, and stabilizes even whole cells against stresses such as UV radiation or cytotoxins. The protective properties of ectoine for proteins can be explained by its strong (kosmotropic) interaction with water and subsequent exclusion from Protein surface, the decrease of the solubility of the peptide backbone and the strengthening of intramolecular hydrogen bonds (secondary structures). The stabilizing and UV-protective properties of ectoine attracted industry, which saw the potential to market ectoine as a novel active component in health care products and cosmetics. In joint efforts of industry and research large-scale fermentation procedures have been developed with the halophilic bacterium Halomonas elongata used as a producer strain. The two key technologies that allow for the annual production of ectoine on a scale of tons are the bacterial milking procedure and the development and application of ectoine-excreting mutants (“leaky” mutant). The details of these two procedures including the strain development and Fermentation processes will be introduced and current and future applications of ectoine will be discussed. Despite the appeal of this mechanism, however, direct supporting evidence is lacking. Here, we use experimental and theoretical methods to demonstrate that an allosteric mechanism of enhanced substrate release is indeed plausible. First, we report the atomic-resolution structure of APO TeaA, the SBP of the Na!-coupled ectoine TRAP transporter TeaBC from Halomonas elongata DSM2581T, and compare it with the substrate-bound structure previously reported. Conformational freeenergy landscape calculations based upon molecular Dynamics simulations are then used to dissect the mechanism that couples ectoine binding to structural change in TeaA. These insights allow us to design a triple mutation that biases TeaA toward apo-like conformations without directly perturbing the binding cleft, thus mimicking the influence of the membrane transporter. Calorimetric measurements demonstrate that the ectoine affinity of the conformationally biased triple mutant is 100-fold weaker than that of the wild type. By contrast, a control mutant predicted to be conformationally unbiased displays wild-type affinity. This work thus demonstrates that substrate release from SBPs onto their Membrane transporters can be facilitated by the latter through a mechanism of allosteric modulation of the former. The importance of social immunity in preventing disease transmission in insect societies is well known. However, we still lack a clear understanding of how collective defence mechanisms are regulated and how they interact with the other regulatory systems of the organism such us the individual immune system. Here we assess how individual immune factors shape the collective immune response to pathogens by inhibiting a key Termite immune effector: gram-negative binding protein (tGNBP-2). tGNBP-2 triggers downstream innate immunity, in addition to cleaving fungal and bacterial pathogens directly in the external environment. We established mini-colonies of Reticulitermes flavipes inside planar arenas where we were able to observe and record hygienic behaviours (i.e. allogrooming and cannibalism) in near-natural nest conditions. Colonies were either treated with the tGNBP-2 inhibitor or a control solution, after which 5 focal workers were introduced into each colony. Focal workers were pre-exposed to either: 1) the fungal pathogen Metharizium anisopliae 2) the t-GNBP-2 inhibitor + M. anisopliae 3) a control solution. Colonies were recorded and the hygienic behaviours performed by nestmates towards focal termites were analysed. By comparison, little attention has been paid to the role of soldiers against pathogenic microorganisms. This is a significant gap in knowledge because in non-hymenopteran societies, such as the eusocial termites, the first-evolved, and arguably most important sterile caste, is the soldier. Here, we investigate whether soldiers of the wood-eating termite, Mastotermes darwiniensis, play a significant role in social immunity. Firstly, we show that uninfected soldiers improve the survival of worker nestmates exposed to a lethal dose of a fungal pathogen, despite the fact that soldiers (unlike workers) cannot allogroom. Hazard ratios of pathogen-exposed workers decreased from 6.616 in infected groups to 4.376 and 2.786 in infected groups containing uninfected workers and soldiers, respectively. To explore the basis of soldier protection, we applied soldier-derived oral secretions to infected worker cuticle, resulting in significantly improved worker survival. Soldiers that contacted infected workers were also more active and produced secretions with significantly greater mass variance than soldiers exposed to uninfected workers. Significantly, we show that secretions produced by soldiers inhibit the growth of a broad range of microorganisms, including gram-negative and gram-positive bacteria, in addition to fungi. Our findings demonstrate that soldiers can contribute powerful antimicrobial factors to external immune defenses, effectively complementing behaviour-based protective activities provided by workers. Ingestion of food and feed containing ZEN causes numerous mycotoxicoses in animals and possibly humans with pronounced estrogenic effects. Due to the trans-configurated double bond, ZEN isomerizes to the cis-configuration upon the influence of light. This work investigates the instrumental analysis, metabolism and toxicity of ZEN and cis- ZEN. The first part focused on the determination of ZEN in edible oils. Due to a maximum level of 400 µg/kg ZEN in the European Union (EU), reliable analytical methods are needed. A comprehensive method comparison proved