Although human activity contributes to the acceleration of the process, bacteria have a self-driven stabilisation mechanism to protect themselves from such and other external threats: biofilm formation. Nonetheless, it is the adhesion of a single bacterial cell to a surface that triggers the formation of such network of biomolecules and microorganisms, as well as its hazardous consequences. The main objective of this work was to quantify the adhesion force of a single E. coli cell on a Ti substrate via the AFM-related single-cell force spectroscopy, with both the cell and the substrate material being of high clinical relevance. A set of 25 x 25 force displacement curves was acquired with a maximum force of 3.2 nN without dwell time, yielding a topography map and an adhesion force map that showed to be correlated. A mean adhesion force of 0.85 ± 0.175 nN was measured and the presence of cell appendages on the bacterial cell wall was verified through individual force-displacement curves. Bacterial viability was assessed after the measurements via live/dead staining. This presents new challenges to industry and the scientific community alike. The storage and transport of hydrogen, which is nowadays mainly realized by austenitic stainless steels, remains problematic, which is due to the degradation of mechanical properties and the possibility of phase transformation by hydrogen diffusion and accumulation. The development of materials and technologies requires a fundamental understanding of these degradation processes. Therefore, studying the behavior of hydrogen in austenitic steel contributes to an understanding of the damage processes, which is crucial for both life assessment and safe use of components in industry and transportation. As one of the few tools that is capable of depicting the distribution of hydrogen in steels, time-of-flight secondary ion mass spectrometry was conducted after electrochemical charging. To obtain further information about the structural composition and cracking behavior, electron-backscattered diffraction and scanning electron microscopy were performed. Gathered data of chemical composition and topography were treated employing data fusion, thus creating a comprehensive portrait of hydrogen-induced effects in the austenite grade AISI 304L. Specimens were electrochemically charged with deuterium instead of hydrogen. This arises from the difficulties to distinguish between artificially charged hydrogen and traces existing in the material or the rest gas in the analysis chamber. Similar diffusion and permeation behavior, as well as solubility, allow nonetheless to draw conclusions from the experiments. Mittels Oberflächenbehandlung, z.B. durch Plasmafeinreinigung bzw. –aktivierung, können die Grenzflächern solcher Polymere - bezogen auf die Eignung als Fügeteilpartner – optimiert werden. Schnelle Prüfverfahren zur Ermittlung von Verbundeigenschaften sind somit erforderlich. Das Messprinzip Centrifugal Adhesion Testing Technologie verwendet die radial gerichtete Zentrifugalkraft als Belastungsgröße des zu testenden Verbunds, die durch Variation der Drehzahl gesteuert wird. Bei Erreichen der kritischen Beanspruchung trennt sich ein aufgeklebter Prüfstempel vom Substrat. Zeitgleich wird ein positionsbezogenes IR-Signal aus dem drehenden Rotor an das Basisgerät gesendet und zusammen mit der Drehzahl an die PC-Software übertragen. Für jede Probe wird die Bruchkraft berechnet und in Echtzeit angezeigt. Der Adhesion Analyser LUMiFrac® untersucht bis zu acht Prüfkörper unter identischen Bedingungen. So kann der Effekt einer Plasmabehandlung von Bauteilen auf die Verbundfestigkeit zeitsparend bestimmt werden. Die gezielte Veränderung unterschiedlicher Plasma-Parameter, wie beispielsweise, die Plasmazusammensetzung, der Abstand vom Plasmabrenner zur Oberfläche oder die Verweildauer unter dem Brenner, beeinflußt die Festigkeit des Klebsystems und ist dem LUMiFrac direkt quantifizierbar. They show exceptional application properties, such as high-strength and ductility or corrosion resistance. Future HEA/MEA-components could be exposed to hydrogen containing environments like vessels for cryogenic or high-pressure storage where the hydrogen absorption and diffusion in these materials is of interest. In our study, we investigated the HEA Co20Cr20Fe20Mn20Ni20 and the MEA Co33.3Cr33.3Ni33.3. For hydrogen ingress, cathodic charging was applied and diffusion kinetic was measured by high-resolution thermal desorption spectros-copy using different heating rates up to 0.250 K/s. Peak deconvolution resulted in high-temperature desorption peaks and hydrogen trapping above 280 °C. A total hydrogen concentration > 40 ppm was identified for the MEA and > 100 ppm for HEA. This indicates two important effects: (1) delayed hydrogen diffusion and (2) considerable amount of trapped hydrogen that must be anticipated for hydrogen assisted cracking phenomenon. Local electrochemical Volta potential maps had been measured for the hydrogen free condition by means of high-resolution Scanning Kelvin Probe Force Microscopy (SKPFM). A material is a nanomaterial if 50 % or more of the particles are in the size range 1-100 nm. The implementation of the definition in a regulatory context challenges measurement methods to reliably identify nanomaterials and ideally also nonnanomaterials as substance or product ingredient as well as in various matrices. The EU FP7 NanoDefine project [www.nanodefine.eu] addressed these challenges by