In this work we want to introduce a first of its kind test artifact. Test artifacts are standardized 3D models with specific geometric feature to evaluate the performance of writing parameters. Test artifacts are already common in other 3D additive manufacturing technologies e.g. Selective Laser Melting. The test artifact introduced in this work was developed in particular to accommodate 1) the high geometrical resolution of DLW structures and 2) the limited possibilities to examine the resulting structure. Geometric accuracy, surface adhesion as well as confocal raman spectroscopy results were considered when evaluating the design of the test artifact. We will explain the individual features and design considerations of our DLW test artifact. The difference between two slicers, Cura and 3DPoli, and the implications on measured feature sizes and the general shape is quantified. The measured geometries are used to derive a general design guide for a specific combination of photoresist, laser power and scanning speed and to analyse the geometric accuracy of a structure produced using these guidelines. Common methods for the quantification of bound protein are measuring protein in the supernatant before and after coating and calculating the difference. This popular approach has the potential for a significant overestimation of the amount of immobilized protein since layers not directly bound to the surface (soft protein corona) are usually lost during washing and handling. Only the layer directly bound to the surface (hard corona) can be used in subsequent assays. A simplified amino acid analysis method based on acidic hydrolysis and RP-HPLC-FLD of tyrosine and phenylalanine (aromatic amino acid analysis, AAAA) is proposed to directly quantify protein bound to the surface of gold nano- and latex microparticles. The results are compared with indirect methods such as colorimetric protein assays, such as Bradford, bicinchoninic acid (BCA), as well as AAAA of the supernatant. For both particle types, these indirect quantification techniques show a protein overestimation of up to 1700% compared to the direct AAAA measurements. In addition, protein coating on latex particles was performed both passively through adsorption and covalently through EDC/sulfo-NHS chemistry. Our results showed no difference between the immobilization methodologies. This finding suggests that usual protein determination methods are no unambiguous proof of a covalent conjugation on particles or beads. This underlines the importance of antibodies as universal bioanalytical reagents. However, little attention is given to the reproducibility crisis that scientific studies are still facing to date. In a recent study, not even half of all research antibodies mentioned in publications could be identified at all. This should spark more efforts in the search for practical solutions for the traceability of antibodies. For this purpose, we used thirty-five monoclonal antibodies against SARS-CoV-2 to demonstrate how sequence-independent antibody identification can be achieved by simple means applied onto the protein. First, we examined the intact and light chain masses of the antibodies relative to the reference material NIST-mAb 8671. Already half of the antibodies could be identified based solely on these two parameters. In addition, we developed two complementary peptide mass fingerprinting methods with MALDI-TOF-MS that can be performed in 45 minutes and had a combined sequence coverage of over 80%. One method is based on the partial acidic hydrolysis of the protein by 5 mM of sulfuric acid at 99 °C. Furthermore, we established a fast way for a tryptic digest without an alkylation step. We were able to show that the distinction of clones is possible simply by a brief visual comparison of the mass spectra. In this work, two clones originating from the same immunization gave the same fingerprints. Later, a hybridoma sequencing confirmed the sequence identity of these sister clones. In order to automate the spectral comparison for larger libraries of antibodies, we developed the online software ABID 2.0 (https://gets.shinyapps.io/ABID/). This open-source software determines the number of matching peptides in the fingerprint spectra. We propose that publications and other documents critically relying on monoclonal antibodies with unknown amino acid sequences should include at least one antibody fingerprint. By fingerprinting an antibody in question, its identity can be confirmed by comparison with a library spectrum at any time and context. The main bottleneck of this method is the choice of regularization parameters which differs for each experiment. To avoid time-consuming manually selected regularization parameters, we propose a learned block-sparse optimization approach using an iterative algorithm