dynamic covalent hydrazine chemistry (DCHC) to be the most suitable approach. Thus, an automated solid phase extraction (SPE) coupled online to high performance liquid chromatography (HPLC) was developed with the novelty of a covalent SPE step comprising the DCHC principle. The automated online system allows an accurate, selective and reliable quantification of ZEN in edible oils in compliance with EU performance criteria while significantly reducing workload and thereby personnel costs. In contrast to ZEN, reference standards and analytical methods are missing for cis- ZEN which causes a lack of data on the occurrence, fate and risks of cis-ZEN. Therefore, a native and an isotopically labeled cis-ZEN standard were synthesized and implemented in an existing stable isotope dilution analysis HPLC tandem mass spectrometry (HPLC-MS/MS) method. Using this method, a large extent of cis-ZEN formation was observed for ZEN contaminated maize germ oils when exposed to daylight which confirms that cis-ZEN can be a relevant food contaminant and should be considered in the analysis of food and feed. Furthermore, this work investigated the in vitro phase I metabolism of ZEN and cis-ZEN in rat and human liver microsomes by using HPLC-MS and -MS/MS analyses. The metabolic pathways of cis-ZEN were found to be essentially similar to ZEN including reduction and oxidation reactions generating α- and β-cis- zearalenol as well as 13- and 15-OH-cis-ZEN. A previously unidentified oxidative metabolic pathway for both isomers of ZEN results in the formation of cis-ZEN-11,12-oxide and ZEN-11,12-oxide in human liver microsomes. The estrogenicity of cis-ZEN and its reductive metabolites was assessed using the E-Screen assay. cis-ZEN proved to be slightly more estrogenic than ZEN. Biotransformation of cis-ZEN to β-cis-ZEL corresponds to a detoxification, whereas metabolism to α-cis-ZEL resembles a metabolic activation as its estrogenicity considerably exceeds that of cis-ZEN. The catecholic metabolites can be expected to show a decreased estrogenicity as demonstrated for 15-OH- ZEN. Independent of the estrogenic effects, the catecholic and epoxidic metabolites identified in this work can be expected to act genotoxic and carcinogenic. The epoxides in particular could fundamentally change the widely accepted view of ZEN causing adverse effects exclusively through endocrine disrupting actions. Hence, threshold values are regulated by law which reference a number of national and international standards for determination of said contaminants. In this context, soil is an especially difficult matrix, because several defining parameters can vary in a wide range. Organic matter content in particular has a crucial influence on extractability of contaminants because it largely governs formation of non-extractable or bound residues. For this study, four soils of different total organic carbon (TOC) content were generated from uncontaminated reference soil (RefeSol 01-A) and compost which were spiked to contaminant levels representative of the Bundes-Bodenschutz-Verordnung (BBodSchV). These test materials were extracted using accelerated solvent extraction (ASE), Soxhlet extraction, and liquid-liquid extraction. For the latter method, samples were initially extracted for 15 minutes employing acetone. The follow-up extraction step was carried out using cyclohexane and the extraction time varied between 15 minutes and 16 hours. After clean-up, extracts were analysed via gas chromatography-mass spectrometry (GC MS) and with a gas chromatograph equipped with an electron capture detector (GC ECD). Statistical analysis of the experimental values showed that recovery was not dependent on matrix TOC content. Also, no clear preference for a single method could be detected. Concerning liquid-liquid-extraction, it is merely observed that prolonged extraction time results in slightly higher recoveries, while moderate extraction times appear to yield the most robust results. Although Soxhlet and ASE show high recoveries, they also lead to larger standard deviations and are not suitable for determination of thermolabile pesticides. In addition to recovery by different extraction methods, the agreement of values obtained by ECD and MS was evaluated. For ECD, standard deviations are generally higher but overall the values obtained by MS and ECD are comparable. Laboratory simulation of SABs permits use of molecular-genetic methods coupled with geochemical and microscopic analyses to study weathering. Our in vitro model includes two free-living and symbiosis-competent, genetically tractable microorganisms: the cyanobacterium Nostoc punctiforme and the microcolonial rock-inhabiting fungus Knufia petricola. This genetically amenable cyanobacterium/fungus model biofilm allows development of quantitative methods tailored to the natural diversity of SABs. Within the framework of a forest soil inventory in Germany polycyclic aromatic hydrocarbons (PAH), polychlorinated biphenyls (PCB) and organochlorine pesticides (OCP) were analyzed in 1500 soil samples taken from 3 soil horizons at 447 sites. An analytical protocol was developed and validated for the simultaneous extraction and measurement of all analytes in humic rich soil at trace level range. It included pressurized liquid extraction (PLE) and a two step cleanup procedure consisting of gel permeation chromatography (GPC) followed by solid phase extraction (SPE). Quantification was carried out using gas chromatography combined with mass spectrometry (GC-MS). In line with the monitoring campaign a comprehensive quality assurance concept was implemented to guarantee constant analytical measurement conditions for two years. The protocol is applicable for field