developing a robust, readily implementable and cost-effective measurement strategy to decide for the widest possible range of materials whether it is a nanomaterial or not. It is based on existing and emerging particle measurement techniques evaluated against harmonized, material-dependent performance criteria and by intra- and inter-lab comparisons. Procedures were established to reliably measure the size of particles within 1-100 nm, and beyond, taking into account different shapes, coatings and chemical compositions in industrial materials and consumer products. Case studies prove their applicability for various sectors, including food, pigments and cosmetics. A main outcome is the establishment of an integrated tiered approach including rapid screening (Tier 1) and confirmatory methods (tier 2), a decision support flow scheme and a user manual to guide end-users, such as manufacturers, in selecting appropriate methods. Another main product is the “NanoDefiner” e-Tool which implements the flow scheme in a user-friendly software and guides the user in a semi-automated way through the entire decision procedure. It allows a cost-effective selection of appropriate methods for material classification according to the EC's nanomaterial definition and provides a comprehensive report with extensive explanation of all decision steps to arrive at a transparent identification of nanomaterials as well as non-nanomaterials for regulatory purposes. However it is difficult to correlate the functional properties of TiO2 nanomaterials to the properties at single nanoparticle level due to the high polydispersity in shape, size and surface features of the currently available TiO2 nanoparticles (NPs). Although intensive experimental and theoretical studies have been conducted on the reactivity of different surfaces of metal oxides such as TiO2 much less attention is paid on the dependence of functional properties, like photocatalytic activity, dye adsorption, open circuit potential and fill factor in dye sensitized solar cells, on crystal facets in different orientations. One of the goal of SETNanoMetro project was the development of design rules to tune crystal facets of TiO2 NPs in order to optimize and control functional properties. In the present work we have developed a series of design rules in order to obtain sets of anatase TiO2 NPs with low polydispersity and to tune their shape and size by hydrothermal processing of Ti(IV)- Triethanolamine complex in presence of different shape controllers. Through a careful experimental design, a predictive soft model was developed. The model is able to predict the synthesis outcome allowing to tune the shape factor from 5 (prisms) to 1.5 (bipyramids) to 0.2 (platelets). This allows to control the main crystal facets exposed ranging from (100) to (001). Due to the dependence of functional properties of nanomaterials on shape distribution and not only size, the availability of NPs sets with uniform and well defined and tunable shapes can be of paramount relevance in order to produce reference nanomaterials for shape measurement. Zu diesen Effekten zählen Wechselwirkungen zwischen den Polymeren und Ionen in der Porenlösung, frühe Phasenbildung, zeitabhängige und kompetitive Adsorption zwischen anionischen Polymeren und Sulfationen, Bildung von Phasen in der Porenlösung sowie Morphologieänderungen an Partikeloberflächen. Die frühe Hydratation von Zement, die durch Lösungs- und Fällungsprozesse angetrieben wird, beeinflusst diese Effekte erheblich. Das permanente Ungleichgewicht der Porenlösung führt zu Veränderungen der Partikeloberflächen, welches widerum zur Folge hat, dass rheometrische Messungen dieser Zementleime anfällig für Streuungen sind. Um die Einflüsse aus der Zementhydratation zu minimieren, wurden die rheometrischen Untersuchungen mit Zement in Porenlösung durchgeführt. Die Experimente wurden mit verschiedenen Feststoffvolumenfraktionen durchgeführt und mit den Ergebnissen identischer Systeme mit Wasser anstelle von Porenlösung verglichen. Zusätzlich wurden die gleichen Systeme mit Zugabe von Polycarboxylatethern untersucht. Die Ergebnisse zeigen, dass die Leime mit Wasser niedrigere Werte für Fließgrenze und plastische Viskosität aufweisen, als die Systeme mit Porenlösung. Während die Polymere eine Verminderung der Fließgrenze zur Folge hatte, war die Wirkung von Polymeren auf die plastische Viskosität vernachlässigbar. The reasons for this lie in the availability of high average powers at pulse repetition rates in the several 100 kHz range. The advantages of using ultrashort laser pulses in terms of processing precision can thus be fully exploited. However, high laser intensities on the workpiece can also lead to the generation of unwanted X-rays. Even if the emitted X-ray dose per pulse is low, the accumulated X-ray dose can become significant for high-repetition-rate laser systems so that X-ray exposure safety limits must be considered. The X-ray emission during ultrashort pulse laser processing was investigated for a pulse duration of 925 fs at 1030 nm wavelength and 400 kHz repetition rate. Industrially relevant materials such as steel,aluminum and glass were treated. Tungsten served as reference. X-ray spectra were recorded, and X-ray dose measurements were performed for laser treatment in air. For laser intensities > 2 × 10^13 W/cm2, X-ray doses exceeding the regulatory exposure limits for members of the public were found. Suitable X-ray protection strategies are proposed.