unfolded into a deep neural network. More precisely, we show the benefits of using a learned block iterative shrinkage thresholding algorithm that is able to learn the choice of regularization parameters. In addition, this algorithm enables the determination of a suitable weight matrix to solve the underlying inverse problem. Therefore, in this paper we present the algorithm and compare it with state of the art block iterative shrinkage thresholding using synthetically generated test data and experimental test data from active thermography for defect reconstruction. Our results show that the use of the learned block-sparse optimization approach provides smaller normalized mean square errors for a small fixed number of iterations than without learning. Thus, this new approach allows to improve the convergence speed and only needs a few iterations to generate accurate defect reconstruction in photothermal super resolution imaging. Photothermal imaging is a well-known technique in active thermography for nondestructive inspection of defects in materials such as metals or composites. A grand challenge of active thermography is to overcome the spatial resolution limitation imposed by heat diffusion in order to accurately resolve each defect. The photothermal SR approach enables to extract high-frequency spatial components based on the deconvolution with the thermal point spread function. However, stable deconvolution can only be achieved by using the sparse structure of defect patterns, which often requires tedious, hand-crafted tuning of hyperparameters and results in computationally intensive algorithms. On this account, Photothermal-SR-Net is proposed in this paper, which performs deconvolution by deep unfolding considering the underlying physics. This enables to super resolve 2D thermal images for nondestructive testing with a substantially improved convergence rate. Since defects appear sparsely in materials, Photothermal-SR-Net applies trained blocksparsity thresholding to the acquired thermal images in each convolutional layer. The performance of the proposed approach is evaluated and discussed using various deep unfolding and thresholding approaches applied to 2D thermal images. Subsequently, studies are conducted on how to increase the reconstruction quality and the computational performance of Photothermal-SR-Net is evaluated. Thereby, it was found that the computing time for creating high-resolution images could be significantly reduced without decreasing the reconstruction quality by using pixel binning as a preprocessing step. This relies on alterations in mitochondrial physiology. Which signals and molecules regulate those alterations remains unclear. We recently reported, that the mitochondrial protein TCAIM inhibits activation-induced changes in mitochondrial morphology and function and thus, CD effector T cell formation. Using conditional TCAIM knock-in (KI) and knockout (KO) mice, w now show that it also applies to CD8+ T cells and more importantly, delineate the molecular processes in mitochondria by which TCAIM controls effector cell differentiation. TCAIM KI resulted in reduced activation-induced HIF1α protein expression. Metabolomics and transcriptional data in combination with mathematical flux modeling revealed an impaired induction of anabolic pathways, especially of the mevalonate pathway and cholesterol biosynthesis in TCAIM KI CD8+ T cells. Addition of cholesterol completely rescued HIF1α protein expression, activation and proliferation of TCAIM KI CD8+ T cells. At the molecular level, TCAIM delayed activation-induced mitochondria-ER contact (MERC) formation by binding to MERC promoting proteins such as RMD3 and VDAC2. In summary, we demonstrate that TCAIM suppresses effector cell differentiation by inhibiting MERC formation, which induce HIF1α-mediated increase in cellular metabolism and cholesterol biosynthesis. T1 - High-purity corundum as support for affinity extractions from complex samples T2 - Preprints N2 - Nonporous corundum powder, known as an abrasive material in the industry, was functionalized covalently with protein binders to isolate and enrich specific proteins from complex matrices. The materials based on corundum were characterized by TEM, ESEM, BET, DLS, and zeta potential measurements. The strong Al-O-P bonds between the corundum surface and amino phosphonic acids are used to introduce functional groups for further conjugations. The common cross-linker glutaraldehyde was compared with a hyperbranched polyglycerol (PG) of around 10 kDa. The latter is oxidized with periodate to generate aldehyde groups that can covalently react with the amines of the surface and the amino groups from the protein via a reductive amination process. The amount of bound protein was quantified via aromatic amino acid analysis (AAAA). This work