laboratories with a high level of automation and for analyzing high sample amounts. At most sampling sites pollutant concentrations gradually decrease with depth. Only few spots showed highest concentrations in the upper mineral soil. In the O-horizon the sum concentrations of 16 PAHs range from 104 to 14.000 µg/kg, of 6 PCBs from <1 to 106 µg/kg and the sum of DDT and metabolites range from <1 to ca. 4.000 µg/kg. The observed concentration of PAH, PCB and OCP reflect an ubiquitous background contamination at most sites. Relatively high concentrations of PAH and PCB are related to industrial areas and peak concentrations only of PAH are associated with open cast mining of brown coal areas. The spatial distribution of the contents of DDT and metabolites in German forest soils can clearly be attributed to historic application. A contribution of ubiquitous background pollution derived from long-range atmospheric transport is likely for some compounds in the studied area, e.g., DDT in the western part of Germany. The results generally reflect the distribution of POPs in densely populated and industrialized countries located in temperate regions]. Train-induced ground vibrations are studied in a measurement campaign where a test train has run with regularly varied speeds. The measured train-induced soil vibration at 2 to 100 m distance from the track is compared with the wave propagation due to hammer excitation and with the theoretical wave field. The strong influence of the soil and the train speed on the amplitudes and frequencies of the vibration has been analysed for passages of the locomotive and the carriages. - The generation of ground vibration by strong explosions has been studied on a large testing area with sandy soil. The propagating waves were measured in a regular grid of measuring points in 10 to 1000 m. Therefore, the dominance of certain waves at certain distances and the changes of compressional waves and Rayleigh waves could clearly be observed. The results are compared with impulse hammer measurements in the range of 5 to 50 m. - A drop test facility has been built on the testing area of the Federal Institute of Materials Research and Testing (BAM). Heavy masses (containers) of up to 200 t can be dropped from 10 m height on a big reinforced concrete foundation. The foundation was instrumented by accelerometers, strain gauges and pressure cells to give information about the loading condition and by geophones to measure the vibration of the surrounding soil and building. NMIs and DIs with proven metrological capabilities for the production and certification of such materials are necessary for the provision of quality data. Earth as a construction material has comparatively weak mechanical properties. It is also susceptible towards liquid water, were it can lose cohesion very quickly if certain moisture contents are exceeded. Repair concepts of structures built with such materials are therefore challenging. Historical earthen structures often exhibit damages in form of extensive cracking, which may have been caused by static or dynamic loads. Frequently these cracks are insufficiently or inappropriately repaired because of lack of knowledge and / or technology. In particular, the behaviour of crack repair by grouting poses a challenge in earthen materials and demands specific requirements for the grouting mortar, such as low water content, good water retention, low shrinkage. Grouting materials require additional specifications such as compatible strengths and Young’s modulus as well as good adhesion to the earthen substrates. In addition, grouts have to be sufficiently flowable to fill small cracks and voids without segregation or bleeding. Therefore, the rheological behaviour has to be well understood and controlled to gain the desired effects.The repair of cracks in earthen buildings is traditionally done by stuffing manually mortar into the gap. Naturally, this method is only usable for cracks with large widths. Another disadvantage is that cracks going through thicker walls cannot be completely reached by the tools used for stuffing the mortar into the crack. Lime based grouts for earthen materials were usually used for re-attachment of plasters but less for structural reasons.Due to the nature of earthen materials grouts based on formulated or hydraulic lime (according to the definition in EN 459-1) have to meet considerable demands on a variety of properties, which are related to compatibility, durability and in particular to the ability of being injected. In the first part of the study the mechanical characterisation of wallets tested under uniaxial compression and diagonal compression and walls tested under in-plane cyclic shear-compression loading is presented. The results were used to implement and validate the finite element simulations. The numerical modelling of the rammed earth samples tested is then discussed in the second part. A non-linear constitutive law based on the total strain rotating crack model (TSRCM) was employed as implemented in the DIANA® software [1]. The aim of the numerical analyses presented here is to simulate the behaviour of rammed earth under different inplane loading conditions. For the wallets, tests under static loading both macro- and micro-modelling approaches were considered for the simulation of the experimental tests. For the walls subjected to cyclic loading only the micro-modelling approach was applied for the simulation of the experimental tests. The respective FEM model was calibrated with the experimental results. The rammed earth layers were represented by continuum elements, the contact surfaces between layers by interface elements. This approach allowed assessing the influence of the apparent weakness of the interfaces between layers on the shear behaviour of rammed earth. The goal of the numerical simulation of the cyclic tests was to establish the adequacy of common analytical methods (e. g. used for masonry) applied to the analysis of rammed earth. Due to their global distribution, potential persistence, bioaccumulation and toxicity most of the BFRs of the first Generation are banned or restricted, and replaced by new ones. However, based on similar properties these new compounds may also pose a serious risk by causing adverse effects to human health and the environment. These alternative BFRs were grouped into two classes: „novel” and „emerging” BFRs. Emerging BFRs are defined as compounds that have been identified in any environmental compartments whereas novel BFRs are only detected in materials and/or goods above 0.1 wt.-%. The environmental fate of these alternative BFRs is partially unknown. Over the past years a multitude of studies on the photodegradation of BFRs in environmental matrices have been conducted to assess the photochemical behavior and fate in the environment. The photolytic debromination reaction is particularly induced among BFRs by UV exposure. The novel brominated triazine-based flame retardant tris-(2,4,6-tribromophenoxy)-s-triazine (TTBPTAZ) (Table 1) is a BFR that is mainly used in acrylonitrile butadiene styrene and high impact polystyrene. Both compounds can be found at low μg/L concentrations in surface water where they are introduced mainly by sewage treatment plants supplied by municipal wastewater. In order to disinfect wastewater in sewage treatment plants or water in swimming pools, chlorine can be added to water as gas or hypochlorite solution. This disinfectant has a high oxidation potential resulting in the formation of disinfection by-products, the estrogenicity and toxicity of which might differ from their precursor. In this work the behaviour of AHTN and HHCB upon treatment with sodium hypochlorite solution was investigated and chlorinated possible disinfection by-products could be identified as well as a carboxylic acids and lactones derived from AHTN and HHCB. The identified species were characterized by GC-MS, LC-MSn and 1H-NMR. The elucidated structures of potential disinfection by-products are verified by X-ray crystallography. Finally the estrogenicity between the parent analytes and generated transformation products were compared. For that task, MCF-7 assay has been chosen for being most relevant to humans. The liquid preservative was used until the end of the 80s. DDT crystal structures are formed on the wood surfaces by the "blooming" of chlorine compounds. Contaminated waste wood with natural biocide ageing, gilded and wood carved elements of an old picture frame and wooden samples with paint layers were provided by the Schlossmuseum Sondershausen. The removal of DDT was evaluated employing femtosecond and nanosecond laser radiation and cold atmospheric plasma technique using different working gases (air, nitrogen, and argon). Before laser application, a chlorine measurement is done by X-ray fluorescence (XRF) analysis as reference. After laser processing, the XRF analysis is used again at the same surface position to determine depletion rates. Additionally, a documentation and characterization of the sample surface is done before and after laser and plasma treatment using optical microscopy. For plasma processing with various systems a chlorine measurement is done by gas chromatographic-mass spectrometry (GCMS) analysis. For laser treatment a depletion of the chlorine concentration of 55-70% and 75% was achieved for 1064-nm nanosecond pulses and 800-nm femtosecond pulses, respectively. For the application of 30-fs laser pulses, no crystalline DDT residues remain on the sample surfaces. This holds also for the plasma processing with nitrogen as working gas. Earth as a construction material has comparatively weak mechanical properties. It is also susceptible towards liquid water, were it can lose cohesion very quickly if certain moisture contents are exceeded. Repair concepts of structures built with such materials are therefore challenging. Historical earthen structures often exhibit damages in form of extensive cracking, which may have been caused by static or dynamic loads. Frequently these cracks are insufficiently or inappropriately repaired because of lack of knowledge and / or technology. In particular, the behaviour of crack repair by grouting poses a challenge in earthen materials and demands specific requirements for the grouting mortar, such as low water content, good water retention, low shrinkage. Grouting materials require additional specifications such as compatible strengths and Young’s modulus as well as good adhesion to the earthen substrates. In addition, grouts have to be sufficiently flowable to fill small cracks and voids without segregation or bleeding. Therefore, the rheological behaviour has to be well understood and controlled to gain the desired effects. The repair of cracks in earthen buildings is traditionally done by stuffing manually mortar into the gap. Naturally, this method is only usable for cracks with large widths. Another disadvantage is that cracks going through thicker walls cannot be completely reached by the tools used for stuffing the mortar into the crack. Lime based grouts for earthen materials were usually used for re-attachment of plasters but less for structural reasons. Due to the nature of earthen materials grouts based on formulated or hydraulic lime (according to the definition in EN 459-1) have to meet considerable demands on a variety of properties, which are related to compatibility, durability and in particular to the ability of