shows that oxidized polyglycerol can be used as an alternative to glutaraldehyde. With polyglycerol, more of the model protein bovine serum albumin (BSA) could be attached to the surface under the same conditions, and lower nonspecific binding (NSB) was observed. As a proof of concept, IgG was extracted with protein A from crude human plasma. The purity of the product was examined by SDS-PAGE. A binding capacity of 1.8 mg IgG per g of corundum powder was achieved. The advantages of corundum are the very low price, extremely high physical and chemical stability, pressure resistance, favorable binding kinetics, and flexible application. For these substances, Environmental Quality Standards (EQS), equivalent to the maximum allowable concentrations, had been defined at very low levels. EQS values refer to the whole water body, i.e. include contaminants which are associated to suspended solids or colloids present in the natural water. For several priority pollutants primary analytical methods capable of measuring the target analytes at EQS level in natural water are still missing. Die Suffosion kann zu Problemen mit der Stabilität des Korngerüstes von Erdstoffen führen. Relevante Anwendungsgebiete sind z.B. im Verkehrswegebau, Hochwasserschutz und allgemeinen Erdbau zu finden. It brought together different stakeholders including sensor manufacturers, end-users, certification bodies and independent evaluators to ensure the optimum use of low-cost and reliable hydrogen sensors. Project partners analysed sensor performance in real-life applications in industrial environments and identified increased requirements for sensors and for regulations, codes and standards. H2SENSE also facilitated the safe use and implementation of hydrogen as an alternative fuel by ensuring the correct use of effective hydrogen detection devices. European scientists worked together with colleagues from the National Renewable Energy Laboratory (NREL) in Colorado, USA. They pooled their knowledge of developments in hydrogen sensor technology as well as deployment and commercialisation strategies. These benefits will be continued through trans-Atlantic inter-laboratory sensor testing programmes in which EU and US laboratories perform complementary tests and exchange results. Mono-incineration of the sludges completely destructs the organic pollutants in a first step. The incineration residues are ashes with a high phosphorus content that still contain heavy metal compounds above the limits for agricultural use. Furthermore, phosphorus in the ashes exhibits low bioavailability - a disadvantage in farming. Therefore, in a second thermochemical step heavy metals are removed and phosphorus transferred into mineral phases available for plants. The principle of the SUSAN-project is the separation of the small heavy metals fraction from the ashes to receive a phosphorus rich product suited as raw material for fertilisers that also contains SiO2, CaO, Al2O3, Fe2O3, MgO and K2O. The thermochemical process is based on evaporation of volatile heavy metal chlorides at 850 - 1000 °C. The sewage sludge ashes are mixed with chlorine donors such as MgCl2 or KCl, grained and treated in a thermal aggregate e.g. a rotary furnace. At the same time a transfer of non soluble P-compounds into Mineral phases available for plants takes place. Besides technological development of the thermochemical process, the products fertilisation performance, the product design, the market for the products and the sustainability of the whole process chain are emphasised in the SUSAN-project. Es soll insbesondere die systematische Entwicklung der Zulassungsanforderungen für diese Art von Druckgefäßen in Deutschland berücksichtigen. Explizit sind im Rahmen des Auftrages zu betrachten: Bauarten von geschweißten Gasflaschen aus Stahl für Gase, wie von der EN 13322-1:2006 und EN 1442:2008 erfasst, deren Zulassung ohne jedwede Art von Auflagen oder Einschränkungen auf der Basis der Druckgasverordnung (vom 20. Juni 1968) oder nachfolgend der Druckbehälterverordnung erteilt wurde. Aufgrund der systematischen Betrachtung können Bauarten, deren Zulassungen unter irgendeiner Art von Betriebsauflagen oder sonstigen Einschränkungen erteilt worden sind, nicht von diesem Gutachten erfasst werden. Dieses Gutachten analysiert die Unterschiede in den Beschaffenheitsanforderungen nach Druckgas- bzw. Druckbehälterverordnung gegenüber dem geltenden RID/ADR 2009. Es soll den benannten Stellen nach Richtlinie 1999/36/EG (TPED) eine Vereinfachung ihrer Bewertungsarbeit zur Neubewertung von in Deutschland zugelassenen Gasflaschen ermöglichen. Für die Anwendung dieses Gutachtens wird auf die sog. „Flaschen-Negativliste“ der EIGA (Doc. 86/02 Gas cylinders with restricted use in the EU WG-2) und ggf. weitere zukünftig verfügbare Erkenntnisquellen verwiesen.