being injected. The goal of this study was to create a grout, which can be used to re-establish structural continuity in cracked earthen masonry or other massive earth walls (rammed earth and cob) with the focus on grouting cracks. Mono-incineration of the sludges completely destructs the organic pollutants in a first step. The incineration residues are ashes with a high phosphorus content that still contain heavy metal compounds above the limits for agricultural use. Furthermore, phosphorus in the ashes exhibits low bioavailability - a disadvantage in farming. Therefore, in a second thermochemical step heavy metals are removed and phosphorus transferred into mineral phases available for plants. The principle of the SUSAN-project is the separation of the small heavy metals fraction from the ashes to receive a phosphorus rich product suited as raw material for fertilisers that also contains SiO2, CaO, Al2O3, Fe2O3, MgO and K2O. The thermochemical process is based on evaporation of volatile heavy metal chlorides at 850 - 1000 °C. The sewage sludge ashes are mixed with chlorine donors such as MgCl2 or KCl, grained and treated in a thermal aggregate e.g. a rotary furnace. Here, we begin to resolve this issue by addressing the interaction between individual immunity and caste identity in the termite Neotermes castaneus. We characterize the transcriptomic responses of different castes to a general immune challenge by sequencing mRNAs from false workers, soldiers and reproductive following injection with a cocktail of heat-killed pathogens. We found many more genes to be differentially regulated in reproductives (1420 and 398 genes down- and upregulated respectively) than soldiers (90 and 108 genes) or false workers (135 and 71 genes). We address this issue by exploring the immune responses of different castes with de novo transcriptome sequencing. After injection with cocktail of heat-killed pathogens (Pseudomonas entomophila, Bacillus thuringiensis, Saccharomyces cerevisiae), the mRNAs from false workers, soldiers and reproductive of Neotermes canstaneus were sequenced. We found much more genes are regulated (1420 genes downregulated and 398 genes upregulated) in reproductives compared to soldiers (90 genes downregulated and 108 genes upregulated) and false workers (135 genes downregulated and 71 genes upregulated). In these upregulated genes, reproductives had 54 same genes as soldiers, while only 7 same genes as workers which almost is same number between soldiers and workers. In the downregulated genes, reproductives shared 50 and 41 same genes as soldiers and workers, respectively. Besides, soldiers and workers had 29 same downregulated genes. Cells respond to nutrient fluctuations by sensing and adapting their physiological state. Recent studies suggest phenotypic heterogeneity in isogenic populations as an alternative strategy in fluctuating environments, where a subpopulation of cells express a function that allows growth under conditions that might arise in the future. It is unknown how environmental factors such as nutrient limitation shape phenotypic heterogeneity in metabolism and whether this allows cells to respond to nutrient fluctuations. Here, we show that substrate limitation increases phenotypic heterogeneity in metabolism, and this heterogeneity allows cells to cope with substrate fluctuations. We subjected the N2-fixing bacterium Klebsiella oxytoca to different levels of substrate limitation and substrate shifts, and obtained time-resolved single-cell measurements of metabolic activities using nanometre-scale secondary ion mass spectrometry (NanoSIMS). We found that the level of NH4+ limitation shapes phenotypic heterogeneity in N2 fixation. In turn, the N2 fixation rate of single cells during NH4+ limitation correlates positively with their growth rate after a shift to NH4+ depletion, experimentally demonstrating the benefit of heterogeneity. The results indicate that phenotypic heterogeneity is a general solution to two important ecological challenges - nutrient limitation and fluctuations - that many microorganisms face. Currently, we use NanoSIMS to develop a new approach that defines functionally-relevant, phenotypic biodiversity in microbial systems. We investigate the causal drivers of microbe-associated shifts in host phenotype by examining how feeding behaviour is modulated by a bacterial infection in an omnivorous cockroach: Blatta orientalis. We conducted food-choice experiments after challenging hosts with the common entomopathogenic soil bacterium Pseudomonas entomophila to understand the impact of bacterial pathogens on host macronutrient preference. We find that immune challenge by this bacterium drives a sharp decline in carbohydrate intake and results in a relative increase in the ratio of protein to carbohydrate consumed. Additionally, infected cockroaches reduce their overall nutrient intake. We show for the first time that cockroach feeding behaviour is dynamically modulated by a pathogen. In contrast to studies on Spodoptera moths, this modulation does not impact any of the immune parameters we measured: it does not affect the abundance of immune related proteins in the hemolymph, hemolymph antimicrobial activity, or survival. This leads to the possibility that as long-lived omnivorous species B. orientalis may be better adapted to unpredictable variation in food availability and quality. An illness-induced anorexia-like response which is thought to assist hosts in limiting nutritional resources available to pathogens is therefore the most likely cause of the observed phenotype. Reduction of carbohydrate intake by sick individuals would be consistent with such an explanation. In another cockroach species Blatella germanica we also investigate whether in addition to pathogens, host microbiota especially gut commensals are causally responsible for shifts in host behavioural phenotype and immune competence. For this purpose, artificial MP/soil mixture samples consisting of standard soils or sand with defined ratios of MP (0,5 – 10 mass% polymer) from polyethylene (PE), polypropylene (PP), polystyrene (PS) and polyethylene terephthalate (PET) were prepared. MP particles with diameters < 2 mm and < 125 µm were obtained from industrial polymer pellets after cryo-milling. Spectra of these mixtures were collected by (i) a process FT-NIR spectrometer equipped with a reflection probe, (ii) by a cw-process Raman spectrometer and (iii) by a time-gated Raman spectrometer using fiber-optic probes. The evaluation of process-spectra was performed by chemometric methods. Whereas MP could be detected by Raman spectroscopy in coastal sand at 0.5 mass%, in standard soil detection of MP was limited to 10 – 5 mass% with the large fraction, but samples containing particles of the 125µm mass-fraction yielded no positive result at all. One reason for the lacking sensitivity could be fluorescence by soil organic matter and thus, in a next test time-gated Raman spectroscopy was applied. However, although being indeed more sensitive to the small particles this method failed at MP < 5 mass% indicating that fluorescence was not the major problem. Finally, FT-NIR was tested. Depending on the polymer, MP contents of 0,5 or 1 mass%, respectively, could be detected in standard soils and polymers identified. The realm of SAB is continually widened by modern materials, and the settlers on these exposed solid surfaces always include melanized, stress-tolerant microcolonial ascomycetes. After their first discovery on desert rock surfaces, these melanized chaetothyrialean and dothidealean ascomycetes have been found on Mediterranean monuments after biocidal treatments, Antarctic rocks and solar panels. New man-made modifications of surfaces (e.g., treatment with biocides or photocatalytically active layers) accommodate the exceptional stress-tolerance of microcolonial fungi and thus further select for this well-protected ecological group. Melanized fungal strains were isolated from a microbial community that developed on highly photocatalytic roof tiles after a long-term environmental exposure in a maritime-influenced region in northwestern Germany. Four of the isolated strains are described here as a novel species, Constantinomyces oldenburgensis, based on multilocus ITS, LSU, RPB2 gene phylogeny. Their closest relative is a still-unnamed rock-inhabiting strain TRN431, here described as C. patonensis. Both species cluster in Capnodiales, among typical melanized microcolonial rock fungi from different stress habitats, including Antarctica. These novel strains flourish in hostile conditions of highly oxidizing material surfaces, and shall be used in reference procedures in material testing. In this study, PAC containing activated sludge from four WWTP were examined by two-step thermogravimetric analysis (TGA) with heating up to 600°C in N2 and subsequently in synthetic air. Direct quantification of PAC according to temperature specific weight losses was possible for one WWTP. Quantification by combining specific mass losses was found to be an alternative direct method, with a detection limit of 1.2% PAC in dry sample mass. Additionally, evolved gas analysis (EGA) by infrared-spectroscopy (FTIR) during TGA revealed interaction mechanisms between PAC and activated sludge. Aliphatic compounds from activated sludge were identified as major substances influenced by PAC. In derivative thermogravimetry (DTG), a typical double peak at approximately 300°C was found to be related to carbonylic species with increased evolution of acetic acid in aged activated sludge. TGA and EGA are promising tools to understand, control and optimise the application of PAC in advanced wastewater treatment. Our approaches rely on the hypothesis that Termite associated bacteria are likely to produce potent antibiotics to defend their hosts against entomopathogenic microorganisms. Termite nests and guts harbor suitable, highly diverse microbiomes in which bacterial taxa are present known to potentially produce natural compounds. In a first step the diversity of Coptotermes species nest microbiomes was assessed carefully by using 16S rDNA amplicon sequencing on the Illumina MiSeq platform and nest material was selected to retrieve viable cells by using Nycodenz density gradient centrifugation. In order to analyze the diversity of the culturable termite nest microbiome, bacterial cells were either distributed in 384-well plates (approach 1) or encapsulated in small spheric agarose beads by an high throughput microfluidics technique (approach 2). Cultures obtained from approach 1 were scaled-up in 96-well Duetz-systems for characterization of diversity and for rapid supernatant screening using the bioluminescence-labeled E. coli pFU166. The generated droplets of approach 2 simultaneously received a small population of GFP-tagged Gram negative screening cells and were sorted for low fluorescence using FACS. After elimination of redundancy we performed a fast scale-up of active strains. Implementation of this pipeline allows us to prioritize antibiotics producing strains in a ultra-high throughput fashion and by cultivation of broad diversity in our approches. N2 - Construction products and waste materials used for construction can be in contact with the environment and may release potential harmful compounds. Information on the total content of these substances in the product is not sufficient to assess its envrionmental impact since it does not consider realistic exposure conditions.The impact of these compounds is often assessed by using the total content in the product. This estimation lacks realistic exposure conditions. Concerning the pathway to soil and groundwater by contact with rain or seepage water leaching tests, which were developed and standardized by the European Committee for Standardization, are available. While for secondary construction products and waste this investigation of the leaching behaviour is standard procedure and is already part of regulation in Germany (draft ordinance on reuse of mineral waste) and within Europe (landfill directive) the release from primary construction products got in the focus more recently. Several monitoring studies found unexpected high concentrations of substances used as herbicides and fungicides in surface water and stormwater originating from urban areas. As some of these compounds are even banned for the use in agricultural applications in Europe alternative sources as roof materials and façade coatings exposed to rain were suggested. Further field and laboratory tests confirmed construction products as sources of these substances in water. The aim of this presentation is to show exemplary results of existing leaching methods and underline the strength and weaknesses of the test system with selected examples from our work. Organics (pesticides, PAHs, PFOS, etc.) and heavy metals (Hg, Cd, Ni, Pb and As) represent target parameters. Laboratories performing sampling and tests in this field regulated by respective EU directives, need strong support in order to establish a quality system. It is necessary to provide appropriate calibrators i.e. matrix CRMs relates to the unique sample matrices representing typical samples in the geomorphological and anthropological sense. In addition to that, bearing in mind the complexity and instability of environmental samples, it is very difficult to obtain appropriate referents materials with no local providers. Our project is aiming to develop capacity to produce CRMs for environmental analysis by transferring the theoretical and practical know-how between the partners and combining their skills to focus on environmental CRM production in accordance with ISO Guide 34. To enable a fast detection and localization of these kind of leaks, we developed a novel robotic platform for aerial remote gas sensing. Spectroscopic measurement methods for remote sensing of selected gases lend themselves for use on mini-copters, which offer a number of advantages for inspection and surveillance over traditional methods. No direct contact with the target gas is needed and thus the influence of the aerial platform on the measured gas plume can be kept to a minimum. This allows to overcome one of the major issues with gas-sensitive mini-copters. On the other hand, remote gas sensors, most prominently Tunable Diode Laser Absorption Spectroscopy (TDLAS) sensors have been too bulky given the payload and energy restrictions of mini-copters. Here, we present the Unmanned Aerial Vehicle for Remote Gas Sensing (UAV-REGAS), which combines a novel lightweight TDLAS sensor with a 3-axis aerial stabilization gimbal for aiming on a versatile hexacopter. The proposed system can be deployed in scenarios that cannot be addressed by currently available robots and thus constitutes a significant step forward for the field of Mobile Robot Olfaction (MRO). It enables tomographic reconstruction of gas plumes and a localization of gas sources. We also present first results showing its performance under realistic conditions. They are used in concrete, mortar and grout in buildings, roads and civil engineering works. LWA are commonly produced by thermal treatment of natural raw materials such as clay, shale and other clayey or igneous rocks at temperatures between 1000 °C and 1300 °C. Expanded clay, expanded shale, vermiculite and expanded perlite are well-known aggregates used in the construction industry today. But porous LWA can also be manufactured from mineral residues, such as waste glass or masonry rubble, by thermal expending process. The source materials must satisfy two conditions. First, their chemical composition must be in balance between fluxing agents, silica and alumina to reach pyroplastic conditions at an economically viable temperature to develop sintering and melting phases of proper viscosity. Second, the material should contain some components, which ensure gas release during the partial melting, to form expanded porous particles. Lightweight aggregates usually show a heterogeneous and macroporous pore structure. Their solid skeleton is continually opened by large bulky pores whereas the skeletal walls contain many small pores. Additionally, the grains consist of partly melted areas. The specific microstructural and morphological characteristics of different LWA are strongly influenced by the great variety of source materials and the particular manufacturing conditions. Recycling granules (RC granules), which are lightweight aggregates made from masonry rubble containing about 50 % brick material, are already described in terms of manufacturing process, engineering properties and microstructure in. The objective of this study is to compare the chemical, physical, mineralogical and microstructural characteristics of RC granules with those of commercially produced expanded clay aggregates. Results of the ICP optical emission spectrometry, X-ray diffraction, thermal analysis, fusibility tests, particle shape analysis, porosity measurements, mercury intrusion porosimetry und environmental scanning electron microscopy are presented. The microstructural characteristics are correlated with engineering properties of the lightweight aggregates, such as water absorption and particle strength. A variety of spectroscopic and spectrometric approaches have been proposed to develop a fast and reliable pollen identification using specific molecular information. Amongst them, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) shows a high potential for the successful investigation of such complex biological samples. Based on optimized MALDI sample preparation using conductive carbon tape, the application of multivariate statistics (e.g. principal components analysis, PCA) yields an enormous improvement concerning taxonomic classification of pollen species compared to common microscopic techniques. Since multivariate evaluation of the recorded mass spectra is of vital importance for classification, it’s helpful to implement the applied sequence of standard Matlab functions into a graphical user interface (GUI). In this presentation, a stand-alone application (GUI) is shown, which provides multiple functions to perform fast multivariate analysis on multiple datasets. The use of a GUI enables a first overview on the measured dataset, conducts spectral pretreatment and can give classification information based on HCA and PCA evaluation. T1 - Optimisation of a Pyrometallurgical Niobium and Tantalum Recyling Process with on-line-LIBS N2 - Tantalum and niobium are essential for the development of electronics towards a more and more compact design, but without reducing their performance today. Especially in smart phones and tablets, tantalum and niobium capacitors with high charge densities have already become almost indispensable. However, tantalum as a critical raw material is still a problem for the sustainable production of electronics. Due to this fact, the existence of efficient recycling processes especially in Europe is becoming even more important nowadays. The Bundesanstalt für Materialforschung und -prüfung (BAM) is currently working on the optimisation of an existing pyrometallurgical tantalum recycling process. Optimisation of this industrial process is carried out in a small-scale electric arc furnace (480 kVA, capacity approx. 150 kg/h) at BAM using a new and innovative equipment for on-line analysis of high temperature processes. The aim of this project is to identify the best timing for an optimum slag tapping, when the slag is lower than minimum targeted tantalum concentration. Hence, LIBS (Laser induced breakdown spectroscopy) is used to identify the chemical composition of the slag layer and during slag tapping. The on-line-LIBS prototype of BAM enables an in-situ measurement of the element distribution in the melt after calibration on the slag system. First results of this joint research project will be presented including on-line-LIBS-measurements and thermodynamic and kinetic aspects of the process. A1 - Koch, Matthias T1 - Investigation of ionophore antibiotics and their transformation products by using electrochemistry coupled to LC-MS N2 - Ionophore antibiotics are used to cure and prevent coccidiosis by chicken especially in broiler farming. The residues are found not only in food products (chicken and eggs) but also in the environment (manure, soil or water). In this work the ionophores monensin (MON), salinomycin (SAL), maduramicin (MAD) and lasalocid (LAS) are investigated aiming to study their transformation products (TPs) through biotransformation processes. Biotransformation can be divided into two phases, phase I: oxidation, reduction or hydrolysis and Phase II: conjugation reactions. It is necessary to further examine the biotransformation pathways to determine TPs to be able to detect residues more specifically in different matrices. The technique of electrochemistry (EC) offers the opportunity to simulate biotransformation processes and to generate TPs for further analysis. The combination of EC with liquid chromatography and mass spectrometry (EC-LC-MS) provide a fast and simple tool to separate and determine the EC-generated TPs. The electrochemical flow through cell is coupled to the (LC)-MS system, allowing the reaction mixture to be separated by a RP-18 column and then analyzed in the MS. The oxidation products are generated at different potentials between 0.0 – 2.5 V vs. Pd/H2 using glassy carbon or boron doped diamond as working electrode materials . The results show a broad spectrum of different TPs depending on used solvents and working electrode materials. Among the generated TPs already known as well as unknown TPs of the drugs can be found. Further investigations on structure elucidation of unkown TPs are planned. The timely identification of these contaminants is of utmost importance, since they directly affect water quality and represent a risk for wildlife and human health even in trace amounts. In this work, we develop a simple system for the on-field detection of total petroleum hydrocarbons (TPH) in water and soil, the "Spectrocube". The test is based on the measurement of the fluorescence signal emitted by the molecular rotor 4-DNS-OH dye. This dye is embedded in a hydrophobic polymeric matrix (PVDF), avoiding interactions of water with the dye and providing a robust support for use in test-strip fashion. The test-strip’s fluorescence intensity increases linearly at low concentrations of TPH, reaching a saturation value at higher concentrations. For excitation and evaluation of the test-strip fluorescence, a simple miniature optical system was designed. The system works semi-quantitatively as solvent-free TPH detection kit, as well as quantitatively when using a simple cyclopentane extraction step. To simplify the fluorescence read-out, the device is coupled to a tablet computer via Bluetooth, running a self-programmed software ("app").
